Abstract
本发明公开了一种防治肝病的中药制剂及其生产方法,它包括以下重量份数的原料:田基黄10-12、垂盆草8-10、积雪草6-8、郁金5-7、女贞子8-10、山楂14-16、蒲公英15-17、猫爪草6-8,其生产方法是将中药经过回流煎煮提浓缩生产而成。该中药制剂参考古今中医学药理研究成就,基于中医学对肝病的认识及治疗原则,具有毒副作用小、不产生耐药性,疏肝理气,健脾消食,用于脂肪肝、酒精肝、药物性肝损伤、慢性肝炎及早期肝硬化等,具有解酒、降脂、解毒、抗炎等效果。
Description
技术领域
本发明涉及中药技术领域,尤其涉及一种防治肝病的中药制剂及其生产方法。
背景技术
肝(liver),人体脏器名,五脏之一。是脊椎动物身体内以代谢功能为主的一个器官,并在身体里面充分扮演着去氧化,储存肝糖,分泌性蛋白质的合成等等。肝脏也制造消化系统中之胆汁。在医学用字上,常以拉丁语字首hepato- 或 hepatic来描述肝脏或肝脏的。大部分的肝脏疾病都会有黄疸的症状,这是由于肝脏无法继续将胆红素排出所以就在体内累积。中医认为:肝与胆相为表里,开窍于目,肝主藏血,肝主疏泄,有贮藏和调节血液的功能。《素问·五脏生成》:“肝之合筋也,其荣爪也。”肝又为将军之官,主谋虑。
肝脏是人体内最大的消化腺,是体内物质能量代谢的中心站。据估计,在肝脏中发生的化学反应有500种以上。首先它分泌胆汁,帮助消化饮食;它把吸收的氨基酸合成蛋白质供给我们机体能量,让我们能够精力充沛地完成一天的工作;它能贮藏和燃烧体内的脂肪,控制我们的体形;它是脂溶性维生素的贮存器官;它还能够氧化、还原、分解体内的毒素,吞噬不小心吃入体内的细菌,是人体最大的解毒器官。实验证明,动物在完全摘除肝脏后即使给予相应的治疗,最多也只能生存50多个小时,这说明肝脏是维持生命活动的一个必不可少的重要器官。
常见的肝病有肝炎、肝硬化、肝脓肿、原发性肝癌等,肝炎主要以慢性肝炎为主,按病因学分为慢性病毒性肝炎、自身免疫性肝炎、药物毒性肝炎、遗传性疾病以及其它原因不明的慢性肝炎。而病毒性肝炎以乙型肝炎最为常见,乙肝病毒是一种世界性传染疾病,据统计全球携带乙肝表面抗原的人数超过2.8亿,中国是乙肝的高发区,目前有现症的慢性乙型肝炎约3000万人,每年国内死于乙肝后肝硬化者有40万人。
肝病症状
1、消化道表现:这是最常见的肝病症状,大多数肝病都会出现,比如:恶心、厌油腻、食欲差、全身乏力等,可出现呕吐,腹泻,脾肿大等症状。此类症状也可能与慢性肝病引起的肝原性溃疡病,门静脉高压性肠病等有关。
2、肝区不适:在所有肝病症状中,肝区不适和肝区疼痛较具有特异性,出现此类症状时首先怀疑是肝病引起的,应排除外伤因素。偶尔正常人也会出现暂时性肝区疼痛不适,比较少见。肝区不适和肝区疼痛往往与肝肿大压迫肝包膜有关,随着病情的转归,肝肿大的加重或减轻,肝区疼痛的性质和程度也不相同。肝癌一般是进行性加重,主要是肝癌肿瘤不断增大压迫肝包膜所致。
3、全身表现:身体乏力、容易疲劳是最常见的全身表现。部分肝病患者可伴有不同程度黄疸,表现为尿黄、眼睛黄和皮肤黄,是最具有特异性的肝病症状(小儿生理性黄疸除外)。黄疸过高时出现皮肤瘙痒。
4、肝掌蜘蛛痣:很多慢性肝病会出现肝掌、蜘蛛痣、肝病面容,尤其肝硬化病人比较多见。但是肝掌和蜘蛛痣没有特异性,在正常人中同样可以见到,因此不能人有蜘蛛痣或肝掌就说是肝病症状。
