Abstract

技术领域
本发明涉及分离的泛素化蛋白组合物和通过分离的泛素化蛋白组合物独自的给药或与补充疗法结合给药激活、增强或促进免疫反应的方法。
背景技术
通过宿主专业抗原呈递细胞(APCs)的外源性抗原的交叉呈递在肿瘤相关的抗原的T细胞免疫反应的激活和发展起到举足轻重的作用,肿瘤相关的抗原包括源自肿瘤细胞的自身抗原或突变自身抗原,以及源自传染源的外源性抗原。已经开发了准癌症疫苗,试图利用外源性抗原的交叉呈递引发针对肿瘤的特异性免疫反应。
自吞噬是一种细胞过程,其中细胞质部分被称为自噬体的尺寸为0.5-2μm的双模泡隔离。这些自噬体的内容分解为裂解室,这便于长寿命蛋白质的周转,并对于保持合成代谢所需的氨基酸池非常重要。自吞噬的引入的重要标记是微管相关蛋白1轻链3(LC3-I)的细胞质形式通过一系列的泛素样偶联步骤转化为与自噬体紧密相关的脂化形式(LC3-PE)。一种最近描述的蛋白,p62/SQSTM1(sequestosome或p62)与多聚泛素和LC3结合,从而便于通过自吞噬进行泛素化蛋白的降解。LC3与p62的相互作用已经增加了自噬网络的复杂性,并表明这种本体降解可以比之前理解地更具选择性。

发明内容

具体实施方式
详细的说明书涉及诱导哺乳动物中的特异性免疫反应的组合物和方法。具体地,说明书涉及组合物,该组合物包括含有一种或多种特异性抗原的分离的泛素化蛋白。例如,泛素化蛋白可以从肿瘤源细胞分离出,该肿瘤源细胞被在抑制蛋白酶体处降解泛素化蛋白。分离的泛素化蛋白可以包括高免疫原性抗原,如短寿命蛋白和缺陷核糖体产物,其可以直接被注射入哺乳动物体内,或者可以与树突状细胞和/或T细胞共培养,以衍生可以用作疫苗或另一种免疫组合物的细胞群。
非常理想的抗癌方法包括开发有效的治疗癌症的疫苗,其能够激活抵抗由多种类型的癌细胞表达或过度表达、而不会被正常健康细胞表达的多种肿瘤排斥抗原的光谱抗肿瘤T细胞反应。由于真实排斥抗原的识别仍是未知的,预期富集这些肿瘤排斥抗原的间接方法有助于开发有效的治疗癌症的疫苗。除了过表达、非突变、短寿命抗原可以作为肿瘤排斥抗原之外,很多癌症细胞产生独特的蛋白或误译的蛋白,误译的蛋白可以是突变、转录、翻译和/或翻译后修饰的错误。这些蛋白可以是短寿命的,且标记为蛋白酶体依赖的降解。详细的说明书公开了一种新型方法,该方法利用用于蛋白酶体介导的降解的通用聚泛素标记分离和富集肿瘤排斥抗原。分离的泛素化蛋白能够激活抗原特异性T细胞,并引发抗肿瘤T细胞介导的免疫反应。实际上,接种这些蛋白质能够介导在乳腺癌的临床前模型中建立的肿瘤的肿瘤消退。
图1A和1B示意地展示了存在或不存在蛋白酶体抑制剂时,用于运输泛素化蛋白的细胞路径。蛋白可以通过自噬路径或蛋白酶体路径有针对性地进行降解。通常,可以用聚泛素标记修饰短寿命蛋白(SLiPs)和缺陷核糖体蛋白(DRiPs),并通过热休克蛋白(例如HSP-90)护送至用于降解的蛋白酶体。蛋白酶体介导的蛋白降解路径在调节细胞信号和提供通过MHC-I限制的抗原呈递进行直接的细胞表面呈递的肽的T细胞中发挥重要作用。
被损坏或错误折叠的长寿命蛋白经常通过自噬路径处理。蛋白和损坏的细胞器被吞噬泡吞噬,吞噬泡随后用溶酶体溶解,发展为自噬体。在抗原呈递细胞中,自吞噬在用于内源性和外源性抗原的、MHC-II限制的抗原呈递中发挥重要作用。在吞噬细胞中,Toll样受体介导的识别和先天免疫反应的激活需要自吞噬。
HSP-90相关肽,如SLiPs和DRiPs包括高免疫原性肽。但是,它们快速被蛋白酶体降解,从而不一定有效地交叉呈递。通过利用抑制剂阻止蛋白酶体,可以保护SLiPs和DRiPs免受蛋白酶体降解,而进行自噬路径。如果也抑制自噬(例如通过氯化铵),那么可以亲和纯化SLiPs和DRiPs,并将其用作免疫原性化合物。
