Abstract

本发明公开了一种基于polyA侨联的双茎环DNA电化学传感器,包括:电极和捕获探针DNA,电极采用金电极,捕获探针DNA为含有两个茎环的单链DNA,DNA链的3’端修饰亚甲基蓝,DNA链的5’端修饰二茂铁,两个茎环的中间以polyA相连接,通过polyA与金电极的作用使捕获探针DNA以双发夹结构立于金电极表面。并公开了其制备方法和用于检测特定序列的方法。发明采用双茎环结构,DNA双颈环通过一段12个腺嘌呤碱基将双茎环DNA固定到金电极表面,即减少了实验的步骤,同时也可以实现等比例的将双目标DNA的捕获探针同时固定在电极表面,从而实现了双组份的同时检测。

技术领域
本发明属于生物电化学技术领域,特别涉及一种基于polyA侨联的双茎环DNA电化学传感器及其制备和应用。
背景技术
DNA作为一种十分重要的生物大分子,它是遗传信息的携带者和基因表达的物质基础。DNA在生物体遗传信息的存储、复制和传递的过程中起着至关重要的作用,因而影响生物的生长、发育、繁殖等正常生命活动。研究DNA的结构和功能,可以从分子水平上了解生命现象的本质。获取目标DNA的序列、浓度等信息,对基因诊断、基因筛选、检测基因变异以及新药开发等领域都具有重大意义。DNA的电化学研究始于20世纪60年代,早期的工作主要集中于DNA的基本电化学行为的研究。随着极谱学的发展,其它电分析化学方法也用于DNA的研究,其范围已经扩大到DNA的结构和形态、DNA探针及传感器的研究。
电化学DNA传感器主要是依据所加电活性指示剂与DNA单、双链作用的不同进行识别,它是将ssDNA探针固定在金电极、碳糊电极或玻璃电极等表面上,然后浸入含有目标ssDNA分子的溶液中,此时电极上的ssDNA探针与溶液中的互补序列的目标DNA单链分子杂交。杂交情况通过可检测的电信号检测。而根据电活性小分子在电化学DNA生物传感器中引起的电化学信号变化的原理不同,可将其分为两类:一类是以非标记型电活性小分子杂交指示剂作为识别元素;另一类是将具有电活性的小分子标记在DNA探针上,利用杂交前后探针上小分子的氧化还原电流的变化进行DNA检测。这种方法将分子信标引入到电极表面,而且不需要任何杂交后处理和外加指示剂,可以方便、直接地指示杂交的进行。
DNA探针在电极上的固定是电化学DNA传感器制备中的关键步骤。主要有吸附法、组合法、生物素-亲和素法、共价键合法、自组装法等。自组装法中DNA在金电极上的固定主要利用Au-S键:一种是在Au电极表面形成特殊官能团的巯基自组装单分子层(SAM),再共价键合或吸附DNA制备修饰电极;一种是将DNA链端巯基化,在通过-SH在Au表面的自组装作用制备DNA修饰电极。马丽等利用SAM技术,将巯己基修饰的具有乙肝病毒DNA固定在Au电极表面,成功地检测到血清乙肝病毒(马丽,白燕,刘仲明,等.血清样品中乙肝病毒的DNA电化学传感器检测.分析测试学报,2003,22:42-44)。其中将DNA固定到电极上的常规方法是通过DNA末端巯基固定到金电极上,AricOpdahl等报道了四大碱基与金电极表面的作用,A>G≈C>>T(AricOpdahl.ControlledandEfficientHybridizationAchievedwithDNAProbesImmobilizedSolelythroughPreferentialDNA-SubstrateInteractions.Anal.Chem.2010,82,2803–2810)。由于腺嘌呤与金电极表面具有强作用力,一系列利用polyA在金电极表面固定的电化学DNA传感器产生(Pei,H.;Li,F.;Wan,Y.;Wei,M.;Liu,H.;Su,Y.;Chen,N.;Huang,Q.;Fan,C.DesignedDiblockOligonucleotidefortheSynthesisofSpatiallyIsolatedandHighlyHybridizableFunctionalizationofDNA-GoldNanoparticleNanoconjugates.J.Am.Chem.Soc.2012,134,11876-11879;Zhang,X.,SurfaceScienceofDNAAdsorptionontoCitrate-CappedGoldNanoparticles.Langmuir2012,28,3896-3902)。
发明内容
本发明的目的之一是提供一种可以双组份同时检测的DNA生物传感器,实现对特定目标序列的特异性检测。
目的之二是提供上述DNA生物传感器的制备方法。
目的之三是提供上述DNA生物传感器的制备方法用于检测特定序列的方法。
为了实现上述目的,本发明采用以下技术方案:一种基于polyA侨联的双茎环DNA电化学传感器,包括:电极和捕获探针DNA,电极采用金电极,捕获探针DNA为含有两个茎环的单链DNA,DNA链的3’端修饰亚甲基蓝,DNA链的5’端修饰二茂铁,两个茎环的中间以polyA相连接,通过polyA与金电极的作用使捕获探针DNA以双发夹结构立于金电极表面。
所述polyA含有12个碱基。
一种基于polyA侨联的双茎环DNA电化学传感器的制备方法,包括以下步骤:
(1)金电极的处理:在硼氢化钠溶液中浸泡电极1h,冲洗后依次用0.3μm和0.05μm氧化铝粉末打磨电极,在超纯水中超声2分钟;
(2)捕获探针的组装:将捕获探针DNA分散在CaCl2-TE缓冲溶液中,滴加捕获探针DNA溶液到已经处理好电极上,盖上电极帽,在恒温箱中培育,取出电极用冲洗缓冲液对电极进行冲洗。
所述硼氢化钠溶液的浓度为1mol/L,为硼氢化钠溶解在质量比1:1的超纯水和乙醇中。
所述CaCl2-TE缓冲溶液的pH值为7.4,采用以下步骤制备:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.4后,加入1mmol/LEDTA和1mol/LCaCl2混合即得CaCl2-TE缓冲溶液。
所述冲洗缓冲液为10mmol/LTris-HCl,采用以下步骤制备:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.4即得冲洗缓冲液。
所述步骤2的培育时间为1~12h。
一种基于polyA侨联的双茎环DNA电化学传感器用于检测特定序列的方法,包括以下步骤:
(1)根据目标物选择茎环DNA制备双茎环DNA电化学传感器,在-0.6~0.6V范围内,对培育冲洗好电极进行方波伏安法测定;
(2)用MCH对电极进行封闭1小时后,清洗并用氮气吹干;将用pH值为7.0的NaCl-TE培育的目标DNA滴加到电极表面,室温培育1h,同样条件进行电化学检测。
本发明设计了一种双颈环探针结构,就可以实现2种目标组分的同时检测。茎环中间一段12个腺嘌呤碱基将双茎环DNA固定到金电极表面。分别在双茎环DNA的3’端和5’端固定亚甲基蓝(MB)和二茂铁(Fc),在未发生杂交反应时,探针处于茎环构型,末端标记的亚甲基蓝分和二茂铁子处于电极的近端,而当溶液中含有靶序列时,由于杂交反应的发生使茎环结构打开,形成双链DNA,末端标记的亚甲基蓝和二茂铁分子就会从电极的近端移动到远端。这种距离的改变导致电子传递效率发生变化,通过检测亚甲基蓝和二茂铁分子氧化还原电流的变化可以方便快速地指示杂交反应的发生。
与现有技术比较,本发明具有以下优异效果:传统的发夹DNA电化学传感器是通过巯基将捕获探针固定到电极表面,而且只能实现一种目标DNA的检测。要实现2种组分的同时检测,需要同时固定2种颈环探针,这样既增加了实验繁琐程度,并且是不能保证两种发夹是1:1的比例固定在金电极表面。本发明采用双茎环结构,DNA双颈环通过一段12个腺嘌呤碱基将双茎环DNA固定到金电极表面,即减少了实验的步骤,同时也可以实现等比例的将双目标DNA的捕获探针同时固定在电极表面,从而实现了双组份的同时检测。

