Abstract
本发明公开了一种具有祛痘美白功效的中药组合提取物,该组合提取物由下述按重量份计的中药原料制备而成:黑果枸杞叶10‑40份、三七叶10‑30份、余甘子10‑40份、藏荆芥10‑30份、变叶海棠叶10‑30份。本发明的中药组合提取物,具有祛痘美白、天然安全、协同增效的特点。

技术领域
本发明属于精细化工日用品技术领域,具体涉及的是一种具有祛痘美白功效的中药组合提取物及应用。
背景技术
皮肤是人类防御系统的第一道屏障,若人体自身的生理作用分泌的皮脂、汗液或者残留在皮肤表面的化妆品以及灰尘得不到及时的清除,会导致毛囊或皮脂腺阻塞、发炎,进而在皮肤表面出现痤疮等。若不及时处理,痤疮不断反复,加之黑色素的沉积,产生色斑,最终导致一系列皮肤病。
导致黑色素产生的原因有很多种,如紫外线废气、活性氧等环境问题和某些遗传因素,其产生的原因相当复杂,普遍研究结果表明黑色素合成的过程如下,以细胞内的酪氨酸作为基质,在酪氨酸酶的作用下生成多巴醌,从多巴醌变成多巴色素之后进一步氧化成5,6-二羟基吲哚和5,6-二羟基-2-羟酸,这些生成物聚合最终生成聚体黑色素。在细胞代谢的层层推动下被转移至肌肤表层,若不能及时被代谢排出体外,就会逐渐累积沉淀形成色斑,最终导致皮肤过黑。而痤疮的产生是由于毛囊堵塞,油脂分泌受阻,加之细菌的作用,形成痤疮,导致皮肤质感和活性下降。
目前研究中,皮肤美白剂通常都是以酪氨酸酶作为作用靶点,通过抑制酪氨酸酶的活性或是阻断酪氨酸氧化生成黑色素的反应来减少皮肤黑色素的生成,从而达到皮肤美白的效果。大多数采用的是过氧化氢、氯化氨基汞和各种酚类衍生物作为皮肤美白剂,这些化合物可以迅速的瓦解黑色素组织达到快速美白的效果,但对皮肤具有腐蚀性、细胞毒性和过敏性等不良影响,安全性较差。对于祛痘,目前大多数还停留在涂抹药膏、配合美容美白面膜使用等的阶段,安全性较差、对皮肤刺激性较强,且实际效果也不太理想。
本发明将五种天然植物提取物组合使用,既能够达到美白祛痘的作用,还能大大降低对皮肤的刺激,提升美白祛痘过程的安全性。
藏荆芥为唇形科植物藏荆芥Nepeta angustifolia C.Y.Wu的全草。其产于西藏(江孜、拉萨),是西藏特有的藏药材。藏荆芥在临床上主要用于神昏痉厥、中风、癫痫、脑溢血、疮伤及疼痛等症的治疗,具有良好的疗效。《晶珠本草》指出:本品“治癫痫,脑溢血,疮伤及疼痛等症”,《藏药志》、《药名荟萃》也有藏荆芥治疗中风、邪病、胸膜炎及皮肤病的记载。现代研究发现,藏荆芥能够发挥消肿止痛、创伤愈合等作用。此外,藏荆芥挥发油的一些组分具有抗氧化、抗炎、镇痛的作用。
变叶海棠叶也称俄色叶,为甘孜藏族自治州藏族民间的药食两用的药材,来源于蔷薇科植物变叶海棠Malus toringoides(Rehd.)Hughes的干燥叶。最早记载于《藏药晶镜本草》和《藏汉大辞典》,据这两本医书记载,该药具有攻坚化积、除腻漆滞、保肝利胆等功效,它的神奇药用功效被录入了最具权威的藏医学著作《四部医典》。对于变叶海棠叶的使用,传统的藏医用法为与酥油同煮口服。主要用于治疗消化不良及一些肝疾,其疗效确切,用法独特。现代药理研究表明,变叶海棠叶中含较高的总黄酮类化合物,其对DPPH自由基和羟基自由基有较好的清除活性,且变叶海棠叶中化学成分根皮苷、根皮素具有抗动脉粥样硬化、抗炎等药理作用。
黑果枸杞叶为茄科枸杞属植物黑果枸杞Lycium Ruthenicum Murr.的茎叶。目前,黑果枸杞的研究多集中于其果实所含色素和多糖的提取和生物活性方面,而对其叶片提取物对美容美白未见报道。现代研究发现,黑果枸杞叶中总黄酮对清除小鼠血清羟基自由基的影响、对红细胞溶血的保护作用以及抑制小鼠肝脏MDA产生方面都具有显著功效,表现出明显的抗氧化活性。本发明研究人员发现黑枸杞叶提取物对酪氨酸酶有抑制作用,表现出一定的美白功效。
三七叶为五加科人参属植物三七PanaxNotoginseng(Burk.)F.H.Chen的茎叶。经研究报道,三七叶中富含大量的黄酮苷类、多糖类及三七叶总皂苷,具有抗炎、抗氧化、解痉、抗癌、抗动脉粥样硬化等作用。动物实验表明,三七叶总皂苷能够抑制血液和脑组织过氧化脂质(LOP)生成和提高超氧化物歧化酶(SOD)活性,对于增加皮肤活力和延缓衰老有较好的作用。此外,三七茎叶提取物能够有效清除氧自由基,对皮肤有较好的抗氧化作用。
