Abstract
抗微生物组合物包含至少一种或多种水中的有机酸或盐,一种或多种水溶性有限的抗微生物剂。所述组合物含有高活性含量并长期稳定。
Description
技术领域
本发明涉及由GRAS物质组成的稳定组合物,用于微生物控制,尤其是在发酵过程中。
背景技术
众所周知,有机羧酸具有抗微生物特性。近期工作显示有机羧酸与其它类型抗微生物剂如啤酒花酸之间有协同效应。例如,美国申请2004/0175480描述若与食品级酸如乳酸、乙酸、丙酸、柠檬酸,钾离子源和抗氧化剂联合,啤酒花β酸作为抗微生物剂更有效。
抗微生物有机酸一般可以高浓度溶于水,但一些有机酸抗微生物剂不溶于水或在电解质存在下盐析。例如,已知啤酒花酸在水中的溶解度有限,现有技术中所述的啤酒花酸的抗微生物组合物活性低,即使采用有机溶剂如乙二醇或悬浮剂。
一种组合不同抗微生物剂的方法是将液体抗微生物剂喷到粉状有机酸或盐上,但所得组合物不易分散于水。尽管能将湿润剂和/或乳化剂配制到固体组合物中,但液体产品有高活性含量对于处理和使用则更为方便。从可持续性的角度而言,具有高活性含量的产品还节省加工、运输和保存费用。
已知表面活性剂可帮助疏水材料溶于水。表面活性剂,如美国专利5374614、7655613和7846889所述的那些,用于从水溶性和非水溶性成分形成微乳液,即热力学稳定的各向同性液体。这类含有机酸的微乳液组合物还描述于美国专利5294364、6251844、7030070和7642227,作为酸性清洁液体。有机酸含量低且所用表面活性剂可能禁止应用于食品或接触食物的物品。
需要有具备全部协同抗微生物组分的单一产品,采用稳定浓缩形式,水作为溶剂。更优选所述抗微生物产物在可燃性、腐蚀性、毒性和法规要求方面顾虑小。
发明详述
本发明提供水性抗微生物组合物,包含:(i)水溶性有机酸或其盐,(ii)水溶性有限的抗微生物剂和(iii)表面活性剂。
本发明提供无害稳定的水性抗微生物组合物,以高活性含量,优选大于10%,更优选大于20%,最优选大于30%的组合物总重量,包含水溶性有机酸和活性水溶性有限的抗微生物剂活性。所述组合物用批准用于食品和饮料的GRAS(公认安全)成分配制,且所述组合物在可燃性和腐蚀性方面归类为非有害物质。该组合物作为均质液体在室温或以下稳定超过1个月,优选超过3个月,最优选超过6个月。
本发明所述组合物设计成作为抗微生物剂,优选降低或控制发酵过程中不需要的微生物的浓度。这种类似组合物还能用于其它目的,如用于抗寄生虫药或用作浓缩清洁、除锈和消毒剂。所述组合物活性组分包含一种或多种有机酸或其盐,和一种或多种水溶性有限的抗微生物剂,总活性含量(有机酸加抗微生物剂)大于5%重量,优选大于15%,最优选大于25%的组合物总重量。该组合物还包含一种或多种表面活性剂增溶剂。这些包括但不限于阴离子表面活性剂、非离子表面活性剂、两性表面活性剂、两性离子表面活性剂和阳离子表面活性剂。也能加入其它成分如中和剂、溶剂以增加总活性含量,而不造成组合物不稳定。还可加入功能助剂,如螯合剂、多价螯合剂、阻蚀剂、酶、表面改性剂、染料和芳香剂。
优选用于本发明的水溶性有机酸选自水溶性单羧酸和聚羧酸,其在分子中有2-8个碳原子,且任选地,由一个或多个羟基取代。合适的类别是烷酸、羟基烷酸、烷基聚羧酸和羟烷基聚羧酸。本文优选涉及第一解离阶段的pKa值小于约6的单羧酸和聚羧酸。这些包括例如乙酸、己二酸、苯磺酸、苯甲酸、柠檬酸、葡萄糖酸、戊二酸、羟基乙酸、乳酸、苹果酸、甲烷磺酸、草酸、丙酸、水杨酸、琥珀酸、酒石酸,和其混合物以及其盐。优选的酸是柠檬酸或其盐。出于本发明目的,啤酒花酸不是有机酸。
水溶性有机酸含量为总组合物重量的5-70%、5-60%、5-50%,优选15-60%,最优选25-50%。
