Abstract
本发明提供一种食品添加剂的制备方法,以海藻类植物为原料,通过酸碱提取、酶解处理、微波处理,可明显提高海藻活性物质的提取效率,并通过菌群发酵对其中的活性成分进行改性使其具有分解大分子团,去除有害物质等功能,通过巴氏杀菌使其符合食用要求,可应用于改善生物体健康。

技术领域
本发明涉及一种食品添加剂的制备方法及其应用,特别涉及海藻类植物功能性活性物质的生物提取方法,以海藻类植物作为原料制备食品添加剂。
背景技术
随着中国产业工业化的发展,环境污染已成为社会十分关注的问题,大量的水污染,空气污染,土壤污染,又通过生物链污染到人类赖于生存的动植物产品及水源,其中重金属污染以隐蔽的方式威胁到食品安全。重金属污染泛指铅、汞、镉、砷等重金属的污染,常以慢性中毒的方式危害于人类。铅中毒是一种蓄积性中毒,主要通过空气、饮水、土壤和被铅污染的食物进入人体内而引起中毒,中毒后往往表现为智力低下,反应迟钝,贫血等慢性中毒症状,尤其影响儿童的生长发育。目前汽车废气、蓄电池污染、电子电器产品废弃物等均是铅污染的主要来源。镉中毒发生的原因主要来自锌冶炼、矿山、电镀、油漆、颜料、陶瓷、塑料和农药等生产中排放的"三废",尤其是废水。镉中毒表现为肾脏损害,实验动物表明,镉有致癌、致畸和致突变作用。汞中毒以有机汞中毒为主,会损害神经系统,引起语言和听觉障碍,由工业三废和药物污染(农药和医药)引起。铝元素的过量摄入会增加患老年痴呆的风险,而油条中的铝超标也是目前社会关注的话题。防治重金属污染要贯彻预防为主的方针,国家定期对环境重金属污染物的监测,采取有效措施,严防工业部门排出的三废污染环境;建立区域性动植物产品定期监测制度或监控计划,对区域性的监测结果进行分析,有的放矢采取综合措施,逐步净化重金属对食品的污染。
海藻属于低等植物,整个藻体都能够从海水中吸收无机物和小分子有机物质作养料,同时还可向周围分泌出有机或无机物质,生长在海水复杂环境中的海藻,其代谢过程和代谢产物与陆地植物相比是极为不同的。大多数海藻含有丰富的碳水化合物(木聚糖、干露聚糖和糖醇等)、含氮化合物(氨基酸、肽类、胺类、细胞色素C、吲哚类化合物和糖酶类等)、色素(叶绿素、类胡萝b素和藻胆蛋白)、脂类化合物(三酰甘油、烃类、蜡酯、脂肪酸、磷脂类和糖脂类等)、酚类化合物、维生素(维生素A和D的前体以及维生素E、K等脂溶性维生素和B族维生素等水溶性维生素)、海藻药类(凝集素等)和无机成分等。因此,海藻具有丰富的营养价值和药用价值。
海藻提取物取材于深海区域的大型经济藻类,它含有丰富的氨基酸、矿物质、多糖、维生素及生理活性物质,对提高作物产量、改善品质和提早成熟并在水果保鲜和抵抗病虫害等方面均有明显的效果。大量的研究表明某些海洋藻类中的提取物能够促进作物生长、增加产量、减少病虫害、增加作物抗寒、抗旱能力。而有关海藻提取物分解大分子团,去除有害物质,如重金属和有机物方面的作用还未见报道。
实际上,海藻提取物的性质和使用效果受提取方式的影响很大。目前,海藻提取物的制备工艺主要有碱提取法、中性水解法、酸提取法和机械破碎提取法。此外,还有超声破碎法、酶解法和热水浸泡法。然而,在这些传统的提取方法中,海藻的生物活性成分受细胞壁主要成分纤维素的阻碍,往往难以被有效提取,本发明恰当地将传统的酸碱提取法、酶解法与微波处理相结合以提高提取效率,同时通过菌群发酵调节提取物中活性成分的含量及性质,使提取物中各组分更好地协同作用,从而促进大分子团的分解,并能吸收或去除重金属和甲醛等有害物质。