图2展示了泛素结合蛋白的例子。多聚泛素链的各单体的C末端上的特定赖氨酸(K)链决定标记蛋白的命运。最充足的多聚泛素链是K48链,其采用封闭结构,作为通过S5a降解的靶蛋白的信号,S5a是26S蛋白酶体的调节复合物(19S)的一种成分。下一个最常用多聚泛素链,K63,采用延长的线状结构,其可作为非蛋白酶体附着点。近年来描述了K63连接蛋白对肿瘤发生和神经退行性疾病中起作用。P62是一种内源性K63结合蛋白,其使这些蛋白穿转变为自噬体。P62也可以结合至K48连接蛋白。当阻止蛋白酶体时,K48连接蛋白的浓度增加,p62可以使K48连接蛋白以及K63连接蛋白转变为自噬体。从而重组S5a和p62可以用于从细胞裂解液中亲和纯化多聚泛素化蛋白。其他泛素结合蛋白,例如TUBES、NEMO、Optineurin、TOLLIP、TOM1、Vx3等可以亲和标记重组表达,以有效回收结合的泛素化蛋白。在本文中,含有结构体的S5a和Vx3用于从细胞裂解液中分离泛素化蛋白,但是,也可以使用其他结构体。具体地,K48和K63特异性蛋白、抗体、和/或其组合也可以用于分离泛素化蛋白的特定种群,用于下游申请。
实施例1:通过分离的泛素化蛋白的交叉呈递激活天然T-细胞
交叉呈递被确认为是这样的一种方式:抗原可以被没有合成这些抗原的细胞呈递,从而避免直接呈递作为激活免疫反应的唯一机理。通过证明在接种过程中,黑色素瘤抗原的交叉呈递对抗肿瘤免疫反应的有效的产生很重要来扩展这些发现。已经讨论过但仍未知的交叉呈递的一个组成部分是抗原源,和输送至专业抗原呈递细胞(APC)的方法。虽然一些团体已经表明抗原源为细胞蛋白,但是其他团体认为它是热休克蛋白(HSPs)陪伴的肽。进一步地,已经证明交叉致敏可以来自蛋白酶体基质的捐赠以及与HSP90结合的稳定细胞质肽。利用短寿命模型抗原系统以及蛋白翻译的抑制剂的实验表明短寿命蛋白(SLiPs)和/或缺陷核糖体产物(DRiPs)是与自噬体依赖性交叉致敏结合的抗原池的不可分割的成分。抗原池通常是低效的交叉呈递,因为SLiPs的短寿命防止它们捐献给APC。有趣的是,正是SLiPs的短暂的特性使得其组成细胞表面上的肽:MHC复合物中的大部分,因为它们迅速被蛋白酶体降解,并通过TAP1/2负载MHC I类分子上。利用蛋白酶体抑制剂(硼替佐米)治疗肿瘤可稳定这些短暂蛋白,并使其转变为其他主要降解路径,自噬体/溶酶体路径。
图3A示意地展示了GFP-Vx3(A7)融合蛋白。Vx3是一种肽,其包括三个相对紧凑的泛素结合Vps27 UIM域。Vx3通过具有较高的特异性的Lys63连接泛素物种结合Lys63连接泛素物种和Lys48连接泛素物种。从而,GFP-Vx3(A7)可用于从细胞裂解液中结合并亲和纯化泛素化蛋白。GFP-Vx3(A7)还包括N末端聚-His标记,其可用于亲和纯化蛋白。
在一个实施例中,使UbiLT3细胞在培养基中生长,并用200nM硼替佐米处理24小时,收获并裂解。将10、30或100μg GFP-Vx3(A7)加入UbiLT3裂解液中,共孵育过夜。然后将镍树脂加入所述混合物中,并共孵育过夜。离心分离镍树脂,保存上清液作为流过馏分。然后用低浓度咪唑洗涤镍树脂,然后用高浓度咪唑溶液洗涤结合蛋白。图3B展示了10、30或100μg的纯化的GFP-Vx3(A7)的输入、流过和洗脱馏分的数字图像。
