技术领域
本发明涉及一种应用反向遗传操作技术生产的VII型新城疫病毒L基因突变的减毒疫苗株及其制备方法,属于生物制品和生物技术领域。
背景技术
新城疫是由新城疫病毒(Newcastle disease virus,NDV)引起,侵害鸡和火鸡,亦可感染其他禽类、鸟类的一种急性、高度传染性疾病,是世界公认的最重要的两大禽类传染病之一(另一个是高致病性禽流感)。 NDV属于副粘病毒科腮腺炎病毒属,具有囊膜结构,基因组为单股负链不分节段的RNA病毒,由6种结构蛋白组成,分别为NP、P、M、F、HN和L蛋白:M、F和HN蛋白与病毒囊膜的形成相关;NP蛋白为核衣壳蛋白,P蛋白是病毒RNA合成的必需因子;L蛋白为病毒最大的蛋白,它在NDV 6种结构蛋白中最为保守,具有RNA聚合酶、转录后修饰等生物学活性,可与NP和P蛋白共同构成病毒复制复合体,该复合体参与病毒基因组的转录和复制,L蛋白除了参与病毒基因组的复制外,还具有5’端加帽、帽依赖的甲基化转移酶活性及3’端添加多聚腺苷酸尾巴等功能。
不同NDV分离株之间的毒力差异很大,根据对鸡和鸡胚的致病性及毒力不同,可将NDV分成三类:即强毒型(速发型)、中等毒力型(中发型)和弱毒型(缓发型)。
以往研究认为NDV的感染性和毒力是由F基因决定,强毒株F蛋白裂解位点为RRQKRF,而弱毒株LaSota的F蛋白裂解位点为GRQGRL。但随着反向遗传学技术的不断发展及在NDV研究中的深入应用,NDV的致病机制又有了新的研究进展。Peeters、De Leeuw和Panda等将弱毒株的F基因裂解位点突变为与强毒株一致的裂解位点后拯救出了新的重组病毒,实验表明虽然其毒力明显提高,但却并没有达到与强毒株毒力一致的水平,这就表明在NDV基因组中除了F基因的裂解位点外,还可能存在其它能影响病毒毒力的基因。最近有研究指出,L、HN和V蛋白也与病毒的毒力和致病性有关,L蛋白可通过提高病毒的复制水平而增强NDV毒力。Rout等将NDV BC株的L基因替换为LaSota的L基因,病毒获救后毒力反而增强。另外,我们成功救获3株基因Ⅶ-97 NDV L突变株(K1756A、K1917A和E1954Q位氨基酸突变)和1株基因Ⅶ-97 NDV F与L共突变株(E1954Q+F),通过病毒毒力指标测定发现,这4株VII型新城疫病毒L基因突变的减毒疫苗株能够使病毒毒力减弱,我们有理由推断NDV L蛋白对病毒的毒力起着至关重要的作用。这些实验充分说明L蛋白与病毒的毒力存在联系。
当前世界上防控NDV主要疫苗株是LaSota和B1两种毒株,LaSota和B1两弱毒株都属于基因II型。但是国内的学者严维巍等在2000年首次报道了基因VII型NDV在中国大陆的存在;2001年,刘华雷等对1994年以来选取的国内不同地区的代表株进行遗传分析证实,所分离的8株毒株在裂解位点的氨基酸序列均与NDV的强毒株相似。通过遗传进化树分析表明,8株分离株中有7株为基因Ⅶ型NDV,这些数据充分表明我国当前主要流行基因Ⅶ型新城疫病毒。LaSota和B1两种弱毒疫苗作为疫苗毒株已经应用了几十年,疫苗尽管能提供一定的免疫保护,但不能阻止家禽的排毒和抑制病毒的传播能力,因此,新城疫依然是威胁养禽业发展的最主要的疫病之一,中国每年由此造成巨大的经济损失。而且当前中国流行的毒株主要是基因Ⅶ型,随着病毒的不断变异,针对基因Ⅶ型的同基因型疫苗的研发成为当前的主要任务,我国Ⅶ型的新城疫疫苗研制已经取得良好进展,一旦产业化后,必将成为规模化养殖场免疫程序中必要的一环。本发明为开展NDV减毒疫苗的研制和NDV相关基础研究奠定了坚实的基础。
发明内容
本发明的目的在于克服现有技术的不足而发明的VII型新城疫病毒L基因突变的减毒疫苗株及其制备方法,从而解决当前我国市场上所用的NDV疫苗株(基因Ⅰ和基因Ⅱ型)与目前NDV流行株(基因Ⅶ型)的基因型不一致而导致的保护效率不完全,不能有效阻止病毒传播的实际问题。
本发明是通过如下技术方案予以实现的:
一种VII型新城疫病毒L基因突变的减毒疫苗株,其特征在于:VII型新城疫病毒L基因突变的减毒疫苗株毒力均明显减弱,同时在鸡胚上具有高滴度、遗传性能稳定、与流行病毒的抗原匹配性高,适合于疫苗的大规模生产,可用于制造疫苗,该VII型新城疫病毒L基因突变的减毒疫苗株的抗原核苷酸序列为SEQ ID NO:1至SEQ ID NO:4所示。
一种所述的VII型新城疫病毒L基因突变的减毒疫苗株的制备方法,其特征在于:
构建覆盖整个基因组的五个cDNA克隆片段,利用各片段间重叠部分的酶切位点,在转录载体质粒pBR322连接组装获得了15192nt的完整cDNA克隆。在全长cDNA片段的5’末端前缀T7RNA聚合酶启动子,在cDNA片段后加入具有自我催化功能的丁肝病毒核酶(RiB)和T7转录终止信号,构建好的质粒命名为prNDV-97;通过PCR基因组将基因组cDNA中的L蛋白编码区第14056位碱基由A同义突变为G,消除14056bp处的Hind Ⅲ酶切位点,并作为拯救病毒的分子标记,构建完成的质粒命名为pmNDV-97;在T7聚合酶启动子于基因组cDNA的5’末端引入两个多余的G,有助于副粘病毒反向遗传操作的病毒拯救。

