The baicalein ketone bioactive action substance and polyhexamethylene biguanide combined disinfectant of the embodiment 3 comprises the following components in parts by weight; 2.5 parts of baicalein ketone bioactive action substance; 15 parts of polyhexamethylene biguanide; 10 of chitosan; 40 parts of azone; 1 part of sodium polyacrylate; 931.5 parts of sterile deionized water, wherein the content of the polyhexamethylene biguanide is 99%; the chitosan has a molecular weight of 1000; the azone is medical grade, and the sodium polyacrylate is food grade.
The method for preparing the disinfectant liquid by combining the baicalone bioactive acting substance and the polyhexamethylene biguanide in the embodiment 3 comprises the following steps;
1) preparation of baicalein biologically active substance
Slicing the carefully selected and processed radix Scutellariae, pulverizing, sieving with a 60-mesh sieve to obtain radix Scutellariae powder, adding 2.5 parts by weight of radix Scutellariae powder into 8 times of deionized water, stirring, heating to 80 deg.C, performing microwave treatment for 4min, leaching for 1h, filtering, and separating to obtain first leaching solution; adding deionized water with the weight 10 times of that of the baical skullcap root powder into the residue, leaching for 1h at 85 ℃, and filtering and separating to obtain a second leaching solution; mixing the two leaching solutions, adding hydrochloric acid to adjust pH to 1-2, keeping the temperature at 80 deg.C for 0.5h, standing and precipitating for 12 h. Separating the supernatant, recycling purified water in a purification system, adding deionized water with the weight of the supernatant into the precipitate, stirring and suspending, adjusting the pH to 7 by using 40% sodium hydroxide, adding 95% ethanol with the same amount, stirring and dissolving, filtering by using a 100-mesh sieve, heating to 60 ℃, adjusting the pH to 1-2 by using hydrochloric acid, preserving heat for 0.5h to fully separate out the precipitate, washing the precipitate to the pH to 7 by using ethanol, separating ethanol water washing liquid, returning the ethanol and water to the purification system for recycling, and drying the pasty precipitate in a vacuum manner at 65 ℃ until the precipitate is constant weight and crushed to obtain the baicalone bioactive action substance.
2) Preparation of solution a
Weighing 631.5 parts of sterile deionized water, adding 15 parts of polyhexamethylene biguanide, heating until the polyhexamethylene biguanide is dissolved, slowly adding 2.5 parts of baicalein ketone bioactive substance when the temperature reaches 90 ℃, stirring, performing microwave treatment for 4min, and adjusting the pH value to 6-7 with citric acid after the baicalein ketone bioactive substance is completely dissolved to obtain solution a;
3) preparation of solution b
Weighing 200 parts of deionized water, diluting and dissolving 10 parts of chitosan to obtain solution b;
4) preparation of solution c
Weighing 100 parts by weight of sterile deionized water, diluting and dissolving 1 part by weight of sodium polyacrylate to obtain solution c;
5) preparation of baicalein bioactive substance and polyhexamethylene biguanide combined disinfectant
Adding azone 40 parts by weight into the solution a prepared in the step 2) at room temperature, and sequentially adding the solution b prepared in the step 3) and the solution c prepared in the step 4) to mix uniformly, thus obtaining a finished product of the baicalein bioactive action substance and the polyhexamethylene biguanide combined disinfectant.
Experimental cases;
(1) through hemorrhagic trauma caused by pedicure of toenails by experimenters, red, swollen and radioactive pain of toes, red and swollen foot surfaces and dare-proof feet, after the secondary spraying treatment of the baical skullcap root ketone bioactive substance and the polyhexamethylene biguanide combined disinfectant for 4 hours, the red and swollen wounds disappear, scabs and the whole feet recover to be normal after 24 hours;
(2) after the experimenter performs spraying treatment on the baicalein ketone bioactive substance and the polyhexamethylene biguanide combined disinfectant for three times at intervals of 2 hours, the toothache feeling gradually disappears after 10 minutes for the first time, and the redness and swelling of the gum disappear after 24 hours.
The experimental cases prove that after the disease-preventing disinfectant liquid containing the baicalone bioactive substance is sprayed on an affected part, the bactericidal effect of the baicalone bioactive substance and the disinfectant on externally damaged skin pathogenic bacteria is good under the synergistic effect. Meanwhile, the gene cell regulation and control effect of the active substance of the main bioactive component of the baicalein is verified. The effects of disinfection, sterilization and disease prevention are realized.
Referring to fig. 1, the content of baicalein in the baicalone bioactive acting substance and polyhexamethylene biguanide combined disinfectant of the present invention was measured by high performance liquid chromatography to be 22.02 mg/l.
Referring to fig. 2, the content of baicalin in the combined disinfectant of biologically active action substance of baicalone and polyhexamethylene biguanide of the present invention was measured to be 29.72683 × 50 ═ 1486.34mg/l by hplc.
Referring to fig. 3, the standard working curve of baicalin drawn by the hplc shows that the ratio of baicalin: 10. 25, 50, 100 and 200 mg/l.
Referring to fig. 4, the standard work curve of baicalein drawn by the high performance liquid chromatograph shows that the content of baicalein: 5. 12.5, 25, 50 and 100 mg/l.
The test result of the baicalein ketone bioactive action substance and polyhexamethylene biguanide combined disinfectant in 2016, 10 and 31 days by the northern China university medical college according to the quantitative sterilization test method of the Ministry of public health of the people's republic of China (2002 edition) is as follows: see table 1.

