发明领域
本发明涉及使用重组克隆型免疫球蛋白(Ig)作为疫苗来治疗HCV相关性和非HCV相关性淋巴组织增生。具体地,本发明提供由来自患有淋巴组织增生的患者的Ig轻链的蛋白质节段VK3-20和VK3-15而衍生出的具有免疫原性的重组蛋白质的用途。
发明背景
丙型肝炎是由特定病毒(丙型肝炎病毒,HCV)引起的肝炎类型。在许多病例中,急性丙型肝炎没有症状并转为慢性,对肝脏造成长期损害,例如导致肝硬化和肝细胞癌。丙型肝炎可引起的其他相关症状例如为甲状腺炎、冷球蛋白血症和某些类型的肾小球性肾炎。
术语“冷球蛋白血症”具体指的是血清中存在一或多种如下免疫球蛋白(Ig),所述免疫球蛋白在低于37℃时发生沉淀,而在再次加热后可再溶解。冷球蛋白血症通常分为3个亚组:单纯型冷球蛋白血症(1型),其特征在于存在单克隆Ig,1型通常与血液系统疾病相关,且经常是无症状的;混合型冷球蛋白血症或MC(2型和3型),特征在于存在循环免疫复合物,所述免疫复合物由作为自身抗原的多克隆IgG和作为相应的自身抗体的单克隆IgM(2型)或多克隆IgM(3型)组成。MC可继发于多种感染或免疫病;当分离的MC代表一种独立的疾病时,称为“原发性”MC。鉴于其与HCV感染的惊人关联性(>90%),术语“预防性”现在仅涉及MC患者的少数(<5%)。HCV可感染淋巴组织,并可触发单-多克隆B淋巴细胞增生,产生不同的自身抗体,包括冷球蛋白。MC综合征是全身性血管炎,其继发于循环免疫复合物和补体在小血管的沉淀。其临床特征为累及补体器官:紫癜、皮肤溃疡、肝炎、肾小球肾炎、周围神经病和/或弥漫性血管炎。一些患者可出现恶性肿瘤,这通常是晚期并发症,特别是B细胞非霍奇金淋巴瘤(10%)、肝细胞癌(<5%)、或甲状腺癌(<1%)。
MC的一线治疗应该是通过干扰素和利巴韦林来清除HCV;但是,这一治疗通常无法清除病毒,并可并发严重的不良反应(肾病、甲状腺炎等等)。应该根据患者临床表现的活动度和严重程度为每一个患者制定病因学治疗(血浆置换、免疫抑制剂和/或肾上腺皮质激素类)。使用干扰素治疗HCV相关性冷球蛋白血症综合征是基于这样的假设,即B-淋巴细胞增生是病毒依赖性的,且由此对降低病毒负荷具有潜在地反应性;因此,其并不是以治疗淋巴组织增生为直接目的的治疗。使用干扰素或聚乙二醇化干扰素(适合于增加药物的血浆半衰期)治疗也伴有毒性和不良反应,例如:易激惹、情绪不稳定、抑郁状态、睡眠障碍、恐惧、躁狂、认知(记忆、注意力)障碍、错乱状态。
因此,有必要寻找用于治疗在此所述的淋巴组织增生的替代性预防和/或治疗手段。
已经使用独特型Ig进行免疫接种来治疗B细胞非霍奇金淋巴瘤(NHL),不过,由于为每个患者产生独特型Ig具有显著的复杂性且成本高昂,严重限制了在大对象群体中应用这一免疫接种策略。
发明内容
本发明的目的是为淋巴组织增生性疾病提供新的疫苗,该疫苗克服了迄今所采用的技术的局限性和不便利之处,并可用于大数量的患者群体。
本发明的另一个目的是提供鉴定、分离和评价那些适合用于制备所述疫苗的蛋白质节段的免疫原性的方法。
还一个目的是提供制备组成所述疫苗的蛋白质节段的方法。
优选实施方式描述
通过使用克隆型重组免疫原性节段治疗HCV相关性和非HCV相关性淋巴组织增生可实现这些和其他目标。
因此,在一个方面,本发明涉及疫苗,所述疫苗包含选自VK3-20和VK3-15中的至少一种重组免疫原性蛋白质节段,其用于治疗和/或预防淋巴组织增生性疾病。
