Experimental example 1
In the experimental example, the experimental observation of the pharmaceutical composition on normal rats and frequent micturition model rats is studied to prove the therapeutic effect of the health food composition for improving the frequent micturition and urinary incontinence of men.
1. Materials and methods
1.1 animals
Wistar rats with the weight of 350-450g are selected and provided by the university of traditional Chinese medicine in Heilongjiang.
1.2 drugs and dosages
The compositions prepared according to comparative examples 1 to 6 and examples 1 to 6 were administered in a dose of 3 g/kg. Control drug: sunitin (tolterodine tartrate tablet), Nanjing Meirui pharmaceutical Co., Ltd., dosage of 0.3 mg/kg.
2. Method of producing a composite material
2.1 Effect of the compositions prepared in comparative examples 1 to 6 and examples 1 to 6 on urination in a frequent urination model rat
Normal male rats were 120 and randomly averaged into 15 groups: sham, model, control (Senecin), comparative examples 1-6, examples 1-6. Performing abdominal anesthesia with sodium pentobarbital, fixing the back, depilating and sterilizing the lower part, cutting a small opening 2cm long along the center of the lower abdomen with a scalpel, searching for the hypogastric nerve, pushing the descending colon and rectum to one side, abutting against the posterior abdominal wall at the center of the mesentery at the moment, separating the hypogastric nerves at two sides, cutting off (only performing incision operation in a sham operation group, without cutting off the nerves), suturing the wound, and normally feeding for 2 weeks. After 2 weeks, the control group, comparative examples 1 to 6 and examples 1 to 6 were administered by gavage with the same dose, the sham operation group and the model group were administered with the same dose of distilled water, the rats were individually placed in rat metabolism cages 1 hour later, and the 1 st urination time, the number of urination times within 2 hours and the change in urine volume within 2 hours of each group of rats were observed, and the experimental results are shown in table 1.

2.2 Effect of the compositions of examples 1-8 on Normal rat urination
Taking 140 normal male rats, randomly and averagely dividing the rats into 14 groups: blank, control, comparative examples 1-6, examples 1-6. Except for normal feeding, the control group, comparative examples 1 to 6, and examples 1 to 6 were administered by gavage daily at the same dose, and the blank group was administered with the same dose of distilled water for 7 days continuously. Day and night urine volume was observed and recorded in rats before and on days 1, 3 and 7 respectively, and the experimental results are shown in table 2.

3. Conclusion
The data results in table 1 show that the time for the model group to urinate 1 st time is shorter than that of the rats in the sham operation group, and the number of times of urination in 2 hours is increased, and the obvious difference is shown, which indicates that the model building is successful. After the rats take the compositions of the groups 1 to 6 of the comparative examples and the groups 1 to 6 of the examples, the frequency and the urgent micturition degree and the urine volume of the rats are reduced, and the rats have obvious difference compared with the model group; it was further observed that the frequency and volume of the urine were lower in the groups of examples 1-3 than in the groups of comparative examples 1-2, the frequency and volume of the urine were lower in the groups of examples 4-6 than in the groups of comparative examples 3-6, and the frequency and volume of the urine were the lowest in the group of example 6, indicating that the components of the formulation of the present invention coordinate with each other to act positively to achieve the desired therapeutic effect. Wherein:
the frequency and volume of urine in the comparative example 2 group are lower than those in the comparative example 1 group, and the frequency and volume of urine in the comparative example 4 group are lower than those in the comparative example 3 group, so that the combination of pumpkin seeds and pumpkin seeds has stronger inhibition effect on prostatic hyperplasia than that of a single component;
the frequency and the volume of urine in the group of example 3 are lower than those in the group of comparative examples 1-2, and the frequency and the volume of urine in the group of comparative examples 5-6 are lower than those in the group of comparative example 4, which shows that the addition of the diaphragma juglandis or the fructus psoraleae has the effect of promoting the drug effect of the composition;
the frequency and volume of urgency of urination in the example 6 group was lower than those in the comparative examples 5-6, indicating that the efficacy of the composition of the present invention is stronger when diaphragma and psoralea are synergistically present than when they are present separately.
The data results in table 2 show that the urine volume of rats after 3 days and 7 days after the administration of the drug to normal rats is significantly different from that before the administration, further proving that the composition of the invention has certain treatment effect on the frequency and the urgent micturition degree and the urine volume.
Experimental example 2 pharmacodynamic test of efficacy of the pharmaceutical composition of the present invention for improving prostate disease
1. Experimental Material
And (3) testing the sample: the compositions prepared according to comparative examples 1 to 6 and examples 1 to 6 were administered in a dose of 2 g/kg. Molding injection: testosterone propionate. Animal species: kunming mouse. Animal sex and number: male, 122 in total. Animal body weight: 18-22 g.
2. Experimental methods
122 male mice are taken and randomly divided into 14 groups according to the body weight, wherein each group comprises 8-10 male mice, and the groups comprise a blank group, a model group, comparative examples 1-6 groups and examples 1-6 groups. Except for normal feeding, except for the blank control group, the mice of each group were injected with testosterone propionate (5mg/kg) 1 time per day; after 1h, groups 1 to 6 of comparative examples and groups 1 to 6 of examples were each subjected to gavage 1 time, and the blank group and the model group were each subjected to the same volume of distilled water for 30 consecutive days. 24h after the last dose, the mice were sacrificed and the prostate was extracted. Calculation of Prostate Index (PI): prostate weight (mg)/mouse body weight 10. The results of the experiment are shown in Table 3.