本研究以海藻类植物为原料,通过酸碱提取、酶解处理、微波处理,可明显提高海藻活性物质的提取效率,并通过菌群发酵对其中的活性成分进行改性使其具有分解大分子团,去除有害物质等功能,扩大其在改善健康或空气、土壤、水环境中的应用,而有关海藻提取液用作食品添加剂来去除有害物质未见报道。
发明内容
本发明的目的是提供一种食品添加剂的制备方法及其应用,特别涉及海藻类植物功能性活性物质的生物提取方法。
本发明提供了一种可分散大团分子的食品添加剂,以重量百分比计,其主要成分为水91.7%,醋酸4.2%,碳水化合物2.4%,蛋白质0.1%。此外,以及微量的脂质、灰分、钠、钾、钙、镁、磷、铁、钛、锌、锆、铑、钯、维他命A、维他命D、维他命B1、维他命B2、烟酸、维他命B6、维他命B12、叶酸、泛酸、维他命C。
本发明的另一方面,提供上述可分散大团分子的海藻提取液的制备方法,其包括以下步骤:
(1)萃取处理:
将海藻洗净切碎,以10~30倍的水或10~95%乙醇提取,温度为50~100℃,共提取3次,合并3次提取液;
(2)微波处理:
对步骤(1)的提取液进行微波处理,辐射时间5-10min、微波功率800-1200W;
(3)酶解:
将步骤(1)的微波处理提取液减压浓缩至原体积的1/3-1/4,加入包含纤维素酶、β-葡聚糖酶、木瓜蛋白酶的复合酶制剂,经恒温30℃水浴8-12小时后取出,置-18℃下速冻成冰,取出置室温下融化,该速冻融化循环重复三次;
(4)菌群发酵:
在步骤(3)的酶解液中按1-5%的体积比接入发酵菌群,并在25~30℃下密闭发酵3~6天,即得海藻提取液;所述发酵菌群为黑曲霉、根霉、里氏木霉、地衣芽孢杆菌、枯草芽孢杆菌、玫瑰微球菌、植物乳酸杆菌、假单胞菌、交替单胞菌、植物酵母菌、富硒酵母菌所组成的发酵菌群;
(5)巴氏杀菌:
将上述步骤的海藻提取液进行巴氏杀菌得到所述食品添加剂。
具体地,所述海藻原料可选自巨藻、墨角藻、马尾藻、海囊藻、泡叶藻、红藻的任一种,或选用两种以上海藻物的混合。优选马尾藻,更优选涠州马尾藻或亨氏马尾藻。
根据本发明的另一个方面,上述步骤(1)中可采用机械冲击剪切磨、气流式超微粉碎磨、球磨机、搅拌磨或振动磨进行超微粉碎。
根据本发明的另一个方面,上述步骤(3)中采用的复合酶制剂由重量份数比为1.2~3.6:2.1~5.2:1.6~4.5的纤维素酶、β-葡聚糖酶、木瓜蛋白酶组成。优选的重量份数比1.2:2.1:1.6。
根据本发明的另一个方面,上述步骤(4)中所述发酵条件为:温度28℃,初始pH值6.5,搅拌速度100r/min,间歇搅拌5min/2h,发酵周期72小时。
根据本发明的又一方面,上述步骤(4)中发酵菌群由重量份数比为1.8:1.2:1.6:1.1:1.5:3.1:2.4:2.7:1.9:1.3:1的黑曲霉、根霉、里氏木霉、地衣芽孢杆菌、枯草芽孢杆菌、玫瑰微球菌、植物乳酸杆菌、假单胞菌、交替单胞菌、植物酵母菌、富硒酵母菌组成。