在另一方面,本发明涉及选自VK3-20、VK3-15以及它们的混合物中的重组免疫原性蛋白质节段的用途。
对于本发明的用途,选自VK3-20、VK3-15以及它们的混合物中的一或多种重组免疫原性蛋白质节段可选地与另一种蛋白质部分或试剂组合,用于制备用于治疗和/或预防淋巴组织增生性疾病的疫苗。
在另一方面,本发明涉及包含重组免疫原性蛋白质节段VK3-20和VK3-15的疫苗在制备用于淋巴组织增生性疾病的药物中的用途。
具体地,术语“重组免疫原性蛋白质节段”指的是通过重组DNA技术获得的、并具有免疫原性活性(即能够诱导免疫应答)的蛋白质部分。
术语VK3-20和VK3-15指的是Igκ型轻链的可变区。
VK3-20和VK3-15的编码基因的核苷酸序列见于NCBI数据库,登录号为:对于VK3-20为AF303897/AAG33824,而对于VK3-15为AAU14891/AY704914。
在另一方面,本发明涉及重组克隆型节段VK3-20和VK3-15的分离、表征、生产和纯化。
根据本发明的一个实施方式,用于生产、实施和纯化重组克隆型节段VK3-20和VK3-15的方法包括以下步骤:
1)将编码所述蛋白质的核苷酸序列插入合适的表达载体;
2)接种至合适的培养基中;
3)根据已知技术进行发酵;和
4)纯化所述片段。
根据本发明的优选实施方式,所述生产方法的特征在于根据以下步骤(1’)至(4’)进行步骤(1)至(4)的至少两个步骤:
1’)将所述序列插入表达载体pET26b,以形成重组菌株pET26b/VK3-20/VK3-15;
2’)以1%的体积比接种细菌测试样品;
3’)使用以磷酸盐缓冲液缓冲并加入葡萄糖或异丙硫半乳糖苷(IPTG)的培养基SB进行发酵;
4’)通过离子交换层析纯化。
根据本发明,术语“pET26b/VK3-20/VK3-15”指的是工程化的表达载体pET26b,其可选地含有编码两种重组独特型蛋白链VK3-20和VK3-15中之一的核苷酸序列。
根据本发明,可选地使用针对VK区域的单克隆抗体(mAb)进行重组独特型蛋白链VK3-20和VK3-15的节段的鉴定和定量。

与已知的方法相比,本发明的改进并优化的方法具有多种优势,事实上其能够:
-加快重组细菌培养物的生长并使得其生物量增加多达50%;
-提高方法的产率;
-降低生产时间和成本;和
-根据生产和质量管理规范(GMP)进行简化、再生产和扩大生产规模。
根据本发明的另一个优选的实施方式,发酵细菌菌株可以是在37℃培养8-12小时,例如过夜。
还根据本发明的优选的实施方式,从培养基或从细菌周质空间纯化所述重组的独特型蛋白质链包括在离子交换柱例如Q Sepharose FF(Amersham-GE)和S Sepharose FF(Amersham-GE)上进行的步骤。
本申请在实验部分给出了本发明方法的详细描述。
如本申请说明书实验部分实施例5所示,已经观察到重组免疫原性节段VK3-20能够识别22种免疫原性表位。
在另一方面,本发明涉及能够识别由蛋白质节段VK3-20所识别的这22种免疫原性表位中的至少一种表位的任何蛋白质节段,其优选地识别一种以上表位,其有利地识别所有22种表位。
本发明的疫苗用于HCV相关性和非HCV相关性淋巴组织增生。事实上已经观察到,由衍生自HCV相关性淋巴瘤的原型轻链VK3-20刺激产生的细胞毒反应表现出显著的交叉反应性,甚至对由同一基因编码但来自不同患者的独特型VK蛋白也是如此,正因如此,本发明所制备的疫苗可有效地施用于大量代表性患者群体,以治疗和/或预防淋巴组织增生性疾病。因此,也可治疗非HCV相关性淋巴组织增生。例如滤泡性淋巴瘤(FL)、慢性淋巴细胞白血病(CLL)、粘膜相关性淋巴组织(MALT)的淋巴瘤以及自身免疫病(例如类风湿性关节炎、干燥综合征)相关性淋巴瘤。
