Abstract
本发明属于中药技术领域,具体涉及一种荆防败毒口服液,其原料包括以下重量份的组分:荆芥8~12份、防风8~12份、羌活6~10份、独活6~10份、党参5~8份、白术5~8份、黄芪5~8份、当归5~8份、川芎4~6份、枳壳3~5份、柴胡4~6份、升麻4~6份、茯苓4~6份、陈皮3~5份、前胡3~5份、桔梗3~5份、甘草3~5份,并根据方中各药物有效成分的性质特点,以化学成分和药理效应为指标,制定合理的制备路线。本发明荆防败毒口服液能在提高免疫力的基础上祛邪,以达到标本兼治的目的。
Description
技术领域
本发明属于中药技术领域,具体涉及一种荆防败毒口服液及其制备方法。
背景技术
传染性法氏囊病是由法氏囊病病毒引起的一种严重危害雏鸡的免疫抑制性、高度接触性疫病。该病发病率高,病程短,呈尖峰式死亡曲线,常伴发大肠杆菌、沙门氏菌、球虫或腺病毒的继发感染,给养鸡场(户)带来巨大的经济损失。一方面是鸡只死亡,影响增重等直接损失;另一方面是免疫抑制,使接种了多种有效疫苗的鸡免疫应答下降或无免疫应答,也由于免疫机能下降,使患病鸡对多种病原的易感性增加。目前,预防该病的主要措施是免疫接种,但由于变异毒株的存在、母源抗体参差不齐、免疫程序不当等多种原因经常导致免疫失败。当发病时,早期紧急注射高免卵黄抗体虽能减少死亡,但该法易人为传播某些疾病;所以要从根本上减少或杜绝该病的发生,除了搞好鸡舍消毒、加强平时的饲养管理等常规措施外,应从增强鸡只的内在免疫功能入手,提高其免疫机能和抗病能力。在临床上,有养殖户于发病早期在饲料或饮水中添加黄芪多糖等增加免疫力的中药提取物,可以有效降低鸡群的死亡率。
经典名方荆防败毒散是临床上防治该病常用的方剂,方中荆芥、防风、羌活、独活四味药物相互协同,旨在直接祛除外邪,使用时拌在饲料中混饲,临床效果也不错。但散剂一般为粗粉直接入药,细胞的破壁率极低。当药物进入胃肠中,有效成分根据简单扩散原理不断地通过细胞壁及细胞膜释放出来,由小肠吸收。因为药物粒粗,位于粒子内部的有效成分将穿过几个或数十个细胞壁及细胞膜方可释放出来,释放速度很慢。又因为禽类的肠道较短,药物在体内停留时间有限,很快就被排出体外,吸收利用率低、见效慢,在临床上单位体重用量大,造成很大浪费;另外,禽类感染该病后,轻者饮食量明显下降,重者饮食基本废绝,在这样的情况下把药物拌在饲料中混饲,药物的服用量会大打折扣,保证不了服用量,临床疗效就会受影响;第三,荆防败毒散主要是祛邪作用突出,对于提高鸡群免疫机能方面没有明显效果,所以也不能从根本上预防该病的发生。
中国发明专利CN102362909公开了一种荆防败毒口服液的制备方法,为纯中药制取、作用迅速、见效快、效果好,但其亦是突出祛邪作用,对于提高鸡群免疫机能方面没有明显效果,所以也不能从根本上预防该病的发生。
发明内容
基于以上几方面的原因,本发明提供一种荆防败毒口服液,能在提高免疫力的基础上祛邪,以达到标本兼治的目的。
为解决上述问题,本发明通过以下技术方案实现:
设计一种荆防败毒口服液,其原料包括以下重量份的组分:荆芥8~12份、防风8~12份、羌活6~10份、独活6~10份、党参5~8份、白术5~8份、黄芪5~8份、当归5~8份、川芎4~6份、枳壳3~5份、柴胡4~6份、升麻4~6份、茯苓4~6份、陈皮3~5份、前胡3~5份、桔梗3~5份、甘草3~5份。
对于上述的荆防败毒口服液,其原料包括以下重量份的组分:荆芥9~11份、防风9~11份、羌活7~9份、独活7~9份、党参6~7份、白术6~7份、黄芪6~7份、当归6~7份、川芎4~5份、枳壳3~4份、柴胡4~5份、升麻5~6份、茯苓4~5份、陈皮3~4份、前胡4~5份、桔梗3~4份、甘草4~5份。

