Abstract
一种具有抗电离辐射作用的复方柿单宁芦荟胶,质量%计,其配方为:脱色芦荟凝胶冻干粉1.6‑3.0%;柿子粉0.5‑5%;金银花提取物 0.5‑1.0%;枸杞提取物 0.5‑1.0%;茶多酚 0.2‑0.55;甘油 2.0‑5.0%;1,3‑丁二醇1.0‑15.0%;高分子透明质酸钠0.0‑1.0%;尿囊素 0.0‑0.4%;卡波姆 0.10%‑2.00%;香精 0.0‑0.2%;防腐剂 0.0‑0.2%;碱液0.1‑8.0%;去离子水 50.0‑90.0%。每100g复方柿单宁芦荟胶产品中,柿单宁含量为150‑250mg,O‑乙酰基大于300mg。将该产品做抗电离辐射实验,采用CCK‑8法检测细胞活性,选用293T细胞作为研究对象,在辐射总剂量为8Gy,辐射后培养12h,复方柿单宁芦荟胶中柿单宁的浓度在2mg/ml时具有最明显的抗电离辐射作用。
Description
技术领域
本发明涉及化妆品领域,尤其是一种具有抗电离辐射作用的外用凝胶产品。
背景技术
涩柿果又名朱果,是一种营养价值很高的水果,享有“果中圣品”的美誉。涩柿果中含有2.0%左右涩柿单宁,还含有糖类、蛋白质、人体必须的氨基酸、微量元素、多种维生素、有机酸等,具有抑制人淋巴细胞白血病、降血压、清除体内自由基、吸附稀有金属元素等多种功效。发明专利申请CN101328194公布了一种柿子粉及其生产工艺;它是将柿子榨汁以后,汁液进行酶解、过滤、灭酶、浓缩、真空冷冻干燥而制得柿单宁含量为1%~99%的粉状物。
枸杞为茄科植物宁夏枸杞Lycium barbarum L.的果实。是我国传统名贵中药材,气微、味甜、微酸。主要成分为枸杞多糖、甜菜碱、玉蜀黍黄素、胡萝卜素及氨基酸等。具有抗衰老、抗突变、抗肿瘤、降血脂、保肝、降血糖、降血压、免疫调节等作用,对造血功能有促进作用。市售枸杞提取物中枸杞多糖的含量为50%左右。
金银花,又名忍冬,其功效主要是清热解毒,主治温病发热、热毒血痢、痈疽疔毒等。现代研究证明,金银花含有绿原酸、木犀草素苷等药理活性成分,对溶血性链球菌、金黄葡萄球菌等多种致病菌及上呼吸道感染致病病毒等有较强的抑制力,另外还可增强免疫力、护肝、抗肿瘤、消炎、解热等。
茶多酚是从茶叶中提取的全天然抗氧化食品,是指茶叶中一大类组成复杂、分子量及其结构差异很大的多酚类及其衍生物混合物,主要由儿茶素、黄酮醇、花色素、酚酸及其缩酚酸等组成的有机化合物,以儿茶素为主。茶多酚能极强的清除有害自由基,阻断脂质过氧化过程,提高人体内酶的活性,从而起到抗突变、抗癌症的功效,对胃癌、肠癌等多种癌症的预防和辅助治疗均有益处。
芦荟含有蒽醌类化合物、芦荟多糖、氨基酸、有机酸、维生素、矿物质等功效成分,可用于食品、药品、化妆品,具有提高免疫力、保湿、美容等保健功效。行业标准QB/T2488-2006规定了脱色芦荟凝胶冻干粉的生产工艺及其质量指标,其中规定了芦荟多糖含量,以O-乙酰基含量计,保留了芦荟的活性成分。公开号为CN103622864A,名称为多功能芦荟胶及其制备方法的发明专利,公开了一种多功能芦荟胶及其制备方法;含有芦荟冻干粉、植物提取物、复配透明质酸钠、尿囊素、D-泛醇、薄荷脑、甜菜碱、β-葡聚糖等有效活性成分,产品具有良好的保湿、镇静、抗衰老、消炎抗菌、缓解刺激、紧致肌肤、促进破损皮肤愈合等功效。
