Abstract
本发明公开了一种双靶向超声造影剂及其制备方法,所述造影剂是Integrin αvβ3/MCP‑1双靶向微泡,所述双靶向微泡包括脂质外壳和气体内芯,所述脂质外壳的外表面连接有Integrin αvβ3单克隆抗体和MCP‑1单克隆抗体。本发明通过不同靶点的联合应用,可以有效提高造影剂的靶向效能,大幅提高其携带基因在靶组织的富集程度。此外,本发明的造影剂是一种兼具显影和治疗双功能的超声造影剂,通过超声成像系统可实时、准确地观察肿瘤成像;通过高强度超声辐照实现基因转染后,可达到肿瘤基因治疗的效果。

技术领域
本发明涉及超声造影剂技术领域,尤其涉及一种双靶向超声造影剂及其制备方法。
背景技术
传统的超声造影剂是以磷脂、白蛋白等膜材料包裹气体制备而成的微泡,这些含气微泡的粒径大小约数个微米,在体内具有与红细胞类似的特征,能够通过体循环及肺循环,在临床上多用以显示整个血池系统的分布及完整性。靶向超声造影剂与普通造影剂最显著的区别就是能够在分子水平上识别靶组织的特异性抗原表位,并与之牢固结合,最终使靶组织特异性显影。为了达到这一目的,就必须对造影剂进行靶向修饰。
目前国内外研究中较常用的方法主要有以下两种:
一种方法是,在造影剂外膜成分中加入对某一疾病特异性位点具有亲和力的成分,以达到靶向的目的。Christiansen JP等人将磷脂酰丝氨酸添加入微泡外膜成分中,使其能够靶向附着炎症区域中活化的中型粒细胞及单核细胞(Christiansen JP,Leong-PoiH,Klibanov AL,et al.Noninvasive imaging of myocardial reperfusion injuryusing leukocyte-targeted contrast echocardiography circulation 2002;105(15):1764-1767)。而刘学兵等人在造影剂膜成分中加入了硫酸脑苷脂,后者对肿瘤细胞外基质的糖蛋白有特殊亲和力,从而成功制备了一种针对肿瘤组织的靶向造影剂(刘学兵,王志刚,许川山,等.纳米级靶向超声造影剂的制备及对兔VX2肝肿瘤的显影试验.中国超声医学杂志,2009;25(1):5-8)。但是总体而言,这种修饰方式适用的靶点有限,并不能在临床上得到广泛应用。微泡与靶细胞并非特异性结合,势必影响其对疾病的诊断及治疗效果。
另一种方法是,运用针对某种疾病特异位点的配体(如抗体、肽等)对造影剂微泡外壳进行修饰。抗原-抗体的结合具有高度的特异性及亲和性,因此,只要找到感兴趣疾病的特征性位点,制备出高特异性及敏感性的靶向造影剂就成为可能。Lindner JR等通过生物素-抗生物素的桥接作用,使结合有抗P-选择素抗体的脂类微泡与活化白细胞表面的P选择素特异结合,提高微泡在炎症区域的滞留,从而对小鼠肾脏的缺血后炎症损伤进行早期检测(Lindner JR,Song J,Christiansen J,et al.Ultrasound assessment ofinflammation and renal tissue injury with microbubbles targeted to P-selectin.Circulation 2001;104(17):2107-2112.)。Weller GE等以细胞黏附分子-1(ICAM-1)为靶点制备出靶向造影剂应用于接受心脏移植的大鼠体内发现,植入组织配型不合心脏的大鼠超声图像上造影剂信号强度明显高于植入组织配型匹配心脏的大鼠(WellerGER,Lu E,Csikari MM,et al.Ultrasound imaging of acute cardiac transplantrejection with microbubbles targeted to intercellular adhesion molecule-1.Circulation 2003;108(2):218-224.)。这种方法虽然通过单克隆抗体等特异性配体的使用,能够实现微泡与靶细胞的特异性结合,但是由于单一靶点的设计,微泡在靶组织内很难达到理想浓度,从而影响其携带基因在靶组织内的浓度,进而影响后续的基因治疗。
