Description

一种肝癌基因NDC80及其应用
技术领域
本发明涉及生物基因工程技术领域,具体涉及一种肝癌基因NDC80及其应用。
背景技术
原发性肝癌(HCC)是常见的恶性肿瘤,其发生率和死亡率分别占全球恶性肿瘤的第5位和第3位1,占我国恶性肿瘤的第3位和第2位,在部分地区和农村其死亡率已经上升到第一位。在全球范围内其发病率逐年上升,每年约新增加60余万病例,其中50%以上病例发生在中国。肝炎病毒感染是肝癌的主要危险因素,高达80%的肝癌归因于乙肝病毒或丙型肝炎病毒感染。在我国,则主要是乙肝病毒感染为主,90%肝癌患者有HBV感染或者HbsAg阳性。但是,目前对于乙肝病毒导致肝细胞癌变的具体机制并不清楚,因此,寻找并确定一种表达肝癌的基因,对于研究肝细胞癌变的具体机制、制定预防或治疗肝癌的方法有重要意义。
发明内容
本发明的目的在于提供一种肝癌基因。
本发明的另一个目的是提供一种肝癌基因的应用。
为了实现上述目的,本发明是通过以下方案予以实现的:
一种肝癌基因NDC80,其核苷酸序列如SEQ ID NO:1所示。该基因的登录号为NC_000018.10。
发明人研究发现乙肝病毒感染肝细胞后,导致NDC80上调,上调后NDC80进一步使乙肝病毒继续扩增,从而形成正反馈,最终,高表达的NDC80促使肝细胞增殖癌变,引起肝癌发生。因此,本发明首次确定NDC80是乙肝病毒感染导致肝癌发生的关键因子。
在确定NDC80基因为乙肝病毒感染导致肝癌发生的关键因子后,本发明同时保护NDC80基因在作为预防或治疗乙肝病毒性肝癌的基因靶点中的应用。
进一步地,本发明还要求保护一种防或治疗乙肝病毒性肝癌的制剂,该试剂含有定量的抑制NDC80基因的抑制剂。
与现有技术相比,本发明具有以下有益效果:
本发明通过研究首次发现NDC80是乙肝病毒感染导致肝癌发生的关键因子。首先,乙肝病毒感染肝细胞后,导致NDC80上调,上调后NDC80进一步使乙肝病毒继续扩增,从而形成正反馈,最终,高表达的NDC80促使肝细胞增殖癌变,引起肝癌发生。该基因有助于进一步了解癌症机制,为肝癌的防治提供新思路及策略。

下面结合说明书附图和具体实施例进一步说明本发明的内容,但不应理解为对本发明的限制。在不背离本发明精神和实质的情况下,对本发明方法、步骤或条件所作的简单修改或替换,均属于本发明的范围,若未特别指明,实施例中所用的技术手段为本领域技术人员所熟知的常规手段。
实施例中所述HBV阴性肝癌是指非乙肝病毒性肝癌,HBV阳性肝癌是指乙肝病毒性肝癌。
实施例1
一、免疫组织化学染色:分别对正常肝组织、HBV阴性肝癌组织、HBV阳性肝癌组织进行染色观察,详细步骤如下:
1.标本来源:分别从正常肝组织、HBV阴性肝癌组织、HBV阳性肝癌组织处收集得到用石蜡包埋的组织块,在显微镜下用薄片切片机将所有的组织块切成4mm的薄片,制成玻片;
2.脱蜡、水化:组织薄片在二甲苯中脱蜡后,再依据标准操作步骤利用分级的酒精(无水、95%、70%)将其水化;
3.抗原修复:将组织浸泡在EDTA(pH=8.0)中,在121℃下高压蒸汽处理5min,再用PBS冲洗3次,每次5min;
4.灭活酶:将经PBS冲洗后的组织放入3%的过氧化氢中,在室温下孵育15min,以阻断内源性过氧化物酶的活性;
5.抗原-抗体反应:在每张组织薄片上滴加一抗(以含有0.5%的BSA的PBS为稀释剂,NDC80抗体(sc-135934)按照1:100稀释),在4℃下孵育过夜,然后用缓冲液冲洗,除去游离抗体;
6.滴加二抗试剂(生物素化二抗工作液),加PBS反应30min;
7.使用试剂盒显色;
8.与苏木精进行对比染色,再依次序在酒精和二甲苯中脱水;
9.以PBS代替一抗作为阴性对照。
染色结果见图1中的A图,由免疫组织化学染色结果可知:正常肝组织很少表达NDC80蛋白,HBV阴性肝癌组织表达中等,HBV阳性肝癌组织表达最多。
二、Western Blot分析
利用商品化的试剂盒提取正常肝组织,HBV阴性肝癌组织和HBV阳性肝癌组织的蛋白,提取步骤按照试剂盒说明操作,粗略步骤如下:
1.将正常肝组织,HBV阴性肝癌组织和HBV阳性肝癌组织剪碎后置入裂解液中进行富集(HBV阳性肝癌组织设为实验组,HBV阴性肝癌组织,正常肝组织设为对照组,),加入0.5mM 原钒酸钠、1mM PMSF、1mg/ml抑肽酶、2ug/ml亮抑肽酶;
2.将裂解物于4℃下14000rpm离心10min;
3.将从癌症或非癌症组织中匀浆提取的组织样品加入到缓冲液中,加入0.2mMPMSF、1ug/ml抑肽酶、1ug/ml亮抑肽酶;
4. BCA试剂盒测定步骤3得到的各种上清液的蛋白浓度,并加入SDS-PAGE凝胶电泳进行western blot实验。
western blot检测结果见图1中的B图,由western blot检测结果可知:正常肝组织很少表达NDC80蛋白,HBV阴性肝癌组织表达中等,HBV阳性肝癌组织表达最多。
三、qRT-PCR定量分析
利用商品化的RNA提取试剂盒分别提取正常肝组织、HBV阴性肝癌组织、HBV阳性肝癌组织的RNA,然后用商品化的RT-PCR试剂盒定量分析各种组织的RNA,检测RNA含量所用PCR引物如下:

RT-PCR的操作步骤按照RT-PCR试剂盒操作进行,检测结果见见图1中的C图,由RT-PCR检测结果可知:正常肝组织很少表达NDC80 mRNA,HBV阴性肝癌组织表达中等,HBV阳性肝癌组织表达最多。
由实施例1中的3项检测结果可知:相对正常肝组织,NDC80基因在原发性肝癌(HBV-)中表达上调,在原发性肝癌(HBV+)上调更加明显。
实施例2
本实施例用到HepG2.2.15细胞和shNDC80细胞共2种细胞,实验组为转染了siRNA的HepG2.2.1.5细胞(简称shNDC80细胞),对照组为HepG2.2.1.5细胞。
一、乙肝病毒DNA和cccDNA的检测
1、乙肝病毒DNA的检测:分别收集培养96h的HepG2.2.15细胞和shNDC80细胞以及其培养上清液;利用商品化的DNA提取试剂盒分别提取2种细胞的DNA,用RT-PCR检测2种HBV感染细胞的DNA。PCR 检测引物如下:
HBV-DAN-F:5’-ATGGAGAACACAACATCAGG-3’,
HBV-DNA-R:5’-GAGGCATAGCAGCAGGATG-3’;
GAPDH-F:5’-GAGTCAACGGATTTGGTCGT-3’,
GAPDH-R-R: 5’-GACAAGCTTCCCGTTCTCAG-3’。
检测结果见图2中的A图。
2、乙肝病毒cccDNA的检测
(1)用PBS冲洗两种细胞,4℃条件下,将两种细胞分别在裂解液(50mMTris,100MmNaCl,0.1%Trion X-100,5mM MgCl2,pH8.0)中裂解10min;
(2)在4℃下,离心2min,将细胞核从270g细胞质中分离;
(3)附加型DNA按照商品化的试剂盒操作从细胞核中提取;
(4)没有链酶蛋白酶的条件下,细胞在裂解液中裂解,另加0.25ml2.5M的KCl沉淀得到蛋白质-洗涤液复合物;
(5)摇晃以混合溶菌产物,离心除去细胞DNA;
(6)上清液中的病毒性cccDNA从等体积的苯酚中提取;
(7)HBVcccDNA用一种新的方法进一步净化。(该方法通过RT-PCR对HBVcccDNA进行定量)
(8)净化后的HBVcccDNA通过RT-PCR检测,PCR引物为:
HBV-cccDNA-F:5’-GTCTGTGCCTTCTCATCTGCC-3’,
HBV-cccDNA-R:5’-ACAGCTTGGAGGCTTGAACAG-3’
之后,按照RT-PCR试剂盒操作进行目标RNA的定量检测。
乙肝病毒的DNA和cccDNA的检测结果见图2中的A图,由图A结果分析可知,在HepG2.2.15 shNDC80细胞系中,干扰NDC80使RNA水平处于低表达。
二、HBsAg 、HBcAg的检测:通过免疫组织化学染色实验间接分析HepG2.2.1.5细胞和shNDC80细胞的HBsAg和HBcAg。具体步骤如下:
1.在4℃下,用4%多聚甲醛将转染细胞固定20min,在Triton X-100中通透;
2.在37℃下,用含有10%山羊血清的PBS孵化细胞;
3.在4℃下,用含有HBsAg(BM0064)或HBcAg(BM0060)的特定的鼠类单克隆抗体孵化细胞过夜;
4.滴加二抗试剂(生物素化二抗工作液),加PCS30min;
5.使用试剂盒显色;
6.与苏木精进行对比染色,再依次序在酒精和二甲苯中脱水;
7.以PBS代替一抗作为阴性对照。
染色结果见图2中的B图。
实施例3
一、细胞生长曲线
1.将HepG2.2.15细胞和shNDC80细胞分别用0.25%的胰蛋白酶消化,制成单细胞悬液,转染了siRNA的HepG2.2.1.5细胞为实验组,未转染siRNA的HepG2.2.1.5为对照组;
2.用Freshney计数板对存活细胞计数:细胞浓度调整至1x105/ml,即1ml细胞中大约有1x105个细胞,置于12孔板中的3个孔内;
3.分别于24h、48h和72h后对存活细胞进行计数,并用平均值绘制细胞生长曲线。
结果见图3中的A图。
二、细胞活性测定
用MTS含量测定不同时间点的细胞活性:在96孔托盘中,通过MTS含量定量测定细胞活性。96孔托盘中,细胞密度为1 x 103。培养24小时后,加入测试药物(测试药物为MTT,全称为 3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐,商品名:噻唑蓝),在37℃下,将培养液孵化不同的时间(0h,24h,48h,72h)。被治疗的细胞以1:10的比例浸在MTS试剂中,在培养基中培养4h。每个孔的吸收值用酶标仪测定,吸收波长设定为490nm。每个样品测定3次,做三次平行实验,并绘制扩散曲线。
测定结果见图3中的B图,从图B中可以看出,在shNDC80细胞系中,肝癌细胞增殖的数量很少,活性很低。
由实施例1-3的结果可知,NDC80是乙肝病毒感染导致肝癌发生的关键因子,该基因有助于进一步了解癌症机制,为肝癌的防治提供新思路及策略。
