Description
[単離ユビキチニル化タンパク質の交差提示による未感作T細胞の刺激]
交差提示は、抗原をその中で合成していない細胞がその抗原を提示できることで、免疫反応を初回抗原刺激するための唯一の機構としての直接提示を不必要にする手段であることが明らかとなった。効果的な抗腫瘍免疫反応の発生には、ワクチン接種の際のメラノーマ抗原の交差提示が不可欠であることが実証されたことで、この知見が広がった。議論され、未だ分かっていない交差提示の要素の1つは、抗原の供給源と、それをプロフェッショナルな抗原提示細胞(APC)へ送達する方法である。いくつかのグループは、抗原供給源が細胞タンパク質であることを示したが、熱ショックタンパク質(HSPs)でシャペロン化されたペプチドであると主張するグループもある。更に、クロスプライミング(cross-priming)は、HSP90と併せて、プロテアソーム基質および安定な細胞質ペプチドの供与の両者から生じることが立証されている。短寿命モデル抗原系、およびタンパク質翻訳の阻害剤の両者を用いた実験は、短寿命タンパク質(SLiPs)および/または欠陥リボソーム産物(DRiPs)が、オートファジー依存型クロスプライミングに関連する抗原プールの不可欠な構成要素であることを示している。SLiPsの半減期が短く、APCへ供与されにくいため、この抗原プールの交差提示は一般に効率が悪い。興味深いことに、SLiPsが細胞表面のペプチド:MHC複合体の大部分を構成できるのは、SLiPsのこの時間的性質であり、これは、SLiPsがプロテアソームによってすぐに分解され、TAP1/2によりMHCクラスI分子上に負荷されるためである。プロテアソーム阻害剤(ボルテゾミブ)による腫瘍の処理は、これらの一時的なタンパク質を安定化して、これらを別の主要な分解径路、オートファゴソーム/リソソーム径路に向かわせる。
[単離ユビキチニル化タンパク質の交差提示による腫瘍特異性T細胞の刺激]
歴史的に、癌ワクチンで誘導したT細胞は腫瘍の根絶にあまり効果がない。誘導T細胞は、腫瘍を認識して腫瘍拒絶抗原を発現できたとしても、しばしば、封鎖され、消散し、アポトーシスを受け、あるいは、単に、腫瘍を根絶できる程までに増殖できない。あるスペクトルの腫瘍拒絶抗原に対するT細胞の刺激に使用可能な、効果の範囲の広い腫瘍拒絶抗原を作ることで、増殖T細胞の治療効果を高めることができる。更に、腫瘍は、多数の抗原の発現を同時に低下させる必要があるため、あるスペクトルの腫瘍抗原に対する広い免疫反応は、腫瘍が免疫異物除去から逃れる可能性を小さくすると考えられる。
[単離ユビキチニル化タンパク質による炎症反応刺激]
免疫学的アジュバントは、免疫反応の増強および/または調節のために、免疫炎症性反応を利用するものである。図8に、単離ユビキチニル化タンパク質が誘導した炎症反応のグラフを示す。末梢血単核細胞(PBMCs)を、96ウェルのプレートに、106細胞/ウェルで播いた。次に、PBMCを、5mMのATPおよび10ng/mlのリポ多糖類(ATP/LPS)、または、B78/H1細胞から単離したユビキチニル化タンパク質(5μg/mlまたは50μg/ml)と共に、一晩培養した。次に、各ウェルの上澄み液を取り出し、ELISAでIL−1βを分析した。図8から分かるように、IL−1βは、B78/H1細胞から単離したユビキチニル化タンパク質で濃度依存的に刺激された。IL−1βは炎症性サイトカインであり、このサイトカインの産生は、ユビキチニル化タンパク質の炎症性を証明している。炎症反応の誘導は、ワクチンアジュバントに良く知られる性質であり、ワクチン特異的免疫反応の発展を増強するよう働く。このように、単離ユビキチニル化タンパク質は、抗原の特性とアジュバントの特性の両者を備えることができる。T細胞が活動する部位において炎症を刺激することで、単離ユビキチニル化タンパク質由来のワクチンは炎症反応を起こし、これがワクチン特異的免疫反応の発生を強めると考えられる。
[単離ユビキチニル化タンパク質が介在する、MHCクラス1a依存性および非依存性T細胞クロスプライミング]
腫瘍が免疫監視から逃れる主なメカニズムの1つは、MHCクラス1a分子の発現を低下または完全に欠失させることである。同時に、NK細胞またはCTLによる排除を逃れるためのメカニズムとして、しばしば、腫瘍細胞上に非古典的MHCクラス1b分子を発現増加する。従って、MHCクラス1aに依存せずに腫瘍細胞を認識するT細胞を初回刺激するワクチンならば、T細胞の標的となり得る拒絶抗原のスペクトルを広げて、腫瘍細胞が免疫異物除去から逃れる可能性を小さくできると考えられる。
[腫瘍ワクチンとしてのユビキチニル化タンパク質]
腫瘍由来オートファゴソームを材料とするワクチン(DRibbleなど)は、全細胞ワクチンよりも広い抗腫瘍免疫反応を起こすために使用してもよいが、免疫原性の高いユビキチニル化タンパク質を単離および再パッケージングすることで、オートファゴソーム内にパッケージされているものより効果が高く、レパートリーの異なるT細胞を初回刺激できる、亜優性(subdominant)の、新しい潜在性抗原を含むワクチンが可能となり、これはオートファゴソーム調製工程の際に共に単離される細胞成分によって拘束されると考えられる。古典的マウス腫瘍モデルの多く(例えば、4T1またはB16BL6−D5)は免疫原性が乏しく、治療が難しい。ましてや、自然発生腫瘍は更に治療が困難となり得る。最近のデータは、腫瘍中に存在する変異の数が内在性免疫原性の程度に影響し得ることを示唆している。このことから、新しく自然発生した腫瘍にはほとんど変異が無く、免疫原性も更に低いと考えられる。