随后分离的泛素化蛋白可呈递至树突状细胞(DCs),其然后可用于T细胞的交叉呈递抗原。在一个实施例中,人类DCs可负载有分离的泛素化蛋白,该分离的泛素化蛋白来自UbiLT3细胞,或设计为表达巨细胞病毒结构蛋白pp65的UbiLT3细胞(UbiLT3pp65)。将纯化的CMV蛋白负载至DCs,作为阳性对照,而单独地培养基(CM)作为阴性对照。DCs被负载6小时,然后添加扩张效应T细胞,并激活16小时。然后分析T细胞,用于通过细胞内染色和布雷菲德菌素-A处理的IFN-γ的生产,随后进行流式细胞术。图3C和3C展示了从UbiLT3pp65细胞(LT3pp65)而非UbiLT3对照细胞(LT3)中分离的泛素化蛋白能够激活CD8+T细胞(图3C)和CD4+T细胞(图3D)中产生的pp65-特异性IFN-γ。这些结果表明除了作为疫苗之外,分离的泛素化蛋白也可以用作体外检测物,并评估对疫苗、癌症和/或感染特定病原的免疫反应,例如通过如下步骤进行:将源自表达特异性抗原的细胞的分离的泛素化蛋白负载到树突状细胞,呈递负载的树突状细胞至从用抵抗特异性抗原的疫苗治疗的患者收获的T细胞,然后通过收获的T细胞评估IFN-γ的产生。
图4A和4B展示了类似实验的图,其中DCs的亚集负载有UbiLT3和UbiLT3pp65Ub-富集的输入和流过馏分。如图4A所示,对于UbiLT3pp65,洗脱的、分离的泛素化蛋白(洗脱液)通过CD8+T细胞激活IFN-γ的产生比单独的UbiLT3pp65细胞裂解液(输入)更有效。通过UbiLT3pp65细胞裂解液的流过馏分或UbiLT3的Ub富集的输入和流过馏分诱导pp65-特异性T细胞的最小化的激活。
图5展示了流式细胞术分布图,证明分离的泛素化蛋白是用于天然CD8+T细胞的抗原的交叉呈递的有效的抗原源。B78/H1的细胞,一类鼠B16黑素瘤细胞设计为表达R-GFP-OVA或R-GFP。使B78/H1细胞在培养基中生长,并用硼替佐米处理,收获并裂解。从B78/H1裂解液以及正常肝组织裂解液中亲和纯化泛素化蛋白。
鼠Mutu-1940 DCs负载有铝纳米离子以及纯化的泛素化蛋白(3,10,或30μg)或者纯化的OVA(10,30,或100μg)。然后该负载的DCs呈递至CFSE标记的OT-1转基因的T细胞。通过在共孵育6天之后,通过CFSE稀释评估T细胞的激活。
如图5所示,脉冲有(pulsed with)来自抗原表达细胞(Ub-Vel-B78H1-OVA)的分离的泛素化蛋白的DCs即使在低抗原浓度时也能有效地激活OT-1 CD8+T细胞,而来自非抗原表达细胞或正常肝组织的分离的泛素化蛋白则无效。实际上,在较低浓度时(3μg/ml),来自Ub-Vel-B78H1-OVA的泛素化蛋白以与浓度为10μg/ml的纯化的OVA相似的效果激活OT-1 CD8+T细胞。图5所示的数据来自一个独立的实验,但是被相似的结果所复制(未展示)。
这些实验证明可以在不存在其他细胞成分时,可以通过树突状细胞占用分离的泛素化蛋白,并将其交叉呈递至T细胞。从而,分离的泛素化蛋白可用于在体内或体外,激活(prime)天然T细胞中的免疫反应。
实施例2:通过分离的泛素化蛋白的交叉呈递激活肿瘤特异性T细胞
过去,癌症疫苗诱导T细胞很少能有效地消灭肿瘤。如果诱导的T细胞能设别肿瘤并代表肿瘤排斥抗原,那么它们往往隔绝、消散、凋亡,或者仅是不能扩展到能够根除肿瘤的水平。通过提供广泛的可用于激活抵抗肿瘤排斥抗原频谱的T细胞的、潜在的肿瘤排斥抗原,可增加扩展的T细胞的治疗功效。进一步地,抵抗肿瘤抗原的广泛的免疫反应将类似地降低肿瘤从免疫清楚逃离的可能性,因为肿瘤将需要同时下调大量抗原的表达。