综上所述,目前的靶向造影剂能够实现与靶细胞的特异性结合,但是单靶点设计的微泡只能与单一靶点结合,受血流剪切力的影响,单靶向微泡与靶组织往往不能紧密黏附,靶向能力低。这种缺陷在针对乳腺癌的造影剂中表现明显。
发明内容
本发明提供一种双靶向超声造影剂及其制备方法,通过不同靶点的联合应用,可以有效提高造影剂的靶向效能,大幅提高其携带基因在靶组织的富集程度。
根据本发明的第一方面,本发明提供一种双靶向超声造影剂,该造影剂是Integrinαvβ3/MCP-1双靶向微泡,该双靶向微泡包括脂质外壳和气体内芯,上述脂质外壳的外表面连接有Integrinαvβ3(整合素αvβ3)单克隆抗体和MCP-1(单核细胞趋化蛋白-1,Monocyte Chemotactic Protein 1)单克隆抗体。
作为本发明的优选方案,上述脂质外壳包括DSPC(二硬脂酰磷脂酰胆碱)、Stearic-PEI600(硬脂酸支链聚醚酰亚胺-600)、DSPE-PEG2000-Biotin(二硬脂酰磷脂酰乙醇胺-聚乙二醇2000-生物素)。更优选地,上述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin的摩尔比为7.5~9.5:0.1~1:0.4~1.5,最优选为8.5:0.5:1。
作为本发明的优选方案,上述气体内芯是全氟丙烷内芯。
作为本发明的优选方案,上述Integrinαvβ3单克隆抗体和MCP-1单克隆抗体为生物素化单克隆抗体;上述脂质外壳与上述单克隆抗体通过亲和素和生物素连接在一起。
作为本发明的优选方案,上述Integrinαvβ3单克隆抗体和MCP-1单克隆抗体的摩尔比为0.5~2:0.5~2,更优选为1:1。
作为本发明的进一步优选方案,上述脂质外壳包括DSPC(二硬脂酰磷脂酰胆碱)、Stearic-PEI600(硬脂酸支链聚醚酰亚胺-600)、DSPE-PEG2000-Biotin(二硬脂酰磷脂酰乙醇胺-聚乙二醇2000-生物素),上述气体内芯是全氟丙烷内芯,上述单克隆抗体是生物素化单克隆抗体,上述DSPE-PEG2000-Biotin连接亲和素,上述亲和素连接上述单克隆抗体。
根据本发明的第二方面,本发明提供一种制备第一方面的双靶向超声造影剂的方法,该方法包括如下步骤:
(1)将DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中;
(2)在惰性气流作用下除去上述三氯甲烷,使上述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin在容器壁上形成均匀薄膜;
(3)加入水性溶液后振荡至透明,然后分装于密封瓶中;
(4)用全氟丙烷置换瓶内空气,振荡后制得生物素化微泡;
(5)向上述生物素化微泡中加入亲和素,并在室温下孵育;
(6)加入Integrinαvβ3和MCP-1的生物素化单克隆抗体,室温下孵育,制得上述双靶向超声造影剂。
作为本发明的优选方案,上述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin的摩尔比为7.5~9.5:0.1~1:0.4~1.5,更优选为8.5:0.5:1。
作为本发明的优选方案,上述Integrinαvβ3和MCP-1的生物素化单克隆抗体的摩尔比为0.5~2:0.5~2,更优选为1:1。
作为本发明的最优选的方案,上述方法包括如下步骤:
(1)将摩尔比为8.5:0.5:1的DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中;
(2)在N2气流作用下除去三氯甲烷,使磷脂在试管壁上形成均匀薄膜;
(3)加入0.1M Tris缓冲溶液后置于水浴式超声振荡器中振荡至透明,分装于洁净西林瓶中密封;
(4)用全氟丙烷置换瓶内空气,置于机械振荡器振荡30s后制得生物素化微泡;
(5)将其加入PBS离心清洗后,取10mL微泡加入50μg亲和素,室温下孵育20min;
(6)再次离心后加入过量的摩尔比为1:1生物素化Integrinαvβ3及MCP-1单克隆抗体,室温下孵育20min,即制得Integrinαvβ3/MCP-1双靶向微泡的造影剂。