对于其治疗和/或预防,本发明的疫苗优选地以合适的药物组合物来施用。
本发明还涉及以下任何蛋白质节段在制备用于治疗和/或预防HCV相关性和非HCV相关性淋巴组织增生的疫苗中的用途,所述蛋白质节段能够识别由蛋白质节段VK3-20所识别的22种免疫原性表位中的至少一种表位,其优选地识别一种以上表位,其有利地识别所有22种表位。
含有所述疫苗的药物组合物是本发明的另一方面。
在本发明的用于口服、皮下、静脉、经皮或局部施用的药物组合物中,所述蛋白质节段优选地以单剂施用,例如作为与常规赋形剂或药用可接受载体的混合物施用。根据患者的年龄、体重和健康状况,或根据疾病严重程度水平和施用途径,可使用不同的剂量(有优选的范围,例如0.1至1mg,优选地0.3至0.7mg,例如每一剂量单元0.5mg)。
本发明的疫苗和/或药物组合物最终可与治疗中所使用的其他药物组合使用,例如与沙格司亭(sargramostim)(GM-CSF,50μg/m2/剂)和/或重组IFN-α2a(1,000,000UI/m2/剂)组合。
在另一方面,本发明包括用于治疗和/或预防淋巴组织增生性疾病的试剂盒,其包括本发明的药物组合物和/或沙格司亭和/或重组IFN-α2a。
本发明的试剂盒优选地包括剂量单元形式的本发明的药物组合物,所述剂量单元含有0.5mg量的有效成分和/或沙格司亭(GM-CSF,50μg/m2/剂)和/或重组IFN-α2a(1.000.000UI/m2/剂)。

实施例1:重组克隆型Ig的分离、制备、体外表征
插入患有II型混合型冷球蛋白血症的6个HCV患者的VH和VK区和伴随的NHL-B并体外重建,产生来自这些序列的单链蛋白(single-filament proteins,ScFv)并通过亲和法纯化。
仅在体外进一步以免疫学方法表征了来自两个患者的VK链的蛋白质,即VK3-20(VK-gal)和VK3-15(VK-gent)。分别从得自肝活检和来自骨髓细胞的肿瘤细胞提取DNA,这两种组织均被Kiel分型中的淋巴浆细胞-淋巴浆细胞样型低度恶性非霍奇金淋巴瘤累及。在这两个病例中,根据以前报道的方法扩增VK区(De Re V等,Blood.2000;96:3578-84)。形成免疫球蛋白轻链可变区的基因为:VK3-20/JK1*01和VK3-15/JK1*01。随后克隆VK序列并以单链形式表达。为此,从琼脂糖凝胶回收PCR产物,以限制性酶BamHI和XhoI消化,并连接至质粒载体pET26b(Novagen,Madison,WI,United States)的BamHI-XhoI限制性位点,形成表达质粒pET26b/VK3-20和pET26b/VK3-15。下游引物FL7(5′-CGG GAT CCGGAA ATT GTG TTG ACG-3′)提供BamHI的限制性位点,FL5引物(5′CCGCTC GAG TCA TTT GAT TTC CAC C-3′)提供3′端XhoI限制性位点。
