Abstract

本发明为药用乙酰水杨酸酯衍生物,其分别为:乙酰水扬酸丹皮酚酯(化学名:2-(乙酰氧基)苯甲酸-2-乙酰基-5-甲氧基苯酯)、乙酰水杨酸-7-羟基异黄酮酯(化学名:7-[2-(乙酰氧基)苯甲酰氧基]-3-苯基-4H-1-苯并吡喃-4-酮)、乙酰水杨酸-4-羟甲基苯酯(化学名:2-(乙酰氧基)苯甲酸-4-羟甲基苯酯)、乙酰水杨酸香草醛酯(化学名:2-(乙酰氧基)苯甲酸-4-甲酰基-2-甲氧基苯酯),这四种新化合物的药理作用与乙酰水杨酸酯类药物相似,但抗血栓形成作用较强,适用于防治心脑血管栓塞性疾病。

乙酰水杨酸酯衍生物
本发明为乙酰水杨酸酯衍生物,是一类具有解热镇痛,抗血栓形成作用的新的乙酰水杨酸酯类药物。
目前现有技术中,乙酰水杨酸(乙酰氧基苯甲酸)酯类化合物主要有下列文献及专利报导:

上述文献中的化合物结构式分别为:

本发明的新化合物与上述三种化合物在基本化学结构及药理作用方面有部份相似之处,即都属于乙酰水杨酸酯衍生物。现有技术中的化合物(I)(III)具有解热镇痛抗炎作用,化合物(II)的药理作用尚在研究之中。而本发明的新化合物在化学结构上与现有技术(I)(II)(III)有明显差异,虽都具有乙酰水杨酸酯的基本化学结构

但由于取代基R不同,而生成了新的乙酰水杨酸酯衍生物。
本发明的目的在于寻找具有药理活性强,毒副作用小,可供临床选用的新乙酰水杨酸酯类药物。新化合物名称为:
(1)乙酰水杨酸丹皮酚酯(化学名:2-(Acetyloxy)benzoic acid2-acetyl-5-methoxy phenyl ester,2-(乙酰氧基)苯甲酸-2-乙酰基-5-甲氧基苯酯);(2)乙酰水杨酸-7-羟基异黄酮酯(化学名:7-[2-(Acetyloxy)benzoxy]-3-phenyl-4H-1-benzopyrane-4-one,7-[2-(乙酰氧基)苯甲酰氧基]-3-苯基-4H-1-苯并吡喃-4-酮);(3)乙酰水杨酸-4-羟甲基苯酯(化学名:2-(Acetyloxy)benzoic acid 4-hydroxy methyl phenyl ester,2-(乙酰氧基)苯甲酸-4-羟甲基苯酯);(4)乙酰水杨酸香草醛酯(化学名:2-(Acetyloxy)benzoic acid 4-formyl-2-methoxy phenyl ester,2-(乙酰氧基)苯甲酸4-甲酰基-2-甲氧基苯酯)。

本发明以乙酰水杨酸、丹皮酚、7-羟基异黄酮、4-羟甲基苯酚、香草醛等为原料,采用下列路线合成:I、乙酰水杨酸丹皮酚酯(1)的合成

II、乙酰水杨酸-7-羟基异黄酮酯(2)的合成

III、乙酰水杨酸-4-羟甲基苯酯(3)的合成

IV、乙酰水杨酸香草醛酯的合成

以上四种乙酰水杨酸酯衍生物及其合成方法,目前尚未见文献报导。
本发明四种新化合物的合成方法简便,原料易得、成本较低,收率较高。药理实验表明四种新乙酰水杨酸酯衍生物除具有乙酰水杨酸类药物的解热镇痛、抗炎、抑制血小板聚集等作用外,还具有扩张血管或改善血液循环等作用,尤其抗血栓形成作用较强,适用于防治心脑血管栓塞性疾病,如冠心病、脑血栓。由于新化合物在体内水解后具有药理活性,故口服给药较乙酰水杨酸类药物对胃肠刺激性小,病人易于接受。
实施例1  乙酰水杨酸丹皮酚酯(1)的制备
取乙酰水杨酸4.5g,二甲基甲酰胺(DMF)2滴,氯化亚砜2.2ml于65-70℃反应约2小时,除去过量氯化亚砜得酰氯,加入少量丙酮。另取氢氧化钠1.48g,丹皮酚3.53g,加水一甲苯及少量新洁尔灭,加入酰氯液,20℃以下反应。分离甲苯层,蒸去甲苯后,沉淀用乙醇重结晶,收率60.2%,mp91-92℃,IR(cm-1):3016,3110(V-C-CH),1770(VCH3COO-),1750(VAr-COO-),1690(V-COCH3)。NMR(ppm):2.29(S,3H,-OCOCH3)。2.49(S,3H,-COCH3),3.89(S,3H,-OCH3)。元素分析:计算值C 65.83%,H 4.91%,实测值C 65.64%,H 4.51%。
实施例2  乙酰水杨酸-7-羟基异黄酮酯(2)的制备
取乙酰水杨酸4.54g,氯化亚砜2.07ml,DMF2滴,65-70°反应得酰氯。另取氢氧化钠1.34g,7-羟基异黄酮5.71g,加水-甲苯,新洁尔灭及酰氯,按实施例1方法得白色沉淀,用丙酮重结晶,收率72.7%,mp170-172℃,IR:1770(VCH3COO-)1750(VArCOO-),1640,1580(VCCOC)。NMR:2.29(S,3H,-OCOCH3),6.48(S,H8’)8.40(S,H2’),7.15-8.37(m,11H,苯环氢)。元素分析:计算值C71.98%;H 4.03%,实测值 C 71.68%,H 4.33%。
实施例3 乙酰水杨酸-4-羟甲基苯酯(3)的制备
取乙酰水杨酸7.21g,氯化亚砜5.81ml,DMF3滴,65-70°反应得酰氯。另取氢氧化钠2.56g,对羟甲基苯酚1.4g,加水一二氯甲烷,少量新洁尔灭及酰氯,参照实施例1法得沉淀,用丙酮重结晶,收率62.8%,mp107-108℃。IR:3400(VOH),1770(VCH3COO-),1755(VAr-COO-)。NMR:2.30(S,3H,-OCOCH3),4.71(S 2H,-CH20-),7.72(q,H4),8.24(q,H6)。元素分析:计算值  C 67.12%,H4.93%,实测值C 67.35% H 4.62%。
实施例4  乙酰水杨酸香草醛酯(4)的制备
取乙酰水杨酸4.32g,氯化亚砜3.2ml,DMF2滴,65-70℃反应得酰氯。另取氢氧化钠2.4g,香草醛3.12g,加水,二氯甲烷及少量新洁尔灭,参照实施例1方法得沉淀,用丙酮重结晶,收率73.85%,mp.118-120℃。IR:1770(VCH3COO-),1750(VArCOO-),1670(V-CHO)。NMR:2.29(S,3H,OCOCH3),3.89(S,3H,-OCH3),9.96(S,1H,-CHO)。元素分析:计算值:C 64.96%,H4.49%,实测值:C 64.65%,H 4.28%。