根据本发明的第三方面,本发明提供一种双靶向载基因超声造影剂,该双靶向载基因超声造影剂包括如第一方面的双靶向超声造影剂以及包裹于上述双靶向超声造影剂的微泡内的重组质粒。
根据本发明的第四方面,本发明提供一种制备第三方面的双靶向载基因超声造影剂的方法,该方法包括将如第一方面的双靶向超声造影剂与重组质粒混合并孵育。
相比现有技术,本发明的超声造影剂具有如下优势:本发明采用Integrinαvβ3和MCP-1双靶向设计,将这两个在乳腺癌组织中高表达的位点结合起来,使得微泡的靶向效率得以提高,能够大幅提高其携带基因在靶组织的富集程度。同时,本发明的造影剂是一种兼具显影和治疗双功能的超声造影剂,通过超声成像系统可实时、准确地观察肿瘤成像;通过高强度超声辐照实现基因转染后,可达到肿瘤基因治疗的效果。此外,在本发明的优选的方案中,将Stearic-PEI600加入成膜材料中,制得的微泡表面带正电荷,与质粒的吸附作用更强。

具体实施方式
下面通过具体实施方式结合附图对本发明作进一步详细说明。
Integrinαvβ3和MCP-1是在乳腺癌细胞中高表达的两个靶点。本发明基于这两个靶点,通过它们的单克隆抗体与微泡的连接,形成本发明的Integrinαvβ3/MCP-1双靶向微泡,即本发明的双靶向超声造影剂。
在本发明中,“双靶向超声造影剂”是指未负载基因的造影剂形式,可以用于通过超声成像系统实时、准确地观察肿瘤成像。而“双靶向载基因超声造影剂”是指负载有基因——如重组质粒——的造影剂形式,该种形式的造影剂不但能够通过超声成像系统实时、准确地观察肿瘤成像,而且能够通过高强度超声辐照实现基因转染后,达到肿瘤基因治疗的效果。
本发明的Integrinαvβ3/MCP-1双靶向微泡,包括脂质外壳和气体内芯,脂质外壳的外表面连接有Integrinαvβ3单克隆抗体和MCP-1单克隆抗体。
脂质外壳可以按照目前这类造影剂通常使用的原料和制备方法来制备。在本发明的一个优选的实施例中,脂质外壳包括DSPC(二硬脂酰磷脂酰胆碱)、Stearic-PEI600(硬脂酸支链聚醚酰亚胺-600)、DSPE-PEG2000-Biotin(二硬脂酰磷脂酰乙醇胺-聚乙二醇2000-生物素)。其中,Stearic-PEI600作为膜材料之一,制得的微泡表面带正电荷,与质粒的吸附作用更强;DSPE-PEG2000-Biotin带有生物素,能够用于结合亲和素,从而介导与生物素化单克隆抗体的连接。
作为内芯的气体只要是生物相容性的即可,例如氮气等惰性气体、全氟丙烷等氟碳类组织相容性气体等。在本发明的一个优选的实施例中,采用全氟丙烷作为气体内芯。
脂质外壳与单克隆抗体的连接方式可能有多种,例如通过抗原与抗体的特异性结合的连接、通过亲和素与生物素结合的连接等。在本发明的一个优选的实施例中,采用亲和素与生物素结合将脂质外壳与单克隆抗体连接在一起。
在本发明的一个最优选的实施例中,脂质外壳包括DSPC(二硬脂酰磷脂酰胆碱)、Stearic-PEI600(硬脂酸支链聚醚酰亚胺-600)、DSPE-PEG2000-Biotin(二硬脂酰磷脂酰乙醇胺-聚乙二醇2000-生物素),气体内芯是全氟丙烷内芯,Integrinαvβ3和MCP-1单克隆抗体是生物素化单克隆抗体,DSPE-PEG2000-Biotin连接亲和素,亲和素连接上述单克隆抗体。
本发明的一个优选的制备上述双靶向超声造影剂的方法,包括如下步骤:
(1)将DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中;
(2)在惰性气流作用下除去上述三氯甲烷,使上述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin在容器壁上形成均匀薄膜;
(3)加入水性溶液后振荡至透明,然后分装于密封瓶中;
(4)用全氟丙烷置换瓶内空气,振荡后制得生物素化微泡;
(5)向上述生物素化微泡中加入亲和素,并在室温下孵育;
(6)加入Integrinαvβ3和MCP-1的生物素化单克隆抗体,室温下孵育,制得上述双靶向超声造影剂。