5、肝腹水:肝腹水一般在肝病晚期或病情极为严重时才会出现,如肝硬化出现肝腹水表示已经进入肝硬化晚期。
6、出血倾向:肝病出血现象体现在肝功能减退,使凝血因子合成减少所致。容易引起肝病患者牙龈出血、痔疮出血、胃肠道出血等,且出血时难以止住。
7、门静脉高压:门脉高压是指门静脉系统压力升高。常引起食管胃底静脉曲张,是造成消化道出血的主要原因。一旦发生出血,结合出血倾向病情往往比较凶险,是肝病(主要指肝硬化、肝癌晚期)死亡的主要原因之一。
8、肝性脑病:这是肝病发展到终末期的表现,症状极为凶险,是造成肝病死亡最主要的原因。
肝病的早期症状
面色晦暗:与太阳晒黑的皮肤不同,该情况面部暗淡而无光泽度;另外严重的黑眼圈都是慢性肝病患者早期症状,其中大多数为慢性乙肝。
皮肤发黄:从未患过肝炎的人,肝病患者早期症状在畏寒、发热、恶心、呕吐、肝痛、极度乏力后,忽然出现眼睛和皮肤发黄,则表明患了急性黄疸型肝炎;慢性肝炎患者若出现黄疸,表明病情加重。
肝掌:与常人的手掌颜色大不相同,普通人的手掌颜色红润,而患有肝病的病人手掌心泛白无血色。
蜘蛛痣:中间有一红点,周围有血丝状,类似于蚊虫叮咬后的症状,当用细棒一端压迫痣中心时、全痣消失,放开后又会出现,这一肝病患者早期症状可与其他血管痣相鉴别。据介绍,男性体表有蜘蛛痣者,85%的人可有不同程度的肝脏组织病变,其中约30%为肝硬化。
我国肝病人群众多,肝病诊断一直处于20年前的水准,治疗上也一直处于“千人一药、万人一方”的状态,而随时增加的用于各类肝病防治的高额治疗费用也越来越将成为患者及社会巨大经济负担之一。而据肝病医生估计,这其中,有一半人员属于过度治疗与盲目治疗。过度与盲目治疗都将使得社会和病人蒙受巨大经济损失,也将加剧病情的恶化及复杂化。有资料显示,药物性肝损害的病人正在逐年迅速增长,其中盲目治疗难逃其咎,市售制剂由于组方和工艺原因,疗效不甚理想,存在没有针对性、疗效低等不足。
发明内容
基于目前存在上述存在的问题,本发明提供了一种防治肝病的中药制剂及其生产方法,本发明的防治肝病的中药制剂,具有毒副作用小、不产生耐药性,疏肝理气,健脾消食,用于脂肪肝、酒精肝、药物性肝损伤、慢性肝炎及早期肝硬化等,具有解酒、降脂、解毒、抗炎等效果。
本发明是采用以下技术方案实现的:
一种保肝护肝片,它包括以下重量份数的原料:田基黄10-12、垂盆草8-10、积雪草6-8、郁金5-7、女贞子8-10、山楂14-16、蒲公英15-17、猫爪草6-8。
优选地,各中药原料的重量份数为:田基黄10、垂盆草8、积雪草6、郁金5、女贞子8、山楂14、蒲公英15、猫爪草6。

所述防治肝病中药制剂的生产方法,包含了以下步骤:
步骤1)按重量份数称取田基黄、垂盆草、积雪草、郁金、女贞子、山楂、蒲公英、猫爪草分别粉碎,混合在一起;
步骤2)加8倍重量的纯化水,回流煎煮2小时,过滤得滤液a;在滤渣中加入4倍重量的纯化水,再回流煎煮2小时,过滤得滤液b;将滤液a和b混合,进行减压浓缩,使总体积缩减为原来的1/4,即为中药水提浓缩液,备用;
步骤3)将中药水提浓缩液用纯化水稀释至处方量1g/mL,即得成品,置于4℃环境保存备用。
本发明选取的中药原料中,各药有效成份分析:
田基黄为藤黄科金丝桃属植物田基黄Hypericum japonicum Thunb. 的干燥全草。一年生纤细小草本,高10~30厘米;茎常有四棱,无毛。叶对生,无柄,基部稍抱茎;叶片卵形或卵状披针形,长7~10毫米,宽1.5~6毫米,顶端钝,基部近心形,全缘,两面均无毛,叶背有稀疏的小黑点,有基出脉5条。花期几全年;聚伞花序生于小枝顶端,疏生;花黄色,直径约6毫米,花梗纤细,长5~10毫米;萼5片,线状长椭圆形或椭圆形,长3~5毫米;花瓣5片,倒卵状长椭圆形,约与花萼等长;雄蕊多数,花丝基部合生;子房卵形至椭圆形,长约2毫米,花柱3枚,柱头头状。蒴果椭圆形,长约 4毫米,成熟时开裂。生态环境:生于田野较湿润处。资源分布:广布于长江流域及其以南各地。性状鉴别:全草长10-40cm。根须状,黄褐色。茎单一或基部分枝,光滑,具4棱,表面黄绿色或黄棕色;质脆,易折断断面中空。叶对生,无柄;完整叶片卵形或卵圆形,全缘,具细小透明腺点,基出脉3-5条。聚伞花序顶生,花小橙黄色。气无,味微苦。以色黄绿,带花者为佳。性味:甘、微苦,微寒。归经:归肝、脾经。功能与主治:清热解毒,利湿退黄,消肿散瘀。用于湿热黄疸,肠痈,目赤肿痛,热毒疮肿;近有用于急慢性肝炎、早期肝硬化、肝区疼痛,阑尾炎,乳腺炎,肺脓肿。
垂盆草(拉丁学名:Sedum sarmentosum Bunge),别名:狗牙瓣、石头菜、佛甲草,为景天科植物垂盆草Sedum sarmentosum Bunge 的新鲜或干燥全草。茎匍匐,易生根,不育枝及花茎细,匍匐而节上生根,直到花序之下,长10-25厘米。3叶轮生,叶倒披针形至长圆形,长15-28毫米,宽3-7毫米。垂盆草一般生长在山坡岩石石隙、山沟边、河边湿润处,极易栽培,对环境要求不严,家前屋后均可种植,也可盆栽,通常采用分株繁殖。夏、秋二季采收,除去杂质。鲜用或干燥。性味:甘、淡,凉。归经: 归肝、胆、小肠经。功能主治: 清利湿热,解毒。用于湿热黄疸,小便不利,痈肿疮疡,急、慢性肝炎。
积雪草为伞形科植物积雪草Centella asiatica (L.)Urb. 的干燥全草,为多年生匍匐草本。别名:十八缺、 崩大碗、马蹄草、雷公根、蚶壳草、铜钱草、落得打。生于阴湿荒地、村旁、路边、水沟边。常卷缩成团状。根圆柱形,长2~4cm,直径1~1.5mm,表面浅黄色或灰黄色。茎细长弯曲,黄棕色,有细纵皱纹,节上常着生须状根。叶片多皱缩、破碎,完整者展平后呈近圆形或肾形,直径1~4cm,灰绿色,边缘有粗钝齿;叶柄长3~6cm,扭曲。伞形花序腋生,短小。双悬果扁圆形,有明显隆起的纵棱及细网纹,果梗甚短。气微,味淡。夏、秋二季采收,除去泥沙,晒干。功能主治: 清热利湿,解毒消肿。用于湿热黄疸,中暑腹泻,砂淋血淋,痈肿疮毒,跌扑损伤。浙江大学药学院进行的一项浙江省中医药重点实验室(药理与毒理)建设项目研究发现,清热利湿解毒药物积雪草具有护肝作用,并找到其护肝作用的有效部位,为积雪草药用资源开发利用提供了实验依据。
郁金(拉丁学名:Curcuma petiolata) 异名:马蒁(《唐本草》),黄郁(《石药尔雅》)。别名:玉金,玉京。为姜科植物温郁金Curcuma wenyujin Y. H. Chen et C. Ling、姜黄Curcuma longa L.、广西莪术Curcuma kwangsiensis S. G. Lee et C. F. Liang或蓬莪术Curcuma phaeocaulis Val.的干燥块根。前两者分别习称“温郁金”和“黄丝郁金”,其余按性状不同习称“桂郁金”或“绿丝郁金”。