具体实施方式
下面结合附图和具体实施例对本发明作进一步的说明。
图1是polyA侨联的双茎环DNA电化学传感器进行单组份检测的原理示意图,图2是polyA侨联的双茎环DNA电化学传感器进行双组份检测的原理示意图。
实施例
捕获探针DNA为5'-(Ferrocene)CCTTT(CAgAggCAgTAACCA)AAAggCCAAAAAAAAAAAACCggATA(CCTggTgCCgTAgAT)TATCC(MB)-3',括号中的序列为茎环序列,根据目标物选择茎环序列。
双茎环DNA电化学传感器的构建:
在硼氢化钠溶液中浸泡电极1h,冲洗后依次用0.3μm和0.05μm氧化铝粉末打磨电极,在超纯水中超声2分钟,在0.5mol/L硫酸中,在-0.1~1.5V内循环扫描30圈处理后,在新的0.5mol/L的硫酸溶液中扫描金电极特征峰。
将双发夹结构的捕获探针分散在pH为7.4的CaCl2-TE缓冲溶液(制备方法:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.4后,加入1mmol/LEDTA和1mol/LCaCl2混合即得CaCl2-TE缓冲溶液)中,为浓度10μmol/L(捕获探针之前是100μmol/L保存在-20℃)。滴加5μL捕获探针DNA溶液到已经处理好电极上,盖上电极帽,防止外界环境的干扰,放在37℃恒温箱中培育12h,捕获探针通过polyA与金表面作用固定到金电极表面。取出电极,用10mmol/LTris-HCl对电极进行适当冲洗。氮气对电化学检测液除氧20分钟后,在-0.6~0.6V范围内,对培育冲洗好电极进行方波伏安法测定。
单组份检测:
在前面实验基础上,用氮气吹干电极,用2mg·mL-1MCH对电极进行封闭1小时。占据电极上未反应掉的活性位点,杜绝后面假阳性信号的产生。清洗换溶液清洗后,氮气轻轻吹干,再把pH7.0NaCl-TE缓冲溶液(制备方法:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.0后,加入1mmol/LEDTA和1mol/LNaCl混合即得NaCl-TE缓冲溶液)培育的一种目标DNA滴加到电极上,室温培育1小时,同样条件进行电化学检测。当一定浓度的一种目标DNA加入到反应体系后,核酸适体与一种目标DNA杂交形成特定的分子结构,一边发夹结构解开,此时MB或Fc远离金电极表面,所产生的电化学信号明显降低。通过电化学信号的变化与目标物浓度之间的变化关系,实现对单组分目标DNA高灵敏、特异性的检测。检测结果见图3.
双组份检测:
在前面实验基础上,同样是MCH封闭后的电极。在电极上滴加含两种目标物的溶液,在室温培育1小时后,同样条件进行电化学检测。当一定浓度的两种目标DNA加入到反应体系后,核酸适体与两种目标DNA杂交形成特定的分子结构,整个发夹结构解开,此时MB和Fc远离金电极表面,所产生的电化学信号明显降低。通过电化学信号的变化与目标物浓度之间的变化关系,实现对两种组分目标DNA高灵敏、特异性的检测。检测结果见图4。
虽然本发明已以较佳实施例公开如上,但实施例并不是用来限定本发明,任何熟悉此技艺者,在不脱离本发明之精神和范围内,自当可作各种变化或润饰,但同样在本发明的保护范围之内。因此本发明的保护范围应当以本申请的权利要求保护范围所界定的为准。