其中,步骤(1)中,DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin的摩尔比可以在比较宽的范围内变化,例如摩尔比为7.5~9.5:0.1~1:0.4~1.5,优选为8.0~9.0:0.2~0.8:0.5~1.2,更优选为8.2~8.8:0.4~0.6:0.7~1.1,例如7.5:1:1.5、8:0.8:1.2、8.5:0.5:1、9:0.2:0.8、9.5:0.1:0.4,最优选8.5:0.5:1。
步骤(2)中,各种惰性气流均可用于除去三氯甲烷,例如氮气、氩气等,优选氮气。
步骤(3)中,水性溶液可以是各种组织相容性的溶液,例如各种缓冲液,优选Tris缓冲溶液,其浓度例如0.05~0.2M,优选0.1M。步骤(3)中的振荡可以采用水浴式超声振荡器;密封瓶可以采用洁净的西林瓶。
步骤(4)中,振荡可以采用机械振荡器实施,振荡时间比如10s~60s,优选30s。
步骤(5)中,在加入亲和素之前,可以先清洗生物素化微泡,例如加入PBS缓冲液离心清洗。生物素化微泡与亲和素的用量可以在较宽的范围内变化,例如10ml微泡加入5~200μg亲和素等,在本发明的一个实施例中,取10ml(1×108/ml)微泡加入50μg亲和素。在室温下孵育的时间例如5~30min,优选20min。
步骤(6)中,Integrinαvβ3和MCP-1的生物素化单克隆抗体,在原则上可以按照任何比例加入,然而考虑到二者发挥恰当的功能的需要,最好是二者用量比较接近,例如Integrinαvβ3和MCP-1的生物素化单克隆抗体的摩尔比为0.5~2:0.5~2,优选为1:1。在室温下孵育的时间例如5~30min,优选20min。
如上已经论述的,本发明的双靶向载基因超声造影剂,是指负载有基因——如重组质粒——的造影剂形式。这样的造影剂包括双靶向超声造影剂以及包裹于双靶向超声造影剂的微泡内的重组质粒。其中,重组质粒上可以携带某些基因或者具有靶向功能的序列。例如,重组质粒上可以携带能够特异性敲除或者沉默特定癌基因的序列,如shRNA序列。在本发明的一个优选的实施例中,成功构建了AKT-shRNA表达质粒,作为重组质粒,包裹到双靶向超声造影剂的微泡内部,能够靶向性地富集到肿瘤部位,并介导重组质粒对肿瘤细胞的转染,进而实现对AKT基因的沉默。图1形象地示出了本发明的一个优选实施例的双靶向载基因超声造影剂的结构示意图。
本发明的双靶向载基因超声造影剂可以通过将双靶向超声造影剂与重组质粒混合并孵育而制得。
以下通过实施例详细说明本发明的技术方案以及其效果。应当理解,以下实施例仅是示例性的,并不能理解为对本发明保护范围的限制,本发明的保护范围以权利要求的范围来界定。
实施例1制备Integrinαvβ3/MCP-1双靶向微泡
1.将摩尔比为8.5:0.5:1的DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中。
2.在N2气流作用下除去三氯甲烷,使磷脂在试管壁上形成均匀薄膜。
3.加入适量0.1M Tris缓冲溶液后置于水浴式超声振荡器中振荡至透明,分装于洁净西林瓶中密封。
4.用全氟丙烷置换瓶内空气,置于机械振荡器振荡30s后制得生物素化微泡。
5.将其加入PBS离心清洗后,取10ml(1×108/ml)微泡加入50μg亲和素,室温下孵育20min。
6.再次离心后加入过量的摩尔比为1:1生物素化Integrinαvβ3及MCP-1单克隆抗体,室温下孵育20min,即制得Integrinαvβ3/MCP-1双靶向微泡。图2示出了本实施例制备的Integrinαvβ3/MCP-1双靶向超声微泡造影剂;经验证,其具有良好的显影效果,平均灰阶强度为156.97±18.05(图3)。
7.微泡基本性质检测:上述制备的微泡加入Accusizer 780粒度分析仪进行分析,分别测定其粒径、浓度;用Marlven电位分析仪测量微泡表面电荷。
实施例2检测Integrinαvβ3/MCP-1双靶向微泡体内、外靶向效能
1.培养乳腺癌细胞(MCF-7):用含10%胎牛血清的DMEM培养基,在37℃,5%CO2,100%饱和湿度条件下常规传代培养。