通过下列引物,分别经Overlapping PCR方法,将NDV亲本毒L蛋白结构域Ⅵ内的1756、1917和1954位氨基酸分别单点突变,将扩增后的目的片段和pmNDV-97载体经Hind Ⅲ和XbaⅠ限制性内切酶分别双酶切,进行核酸电泳,胶回收目的片段和载体pmNDV-97后进行连接,所获得的突变质粒分别命名为pK1756A、pK1917A和pE1954Q。

其中,AF1+AF6、AF1+AF7和AF1+AF8引物利用PCR方法分别扩出目的条带,分别命名为A1、B1和C1;AF5+AF2、AF5+AF3和AF5+AF4引物利用PCR方法分别扩出目的条带,分别命名为A2、B2和C2。在此基础上,利用AF1+AF5引物分别经Overlapping PCR方法,以A1+A2、B1+B2和C1+C2分别为模板,扩增约6000bp(含突变位点)的目的片段,分别获得L突变片段L1756、L1917和L1954。
一种NDV Ⅶ-97 L基因突变及F与L基因共突变致弱株的构建方法,具体为:以分离株NDV Ⅶ-97的基因组L基因进行定点突变,对L蛋白结构域Ⅵ内进行单点氨基酸突变,得到高度致弱的NDV Ⅶ-97 L减毒株K1756A、K1917A和E1954Q。在E1954Q的基础上,对F蛋白裂解位点进行突变,将强毒株的F蛋白裂解位点(RRQKRF)突变为LaSota弱毒株的F蛋白裂解位点(GRQGRL),所获得的减毒株为E1954Q+F。
用于突变F蛋白裂解位点的引物如下:

其中,AF9+AR10和AF10+AR9 2对引物利用PCR方法分别扩出目的条带,胶回收后再以胶回收产物为模板,以AF9+AR9这1对引物利用Overlapping 方法进行PCR扩增,获得含有F蛋白突变的片段长约5458bp ,以此获得的突变体为E1954Q +F。
一种VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q和E1954Q+F的构建方法,具体为:
1)NDV Ⅶ-97株基因组全长cDNA的构建;
2)NDV Ⅶ-97 L 蛋白结构域Ⅵ内氨基酸定点突变和突变体全长cDNA的连接;
3)在E1954Q突变的基础上,NDV Ⅶ-97 F蛋白裂解位点的突变和突变体全长cDNA的连接;
4)VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q
和E1954Q +F的拯救;
将质粒pK1756A、pK1917A、pE1954Q和pE1954Q+F分别与pBSK-NP、pBSK-P和pBSK-L三个真核表达质粒共转染BHK-21细胞(仓鼠肾细胞),转染72h后将细胞上清接种9-11日龄SPF鸡胚,每份样品接种3枚,0.2mL/枚,成功救获VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A和E1954Q及F蛋白与L蛋白共突变减毒疫苗株E1954Q +F。
本发明具有如下积极效果:
本发明利用已建立的NDV反向遗传操作平台,通过对相关基因改造,构建了病毒滴度高、遗传性能稳定、抗原匹配性高以及对宿主鸡等禽类致病力低的减毒疫苗株,这将为研制高效疫苗种毒奠定基础。与常规疫苗(基因Ⅰ和基因Ⅱ型)相比,VII型新城疫病毒L基因突变的减毒疫苗株将在控制新城疫的发病和流行方面显示出广阔的应用前景。

图1为利用高保真RT-PCR产生的亚基因组重叠cDNA片段装配全长NDV cDNA。将cDNA片段在共有的限制性酶切位点连接,并且在转录质粒pBR322中装配,在转录质粒pBR322中将PBZ和T7终止子序列预先克隆在其中。图1代表了连接全长cDNA的模式图。

图5为VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q、L与F共突变减毒株E1954Q+F和rNDV的间接免疫荧光分析。用K1756A、K1917A、E1954Q、E1954Q+F和rNDV 分别感染BHK-21细胞(同时设置阴性对照)。感染剂量为1 MOI,感染后24h将细胞固定,透膜,用鸡抗-NDV抗血清检测,接着与FITC-偶联的兔抗鸡IgG抗体孵育。在Leica荧光显微镜下观察荧光。
具体实施方式
本发明结合近年来的新城疫病毒株积累和对其全基因组序列的分析研究,将我国VII 型新城疫毒株L基因进行定点氨基酸突变及F与L基因的氨基酸共突变,并对L基因突变和L基因与F基因共突变的全长NDV基因组cDNA拯救,获救出高度致弱的VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q和E1954Q+F,这4株VII型新城疫病毒L基因突变的减毒疫苗株毒力明显减弱,同时在鸡胚上具有高滴度、遗传性能稳定、与流行病毒的抗原匹配性高,适合于疫苗的大规模生产,可用于疫苗的生产,该VII型新城疫病毒L基因突变的减毒疫苗株的抗原核苷酸序列为SEQ ID NO:1至SEQ ID NO:4所示。
下面结合具体实施实例对本发明做进一步地描述。
实施例1.转录载体的构建:
本发明人所用BHK-21细胞为仓鼠肾细胞;培养基为含10%胎牛血清(Gibco);NDVⅦ-97株由中国农业科学院兰州兽医研究所禽病研究室保存;SPF鸡胚及SPF雏鸡均购买于北京梅里亚维通生物技术有限公司。
NDV Ⅶ-97株的反向遗传操作平台是在pBR322质粒的基础上改造而成,分别在NDV全基因组的两端分别含有T7启动子和T7核酶转录终止序列,同时插入具有自我剪切功能的丁肝病毒核酶序列。克隆在T7启动子和核酶之间的DNA片段可以在T7RNA聚合酶的作用下得到转录,并且由于丁肝病毒核酶的自身催化功能,可以保证转录产物的3’末端与克隆的DNA片段精确一致。
实施例2.NDV基因组全长cDNA的构建:
构建覆盖整个基因组的5个cDNA克隆片段,利用各片段间重叠部分的酶切位点,在转录载体质粒pBR322连接组装获得了15192nt的完整cDNA克隆。在全长cDNA片段的5’末端前缀T7RNA聚合酶启动子,在cDNA片段后加入具有自我催化功能的丁肝病毒核酶和T7转录终止信号。构建完成的质粒命名为prNDV。为消除14056bp处Hind Ⅲ酶切位点,通过PCR基因组将基因组cDNA中的L蛋白编码区第14056位碱基由A同义突变为G,并作为拯救病毒的分子标记,我们同时在T7聚合酶启动子于基因组cDNA的5’末端引入两个多余的G,这可能有助于副粘病毒反向遗传操作的病毒拯救。