本发明荆防败毒口服液的制备方法,包括以下步骤:
(1)原料药浸润:
将上述原料药投入提取罐,加入其重量8~10倍的纯化水,加热使水温保持在35~40℃,保持30~45分钟,得浸润药液;
(2)提取:
第一次煎煮:将浸润药液加热至沸腾,保持2小时,将药液过滤至储液罐,药渣回收至提取罐;
第二次煎煮:向药渣中加入其重量6~8倍的纯化水,加热至沸腾,保持1.5小时,再将药液过滤至储液罐,药渣回收至提取罐;
第三次煎煮:向药渣中加入其重量4~6倍的纯化水,加热至沸腾,保持1小时,再将药液过滤至储液罐;
以上三次煎煮过程中收集挥发油(提取罐挥发油收集器);
(3)浓缩:
将三次煎煮提取的药液合并,浓缩至密度1.1~1.2kg/L,得浓缩药液;
(4)分离:
将浓缩药液进行离心分离,使离心转速16500~17000转,将杂质分离出来(收率不低于98%);
(5)精制:
在分离后的浓缩药液中加入无水乙醇,使乙醇含量达到65~70wt%,静置、冷藏24小时后,抽滤,除去药液中的乙醇(至无醇味),得到精制药液;
(6)挥发油回收:
在收集到的挥发油中加入等体积的无水乙醇,充分搅拌后,再加入温度为40~55℃的β-环糊精饱和溶液(挥发油体积的4~8倍)中,搅拌1.5小时,冷却、过滤、干燥,得到挥发油粉末;
(7)混合:将所得挥发油粉末加入所得精制药液中,溶解,即得本发明荆防败毒口服液。
本发明的积极有益效果:
(1)本发明方内含荆芥、防风、茯苓、独活、柴胡、前胡、川芎、枳壳、羌活、桔梗、甘草,在于祛邪;内含黄芪、白术、党参、当归、陈皮、升麻,在于扶正;内含陈皮、前胡,在于和解和调理机体全身气机;故本发明充分体现扶正祛邪并用、补泻与调理并用的组方思想,在控制外感病邪,尤其在防治疫疠之气方面充分考虑到控制外因,协调内因,在临床上用于防治鸡法氏囊病的应用上取得了较好的效果。
(2)本发明口服液除可以有效防治鸡传染性法氏囊病外,对禽流感、鸡传染性喉气管炎等病毒性疾病也有非常好的防治效果,发病初期效果尤佳,可配合西药应用,治疗混合感染。
(3)本发明根据方中各药物有效成分的性质特点,以化学成分和药理效应为指标,制定合理的制备路线,制成口服液后,应用到临床后,能够有效提高该方的利用率,增强临床疗效,为养殖户减少损失,创造更大的经济效益。
(4)本发明口服液应用于禽病的防治后,即可混饲,又可混饮,使用方便。首先不会因为病鸡的食欲下降不采食而影响药物的服用量,因为病鸡还要正常饮水,把口服液拌在饮水中就能解决该问题;另外,本发明口服液到体内后能够直接被吸收,速度较快,避免由于鸡的肠道短、药物排出快而造成的药物吸收不完全。本发明制备工艺对提高中兽药的临床效果、节省药材资源、方便服用、降低成本等具有重要意义。

一种荆防败毒口服液,其原料由以下重量份原料组成:荆芥10份、防风10份、羌活8份、独活8份、党参6份、白术6份、黄芪6份、当归6份、川芎5份、枳壳4份、柴胡5份、升麻5份、茯苓5份、陈皮4份、前胡4份、桔梗4份、甘草4份。