Claims (8)

1.一种基于polyA侨联的双茎环DNA电化学传感器,其特征在于包括:电极和捕获探针DNA,电极采用金电极,捕获探针DNA为含有两个茎环的单链DNA,DNA链的3’端修饰亚甲基蓝,DNA链的5’端修饰二茂铁,两个茎环的中间以polyA相连接,通过polyA与金电极的作用使捕获探针DNA以双发夹结构立于金电极表面。
2.根据权利要求1所述的传感器,其特征在于:所述polyA含有12个碱基。
3.一种根据权利要求1或2所述电化学传感器的制备方法,其特征在于包括以下步骤:
(1)金电极的处理:在硼氢化钠溶液中浸泡电极1h,冲洗后依次用0.3μm和0.05μm氧化铝粉末打磨电极,在超纯水中超声2分钟;
(2)捕获探针的组装:将捕获探针DNA分散在CaCl2-TE缓冲溶液中,滴加捕获探针DNA溶液到已经处理好电极上,盖上电极帽,在恒温箱中培育,取出电极用冲洗缓冲液对电极进行冲洗。
4.根据权利要求3所述的制备方法,其特征在于:所述硼氢化钠溶液的浓度为1mol/L,为硼氢化钠溶解在质量比1:1的超纯水和乙醇中。
5.根据权利要求3所述的制备方法,其特征在于:所述CaCl2-TE缓冲溶液的pH值为7.4,采用以下步骤制备:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.4后,加入1mmol/LEDTA和1mol/LCaCl2混合即得CaCl2-TE缓冲溶液。
6.根据权利要求3所述的制备方法,其特征在于:所述冲洗缓冲液为10mmol/LTris-HCl,采用以下步骤制备:配制10mmol/L的Tris-base,用0.2mol/LHCl调节pH至7.4即得冲洗缓冲液。
7.根据权利要求3所述的制备方法,其特征在于:所述步骤2的培育时间为1~12h。
8.一种根据权利要求1或2所述电化学传感器用于检测特定序列的方法,其特征在于包括以下步骤:
(1)根据目标物选择茎环DNA制备双茎环DNA电化学传感器,在-0.6~0.6V范围内,对培育冲洗好电极进行方波伏安法测定;
(2)用MCH对电极进行封闭1小时后,清洗并用氮气吹干;将用pH值为7.0的NaCl-TE培育的目标DNA滴加到电极表面,室温培育1h,同样条件进行电化学检测。