电磁辐射是一种重要的能源污染,是由直接或间接电离粒子或二者混合组成的辐射。电离辐射是一切能引起物质电离的辐射总称,其种类很多,主要是波长短、频率高、能量高的射线。电磁辐射现象不仅会出现在人们关注的手机、寻呼机等移动通讯器材上,还包括广播电视、通讯、医疗、交通、电力输送等。例如,广播电视发射塔、家用微波炉、可调光台灯等都会放出电磁波,只是功率有大有小。电离辐射可引起放射病,对机体的损伤可分为急性放射损伤和慢性放射性损伤。短时间内接受大剂量的照射,可引起机体的急性损伤,平时见于核事故和放射治疗病人。而较长时间内分散接受一定剂量的照射,可引起慢性放射性损伤,如皮肤损伤、造血障碍,白细胞减少、生育力受损等。另外,辐射还可以致癌和引起胎儿的死亡和畸形。开发具有抗电离辐射的外用产品是大势所趋。
发明内容
本发明所要解决的技术问题是提供一种具有抗电离辐射作用的复方柿单宁芦荟胶产品。
为了解决这个技术问题,具有抗电离辐射作用的复方柿单宁芦荟胶,按质量百分比,包括以下各组分:
脱色芦荟凝胶冻干粉 1.6-3.0%;
柿子粉 0.5-5.0%;
金银花提取物 0.5-1.0%;
枸杞提取物 0.5-1.0%;
茶多酚 0.2-0.5%;
甘油 2.0-5.0%;
1,3-丁二醇 1.0-15.0%;
高分子透明质酸钠 0.0-1.0%;
尿囊素 0.0-0.4%;
卡波姆 0.10-2.0%;
香精 0.0-0.2%;
防腐剂 0.0-0.2%;
碱液 0.1-8.0%;
去离子水 50.0-90.0%。
所述柿子粉中柿单宁含量为2.0-10%;
所述金银花提取物、枸杞提取物和茶多酚是在市场上购买的;
所述脱色芦荟凝胶冻干粉是满足轻工行业标准QB/T2488-2006规定;
所述防腐剂是甲基异噻唑啉酮及碘代丙炔基氨甲酸丁酯中的一种或两种混合;
所述碱液是浓度为5%的氢氧化钾或氢氧化钠溶液中的一种。
每100g产品中,柿单宁含量为150-250mg,O-乙酰基大于300mg。
具有抗电离辐射作用的复方柿单宁芦荟胶的制备方法,包括以下步骤:
A 按质量百分比称取各组分,备用;
B 将脱色芦荟凝胶冻干粉、柿子粉、金银花提取物、枸杞提取物、茶多酚可以先用适量的水溶解,待用;
C 去离子水、高分子透明质酸钠、尿囊素、卡波姆分别投入乳化锅中,抽真空,以30转/分-50转/分混合搅拌,加热至70℃温度后,控制加热,继续搅拌直至完全溶解后,冷却至温度50℃,加入B步骤所述提取物的溶液,抽真空,30转/分搅拌,继续冷却到温度下降到45℃以下;
D 将甘油、1,3-丁二醇、防腐剂、香精混合均匀并溶解后投入乳化锅,搅拌5分钟,直至完全溶解并混合均匀;
E 加入浓度为5%的碱液,搅拌冷却至40℃以下;
F 出料。
该凝胶产品是一种性质温和、使用安全、具有优越的保湿性能、既能发挥柿单宁和芦荟自身的护肤效果、又能与其它多种护肤产品搭配使用发挥多种护肤美容效果的凝胶产品。使用时,涂抹在脸上或者相应的皮肤表面,并轻柔片刻。
将该产品做抗电离辐射实验,采用CCK-8法检测细胞活性。
由于293T对X射线的敏感性高,选用293T细胞作为研究对象,将复方柿单宁芦荟胶作为受试药物,通过细胞形态观察、CCK-8检测细胞活力、Hoechst33258染色检测细胞凋亡、流式细胞仪检测细胞凋亡来验证复方柿单宁芦荟胶对辐射损伤的保护作用。实验表明,随着辐射剂量的增大,细胞的活性逐渐降低。在辐射总剂量为8Gy,辐射后培养12h,在辐射前添加不同浓度的复方柿单宁芦荟胶对细胞都具有一定的防护作用,柿单宁浓度在2mg/ml时具有最明显的防护作用。
