技术领域
本发明涉及一种制备用于局部施用的脂质体复合物的新方法及使用该脂质体复合物的抗肿瘤剂。
背景技术
脂质体由构成生物体的细胞膜的磷脂组成,生物相容性高,且能够在生物体内保护药物或活性成分免受分解酶等的同时进行运送,作为药物递送系统的有用工具而备受关注。
另一方面,引起RNA干扰(以下,描述为“RNAi”)的RNAi分子作为用于治疗肿瘤等的有用的工具备受关注,开发了能够抑制肿瘤的增殖的各种RNAi分子。另外,开发了使用由RNAi分子和脂质混合物构成的复合物(脂质体复合物)、将作为活性成分的RNAi分子递送到肿瘤细胞的方法(非专利文献1-3)。
本发明人等迄今为止开发了将涉及肿瘤的增殖的胸苷酸合成酶(以下,描述为“TS”)作为靶的RNAi分子(专利文献1),报道了:使用将该RNAi分子负载于由规定的比例构成的阳离子性脂质混合物的复合体(脂质体复合物),例如通过静脉内施用并递送至肿瘤,由此可以抑制表达TS的肿瘤的增殖,进一步通过将该脂质体复合物与化疗剂并用,可以提高对肿瘤的靶向作用,显著地提高RNAi分子引起的抗肿瘤效果(专利文献2)。

发明内容
发明所要解决的课题
在现有的脂质体复合物的制备中,制备脂质混合物的方法包括将作为构成成分的磷脂等预先溶解于氯仿或环己烷等有机溶剂的工序。但是,在利用氯仿或环己烷等有机溶剂的工业制备方法中,需要危险物操作设备或防爆装置等大规模设备。另外,目前在脂质体复合物的制备时,在医院的药剂部门中,在对癌症患者的给药前制备RNAi分子的水溶液,另外,在制备脂质混合物之后,需要将两者进行混合的繁杂的工序,需要大量劳动力。
因此,本发明的目的在于,提供一种脂质体复合物的工业制备方法,其不需要使用大量的氯仿或环己烷等有机溶剂、进一步进行冷冻干燥之后、将生成的脂质混合物分散于水等溶剂中、与RNAi分子的水溶液等混合的繁杂的脂质体复合物制备工序,直接制备通过仅悬浮于水等溶剂而容易地生成脂质体复合物的粉末。
另外,目的在于取消医院的药剂部门中的繁杂且需要大量劳动力的脂质体复合物的配制工序。
用于解决课题的技术方案
本发明人等为了解决上述课题重复进行了深入研究,结果发现:将二油酰磷脂酰乙醇胺(DOPE)、磷脂酰胆碱及阳离子性脂质溶解于生物相容性高、没有爆炸性的醇,一边搅拌该醇溶液一边滴加于RNAi分子的溶液中之后,将该溶液进行冷冻干燥并除去醇等溶剂,由此可以制备脂质体复合物的冷冻干燥产物。而且,将通过该方法得到的脂质体复合物的冷冻干燥产物在适当的缓冲液中混合,进行对患有癌症的靶器官的局部施用,由此可以有效地治疗该癌症患者。本发明基于上述见解。

根据本发明,可以提供一种脂质体复合物的工业制备方法,其不需要使用大量的氯仿或环己烷等有机溶剂、进一步进行冷冻干燥之后、将生成的脂质混合物分散于水等溶剂中、与RNAi分子的水溶液等混合的繁杂的脂质体复合物制备工序,直接制备仅通过悬浮于水等溶剂而容易地生成脂质体复合物的粉末。
另外,根据本发明,可以消除医院的药剂部门中的繁杂且需要大量劳动力的脂质体复合物的配制工序。
此外,根据由本发明得到的脂质体复合物,可以将能够抑制肿瘤的增殖的RNAi分子有效地递送到肿瘤细胞。

具体实施方式
本发明的脂质体复合物含有由二油酰磷脂酰乙醇胺(DOPE)、磷脂酰胆碱及阳离子性脂质构成的脂质混合物和RNAi分子、或为由它们构成的复合体。
作为可在本发明中利用的“磷脂酰胆碱”,具有选自以下的(i)-(iii)中的一个以上的特征:
(i)包含至少一个含有碳-碳双键的不饱和脂肪链;
(ii)包含至少一个含有顺式的碳-碳双键的不饱和脂肪链;
(iii)相变温度低(例如低于0℃、低于-10℃、低于-20℃)。
作为这种“磷脂酰胆碱”,可列举:1,2-二油酰基-sn-甘油-3-磷酸胆碱(DOPC)、棕榈酰油酰磷脂酰胆碱(POPC)、1,2-二芥酰基-sn-甘油-3-磷酸胆碱(DEPC)。优选为DOPC。
作为可在本发明中利用的“阳离子性脂质”,可以利用选自O,O’-双十四碳酰基-Ν-(α三甲基铵基乙酰基)二乙醇胺氯化物(DC-6-14)、N,N-二油基-N,N-二甲基氯化铵(DODAC)、N,N-二硬脂基-N,N-二甲基溴化铵(DDAB)、N-(1-(2,3-二油酰氧基)丙基)-N,N,N-三甲基氯化铵(DOTAP)、N-(1-(2,3-二油基氧基)丙基)-N,N,N-三甲基氯化铵(DOTMA)、N,N-二甲基-(2,3-二油基氧基)丙胺(DODMA)及它们的衍生物中的任一种。优选阳离子性脂质为DC-6-14。
优选本发明中的脂质体复合物包含由DOPE、DOPC及DC-6-14构成的脂质混合物。
脂质体复合物中含有的DOPE、磷脂酰胆碱及阳离子性脂质的比(摩尔比)可以从DOPE:磷脂酰胆碱:阳离子性脂质=2~4:1~3:4~6的范围中选择。优选为DOPE:DOPC:DC-6-14=3:2:5。