合适的溶解度有限的抗微生物剂包括啤酒花酸或乳酸链球菌肽。本文所用的术语“啤酒花酸”包括啤酒花β酸、啤酒花α酸,其异构化或衍生产物和其盐。其还包括啤酒花提取物,如US 5286506所述的那些。α和β啤酒花酸的化学名称(chemical identity)包括类葎草酮(cohumulone)、律草酮(humulone)、伴葎草酮(adhumulone)、类蛇麻酮(colupulone)、蛇麻酮(lupulone)和伴蛇麻酮(adlupulone)。这些都纳入本文所用的术语“啤酒花酸”。啤酒花酸能通过从天然啤酒花提取和纯化或者化学合成来制备。优选的抗微生物剂是啤酒花酸。
疏水抗微生物组分,优选啤酒花酸,的活性含量为总组合物重量的0.05-15%,优选0.1-10%,最优选0.5-5%。
用于低水溶性抗微生物组分的表面活性增溶或乳化剂包括非离子、阴离子、阳离子、两性和两性离子表面活性剂。美国专利7846889、7030070、6251844和5294364所述的表面活性剂通过引用纳入本文。优选的表面活性剂是批准用作食品和药物成分或用于在可能接触食品的物品中的那些。这些表面活性剂包括聚山梨醇酯(聚氧乙烯脱水山梨醇脂肪酸酯)如吐温(禾大公司(Croda Inc)),磺基琥珀酸单烷基酯或磺基琥珀酸二烷基酯如Aerosol OT(氰特公司(Cytec)),烷基聚葡萄糖苷如Glucopons(巴斯夫(BASF))和硫酸烷基酯。用于本发明的优选表面活性剂包括聚氧乙烯(20)脱水山梨醇单油酸酯和烷基聚葡萄糖苷。
所述组合物可包含一种或多种来自单一组或不同组的表面活性剂。组合物中一种或多种表面活性剂含量为总组合物重量的0.1-30%,优选1-20%,最优选2-15%。
还能使用有限量的有机溶剂或助表面活性剂,从而可减少表面活性剂的量。示例是脂肪醇,如甲醇、乙醇、丙醇、丁醇、乙二醇的二醇或多元醇、丙二醇、甘油;脂肪酸或二酸如丁酸、己二酸、辛酸。优选的助表面活性剂包括乙醇、丙二醇和己二酸。
所述组合物可包含来自单一组或不同类型的一种或多种助表面活性剂或有机溶剂。组合物中一种或多种助表面活性剂或有机溶剂含量为组合物重量的0.1-30%,优选1-20%,最优选2-10%。
有机酸能用碱金属或碱土金属氢氧化物、氢氧化铵或有机胺总体或部分中和。示例是氢氧化钠、氢氧化钾和氨水。中和度取决于有机酸类型和量。通常,组合物应中和到pH高于2以减少有机酸腐蚀性,但不超过pH 4以保留可能的最高活性含量。然而,为了获得组合物最高稳定性,优选中和到碱性pH。
所述组合物优选配制成微乳液,即含表面活性剂的油和水的热力学稳定的各向同性液体混合物,优选有助表面活性剂或溶剂。有机酸首先溶于水并中和到所需pH,然后与至少一种表面活性剂、至少一种助表面活性和至少一种水溶解性有限的抗微生物剂混合。优选疏水抗微生物剂首先溶于至少一种表面活性剂和至少一种助表面活性或至少一种溶剂以制备预混合物,其随后用中和的有机酸水性溶液稀释。
优选抗微生物组合物的所有组分被FDA批准为食品配料成分或授予GRAS(公认安全)状态。还优选组合物在腐蚀性和可燃性方面无害。
所述组合物提供有力的、非抗生素的抗微生物处理。该组合物能用于在水性体系中减少不需要的微生物浓度,促进所需微生物繁殖,并提高所需微生物效率。
在前述方法中,意在减少的“不需要的”微生物是与促进所需发酵过程的想要的微生物竞争营养物的那些。发酵中不想要或不需要的微生物包括产乳酸细菌(LAB)和产醋酸细菌,其中乳杆菌(Lactobacillus)和醋杆菌(Acetobacter)是主要代表。竞争可发酵物,不让其进入预期发酵微生物并因而降低产率的任何微生物可视作不需要。在此方面,用于本方法的啤酒花酸提取物和有机酸不对所需要的、促进发酵的微生物的生长和活力造成不利影响,但确实消除或抑制干扰发酵过程的不需要微生物生长。此外,消除或抑制不需要的微生物对所需微生物生长和活力产生有利影响。
