Abstract

本发明公开了一种肺癌辅助诊断或者疗效预测用的生物标记物及其应用,主要是用于制备肺癌辅助诊断或者疗效预测的实时荧光定量检测试剂。通过研究证实高表达LINC01420的肺癌患者比低表达LINC01420的肺癌患者预后差,因此,将LINC01420的表达用于肺癌患者的预后预测,可以为肺癌患者的预后提供强有力的分子生物学依据,具有深远的临床意义和重要的推广应用前景。

长链非编码RNA LINC01420的应用方法
技术领域
本发明属于肿瘤分子生物学领域,具体涉及一种肺癌辅助诊断或者疗效预测用的生物标记物长链非编码RNA LINC01420,以及检测该RNA表达量的试剂用于制备肺癌辅助诊断或者疗效预测制剂的用途。
背景技术
人类基因组计划及其后续的DNA元件百科全书计划(The Encyclopedia of DNAElements Project,ENCODE)研究成果表明,蛋白编码基因序列仅占人类基因组序列的1-3%,而人基因组中绝大部分可转录的序列为长链非编码RNA(Long non-coding RNA,lncRNA)。LncRNA广泛地存在于各种生物中,且随着生物复杂程度的升高,基因组中lncRNA序列的比例也相应地增大,提示lncRNA在生物进化过程中有重要意义。随着lncRNA不断被发现,它们的功能逐渐被诠释,科学家们发现lncRNA广泛而活跃地参与到生命活动不同层面的功能调控中,作为一个崭新的领域,已经成为国际生命科学研究领域新的前沿与热点。
LncRNA可作为功能蛋白质的信号(signal)、诱导(guide)、诱饵(decoy)或支架(scaffold)分子等多种方式,在染色质重构、基因转录、翻译及蛋白修饰等多重水平调控基因的表达,并在包括发育、免疫、生殖等基本生理过程中扮演了不可替代的角色,更重要的是,lncRNA的表达与功能失调已经与包括恶性肿瘤在内的人类多种疾病紧密联系在了一起。因此,深入研究lncRNA的功能,揭示由lncRNA介导的遗传信息传递方式和表达调控网络,不仅可以从蛋白编码基因以外的角度重新注释和阐明基因组的结构与功能,深入发现生命活动的本质和规律,还有望从一个新的视角认识包括肿瘤在内的多种人类常见疾病的发病机理,并为这些疾病的诊断与治疗提供新的分子标志物与治疗靶点。
肺癌是最常见的肿瘤,发病率和死亡率增长最快,对人群健康和生命威胁最大的恶性肿瘤之一。男性肺癌发病率和死亡率均占所有恶性肿瘤的第一位,女性发病率占第二位,死亡率占第二位。肺癌的病因至今尚不完全明确,长期大量吸烟与肺癌的发生有非常密切的关系。城市居民肺癌的发病率比农村高,这可能与城市大气污染和烟尘中含有致癌物质有关。LncRNA在肺癌的发生发展过程中可能也起到了重要的作用。最近我们在肺癌中发现lncRNA LINC01420(NCBI accession number:NR_015367)表达高的患者更容易发生转移,其生存时间短于该lncRNA表达低的患者,因此针对该lncRNA的检测制剂也可以用于肺癌的预后判断。
发明内容
本发明的目的是提供一种肺癌辅助诊断或者疗效预测用的生物标记物长链非编码RNA LINC01420,以及检测肺癌组织中该RNA表达量的试剂用于制备肺癌辅助诊断或者疗效预测制剂的用途。
一种肺癌辅助诊断或者疗效预测用的生物标记物为长链非编码RNA LINC01420,该长链非编码RNA LINC01420的序列如SEQ NO:1所示。
检测肺癌组织中长链非编码RNA LINC01420表达量的试剂的应用,所述的试剂用于制备肺癌辅助诊断或者疗效预测制剂;该长链非编码RNA LINC01420的序列如SEQ NO:1所示。
所述的肺癌辅助诊断或者疗效预测制剂为实时荧光定量检测试剂。