本发明具有下列优点和特性:
1、品质好。以柿子粉中的柿单宁为主要活性成分,采用柿子粉,枸杞提取物、脱色凝胶芦荟冷冻干燥粉,金银花提取物,茶多酚进行复方,制成外用凝胶,具有明显的抗辐射作用。尤其在辐射总剂量为8Gy时,控制产品中功效成分含量为:每100g产品中,柿单宁含量为150-250mg,O-乙酰基大于300mg时,具有良好的抗电离辐射效果。按照配方复配,浓缩了各种活性物质精华,品质好。
2、多种组分同时使用,既能充分发挥每个组分的独特效果,又能保证整体产品的最佳抗电离辐射效果,共同作用凸显柿单宁和芦荟的抗电离辐射作用,并且副作用低。
具体实施方式
以下所述实例详细地说明了本发明。在实例中,除另有说明外,所有份数和百分比均按质量计。
实施例1
1、按照配方,将符合要求的各种提取物原料准备好,备用。
2、按10kg具有抗电离辐射作用的复方柿单宁芦荟胶成品称取各种成分:
脱色芦荟凝胶冻干粉200g;柿子粉400g;金银花提取物50g;枸杞提取物50g;茶多酚20g;甘油500g,1,3-丁二醇1000g;高分子透明质酸钠50g;尿囊素20g;卡波姆150g;香精100g;碘代丙炔基氨甲酸丁酯防腐剂100g;5%氢氧化钾溶液500g;去离子水6860g;备用;
3、首先将高分子透明质酸钠、去离子水、尿囊素、卡波姆分别投入乳化锅中,抽真空,以30转/分-50转/分,混合搅拌,同时加热至70℃后,控制加热,继续搅拌直至完全溶解后,冷却至温度50℃,加入步骤2所述的各种植物提取物,抽真空,以30转/分搅拌,继续冷却到温度下降到45℃以下;再将甘油、1,3-丁二醇、防腐剂、香精混合均匀并溶解后投入乳化锅,搅拌5分钟,直至完全溶解并混合均匀;然后加入浓度为5%的碱液,搅拌冷却至40℃以下;出料。即得具有抗电离辐射作用的复方柿单宁芦荟胶。经检测,每100g产品中,柿单宁含量为205g,O-乙酰基含量为420mg。
抗电离辐射实验。采用CCK-8法检测细胞活性;
(1)选取对数生长期的293T细胞,制成1×104个/ml的单细胞悬液,然后将细胞接种到96孔板中,每个孔加入100μl含10%新生牛血清的DMEM新鲜培养液,96孔板的边缘用无菌的PBS填充,然后置于37℃、CO2含量为5%恒温密闭的细胞培养箱中培养至细胞单层铺满孔底。
(2)细胞培养24h至贴壁后,倒去旧的培养液,然后加入100μl含10%新生牛血清的DMEM新鲜培养液,培养12h,使293T细胞同步化。
(3)待细胞同步化后,弃旧培养液。每孔加入90μl含l0%新生牛血清的DMEM培养液和10μl药液。药物组按每孔柿单宁浓度分设6个浓度亚组(0mg/ml、0.5mg/ml、1mg/ml、2mg/ml、4mg/ml、8mg/ml),同时设置空白组,空白组为无细胞的培养基,每亚组设4个平行,培养24小时后,293T细胞接受不同剂量的X射线(4GY、8GY、12GY、16GY、20GY)照射。同时设置不接收X射线照射的对照组。
(4)辐射后,分别于12h、24后,将96孔培养板中的旧液弃除,用无菌的PBS溶液洗涤3次,然后每孔分别加入90μl新鲜的DMEM培养基及10μl CCK-8液继续培养4h。
(5)4h后将96孔细胞培养板置于多功能酶标仪下(490nm,1S)测定各孔的吸光度(OD)值。
