本发明公开了一种用于防治羊白肌病的中药组合物及其制备方法,其中药组合物包括以下原料药材:陈皮、木香、甘松、神曲、麦芽、鸡矢藤、党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉、菟丝子、沙苑子、海马、女贞子、当归、熟地黄、何首乌、龙眼肉、酸枣仁、野料豆、蚂蝗七、人面子、卷茎蓼和独脚柑。本发明的有益效果是:本发明的中药组合物治疗羊白肌病可健脾消食,益气养血,增强气血的生成及循环,加强羊脏腑正常功能的运行,促进正常肌肉组织的生成,维持肌肉、关节的正常功能;疗程短、见效快、成本价低,纯天然无污染。

技术领域
本发明涉及中兽药技术领域,特别涉及一种用于防治羊白肌病的中药组合物及其制备方法。
背景技术
白肌病是幼畜的一种以骨骼肌、心肌纤维以及肝组织发生变性、坏死为主要特征的疾病,因病变部位肌肉色淡,甚至苍白而得名。各种动物特别是幼畜、幼禽均可发生,山羊羔的发病率可达90%以上,死亡率也很高。挪威、瑞典、英国、德国、日本、新西兰、俄罗斯、美国等都有本病的发生。我国的西北、华北、西南等地,特别是山区、丘陵地带都有本病的报道。个体营养良好与否均可发病,且常呈地方性发生。
病因
白肌病早于1863年就有记载,对其病因发病学及防治的研究,近30多年来进展较快。据目前的研究资料及防治效果来看,硒和维生素E的缺乏为主要原因。
缺硒已成为世界性的畜牧业问题,迄今人们发现约有40种动物具有硒缺乏病。缺硒不仅引起山羊羔白肌疡,而且可引起母羊流产,胎衣不下,受精率降低,妊娠率下降。因此,已引起人们的高度重视。因为许多国家都有缺硒地带,使生长在该地区的饲料缺硒;同时由于工业的不断发展,煤炭等燃烧后有大量的硫散落在土壤中,从而引起土壤中的硫、硒比例失调,使硒缺乏。
缺硒可分为两种情况:一为土壤缺硒;二为条件性硒缺乏。但由于饲料配合不当,长期饲喂缺硒饲料或因阴雨过多,使饲料的硒含量减少所致。此外,本病的发生与含硫氨基酸及维生素A,B,C缺乏等因素有关。
通过大量的研究证明是属于硒与维生素E缺乏引起的一种营养代谢病,特征是心肌和骨骼肌发生变性与坏死。硒是动物机体必需的微量元素和重要的辅酸物质,直接参 与蛋白质的合成和细胞的抗氧化过程。硒是生物膜的组成部分,硒和维生素E都是动物体内 的抗氧化剂,在保护膜不受损害上有重要的作用。缺乏时细胞或亚细胞结构的脂质膜破坏,机体在代谢中产生的内源性过氧化物引起细胞变性、坏死,从而发生白肌样病变,色泽变淡似煮肉样。
临床症状
本病的临床特点是心脏机能衰退(心音混浊,节律不齐,心搏过速,有时可闻缩期杂 音)。急性病例,常突然死亡,死前无任何临床症状,山羊羔有时在白天仅表现沉郁,呻吟,不食,而夜间死亡。生前检查心跳极快,心律不齐,死于心脏麻痹。也有呈慢性经过的,但一般多呈亚急性经过,死亡率可达50%一70%。
病羊羔精神萎顿,食欲减少,常有腹泻,踱行,拱背站立或卧地不起现象。若驱赶运动,则步态僵硬,关节不能伸直,触诊四肢及腰部肌肉感到硬而肿胀且有痛感,骨骼肌弹性降低。心区有压痛,脉搏150-200次/分钟,呼吸90一100次/分钟。病羊羔常发生结膜炎,角膜混浊、软化,终至失明。四肢及胸腹下出现水肿。尿液往往呈红褐色。常由于咬肌及舌肌机能丧失而无法采食,心肌及骨骼肌严重损害时导致死亡。
白肌病病变常发生于半腱肌、半膜肌、股二头肌、背最长肌、臂三头肌及心肌等。发生病损的骨骼肌呈白色条纹或斑块,严重的整个肌肉呈弥漫性黄白色,切面干燥,似鱼肉样外观, 常呈对称性损害。个别病例,变性坏死的肌纤维发生退变,剖面呈线条状,俗称"线猪肉"。心肌的病损主要发生于心内膜、乳头肌及中膈,呈灰白色条纹或斑块,病变可深入心肌 ,使心肌纤维发生变性、坏死及钙化。当心脏受到严重损害时,可见肺水肿及胸腔积液。镜检:肌纤维肿胀、断裂、溶解,为典型透明变性或蜡样坏死,有时坏死的肌纤维可发生钙化。部分肌纤维中细胞核消失。肌纤维之间缺乏血管,结缔组织明显增生,并有较多的淋巴细胞浸润。机体其他脏器和淋巴结均无明显的可见病变。
