Abstract

【技术领域】
本发明涉及中兽药技术领域,尤其是一种治疗鸡慢性呼吸道病的中药制剂及其制备方法。
【技术背景】
鸡慢性呼吸道病是由鸡败血支原体感染引起的慢性呼吸道传染病,又称鸡败血霉形体病、鸡败血支原体病,其是养鸡生产中较为常发且影响面较广的一种慢性、消耗性、条件性疾病,发病率为30%-60%,死亡率一般有10%-30%,是养鸡业和兽医诊疗人员在临床上很棘手的问题之一。鸡慢性呼吸道病全年各季均可发生,但以寒冬及早春最为严重,生产性强应激或遇寒感冒是促发该病的主要诱因,其主要发病症状表现为呼吸罗音、咳嗽、流泪、眼擦翅膀、爪抓眼、鼻流清液、甩鼻,后期可见颜面部肿胀,特别是眶下窦肿胀。解剖病理变化主要是鼻窦、气管、气囊病变;鼻窦有黏液,粘膜淤血;眼结膜淤血;气管黏膜充血、有黏液;肺脏一侧或两侧近支气管端淤血,后期喉头、气管、支气管内有灰白色或淡黄色干酪物;气囊浑浊、污秽变厚,有时有黄色干酪物附着;发病后期多继发大肠杆菌、沙门氏菌等肠道致病菌感染,继发大肠杆菌病后,出现纤维素性心包炎、肝周炎和腹膜炎、输卵管炎、卵巢炎等。鸡慢性呼吸道病可引起鸡生长速度明显降低、料肉比明显提高、鸡蛋产蛋率降低、畸形蛋增多、种蛋孵化率降低、健雏率下降等严重后果。
目前,治疗鸡慢性呼吸道病的药物很多,采用较多的是抗菌素类西药,该类药虽然具有一定的疗效,但长期使用会产生较强的耐药性,而且易在鸡体内留下化学药物残留,有些还会产生一定的副作用,不仅影响鸡的生长和生产性能,也给鸡产品食用者的健康带来潜在威胁。中草药治疗强调从整体上调节机体功能、提高机体自身的抗病能力,且中草药具有天然、毒副作用小、无化学残留、不易产生抗药性的特点,长期使用安全可靠,故将相应的中草药应用于养鸡业疾病的防治,具有广泛的应用前景。但目前市售的用于鸡慢性呼吸道病的中草药制剂还较少,而现有的中药制剂使用效果差,治愈率低,疗程长且成本高。
【发明内容】
针对以上提出的问题,本发明提供了一种治疗鸡慢性呼吸道病的中药制剂及其制备方法,该中药制剂以纯天然的中草药原料制备而成,可预防可治疗,成本低廉,效果明显,治愈率高,疗程短,安全可靠,毒副作用低,无残留,且不易产生抗药性。
为了实现以上目的,本发明采取的技术方案为:
一种治疗鸡慢性呼吸道病的中药制剂,所述中药制剂包括如下重量份的原料:蒲公英10-30份、野菊花10-15份、白头翁15-20份、秦皮6-12份、芙蓉叶10-18份、甘草2-10份、车前子5-15份、天浆壳6-9份、仙人球10-20份、水杨梅根15-30份、大血藤9-15份、野马追15-20份、椿皮6-15份、龙眼叶9-15份。
进一步地,所述中药制剂包括如下重量份的原料:蒲公英15-25份、野菊花12-14份、白头翁17-19份、秦皮8-10份、芙蓉叶12-16份、甘草4-8份、车前子8-12份、天浆壳7-8份、仙人球13-17份、水杨梅根20-25份、大血藤11-13份、野马追16-18份、椿皮9-12份、龙眼叶11-13份。
进一步地,所述中药制剂包括如下重量份的原料:蒲公英20份、野菊花13份、白头翁18份、秦皮9份、芙蓉叶14份、甘草6份、车前子10份、天浆壳7份、仙人球15份、水杨梅根22份、大血藤12份、野马追17份、椿皮11份、龙眼叶12份。
进一步地,所述中药制剂为颗粒剂、片剂或粉剂。
本发明还提供上述中药制剂的制备方法,包括以下步骤:
1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:将步骤1)所取并处理后的原料用体积浓度为60-80%的乙醇回流提取,过滤,收集提取液,再回收乙醇,进而浓缩成25℃时相对密度为1.1-1.2的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;
3)水提取:向步骤2)醇提取所剩余的药渣中加水煎煮提取,而后过滤,再将所得药液浓缩成25℃时相对密度为1.1-1.2的清膏,得水提取物;
合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入药学上可以接收的辅料,制成颗粒剂、片剂或粉剂。
