本发明涉及一种以无名葵、刺五加、五味子、菌陈、瓜蒌、柴胡、枳实、夏枯草、葛根、川芎、山茱萸、当归、甘草、甜叶菊、黄芪、马齿苋等药食同源中草药为主要原料具有润肠通便、健脾补肺、益胃补肾、防治便秘功效的无名葵保健饮料及制备方法,属于保健食品领域。
背景技术
随着人民生活水平的提高,生活方式有了相应的转变,特别体现在饮食精细、热量充足,体力活动减少,随之而来的却是慢性非传染性疾病患病率的增加。除了人们熟知的心血管疾病、糖尿病等疾病外,便秘的患病率也逐渐升高。便秘是一种常见病,以粪便干燥、排便困难或不畅、排便次数减少为特征。便秘带给人类的健康危害是多方面的,不仅能诱发肛肠的痔、裂,而且是结肠癌、肝性脑病、乳腺疾病的危险因素;便秘经常导致心血管病人的碎死。我国便秘的患病率接近20%,美国的患病率高达28%。流行病学调查资料显示我国60岁以上老人便秘的患病率为8.7%~19.5%;7~12岁儿童的患病率为4.4%;社区人群中便秘患者占17.6%;女性患病率是男性的4倍以上。这些数字表明便秘已成为我国病源群最广、发病率最高的病种之一。
无名葵,锦葵科,草本一年生,在200多个秋菊植物中最具食用,药用,保健功效,有极高的利用价值。除观赏绿化环境外,珍惜之处在于全株无一废弃物,芽、苗、嫩尖、花蕾、嫩果富含蛋白质、高聚合原胶、Ve、Vc及矿物质,可作为新品种供应市场,家庭及饭店可炒菜,过水凉拌,煲汤等,荤素皆宜。口感风味独特,粘甜滑溜,无任何蔬菜与其相雷同,食之助消化,润肠理气使胃肠中废祛物通畅排出,有效的抑制毒素被吸收,控制低密度蛋白质提高,或脂肪堆积对人体的危害;尤其治疗老年人便秘有立竿见影的效果,解脱排便困难之苦,强力清洗肠胃过盛的脂肪,利于老年人的身心健康。无名葵初放的花朵和籽仁提取生物总黄酮是一项重要新发现,无名葵黄酮含量居所有现在已知含黄酮植物之首,在此之前世界公认银杏和大豆黄酮含量最高,而新发现的无名葵却高出银杏和大豆500倍。生物黄酮对人体的重要性,早已被提高到非常重要的地位,有效抑制杀灭癌细胞和止血消炎作用,可全面预防和除治各种“混血”疾病,对人体产生的强大深远影响,且具有显著的补肾壮阳功效,因为无名葵被誉为天然“伟哥”;同时,VE的含量也比其它物种胜出,高出牛奶2000倍、鲜橙汁3000倍,独占鳌头;此外,Vc、蛋白、聚糖胶、植物纤维、不造化矿物硒、铁、锌等人体必须的微量元素也极其丰富。因此进行无名葵的综合利用,开发预防便秘的无名葵保健饮料,能够提高无名葵的加工附加值。
发明内容
本发明针对上述问题,提供一种基于无名葵的综合利用与保健饮料相结合的具有预防便秘的无名葵保健饮料。
为实现上述目的,本发明采用如下技术方案:
本发明提供一种具有治疗便秘功效的无名葵保健饮料,其特征在于:无名葵保健饮料由以下质量份数的原料组成:无名葵200份、瓜蒌10~20份、五味子10~20份、黄芪5~10份、刺五加10~20份、甜叶菊8~12份、枳实5~10份、夏枯草5~10份、葛根3~5份、川芎3~5份、当归3~5份、菌陈2~5份、甘草5~10份、山茱萸2~5份、党参2~3份、马齿苋1~3份、柴胡1~3份。
一种具有治疗便秘功效的无名葵保健饮料的制备方法,其特征在于具体包括以下步骤:
(1)无名葵预处理:将无名葵原料粉粹,过100目筛,用流动的饮用水清洗中药材原料3次,;