通过使用以独特型蛋白VK3-20和VK3-15脉冲式刺激的自体树突状细胞刺激来自13个健康供者的外周血单个核细胞(PBMC)而获得多克隆细胞毒T淋巴细胞(CTL)培养物,这些供者经主要组织相容性系统基因型分析证实彼此之间没有关联。以这种方式获得了9个经VK3-20刺激的CTL细胞系和4个经VK3-15刺激的CTL细胞系。为了体外评估免疫原性,使用放射性同位素(51Cr)方法和非放射性方法(钙黄绿素释放)进行细胞毒性测试。对这些蛋白质的体外免疫原性的另一测试是在存在商品化抗体W6/32的情况下评估特异性细胞毒性被抑制的百分数,目的在于评估所诱导的免疫应答的抗原特异性,结果证实所用的机制是I类HLC限制性的(图1)。
实施例2:重组独特型蛋白的交叉识别
已经发现,以来自HCV相关性淋巴瘤的原型轻链VK3-20刺激的细胞毒性应答出现了交叉反应性,甚至对由同一基因(VK3-20)编码但来自不同患者的淋巴瘤的独特型VK蛋白也是如此(图2)。图2的数据得自4个不相关的供者(通过不同的I类HLA单元型加以区分),这些数据显示由重组原型蛋白VK3-20诱导的细胞毒性免疫应答为何在患有表达独特型免疫球蛋白(包括轻链VK3-20)的淋巴瘤的无关患者中也具有显著的免疫治疗潜能。实施例3:评估VK3-20-特异性CTL和VK3-15-特异性CTL的交叉反应性
进行进一步的实验以评估VK3-20-特异性CTL和VK3-15-特异性CTL识别并杀伤自体的匹配靶标(或其携带相应的用于产生所述CTL的蛋白质)和自体的不匹配靶标(或其携带分子相关性蛋白质VK3)的能力。在所考虑的3个供者中,如图3所示,VK3-20-特异性CTL和VK3-15-特异性CTL甚至能够以I类限制性方式杀伤分别携带VK3-15和VK3-20的靶标(不匹配靶标)。实际上,这两种VK3蛋白的序列同源性超过80%,这可以解释它们的交叉反应性。这为将这些重组蛋白用作治疗和预防表达分子相关性独特型的淋巴组织增生的疫苗提供了依据。已经显示了VK3-20-特异性CTL是如何以I类限制性方式识别并杀伤天然表达蛋白VK3-20的淋巴瘤B细胞(DG75)和天然表达分子相关性蛋白VK3-15的B-类淋巴母细胞系(SH9)(图4)。这说明,由淋巴组织增生B细胞表达的VK3-20和VK3-15可被天然地加工,并以生理学方式产生能够介导具有潜在临床价值的细胞介导的免疫应答的免疫原性表位。
实施例4:鉴定记忆应答
为了鉴定淋巴肿瘤患者内针对选定的独特型Ig的特异性记忆细胞,基于鉴定分泌干扰素-γ的CD8+T-淋巴细胞而进行ELISPOT分析。该分析显示,在患有VK+淋巴瘤的患者中存在针对VK3-15和VK3-20的特异性记忆应答,见图5。以ELISPOT方法在48小时对13个患有HCV相关性淋巴瘤的患者的PBMC进行测定,其中使用携带所述选定的重组蛋白质的自体单核细胞作为抗原呈递细胞,以CD8+T-淋巴细胞作为“应答”细胞。实施例5:鉴定并验证免疫原性表位
为了鉴定能够结合最常见的I类HLA等位基因的VK3-20肽,使用预测表位的计算机方法(Syfpeithi,Net-MHC,Bimas)分析来自HCV-相关性淋巴组织增生的克隆型Ig的VH和VL区域。仅对于HLA-A*0201等位基因就鉴定到了35个能够结合这一限制元件的潜在表位。为了鉴定和验证蛋白VK3-20的免疫原性表位,使用了iTopiaTM high-throughput EpitopeDiscovery System(Beckman Coulter)。随后鉴定了蛋白VK3-20的免疫原性表位。通过对肽与重组形式HLA分子的结合、不结合以及亲和力的一系列ELISA样测试,分析了来自VK3-20蛋白的100个九聚物的文库,所述九聚物彼此重叠一个氨基酸。随后鉴定到22个肽,它们对7种不同的HLA-A和-B等位基因的每一个具有中等结合能力(图6和表1)。还已证实,以蛋白VK3-20在不同供者中诱导的CTL可以A2限制性方式识别并杀伤携带A*0201肽的靶标以及天然表达蛋白VK3-15的SJ9类淋巴母细胞系。这进一步证实蛋白VK3-20的表位所诱导的免疫应答的交叉反应性。