所述的实时荧光定量检测试剂中包含用于实时荧光定量检测LINC01420表达的引物序列:
正向引物:5'-CACTCTACCCTCCGCACC-3',
反向引物:5'-AGGAAGTGAAATCGTGCTGA-3'。
所述的实时荧光定量检测试剂中包含内参基因GAPDH特异性PCR引物:
正向引物5’-ACCACAGTCCATGCCATCAC-3’
反向引物5’-TCCACCACCCTGTTGCTGTA-3’。
本发明通过实时荧光定量PCR技术检测lncRNALINC01420在肺癌患者(226例)中的表达情况;发现LINC01420的表达与预后密切相关,LINC01420表达高的患者生存时间更短,提示LINC01420可作为肺癌预后预测的分子标志。本发明为肺癌的辅助诊断和预后预测提供了强有力的分子生物学工具,具有深远的临床意义和重要的推广应用前景。
附图说明
图1为实时荧光定量PCR技术检测lncRNA LINC01420的表达与肺患者预后的关系,肺癌组织中LINC01420的高表达与肺癌患者的不良预后相关,即LINC01420表达最高的13名患者总的生存时间(Over-all survival)要明显低于LINC01420低表达或不表达的患者。
具体实施方式
以下结合具体实施方式进一步说明本发明,而非限制本发明。
实施例1,实时荧光定量PCR法检测证实LINC01420在肺癌中表达上调
1.材料与方法:
收集226例肺癌组织,抽提总RNA,1μg RNA经逆转录成cDNA后,进行实时荧光定量PCR。LINC01420正向引物:5'-CACTCTACCCTCCGCACC-3',SEQ NO:2:反向引物:5'-AGGAAGTGAAATCGTGCTGA-3'。如SEQ NO:3所示。
用于对照的GAPDH正向引物为5’-ACCACAGTCCATGCCATCAC-3’如SEQ NO:4所示,和反向引物5’-TCCACCACCCTGTTGCTGTA-3’,如SEQ NO:5所示。
实时荧光定量PCR反应体系
实时荧光定量PCR反应步骤
反应结束后确认实时荧光定量PCR的扩增曲线和熔解曲线,各基因的表达强度根据CT值(threshold cycle values)、内参基因(GAPDH)标化后,采用group t-test检验计算P值。
2.结果
LINC01420高表达的肺癌患者预后较差
对226例肺癌患者进行了电话随访,详细询问了他们的首发时间、治疗情况、有无复发、有无再患其他疾病、复发及死亡时间等,并登记了生存时间和状态,并对肺癌组织中LINC01420的表达与病人的生存时间和状态进行的生存分析,发现LINC01420高表达最高的13名患者平均生存时间明显短于LINC01420低表达或不表达的患者(图1)。说明LINC01420是一个与肺癌预后相关的分子标记,该lncRNA表达高,病人预后差。

1.检测肺癌组织中长链非编码RNA LINC01420表达量的试剂的应用,其特征在于,所述的试剂用于制备肺癌辅助诊断或者疗效预测制剂;该长链非编码RNALINC01420的序列如SEQ NO:1所示。
2.根据权利要求1所述的应用,其特征在于,所述的肺癌辅助诊断或者疗效预测制剂为实时荧光定量检测试剂。
3.根据权利要求2所述的应用,其特征在于,所述的实时荧光定量检测试剂中包含用于实时荧光定量检测LINC01420表达的引物序列:
正向引物:5’ -CACTCTACCCTCCGCACC-3’,
反向引物:5’ -AGGAAGTGAAATCGTGCTGA-3’。
4.根据权利要求2或3所述的应用,其特征在于,所述的实时荧光定量检测试剂中包含内参基因GAPDH特异性PCR引物:
正向引物5’-ACCACAGTCCATGCCATCAC-3’
反向引物5’-TCCACCACCCTGTTGCTGTA-3’。