2.检验双靶向微泡的体外寻靶效率:将Integrinαvβ3、MCP-1的单克隆抗体以不同荧光标记,分别制备Integrinαvβ3单靶向、MCP-1单靶向、Integrinαvβ3/MCP-1双靶向以及非靶向对照纳米微泡,方法同实施例1。将制备的各组微泡加入体外培养的乳腺癌细胞(MCF-7)培养皿中,室温反应30min,PBS冲洗2次,荧光显微镜下观察各组微泡与癌细胞的靶向结合情况并通过其荧光强度进行比较靶向能力差异。图4所示的结果显示,本发明设计的双靶向超声造影剂微泡具备与MCF-7靶向结合能力,且较之Integrinαvβ3或MCP-1单靶向微泡,双靶向超声造影剂的靶向效能明显增高。
3.建立乳腺癌裸鼠皮下移植瘤模型:取对数生长期的MCF-7细胞,制成单细胞悬液。按照无菌手术操作原则,将MCF-7细胞悬液注射于裸鼠背部皮下,以诱导肿瘤形成。术后,于SPF条件下继续饲养,密切观察,待肿瘤直径达5-10mm时开始实验。
4.检验双靶向微泡的体内寻靶效率:将荷瘤鼠随机分为四组,分别将制备的不同荧光标记的Integrinαvβ3单靶向、MCP-1单靶向、Integrinαvβ3/MCP-1双靶向以及非靶向对照微泡,经尾静脉注入四组荷瘤鼠体内,利用超声诊断系统评价各组微泡靶向显影效果。图5示出了4组超声造影剂的靶向显影效果情况,其中图5中的白色箭头指示原始荧光图像中的绿色荧光信号。图5所示的结果显示,本发明设计的双靶向微泡靶向显影效果明显优于空白对照及Integrinαvβ3或MCP-1单靶向微泡组。然后取肿瘤组织行冰冻切片,利用共聚焦显微镜观察各组微泡在肿瘤内荧光显示强度,比较各组微泡靶向效率。
实施例3成功构建AKT-shRNA表达质粒,并将其包裹于双靶向微泡上
1.重组质粒的鉴定及筛选:委托基因公司根据基因库中AKT基因序列设计两对RNA干扰的靶点序列,合成shRNA重组质粒AKT2-1和AKT2-2(序列5'-3':CACCTTTGTCATACGCTGC和GGGCTAAAGTGACCATGAATG),采用脂质体法(Lipofectamine 2000)转染MCF-7乳腺癌细胞,运用荧光显微镜、流式细胞仪、CCK8法检测转染效果,筛选最佳质粒用于后续实验中。
2.载质粒双靶向微泡的制备:制备Integrinαvβ3/MCP-1双靶向微泡,方法同实施例1。将制备的AKT-shRNA重组质粒与双靶向微泡混合,充分孵育30min,制备双靶向载基因微泡。
实施例4双靶向载基因微泡在超声辐照下介导AKT-shRNA转染的体外实验
1.培养MCF-7细胞,方法同实施例2。
2.实验分组:①MCF-7+AKT-shRNA裸质粒组;②MCF-7+空白微泡;③MCF-7+Integrinαvβ3单靶向微泡携带AKT-shRNA重组质粒转染组;④MCF-7+MCP-1单靶向微泡携带AKT-shRNA重组质粒转染组;⑤MCF-7+Integrinαvβ3/MCP-1双靶向微泡携带AKT-shRNA重组质粒转染组。
3.根据分组将质粒(2μg)、单靶向微泡(3×107)或微泡复合物(2μg+3×107)加入培养板孔中,培养板于培养箱中静置孵育15分钟,使微泡附着于细胞。
4.通过聚焦超声探头(探头直径2.0cm,焦距2.5cm)给予超声辐照,辐照时间20s/孔。所用的超声参数为:频率1MHz,脉冲重复频率1kHz,占空比20%,声强1.65W/cm2。
5.孵育6h后吸弃转染复合溶液,更换为含血清和抗生素的培养基,放入37℃、5%CO2培养箱中继续培养。
6.转染效应检测方法:利用RT-PCR、Western-blot印迹法观察AKT mRNA及蛋白表达水平,运用MTT比色法检测细胞增殖,流式细胞仪检测细胞凋亡。
图6示出了不同的载基因微泡在超声辐照下介导AKT-shRNA转染的体外实验结果,其中图6中的白色箭头指示原始荧光图像中的绿色荧光信号。根据实验结果,本实施例设计的双靶向微泡靶向基因转染效果明显优于空白对照及Integrinαvβ3或MCP-1单靶向微泡组。
实施例5双靶向载基因微泡在超声辐照下介导AKT-shRNA转染的体内实验
1.建立乳腺癌裸鼠皮下移植瘤模型,方法同实施例2。
2.实验分组:①AKT-shRNA裸质粒组;②空白微泡对照组;③Integrinαvβ3单靶向微泡携带AKT-shRNA重组质粒转染组;④MCP-1单靶向微泡携带AKT-shRNA重组质粒转染组;⑤Integrinαvβ3/MCP-1双靶向微泡携带AKT-shRNA重组质粒转染组。