进一步地,在步骤2)中,所述醇提取的具体方法为:向所述步骤1)所取并处理后的原料中加入原料总质量6-8倍且体积浓度为60-80%的乙醇浸泡1-3h,随后回流提取两次,每次1-2h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.1-1.2的浸膏,得醇提取物。
进一步地,在步骤3)中,所述水提取的具体方法为:向步骤2)醇提取所剩余的药渣中加入原料总重量5-7倍的水,武火煮沸后转文火煎煮20-30min,过滤取药液,药渣则采用相同的方法继续煎煮两次,合并三次煎煮所得药液,再将合并后的药液浓缩成25℃时相对密度 为1.1-1.2的清膏,得水提取物。
进一步地,在步骤4)中,所述药学上可接受的辅料为乳糖、可压性淀粉、糊精、微晶纤维素和羟丙基纤维素中的两种或以上。
本发明所采用的中药详细情况如下:
蒲公英:为菊科植物蒲公英的带根全草。其中含有蒲公英醇、蒲公英素、胆碱、有机酸、菊糖等多种健康营养成分,同时含有蛋白质、脂肪、碳水化合物、微量元素及维生素等,营养价值丰富。其味苦、甘,性寒。归肝经、胃经。具有清热解毒、利尿散结、缓泻、清肺、利嗽化痰、散结消痈、养阴凉血、舒筋固齿的功效;对金黄色葡萄球菌耐药菌株、溶血性链球菌有较强的杀菌作用,对肺炎双球菌、脑膜炎球菌、白喉杆菌、绿脓杆菌、变形杆菌、痢疾杆菌、伤寒杆菌等及蒲公英卡他球菌亦有一定的杀菌作用。可用于治淋巴腺炎,瘰疬,疔毒疮肿,急性结膜炎,感冒发热,急性扁桃体炎,急性支气管炎,胃炎,肝炎,胆囊炎,尿路感染。
野菊花:为菊科植物野菊的干燥头状花序。味苦、辛,性微寒。归肝经、心经。具有疏风清热、降火解毒、破血疏肝、消肿排脓和抗菌消炎的功效;对金黄色葡萄球菌、白喉杆菌、链球菌、绿脓杆菌、蒺疾杆菌、流感病毒均有抑制作用。可治风热感冒,肺炎、白喉,胃肠炎,疔,痈,口疮,丹毒,湿疹,天泡疮。
白头翁:为毛茛科植物白头翁的干燥根。其中含有原白头翁素,根含三萜皂苷;味苦,性寒。归胃经、大肠经。具有清热解毒、凉血止痢、消瘀、抗菌抗病毒和提高机体免疫力的功效,可抑制溶组织阿米巴原虫生长,对金黄色葡萄球菌绿脓杆菌、痢疾杆菌、志贺氏菌有抑制作用。可治热毒血痢,温疟寒热,鼻衄,血痔。
秦皮:为木犀科植物苦枥白蜡树、白蜡树、尖叶白蜡树或宿柱白蜡树的干燥枝皮或干皮。味苦,性寒。入肝经、胆经。具有清热燥湿、平喘止咳、洗肝明目的功效;对金黄色葡萄球菌、大肠杆菌、福氏痢疾杆菌、宋内痢疾杆菌均有抑制作用,对流感病毒、疱疹病毒等病毒亦有一定的抑制作用。可用于治细菌性痢疾,肠炎,慢性气管炎,目赤肿痛,迎风流泪,牛皮癣。
芙蓉叶:为锦葵科植物木芙蓉的叶。含黄酮甙、酚类、氨基酸、还原糖、粘液质。味微辛,性凉。归肺、肝经。具有清肺凉血、散热解毒、消肿排脓的功效。用于肺热咳嗽,瘰疬,肠痈。
甘草:为豆科甘草属植物甘草的根和根状茎。味甘,性平。入脾经、胃经、肺经。具有益气和中、缓急止痛、润肺止咳、泻火解毒、调和诸药以及抗炎、抗过敏的功效。可治脾胃虚弱、食少、腹痛便溏、劳倦发热、肺痿咳嗽、心悸、惊痫;以及咽喉肿痛、消化性溃疡、痈疽疮疡、解药毒和食物中毒。
车前子:为车前草科植物车前或平车前的干燥成熟种子。味甘,性寒。入肾经、膀胱经。具有利水通淋、利尿止泻、清肝明目、清肺化痰、镇咳平喘的功效;对同心性毛癣菌、许兰黄癣菌、奥杜盎小芽胞癣菌、铁锈色小芽胞癣菌、羊毛状小芽胞癣菌、星形奴卡菌等皮肤真菌有不同程度的抑制作用;同时,对多种致病菌,如金黄色葡萄球菌、宋内痢疾杆菌、大肠杆菌、绿脓杆菌及伤寒杆菌亦有不同程度的抑制作用。用于水肿胀满,热淋涩痛,暑湿泄泻,目赤肿痛,痰热咳嗽。
天浆壳:为萝藦科植物萝藦的干燥果壳。味甘、辛,性温。归肺、肾经。具有清肺化痰、止咳平喘、散瘀止血的功效。主咳嗽痰多,气喘,百日咳,惊痫,麻疹不透。
仙人球:为仙人掌科植物仙人球的茎。含有三萜、苹果酸、琥珀酸、碳酸钾等成分。味甘淡,性平。归肺、胃经。具有清热止咳、凉血解毒、消痈散结、消肿止痛、行气活血、趋势退热的功效。可治肺热咳嗽,痈肿。
水杨梅根:为双子叶植物药茜草科植物细叶水团花的根。含水杨梅甙,味苦、辛,性凉。归肝、肺、肾经。具有清热解表、活血解毒、抗菌消炎的功效。主治感冒发热,咳嗽,腮腺炎,咽喉肿痛。