图6A-6B展示了通过分离的泛素化蛋白激活的肿瘤特异性、Dribble致敏的T细胞的图。Nu77-GFPCre小鼠(C57BL/6小鼠的菌株,其表达脾、以及在T和B淋巴细胞中的髓样细胞中的GFP)接种零矢量(图6A)或源自B78/H1细胞的Dribbles(图6B)。接种7天后,处死小鼠,并收获脾。将铝纳米离子,以及PBS,或5、10或30μg的来自正常肝细胞的分离的泛素化蛋白(Ub-肝HS),或5、10或30μg的来自B78/H1细胞的分离的泛素化蛋白(Ub-B78H1 HS),5、10或30μg的来自硼替佐米处理的B78/H1细胞的分离的泛素化蛋白(VelUb-B78H1 HS),或5、10或30μg的源自B78/H1-OVA细胞的Dribbles负载至Mutu 1940 DCs。然后将负载的DC’s与分离的脾共孵育。然后通过CD8+、CD69+、GFP+细胞的流式细胞分析检查效应T细胞的活化。对于从此前接种的小鼠分离的脾,可以看到对于来自B78/H1细胞的分离的泛素化蛋白,效应T细胞的浓度依赖活化,以及对于来自硼替佐米处理的B78/H1细胞的分离的泛素化蛋白更加显著的效应T细胞的浓度依赖活化。
该实验表明分离的泛素化蛋白可以用作疫苗增强剂(vaccine booster)。例如,异体自噬体富集的组合物可以提供给荷瘤患者,然后提供包括源自患者自身原发肿瘤细胞的分离的泛素化蛋白的组合物。这样,可以增加疫苗的功效,因为在接种之后可以触发T细胞扩展。
图7为通过接种分离的泛素化蛋白介导的肿瘤特异性T细胞的体内致敏。为C57BL/6小鼠接种培养基(HS)、来自正常肝细胞的分离的泛素化蛋白(HSUb肝)或来自B78/H1细胞的分离的泛素化蛋白(HS Ub B78H1)。接种7天后,处死小鼠,并收获脾。然后将脾与B78/H1细胞、设计为表达H2-Db的B78/H1细胞(B78H1Db)、设计为表达H2-Kb的B78/H1细胞(B78H1Kb)、设计为表达H2-Db和H2-Kb的B78/H1细胞(B78H1DbKb)、或抗-CD3抗体(CD3)共孵育。然后通过CD8+,IFN-γ+细胞的流式细胞分析确定T细胞的活化。
如图7所示,小鼠接种有来自B78/H1细胞的分离的泛素化蛋白,其致敏T细胞,且该T细胞识别设计为表达H2-Db、H2-Kb或表达这两者的B78/H1细胞。该结果表明,接种来自B78/H1细胞的分离的泛素化蛋白有效地在体内致敏T细胞,该T细胞以MHC-I限制方式特异性地用于肿瘤类型。
实施例3:分离的泛素化蛋白激活的炎症反应
免疫佐剂依赖免疫炎症反应,以加强和/或调解免疫反应。图8展示了由分离的泛素化蛋白诱导的炎症反应。在106细胞/孔的96孔板中接种外周血单核细胞。然后用浓度为5μg/ml或50μg/ml的5mM ATP+10ng/ml脂多糖(ATP/LPS)或来自B78/H1细胞的分离的泛素化蛋白将PBMCs孵育过夜。然后取每个孔的上清液,并通过ELISA测试IL-1β。如图8所示,通过来自B78/H1细胞的分离的泛素化蛋白一浓度依赖的方式激活IL-1β。IL-1β促炎细胞因子,这种细胞因子的产生证明泛素化蛋白的炎症性。炎症反应的诱导是疫苗佐剂的共有性能,并用于增加疫苗特异性免疫反应的发展。从而分离的泛素化蛋白具有抗原和佐剂的性能。通过激活T细胞作用部位的炎症,源自分离的泛素化蛋白的疫苗将提供炎症反应,该炎症反应可增强疫苗特异性免疫反应的产生。
实施例4:通过分离的泛素化蛋白介导的MHC类1a依赖和独立的T细胞的交叉致敏