实施例6:产生重组克隆型Ig
为了产生重组克隆型Ig VK3-20和VK3-15并实施所述生产和纯化方法,将这些衍生自HCV相关性淋巴瘤的片段插入细菌表达载体pET26b,后者适合用于临床目的的生产。
然后进行以下操作:
-将编码VK3-20和VK3-15的核苷酸序列插入细菌载体pET26b
-以占总体积1%的浓度接种在含有1%葡萄糖的缓冲SB培养基中
-37℃发酵8-12小时
-纯化所述片段
在片段插入步骤中,将片段稳定插入至卡那霉素抗性细菌表达载体pET26b中,并使用针对VK区的单克隆抗体(mAb)鉴定并定量所产生的片段。在接种步骤中,将细菌测试样品,pET26b/VK3-20/VK3-15,其相当于总体积的1%,接种至添加了磷酸盐缓冲液和1%葡萄糖的改良SB培养基中,以IPTG诱导并将培养物在37℃过夜培养发酵。在纯化步骤中由培养基和细菌周质空间纯化目的片段。该方法包括使提取物和培养基通过离子交换树脂Q Sepharose FF(Amersham-GE)以结合不需要的产物,而将目的片段VK3-20和VK3-15作为柱的洗脱产物而收集,并进一步在阳离子交换树脂S Sepharose FF(Amersham-GE)上处理。通过SDS-PAGE分析评估VK3-20和VK3-15片段的纯度。上述过程在默认范围内可从周质空间提取物中纯化大约37mg的片段,产率为4mg/L,从培养基中纯化大约30mg的片段,产率为20mg/L。
实施例7:由特异性肽CTL VK3-20诱导的针对与VK蛋白不相关的类似表位的交叉反应性细胞毒应答
特异性CTL VK3-20可诱导针对VK3-15的交叉反应性细胞毒应答这一结果提示这样一种假设,即VK3-20可能含有针对同样属于VK-III家族或其他家族轻链的类似肽的潜在交叉反应性免疫原性表位。因此进行了免疫信息学分析,即通过ImMunoGeneTics information system来比对VK3-20的表位T HLA-A*0201的序列与所收集的所有VK链的序列。
就保守性而言,仅考虑了具有单个氨基酸变异的肽。所做的分析似乎提示表位VK3-20在VK-III家族内具有高度的保守性。对于肽P20,鉴定到了两种属于VK-III家族的其他蛋白质的潜在交叉反应性表位(表2)。

令人感兴趣的是指出VK3-20的表位P33如何与属于其他家族的VK蛋白(VK-1、VK-V和VK-V1)的一些具有潜在交叉反应性的类似表位相对应(表3),此外,它们在淋巴肿瘤中的表达并不少见。

为了证明交叉反应性的实际效力,从2个HLA-A*0201供者获得了对VK3-20的肽P20和P33具有特异性的2个CTL细胞系。所获得的细胞系能够以特异性方式和HLA-A2限制性方式裂解经诱导肽或经完整VK3-20脉冲式刺激的自体类淋巴母细胞系、以及细胞系DG75(VK3-20+)和SH9(VK3-15+)(图7)。这些结果进一步支持蛋白Vk3-20的免疫原性,且同时表明,可以比较容易地从健康供者的外周血单个核细胞获得对VK3-20的表位HLA-A*0201具有特异性的离体CTL。随后分析了以这种方式获得的CTL细胞系裂解自体靶标(LCL)的能力,所述自体靶标可选地呈递诱导肽和一系列属于不同VK蛋白的“交叉反应性”肽(表2和3)。该分析证实,对P20或P33具有特异性的CTL细胞系能够对所有被研究的“交叉反应性”肽施加HLA-A*0201限制性高细胞毒性(图8和9)。