The detection result shows that: the baicalein bioactive action substance and polyhexamethylene biguanide combined disinfectant is superior to the national standard escherichia coli (ATCC8099), staphylococcus aureus (ATCC6538), pseudomonas aeruginosa (ATCC15442) and candida albicans (ATCC10231) of the biguanide disinfectant in the prior art; the killing rate logarithmic value of the carrier method is more than or equal to 3; the suspension method has the advantages that the sterilization logarithm value of other sterilization logarithm values are more than or equal to 5 except that the sterilization logarithm value of the candida albicans is more than or equal to 4, and the defect that the polyhexamethylene biguanide kills bacillus subtilis spores is overcome; the baicalein and baicalin which are main components of the baicalein and baicalin of the baicalone biological active matter can perform gene cell regulation and control on cell disorder, virus generation and oxygen radical formation under the action concentration of effective active matters, particularly can perform cell regulation and control under gene control on people in a severe disease pathological change stage, and has an important effect on prevention and control of severe diseases.

Claims (3)

1. A baicalein bioactive action substance and polyhexamethylene biguanide combined disinfectant is characterized by comprising the following components in parts by weight; 1.5-2.5 parts of baicalein biologically active substance; 5-15 parts of polyhexamethylene biguanide; 10 parts of chitosan; 25-60 parts of azone; 1 part of sodium polyacrylate; 957.5-911.5 parts of sterile deionized water; the polyhexamethylene biguanide content is 99%; the chitosan has a molecular weight of 1000; the azone is of medical grade, and the sodium polyacrylate is of food grade.
2. The combined disinfectant of the baicalone bioactive substrate and the polyhexamethylene biguanide as claimed in claim 1, which is characterized by comprising the following components in parts by weight; 2 parts of baicalein biologically active substance; 10 parts of polyhexamethylene biguanide; 10 parts of chitosan; 25 parts of azone; 1 part of sodium polyacrylate; 952 parts of sterile deionized water; the polyhexamethylene biguanide content is 99%; the molecular weight of the chitosan is 1000; the azone is of medical grade, and the sodium polyacrylate is of food grade.
3. The disinfectant liquid as claimed in claim 2, wherein the preparation method comprises the following steps;
1) preparation of baicalein biologically active substance
Slicing and pulverizing processed Scutellariae radix, sieving with 60 mesh sieve to obtain Scutellariae radix powder, adding 1 part of Scutellariae radix powder into deionized water 8 times of Scutellariae radix powder, stirring, placing in microwave heater, heating at 80 deg.C for 4min, leaching for 1 hr, and filtering with 80 mesh sieve to obtain first leaching solution; adding deionized water 10 times the weight of the Scutellariae radix powder into the residue, placing in a microwave heater, leaching at 85 deg.C for 1 hr, and filtering with 100 mesh sieve for separation to obtain second leaching solution; mixing the waste residues, mixing the secondary leaching liquor, adding hydrochloric acid to adjust pH to 1-2, keeping the temperature at 85 ℃ for 0.5h, standing and precipitating for 12h, separating supernatant, recycling purified water in a purification system, adding deionized water for separating the weight of the supernatant into the precipitate, stirring and suspending, adding 40% sodium hydroxide for adjusting pH to 7, adding deionized water for separating the weight of the supernatant into the precipitate, adding 40% sodium hydroxide for adjusting pH to 7, adding 95% ethanol with equivalent concentration, stirring to dissolve, filtering through a 100-mesh sieve, placing in a microwave heater, heating to 60 deg.C, adjusting pH to 1-2 with hydrochloric acid, maintaining the temperature for 0.5h to fully separate out precipitate, washing the precipitate with ethanol to pH7, separating ethanol water washing solution, sending back to purification system for treating ethanol and water for recycling, vacuum drying the pasty precipitate at 65 deg.C to constant weight, and pulverizing to obtain bioactive active substance of baicalein;
2) preparation of solution a
Weighing 652 parts by weight of sterile deionized water, adding 10 parts by weight of polyhexamethylene biguanide, heating until the polyhexamethylene biguanide is dissolved, slowly adding 2 parts by weight of baicalein ketone bioactive substance when the temperature reaches 80 ℃, stirring, performing microwave treatment for 2min, and adjusting the pH value of 6-7 by using citric acid after the baicalein ketone bioactive substance is completely dissolved to obtain solution a;
3) preparation of solution b
Weighing 200 parts of deionized water, diluting and dissolving 10 parts of chitosan to obtain solution b;
4) preparation of solution c
Weighing 100 parts by weight of sterile deionized water, diluting and dissolving 1 part by weight of sodium polyacrylate to obtain solution c;
5) preparation of baicalein bioactive substance and polyhexamethylene biguanide combined disinfectant
Adding 25 parts by weight of azone into the solution a prepared in the step 2) at room temperature, and sequentially adding the solution b prepared in the step 3) and the solution c prepared in the step 4) for uniformly mixing to obtain a finished product of the baicalein bioactive action substance and the polyhexamethylene biguanide combined disinfectant.