肿瘤逃离免疫监视的一个主要机理在于减少或完全消除MHC类1a分子的表达。同时,非经典MHC类1b分子经常在肿瘤细胞中被上调,作为通过NK细胞或CTL逃离消除的机理。致敏T细胞(识别独立于MHC类1a的肿瘤细胞)的疫苗从而可加宽被T细胞靶向对准的排斥抗原的频谱,从而降低肿瘤细胞逃离免疫消除的可能性。
B16黑色素瘤细胞的B78/H1克隆具有经典MHC类I分子(MHC类1a)的缺陷表达,但是保留非多态性MHC类I分子(MHC类1b)的表达。图9A-9D展示了流式细胞分析图,证明MHC类1a依赖和独立的CD8+T细胞可以被分离的泛素化蛋白交叉致敏。使Nurr77GFP记者小鼠接种15μg的Dribbles或来自B78H1黑色素瘤细胞的分离的泛素化蛋白。Dribbles通过内部(intra-nodally)给药,而分离的泛素化蛋白与铝纳米离子结合通过皮下给药。然后分析致敏CD8+T细胞识别亲代B78H1细胞和转染为表达外源性H2-Kb或Db分子的B78H1细胞的能力。免疫7-10天后收集被注射的或引流淋巴结,并用B78H1或表达H2-Db或Kb的B7H1肿瘤细胞激活淋巴细胞。阴性对照和阳性对照是单独的培养基(CM)和抗-CD3抗体(CD3)。进行流式细胞分析,以确定产生IFN-γ(9A-9C)的CD8+T细胞的百分比(9A-9C),或上调GFP和CD69表达(9D)。当在体外用MHC类1a-B78/H1激活时,大约4%的、通过DRibbles(图9B)或Ub-蛋白(图9C)致敏的CD8+T细胞可产生IFN-γ。Db的表达(和Kb在较小程度上)进一步增加了IFN-γCD8+T细胞的百分比。当用来自正常小鼠肝的分离的泛素化蛋白使小鼠免疫时(图9A),当用B78H1或B78H-Db激活时,没有检测到IFN-γ产生CD8+T细胞。因为B78H1细胞是有TAP-2缺陷的,这些T细胞识别通过TAP-2-独立的机理呈递的表位。检查GFP的表达揭示了MHC类1a分子不会限制大部分的B78H1反应性IFN-γ产生CD8+T细胞。
实施例5:作为肿瘤疫苗的泛素化蛋白
虽然基于疫苗的肿瘤源自噬体(例如DRibbles)可用于产生比全细胞疫苗更宽的抗肿瘤免疫反应,但是高免疫原性泛素化蛋白的分离和重新包装可允许更有效的疫苗,其包括次属群、新的和神秘的抗原,该抗原能够致敏与包装在自噬体中的抗原不同的T细胞细胞库,这可能受到自噬体制备过程中共分离的细胞成分的限制。很多经典的小鼠肿瘤模型(例如4T1或B16BL6-D5)免疫原性不佳,且难以治疗。自发性肿瘤可能更难以治疗。目前的数据表明,肿瘤中存在的突变数可能影响内源性免疫原性的水平。因此,新的、自发产生的肿瘤可具有非常少的突变和甚至更少的免疫原性。
图10展示了接种有分离的泛素化蛋白的荷瘤小鼠的生存曲线。小鼠患有4T1乳腺肿瘤,然后接受培养基(无疫苗,曲线1010)、源自4T1细胞的完整的Dribbles(曲线1020)、耗尽泛素化蛋白的4T1 Dribble馏分(曲线1030)、或从4T1 Dribbles分离的泛素化蛋白(曲线1040)的免疫治疗。用在培养基中生长的生长并收获的25,000活的4T1肿瘤细胞挑战小鼠。接种5天后,小鼠接受包括上述疫苗类型之一的10μg每节点的免疫治疗。在接种7天和9天后,小鼠接受同等浓度的疫苗增强剂。
对于该实验,Dribbles源自4T1细胞。简要地,硼替佐米(200nmol/L)和氯化铵(10-20mmol/L)处理细胞24小时。在300×g离心沉淀细胞和细胞碎片7分钟。通过剧烈拍打,Dribbles从细胞或细胞碎片的团块中脱落。然后在7500×g下离心悬浮液,以沉淀Dribbles。抛弃含有纳米囊泡和外来体的上清液。