对从2个不同供者获得的肽特异性CTL进行分析,针对所有被研究的“交叉反应性”肽HLA-A*0201得到了相当的结果。
所获得的数据支持如下结论,即可以认为蛋白VK3-20是由最常见的I类HLA所呈递的众多免疫原性表位的“载体”。由淋巴瘤VK3-20+细胞天然呈递的这些表位所介导的免疫应答很可能不仅对于表达VK-III家族轻链的淋巴瘤是有效的,而且对于表达不同家族的VK蛋白的淋巴瘤也是有效的。因此,这些结果为基于使用不同B细胞淋巴组织增生之间所共有的Id来构建用于“交叉反应性”免疫治疗的重组疫苗提供了坚实的临床前期依据。
实施例8:在生物反应器Wave内产生蛋白VK3-20
为了获得能够在GMP竞争中快速转化的蛋白VK3-20生产方法,建立了在一次性生物反应器Wave EHTD 20/50(GE Healthcare)中的发酵方法。该方式采用无菌一次性袋,因此与传统发酵罐相比能够消除不同批次之间交叉污染的风险。出于同样的原因也不需要清洁和无菌化验证过程。
根据以下方案进行发酵:
-制备预培养物:
将单个袋连接于生物反应器振荡板,并在无菌条件下充入100ml的SBT培养基。从甘油储存液中取50μl细菌,移液至1ml的SBT培养基中,然后通过注射器接种在袋内。该预培养物在37℃摇动培养过夜,该生物反应器被设置为摆动12°,速度为40rpm。
-发酵:
次日,通过充入10L的SBT培养基制备新的一次性袋。使用袋上所装的具有路厄连接(luer junction)的管子将100ml预培养物接种在10L培养基内。然后在37℃发酵该培养物3小时,摇动(12°,40rpm)。
3小时后使用注射器向培养物内加入诱导剂IPTG,终浓度为1mM,然后使培养物在相同的温度和摇动条件下生长过夜。
通过在600nm读取光密度而监测细菌生长,结果如下:

这些数据说明细菌群体在生物反应器Wave内的生长趋势与在传统系统(摇瓶)内的生长相当。
发酵方法获得的生物量为每升细菌培养物平均18克沉淀物,且蛋白VK3-20的产量与传统发酵方法获得的产量相当。
因此,可以使用发酵用生物反应器Wave产生适合于GMP生产的具有可重复性的理想方法。该生物反应器还允许处理规模线性扩大至500L,且因此能够产生足够用于临床的原料。
实施例9:蛋白VK3-20与KLH偶联
为了增强VK3-20蛋白的免疫原性,建立了将多肽偶联于血蓝蛋白(钥孔血蓝蛋白,KLH,Vacmune)的方法。所述偶联蛋白用于免疫方案以产生单克隆抗体。
根据以下方法将纯化的蛋白质偶联于KLH。
在PBS中制备浓度为4.8mg/ml的Sulfo SMCC溶液。取260μl的KLH储存液(浓度20mg/ml)并加入44μl所制备的Sulfo SMCC溶液,并加入200μl的PBS:然后将混合物室温温育30分钟。使用Centricon(50KDa截断值,Millipore)去除多余的交联剂。向制备的溶液中加入6mg预先纯化并在PBS中透析的蛋白VK3-20,然后室温温育30分钟。通过在将终产物通过具有50KDa截断值的膜进行过滤之后定量流通部分和保留部分中的蛋白质含量而评估该偶联方法的效力。