(6)计算细胞活力值,结果见表1、表2。

可见,在辐射总剂量为8Gy,辐射后培养12h,在辐射前添加不同浓度的柿单宁芦荟胶对293T细胞都具有一定的防护作用,柿单宁的浓度在2mg/ml时具有最明显的抗电离辐射作用。
实施例2
1、按照配方,将符合要求的各种提取物原料准备好,备用。
2、按10kg复方柿单宁芦荟胶成品计,称取各种成分:
脱色芦荟凝胶冻干粉300g;柿子粉500g;金银花提取物100g;枸杞提取物50g;茶多酚30g;甘油200g,1,3-丁二醇1500g;高分子透明质酸钠100g;尿囊素40g;卡波姆100g;香精50g;甲基异噻唑啉酮防腐剂100g;5%氢氧化钠溶液800g;去离子水6030g;将脱色芦荟凝胶冻干粉300g;柿子粉500g;金银花提取物100g;枸杞提取物50g;茶多酚30g先用2000g去离子水溶解;待用;
3、首先将高分子透明质酸钠、剩余去离子水、尿囊素、卡波姆分别投入乳化锅中,抽真空,以30转/分-50转/分,混合搅拌,同时加热至70℃温度后,控制加热,继续搅拌直至完全溶解后;冷却至温度50℃;加入步骤2所述的各种植物提取物,抽真空,以30转/分搅拌,继续冷却到温度下降到45℃以下;再将甘油、1,3-丁二醇、防腐剂、香精混合均匀并溶解后投入乳化锅,搅拌5分钟,直至完全溶解并混合均匀;然后加入浓度为5%的碱液,搅拌冷却至40℃以下;出料。即得具有抗电离辐射作用的复方柿单宁芦荟胶。经检测,每100g产品中,柿单宁含量为245g,O-乙酰基含量为620mg。
实施例3
1、按照配方,将符合要求的各种提取物原料准备好,备用。
2、按10kg复方柿单宁芦荟胶成品计,称取各种成分:
脱色芦荟凝胶冻干粉160g;柿子粉50g;金银花提取物50g;枸杞提取物100g;茶多酚30g;甘油500g,1,3-丁二醇850g;卡波姆100g;甲基异噻唑啉酮防腐剂50g;代丙炔基氨甲酸丁酯防腐剂50g;5%氢氧化钾溶液500g;去离子水6030g;将脱色芦荟凝胶冻干粉、柿子粉、金银花提取物、枸杞提取物、茶多酚先用2000g去离子水溶解;待用;
3、首先将剩余去离子水、卡波姆分别投入乳化锅中,抽真空,以30转/分-50转/分,混合搅拌,同时加热至70℃温度后,控制加热,继续搅拌直至完全溶解后;冷却至温度50℃;加入步骤2所述的各种植物提取物,抽真空,以30转/分搅拌,继续冷却到温度下降到45℃以下;再将甘油、1,3-丁二醇、防腐剂混合均匀并溶解后投入乳化锅,搅拌5分钟,直至完全溶解并混合均匀;然后加入浓度为5%的碱液,搅拌冷却至40℃以下;出料。即得具有抗电离辐射作用的复方柿单宁芦荟胶。经检测,每100g产品中,柿单宁含量为152mg;O-乙酰基含量为365mg。

脱色芦荟凝胶冻干粉1.6-3.0%;
柿子粉0.5-5%;
金银花提取物 0.5-1.0%;
枸杞提取物 0.5-1.0%;
茶多酚 0.2-0.5%;
甘油 2.0-5.0%;
1,3-丁二醇1.0-15.0%;
高分子透明质酸钠0.0-1.0%;
尿囊素 0.0-0.4%;
卡波姆 0.1%-2.0%;
香精 0.0-0.2%;
防腐剂 0.0-0.2%;
碱液0.1-8.0%;
去离子水 50.0-90.0%;
每100g复方柿单宁芦荟胶产品中,柿单宁含量为150-250mg,O-乙酰基大于300mg。