在冰上的5x-RIPA缓冲液中裂解分离的Dribbles的馏分,然后超声破坏细胞膜。通过在13,000xg,4℃的条件下,离心12分钟去除溶液中的细胞膜。将所得溶液以1:10稀释于PBS中。然后使用市售的S5a-连接珠通过在4℃过夜孵育从溶液中俘获泛素化蛋白。在90%PBS/10%细胞裂解液缓冲液中洗涤具有S5a-连接珠的S5a 2x。在56℃,在含有2M NaCl,pH 6.0的溶液(磷酸盐)洗脱结合至S5a-连接珠的蛋白30分钟。将洗脱液多次透析到PBS中,然后浓缩为1mg/ml的溶液。虽然S5a对K-48连接的泛素具有较高的亲和性,但是通过免疫印迹法(未展示)在洗脱液中也检测到K-48和K-63连接的泛素物种。
如曲线1040所示,接种有Ub-富集的Dribble馏分的6只小鼠中的5只在接种42天之后无肿瘤,而对照组、完全Dribble组或Ub-耗尽的Dribble馏分组的全部小鼠肿瘤恶化。该结果表明,Dribbles中含有的泛素化蛋白对于Dribble疫苗的治疗性能很重要。通过分离泛素化蛋白,可将疫苗浓缩,且疫苗可以更有效地激活抗肿瘤活性。
实施例6:来自表达病原相关蛋白的细胞的疫苗来源
用于导出目标-特异性泛素化蛋白的另一种方法是将特异性人类或动物病原体的基因组、或基因组成分插入用于产生泛素化蛋白的细胞系,例如HIV中。从表达HIV蛋白的细胞中分离的泛素化蛋白可用作疫苗,以增强免疫力并潜在地清楚HIV基因组。这种疫苗可以与针对这种疾病的CMV载体结合使用。在其他情况下,特异性人类病原体的基因组,或基因组的成分(例如结核分枝杆菌或埃博拉)可以插入用于产生泛素化蛋白的细胞系中。在其他实施例中,特异性动物病原体的基因组,或基因组的成分(例如猪繁殖与呼吸综合征病毒(PRRSV))可以插入用于产生泛素化蛋白的细胞系中。
图11展示了证明从有效表达CMV特异性抗原的细胞中分离的Dribbles携带并交叉呈递这些抗原的图和曲线。负载有CEF-Dribbles的单核细胞比对照E6E7-DRibbles(2.4%vs 0.06%;p<0.01;曲线1110)和CD4+T细胞(0.56%vs0.03%;p<0.05;曲线1120)激活更多的CD8+T细胞。用E6E7-Dribbles的激活不会增加超越无抗原组中的观察到的频率的反应。这些结果表明,当负载至单核细胞时,HEK 293 T源Dribbles将病毒抗原交叉呈递至抗原特异性CD8+T细胞和CD4+T细胞反应。通过源自缺乏CEF抗原肽的HEK 293 T细胞的Dribbles没有明显地非特异性激活记忆T细胞。
我们也是用源自UbiLT3细胞系的Dribbles,其源自患有癌症的患者。产生表达CMV的pp65蛋白的UbiLT3细胞系,UbiLT3细胞系用于制备作为CMVpp65抗原源的Dribbles,以激活来自24个捐献者的PBMCs。在上述细胞内染色协议之后,计算IFN-γ+CD8+T细胞和IFN-γ+CD4+T细胞的百分比。UbiLT3pp65 DRibbles激活组的IFN-γ+CD8+T细胞的平均百分比为0.24%,相比而言,用对照组UbiLT3 GFP Dribbles处理的T细胞的配对组中的IFN-γ+CD8+T细胞的平均百分比为0.08%(p<0.01;曲线1130,流程图1150)。根据pp65 Dribbles激活产生IFN-γ的CD4+T细胞的平均百分比为0.54%,相比而言,用对照Dribbles激活后的为0.04%(p<0.01;曲线1140,流程图1150)。与纯化的重组CMVpp65相比,UbiLT3 pp65 Dribbles是记忆CD4+T细胞的更好的激活剂(0.54%vs0.39%,p<0.05,图2D,E),但是更少地激活记忆CD8+T-细胞(0.24%vs 0.58%,p<0.01,图2C,E)。这些数据表明,当负载到单核细胞或直接添加至人类PBMCs中时,来自HEK293T细胞和UbiLT3细胞的Dribbles是用于CD8+T-细胞和CD4+T细胞的抗原特异性活化的强效免疫原。