Claims (25)
1.疫苗,其包含选自VK3-20和VK3-15中的至少一种蛋白质节段。
2.权利要求1的疫苗,其包含蛋白质节段VK3-20和VK3-15。
3.选自VK3-20、VK3-15 VK3-20、VK3-15及其混合物的蛋白质节段在制备用于治疗和/或预防淋巴组织增生性疾病的药物或疫苗中的用途。
4.权利要求3的用途,用于治疗和/或预防HCV相关性和非HCV相关性淋巴组织增生性疾病。
5.用于治疗淋巴组织增生的药物组合物,其包含选自VK3-20和VK3-15中的至少一种蛋白质节段作为有效成分,所述蛋白质节段与至少一种药用可接受赋形剂混合。
6.权利要求5的药物组合物,其包含VK3-20和VK3-15这两种蛋白质节段作为有效成分。
7.权利要求5或6的药物组合物,用于口服、胃肠外或局部施用,施用的有效成分为0.1至1mg。
8.制备选自VK3-20和VK3-15中的至少一种蛋白质节段的方法,其包括将核苷酸序列插入合适的表达载体,发酵、分离并纯化所述蛋白质节段。
9.权利要求8的方法,其中所述选自VK3-20和VK3-15中的至少一种蛋白质节段被插入卡那霉素抗性细菌表达载体pET26b以形成重组菌株pET26b/VK3-20/VK3-15。
10.权利要求8或9的方法,其中使用抗VK的mAb鉴定并定量所述选自VK3-20和VK3-15中的至少一种蛋白质节段。
11.权利要求9或10中任一项的方法,其中所述发酵包括在SB培养基中接种1%的所述重组菌株pET26b/VK3-20/VK3-15。
12.权利要求11的方法,其中所述SB培养基是缓冲的。
13.权利要求12的方法,其中所述培养基以磷酸盐缓冲液进行缓冲并含有1%(w/w)的葡萄糖。
14.权利要求11-13中任一项的方法,其中所述重组菌株在37℃培养8-12小时。
15.权利要求8-14中任一项的方法,其中通过在离子交换树脂QSepharose FF(Amersham-GE)上进行洗脱从培养基或细菌周质空间分离所述选自VK3-20和VK3-15中的至少一种蛋白质节段。
16.权利要求15的方法,其中在阳离子交换树脂S Sepharose FF(Amersham-GE)上纯化所述洗脱的产物。
17.试剂盒,其包含权利要求5至7中任一项的药物组合物和/或沙格司亭和/或重组IFN-α2a。
18.试剂盒,其包含含有0.5mg有效成分的权利要求5至7中任一项的药物组合物和/或沙格司亭(GM-CSF,50μg/m2/剂)和/或重组IFN-α2a(1.000.000UI/m2/剂)。
19.疫苗,其包含蛋白质节段,所述蛋白质节段能够识别蛋白VK3-20的22个已鉴定的免疫原性表位中的至少一种。
20.权利要求19的疫苗,其中所述蛋白质节段能够识别一种以上所述22个已鉴定的表位。
21.权利要求19或20的疫苗,其中所述蛋白质节段能够识别所有所述22个已鉴定的表位。
22.能够识别蛋白VK3-20的22个已鉴定的免疫原性表位中的至少一种的至少一种蛋白质节段在制备用于治疗和/或预防淋巴组织增生性疾病的疫苗中的用途。
23.权利要求22的用途,用于制备用于治疗和/或预防HCV相关性和非HCV相关性淋巴组织增生性疾病的疫苗。
24.权利要求22或23的用途,其中所述蛋白质节段能够识别一种以上所述22个已鉴定的表位。
25.权利要求22或23的用途,其中所述蛋白质节段能够识别所有所述22个已鉴定的表位。