本发明的脂质体复合物的粒径为200nm~2000nm,优选为大约400nm~700nm。另外,本发明的脂质体复合物的Zeta电位为30~60mV,优选为大约30~40mV。
本发明的脂质体复合物可以通过包含以下的工序的方法来制备。
即,将磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质的混合物的冷冻干燥产物或各自的成分溶解于醇。
作为醇,可以利用乙醇、甲醇,特别优选为生物相容性高的乙醇。磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质可以利用任何形式,例如可以分别利用粉末的形式。
磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质既可以分别预先单独地溶解于醇之后,分别取每一种混合以达到上述规定的量,或者也可以将由作为上述规定量的磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质构成的脂质混合物的冷冻干燥产物溶解于醇。
在含有磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质醇的醇混合液中,可以分别独立地以1~100mM、优选10~80mM含有磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质。
磷脂酰乙醇胺、磷脂酰胆碱及阳离子性脂质在醇中的溶解可以通过加热而进行。例如,可以加热至35℃~60℃、优选40℃~50℃而进行。
接着,在溶解有RNAi分子的溶液(例如用水溶解的水溶液)中在搅拌下滴加得到的醇溶液,将得到的溶液进行冷冻干燥,由此可以得到脂质体复合物的冷冻干燥产物。醇溶液和溶解有RNAi分子的溶液优选以1~3:7~9的比例进行混合。
得到的冷冻干燥产物进而通过加入缓冲液等水溶液并进行混合,可以得到脂质体复合物的悬浮液,可以将其施用于生物体。作为缓冲液,只要是可施用于生物体的水溶液即可,例如,可以利用生理盐水或糖质输液剂等。就混合而言,优选用涡旋搅拌机等搅拌1~15分钟、优选约5分钟。通过施加该搅拌,可以将脂质体复合物的粒径调整为上述所需要的尺寸。
本发明中的脂质体复合物的制备方法由于不需要使用毒性高、爆炸性高的氯仿或环己烷,因此,不需要在该制剂的制备工场内修筑防爆用的重型设备,另外,即使有极少量的残留溶剂,也仅为生物相容性高的乙醇,在对患者的施用方面问题少。
另外,根据本发明,可以在医院的配药部等医疗现场简便地制备脂质体复合物。即,通过在医疗现场提供填充有上述脂质体复合物的冷冻干燥产物的注射小瓶等,仅在对患者的施用前在医院的配药部向其中加入缓冲液,就可以在使用前制备含有脂质体复合物的施用溶液(水分散溶液)。
本发明的脂质体复合物可以用于对患者进行局部施用。本发明中“局部施用”并不是指静脉注射等全身施用。局部施用可列举例如:腹腔内施用、胸腔内施用、肌内施用、皮下施用、皮内施用、眼内施用、脑内施用、脑脊髓腔内施用、阴道内施用、直肠内施用、脏器内施用、对表皮的涂布等,但并不限定于这些。优选“局部施用”为腔内施用,进一步优选为胸腔内施用或腹腔内施用。
本发明的脂质体复合物含有能够通过RNAi作用抑制在肿瘤细胞中表达且编码参与肿瘤细胞的增殖的因子的基因的表达的RNAi分子(siRNA或shRNA等)作为活性成分。作为“在肿瘤细胞中表达且编码参与肿瘤细胞的增殖的因子的基因”,可列举例如编码胸苷酸合成酶、VEGF、EGFR、PDGF、HGF、Wint、Bcl-2、存活素等生长调节因子的组,核糖核苷酸还原酶、DNA聚合酶等核酸合成相关的酶的组等的基因,但并不限定于这些。这些基因的基因信息在GenBank等公知的数据库中被公开,可以基于这些基因信息对siRNA或shRNA进行设计、合成。关于能够通过RNAi作用抑制胸苷酸合成酶的表达的shRNA,可以使用下面详述的shRNA。另外,作为活性成分,也可以使用癌症化疗剂。
在本发明的脂质体复合物中,RNAi分子既可以被包含在脂质混合物的脂质双层包围的中空部分,或者可以被键合在脂质双层的外膜表面,优选被键合在脂质双层的外膜表面。RNAi分子与脂质双层的外膜表面的键合可以通过上述制备方法来实现。
本发明的脂质体复合物可以作为抗肿瘤剂使用。
在一个实施方案中,本发明的脂质体复合物包含通过RNAi作用能够抑制胸苷酸合成酶(以下,描述为“TS”)的表达的shRNA。
本发明中的能够抑制TS的表达的shRNA通过将TS的mRNA作为靶,起到TS特异性的RNAi作用,能够显著地抑制TS的表达。在此“将mRNA作为靶”是指:下述中详述的shRNA的反义链能够与靶mRNA在严格的条件下杂交。
严格的条件可以按照常规方法,基于形成杂交的核酸的解链温度(Tm)而得出。例如,作为能够维持杂交状态的洗涤条件,可列举通常约“1×SSC、0.1%SDS、37℃”的条件、更严格而言约“0.5×SSC、0.1%SDS、42℃”的条件、进一步严格而言约“0.1×SSC、0.1%SDS、65℃”的条件。