大血藤:为木通科植物大血藤的干燥藤茎。味苦,性平。归肝经、大肠经。具有败毒消痈、活血通络、祛风杀虫、抗菌消炎、消肿散结、强筋壮骨的功效;对金黄色葡萄球菌、乙型链球菌有极敏感抑菌作用;对大肠杆菌、绿脓杆菌、甲型链球菌、卡他球菌、白色葡萄球菌均有高敏感抑菌作用。可治急、慢性阑尾炎,风湿痹痛,赤痢,虫痛。
野马追:为菊科植物轮叶泽兰的干燥地上部分。含黄酮类、生物碱、香豆粗等,味苦,性平。归肺经。具有化痰、止咳平喘的功效,对流感杆菌、甲型链球菌、金黄色葡萄球菌及卡他球菌有抑制作用。用于痰多咳嗽气喘,慢性气管炎,支气管炎。
椿皮:为苦木科植物臭椿的干燥根皮或干皮。味苦、涩,性寒。归大肠经、胃经、肝经。具有除热、可燥湿、涩肠、止血、杀虫的功效,对福氏痢疾杆菌、宋氏痢疾杆菌和大肠杆菌有抑制作用。治久泻,久痢,肠风便血,蛔虫。
龙眼叶:为无患子科植物龙眼的叶或嫩芽。含谷甾醇、豆甾醇、豆甾醇葡萄糖甙,表无羁萜醇、16-卅一烷醇、槲皮素、槲皮甙。味甘淡,性平。具有发表清热、解毒、燥湿的功效。可治感冒发热,疟疾。
本发明提供了一种治疗鸡慢性呼吸道病的中药制剂及其制备方法,与现有技术相比,具有以下有益效果:

一种治疗鸡慢性呼吸道病的中药颗粒制剂,其包括如下重量份的原料:蒲公英10份、野菊花10份、白头翁15份、秦皮6份、芙蓉叶10份、甘草2份、车前子5份、天浆壳6份、仙人球10份、水杨梅根15份、大血藤9份、野马追15份、椿皮6份、龙眼叶9份。具体制备方法包括以下步骤:
1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:向步骤1)所取并处理后的原料中加入原料总质量6倍且体积浓度为80%的乙醇浸泡1h,随后回流提取两次,每次1h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.1的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;

1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:向步骤1)所取并处理后的原料中加入原料总质量6倍且体积浓度为75%的乙醇浸泡1.5h,随后回流提取两次,每次1h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.1的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;

4)合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入适量辅料可压性淀粉、糊精和微晶纤维素,混匀后采用湿法制粒、干燥,经整粒后压片制成片剂,每片0.3g。
实施例3
一种治疗鸡慢性呼吸道病的中药粉剂,其包括如下重量份的原料:蒲公英20份、野菊花13份、白头翁18份、秦皮9份、芙蓉叶14份、甘草6份、车前子10份、天浆壳7份、仙人球15份、水杨梅根22份、大血藤12份、野马追17份、椿皮11份、龙眼叶12份。具体制备方法包括以下步骤:
1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:向步骤1)所取并处理后的原料中加入原料总质量7倍且体积浓度为70%的乙醇浸泡2h,随后回流提取两次,每次1.5h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.15的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;
3)水提取:向步骤2)醇提取所剩余的药渣中加入原料总重量6倍的水,武火煮沸后转文火煎煮25min,过滤取药液,药渣则采用相同的方法继续煎煮两次,合并三次煎煮所得药液, 再将合并后的药液浓缩成25℃时相对密度为1.15的清膏,得水提取物;
4)合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入适量辅料可压性淀粉和糊精,混匀后干燥、粉碎成粉剂。
实施例4