控制发酵过程所用水性溶液中不需要的微生物浓度的方法包括以下步骤:向水性溶液引入可发酵的碳水化合物,向所述水性溶液引入至少一种酵母,和向所述水性溶液引入组合物,所述组合物含有(a)水溶性有机酸或其盐,(b)水溶性有限的抗微生物剂和(c)表面活性剂。
向发酵体系加入组合物以控制不需要的微生物的浓度,从而发酵体系中有机酸的量为12500ppm下至100ppm,6250下至100ppm,或4000下至100ppm,或4000下至200ppm。一般,使用至少100ppm或至少200ppm或至少300ppm有机酸。向发酵体系加入组合物,从而发酵体系中水溶性有限的抗微生物剂的量为0.5ppm-200ppm,0.5ppm-150ppm,或2-150ppm,或2-100ppm。一般,所用啤酒花酸的量是至少2ppm或至少5ppm。
实施例
实施例1
将10.82份吐温80(聚氧乙烯(20)脱水山梨醇单油酸酯),3.74份无水乙醇和3.15份啤酒花酸钾盐(30%活性酸),以所述顺序混合,产生均质液体。在单独容器中,将34.49份无水柠檬酸溶于34.49份去矿物质水,随后通过加入6.02份28-30%氨含量的氢氧化铵来中和冷溶液。然后,向含啤酒花酸的有机相加入所述部分中和的柠檬酸,产生约35%活性组合的抗微生物剂的各向同性液体。发现组合物在4℃稳定而没有柠檬酸盐结晶,并在50℃保持均匀1个月。用所述配制物作为抗微生物剂没有观察到任何负面影响。

实施例2
应用与实施例1相同的过程,用表1所列的参数(charge)以产生38%活性组合的活性抗微生物剂的组合物。发现组合物在4℃和室温稳定而没有柠檬酸盐结晶,并在50℃保持均匀37天(表1)。再次,用该配制物没有观察到抗微生物特性方面的任何不良反应。
实施例3-11
3种GRAS状态表面活性剂即吐温80、Aerosol OT-70PG(氰特公司)和Triton BG-10(美国陶氏(Dow))或Glucopon 215UP(巴斯夫)的烷基聚葡萄糖苷,在这些实施例中根据表2所列参数配制。柠檬酸首先溶于水,随后用氨水部分中和。表面活性剂吐温80、Aerosol OT-70PG、Triton BG-10或Glucopon 215UP如所列加入,接着是啤酒花酸和乙醇。充分混合后,通常获得均质液体,保存期如表2所列。

实施例12-13
在这些实施例中,氢氧化钾用作中和剂。遵循与实施例3-10相同的过程,用表3所列重量参数。抗微生物剂的活性含量增加到40%以上,但保存期有限。认为不稳定由酸倾向性表面活性增溶剂导致。
实施例14-16
在这些实施例中,为了高稳定性用氢氧化钾中和酸,趋向碱性pH。也使用不燃性丙烯和/或己二酸的替代性助表面活性剂,如这些组合物的表4所示。遵循与实施例3-10相同的过程,用表4所列重量参数。预期所用表面活性剂无水解,在4、22和50℃老化大于6周后没有观察到啤酒花酸不稳定且变色很少。

实施例21:柠檬酸/啤酒花酸组合物的MIC测定
在以下实施例中,用于测量抗微生物活性水平的终点被称为最低抑菌浓度或MIC。这是能实现完全生长抑制的一种或多种物质最低浓度。
5毫升生长培养基接种有合适的测试微生物,振荡孵育过夜。这些过夜培养物(ON)1:100稀释(10mL PBS中100μl ON)。采用康宁(Corning)96孔微量滴定板并如下准备:分配252μl培养基到列1和140μl到列2-12。其次,28μl配制物分配到列1以实现1:10稀释和待测试的最高组合物浓度。列1通过移液管上下抽吸来混合,完成整个板的2倍连续稀释(每次转140μl培养基+组合物)。列12用作无组合物的对照,因此,取出来自列11的140μl培养基+组合物以废弃。最后,向各孔加入5μl细菌,产生~5x 105cfu/ml。96孔板用石蜡膜覆盖、密封并在合适温度孵育18-24小时。根据目测检查混浊孔将孔定为阳性或阴性,浊度作为生长指示物。完全抑制生长(如清澈孔)的最低抗微生物剂浓度指定为最低抑菌浓度。