本发明中的shRNA包含具有编码TS的ORF的碱基序列或与其部分相同的碱基序列的有义链、和与该有义链在严格的条件下杂交的反义链。在此,“ORF的碱基序列或与其部分相同的碱基序列”是指:将ORF的碱基序列中的胸腺嘧啶替换为尿嘧啶而表示的碱基序列或与其部分相同的碱基序列。
该有义链由15~25个碱基、优选19个碱基构成。有义链的碱基序列优选与编码TS的ORF的碱基序列相同,但可以为基本上相同、即同源序列。即,有义链的碱基序列可以在ORF的碱基序列中具有1个或更多个、即1~3个碱基、优选1~2个碱基、更优选1个碱基的置换、缺失、插入和/或添加。
该反义链具有能够与有义链在严格的条件下杂交的碱基序列。反义链可以具有含有1~3个碱基、优选1~2个碱基、更优选1个碱基的置换、缺失、插入和/或添加的错配,只要能够在严格的条件下杂交即可。优选反义链由与有义链完全互补的碱基序列构成。
有义链及反义链的碱基序列可以基于公知的编码TS的碱基序列(GenBank:CR601528.1)而选择。已知作为选择这些碱基序列的方法的各种方法,例如可以使用siRNADesign Support System(Takara Bio株式会社)等。
在本发明中,作为有义链,可列举由以下的碱基序列构成的有义链,但并不限定于这些:5’-GUAACACCAUCGAUCAUGA-3’(序列号1);5’-GAAUACAGAGAUAUGGAAU-3’(序列号3);5’-CGAUCAUGAUGUAGAGUGU-3’(序列号5)。
优选本发明中的shRNA包含有义链5’-GUAACACCAUCGAUCAUGA-3’(序列号1)和反义链5’-UCAUGAUCGAUGGUGUUAC-3’(序列号2);有义链5’-GAAUACAGAGAUAUGGAAU-3’(序列号3)和反义链5’-AUUCCAUAUCUCUGUAUUC-3’(序列号4);或有义链5’-CGAUCAUGAUGUAGAGUGU-3’(序列号5)和反义链5’-ACACUCUACAUCAUGAUCG-3’(序列号6)。
进一步优选本发明中的shRNA包含由序列号1表示的碱基序列构成的有义链和由序列号2表示的碱基序列构成的反义链。
有义链和反义链经由接头部分而连接,该接头部分通过形成环而被折叠,该反义链和该有义链杂交而形成双链部分。shRNA分子中所含的接头部分只要可以连接有义链和反义链而形成茎环结构即可,可以为多核苷酸接头,也可以为非多核苷酸接头,没有特别限定,优选本领域技术人员所公知的2~22个碱基的多核苷酸接头。具体而言,可以例示UAGUGCUCCUGGUUG(序列号7)、UUCAAGAGA、CCACC、CUCGAG、CCACACC、UUCAAGAGA、AUG、CCC及UUCG,优选UAGUGCUCCUGGUUG(序列号7)。
本发明中的shRNA在3’末端具有由至少2个碱基构成的突出端。
在本发明中,突出端是指:为在反义链的3’末端所添加的碱基、可以在有义链的对应的位置不存在互补地键合的碱基的碱基。与具有突出端的情况相比,在反义链的3’末端不具有突出端的情况下shRNA引起的TS的表达抑制的程度降低约40~60%。该突出端的碱基的种类、数量没有限定,可列举例如由1~5个、优选1~3个、进一步优选1个或2个碱基构成的序列,可列举例如TTT、UU或TT。优选为UU。
在本发明中,优选的shRNA为由序列号8表示的碱基序列构成的单链RNA。
另外,有义链或反义链根据需要可以将5’末端进行磷酸化,也可以在5’末端键合三磷酸(ppp)。
除了能够抑制TS的表达的shRNA之外,本发明中的具有shRNA的脂质体复合物还可以含有通过RNAi作用能够抑制“在肿瘤细胞中表达且编码参与肿瘤细胞的增殖的因子的基因”的表达的siRNA或shRNA。作为此类siRNA或shRNA,可以使用以上所定义的siRNA或shRNA。能够抑制TS的表达的shRNA和其它的siRNA或shRNA既可以存在于相同的脂质体复合物中,或者也可以分别存在于单独的脂质体复合物中。
如下述实施例中所详述的那样,通过局部施用上述具有shRNA的脂质体复合物能够抑制肿瘤细胞的增殖,可以用于治疗癌症。
作为能够使用本发明的抗肿瘤剂治疗的癌症,可列举表达TS的大量癌症,没有特别限制,可列举例如:结肠·直肠癌、肝癌、肾癌、头颈癌、食道癌、胃癌、胆道癌、胆囊·胆管癌、胰腺癌、肺癌、乳癌、卵巢癌、子宫颈癌、子宫体癌、膀胱癌、前列腺癌、恶性胸膜间皮瘤、睾丸癌、卵巢癌、骨·软组织肉瘤、皮肤癌、脑肿瘤等。另外,癌性胸膜炎或癌性腹膜炎也可以用本抗肿瘤剂治疗。作为治疗对象,优选为胃癌、肺癌、胆道癌、肝癌、恶性胸膜间皮瘤、卵巢癌、癌性胸膜炎或腹膜炎,特别优选为胃癌、卵巢癌或胰腺癌的腹膜转移、胸膜恶性间皮瘤、在胸腔内存在主病灶的肺癌及癌性肺炎。