一种治疗鸡慢性呼吸道病的中药颗粒剂,其包括如下重量份的原料:蒲公英25份、野菊花14份、白头翁19份、秦皮10份、芙蓉叶16份、甘草8份、车前子12份、天浆壳8份、仙人球17份、水杨梅根25份、大血藤13份、野马追18份、椿皮12份、龙眼叶13份。具体制备方法包括以下步骤:
1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:向步骤1)所取并处理后的原料中加入原料总质量7倍且体积浓度为65%的乙醇浸泡3h,随后回流提取两次,每次2h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.2的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;
3)水提取:向步骤2)醇提取所剩余的药渣中加入原料总重量7倍的水,武火煮沸后转文火煎煮30min,过滤取药液,药渣则采用相同的方法继续煎煮两次,合并三次煎煮所得药液,再将合并后的药液浓缩成25℃时相对密度为1.2的清膏,得水提取物;
4)合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入适量辅料乳糖、糊精和羟丙基纤维素并混匀,而后采用湿法制粒、干燥,经整粒后装袋,每袋6g,制成颗粒制剂。
实施例5
一种治疗鸡慢性呼吸道病的中药粉剂,其包括如下重量份的原料:蒲公英30份、野菊花15份、白头翁20份、秦皮12份、芙蓉叶18份、甘草10份、车前子15份、天浆壳9份、仙人球20份、水杨梅根30份、大血藤15份、野马追20份、椿皮15份、龙眼叶15份。具体制备方法包括以下步骤:
1)取原料:按照上述重量份称取所述原料,洗净晾干后,切断至长度小于1cm;
2)醇提取:向步骤1)所取并处理后的原料中加入原料总质量8倍且体积浓度为60%的乙醇浸泡3h,随后回流提取两次,每次1.5h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.15的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;
3)水提取:向步骤2)醇提取所剩余的药渣中加入原料总重量7倍的水,武火煮沸后转文火煎煮20min,过滤取药液,药渣则采用相同的方法继续煎煮两次,合并三次煎煮所得药液, 再将合并后的药液浓缩成25℃时相对密度为1.15的清膏,得水提取物;
4)合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入适量辅料乳糖、糊精和微晶纤维素,混匀后干燥、粉碎成粉剂。

2)醇提取:将步骤1)所取并处理后的原料用体积浓度为60-80%的乙醇回流提取,过滤,收集提取液,再回收乙醇,进而浓缩成25℃时相对密度为1.1-1.2的浸膏,得醇提取物;醇提取所剩余的药渣保留,备用;
3)水提取:向步骤2)醇提取所剩余的药渣中加水煎煮提取,而后过滤,再将所得药液浓缩成25℃时相对密度为1.1-1.2的清膏,得水提取物:
4)合并制药:将步骤2)所得醇提取物和步骤3)所得水提取物混合均匀,加入药学上可以接收的辅料,制成颗粒剂、片剂或粉剂。
6.如权利要求5所述的治疗鸡慢性呼吸道病的中药制剂的制备方法,其特征在于:在步骤2)中,所述醇提取的具体方法为:向步骤1)所取并处理后的原料中加入原料总质量6-8倍且体积浓度为60-80%的乙醇浸泡1-3h,随后回流提取两次,每次1-2h,过滤,收集并合并两次提取液,而后减压回收乙醇,再浓缩成25℃时相对密度为1.1-1.2的浸膏,得醇提取物。
7.如权利要求5所述的治疗鸡慢性呼吸道病的中药制剂的制备方法,其特征在于:在步骤3)中,所述水提取的具体方法为:向步骤2)醇提取所剩余的药渣中加入原料总重量5-7倍的水,武火煮沸后转文火煎煮20-30min,过滤取药液,药渣则采用相同的方法继续煎煮两次,合并三次煎煮所得药液,再将合并后的药液浓缩成25℃时相对密度为1.1-1.2的清膏,得水提取物。
8.如步骤5所述的治疗鸡慢性呼吸道病的中药制剂的制备方法,其特征在于:在步骤4)中,所述药学上可接受的辅料为乳糖、可压性淀粉、糊精、微晶纤维素和羟丙基纤维素中的两种或以上。