这些结果表明,表达的病毒蛋白转变为Dribbles,其中它们可以用作用于交叉呈递的抗原。通过从表达病毒蛋白的细胞中分离泛素化蛋白,可以衍生出具有增强效力的免疫原。可以培养表达包括肿瘤抗原、病毒抗原、细菌抗原和其他病原体抗原的多种抗原的细胞类型,并抑制经蛋白酶体降解蛋白。然后从这些细胞中分离泛素化蛋白,用作疫苗、免疫增强剂、T细胞引物、免疫监测试剂、或其免疫原性或治疗化合物。
如上所述的实验数据和方法能够实现一种或多种组合物,和一种或多种方法。在一个实施例中,提供一种设计为诱导哺乳动物中的特异性免疫反应的组合物,该组合物包括分离的泛素化蛋白,其在溶液中,且不含有膜结合细胞器,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白(SLiPs)和多聚泛素化缺陷核糖体产物(DRiPs)。SLiPs和DRiPs的临界量可以是在体内和/或体外足以激活免疫反应的量。SLiPs和DRiPs的临界量可以是足以激活肿瘤特异性T细胞的增殖的量。SLiPs和DRiPs的临界量可以是在体内或体外足以负载树突状细胞,以使抗原可交叉呈递至T细胞的量。SLiPs和DRiPs的临界量可以是足以激活独立于MHC类1a的T细胞的交叉致敏的量。SLiPs和DRiPs的临界量可以是在体内足以激活抗肿瘤活性的量。SLiPs和DRiPs的临界量还可以是足以体内激活炎症反应的量。
分离的泛素化蛋白可以从在培养基中生长的肿瘤源细胞亲和纯化。当在培养基中生长时,例如可通过使用蛋白酶体抑制剂(硼替佐米)来抑制肿瘤源细胞通过蛋白酶体降解泛素化蛋白。在一些实施例中,当在培养基中生长时,例如可通过使用溶酶体抑制剂来抑制肿瘤源细胞通过溶酶体降解泛素化蛋白。
肿瘤源细胞可以源自哺乳动物的肿瘤。在一些实施例中,肿瘤源细胞可以是源自哺乳动物的肿瘤的原代培养细胞。在一些实施例中,肿瘤源细胞可以源自肿瘤细胞系。肿瘤源细胞可以设计为表达肿瘤源细胞的非内源性的一种或多种特异性抗原,例如肿瘤特异性抗原、病毒抗原、细菌抗原、病原体抗原等。
在另一个实施例中,提供一种用于制备免疫原性试剂的方法,该方法包括:抑制在培养基中生长的肿瘤源细胞中的泛素化蛋白的蛋白酶体降解;从在培养基中生长的所述肿瘤源细胞中分离不含有膜结合细胞器的泛素化蛋白,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白和多聚泛素化缺陷核糖体产物。该方法可进一步包括裂解肿瘤细胞;亲和纯化来自具有重组多聚泛素结合肽的所得细胞裂解液的泛素化蛋白;和洗脱来自所述重组多聚泛素结合肽的所述泛素化蛋白。
在另一个实施例中,提供一种用于诱导哺乳动物中的特异性免疫反应的方法,该方法包括:提供第一组合物,所述第一组合物包括在溶液中的分离的泛素化蛋白,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白和多聚泛素化缺陷核糖体产物;和将所述一种或多种特异性抗原交叉呈递至T细胞。该方法进一步包括:在体外,用所述第一组合物负载树突状细胞,以产生负载的树突状细胞;和将抗原从所述负载的树突状细胞交叉呈递至T细胞。抗原可以在体内或体外被从负载的树突状细胞交叉呈递至T细胞。可以通过皮下注射、节内(intra-nodally)给药、鼻内给药、静脉内给药、或通过任何其它合适的方式将第一组合物给药至哺乳动物。可以在注射能够诱导哺乳动物中的特异性免疫反应的第二组合物之后的一段时间之后,给哺乳动物注射第一组合物。第二组合物是异体自噬体富集的组合物,例如Dribble疫苗。特异性免疫反应可以针对经典和/或非经典MHC限制抗原。