(2)中药草预处理:将各中药材原料中的杂质剔除,切碎成2~3mm;
(3)配料:根据无名葵保健饮料产品配方准确配料,加入原料质量比20~30倍纯净水;
(4)超声波辅助酶解:调整悬浮液的pH至5.5~7.5,按比例加入100~120U/gα-淀粉酶、150~200U/g风味蛋白酶、100~200U/g纤维素酶、200~250U/g木聚糖酶,在超声波辅助作用下酶解,酶解温度70~80℃,超声功率300W,期间采用搅拌机匀速搅拌,酶解时间4~6h;
(5)微生物发酵:超声波辅助酶解后,加入0.15%硫酸铵、0.10%磷酸二氢钾,接种活化后的酿酒酵母和乳酸杆菌,接种比例分别为3~5%、1~2%,在搅拌条件下发酵12~24h;
(6)超声波辅助二次浸提:发酵结束后在超声波辅助作用下二次浸提10~20min,浸提温度控制在60~80℃;
(7)离心:离心得上清液,滤渣加入3~5倍纯净水,离心,合并上清液即为浸提液;
(8)配料:向浸提液中按1~1.5g/L蜂蜜、1~2g/L乳酸钙、5~10g/L柠檬酸、10~20g/L黄原胶、50mg/L六偏磷酸钠、50mg/L Vc、50mg/L山梨酸钾比例加入配料并搅拌均匀;
(9)微孔膜过滤:用孔径为1.5μm的微孔膜过滤浸提液即得具有预防便秘无名葵保健饮料成品;
(10)无菌灌装封口:在无菌灌装生产线中灌装封口。
本发明提供一种具有治疗便秘功效的无名葵保健饮料,饮料色泽均匀一致,呈浅黄色,澄清透明,有光泽,酸甜适中,无异味;饮料中固形物含量大于8.1%,低聚木糖含量2.52mg/mL,蛋白质含量4.12 mg/mL,菌落总数≤10cfu/m L,大肠菌群≤3 MPN/100mL,霉菌≤3cfu/mL,酵母≤3cfu/mL,无致病菌检出。
本发明提供一种具有治疗便秘功效的无名葵保健饮料,通过动物试验进行无名葵保健饮料的毒理学试验及安全性评价。小鼠急性经口毒性试验结果表明,本无名葵保健饮料对小鼠急性经口最大耐受剂量大于高剂量组75mg/kg·bw; 遗传毒性试验小鼠骨髓细胞微核试验和小鼠精子畸形试验两项试验结果均为阴性,表明该无名葵保健饮料无致突变作用;由大鼠30d喂养试验可知,大鼠体征行为正常、体态健康、血液及血液生化指标也无异常变化,因此75mg/kg·bw的剂量(人体推荐量的150倍)属于安全剂量;遗传毒性试验和大鼠30 d喂养试验结果均为阴性,因此确定本保健饮料属无毒类食品,人可以安全放心食用。
本发明提供一种具有治疗便秘功效的无名葵保健饮料,不同的中草药性味各异,功效存在着巨大的差异,不同的中草药组合后存在复杂的相互作用关系,且中草药之间的配比比较密切,不同配比也将使君臣佐使关系发生实质性的变化,从而使复合中草药的关系发生实质性改变。本申请,而且通过无名葵与瓜蒌、五味子、黄芪、刺五加、甜叶菊、当归、菌陈、甘草、山茱萸、马齿苋等中草药彼此间存在复杂的相互作用关系,合理配比,取得了预料不到的技术效果,使具有预防便秘无名葵保健饮料的关系发生实质性改变,无名葵复合发酵型无名葵保健饮料的总抗氧化能力达到455.87±21.55U/mL,改变中草药配比或中草药组成会降低无名葵保健饮料的抗氧化能力。本申请通过小鼠试验进行了饮料的缓解便秘效果,试验结果表明
本发明产品经口给予便秘小鼠模型后,能够缩短便秘小鼠首粒黑便的时间23.76%以上,增加排便的粒数32.17,证明具有较好的辅助通便、缓解便秘的功能。此外动物试验结果表明改变原料组成对中草药功效具有较大的影响(P<0.01)。
