本发明涉及一种防治家禽大肠杆菌疾病的注射液，其注射液的组分及其重量份数比分别为：头孢噻呋2-6份；当归1-5份；丹参2-6份；延胡索1-5份；三棱3-7份；土茯苓1-5份；苦参4-8份；赤芍4-8份；金银花4-8份；莪术1-5份；豆豉1-5份；苍术1-5份。本发明配方组成科学合理，工艺简单，使用方便，成本较低，对于鸡大肠杆菌感染引起的大肠杆菌病、白痢、肠炎、气囊炎等疾病有奇效，见效快，疗效好。

技术领域
本发明属于兽药领域，尤其是一种防治家禽大肠杆菌疾病的注射液及其制备方法。
背景技术
近年来，家禽的养殖环境恶劣，疾病不断侵害养殖鸡群，让养殖成本增加，尤其是耐药的大肠杆菌的危害最为严重，鸡感染大肠杆菌病感染肠道。鸡感染肠道疾病、拉痢鸡采食量下降、精神沉郁、拉黄白痢，长期使用抗菌药而治疗无效，鸡各种应激和继发感染大肠杆菌病很多地方已呈爆发性流行的趋势，严重威胁着畜禽业，造成了巨大的经济损失。
发明内容
本发明的目的在于提供一种防治家禽大肠杆菌疾病的注射液，该组合物配方组成科学合理，工艺简单，使用方便，成本较低，对于鸡大肠杆菌感染引起的大肠杆菌病、白痢、肠炎、气囊炎等疾病有奇效，见效快，疗效好。
本发明的另一个目的在于提供一种防治家禽大肠杆菌疾病的注射液的制备方法。
本发明主要通过以下技术方案实现的：

一种防治家禽大肠杆菌疾病的注射液的制备方法，其步骤为：
(1)按配比重量称取当归、丹参、延胡索、三棱、土茯苓、苦参、赤芍、金银花、莪术、豆豉、苍术，经洗净润药烘干后投入中药提取罐内，加1:1投料量的注射用水，在室温下浸泡5小时后，再继续加热至水沸腾煎煮6小时，将提取液0.45um过滤后吸入配液罐内储罐；
(2)中药提取罐内再加入3倍投料量的注射用水，加热至沸腾煎煮2小时将提取液过滤后吸入配液罐内储罐，和第一次提取液合并；
(3)将提取液进行浓缩，将配液罐加热以蒸汽压力0.09Mpa，蒸发器15分钟排放冷凝水一次，每两小时排放加热器的冷凝汽一次；
(4)提取液浓缩至原生药1ml将提取液吸入醇沉罐内，室内自然冷却，备用；
(5)将上清液储罐中的提取液过滤后吸入醇沉罐，自然醇沉10小时收取上清液，弃去残渣，直至原生药1ml；
(6)加入0.3％活性炭，用0.22um聚醚砜滤芯过滤；
(7)在称量区称取头孢噻呋，向配液罐内缓慢加入头孢噻呋，再加入65℃的注射水，加至200L，调节药液PH值：4.0-6.0，搅拌30分钟以上，用0.22um聚醚砜滤芯过滤，药液通过管道直接进入灌装机；
(8)打开灌装机进行灌装，灌装到安瓿瓶内，用注射用水清洗安瓿瓶，将安瓿瓶放置于灭菌柜内100℃流通蒸汽灭菌30分钟，室内自然冷却至室温，即可得到一种防治家禽大肠杆菌疾病的注射液。
本发明的优点及有益效果是：
1.注射液通过抑制细菌DNA旋转酶，阻断DNA的复制，通过打破细菌细胞壁让其他药物直接作用于细胞核，是生物物质外漏来达到杀菌效果，抗菌后效应显著。本品对革兰氏阴性杆菌的抗菌作用强大。
2.注射液对于鸡大肠杆菌感染引起的大肠杆菌败血症、白痢、肠炎、气囊炎、心包炎、肝周炎等疾病有奇效。见效快，疗效好。
3.本发明无毒副作用，使用安全方便。
4.本发明的整个制备过程中，所产生的废弃物不会污染环境，易于处理。
5.本品吸收完全，在1小时内达到血药高峰，半衰期可达48小时，无药物残留，在动物的肝脏、血液、肉组织中的药物残留小于0.001ppM，远远低于出口日本欧盟的药物残留指标。
6.本品为复方制剂，利用药物的相互作用，对大肠杆菌层层破壁，彻底杀灭，降低了耐药性的发生。
具体实施方式
本发明通过以下实施例进一步详述。需要说明的是：下述实施例是说明性的，不是限定性的，不能以下述实施例来限定本发明的保护范围。
实施例1.