余甘子为大戟科植物余甘子Phyllanthus emblica L.的果实,明《本草纲目》记载:“余甘子果,主补益气,久服轻身,延年益寿”。可用于防治脂肪肝、糖尿病、高血脂、高血压、肥胖、癌症,可增强人体的免疫力,从根本上提高人体脂类的分解能力而达到平衡代谢的目的。动物实验研究发现,余甘子提取液具有抗氧化作用和清除氧自由基能力,可提高红细胞超氧化物歧化酶SOD活性、降低血清中过氧化脂质LOP含量。另外,有研究表明余甘子提取物对引起皮炎的真菌有较强的抑制作用,对祛痘有一定的疗效。
发明内容
本发明的目的是克服现有技术的不足,提供一种具有祛痘美白、天然安全、协同增效的中药组合提取物以及应用。
为了实现上述发明目的,本发明的技术方案如下:
一种具有祛痘美白功效的中药组合提取物,该组合提取物由下述按重量份计的中药原料制备而成:黑果枸杞叶10-40份、三七叶10-30份、余甘子10-40份、藏荆芥10-30份、变叶海棠叶10-30份。
优选地,该中药复方提取物由下述按质量份数计的中药原料制备而成:黑果枸杞叶20-40份、三七叶10-20份、余甘子20-40份、藏荆芥10-20份、变叶海棠叶10-20份。
所述具有祛痘美白功效的中药组合提取物的制备方法,包括以下步骤:
取干燥的黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶药材,粉碎,过20目筛,按照体积质量比加入5-8倍量(V/m)50%-80%乙醇,回流提取1-3次,每次提取1-3小时,滤过,滤液合并,在55-75℃条件下减压回收乙醇至无醇味,再上D101型或AB-8型大孔树脂柱纯化,收集40%-70%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,50-70℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
上述的一种具有祛痘美白功效的中药组合提取物在制备护肤品中的应用。
所述具有祛痘美白功效的中药组合提取物在祛痘美白护肤品中的质量百分含量为0.1-10%,优选质量百分含量为1-8%,其余为化妆品外用剂型的常用基质以及去离子水或纯化水或蒸馏水。通过常规工艺,可制成祛痘美白洗面奶、祛痘美白洗面乳、祛痘美白洗面霜、祛痘美白膏、祛痘美白霜、祛痘美白乳液、祛痘美白水、祛痘美白面膜、祛痘美白凝胶或祛痘美白皂等化妆品外用剂型。
本发明的解决方案是基于相关的医学传统理论,结合现代医药学研究技术与手段,通过大量实验优选与验证,发现了黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶等五味药材配合使用,可起到明显的祛痘美白护肤作用。本发明所优选配方,通过国内外文献检索,未发现相关应用,加之通过大量实验筛选,所得配方比例为最佳配方比例,配方科学,且安全性高,可用于日常皮肤护理。
本发明具有以下优点:
(1)发现了黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶配合使用的祛痘美白作用,扩大了上述原料的应用范围,特别是在皮肤护理中的应用;
(2)通过大量的实验筛选,确定了原料的最佳比例范围,确保本发明产品使用的效果;
(3)本发明配方效果明确且稳定,有效克服了天然产物护肤品效果不稳定,也克服了单纯化学成分安全系数较低等缺点。
具体实施方式
下面结合具体实施例进一步说明本发明的技术解决方案,这些实施例不能理解为是对技术方案的限制。
实施例1
黑果枸杞叶30份、三七叶10份、余甘子40份、藏荆芥10份、变叶海棠叶10份
取干燥的黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶药材,粉碎,过20目筛,按照体积质量比加入8倍量(V/m)60%乙醇,回流提取2次,每次提取2小时,滤过,滤液合并,在65℃条件下减压回收乙醇至无醇味,再上AB-8型大孔树脂柱纯化,收集50%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,70℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