3.根据分组将质粒(5mg)、单靶向微泡(1×109)或微泡复合物(5mg+1×109)经尾静脉注入荷瘤鼠体内。
4.通过聚焦超声探头(探头直径2.0cm,焦距1.5cm)置于肿瘤处给予超声辐照,辐照时间50s。所用的超声参数为:频率1MHz,脉冲重复频率1kHz,占空比30%,声强1.85W/cm2。
5.肿瘤抑制效果观察。具体观察指标如下:①肿瘤体积及瘤重测量:于实验后不同时间点(3、6、9、12、15、18,21天)定期用游标卡尺测量肿瘤长径及短径。按以下公式计算肿瘤体积:肿瘤体积=0.5×肿瘤长径×肿瘤短径2,并绘制肿瘤生长曲线。实验最后处死裸鼠,摘出移植瘤并称取瘤重,计算抑瘤率:抑瘤率=(对照组瘤重-治疗组瘤重)/对照组瘤重×100%。②评价AKT mRNA含量:实时定量PCR法(RT-PCR)分析肿瘤组织中AKT mRNA的表达量。③评价AKT蛋白表达情况:免疫组化染色或蛋白印迹杂交法分析瘤组织中AKT蛋白的表达。④微血管密度测量(MVD):每一肿瘤随机观察三张切片,每张切片选取密度较高的三个视野观察,最终以每高倍视野内微血管数的平均值作为微血管密度。⑤TUNEL染色检测肿瘤组织细胞凋亡:TUNEL染色,光镜下观察细胞凋亡情况并计算凋亡指数:凋亡指数=凋亡细胞数/细胞总数×100%。
5.安全性评价:行半数致死量(LD50)测定及最大耐受量试验,用以评估体内生物毒性作用。

5.根据权利要求4所述的双靶向超声造影剂,其特征在于,所述Integrinαvβ3单克隆抗体和MCP-1单克隆抗体的摩尔比为1:1。
6.根据权利要求1所述的双靶向超声造影剂,其特征在于,所述气体内芯是全氟丙烷内芯。
7.一种制备权利要求1-6任一项所述的双靶向超声造影剂的方法,其特征在于,所述方法包括如下步骤:
(1)将DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中;
(2)在惰性气流作用下除去所述三氯甲烷,使所述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin在容器壁上形成均匀薄膜;
(3)加入水性溶液后振荡至透明,然后分装于密封瓶中;
(4)用全氟丙烷置换瓶内空气,振荡后制得生物素化微泡;
(5)向所述生物素化微泡中加入亲和素,并在室温下孵育;
(6)加入Integrinαvβ3和MCP-1的生物素化单克隆抗体,室温下孵育,制得所述双靶向超声造影剂。
8.根据权利要求7所述的方法,其特征在于,所述DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin的摩尔比为7.5~9.5:0.1~1:0.4~1.5;
所述Integrinαvβ3和MCP-1的生物素化单克隆抗体的摩尔比为0.5~2:0.5~2。
9.根据权利要求8所述的方法,其特征在于,所述方法包括如下步骤:
(1)将摩尔比为8.5:0.5:1的DSPC、Stearic-PEI600、DSPE-PEG2000-Biotin溶解在三氯甲烷中;
(2)在N2气流作用下除去三氯甲烷,使磷脂在试管壁上形成均匀薄膜;
(3)加入0.1M Tris缓冲溶液后置于水浴式超声振荡器中振荡至透明,分装于洁净西林瓶中密封;
(4)用全氟丙烷置换瓶内空气,置于机械振荡器振荡30s后制得生物素化微泡;
(5)将其加入PBS离心清洗后,取10mL微泡加入50μg亲和素,室温下孵育20min;
(6)再次离心后加入过量的摩尔比为1:1生物素化Integrinαvβ3及MCP-1单克隆抗体,室温下孵育20min,即制得Integrinαvβ3/MCP-1双靶向微泡的造影剂。
10.一种双靶向载基因超声造影剂,其特征在于,所述双靶向载基因超声造影剂包括如权利要求1-6任一项所述的双靶向超声造影剂以及包裹于所述双靶向超声造影剂的微泡内的重组质粒。
11.一种制备如权利要求10所述的双靶向载基因超声造影剂的方法,其特征在于,所述方法包括将如权利要求1-6任一项所述的双靶向超声造影剂与重组质粒混合并孵育。