温郁金:呈长圆形或卵圆形,稍扁,有的微弯曲,两端渐尖。长3.5~7cm,直径1.2~2.5cm。表面灰褐色或灰棕色,具不规则的纵皱纹,纵纹隆起处色较浅。质坚实,断面灰棕色,角质样;内皮层环明显。气微香,味微苦。黄丝郁金:呈纺锤形,有的一端细长,长2.5~4.5cm,直径1~1.5cm。表面棕灰色或灰黄色,具细皱纹,断面橙黄色,外周棕黄色至棕红色。气芳香,味辛辣。桂郁金:呈长圆锥形或长圆形,长2~6.5cm,直径1~1.8cm。表面具疏浅纵纹或较粗糙网状皱纹。气微,味微辛苦。绿丝郁金:呈长椭圆形,较粗壮,长1.5~3.5cm,直径1~1.2cm。气微,味淡。冬季茎叶枯萎后采挖,除去泥沙及细根,蒸或煮至透心,干燥。性味:辛、苦,寒。归经:归肝、心、肺经。功能主治: 行气化瘀,清心解郁,利胆退黄。用于经闭痛经,胸腹胀痛、刺痛,热病神昏,癫痫发狂,黄疸尿赤。
女贞子(拉丁学名Fructus Ligustri Lucidi),为木犀科植物女贞Ligustrum lucidum Ait.的干燥成熟果实。常绿大灌木或乔木,高达10米。树干直立,树皮灰绿色,光滑不裂,枝开展,平滑无毛,具明显皮孔。叶对生,卵形至卵状披针形。夏季开白色小花,圆锥花序顶生,花芳香,密集,几乎无梗。浆果状核果,长圆形,一侧稍凸,长约1厘米,熟时蓝黑色。女贞子为女贞果实,呈卵形、椭圆形或肾形,长6~8.5mm,直径3.5~5.5mm。表面黑紫色或灰黑色,皱缩不平,基部有果梗痕或具宿萼及短梗。体轻。外果皮薄,中果皮较松软,易剥离,内果皮木质,黄棕色,具纵棱,破开后种子通常为1粒,肾形,紫黑色,油性。无臭,味甘、微苦涩。性味:甘、苦,凉。归经:归肝、肾经。功能主治:滋补肝肾,明目乌发。用于眩晕耳鸣,腰膝酸软,须发早白,目暗不明。
山楂,(拉丁学名Crataegus pinnatifida Bunge),又名山里果、山里红,为蔷薇科植物山里红Crataegus pinnatifida Bge.var.major N.E.Br.或山楂Crataegus pinnatifida Bge.的干燥成熟果实。落叶乔木或大灌木,高达8米.树皮暗棕色,多分枝,枝条无刺或具稀刺。单叶互生;具托叶,托叶卵圆形至卵状披针形,边缘具锯齿;叶柄长2~4匣米;叶片阔卵形、三角卵形至菱状卵形,长6~12厘米,宽5~8厘米,先端尖,基部楔形,边缘有5~9羽状裂片,裂片有尖锐和不整齐的锯齿,上面绿色,有光泽,下面色较淡,两面脉上均被短柔毛。花10~12朵成伞房花序;花梗被短柔毛;萼片5,绿色,基部连合成杯状,上部5齿裂;花冠白色或带淡红色,直径8~13毫米,花瓣5,离生,倒宽卵形,长和宽均为6毫米;雄蕊20,不等长;心皮5,子房下位,5室,各室具一胚珠,花柱5,柱头圆形。梨果球形或圆卵形,直径约2.5厘米,深红色,具多数白色斑点,果之顶端有外曲的宿存花萼。种子5枚。花期5月。果期8~10月。果呈圆形片,皱缩不平,直径1~2.5cm,厚0.2~0.4cm。外皮红色,具皱纹,有灰白小斑点。果肉深黄色至浅棕色。中部横切片具5粒浅黄色果核,但核多脱落而中空。有的片上可见短而细的果梗或花萼残迹。生于河岸的砂土或干燥多砂石的山坡上.栽培于东北南部、华北以至江苏一带。气微清香,味酸、微甜。秋季果实成熟时采收,切片,干燥。性味: 酸、甘、微温。归经: 归脾、胃、肝经。功能主治: 消食健胃,行气散瘀。用于肉食积滞,胃脘胀满,泻痢腹痛,瘀血经闭,产后瘀阻,心腹刺痛,疝气疼痛;高脂血症。焦山楂消食导滞作用增强。用于肉食积滞,泻痢不爽。