利用Trizol法提取NDV Ⅶ-97鸡胚接种尿囊液中的基因组RNA,详细步骤如下:
① 取鸡胚尿囊液250uL,加入750 uL Trizol(Invitrogen,USA),震荡混匀,室温放置5分钟;
② 每管加入氯仿200uL,盖紧离心管,剧烈摇荡离心管15秒;室温放置10min,12000rpm离心15min;
③ 取上层水相放置于新的离心管内,加700uL异丙醇,4度放置10分钟,12000rpm离心10分钟;
④ 弃去上清液,按每毫升 Trizol液加入至少1ml的比例加入75%乙醇,混匀,4℃下7500g离心5分钟;
⑤小心弃去上清液,然后室温干燥5-10分钟,注意不要干燥过分,否则会降低RNA的溶解度;最后将RNA溶于水中。将RNA反转录成cDNA后,利用PCR方法扩增各基因片段,切胶回收目的片段,备用 。
整个基因组分为末端部分重叠的5个片段(F1-F5)进行PCR扩增,cDNA片段克隆至pBR322真核表达载体上;核衣壳蛋白(NP)、磷蛋白(P)和大聚合酶蛋白(L)基因的开放阅读框(ORF)cDNA分别克隆在pBluescriptⅡ(+/-)质粒T7启动子下游,分别构成转录辅助质粒pBSK-NP、pBSK-P和pBSK-L。
克隆的相应基因组的全长cDNA是建立NDV反向遗传操作系统的基础,作为基因组负链RNA转录模板。
实施例3.NDV Ⅶ-97株 L基因进行定点氨基酸突变和L突变体全长cDNA的连接
通过引物分别经Overlapping PCR方法,将NDV Ⅶ-97株L蛋白结构域Ⅵ内的1756、1917和1954位氨基酸分别单点突变(表1),将扩增后的目的片段和prNDV-97载体经Hind Ⅲ和XbaⅠ限制性内切酶分别双酶切,进行核酸电泳,胶回收目的片段和载体prNDV-97后进行连接,所获得的突变质粒分别命名为pK1756A、pK1917A和pE1954Q。
在pE1954Q的基础上,对F蛋白裂解位点进行突变,将强毒株的F蛋白裂解位点(RRQKRF)突变为LaSota弱毒株的F蛋白裂解位点(GRQGRL)。将F蛋白突变位置通过引物分别经Overlapping PCR方法扩增,扩增后的目的片段和pE1954Q载体经SalⅠ和AfeⅠ限制性内切酶分别双酶切,进行核酸电泳,胶回收目的片段和载体pE1954Q后进行连接,所获得的突变质粒分别命名为pE1954Q +F。用于突变F蛋白裂解位点的引物如下:

实施例4.VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q和E1954Q+F的拯救
在转染前先用痘病毒感染BHK-21细胞,1h后洗细胞2-3遍,随后转染拯救病毒。将质粒pK1756A、pK1917A、pE1954Q和pE1954Q+F分别与pBSK-NP、pBSK-P和pBSK-L三个真核表达质粒共转染BHK-21细胞,72h后将细胞轻轻刮下,并与细胞上清充分混合过滤后接种9-11日龄SPF鸡胚,每份样品接种3枚,0.2mL/枚,接种后的SPF胚继续培养3天,取鸡胚尿囊液50uL按常规进行NDV的血凝(HA)和血凝抑制(HI)试验,获得VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q和E1954Q +F。
实施例5.VII型新城疫病毒L基因突变的减毒疫苗株K1756A、K1917A、E1954Q和E1954Q +F的鉴定
5.1遗传稳定性分析
实验方法:将VII型新城疫病毒L基因突变的减毒疫苗株(K1756A、 K1917A、E1954Q和E1954Q +F)分别使用9日龄SPF鸡胚传代至第10代,提取第10代病毒RNA,反转录成cDNA后扩增目的片段,并切胶回收目的片段,送样测序。
实验结果:各突变株突变位点依然存在,遗传稳定性好(图4)。

