Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to application of radix sophorae tonkinensis and an extract thereof.
Background
Streptococcus suis (Streptococcus suis) is a general term for epidemic diseases in pigs caused by Streptococcus equi subsp zooepidemicus, Streptococcus equi subsp, group D, E, L streptococci in Lancefield grouping, and Streptococcus suis. Clinically, the symptoms are mainly lymphadenectasis, meningitis, arthritis, septicemia and the like. The streptococcus suis is the main pathogen causing streptococcus suis, and can cause epidemic diseases such as swine meningitis and septicemia, and people can also infect the streptococcus suis through a specific transmission path. Kilpper-Balz and Schleifer J suggested a new group of S.suis in 1987. In recent years, pig farms in the Wuzhishan mountain of Hainan have mixed infection of streptococcicosis and other diseases, which causes certain loss. Although there are theoretically many drugs that can treat swine streptococcosis, such as penicillin, the drug resistance of pathogenic bacteria is becoming more serious with the widespread use of antibacterial drugs and the unreasonable clinical application.
Chinese patent CN105267916A discloses a veterinary drug for treating swine streptococcosis, which is a pure traditional Chinese medicine preparation prepared from 50g of astragalus root, 40g of emblic leafflower fruit, 15g of finger citron, 10g of great burdock achene, 8g of citron, 7g of sea-buckthorn, 7g of black sesame, 5g of nutmeg, 100g of houttuynia cordata, 45g of dried ginger, 20g of medlar, 15g of dandelion, 10g of plantain herb, 6g of fenugreek, 5g of rhodiola rosea, 4g of fructus evodiae, 4g of gordon euryale seed and 3g of pepper, and can effectively treat swine streptococcosis and avoid the drug resistance problem of current antibacterial drugs. However, the problem is that the existing traditional Chinese medicine products use more traditional Chinese medicine components, about 20 traditional Chinese medicine components are used in the above patents, which are disadvantageous in terms of production simplicity and cost, and the similar patents also contain CN105267797A and CN 105147972A. Therefore, there is a need to provide a simple and effective drug for the control of streptococcus suis.
Disclosure of Invention
In view of the above, the present invention aims to provide application of croton argentea and its extract, so that the croton argentea and its extract have significant efficacy in treating streptococcus suis, have high sensitivity to streptococcus suis, and can be used for preparation of related medicines.
Croton cascarlloides Raeusch, Euphorbiaceae (Euphorbiaceae) Croton L, the alias of the leaf of Croton L, is the common herb in Hainan Li. Growing in river valleys or seaside shrubs or sparse forests below 500 m in elevation. Produced in Taiwan, Fujian, Guangdong, Hainan, Guangxi and Yunnan provinces. According to the application, the invention provides a medicament for treating and/or preventing swine streptococcosis, which comprises the silver leaf croton root or the silver leaf croton root extract. Wherein, the radix crotonis argenteae extract is preferably water extract, organic solvent extract or mixture of the two, and the organic solvent extract is more preferably ethanol extract of radix crotonis argenteae. The medicine is preferably injection, capsule, tablet or powder injection.
In addition, the invention also provides a croton argentea extract, which is a water extract, an organic solvent extract or a mixture of the water extract and the organic solvent extract of the croton argentea. Preferably, the organic solvent extract is an ethanol extract.
Meanwhile, the invention also provides an extraction method of the croton argentea extract, which is characterized in that the croton argentea is extracted by adding water or an organic solvent, and the obtained water extract, organic solvent extract or the combination of the water extract and the organic solvent extract is the croton argentea extract.
The further extraction scheme is that the croton argentea extract is obtained by soaking croton argentea in ethanol, heating for extraction, repeating extraction twice, mixing filtrates after each filtration, concentrating under reduced pressure, and recovering ethanol solution to fluid extract.