本发明的抗肿瘤剂含有脂质体复合物同时另外可以含有在医药的制备中通常所使用的赋形剂、粘合剂、崩解剂、润滑剂、稀释剂、增溶剂、悬浮剂、等张剂、pH调节剂、缓冲剂、稳定剂、着色剂、矫味剂、矫臭剂、组氨酸等。
作为赋形剂,可列举例如:乳糖、蔗糖、氯化钠、葡萄糖、麦芽糖、甘露醇、赤藓醇、木糖醇、麦芽糖醇、肌醇、葡聚糖、山梨糖醇、白蛋白、尿素、淀粉、碳酸钙、高岭土、结晶纤维素、硅酸、甲基纤维素、甘油、海藻酸钠、阿拉伯树胶及它们的混合物等。作为润滑剂,可列举例如:精制滑石、硬脂酸盐、硼砂、聚乙二醇及它们的混合物等。作为粘合剂,可列举例如:单糖浆、葡萄糖溶液、淀粉溶液、明胶溶液、聚乙烯醇、聚乙烯醚、聚乙烯基吡咯烷酮、羧甲基纤维素、虫胶、甲基纤维素、乙基纤维素、水、乙醇、磷酸钾及它们的混合物等。作为崩解剂,可列举例如:干燥淀粉、海藻酸钠、琼脂粉、褐藻粉(ラミナラン末,laminaran powder)、碳酸氢钠、碳酸钙、聚氧乙烯山梨糖醇酐脂肪酸酯类、月桂基硫酸钠、硬脂酸单甘油酯、淀粉、乳糖及它们的混合物等。作为稀释剂,可列举例如:水、乙醇、聚乙二醇、丙二醇、乙氧基化异硬脂醇、聚氧化异硬脂醇、聚氧乙烯山梨糖醇酐脂肪酸酯类及它们的混合物等。作为稳定剂,可列举例如:焦亚硫酸钠、乙二胺四乙酸、巯基乙酸、硫代乳酸及它们的混合物等。作为等张剂,可列举例如:氯化钠、硼酸、葡萄糖、甘油及它们的混合物等。作为pH调节剂及缓冲剂,可列举例如:柠檬酸钠、柠檬酸、醋酸钠、磷酸钠及它们的混合物等。
本发明的抗肿瘤剂可以通过局部施用而施用。局部施用可列举上述所定义的施用方式。另外,本发明的组合物可以制备成适于局部施用的剂型,可以制备成注射剂、悬浮剂、乳化剂、喷雾剂等各种制剂形式。另外,本发明的抗肿瘤剂可以以冷冻干燥形式提供,在使用时可以加入适当的缓冲液(例如生理盐水)而使用。
就本发明的抗肿瘤剂的效果而言,对源自上述癌症的细胞或组织及患有上述癌症的个体施用该抗肿瘤剂,肿瘤大小与没有施用该抗肿瘤剂的(或施用前的)细胞或组织及个体中的肿瘤大小相比,可以以肿瘤缩小或消失为指标进行评价。另外,就本发明的抗肿瘤剂的效果而言,对源自上述癌症的细胞或组织及患有上述癌症的个体施用该抗肿瘤剂,与没有施用该抗肿瘤剂的个体相比,可以以存活率的提高(存活效果)、胸水或腹水减少或消失为指标进行评价。
本发明的抗肿瘤剂可以与现有的癌症化疗法或癌症化疗剂一起使用。作为可与本发明的抗肿瘤剂并用的癌症化疗法或癌症化疗剂,只要可以以本发明的脂质体复合物容易穿透肿瘤组织的方式改变肿瘤环境即可,例如,作为现有的癌症化疗剂,可列举认为对胃癌或卵巢癌的腹膜转移患者有效的Taxol(注册商标)(Bristol-Myers Squibb)或Taxotere(注册商标)(Sanofi-Aventis)等具有微管蛋白的解聚抑制作用的抗肿瘤剂、由对胰腺癌及其腹膜转移的治疗有用的gemcitabine(注册商标)(Eli Lilly)等脱氧胞苷衍生物构成的抗肿瘤剂。
此外,除上述癌症化疗剂之外,本发明的抗肿瘤剂也可以与其它现有的癌症化疗剂一起使用。作为这种癌症化疗剂,可列举:环磷酰胺、氮芥N-氧化物、异环磷酰胺、美法仑、白消安、二溴甘露醇、卡波醌、噻替派、雷莫司汀、尼莫司汀、替莫唑胺、卡莫司汀、培美曲塞二钠、甲氨蝶呤、6-巯基嘌呤核苷、巯基嘌呤、去氧氟尿苷、卡莫氟、阿糖胞苷、阿糖胞苷十八烷基磷酸盐、依诺他滨、氟达拉滨、培美曲塞、顺铂、卡铂、奥沙利铂、多西他奇、盐酸伊立替康、卡培他滨等,可以使用选自这些物质中的一种或更多种癌症化疗剂。与上述癌症化疗剂相同,这些癌症化疗剂也通过与本发明的抗肿瘤剂的并用,能够将shRNA有效地递送到肿瘤细胞,另外,与单独使用该癌症化疗剂或本发明的抗肿瘤剂的情况相比,可以得到显著提高的抗肿瘤效果。
本发明的抗肿瘤剂和上述现有的癌症化疗剂并用施用时,可以作为组合物提供。
本发明的抗肿瘤剂可以与上述现有的癌症化疗剂同时作为“组合物”的形式。“组合物”既可以为含有本发明的抗肿瘤剂和上述现有的癌症化疗剂作为有效成分的配合剂,且也可以为使本发明的抗肿瘤剂和上述现有的癌症化疗剂作为适于并用施用的单一的包装(试剂盒制剂)制备、包装、流通。
在“并用施用”中,不仅包含同时施用本发明的抗肿瘤剂和上述现有的癌症化疗剂的情况,而且也包含将本发明的抗肿瘤剂及上述现有的癌症化疗剂隔开间隔施用的情况。本发明的抗肿瘤剂和上述现有的癌症化疗剂的施用途径及施用方法既可以相同,也可以不同。
本发明的抗肿瘤剂的施用量及施用次数可以根据患者的年龄、体重、疾病的严重程度等主要因素而变化,但作为shRNA的量,可以以每次每1kg体重从0.0001mg~100mg的范围中适当选择的量、1天1~3次、每天或每1~21天施用。
