Abstract
本发明公开了一种用于黄鳝养殖水的微生物净水剂及其制备方法。所述制备方法:将30-50重量份表面修饰C8和C18烷基链的磁性硅球加入到150-250重量份的水中;加入150-250重量份微生物试剂:红螺菌40-50份、红假单胞菌40-60份、亚硝化叶菌30-50份、硝化球菌30-40份、脱氮硫杆菌20-30份、浮游球衣菌30-40份、透明颤菌2-4份、硫杆菌2-4份、草履虫30-50份、线虫40-60份、扁藻40-50份、蓝藻40-50份;再加入60-80重量份牛肉膏蛋白胨琼脂培养基培养。将本发明的微生物净水剂,投入黄鳝养殖池中,能达到净化养殖池水的目的。

本发明涉及一种微生物净水剂及其制备方法,尤其涉及一种用于黄鳝养殖水的微生物净水剂及其制备方法。
背景技术
近年来,高效农业的快速推进,使得特种水产养殖业发展迅猛,河蟹、青虾、克氏原螯虾、罗氏沼虾、南美白对虾、中华鳖、鳜鱼、鲈鱼、河鲀、黄鳝、泥鳅等名特优新水产养殖品种已达到规模化生产的水平,形成了自身的特点。
黄鳝属鱼纲、合鳃目、合鳃科、黄鳝亚科,亦称鱓鱼、罗鳝、蛇鱼、长鱼(苏北一带)。黄鳝分布广泛,在中国几乎各地都有分布。黄鳝营养价值极高,根据美国试验研究资料,经常摄取卵磷脂,记忆力可以提高20%。故食用鳝鱼肉有补脑健身的功效。它所含的特种物质“鳝鱼素”,有清热解毒、凉血止痛、祛风消肿、润肠止血、健脾等功效,能降低血糖和调节血糖,对痔疮、糖尿病有较好的治疗作用,加之所含脂肪极少,因而是糖尿病患者的理想食品。鳝鱼含有的维生素A量高得惊人。维生素A可以增进视力,促进皮膜的新陈代谢。每100克鳝鱼肉中蛋白质含量达17.2~18.8克,脂肪0.9~1.2克,钙质38毫克,磷150毫克,铁1.6毫克;此外还含有硫胺素(维生素B1)、核黄素(B2)、尼克酸(维生素PP)、抗坏血酸(维生素C)等多种维生素。黄鳝不仅被当作名菜用来款待客人,还能活运出口,畅销国外,更有冰冻鳝鱼远销美洲等地。黄鳝一年四季均产,但以小暑前后者最佳,民间有“小暑黄鳝赛人参”的说法。黄鳝的经济价值和食用价值都非常高,深受老百姓的欢迎,而天然的产量远远满足不了人们的需求,因此人工饲养黄鳝的发展方兴未艾,已经蓬勃发展了很多年。
人工饲养黄鳝的食物以小杂鱼、蚯蚓、螺蚬、蚌肉等为主,以蚯蚓为最佳,也可喂水蚤、豆渣、菜籽饼、瓜果等人工配合饲料。但是由于饲料投放量往往大于黄鳝的进食量,多余的饲料得不到有效处理容易造成水质下降,并且黄鳝排泄物相对量比较大,如果不对养殖水实施处理,容易造成黄鳝发病率升高、存活率降低,产量下降等问题。常规的解决方式是间隔一段时间进行换水处理,但是换水一方面提高生产及管理成本,另一方面也容易引起黄鳝的不适应症状。
发明内容
针对现有黄鳝养殖技术中存在的上述不足,本发明所要解决的技术问题之一是提供一种黄鳝养殖水的微生物净水剂的制备方法。
本发明所要解决的技术问题之二是提供一种黄鳝养殖水的微生物净水剂,采用上述方法制备而成。
本发明所要解决的技术问题,是通过如下技术方案实现的:
一种用于黄鳝养殖水的微生物净水剂的制备方法,由下述步骤组成:
(1)将30-50重量份表面修饰烷基链的磁性硅球加入到150-250重量份的水中;
(2)在2-10转/秒搅拌条件下,加入150-200重量份微生物试剂,所述微生物试剂由下述重量份组分组成:红螺菌(Rhodospirillum)40-50份、红假单胞菌(Rhodopseudomonas)40-60份、亚硝化叶菌(Nitrosolobus)30-50份、硝化球菌(Nitrococcus)30-40份、脱氮硫杆菌(Thiobacillus denitrificans)20-30份、浮游球衣菌(Sphaerotilus natans)30-40份、透明颤菌(Vitreoscilla)2-4份、硫杆菌(Thiobacillus)2-4份、草履虫(Paramoecium)30-50份、线虫(nematode)40-60份、扁藻(Tetraselmis Chui)40-50份、蓝藻(Cyanobacteria)40-50份;
(3)再加入60-80重量份牛肉膏蛋白胨琼脂培养基常温下培养20-30小时,即可制得本发明的用于黄鳝养殖水的微生物净水剂。
优选地,