实施例2
黑果枸杞叶30份、三七叶20份、余甘子30份、藏荆芥10份、变叶海棠叶10份
取干燥的黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶药材,粉碎,过20目筛,按照体积质量比加入5倍量(V/m)80%乙醇,回流提取3次,每次提取1小时,滤过,滤液合并,在55℃条件下减压回收乙醇至无醇味,再上D101型大孔树脂柱纯化,收集70%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,60℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
实施例3
黑果枸杞叶40份、三七叶20份、余甘子20份、藏荆芥10份、变叶海棠叶10份
取干燥的黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶药材,粉碎,过20目筛,按照体积质量比加入8倍量(V/m)50%乙醇,回流提取1次,每次提取3小时,滤过,滤液合并,在75℃条件下减压回收乙醇至无醇味,再上AB-8型大孔树脂柱纯化,收集60%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,55℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
实施例4
黑果枸杞叶20份、三七叶10份、余甘子40份、藏荆芥20份、变叶海棠叶10份
取干燥的黑果枸杞叶、三七叶、余甘子、藏荆芥、变叶海棠叶药材,粉碎,过20目筛,按照体积质量比加入7倍量(V/m)70%乙醇,回流提取3次,每次提取1.5小时,滤过,滤液合并,在70℃条件下减压回收乙醇至无醇味,再上D101型大孔树脂柱纯化,收集40%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,65℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
实施例5
黑果枸杞叶25份、三七叶15份、余甘子35份、藏荆芥15份、变叶海棠叶10份
取干燥的黑果枸杞叶、三七叶、藏荆芥、变叶海棠叶、独一味药材,粉碎,过20目筛,按照体积质量比加入8倍量(V/m)60%乙醇,回流提取2次,每次提取2小时,滤过,滤液合并,在60℃条件下减压回收乙醇至无醇味,再上AB-8型大孔树脂柱纯化,收集60%乙醇洗脱部分,60℃条件下减压回收乙醇,得浸膏,70℃条件下真空干燥,粉碎,即为本发明具有祛痘美白功效的中药组合提取物。
实施例6

1材料与方法:
1.1样品
实施例1提取物、实施例2提取物、实施例3提取物、熊果苷(阳性对照)。
1.2实验方法
(1)磷酸缓冲液的配制:精密称取Na2HPO4·12H2O 26.797g,NaH2PO4·2H2O13.803g,分别用去离子水溶解后,定容至500mL容量瓶中。取磷酸二氢钠和磷酸氢二钠溶液各100mL至500mL容量瓶中,去离子水定容,摇匀,即为pH=6.8的磷酸缓冲液。
(2)样品溶液的配制:将上述四种样品用DMSO分别配制成浓度0.1mg/mL、0.2mg/mL、0.3mg/mL、0.4mg/mL、0.5mg/mL、0.6mg/mL、0.7mg/mL、0.8mg/mL待测样品溶液。
(3)底物L-酪氨酸溶液的配制:底物L-酪氨酸用磷酸缓冲液配制成质量浓度为0.05%的L-酪氨酸溶液。
(4)酪氨酸酶溶液的配制:酪氨酸酶用磷酸缓冲溶液配制成100U/mL的溶液。
(5)酪氨酸酶抑制率的测定:按表1准确吸取除酪氨酸酶溶液的C1、C2、T1、T2四组反应样液,在37℃水浴恒温10分钟后,再加入酪氨酸酶溶液,反应10分钟,以C2为C1的参比,测出体系在475nm处吸光度A1,以T2为T1的参比,测出体系吸光度A2。为平行样品带来的系统误差,在C1和C2中分别加入了ImL的DMSO。为消除样品在475nm处吸光度误差,在T2中也加入了lmL样品溶液。按下列公式计算酪氨酸酶的抑制率,结果见表2。