上述现有的癌症化疗剂的施用量可以根据作为有效成分的化学物质的种类、患者的年龄、体重、疾病的严重程度等主要因素而变化,但可以以每次每1kg体重从0.0001mg~1000mg的范围中适当选择的量、1天1~3次、每天或每1~14天施用。例如,现有的癌症化疗剂为紫杉醇时,可以以1天500~1000mg、每1~21天腹腔内或静脉内施用。现有的化疗剂为培美曲塞二钠水合物时,可以以1天500~1000mg、每1~21天静脉内施用。现有的化疗剂为5-FU系经口抗癌剂的TS-1时,可以1天1~3次、每天或每2~3天施用。与单独使用的情况相比,可以以低用量且多次施用上述现有的癌症化疗剂。由此,可以抑制或延缓通过上述现有的癌症化疗剂的施用而引起的副作用(例如骨髓抑制、溶血性贫血、弥散性血管内凝血综合征、爆发性肝炎、脱水、肠炎、间质性肺炎、口腔溃疡、消化道溃疡、消化道出血、消化道穿孔、急性肾功能衰竭、皮肤粘膜眼综合征、中毒性表皮坏死松解症、神经精神障碍、急性胰腺炎、横纹肌溶解症、嗅觉丧失等,但并不限定于这些)的发作。
本发明还涉及一种使用本发明的上述抗肿瘤剂治疗癌症的方法。作为可以利用该方法治疗的癌症,包括以上所定义的癌症。另外,在该方法中,本发明的上述抗肿瘤剂及现有的癌症化疗剂的用法及用量如上所述。
实施例
以下,示出实施例,对本发明进一步详细地进行说明。但是,本发明并不限于这些实施例。
实施例1:脂质体复合物的制备
在本实施例中,试剂使用以下描述的试剂。

(将TS作为靶的shRNA)
将TS作为靶的shRNA(以下,描述为“TS-shRNA”)为基于能够抑制TS的表达、且已经确认了抗肿瘤效果的公知的shRNA(参照WO2012/161196)合成的具有以下序列的shRNA。
TS-shRNA:

(1)利用本发明方法的脂质体复合物(新型制剂)的制备
(1-1)来自脂质混合物的冷冻干燥产物的脂质体复合物(新型制剂(1))的制备
称量日本精化制Presome DF1(以3/2/5的摩尔比含有DOPE/DOPC/DC-6-14的脂质混合物)(100mg),在室温下溶解于1.842mL乙醇,由此得到脂质混合物的乙醇溶液。
在无RNA酶的水(66.2μL)中加入3.8μL 300μM TS-shRNA,得到TS-shRNA水溶液。
接着,缓慢搅拌的同时,在上述的TS-shRNA水溶液(70μL)中缓慢地滴加上述脂质混合物的乙醇溶液(30μL),由此得到脂质体复合物的乙醇/水溶液。
使得到的脂质体复合物的乙醇/水溶液在-40℃以下冷冻之后,使用真空冷冻干燥机(LABCONCO FZ-4.5,朝日生命科学)进行冷冻干燥。
在得到的冷冻干燥品中加入100μL由无RNA酶的水制备的生理盐水(0.9v/w%氯化钠水溶液),用PresentMixer2013搅拌10秒,由此得到脂质体复合物(TS-shRNA量为20μg/100μL)。
以下,将这样得到的脂质体复合物描述为“新型制剂(1)”。
(1-2)来自DOPE、DOPC及DC-6-14的各脂质粉末的脂质体复合物(新型制剂(2))的制备
分别称量DOPE(55.8mg)、DOPC(78.6mg)及DC-6-14(66.0mg)的粉末,分别添加1mL乙醇后,加热至40℃,由此得到75mM DOPE、100mM DOPC、100mM DC-6-14的溶液。
向乙醇(4.777μL)中加入上述75mM DOPE溶液(9.172μL)、100mM DOPC溶液(4.586μL)及100mM DC-6-14溶液(11.465μL),得到以3/2/5的摩尔比含有DOPE/DOPC/DC-6-14的脂质混合物的乙醇溶液。
接着,缓慢搅拌的同时,在上述TS-shRNA水溶液(70μL)中缓慢地滴加上述脂质混合物的乙醇溶液(30μL),由此得到脂质体复合物的乙醇/水溶液。
将得到的脂质体复合物的乙醇/水溶液与上述(1-1)同样地进行冷冻干燥,加入用无RNA酶的水制备的生理盐水并进行搅拌,由此得到脂质体复合物(TS-shRNA量为20μg/100μL)。
以下,将这样得到的脂质体复合物描述为“新型制剂(2)”。
(2)利用现有公知的方法的脂质体复合物的制备
分别称量DOPE(0.51mg)、DOPC(0.36mg)及DC-6-14(0.76mg)的粉末,在1.0mL环己烷/乙醇混液(95%/5%(V/V))中进行混合并溶解。接着,将得到的溶液进行冷冻干燥处理而除去环己烷、乙醇之后,加入生理盐水(50μL),使用涡旋搅拌机激烈地搅拌10分钟并使其悬浮之后,加入含有TS-shRNA的(0.4μg/μL)水溶液(50μL)并混合,由此得到脂质体复合物(TS-shRNA量为20μg/100μL)。
以下,将这样得到的脂质体复合物描述为“现有制剂”。
实施例2:各脂质体复合物的物理性质的同等性比较
使用Zetasizer Nano ZS(Malvern)测定实施例1中得到的新型制剂(1)、新型制剂(2)及现有制剂的粒径(d.nm)、多分散性指数(polydispersity index:PdI)、及Zeta电位(mV)。
将结果示于图1。