根据本发明的又一方面,可进一步包括如下步骤:将由步骤(5)制得的15~20份食品添加剂与硬脂酰乳酸钠0~10份、卡拉胶5~15份、蔗糖酯5~10份、硬脂酸钠1~3份、麦芽糖糊精1~3份、淀粉酶0.09~0.11份、淀粉9~11份,在25-35℃下搅拌溶解混匀后静置45~60分钟,经过微波干燥灭菌机进行65℃~70℃的干燥灭菌处理,处理后的食品添加剂含水量为20~25%,然后采用粉碎机粉碎至200目的食品添加剂,既得粉状食品添加剂。
一种食品添加剂的应用,将所述食品添加剂添加到液态食品如饮料中。
一种食品添加剂的应用,将所述食品添加剂添加到面粉或米粉中混合,通过干燥或烘开的方式得到面制食品或米制食品。
本发明提供的食品添加剂,其中的金属元素在发酵过程中可形成金属氧化物或金属络合物,将上述发酵后得到的提取物在超临界状态下制成金属氧化物的超微纳米粒子,该超微纳米粒子具有高的远红外发射能力,其辐射的远红外线作用于附近接触或非接触的物质,如大的水分子团,使水分子产生共振吸收,从而使得部分水分子之间的氢键断裂,使该部分水分子会分离,进而使水分子团簇变小而得到活化。当该食品添加剂应用于食品时,能有效分解生物体中的大分子团簇,使该食品更易被生物体吸收,更具口感。且该食品添加剂随食品被生物体(如人)吸收时,能将有效分解或降低生物体中血液中的有害物质,平衡生物体的酸碱性,从而促进人体的健康。
具体实施方式
实施例1:
含红藻提取液的食品添加剂的制备
取100g红藻洗净切碎,按照下述步骤制备:
(1)微波萃取处理:
将海藻洗净切碎,以40倍的重量的水进行微波提取,辐射时间5-10min,微波功率800-1200W,温度为35℃,共提取3次,合并3次提取液;
(2)酶解:
将步骤(1)的微波处理提取液减压浓缩至原体积的1/3,加入重量份数比1.2:2.1:1.6的纤维素酶、β-葡聚糖酶、木瓜蛋白酶组成的复合酶制剂,经恒温30℃水浴8小时后取出,置-18℃下速冻成冰,取出置室温下融化,该速冻融化循环重复三次;
(3)菌群发酵:
在步骤(2)的酶解液中按1%的体积比接入发酵菌群,并在25~30℃下密闭发酵3天,即得海藻提取液;所述发酵菌群为黑曲霉、根霉、里氏木霉、地衣芽孢杆菌、枯草芽孢杆菌、玫瑰微球菌、植物乳酸杆菌、假单胞菌、交替单胞菌、植物酵母菌、富硒酵母菌所组成的发酵菌群。
(4)巴氏杀菌:
将上述步骤的海藻提取液进行巴氏杀菌得到所述食品添加剂。
实施例2:巨藻提取液的制备
(1)微波萃取处理:
取100g巨藻洗净切碎,按照下述步骤制备:
将巨藻洗净切碎,以10倍的重量的95%乙醇微波提取,辐射时间5-10min,微波功率800-1200W,温度为50℃,共提取3次,合并3次提取液;
(2)酶解:
将步骤(1)的微波处理提取液减压浓缩至原体积的1/4,加入重量份数比3.6:5.2:4.5的纤维素酶、β-葡聚糖酶、木瓜蛋白酶组成的复合酶制剂,经恒温30℃水浴8小时后取出,置-18℃下速冻成冰,取出置室温下融化,该速冻融化循环重复三次;
(4)菌群发酵:
在步骤(3)的酶解液中按2%的体积比接入发酵菌群,并在30℃下密闭发酵6天,即得提取液;所述发酵菌群为黑曲霉、根霉、里氏木霉、地衣芽孢杆菌、枯草芽孢杆菌、玫瑰微球菌、植物乳酸杆菌、假单胞菌、交替单胞菌、植物酵母菌、富硒酵母菌所组成的发酵菌群。
(5)巴氏杀菌:
将上述步骤的海藻提取液进行巴氏杀菌得到所述食品添加剂。