其中A1为未加样品的反应液的吸光度值的改变,A2为加入样品后反应液的吸光度值的改变。

结果表明,本发明三种实施例提取物对酪氨酸酶活性均有较好的抑制作用,其中,实施例2提取物的抑制效果最好,浓度在0.3mg/mL时,其抑制率达到55.08%,实施例1提取物的浓度在0.4mg/mL时,其抑制率为53.73%,实施例3提取物的浓度在0.4mg/mL时,其抑制率为57.11%,三种实施例提取物随着浓度的增加,酪氨酸酶的抑制率也相应增加,其中,实施例2的酪氨酸酶抑制率总体高于实施例1和实施例3。结果说明三种实施例提取物对酪氨酸酶的抑制效果明显,三种实施例提取物均具有良好的美白效果。

1.1材料与方法
1.1.1样品
实施例1提取物、实施例2提取物、实施例3提取物、维生素C(阳性对照)。
1.1.2实验方法
将上述四种样品用无水乙醇分别配制成浓度0.05mg/mL、0.06mg/mL、0.07mg/mL、0.08mg/mL、0.09mg/mL、0.1mg/mL、0.2mg/mL、0.3mg/mL、0.4mg/mL、0.5mg/mL待测样品溶液。取2.5mL样品溶液,加入2.5mL浓度为50mg/L的DPPH标准溶液,立即混匀,在室温下放入暗室反应30min,同时,取2.5mL样品溶液,加入95%的乙醇溶液2.5ml混合均匀,在517nm处测定吸光度。按下列公式计算DPPH自由基清除率,结果见表3。
As:2.5mlLDPPH溶液与2.5mL不同浓度的样品溶液组成的混合液的吸光度;
A:为2.5mL不同浓度的测试液与2.5mL无水乙醇组成的混合液的吸光度;
A0:为2.5mL DPPH溶液与2.5mL无水乙醇组成的混合溶液的吸光度。

2实施例1提取物、实施例2提取物、实施例3提取物清除ABTS自由基实验
2.1材料与方法
2.1.1样品
实施例1提取物、实施例2提取物、实施例3提取物、维生素C(阳性对照)。
2.1.2实验方法
将上述四种样品用95%乙醇分别配制成浓度0.05mg/mL、0.06mg/mL、0.07mg/mL、0.08mg/mL、0.09mg/mL、0.1mg/mL、0.2mg/mL、0.3mg/mL、0.4mg/mL、0.5mg/mL待测样品溶液。精密量取7mmol/L的ABTS溶液(由蒸馏水配制)0.20ml,与0.20ml的2.6mmol/L的过硫酸钾溶液混合,稀释40-50倍,使溶液754nm处的吸光度值保持在0.7±0.02,既得ABTS自由基工作液,室温下避光保存。分别吸取4mlABTS与1ml茶叶样品充分混合均匀,静置6min,以95%乙醇作为空白照,754nm处测定吸光度值。按照下列公式计算清除率:
其中,A0为ABTS自由基本身的吸光度值,A为样品对ABTS自由基作用后的吸光度值。结果见表4。
2.2实验结果

上述抗氧化实验结果表明,本发明三种实施例提取物均有良好的抗氧化作用,其中,实施例2提取物的抗氧化作用优于实施例1和实施例3提取物。现代研究表明,黑色素形成与氧自由基有关,故通过清除氧自由基,可有效减少黑色素的生成,起到美白作用。上述抗氧化实验表明,实施例2提取物的抗氧化活性强于其他实施例,结果与L-酪氨酸酶活性抑制实验一致。
实施例8
实施例1提取物、实施例2提取物、实施例3提取物抑菌实验
痤疮丙酸杆菌、表皮葡萄球菌和金黄色葡萄球菌是导致痤疮的主要三种病原菌,通过对本发明三种实施例的抑菌实验,可评价本发明的祛痘效果。
1材料与方法
1.1样品
实施例1提取物、实施例2提取物、实施例3提取物、甲硝唑(阳性对照)。
1.2实验方法
称取实施例提取物和甲硝唑各1.0g,加入去离子水50mL,溶解,过滤,取滤液备用(现配现用),将灭菌后的直径6mm的滤纸片,用无菌镊子夹取浸泡在样品液中,取出,晾干,置于无菌培养皿中,以灭菌后的生理盐水作阴性对照。移取200μL痤疮丙酸杆菌、表皮葡萄球菌和金黄色葡萄球菌菌悬液至固体培养皿上,涂布均匀,将准备好的滤纸片贴于培养皿上,37℃培养18-24h,观察结果,测定抑菌圈大小,每个样品设三个平行,结果取平均值。结果见表5。