本发明提供一种具有治疗便秘功效的无名葵保健饮料,其优势在于:一是采用超声波辅助酶解和微生物发酵相结合的工艺提取中草药活性物质,提高了原料纤维素、木聚糖的降解率和功能性活性物质的提取率,提高了具有预防便秘无名葵保健饮料的营养价值;二是产品具有无名葵低聚木糖及瓜蒌、五味子等中草药的润肠通便、健脾补肺、益胃补肾、防治便秘功效等功效;三是具有预防便秘无名葵保健饮料产品中添加了蜂蜜、乳酸钙、柠檬酸、黄原胶、六偏磷酸钠、Vc、山梨酸钾等配料,产品适口性好,延长了产品的货架期;四是用微孔膜过滤替代传统的巴士灭菌法,灭菌效果彻底,避免了高温带来的营养物质损失,产品澄清度高;五是生产工艺提取率高,对设备要求相对较低,便于大规模工业化生产。
具体实施方式
实施例1:
一种具有治疗便秘功效的无名葵保健饮料的制备方法,具体包括以下步骤:
(1)无名葵预处理:将无名葵原料粉粹,过100目筛,用流动的饮用水清洗中药材原料3次,;
(2)中药草预处理:将各中药材原料中的杂质剔除,切碎成2~3mm;
(3)配料:根据无名葵200份、瓜蒌15份、五味子15份、黄芪7.5份、刺五加15份、甜叶菊10份、枳实7.5份、夏枯草7.5份、葛根4份、川芎4份、当归4份、菌陈3.5份、甘草7.5份、山茱萸3.5份、党参2.5份、马齿苋2份、柴胡2份比例准确配料,加入原料质量比25倍纯净水;
(4)超声波辅助酶解:调整悬浮液的pH至5.5~7.5,按比例加入110U/gα-淀粉酶、180U/g风味蛋白酶、150U/g纤维素酶、225U/g木聚糖酶,在超声波辅助作用下酶解,酶解温度70~80℃,超声功率300W,期间采用搅拌机匀速搅拌,酶解时间5h;
(5)微生物发酵:超声波辅助酶解后,加入0.15%硫酸铵、0.10%磷酸二氢钾,接种活化后的酿酒酵母和乳酸杆菌,接种比例分别为4%、1.5%,在搅拌条件下发酵18h;
(6)超声波辅助二次浸提:发酵结束后在超声波辅助作用下二次浸提15min,浸提温度控制在60~80℃;
(7)离心:离心得上清液,滤渣加入4倍纯净水,离心,合并上清液即为浸提液;
(8)配料:向浸提液中按1.2g/L蜂蜜、1.5g/L乳酸钙、7.5g/L柠檬酸、15g/L黄原胶、50mg/L六偏磷酸钠、50mg/L Vc、50mg/L山梨酸钾比例加入配料并搅拌均匀;
(9)微孔膜过滤:用孔径为1.5μm的微孔膜过滤浸提液即得具有预防便秘无名葵保健饮料成品;
(10)无菌灌装封口:在无菌灌装生产线中灌装封口。
实施例2:
一种具有治疗便秘功效的无名葵保健饮料的制备方法,具体包括以下步骤:
(1)无名葵预处理:将无名葵原料粉粹,过100目筛,用流动的饮用水清洗中药材原料3次,;
(2)中药草预处理:将各中药材原料中的杂质剔除,切碎成2~3mm;
(3)配料:根据无名葵200份、瓜蒌20份、五味子10份、黄芪10份、刺五加10份、甜叶菊12份、枳实5份、夏枯草5份、葛根3份、川芎5份、当归3份、菌陈5份、甘草5份、山茱萸5份、党参2份、马齿苋3份、柴胡1份比例准确配料,加入原料质量比30倍纯净水;