Claims (20)

分离的泛素化蛋白,其在溶液中,且不含有膜结合细胞器,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白和多聚泛素化缺陷核糖体产物。
2.根据权利要求1所述的组合物,其特征在于,所述分离的泛素化蛋白是从在培养基中生长的肿瘤源细胞亲和纯化的。
3.根据权利要求2所述的组合物,其特征在于,在培养基中生长时,抑制所述肿瘤源细胞通过蛋白酶体降解泛素化蛋白。
4.根据权利要求2所述的组合物,其特征在于,所述肿瘤源细胞为源自哺乳动物的培养细胞。
5.根据权利要求4所述的组合物,其特征在于,所述肿瘤源细胞设计为表达所述肿瘤源细胞的非内源性的一种或多种特异性抗原。
6.根据权利要求5所述的组合物,其特征在于,所述肿瘤源细胞的非内源性的所述一种或多种特异性抗原包括病毒抗原。
7.根据权利要求5所述的组合物,其特征在于,所述肿瘤源细胞的非内源性的所述一种或多种特异性抗原包括细菌抗原。
8.一种用于制备免疫原性试剂的方法,该方法包括:
抑制在培养基中生长的肿瘤源细胞中的泛素化蛋白的蛋白酶体降解;
从在培养基中生长的所述肿瘤源细胞中分离不含有膜结合细胞器的泛素化蛋白,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白和多聚泛素化缺陷核糖体产物。
9.根据权利要求8所述的方法,进一步包括:
裂解所述肿瘤细胞;
对来自具有重组多聚泛素结合肽的所得细胞裂解液的泛素化蛋白进行亲和纯化;和
洗脱来自所述重组多聚泛素结合肽的所述泛素化蛋白。
10.根据权利要求8所述的方法,其特征在于,所述肿瘤源细胞设计为表达未被所述肿瘤源细胞内源性表达的一种或多种特异性抗原。
11.根据权利要求10所述的方法,其特征在于,所述一种或多种特异性抗原包括病毒抗原。
12.根据权利要求11所述的组合物,其特征在于,所述一种或多种特异性抗原包括细菌抗原。
13.根据权利要求9所述的组合物,其特征在于,所述肿瘤源细胞为源自哺乳动物的肿瘤的原代培养细胞。
14.一种用于诱导哺乳动物中的特异性免疫反应的方法,该方法包括:
提供第一组合物,所述第一组合物包括在溶液中的分离的泛素化蛋白,所述分离的泛素化蛋白包含一种或多种特异性抗原,并进一步包含临界量的多聚泛素化短寿命蛋白和多聚泛素化缺陷核糖体产物;和
将所述一种或多种特异性抗原交叉呈递至T细胞。
15.根据权利要求14所述的方法,进一步包括:
在体外,用所述第一组合物负载树突状细胞,以产生负载的树突状细胞;和
将抗原从所述负载的树突状细胞交叉呈递至T细胞。
16.根据权利要求15所述的方法,其特征在于,在体内,使抗原从所述负载的树突状细胞交叉呈递至T细胞。
17.根据权利要求14所述的方法,其特征在于,将所述第一组合物皮下注射入哺乳动物。
18.根据权利要求14所述的方法,其特征在于,在注射能够诱导哺乳动物的特异性免疫反应的第二组合物一段时间之后,将所述第一组合物注射入哺乳动物。
19.根据权利要求18所述的方法,其特征在于,所述第二组合物为异体自噬体富集的组合物。
20.根据权利要求14所述的方法,其特征在于,所述特异性免疫反应针对经典和/或非经典MHC限制的抗原。