可以理解,为了便于存储和应用,所述食品添加剂还可支撑粉状形式,具体可包括如下步骤:将由实施例1或2中所制得的15~20份食品添加剂与硬脂酰乳酸钠0~10份、卡拉胶5~15份、蔗糖酯5~10份、硬脂酸钠1~3份、麦芽糖糊精1~3份、淀粉酶0.09~0.11份、淀粉9~11份,在25-35℃下搅拌溶解混匀后静置45~60分钟,经过微波干燥灭菌机进行65℃~70℃的干燥灭菌处理,处理后的食品添加剂含水量为20~25%,然后采用粉碎机粉碎至200目的食品添加剂,既得粉状食品添加剂。
所述食品添加剂的应用方式不限,譬如可将食品添加剂添加到液态食品如饮料或水中。也可将所述食品添加剂的制备方法添加到面粉或米粉中混合,通过干燥或烘开的方式得到面制食品或米制食品。
实施例3:活化水的效果测试
A组:取未经处理的普通10mL自来水中,10min后过滤,离心分离,取上清液测试水分子17O核磁共振半高宽(NMRFWHM)。
B组:取实施例1的食品添加剂0.001mL加入未经处理的普通10mL自来水,静止12个小时后过滤,离心分离,取上清液测试水分子17O核磁共振半高宽(NMRFWHM)。
C组:取实施例2的食品添加剂0.001mL加入未经处理的普通10mL自来水,,静止12个小时后过滤,离心分离,取上清液测试水分子17O核磁共振半高宽(NMRFWHM)。
测试仪器为BruckerAVANCEIII400MHz的超导核磁共振仪,依据JY/T007-1996超导脉冲傅立叶变换核磁共振谱方法进行测定。
测试结果:3组样品的半峰高如表所示:

由此表明,实施例1、2中的食品添加剂均能够降低水分子的17ONMR半高宽,即提高了水中具有5~6个水分子缔合的小分子团的比例,水分子活性得到了提高。
实施例4:采用本食品添加剂抑制酸碱性的动物试验报告
选用小白鼠六只,分成三组,A组为试验1组(两只),B组为试验2组(两只),C组为对比组(两只)。
A组试验组
称重两只小白鼠的体重,分别为160g,试验前进行体检,分别抽取血液样本10ml,进行化验,检测结果属正常,血液中酸碱性正常。
然后每天早上给予每只小白鼠喂食含10g实施例1制备的提取液的食物30g,其中添加有铅0.003mg给小白鼠吃;中午再喂给相同量的相同食物给白鼠吃,下午喂的食物与之前两次喂的食物均相同。连续喂食两周后,抽血化验,检测结果:血液中酸碱性正常。
B组对比组
对试验小白鼠事先喂食了含有重金属铅的食物,两周后,抽血化验检测两只小白鼠都不同程度含有铅重金属。一只铅≥0.054mg,另一只铅≥0.028mg。经过喂食实施例2制备的提取液的食物30g,其中不添加铅,两周后再次检测,其中一只铅含量降至0.035mg,另一只铅降至0.024mg,证明提取物有协同置换重金属作用。这是提取液中包含的蛋白质和微量元素、氨基酸等生物活性物质协同作用的效果。
C组对比组
与A组试验内容相同,只是其食物中,不含有实施例制备的提取液,仅含与A组食物相同的重金属铅。喂食量和次数均相同,连续喂食两周后,两只小白鼠分别抽血5ml化验,检测结果:血液中含有铅≥0.052mg、≥0.064mg。证明铅已被血液吸收至体内。
结论:上述试验证明本发明制备的食品添加剂可抑制重金属在血液中吸收,平衡血液中的酸碱性。
实施例5:以面包用食品添加剂为例
食品添加剂的配制:分别取实施例1和2中的食品添加剂,按重量0.2-0.5%加入面粉中,充分搅拌即可。