(4)超声波辅助酶解:调整悬浮液的pH至5.5~7.5,按比例加入120U/gα-淀粉酶、200U/g风味蛋白酶、200U/g纤维素酶、250U/g木聚糖酶,在超声波辅助作用下酶解,酶解温度70~80℃,超声功率300W,期间采用搅拌机匀速搅拌,酶解时间4h;
(5)微生物发酵:超声波辅助酶解后,加入0.15%硫酸铵、0.10%磷酸二氢钾,接种活化后的酿酒酵母和乳酸杆菌,接种比例分别为5%、2%,在搅拌条件下发酵12h;
(6)超声波辅助二次浸提:发酵结束后在超声波辅助作用下二次浸提10min,浸提温度控制在60~80℃;
(7)离心:离心得上清液,滤渣加入5倍纯净水,离心,合并上清液即为浸提液;
(8)配料:向浸提液中按1.5g/L蜂蜜、1g/L乳酸钙、10g/L柠檬酸、10g/L黄原胶、50mg/L六偏磷酸钠、50mg/L Vc、50mg/L山梨酸钾比例加入配料并搅拌均匀;
(9)微孔膜过滤:用孔径为1.5μm的微孔膜过滤浸提液即得具有预防便秘无名葵保健饮料成品;
(10)无菌灌装封口:在无菌灌装生产线中灌装封口。
实施例3:
一种具有治疗便秘功效的无名葵保健饮料的制备方法,具体包括以下步骤:
(1)无名葵预处理:将无名葵原料粉粹,过100目筛,用流动的饮用水清洗中药材原料3次,;
(2)中药草预处理:将各中药材原料中的杂质剔除,切碎成2~3mm;
(3)配料:根据无名葵200份、瓜蒌10份、五味子20份、黄芪5份、刺五加20份、甜叶菊8份、枳实10份、夏枯草5份、葛根5份、川芎3份、当归5份、菌陈2份、甘草10份、山茱萸2份、党参3份、马齿苋1份、柴胡3份比例准确配料,加入原料质量比20倍纯净水;
(4)超声波辅助酶解:调整悬浮液的pH至5.5~7.5,按比例加入100U/gα-淀粉酶、150U/g风味蛋白酶、100U/g纤维素酶、200U/g木聚糖酶,在超声波辅助作用下酶解,酶解温度70~80℃,超声功率300W,期间采用搅拌机匀速搅拌,酶解时间6h;
(5)微生物发酵:超声波辅助酶解后,加入0.15%硫酸铵、0.10%磷酸二氢钾,接种活化后的酿酒酵母和乳酸杆菌,接种比例分别为3%、1%,在搅拌条件下发酵24h;
(6)超声波辅助二次浸提:发酵结束后在超声波辅助作用下二次浸提20min,浸提温度控制在60~80℃;
(7)离心:离心得上清液,滤渣加入3倍纯净水,离心,合并上清液即为浸提液;
(8)配料:向浸提液中按1g/L蜂蜜、2g/L乳酸钙、5g/L柠檬酸、20g/L黄原胶、50mg/L六偏磷酸钠、50mg/L Vc、50mg/L山梨酸钾比例加入配料并搅拌均匀;
(9)微孔膜过滤:用孔径为1.5μm的微孔膜过滤浸提液即得具有预防便秘无名葵保健饮料成品;
(10)无菌灌装封口:在无菌灌装生产线中灌装封口。

1.一种具有治疗便秘功效的无名葵保健饮料的制备方法,其特征在于具体包括以下步骤:
(1)无名葵预处理:将无名葵原料粉粹,过100目筛,用流动的饮用水清洗中药材原料3次;
(2)中药草预处理:将各中药材原料中的杂质剔除,切碎成2~3mm;
(3)配料:根据无名葵200份、瓜蒌10~20份、五味子10~20份、黄芪5~10份、刺五加10~20份、甜叶菊8~12份、枳实5~10份、夏枯草5~10份、葛根3~5份、川芎3~5份、当归3~5份、菌陈2~5份、甘草5~10份、山茱萸2~5份、党参2~3份、马齿苋1~3份、柴胡1~3份配方准确配料,加入原料质量比20~30倍纯净水;