步骤(1)中,所述表面修饰烷基链的磁性硅球为表面修饰C8烷基链的磁性硅球和/或表面修饰C18烷基链的磁性硅球。
表面修饰C8烷基链的磁性硅球,具体可以采用中国发明专利CN101185874A实施例1所公开的方法进行制备。
进一步地,所述表面修饰烷基链的磁性硅球为表面修饰C8和C18烷基链的磁性硅球,采用下述方法制备而成:
(1-1)用水热法合成Fe3O4磁性纳米粒子:将1克FeCl3·6H2O,溶于30ml乙二醇中,添加4克无水乙酸钠,反应温度为200℃,反应时间为12小时,生成Fe3O4磁性纳米粒子;
(1-2)将上述Fe3O4磁性纳米粒子用水洗涤,除去水溶性杂质,然后将其分散在10g水中,得分散液A,取离心管,加3 g上述分散液A,加入5ml 2M HCl溶液超声8min,再用磁铁分离除去绿色清液,然后用10ml水分散,再用磁铁收集粒子,去除浅绿色清液,用5 ml 10wt%的柠檬酸三钠溶液超声分散,静置2h,重新用5 ml水分散后得分散液B;
(1-3)取上述分散液B 1g,用5ml水和30ml乙醇稀释后,再加20wt%氨水0.5ml,超声分散10分钟,搅拌,加入1ml正硅酸乙酯,室温搅拌12小时,用磁铁收集,洗涤,干燥,得到复合微球;
(1-4)将0.01g上述复合微球加入到1.0g无水吡啶中,超声分散,然后再加入表面修饰剂,所述表面修饰剂为0.1g二甲基辛基硅烷和0.2g十八烷基三乙氧基硅烷组成的混合物,室温搅拌12小时,用乙醇和水洗涤,再真空干燥,制得表面修饰C8和C18烷基链的磁性硅球。
步骤(3)中,所述牛肉膏蛋白胨琼脂培养基由牛肉膏0.5g、蛋白胨1g、NaCl 0.5g、琼脂2.0g和水 100 ml采用常规方法配制而成。
本发明还提供了一种用于黄鳝养殖水的微生物净水剂,由上述方法制备而成。
将本发明的用于黄鳝养殖水的微生物净水剂,投入到养殖有黄鳝的水池中,净水剂可以沉到池底的水-沉积物交界面,能达到净化养殖池水的目的。由于采用了比表面积大的磁性纳米材料做载体,该微生物净水剂具有易于沉淀、净化效率高、效果稳定且易于处理等优点,非常适合黄鳝养殖水的在线处理。
具体实施方式
下面结合实施例对本发明作进一步描述。
实施例1:
(1)表面修饰C8和C18烷基链的磁性硅球的制备:
(1-1)用水热法合成Fe3O4磁性纳米粒子:将1克FeCl3·6H2O,溶于30ml乙二醇中,添加4克无水乙酸钠,反应温度为200℃,反应时间为12小时,生成Fe3O4磁性纳米粒子;
(1-2)将上述Fe3O4磁性纳米粒子用水洗涤,除去水溶性杂质,然后将其分散在10g水中,得分散液A,取离心管,加3 g上述分散液A,加入5ml 2M HCl溶液超声8min,再用磁铁分离除去绿色清液,然后用10ml水分散,再用磁铁收集粒子,去除浅绿色清液,用5 ml 10wt%的柠檬酸三钠溶液超声分散,静置2h,重新用5 ml水分散后得分散液B;
(1-3)取上述分散液B 1g,用5ml水和30ml乙醇稀释后,再加20wt%氨水0.5ml,超声分散10分钟,搅拌,加入1ml正硅酸乙酯,室温搅拌12小时,用磁铁收集,洗涤,干燥,得到复合微球;
(1-4)将0.01g上述复合微球加入到1.0g无水吡啶中,超声分散,然后再加入表面修饰剂,所述表面修饰剂为0.1g二甲基辛基硅烷和0.2g十八烷基三乙氧基硅烷组成的混合物,室温搅拌12小时,用乙醇和水洗涤,再真空干燥,制得表面修饰C8和C18烷基链的磁性硅球。
(2)将40重量份上述表面修饰C8和C18烷基链的磁性硅球加入200重量份的水中;
(3)在5转/秒搅拌条件下加入170重量份的微生物试剂,所述微生物试剂由下述重量份组分组成:红螺菌(Rhodospirillum)45份、红假单胞菌(Rhodopseudomonas)50份、亚硝化叶菌(Nitrosolobus)35份、硝化球菌(Nitrococcus)40份、脱氮硫杆菌(Thiobacillus denitrificans)25份、浮游球衣菌(Sphaerotilus natans)35份、透明颤菌(Vitreoscilla) 4份、硫杆菌(Thiobacillus)2份、草履虫(Paramoecium)40份、线虫(nematode)55份、扁藻(Tetraselmis Chui)45份、蓝藻(Cyanobacteria)45份;