一种荆防败毒口服液,其原料由以下重量份原料组成:荆芥8份、防风12份、羌活6份、独活10份、党参5份、白术8份、黄芪5份、当归8份、川芎4份、枳壳5份、柴胡4份、升麻6份、茯苓4份、陈皮5份、前胡3份、桔梗5份、甘草3份。

一种荆防败毒口服液,其原料由以下重量份原料组成:荆芥11份、防风9份、羌活9份、独活7份、党参7份、白术6份、黄芪7份、当归6份、川芎5份、枳壳3份、柴胡5份、升麻5份、茯苓5份、陈皮3份、前胡5份、桔梗3份、甘草5份。
实施例6
实施例1~5荆防败毒口服液的制备方法,包括以下步骤:
(1)原料药浸润:
将上述原料药投入提取罐,加入其重量8~10倍的纯化水,加热使水温保持在35~40℃,保持30~45分钟,得浸润药液;
(2)提取:
第一次煎煮:将浸润药液加热至沸腾,保持2小时,将药液过滤至储液罐,药渣回收至提取罐;
第二次煎煮:向药渣中加入其重量6~8倍的纯化水,加热至沸腾,保持1.5小时,再将药液过滤至储液罐,药渣回收至提取罐;
第三次煎煮:向药渣中加入其重量4~6倍的纯化水,加热至沸腾,保持1小时,再将药液过滤至储液罐;
以上三次煎煮过程中收集挥发油(提取罐挥发油收集器);
(3)浓缩:
将三次煎煮提取的药液合并,浓缩至密度1.1~1.2kg/L,得浓缩药液;
(4)分离:
将浓缩药液(经三足式离心机)进行离心分离,使离心转速16500~17000转,将杂质分离出来(收率不低于98%);
(5)精制:
在分离后的浓缩药液中加入无水乙醇,使乙醇含量达到65~70wt%,静置、冷藏24小时后,抽滤,除去药液中的乙醇(至无醇味),得到精制药液;
(6)挥发油回收:
在收集到的挥发油中加入等体积的无水乙醇,充分搅拌后,再加入温度为40~55℃的β-环糊精饱和溶液(挥发油体积的6倍)中,搅拌1.5小时,冷却、过滤、干燥,得到挥发油粉末;
(7)混合:将所得挥发油粉末加入所得精制药液中,溶解,即得本发明荆防败毒口服液。
鸡传染性法氏囊病效果试验
1、试验动物
15日龄海兰蛋鸡490羽,河南省农科院种鸡场提供。在发病之前鸡场已按照免疫程序于7日龄做过新城疫支气管二联疫苗的饮水免疫,14日龄做IBD弱毒疫苗的饮水免疫。免疫后1d发病,经临床症状,病理剖检变化确诊为传染性法氏囊病。
2、试验药品
实施例1荆防败毒口服液,以及利巴韦林、鸡传染性法氏囊病、卵黄抗体(抗体交效价1∶128)、多种维生素、阿莫西林等药品由河南省农科院种鸡场提供。
3、试验方法
将患病鸡随机分为4组。由于病鸡食欲下降,饮欲增加,所以均采用饮水给药。
高剂量组用荆防败毒口服液,按全天量集中1次饮水。
中剂量组用荆防败毒口服液,按全天量集中1次饮水。
低剂量组用荆防败毒口服液,按全天量集中1次饮水。
利巴韦林组,利巴韦林每50g溶于100kg水中集中1次饮水。
法氏囊卵黄抗体组,每只肌注1.5mL。(注射时,要将卵黄抗体预温至鸡体温,以防注射时引起应激死亡现象。)
对照组不加任何药物。
全群饮水中加阿莫西林,集中在2h内饮完,预防继发感染;全天饮水添加水溶性多种维生素。

对表2数据进行显著性检验,结果表明:采用荆防败毒口服液治疗IBD病鸡,高剂量组有效率91.4%,治愈率达77.1%,低剂量组有效率72.8%,治愈率达70%,与利巴韦林组和法氏囊卵黄抗体对照组相比,差异显著(P<0.05),与感染对照组差异极显著(P<0.01)。