为检测减毒株(K1756A、K1917A、E1954Q和E1954Q+F)在BHK-21细胞内的复制,尿囊毒分别以MOI为1的病毒量感染约70%-80%的单层BHK-21细胞,在感染后24h,以NDV高免SPF鸡阳性血清为检测抗体进行间接免疫荧光试验,并设阴性对照。通过间接免疫荧光试验发现,获救病毒均能在BHK-21细胞上增殖。
详细步骤如下:
减毒株鸡胚尿囊毒分别以DMEM适当倍数稀释,按照MOI为1、300uL体积感染生长于六孔板的BHK-21细胞,37℃孵育1h后用DMEM洗涤三遍,然后加入完全DMEM继续培养,24h后用4%的多聚甲醛固定细胞10min,PBS洗细胞后用5%的BSA进行封闭30min,然后以NDV高免SPF鸡阳性血清为一抗,1:500倍稀释,37℃作用1h后用PBS洗5遍,加入1:800稀释的荧光素(FITC)标记的二抗(Sigma),作用45min,PBS洗后荧光显微镜观察结果(图5)。
实施例6.减毒株的生长特性及致病特性
实验目的:检测利用反向遗传技术救获的VII型新城疫病毒L基因突变的减毒疫苗株在鸡胚上的生长特性及其对SPF鸡的致病性。
实验方法:将K1756A、K1917A、E1954Q和E1954Q+F减毒株F4代尿囊液及rNDV分别按106EID50剂量接种9日龄SPF鸡胚尿囊腔,96小时内无死胚;96小时后将鸡胚冻死,收集尿囊液,HA滴度分别为25、26、26 、28和28,而每毫升尿囊液EID50则分别为2.15×109、3.16×109、1.78×109、1.78×109和1×1010(表2)。按照OIE标准测定rNDV及其减毒株的鸡胚半数致死量(MDT)、脑内接种致病指数(ICPI)和静脉接种致病指数(IVPI)的毒力指数测定。结果见表3(OIE 编著[农业部畜牧兽医局译]. 哺乳动物、禽类和蜜蜂 A 类和 B 类疾病诊断试验和疫苗标准手册. 北京:中国农业科技出版社,1996:140-146)。
实验结果:通过以上数据表明利用反向遗传操作技术救获的VII型新城疫病毒L基因突变的减毒疫苗株毒力显著低于亲本毒株,在鸡胚中具有较高的生长滴度和对SPF鸡毒力明显减弱的生物学特性。

* 病毒经适当比例稀释后,接种9日龄SPF鸡胚(100uL/只),能够使所有鸡胚致死的病毒最大稀释倍数即为病毒的最小致死量(MLD),MDT 是最小致死量(MLD)引起鸡胚死亡的平均时间,利用此方法测定MDT。
**病毒种毒经10倍稀释后脑内接种1日龄SPF雏鸡(50uL/只),测定ICPI。
***翅下静脉注射(1×106EID50/只),测定IVPI。
实施例7.诱导动物保护性抗体的免疫效果
实验目的:测定利用反向遗传操作系统救获的VII型新城疫病毒L基因突变的减毒疫苗株对2周龄SPF雏鸡的免疫保护性。
实验方法:利用9日龄SPF鸡胚分别扩增的病毒,1×106EID50剂量经滴鼻加点眼途径分别人工免疫10只白色来亨SPF雏鸡,另设非免疫对照组10只,免疫组和非免疫组分别饲养于空气负压过滤隔离器中。21天后翅静脉采血分离血清按常规检测NDV特异HI抗体。另外,在免疫后21天用强毒NDVⅦ-74(本室保存用种毒)以1×106EID50剂量经滴鼻加点眼途径进行攻毒,观察两周,统计发病与死亡情况。
实验结果:尿囊病毒液F4代以1×106EID50剂量经滴鼻加点眼途径人工免疫白色来亨SPF雏鸡,免疫后观察3周,期间4组免疫组所有雏鸡无任何异常,饲料消耗及生长发育与非免疫对照组无明显差异。免疫后3周检测血清NDV特异HI抗体水平(表4)。4组免疫组攻毒后未出现任何发病或死亡,完全保护(10/10),同日龄非免疫对照组则全部发病死亡(0/10)(表5)。结果显示,突变减毒株一次免疫雏鸡即可诱导高水平的保护性抗体反应,具有良好的保护效果。