3. Conclusion
As can be seen from Table 3, the compositions of comparative examples 1 to 6 and examples 1 to 6 each had an inhibitory effect on prostatic hyperplasia. Further studies have shown that the prostate indices of the groups of examples 1-3 are lower than those of the groups of comparative examples 1-2, the prostate indices of the groups of examples 4-6 are lower than those of the groups of comparative examples 3-6, and the prostate index of the group of example 6 is the lowest, indicating that the components of the formulation of the present invention are mutually coordinated and together play a positive role, achieving the desired therapeutic effect. Wherein:
the prostate index of the group of comparative example 2 is lower than that of the group of comparative example 1, and the prostate index of the group of comparative example 4 is lower than that of the group of comparative example 3, which shows that the combination of pumpkin seeds and pumpkin seeds has stronger inhibition effect on prostate hyperplasia than that of a single component;
the prostate index of the group of example 3 was lower than that of the groups of comparative examples 1-2, and the prostate index of the groups of comparative examples 5-6 was lower than that of the group of comparative example 4, indicating that the addition of diaphragma juglandis or psoralea corylifolia contributes to the efficacy of the composition of the present invention;
the prostate index of the example 6 group was lower than that of the comparative examples 5-6, indicating that the efficacy of rosewood and psoralea was stronger when they were present synergistically in the compositions of the present invention than when they were present separately.
Experimental example 3 clinical study of the pharmaceutical composition of the present invention on male pollakiuria, urgency and urinary incontinence
Frequent micturition and urgent micturition are common diseases, the incidence rate of the diseases is gradually increased with the increase of the age, the pathogenesis is more complex, and an effective treatment method is lacked at present. The frequency and urgency of urination can reach 20-30 times/day without increasing the total amount of urine. The patient feels urgent micturition but does not feel pain, and the daily life of the patient is seriously affected.
1. Clinical data and methods
1.1 clinical data and grouping
60 male patients with frequent and urgent micturition with disease course more than 1 month were selected from 2016 to 9, and randomly divided into 3 groups of 20 persons, namely a control group, an example 3 group and an example 6 group.
1.2 inclusion criteria
Gender is male, the age is 35-55 years, and the number of urination at night is more than 3 on average.
1.3 diagnostic criteria
The traditional Chinese medicine symptoms comprise intolerance of cold, cold limbs, frequent micturition, qi deficiency, weakness, soreness of waist and knees, pale tongue with white fur, and patients with bladder and urethra organic lesions and frequent micturition and urgent micturition caused by tumor, renal failure and the like.
1.4 methods of treatment
The control group adopts conventional western medicine treatment, and racanisodamine tablet is orally taken according to dosage (5 mg/tablet, 2 tablets once, 3 times a day); the composition of example 3 was administered in a dose (25 g/time, three times a day) in the group of example 3, and the composition of example 6 was administered in a dose (25 g/time, three times a day) in the group of example 6.
1.5 Observation index
According to the usual work and rest of the patient, the patient can relieve the urine from getting up to sleep to the next morning, and the urine frequency, the urine volume and the continuous recording are carried out for 7 days. And (6) averaging. The observation is carried out for 1 time respectively before taking medicine, 3 months after taking medicine and after the treatment course is finished.
1.6 Effect assessment
The symptom is completely disappeared to be cured, the symptom is obviously relieved to be effective, the symptom which is relieved but not reach 50 percent is effective, and the symptom which is not relieved or even aggravated is ineffective.

1. A health food composition for improving male pollakisuria and urinary incontinence is characterized by comprising the following raw material components in parts by weight: 5-20 parts of rape pollen, 0.4-5 parts of pumpkin seed oil, 0.4-4 parts of pumpkin seed water extract, 0.0175-0.0702 parts of zinc gluconate, 0.5-3 parts of fructus psoraleae extract and 0.5-3 parts of diaphragma juglandis water extract.