The invention takes the traditional Chinese medicine preparation in the patent CN105267916A as a contrast, and clinical observation and treatment are carried out on sick pigs with streptococcus suis by adopting the croton argentea ethanol extract extracted by the invention, and the result shows that the total effective rate and the contrast of the treatment of the croton argentea ethanol extract provided by the invention are both 100%, but the invention has more advantages in terms of the dosage. Meanwhile, the invention also detects the drug sensitivity to the streptococcus suis together with penicillin G, compound dixin and Junbizhi, and the result shows that the ethanol extract of the croton argentea has high sensitivity to the streptococcus suis, while penicillin G generates drug resistance.
According to the technical scheme, the application of the croton argentea roots and the extract thereof in preparing the medicament for treating and/or preventing the streptococcus suis is provided, the remarkable treatment effect is achieved by using a single traditional Chinese medicine component, the sensitivity to the streptococcus suis is high, and a new way is developed for preventing and treating the streptococcus suis.
Detailed Description
The invention discloses application of the radix sophorae tonkinensis and the extract thereof, and can be realized by appropriately improving process parameters by taking the contents as reference by the technical personnel in the field. Example 2: animal testing
Control group 1: the patent CN105267916A provides a veterinary drug for treating swine streptococcicosis, which comprises 50g of astragalus, 40g of emblic leafflower fruit, 15g of fingered citron, 10g of great burdock achene, 8g of citron, 7g of sea-buckthorn, 7g of black sesame, 5g of nutmeg, 100g of houttuynia cordata, 45g of dried ginger, 20g of medlar, 15g of dandelion, 10g of plantain herb, 6g of fenugreek, 5g of rhodiola rosea, 4g of fructus evodiae, 4g of gordon euryale seed and 3g of pepper.
Putting the traditional Chinese medicinal materials in the weight into a decocting tool, and adding 5000 milliliters of purified water; soaking for 30 min to thoroughly moisten Chinese medicinal materials, boiling with strong fire, decocting with slow fire for 1 hr, filtering to remove residue to obtain Chinese medicinal liquid; heating and concentrating the traditional Chinese medicine liquid to 2000 ml to obtain the veterinary medicine for treating the swine streptococcosis.
Control group 2: 50g of astragalus membranaceus, 40g of phyllanthus emblica, 15g of fingered citron, 10g of burdock, 8g of citron, 7g of sea-buckthorn, 7g of black sesame and 5g of nutmeg. The preparation method is the same as that of the control group 1.
Test groups: 10g of the drug obtained by the production method of example 1 was added with 10ml of sterile purified water to prepare a drug solution having a concentration of about 1 g/ml.
Clinical data: the clinical observation and treatment are carried out on 62 sick pigs suffering from the streptococcus suis disease in Wuzhishan city, Hainan province, and the main disease symptoms are as follows: fever, decreased appetite, lameness or incapability of standing, shortness of breath, rhinorrhea, conjunctival flushing and the like, swine infectious atrophic rhinitis, swine contagious pleuropneumonia and porcine reproductive and respiratory syndrome are eliminated, and the diagnosis can be confirmed by finding gram-positive streptococcus through pathogenic microorganism detection. The sick pigs diagnosed with streptococcicosis suis were divided into 3 treatment groups, 21 pigs in the first group, 21 pigs in the second group, and 20 pigs in the third group. The first group was treated with the test group drugs; the second group was treated with the veterinary drug of control group 1; the third group was treated with the veterinary drug of control group 2.

(3) The treatment effect is as follows:
a first group: 18 healed heads, accounting for 85%; effective 3 heads, accounting for 15%; invalid 0 head; the total effective rate is 100%.
Second group: cure 19 heads, accounting for 90%; 2 effective heads, accounting for 10%; invalid 0 head; the total effective rate is 100%.
Third group: 12 heads are cured, accounting for 60%; effective 6 heads, accounting for 30%; invalid 2 heads; the total effective rate is 90%.
The clinical tests show that the radix crotonalis argenteae has obvious curative effect on the swine streptococcosis, the cure rate is as high as 85%, the total effective rate is 100%, the effect is equivalent to that of the second group, but the traditional Chinese medicine components are far less than that of the existing product.
Example 3: in vitro drug susceptibility testing

2. Test method
A paper diffusion method is adopted. The stored purified strains were taken up at 37 days respectivelyCulturing in nutritive bouillon of 'shi' degree for 18-24 hr, diluting with aseptic normal saline to 2X 106pieces/mL, spread evenly on nutrient agar plates. Each flat plate is stuck with 4 pieces of drug sensitive paper, the distance between the two paper pieces is 25mm, the distance between the paper piece and the edge of the flat plate is 10 mm-15 mm, the center of the paper piece is lightly pressed to ensure tight adhesion, and the flat plate is cultured for 24 hours at the constant temperature of 37 ℃.