实施例8.减毒株K1756A、K1917A、E1954Q和E1954Q+F排毒情况分析
实验目的:测定利用反向遗传操作系统救获的3株NDV Ⅶ-97 L突变减毒株和1株L与F共突变减毒株免疫后的排毒情况。
实验方法:本实验利用9日龄SPF鸡胚分别扩增的病毒,1×106EID50剂量经滴鼻加点眼途径分别人工免疫10只2周龄来亨SPF雏鸡,另设LaSota对照组10只,免疫组和非免疫组分别饲养于空气负压过滤隔离器中。在免疫后21天用强毒NDVⅦ-74(本室保存用种毒)以1×106EID50剂量经滴鼻加点眼途径进行攻毒,攻毒后的第3d和第6d分别采集喉拭子和泄殖腔拭子,利用SPF鸡胚进行滴定,检测病毒的排毒情况。
实验结果:免疫组及对照组免疫后攻毒,所用鸡均获得免疫保护(空白鸡除外),但免疫攻毒后检测排毒情况,LaSota喉拭子的排毒量高于其它4组L突变减毒株(表6)。

本发明采用稳定表达T7 RNA聚合酶系统的重组痘病毒感染BHK-21细胞,在转染细胞中转录全长基因组负链RNA,同时表达启动病毒复制所需的NP、P和L蛋白,取得了很高的病毒拯救效率。
利用反向遗传操作技术获救的3株NDV Ⅶ-97 L突变减毒株K1756A、K1917A和E1954Q和1株F蛋白裂解位点与L共突变减毒株E1954Q+F均可在鸡胚内达到与亲本rNDV相似的生长滴度,并且获救病毒以106EID50剂量尿囊腔内接种9日龄SPF鸡胚,96h内不致死鸡胚,HA分别为25、26、26和 28;每毫升尿囊液EID50分别为2.15×109、3.16×109、1.78×109、1.78×109,免疫14日龄SPF雏鸡可诱导高水平的保护性抗体反应,免疫后第21d抗体效价滴度分别为25、24、24和24,充分显示本发明的反向遗传操作系统获救4株VII型新城疫病毒L基因突变的减毒疫苗株可作为疫苗株。该减毒疫苗株具有良好的鸡胚生长特性及较高的病毒生长滴度,生产成本低廉且使用方便。NDV减毒疫苗免疫在我国养禽业几乎是所有新生雏鸡必不可少的免疫程序,且基因型与目前我国的流行株相一致,理所当然获救4株VII型新城疫病毒L基因突变的减毒疫苗株成为最具吸引力的疫苗候选者。

2.一种VII 型新城疫病毒L 基因突变的减毒疫苗株的制备方法,其特征在于:构建了覆盖整个基因组的五个cDNA 克隆片段,利用各片段间重叠部分的酶切位点,在转录载体质粒pBR322 连接组装获得了15192nt 的完整cDNA 克隆;在全长cDNA片段的5’末端前添加T7RNA聚合酶启动子,在全长cDNA片段后加入具有自我催化功能的丁肝病毒核酶(RiB)序列和T7转录终止信号,构建好的质粒命名为prNDV-97;通过PCR方法将全长基因组cDNA中的L蛋白编码区第14056位碱基由A同义突变为G,消除14056bp处的Hind Ⅲ酶切位点,并作为拯救病毒的分子标记,构建完成的质粒命名为pmNDV-97;同时在T7RNA聚合酶启动子与全长基因组cDNA的5’末端之间引入两个多余的G,这有助于副粘病毒反向遗传操作的病毒拯救;

1)NDV Ⅶ-97株基因组全长cDNA的构建;
2)NDV Ⅶ-97 L 蛋白结构域Ⅵ内氨基酸定点突变和突变体全长cDNA的连接;
3)VII 型新城疫病毒L 基因突变的减毒疫苗株E1954Q的拯救;
将质粒pE1954Q与pBSK-NP、pBSK-P 和pBSK-L三个真核表达质粒共转染BHK-21 细胞,转染72h 后将细胞上清接种9-11 日龄SPF 鸡胚,每份样品接种3 枚,0.2mL/ 枚,成功救获NDV Ⅶ -97 L 基因突变减毒株E1954Q。