表5结果表明,本发明三个实施例提取物对痤疮丙酸杆菌、表皮葡萄球菌和金黄色葡萄球菌等三种细菌具有很好的抑制作用,其抑菌圈的直径均在20cm以上,抑菌效果优良。说明本发明的实施例均有很好的抑菌作用,可用于祛痘美容。其中,实施例2提取物的抑菌效果最为显著,对痤疮丙酸杆菌、表皮葡萄球菌和金黄色葡萄球菌的抑制作用均显著优于其他两种实施例提取物。
实施例9
本发明实施例1产品、实施例2产品、实施例3产品人体试用评价实验
1祛痘美白霜的制备
本发明取上述复方中药提取物配制成祛痘美白霜,配方如下(按质量百分比计):
A相:硬脂酸 3.0%
凡士林 8.0%
单硬脂酸甘油酯 3.0%
液状石蜡 10.0%
十六醇 2.0%
苯甲酸 0.2%
B相:纯化水 55.0%
丙二醇 5.0%
聚乙二醇1500 5.0%
吐温-40 3.0%
三乙醇胺 0.5%
亚硫酸钠 0.1%
C相:复方中药提取物 5.0%
D相:香精 0.2%
制备工艺如下:
将A相和B相分别加热至80℃,充分搅拌混合均匀,在80℃条件下,将B相搅拌加入A相中,再加入C相,继续搅拌,至温度降至45℃时,加入D相,搅拌至室温,即得祛痘美白霜。
将实施例1提取物、实施例2提取物、实施例3提取物按照上述方法制成相应的祛痘美白霜,进行相应的人体实验。
2材料与方法
2.1样品
实施例1提取物祛痘美白霜、实施例2提取物祛痘美白霜、实施例3提取物祛痘美白霜、市售祛痘美白霜(主要成分:金银花、芦荟、灵芝等)。
2.2实验方法
选取自愿试用者160人(男性80人,女性80人,年龄18-25岁),脸部均有丘疹、脓包、痘痕,身体状况正常,无化妆品过敏史。将志愿者分成4组,每组40人,男女比例各半。第一组,试用实施例1产品;第二组,试用实施例2产品;第三组,试用实施例3产品,第四组,试用市售祛痘美白霜。
使用方法:早晚清洁面部后,取适量试用产品均匀涂抹于面部,轻轻拍打吸收,并轻轻按摩3-5分钟,让肌肤充分吸收,连续试用8周。通过问卷形式,调查试用者对所使用产品的主观评价,包括美白效果以及产品干涩、刺激感等指标。结果见表6。同时,观察与统计志愿者脸部皮肤改善情况,结果见表7。
显效:脸部丘疹、脓包、痘痕消退60%~90%;
有效:脸部丘疹、脓包、痘痕消退20%~60%;
无效:脸部丘疹、脓包、痘痕消退<20%。
3实验结果

上述两表为试用者对试用产品的评价表,表中结果显示,本发明实施例1、实施例2和实施例3祛痘美白霜产品具有较好的祛痘和美白作用。其中,表6结果表明,本发明三个实施例产品的美白作用明显,试用者对美白效果的评价明显优于市售祛痘美白霜,其中实施例2产品的美白效果最佳;表7结果显示,本发明实施例三个产品均能明显改善受试志愿者脸部皮肤的状态,丘疹、脓包、痘痕等症状得到明显改善,有效率均在85.0%以上,说明本发明产品具有明显的祛痘效果,其中,实施例2产品的效果最为明显,其有效率达到95.0%,优于其他两个实施例产品,也显著优于市售祛痘美白霜。此外,三个实施例产品均无过敏和刺激感,产品安全性高。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (6)
1.一种具有祛痘美白功效的中药组合提取物,其特征在于,该组合提取物由下述按重量份计的中药原料制备而成:黑果枸杞叶10-40份、三七叶10-30份、余甘子10-40份、藏荆芥10-30份、变叶海棠叶10-30份。
2.根据权利要求1所述的一种具有祛痘美白功效的中药组合提取物,其特征在于,该组合提取物由下述按重量份计的中药原料制备而成:黑果枸杞叶20-40份、三七叶10-20份、余甘子20-40份、藏荆芥10-20份、变叶海棠叶10-20份。
3.根据权利要求1所述的一种具有祛痘美白功效的中药组合提取物在制备祛痘美白护肤品中的应用。
4.根据权利要求3所述的应用,其特征在于:所述具有祛痘美白功效的中药复方提取物在祛痘美白护肤品中的质量百分含量为0.1-10%。
5.根据权利要求3所述的应用,其特征在于:所述具有祛痘美白功效的中药复方提取物在祛痘美白护肤品中的质量百分含量为1-8%。
6.根据权利要求3所述的应用,其特征在于:所述祛痘美白护肤品为祛痘美白洗面奶、祛痘美白洗面乳、祛痘美白洗面霜、祛痘美白膏、祛痘美白霜、祛痘美白乳液、祛痘美白水、祛痘美白面膜、祛痘美白凝胶或祛痘美白皂等化妆品外用剂型。