蒲公英(Taraxacum mongolicum Hand.-Mazz.),别名黄花地丁、婆婆丁,呈皱缩卷曲的团块。根呈圆锥形,多弯曲,长3~7cm;表面棕褐色,抽皱;根头部有棕褐色或黄白色的茸毛,有的已脱落。叶基生,多皱缩破碎,完整叶片呈倒披针形,绿褐色或暗灰色,先端尖或钝,边缘浅裂或羽状分裂,基部渐狭,下延呈柄状,下表面主脉明显。花茎1至数条,每条顶生头状花序,总苞片多层,内面一层较长,花冠黄褐色或淡黄白色。有的可见多数具白色冠毛的长椭圆形瘦果。气微,味微苦。春至秋季花初开时采挖,除去杂质,洗净,晒干。性味: 苦、甘,寒。功能主治: 清热解毒,消肿散结,利尿通淋。用于疔疮肿毒,乳痈,瘰疠,目赤,咽痛,肺痈,肠痈,湿热黄疸,热淋涩痛。
猫爪草(拉丁学名Ranunculus ternatus Thunb.),为毛茛科植物小毛茛Ranunculus ternatus Thunb. 的干燥块根。因其块根肉质,数个簇生,近纺锤形,外皮黄褐色,形似猫爪而得名。幼株疏被灰白色的细柔毛,后变秃净或稍具柔毛。块根肉质,纺锤形,常数个聚集。茎高5~15厘米,具分枝;基生叶为3出复叶或3深裂,小叶片卵圆形或阔倒卵形,长0.5~1.5厘米,宽0.5~1厘米,先端3浅裂或齿裂,基部楔形,有时裂成线形或线状披针形,中央裂片较两侧者略大;具叶柄,柄长3~6厘米,基部扩大,边缘膜质;茎生叶互生,通常无柄,3裂,裂片线形,长约1.5厘米,宽约1毫米。花单生于茎端,与叶对生,直径达1.5厘米,花柄长0.5~2厘米,有短细毛:萼片5,长圆形或倒卵形,膜质,绿色,边缘淡黄色,向下反曲,外有细毛;花瓣5,阔倒卵形,黄色,无毛;雄蕊多数,花药长圆形,纵裂,花丝扁平;心皮多数,离生,丛集于膨大的花托上;柱头短小,单一。聚合果球形;瘦果扁卵形,细小,表面淡棕色,平滑,顶端有短喙。花期4~5月。果期5~6月。生境分布:生于田边、路旁、洼地及山坡草丛中。分布浙江、江苏、安徽、江西、广西、河南、湖北、四川、云南、贵州等地。产河南、江苏、浙江、广西等地。春、秋二季采挖,除去须根及泥沙,晒干。性味:甘辛,温。①《中药材手册》:"味微甘。"②《广西中药志》:"味酸甘,性平,无毒。"③《河南中草药手册》:"味辛,性温。"归经:《广西中药志》:"入肝、肺二经。"功能主治:治瘰疬,肺结核,疟疾。中药材手册》:"治颈上瘰疬结核。"《河南中草药手册》:"消肿,截疟。治瘰疬,肺结核。"
本发明的突出的实质性特点和显著的进步是:
1、本发明的中药制剂按中医理论将田基黄、垂盆草、积雪草、郁金、女贞子、山楂、蒲公英、猫爪草按常规制剂工艺生产而成,参考古今中医学药理研究成就,基于中医学对肝病的认识及治疗原则,具有毒副作用小、不产生耐药性,疏肝理气,健脾消食,用于脂肪肝、酒精肝、药物性肝损伤、慢性肝炎及早期肝硬化等,具有解酒、降脂、解毒、抗炎等效果;
2、中医学认为,肝炎的定位在肝,慢性肝炎、肝纤化的病因是疫毒侵入人体,湿热留恋不去,出现气滞血瘀和气血不足等症状,本发明的中药制剂在活血化瘀的同时,辅以清热解毒祛湿,对肝的阴阳气血来进行调整,可取得满意的治疗效果。
3、本发明的中药制剂相比现有技术具有天然、安全、无毒、高效、提高人体免疫力的优点,可以调整人体的失衡状态,达到养生护肝的功效;