由该结果可以确认:利用现有公知的方法得到的现有制剂和用本发明方法得到的新型制剂(1)及新型制剂(2),在其物理性质上没有显著的差异,利用现有公知的方法得到的脂质体复合物和用本发明方法得到的脂质体复合物,在物理性质上没有差异。
实施例3:脂质体复合物的TS-shRNA保持能力的评价
在本实施例中,试剂使用以下描述的试剂。

溴化乙锭(EtBr):和光纯药工业
分别对实施例1中得到的新型制剂(1)、新型制剂(2)及现有制剂的shRNA保持能力进行评价。
分别称量Tris(5.4g)、硼酸(2.75g)、EDTA·2Na(0.185g),用蒸馏水定容为1L,由此制备TBE缓冲液。在琼脂糖(0.4g)中加入TBE缓冲液(40mL),煮沸而使琼脂糖完全溶解之后,加入1mg/mL EtBr(4μL),将其倒入凝胶板,冷却到室温,由此制作1%琼脂糖凝胶。
在电泳装置(PLUS-2、CIMA Biotechnology)中设置凝胶板,将现有制剂、新型制剂(1)及新型制剂(2)分别稀释10倍并加样于琼脂糖凝胶,在电泳槽中加入TBE缓冲液之后,在100V下电泳15分钟。其后,用AE-9000N E-Graph(ATTO)观察TS-shRNA条带。另外,在制备各脂质体复合物1天后及3天后,也同样地进行评价。
将结果示于图2。
在制备之后的现有制剂以及新型制剂(1)及新型制剂(2)中,在与游离TS-shRNA相同的位置上完全没有观察到条带,表明含有各脂质体复合物的样品中几乎不存在游离TS-shRNA。另外,在至制备3天后,在与TS-shRNA相同位置也没有观察到条带,因此,可以确认:现有制剂以及新型制剂(1)及新型制剂(2)可连续保持TS-shRNA至制备3天后。
由上述结果可以确认:在利用现有公知的方法得到的脂质体复合物和用本发明方法得到的脂质体复合物之间,在shRNA保持能力上没有显著的差异。
实施例4:各脂质体复合物的腹腔内施用导致的肿瘤中的靶基因抑制效果的评价
(实时RT-PCR)

对实施例1中得到的新型制剂(1)、新型制剂(2)及现有制剂的肿瘤中的靶基因抑制效果,利用实时RT-PCR法进行评价。
培养MKN45人胃癌细胞(理化学研究所),以5×106个(细胞)/小鼠移植到BALB/cnu/nu小鼠(5周龄、雄性)的腹腔内。分别测定移植当天及移植6天后的小鼠的体重,将体重减少(认为移植细胞存活)的小鼠作为MKN45腹膜转移模型小鼠用于实验。
在细胞移植后7天、9天、11天的总计3次,分别对各模型小鼠腹腔内施用现有制剂、新型制剂(1)或新型制剂(2)以实现TS-shRNA的施用量为20μg/小鼠/天。在细胞移植13天后,从各模型小鼠中切除肿瘤,用MULTI-BEADS SHOCKER(注册商标)(安井机械)将肿瘤进行匀浆处理后,使用RNeasy(注册商标)Mini Kit(QIAGEN)提取RNA。
用NanoDrop 8000(Thermo Fisher Scientific)测定得到的RNA提取液的RNA浓度之后,用无RNA酶的水稀释以获得RNA浓度为1μg/7.7μL,使用TaqMan(注册商标)ReverseTranscription Reagents进行逆转录(在16℃下30分钟、在42℃下30分钟、在85℃下5分钟)。
向得到的cDNA溶液5μL中加入15μL以下所示的TS PCR混合物或GAPDH PCR混合物,使用StepOnePlusTM(Life Technologies)进行实时RT-PCR(1循环:在50℃下2分钟、在95℃下10分钟;40循环:在95℃下15秒、在60℃下1分钟),由此测定TS mRNA量及GAPDH mRNA量。

将结果示于图3。图3将用实时RT-PCR测定的TS mRNA的值的数据用GAPDH mRNA的值校正并设为“TS mRNA表达水平”,所述“TS mRNA表达水平”以没有施用脂质体复合物而施用9%蔗糖溶液的模型小鼠(对照组)的值设为100%的相对值表示。
与对照组相比,在现有制剂、新型制剂(1)及新型制剂(2)的所有组中,观察到约30%的TS mRNA表达水平的显著降低。由此表明,利用现有公知的方法得到的脂质体复合物及利用本发明方法得到的脂质体复合物,能够显著地抑制转移至腹膜的肿瘤的靶基因表达,另外得知:其效果在利用现有公知的方法得到的脂质体复合物和利用本发明方法得到的脂质体复合物之间没有显著差异。
实施例5:各脂质体复合物的腹腔内施用导致的肿瘤的靶基因抑制效果的评价
(体内成像系统(In Vivo Imaging Systems:IVIS))
在本实施例中,试剂使用以下描述的试剂。

将Luc作为靶的shRNA(以下,描述为“Luc-shRNA”)具有以下的序列。

在本实施例中,使用针对萤光素酶的shRNA(Luc-shRNA),在上述实施例1中描述的脂质体复合物的制备方法中,将shRNA替换为Luc-shRNA,除此之外,同样地操作,制备负载Luc-shRNA的脂质体复合物,对新型制剂(1)、新型制剂(2)及现有制剂的肿瘤的靶基因抑制效果,利用IVIS进行评价。