对表3数据进行显著性检验,结果表明:采用荆防败毒口服液治疗IBD病鸡,无论是高剂量组、中剂量组还是低剂量组均能使试验鸡的体重显著增加,和利巴韦林组相比,差异极显著(P<0.01);和法氏囊卵黄抗体对照组相比,差异显著(P<0.05)。
应用于其它疾病的典型病例
1、鹤壁市腾达禽业养殖场,养殖海兰褐青年鸡,3栋鸡舍20000多只因受寒感冒,采食量下降,日死亡300多只,使用荆防败毒口服液饮水后,连用5天,死亡率明显下降,日死亡大幅度降低,采食量恢复正常。
2、郑州隆成养殖公司,养殖肉鸡30000只,30日龄发病,病鸡体温42~43℃,精神不振,缩头呆立,羽毛蓬松,呼吸困难,采食量减少。个别鸡出现下痢。急性表现为鼻孔有分泌物和呼吸时发出湿罗音,而后出现痉挛性咳嗽和喘气。严重时出现明显的呼吸困难、咳嗽、举颈、甩头,咳出含有血液或血样的渗出物。有的病鸡结膜发炎、红肿、流泪,眼有浆液性或脓性渗出物。口腔检查可见喉部黏膜肿胀并有淡黄色凝固物附着,不易撕去。初步诊断为传染性喉支气管炎,随使用荆防败毒口服液饮水,连用5日,鸡群精神状况明显好转,呼吸道症状减轻,死亡率明显降低,7日后,鸡群鸡群恢复正常。

1.一种荆防败毒口服液,其特征在于,包括以下重量份的组分:荆芥8~12份、防风8~12份、羌活6~10份、独活6~10份、党参5~8份、白术5~8份、黄芪5~8份、当归5~8份、川芎4~6份、枳壳3~5份、柴胡4~6份、升麻4~6份、茯苓4~6份、陈皮3~5份、前胡3~5份、桔梗3~5份、甘草3~5份。
2.根据权利要求1所述的荆防败毒口服液,其特征在于,包括以下重量份的组分:荆芥9~11份、防风9~11份、羌活7~9份、独活7~9份、党参6~7份、白术6~7份、黄芪6~7份、当归6~7份、川芎4~5份、枳壳3~4份、柴胡4~5份、升麻5~6份、茯苓4~5份、陈皮3~4份、前胡4~5份、桔梗3~4份、甘草4~5份。

4.一种荆防败毒口服液的制备方法,其特征在于,包括以下步骤:
(1)原料药浸润:
将权利要求1所述原料药投入提取罐,加入其重量8~10倍的纯化水,加热使水温保持在35~40℃,保持30~45分钟,得浸润药液;
(2)提取:
第一次煎煮:将浸润药液加热至沸腾,保持2小时,将药液过滤至储液罐,药渣回收至提取罐;
第二次煎煮:向药渣中加入其重量6~8倍的纯化水,加热至沸腾,保持1.5小时,再将药液过滤至储液罐,药渣回收至提取罐;
第三次煎煮:向药渣中加入其重量4~6倍的纯化水,加热至沸腾,保持1小时,再将药液过滤至储液罐;
以上三次煎煮过程中收集挥发油;
(3)浓缩:
将三次煎煮提取的药液合并,浓缩至密度1.1~1.2kg/L,得浓缩药液;
(4)分离:
将浓缩药液进行离心分离,使离心转速16500~17000转,将杂质分离出来;
(5)精制:
在分离后的浓缩药液中加入无水乙醇,使乙醇含量达到65~70wt%,静置、冷藏24小时后,抽滤,除去药液中的乙醇,得到精制药液;
(6)挥发油回收:
在收集到的挥发油中加入等体积的无水乙醇,充分搅拌后,再加入温度为40~55℃的β-环糊精饱和溶液中,搅拌1.5小时,冷却、过滤、干燥,得到挥发油粉末;
(7)混合:将所得挥发油粉末加入所得精制药液中,溶解,即得本发明荆防败毒口服液。
5.根据权利要求4所述荆防败毒口服液的制备方法,其特征在于:所述β-环糊精饱和溶液用量是挥发油体积的4~8倍。