Claims (20)
1.一种水性抗微生物组合物,包含
(i)至少一种水溶性有机酸或其盐,
(ii)至少一种水溶性有限的抗微生物剂,
(iii)至少一种表面活性剂,
其中所述
(a)水溶性有机酸或其盐包含组合物总重量的5-70%,
(b)水溶性有限的抗微生物剂包含组合物总重量的0.05-15%,
(c)表面活性剂包含组合物总重量的0.1-30%。
2.如权利要求1所述的抗微生物组合物,其中所述有机酸包括单羧酸和聚羧酸或其盐,所述单羧酸和聚羧酸涉及第一解离阶段的pKa值小于约6。
3.如权利要求1或2所述的抗微生物组合物,其中所述有机酸选自乙酸、己二酸、苯磺酸、苯甲酸、柠檬酸、葡萄糖酸、戊二酸、羟基乙酸、乳酸、苹果酸、甲烷磺酸、草酸、丙酸、水杨酸、琥珀酸、酒石酸,和其混合物以及其盐。
4.如权利要求1-3中任一项所述的抗微生物组合物,其中所述抗微生物剂选自啤酒花酸、乳酸链球菌肽或其组合。
5.如权利要求1-3中任一项所述的抗微生物组合物,其中所述抗微生物剂包含啤酒花酸。
6.如权利要求1-5中任一项所述的抗微生物组合物,还包含
(iv)根据组合物总重量,0-20%重量的助表面活性剂或溶剂。
7.如权利要求1-6中任一项所述的抗微生物组合物,还包含中和剂。
8.如权利要求1-7中任一项所述的抗微生物组合物,其中所述有机酸包含柠檬酸或其盐。
9.如权利要求1-8中任一项所述的抗微生物组合物,其中所述抗微生物剂的重量是组合物重量的0.1-10%或0.5-5%。
10.如权利要求1-9中任一项所述的抗微生物组合物,其中所述表面活性剂重量是组合物重量的1-20%,优选2-15%。
11.如权利要求1-10中任一项所述的抗微生物组合物,其中至少一种表面活性剂选自聚山梨醇酯(聚氧乙烯脱水山梨醇脂肪酸酯)、磺基琥珀酸单烷基酯或磺基琥珀酸二烷基酯、烷基聚葡萄糖苷和硫酸烷基酯或其组合。
12.如权利要求1-10中任一项所述的抗微生物组合物,其中至少一种表面活性剂选自聚氧乙烯(20)脱水山梨醇单油酸酯和烷基聚葡萄糖苷。
13.如权利要求1-12中任一项所述的抗微生物组合物,其中所述至少的有机酸的量是5-50%。
14.如权利要求1-12中任一项所述的抗微生物组合物,其中所述至少的酸的量是15-60%。
15.如权利要求1-12中任一项所述的抗微生物组合物,其中所述至少的酸的量是组合物重量的25-50%。
16.一种水性抗微生物组合物,包含
(i)柠檬酸或其盐,
(ii)选自啤酒花酸和乳酸链球菌肽的至少一种水溶性有限的抗微生物剂,
(iii)至少一种表面活性剂,
其中所述
(a)柠檬酸或其盐包含组合物总重量的5-50%,
(b)水溶性有限的抗微生物剂包含组合物总重量的0.1-10%,
(c)表面活性剂包含组合物总重量的1-30%。
17.如权利要求16所述的水性抗微生物组合物,其中所述水溶性有限的抗微生物剂包含乳酸链球菌肽。
18.如权利要求17所述的水性抗微生物组合物,其中所述水溶性有限的抗微生物剂包含啤酒花酸。
19.一种控制发酵过程所用水性溶液中不需要的微生物的浓度的方法,所述方法包括步骤:
(a)向水性溶液引入可发酵的碳水化合物;
(b)向所述溶液引入至少一种酵母;和
(c)向所述水性溶液引入权利要求1-18中任一项所述的组合物。
20.如权利要求18所述的方法,其中所述发酵体系中的有机酸为12500ppm下至100ppm,6250下至100ppm,或4000下至100ppm,或4000下至200ppm,且所述发酵体系中水溶性有限的抗微生物剂的量为0.5ppm-200ppm,或0.5ppm-150ppm,或1-150ppm,或2-100ppm,优选柠檬酸是6250下至100ppm且水溶性有限的抗微生物剂是1-150ppm。