培养表达萤光素酶的NCI-N87人胃癌细胞(NCI-N87-Luc、住商PharmaceuticalsInternational),以4×106个(细胞)/小鼠移植到BALB/c nu/nu小鼠(5周龄、雄性、日本SLC)的腹腔内,由此制作NCI-N87-Luc腹膜转移模型小鼠。
在细胞移植后8天、11天、14天、17天、20天总计5次,分别对各模型小鼠腹腔内施用现有制剂、新型制剂(1)或新型制剂(2)以实现Luc-shRNA的施用量为20μg/小鼠/天。另外,用PBS溶解荧光素以获得7.5mg/mL,在细胞移植后7天、10天、13天、16天、19天、22天,分别进行腹腔内施用荧光素10μL之后,用IVIS(Xenogen,Alameda,CA,USA)观察肿瘤的萤光素酶活性。将没有施用脂质体复合物而施用9%蔗糖溶液的模型小鼠用作对照组。
图4表示使用得到的成像数据结果将腹腔中的萤光素酶活性定量化的曲线图。
在对照组中,随着时间推移,观察到萤光素酶活性的上升,显示肿瘤在腹腔内增殖。另一方面,在负载Luc-shRNA的现有制剂、新型制剂(1)及新型制剂(2)的各腹腔内施用组中,从第1次施用后观察到萤光素酶活性的显著降低,在实施实验期间(至细胞移植22天后),萤光素酶活性没有再上升(图4)。
在本实施例中,使用了针对萤光素酶的Luc-shRNA,但其不大可能对肿瘤的增殖有影响。实际测定细胞移植24天后的各组的肿瘤重量,结果,在组间没有看到显著的差异。由此显示:通过施用负载Luc-shRNA的各脂质体复合物,仅抑制肿瘤的萤光素酶的活性。由上述结果得知:在利用现有公知的方法得到的脂质体复合物和用本发明方法得到的新型的脂质体复合物之间保持相同的TS-shRNA活性。
实施例6:利用脂质体复合物(新型制剂(2))的腹腔内施用的针对卵巢癌腹膜转移的治疗评价
对与上述实施例1同样地制备的负载TS-shRNA的脂质体复合物(新型制剂(2))的针对卵巢癌腹膜转移的治疗效果进行评价。
培养表达萤光素酶的OVCAR-3人卵巢癌细胞(OVCAR-3-luc、Pharmaron),以1.5x107个(细胞)/小鼠移植到BALB/c nu/nu小鼠(6-8周龄、雌性、Beijing HFK Bio-Technology Co.,LTD)的腹腔内,由此产生OVCAR-3-luc腹膜转移模型小鼠。
在本实施例中,向模型小鼠施用脂质体复合物(新型制剂(2))、紫杉醇(Taxol、Pharmaron)、或脂质体复合物(新型制剂(2))及紫杉醇。在对照中施用9%蔗糖溶液。
在细胞移植后8天、11天、14天、17天的总计4次,对模型小鼠腹腔内施用脂质体复合物(新型制剂(2))以实现TS-shRNA的施用量为20μg/小鼠/天。在施用紫杉醇的组中,在细胞移植后8天、11天、14天、17天的总计4次,以15mg/kg体重的方式对模型小鼠腹腔内施用。
用PBS溶解荧光素以获得7.5mg/mL,在细胞移植后7天、14天、21天、26天分别进行腹腔内施用荧光素10μL之后,用IVIS观察肿瘤的萤光素酶活性。
将结果示于图5-1。另外,使用图5-1的成像数据,分别示出将腹腔中的萤光素酶活性定量化的曲线、图(图5-2)及其对数图(图5-3)。
在对照组中,随着时间推移,观察到萤光素酶活性的上升,显示肿瘤在腹腔内增殖。另一方面,与对照组相比,在腹腔内施用负载TS-shRNA的脂质体复合物的组抑制了萤光素酶活性的上升,即,确认能够抑制腹腔内的肿瘤的增殖。此外,在脂质体复合物和紫杉醇并用的组中,观察到最高的肿瘤抑制效果。
此外,将各模型小鼠的存活期示于图5-4。
与对照组相比,在脂质体复合物和紫杉醇并用的组中,观察到显著提高的存活效果。
此外,将用RT-PCR法测定施用脂质体复合物的模型小鼠的血液中及腹水中的TS-shRNA的浓度变化得到的结果示于图5-5。
结果在腹水中观测到约8小时的长的半衰期,同时,确认在血液中不存在TS-shRNA渗漏。
实施例7:利用脂质体复合物(新型制剂(2))的腹腔内施用的针对胰腺癌腹膜转移的治疗评价
对与上述实施例1同样地制备的负载TS-shRNA的脂质体复合物(新型制剂(2))的针对胰腺癌腹膜转移的治疗效果进行评价。
培养表达萤光素酶的PANC-1人胰腺癌细胞(PANC-1-luc、Pharmaron),以1.0×107个(细胞)/小鼠移植到BALB/c nu/nu小鼠(6-8周龄、雌性、Beijing HFK Bio-TechnologyCo.,LTD)的腹腔内,由此产生PANC-1-luc腹膜转移模型小鼠。
在本实施例中,对模型小鼠施用脂质体复合物(新型制剂(2))、紫杉醇(Taxol、Pharmaron)、或脂质体复合物(新型制剂(2))及紫杉醇。在对照中施用9%蔗糖溶液。