(4)再加入75重量份普通牛肉膏蛋白胨培养基(由牛肉膏0.5g,蛋白胨1g,NaCl 0.5g,琼脂2.0g, 水 100 ml采用本行业通用方法配制而成)常温下培养24小时,即可制得用于黄鳝养殖水的微生物净水剂。
实施例2:
与实施例1基本相同,制得用于黄鳝养殖水的微生物净水剂,区别仅在于步骤(1-4)中表面修饰剂不同。实施例2中采用0.3g二甲基辛基硅烷为表面修饰剂。
实施例3:
与实施例1基本相同,制得用于黄鳝养殖水的微生物净水剂,区别仅在于步骤(1-4)中表面修饰剂不同。实施例3中采用0.3g十八烷基三乙氧基硅烷为表面修饰剂。
测试例:
取四块大小及水质完全相同的黄鳝养殖水池(2m×2m×1.2 m)进行测试,每块水池投40g左右的黄鳝幼苗400只。其中一块泼洒实施例1的微生物净水剂;一块泼洒实施例2的微生物净水剂;一块泼洒实施例3的微生物净水剂;还有一块不投放任何净水剂,作为空白对照组。
将40克微生物净水剂分别泼洒至黄鳝养殖水池中,7天后,对所有的黄鳝养殖水池进行水质测定。水质测量的依据为GB11607-89中华人民共和国国家标准渔业水质标准和NY5051-2001无公害食品淡水养殖用水水质,测定指标及数据如下表所示。

由表中数据可以看出,实施例1的各项水质指标均优于实施例2和实施例3,更明显优于对照池。
以上所述,仅为本发明的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本领域的技术人员在本发明所揭露的技术范围内,可不经过创造性劳动想到的变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应该以权利要求书所限定的保护范围为准。
Claims (4)
1.一种用于黄鳝养殖水的微生物净水剂的制备方法,其特征在于,由下述步骤组成:
(1)将30-50重量份表面修饰烷基链的磁性硅球加入到150-250重量份的水中;
(2)在2-10转/秒搅拌条件下,加入150-200重量份微生物试剂,所述微生物试剂由下述重量份组分组成:红螺菌40-50份、红假单胞菌40-60份、亚硝化叶菌30-50份、硝化球菌30-40份、脱氮硫杆菌20-30份、浮游球衣菌30-40份、透明颤菌2-4份、硫杆菌2-4份、草履虫30-50份、线虫40-60份、扁藻40-50份、蓝藻40-50份;
(3)再加入60-80重量份牛肉膏蛋白胨琼脂培养基常温下培养20-30小时。
2.如权利要求1所述的用于黄鳝养殖水的微生物净水剂的制备方法,其特征在于:步骤(1)中,所述表面修饰烷基链的磁性硅球为表面修饰C8烷基链的磁性硅球和/或表面修饰C18烷基链的磁性硅球。
3.如权利要求2所述的用于黄鳝养殖水的微生物净水剂的制备方法,其特征在于,步骤(1)中,所述表面修饰烷基链的磁性硅球为表面修饰C8和C18烷基链的磁性硅球,采用下述方法制备而成:
(1-1)用水热法合成Fe3O4磁性纳米粒子:将1克FeCl3·6H2O,溶于30ml乙二醇中,添加4克无水乙酸钠,反应温度为200℃,反应时间为12小时,生成Fe3O4磁性纳米粒子;
(1-2)将上述Fe3O4磁性纳米粒子用水洗涤,除去水溶性杂质,然后将其分散在10g水中,得分散液A,取离心管,加3 g上述分散液A,加入5ml 2M HCl溶液超声8min,再用磁铁分离除去绿色清液,然后用10ml水分散,再用磁铁收集粒子,去除浅绿色清液,用5 ml 10wt%的柠檬酸三钠溶液超声分散,静置2h,重新用5 ml水分散后得分散液B;
(1-3)取上述分散液B 1g,用5ml水和30ml乙醇稀释后,再加20wt%氨水0.5ml,超声分散10分钟,搅拌,加入1ml正硅酸乙酯,室温搅拌12小时,用磁铁收集,洗涤,干燥,得到复合微球;
(1-4)将0.01g上述复合微球加入到1.0g无水吡啶中,超声分散,然后再加入表面修饰剂,所述表面修饰剂为0.1g二甲基辛基硅烷和0.2g十八烷基三乙氧基硅烷组成的混合物,室温搅拌12小时,用乙醇和水洗涤,再真空干燥,制得所述表面修饰C8和C18烷基链的磁性硅球。
4.一种用于黄鳝养殖水的微生物净水剂,其特征在于:采用权利要求1和2和3所述方法制备而成。