[病原体関連タンパク質を発現している細胞からのワクチンの誘導]
標的特異性ユビキチニル化タンパク質を得るもうひとつの方法は、特定のヒトまたは動物病原体(例えばHIV)のゲノムまたはゲノム成分を、ユビキチニル化タンパク質の産生に使用する細胞株に挿入することである。HIVタンパク質を発現している細胞から単離したユビキチニル化タンパク質をワクチンとして使用すると、免疫性を高め、またHIVゲノムを一掃できる可能性もある。このようなワクチンを、この病気に対抗するCMVベクターと組み合わせて使用してもよい。別のシナリオでは、特定のヒト病原体(例えば、ヒト型結核菌またはエボラ)のゲノムまたはゲノム成分を、ユビキチニル化タンパク質の産生に使用する細胞株に挿入してもよい。別の例では、特定の動物病原体(例えば、豚繁殖・呼吸障害症候群ウイルス:PRRSV)のゲノムまたはゲノム成分を、ユビキチニル化タンパク質の産生に使用する細胞株に挿入してもよい。
Claims (27)
- 哺乳動物に特異的免疫反応を誘発するための組成物であって、
前記組成物は、二重膜を含まない、単離ユビキチニル化タンパク質の溶液を含み、
前記単離ユビキチニル化タンパク質は、1つ以上の特異性抗原を含み、更に、閾値量のK48結合型ポリユビキチニル化短寿命タンパク質およびK48結合型ポリユビキチニル化欠陥リボソーム産物を含むことを特徴とする、組成物。 - 前記単離ユビキチニル化タンパク質は、培養成長させた腫瘍由来細胞からアフィニティ精製されたものであることを特徴とする、請求項1に記載の組成物。
- 前記腫瘍由来細胞は、培養成長する間、プロテアソームによるユビキチニル化タンパク質の分解が抑制されたものであることを特徴とする、請求項2に記載の組成物。
- 前記腫瘍由来細胞が、哺乳動物に由来する培養細胞であることを特徴とする、請求項2又は3に記載の組成物。
- 前記腫瘍由来細胞が、前記腫瘍由来細胞に対して内在性でない1つ以上の特異性抗原を発現するよう改変されていることを特徴とする、請求項4に記載の組成物。
- 前記腫瘍由来細胞に対して内在性でない前記1つ以上の特異性抗原が、ウイルス性抗原を含むことを特徴とする、請求項5に記載の組成物。
- 前記腫瘍由来細胞に対して内在性でない前記1つ以上の特異性抗原が、細菌性抗原を含むことを特徴とする、請求項5に記載の組成物。
- 免疫原性試薬の製造法であって、
培養成長させている腫瘍由来細胞中のユビキチニル化タンパク質のプロテアソーム分解を抑制する工程と、
培養成長させた前記腫瘍由来細胞から、二重膜の不在下で、ユビキチニル化タンパク質を単離する工程と、
を含み、
前記単離ユビキチニル化タンパク質は、1つ以上の特異性抗原を含み、更に、閾値量のK48結合型ポリユビキチニル化短寿命タンパク質およびK48結合型ポリユビキチニル化欠陥リボソーム産物を含むことを特徴とする、製造法。 - 更に、
前記腫瘍由来細胞を溶解する工程と、
得られた細胞溶解物から、組み換えポリユビキチン結合ペプチドを用いてユビキチニル化タンパク質をアフィニティ精製する工程と、
前記組み換えポリユビキチン結合ペプチドから前記ユビキチニル化タンパク質を溶離する工程と、
を含むことを特徴とする、請求項8に記載の製造法。 - 前記腫瘍由来細胞が、前記腫瘍由来細胞によって内因性発現しない1つ以上の特異性抗原を発現するよう改変されていることを特徴とする、請求項8又は9に記載の製造法。
- 前記1つ以上の前記特異性抗原が、ウイルス性抗原を含むことを特徴とする、請求項10に記載の製造法。
- 前記1つ以上の前記特異性抗原が、細菌性抗原を含むことを特徴とする、請求項11に記載の製造法。
- 前記腫瘍由来細胞が、哺乳動物の腫瘍に由来する初代培養細胞であることを特徴とする、請求項9に記載の製造法。
- 前記閾値量は、インビボおよび/またはインビトロで、未感作T細胞に免疫反応を初回抗原刺激するのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、腫瘍特異性および/または癌特異性のT細胞の増殖を刺激するのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、抗原がT細胞に交差提示されるよう、インビボまたはインビトロで樹状細胞に負荷するのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、主要組織適合性複合体(MHC)クラス1aに依存しないT細胞のクロスプライミングを行うのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、インビボで抗腫瘍活性を刺激するのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、インビボで炎症反応を刺激するのに十分な量を含むことを特徴とする、請求項1に記載の組成物。
- さらに、K63結合型ポリユビキチニル化短寿命タンパク質およびK63結合型ポリユビキチニル化欠陥リボソーム産物を含むことを特徴とする、請求項1に記載の組成物。
- 前記閾値量は、インビボおよび/またはインビトロで、未感作T細胞に免疫反応を初回抗原刺激するのに十分な量を含むことを特徴とする、請求項8に記載の方法。
- 前記閾値量は、腫瘍特異性および/または癌特異性のT細胞の増殖を刺激するのに十分な量を含むことを特徴とする、請求項8に記載の方法。
- 前記閾値量は、抗原がT細胞に交差提示されるよう、インビボまたはインビトロで樹状細胞に負荷するのに十分な量を含むことを特徴とする、請求項8に記載の方法。
