Abstract

一种中国冬虫夏草真菌的发酵生产方法,其特点是将菌种植入液体培养基置于摇床上,温度20℃以下培养8天,再植入一级种子罐,温度20℃以下发酵5-8天,然后以培养基量的10倍扩大,逐级发酵,直至所需的量,出罐过滤而成。液体培养基为:含碳原0.5-5%,氮原0.2-5%,微量元素、维生素少许,水。该方法实现了冬虫夏草真菌的工业化生产,其生产成本低,生产速度快,真菌纯净,无杂菌。

Description

中国冬虫夏草真菌的发酵生产方法
本发明涉及一种中国冬虫夏草无性阶段真菌(Hirsutella bepiali Chen etSben,SP.nov-由虫草分类专家陈庆涛研究员1985年定名)的发酵生产方法。
中国专利(CN851019714)公开了一种“中国冬虫夏草真菌的人工培养方法”,该方法是在固体培养上培养。也就是在实验室中用试管三角瓶等作容器在固体培养基上培养。这种方法获得的菌丝数量很少。不能大规模生产面形成产品。也有人在20℃以上的条件下培养,实际上在20℃以上培养出来的并非中国冬虫夏草真菌,因为此条件下中国冬虫夏草真菌是不生长的。
本发明的目的是要提供一种能大规模工业化生产中国冬虫夏草真菌,且生产速度快、成本低。真菌纯的发酵生产方法。
本发明的技术方案是:将菌种植入液体培养基置于摇床上,温度12-20℃培养8天。然后植入一级种子罐,温度12-20℃发酵5-8天,然后以培养基量(体积)的10倍扩大,逐级发酵,直至达到所需要的量,出罐过滤而成。上述发酵过程均在液体培养基中进行,液体培养基为(按重量百分比):碳原0.5-5%,氮原0.2-5%,微量元素、维生素少许,其余为水份。
本发明的特点是,所述的培养基为(按重量百分比):蛋白胨0.5-2%,遮糖0.5-5%,奶粉0.5-2%,磷酸氢二钾、硫酸镁少许,其余为水。
本发明的特点是,所述的培养基为(按重量百分比):蚕蛹粉0.5-2%,蛋白胨0.3-2%,玉米粉0.5-2%,遮糖0.5-2%,磷酸氢二钾、硫酸镁少许,其余为水。
本发明的特点是,所述的培养基为(按重量百分比):牛奶5-15%,牛肉汤5-20%,蛋白胨0.2-3%, 燕麦粉0.5-2%,遮糖0.5-2%,磷酸氢二钾。硫酸镁少许,复合维生素少许,其余为水。
上述发酵过程都必须在20℃以下温度下发酵,因为中国冬虫夏草真菌的一个非常重要特征。就是只能在20℃以下的温度下才能生长,超过20℃就不是此物了。
上述发酵过程都必须通入一定量的无菌空气。
本发明的优点及效果是:实现了中国冬虫夏草真菌的大规模工业化生产,使用该方法生产成本低,生产速度快,生产出的菌纯净,无杂菌。到目前为止已发现该产品具有治疗各种虑症以及器官移植手术的抗排异作用,对内分泌有双向调节作用。其它作用有待进一步挖掘。
实施例1,工艺流程如前所述,实施四级发酵,温度为18℃,培养基为:蛋白胨2%,遮糖2%,奶粉1%,磷酸氢二钾、硫酸镁少许,其余为水。
实施例2,培养基为:蚕蛹粉1.5 %,蛋白胨1%,玉米粉2%,遮糖2%,磷酸氢二钾、硫酸镁少许,其余为水。工艺流程如前所述四级发酵,温度为14-16℃。
实施例3,培养基为:牛奶10%。牛肉汤20%,蛋白胨1%,燕麦粉2%,遮糖2%,磷酸氢二钾、硫酸镁、复合维生素少许。其余为水。工艺过程如前所述,实施三级发酵,温度为14-18℃。
上述实施例,均须通入无菌空气。

Claims (4)

1.一种中国冬虫夏草真菌的发酵生产方法,其特征在于将菌种植入液体培养基置于摇床上。温度20℃以下培养8天,然后植入一级种子罐。温度20℃以下发酵5-8天,然后以培养基量10倍扩大,逐级发酵,直至达到所需要的量。出罐过滤而成;上述发酵过程均在液体培养基中进行,液体培养基为(按重量百分比):碳原0.5-5%,氮原0.2-5%。微量元素、维生素少许,其余为水份。
2.如权利要求1所述的一种中国冬虫夏草真菌的发酵生产方法,其特征在于所述的培养基为(按重量百分比):蛋白胨0.5-2%,遮糖0.5-5%,奶粉0.5-2%,磷酸氢二钾、硫酸镁少许,其余为水。
3.如权利要求1所述的一种中国冬虫夏草真菌的发酵生产方法,其特征在于所述的培养基为(按重量百分比):蚕蛹粉0.5-2%,蛋白胨0.3-2%,玉米粉0.5-2%,遮糖0.5-2%,磷酸氢二钾、硫酸镁少许,其余为水。
4.如权利要求1所述的一种中国冬虫夏草真菌的发酵生产方法,其特征在于所述的培养基为(按重量百分比):牛奶5-15%,牛肉汤5-20%,蛋白胨0.2-3%,燕麦粉0.5-2%,遮糖0.5-2%,磷酸氢二钾、硫酸镁少许,复合维生素少许,其余为水。