在细胞移植后8天、11天、14天、17天的总计4次,对模型小鼠腹腔内施用脂质体复合物(新型制剂(2))以实现TS-shRNA的施用量为20μg/小鼠/天。在施用紫杉醇的组中,细胞移植后8天、11天、14天、17天的总计4次,以10mg/kg体重的方式对模型小鼠腹腔内施用。
用PBS溶解荧光素以获得7.5mg/mL,在细胞移植后7天、14天、21天、25天,分别进行腹腔内施用荧光素10μL之后,用IVIS观察肿瘤的萤光素酶活性。
分别示出基于IVIS的成像数据将腹腔中的萤光素酶活性定量化的曲线图(图6-1)及其对数图(图6-2)。
在对照组中,随着时间推移,观察到萤光素酶活性的上升,显示肿瘤在腹腔内增殖。另一方面,与对照组相比,施用脂质体复合物(新型制剂(2))、紫杉醇(Taxol、Pharmaron)、或脂质体复合物(新型制剂(2))及紫杉醇的组抑制了例如萤光素酶活性的上升,即,确认能够抑制腹腔内的肿瘤的增殖。特别是在施用紫杉醇的组以及脂质体复合物和紫杉醇并用的组中,观察到高的肿瘤抑制效果。
另外,将各模型小鼠的存活期示于图6-3。
与对照组相比,在并用了脂质体复合物和紫杉醇的组中,看到显著提高的存活效果。
工业上的可利用性
根据本发明,可以不使用如氯仿或环己烷那样毒性高、爆炸危险性高的有机溶剂,仅使用生物相容性高的溶剂制备脂质体复合物,因此,是理想的药物制剂的工业制备方法。另外,避免了在医院的药剂部门等医疗现场中分别制备脂质混合物的水分散液和RNAi分子的水溶液、将两者混合的复杂的配药过程,仅通过在脂质体复合物的粉末中加入盐水或糖质输液剂、轻轻地搅拌即可以制备用于患者的施用液,因此,可以大幅降低医院的药剂部门的配药的劳力及成本。
此外,针对胃癌或卵巢癌或胰腺癌等的腹膜转移(接近于每种癌症的末期状态),进行腹腔内施用,另外,针对在胸腔内存在主病灶的胸膜恶性间皮瘤或肺癌或者癌性肺炎进行胸腔内施用等适宜的局部施用,由此可以将RNAi分子有效地递送到肿瘤细胞,可以有效地抑制该肿瘤的增殖。本发明预期在药物递送领域以及在癌症治疗领域做出大的贡献。

Claims (15)
1.一种制备脂质体复合物的方法,所述脂质体复合物含有二油酰磷脂酰乙醇胺(DOPE)、磷脂酰胆碱及阳离子性脂质以及RNAi分子,所述方法包括:
(a)将二油酰磷脂酰乙醇胺(DOPE)、磷脂酰胆碱及阳离子性脂质溶解于醇中的工序;
(b)在溶解有RNAi分子的溶液中在搅拌下滴加(a)中得到的醇溶液的工序;及
(c)将(b)中得到的溶液进行冷冻干燥的工序,
所述磷脂酰胆碱具有选自以下的(i)-(iii)中的一个以上的特征:
(i)包含至少一个含有碳-碳双键的不饱和脂肪链;
(ii)包含至少一个含有顺式的碳-碳双键的不饱和脂肪链;
(iii)相变温度低于0℃。
2.根据权利要求1所述的方法,其中,阳离子性脂质为O,O’-双十四碳酰基-Ν-(α三甲基铵基乙酰基)二乙醇胺氯化物(DC-6-14)。
3.根据权利要求1或2所述的方法,其中,磷脂酰胆碱为1,2-二油酰基-sn-甘油-3-磷酸胆碱(DOPC)、棕榈酰油酰磷脂酰胆碱(POPC)、或1,2-二芥酰基-sn-甘油-3-磷酸胆碱(DEPC)。
4.根据权利要求3所述的方法,其中,磷脂酰胆碱为DOPC。
5.根据权利要求1所述的方法,所述方法包括:在工序(a)中将DOPE、DOPC及DC-6-14溶解于醇中并进行混合。

7.一种用于局部施用的药物组合物,所述药物组合物用于治疗胃癌、卵巢癌及胰腺癌的腹膜转移,所述药物组合物包含由二油酰磷脂酰乙醇胺(DOPE)、磷脂酰胆碱及阳离子性脂质构成的脂质混合物和能够通过RNAi作用而抑制胸苷酸合成酶的表达的短发夹RNA(TS-shRNA),其中,所述磷脂酰胆碱具有选自以下的(i)-(iii)中的一个以上的特征:
(i)包含至少一个含有碳-碳双键的不饱和脂肪链;
(ii)包含至少一个含有顺式的碳-碳双键的不饱和脂肪链;
(iii)相变温度低于0℃。
8.根据权利要求7所述的药物组合物,其中,阳离子性脂质为O,O’-双十四碳酰基-Ν-(α三甲基铵基乙酰基)二乙醇胺氯化物(DC-6-14)。
9.根据权利要求7所述的药物组合物,其中,磷脂酰胆碱为DOPC。
10.根据权利要求7所述的药物组合物,其中,脂质混合物由DOPE、DOPC及DC-6-14构成。
11.根据权利要求10所述的药物组合物,所述药物组合物以3:2:5的摩尔比含有DOPE、DOPC及DC-6-14。
12.根据权利要求7~11中任一项所述的药物组合物,其中,shRNA由以序列号8表示的碱基序列构成。
13.根据权利要求7~12中任一项所述的药物组合物,所述药物组合物与癌症化疗剂并用。
14.一种组合物,所述组合物含有权利要求7~13中任一项所述的药物组合物和癌症化疗剂。
15.根据权利要求14所述的组合物,其中,癌症化疗剂选自由具有微管蛋白的解聚抑制作用的抗肿瘤剂、脱氧胞苷衍生物及具有TS抑制作用的抗肿瘤剂构成的组。