根据GB2760-2011食品安全国家标准食品添加剂使用标准,检测方法对所配制的添加剂分别对排重金属和酸碱性进行检验。(实验条件:温度20℃±2℃;相对湿度50%±10%),检测结果见下表:

备注:0.2%、0.5%表明食品添加剂占面粉重量的百分比。
结论:由检测结果可以看出,本食品添加剂在使用时,对排除铅镉汞具有很好的效果,并且食品添加剂使用量小。

Claims (7)
1.一种食品添加剂的制备方法,其特征在于包括以下步骤:
(1)萃取处理:
将海藻洗净切碎,以10~30倍的水或10~95%乙醇提取,温度为50~100℃,共提取3次,合并3次提取液;
(2)微波处理:
对步骤(1)的提取液进行微波处理,辐射时间5-10min、微波功率800-1200W;
(3)酶解:
将步骤(2)的微波处理提取液减压浓缩至原体积的1/3-1/4,加入包含纤维素酶、β-葡聚糖酶、木瓜蛋白酶的复合酶制剂,经恒温30℃水浴8-12小时后取出,置-18℃下速冻成冰,取出置室温下融化,该速冻融化循环重复三次,其中所述复合酶制剂的组成为:纤维素酶、β-葡聚糖酶、木瓜蛋白酶重量份配比为1.2~3.6:2.1~5.2:1.6~4.5;
(4)菌群发酵:
在步骤(3)的酶解液中按1-5%的体积比接入发酵菌群,并在25~30℃下密闭发酵3~6天,即得海藻提取液;所述发酵菌群为黑曲霉、根霉、里氏木霉、地衣芽孢杆菌、枯草芽孢杆菌、玫瑰微球菌、植物乳酸杆菌、假单胞菌、交替单胞菌、植物酵母菌、富硒酵母菌所组成的发酵菌群;
(5)将上述发酵后的海藻提取液在超临界状态下制成金属氧化物的超微纳米粒子;
(6)巴氏杀菌:
将上述步骤(5)的海藻提取液进行巴氏杀菌得到所述食品添加剂。
2.如权利要求1所述的食品添加剂的制备方法,其特征在于所述海藻为巨藻、墨角藻、马尾藻、海囊藻、泡叶藻、红藻的任一种,或选用两种以上海藻物的混合。
3.一种如权利要求1所述的食品添加剂的制备方法,其特征在于步骤(4)中所述发酵条件为:温度28℃,初始pH值6.5,搅拌速度100r/min,间歇搅拌5min/2h,发酵周期72小时。
4.一种如权利要求1所述的食品添加剂的制备方法,其特征在于进一步包括如下步骤:将由步骤(6)制得的15~20份食品添加剂与硬脂酰乳酸钠0~10份、卡拉胶5~15份、蔗糖酯5~10份、硬脂酸钠1~3份、麦芽糖糊精1~3份、淀粉酶0.09~0.11份、淀粉9~11份,在25-35℃下搅拌溶解混匀后静置45~60分钟,经过微波干燥灭菌机进行65℃~70℃的干燥灭菌处理,处理后的食品添加剂含水量为20~25%,然后采用粉碎机粉碎至200目的食品添加剂,即得粉状食品添加剂。
5.一种如权利要求1所述食品添加剂的制备方法,其特征在于,所述食品添加剂包括以下主要成分:
以及微量的脂质、灰分、钠、钾、钙、镁、磷、铁、钛、锌、锆、铑、钯、维他命A、维他命D、维他命B1、维他命B2、烟酸、维他命B6、维他命B12、叶酸、泛酸、维他命C。
6.一种食品添加剂的应用,其特征在于,将由权利要求1所述食品添加剂的制备方法所制备的食品添加剂添加到液态食品中。
7.一种食品添加剂的应用,其特征在于,将由权利要求1或2所述食品添加剂的制备方法所制备的食品添加剂添加到面粉或米粉中混合,通过干燥的方式得到面制食品或米制食品。