4、本发明的中药制剂,其采用纯天然中草药配方,饮用后无残留药物,保障了药品的安全性和人体健康;

具体实施方式
实施例1
步骤1)按重量份数称取田基黄10、垂盆草8、积雪草6、郁金5、女贞子8、山楂14、蒲公英15、猫爪草6分别粉碎,混合在一起;
步骤2)加8倍重量的纯化水,回流煎煮2小时,过滤得滤液a;在滤渣中加入4倍重量的纯化水,再回流煎煮2小时,过滤得滤液b;将滤液a和b混合,进行减压浓缩,使总体积缩减为原来的1/4,即为中药水提浓缩液,备用;
步骤3)将中药水提浓缩液用纯化水稀释至处方量1g/mL,即得成品,置于4℃环境保存备用。
实施例2

本发明防治肝病的中药制剂的应用效果
本发明的防治肝病的中药制剂于2013年研究并试制出来的产品给部分人群进行服用,志愿者35名,其中有酒精性肝病、脂肪性肝病等病患,伴有身体乏力、容易疲劳,部分肝病患者可伴有不同程度黄疸,表现为尿黄、眼睛黄和皮肤黄等症状。服用本发明的中药制剂,开水冲服本发明中药制剂,一次10g,一日2次,结果患者服用后有效率达到95%以上,一个月左右症状大为减轻,3-5个月基本痊愈,除了3名症状较严重者需要到医院治疗以外,32名均不用去医院,也不服用西药。
典型病例
案例一:陈女士得肝炎有两三年了,最近一次单位体检同事都知道她有肝炎病,很多同事都躲着她,大家都怕被传染。无奈下只好回老家了,天天躲在屋里不想出来,在得知她的经济条件不很宽裕,于是服用本发明的中药制剂,开水冲服本发明中药制剂,一次10g,一日2次,三个月后复查,所有化验项目全部正常。遂再吃一个月以资巩固,从此未见再有复发。
案例二:李女士半年前无诱因出现皮肤黄,尿黄,伴乏力,纳差,间断咳嗽,发热,体温38-39℃,无腹痛,腹胀,无恶心,呕吐,无腹泻,无皮肤瘙痒,无关节疼痛,在当地医院治疗后,咳嗽好转,肤黄,尿黄,目黄明显。诊断:肝脾肿大,黄疸原因。于是服用本发明的中药制剂,开水冲服本发明中药制剂,一次10g,一日2次,服用了一个月后,再去复查,各项指标都已正常。

1.一种防治肝病的中药制剂,其特征在于,它包括以下重量份数的原料:田基黄10-12、垂盆草8-10、积雪草6-8、郁金5-7、女贞子8-10、山楂14-16、蒲公英15-17、猫爪草6-8。
2.根据权利要求1所述的防治肝病的中药制剂,其特征在于,各中药原料的重量份数为:田基黄10、垂盆草8、积雪草6、郁金5、女贞子8、山楂14、蒲公英15、猫爪草6。
3.根据权利要求1所述的保肝护肝片,其特征在于,各中药原料的重量份数为:田基黄11、垂盆草9、积雪草7、郁金6、女贞子9、山楂15、蒲公英16、猫爪草7。
4.根据权利要求1所述的防治肝病的中药制剂,其特征在于,各中药原料的重量份数为:田基黄12、垂盆草10、积雪草8、郁金7、女贞子10、山楂16、蒲公英17、猫爪草8。
5.根据权利要求1所述防治肝病中药制剂的生产方法,其特征在于,包含了以下步骤:
步骤1)按重量份数称取田基黄、垂盆草、积雪草、郁金、女贞子、山楂、蒲公英、猫爪草分别粉碎,混合在一起;
步骤2)加8倍重量的纯化水,回流煎煮2小时,过滤得滤液a;在滤渣中加入4倍重量的纯化水,再回流煎煮2小时,过滤得滤液b;将滤液a和b混合,进行减压浓缩,使总体积缩减为原来的1/4,即为中药水提浓缩液,备用;
步骤3)将中药水提浓缩液用纯化水稀释至处方量1g/mL,即得成品,置于4℃环境保存备用。