一种防治家禽大肠杆菌疾病的注射液，该注射液组分的重量分别为：头孢噻呋4kg、当归3kg、丹参4kg、延胡索3kg、三棱5kg、土茯苓3kg、苦参6kg、赤芍6kg、金银花6kg、莪术3kg、豆豉3kg、苍术3kg。
一种防治家禽大肠杆菌疾病的注射液的制备方法，该注射液制备工艺步骤如下：
(1)亳州凤彦采购购筛选净药材，在称重区按配比重量称取当归、丹参、延胡索、三棱、土茯苓、苦参、赤芍、金银花、莪术、豆豉、苍术，经洗净润药烘干后投入中药提取罐内，加1:1投料量的注射用水，在室温下浸泡5小时后，再继续加热至水沸腾煎煮6小时，将提取液0.45um过滤后吸入配液罐内储罐；
(2)中药提取罐内再加入3倍投料量的注射用水，加热至沸腾煎煮2小时将提取液过滤后吸入配液罐内储罐，和第一次提取液合并；(3)将提取液进行浓缩，将配液罐加热以蒸汽压力0.09Mpa为宜，蒸发器15分钟排放冷凝水一次，每两小时排放加热器的冷凝汽一次。
(4)提取液浓缩至原生药1ml相当于原生药0.5g将提取液吸入醇沉罐内，室内自然冷却，备用；
(5)将上清液储罐中的提取液过滤后吸入醇醇沉罐，自然醇沉10小时收取上清液，弃去残渣，直至原生药1ml相当于原生药1g；
(6)加入0.3％活性炭，用0.22um聚醚砜滤芯过滤；
(7)在称量区称取烟酸诺氟沙星，向配液罐内缓慢加入头孢噻呋，再加入65℃的注射水，加至200L，调节药液PH值：4.0-6.0，搅拌30分钟以上，用0.22um聚醚砜滤芯过滤，药液通过管道直接进入灌装机；
(8)打开灌装机进行灌装，灌装到安瓿瓶内，用注射用水清洗安瓿瓶，将安瓿瓶放置于灭菌柜内100℃流通蒸汽灭菌30分钟，室内自然冷却至室温，用纯化水清洗安瓿瓶后，印字包装，即可得到一种鸡用大肠杆菌注射液。
实施例2.
一种防治家禽大肠杆菌疾病的注射液，该注射液组分的重量分别为：头孢噻呋2kg、当归1kg、丹参2kg、延胡索1kg、三棱3kg、土茯苓1kg、苦参4kg、赤芍4kg、金银花4kg、莪术1kg、豆豉1kg、苍术1kg。
其制备工艺的步骤同实施例1。
实施例3.
一种防治家禽大肠杆菌疾病的注射液，该注射液组分的重量分别为：头孢噻呋3kg、当归2kg、丹参3kg、延胡索2kg、三棱4kg、土茯苓2kg、苦参5kg、赤芍5kg、金银花5kg、莪术2kg、豆豉2kg、苍术2kg。
其制备工艺的步骤同实施例1。
实施例4.
一种防治家禽大肠杆菌疾病的注射液，该注射液组分的重量分别为：头孢噻呋5kg、当归4kg、丹参5kg、延胡索4kg、三棱6kg、土茯苓4kg、苦参7kg、赤芍7kg、金银花7kg、莪术4kg、豆豉4kg、苍术4kg。
其制备工艺的步骤同实施例1。
实施例5.
一种防治家禽大肠杆菌疾病的注射液，该注射液组分的重量分别为：头孢噻呋6kg、当归5kg、丹参6kg、延胡索5kg、三棱7kg、土茯苓5kg、苦参根8kg、赤芍8kg、金银花8kg、莪术5kg、豆豉5kg、苍术5kg。
其制备工艺的步骤同实施例1。
临床试验
注射液在家禽中的临床试验，病鸡表现为精神沉郁，食欲下降，羽毛粗乱，消瘦。侵害呼吸道后会出现呼吸困难，黏膜发绀。侵害消化道后会出现腹泻，排绿色或黄绿色稀便。侵害关节后表现为跗关节和指关节肿大，在关节和附近有大小不一的水泡或脓包。侵害眼睛时，眼肿胀，有黄白色渗出物。侵害大脑时，出现神经症状，表现为头颈震颤，角弓反张。肝周炎，气囊炎，纤维素心包炎，关节炎，全眼球炎，输卵管炎，也有可能在心脏，胰脏，肠道出现黄白色肉芽肿。试验时间：2014.03.28日，试验鸡群，莱阳慧和AA肉仔鸡，35日龄，体重2.1公斤，300只，分为4组，对照组，试验组，常规治疗I组，常规治疗II组每组75只，用法用量：治疗组肌内注射一次量每1kg体重，鸡0.05ml，一日一次，仅用一次。
对照组：为空白组，不使用任何药物治疗。
本发明实验组：为注射液治疗组。1kg体重0.05ml
常规治疗I组：为市售药物治疗组，氟苯尼考溶液10％，2ml兑水1000ml,连用3天。
常规治疗II组：为市售药物治疗组硫酸庆大霉素可溶性粉5％，1g兑水1000ml，连用3天。
试验结果如下：
试验表明：
综上所述，本发明注射液对鸡大肠杆菌引起的心包炎、肝周炎、肠炎腹泻，见效快，疗效好。