中兽医认为,羊白肌病主要是营养不良造成,属于“痿症”等范畴。痿病系指外感或内伤,使精血受损,肌肉筋脉失养以致肢体弛缓、软弱无力,甚至日久不用,引起肌肉萎缩或瘫痪的一种病证。痿者萎也,枯萎之义,即指肢体痿弱,肌肉萎缩。凡手足或其他部位的肌肉痿弱无力,弛缓不收者均属痿病范畴。
西医在治疗白肌病方面,可供选择的药物种类有限,治疗方案局限,临床效果不甚明显。并且,长期应用药物残留量较高,消费者食用此类羊肉对其机体免疫力可产生不利影响等。
但是,中医药不仅具有西药的疗效,并且还具有西医所不可及的优势特点。采用传统中医理论为指导,以整体观为基础,根据不同的临床症状采用“异病同治”、“同病异治”的原则,进行辨证论治,治愈率高,无污染,可健脾消食,益气养血, 增强气血的生成及循环,加强羊脏腑正常功能的运行,促进正常肌肉组织的生成,维持肌肉、关节的正常功能等。中药治疗具有见效快、疗效确实、不易产生抗药性、廉价、不易复发、副作用少、畜产品药物残留量低、有利于人畜安全等优点。
发明内容
本发明所要解决的技术问题是在于提供一种用于防治羊白肌病的中药组合物,采用这种中药组合物能够克服现有技术中西药和中药的缺陷,治疗羊白肌病,能够健脾消食,益气养血, 增强气血的生成及循环,加强羊脏腑正常功能的运行,促进正常肌肉组织的生成,维持肌肉、关节的正常功能等。
为了实现上述发明目的,本发明提供了一种用于防治羊白肌病的中药组合物,所述中药组合物包括以下原料药材:陈皮、木香、甘松、神曲、麦芽、鸡矢藤、党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉、菟丝子、沙苑子、海马、女贞子、当归、熟地黄、何首乌、龙眼肉、酸枣仁、野料豆、蚂蝗七、人面子、卷茎蓼和独脚柑。

所述中药组合物的剂型为片剂,其制备方法包括以下步骤:
第一步,将陈皮、木香、甘松、神曲、麦芽和鸡矢藤混合,加相对于混合物3倍~5倍量醇浓度为55%~65%的乙醇回流提取2次~4次,每次2小时~3小时,过滤,滤液合并,回收乙醇,浓缩至70℃相对密度为1.30~1.32的膏体,备用;
第二步,将党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉和菟丝子按比例混合,加相对于混合物3~5倍量水,煮沸2~4小时,过滤,浓缩至70℃相对密度为1.27~1.29的膏体;
第三步,将第一步、第二步获得的膏体混合,55℃~65℃减压真空干燥,得干膏粉;
第四步,将所述的干膏粉与淀粉进行混匀处理,得到膏体混合物;所述的膏体和淀粉的重量比为1∶0.01~0.02;将所述膏体混合物进行制粒处理、干燥处理,得到的膏体混合物颗粒;
第五步,将所述的膏体混合物颗粒与滑石粉、硬脂酸镁依次进行混匀处理、压片处理、包衣处理,即得中药制剂片剂成品;所述的颗粒、滑石粉、硬脂酸镁的重量比为100∶1~3∶0.15~0.3。
所述药物组合物的剂型为散剂,其制备方法包括以下步骤:
将各原料药按重量分数比混合碎成粗粉,再进一步粉碎,过50目筛,使能通过50目筛的细粉量在达到60%,收集通过50目筛的细粉得细粉a;再将不能通过50目筛的粗粉投入多功能提取罐提取两次,第一次加6倍量水煎煮1.5-2小时,第二次加5倍量水煎煮1-2.0小时,合并两次煎液,过滤得滤液;加热浓缩至糊状,放入烘箱95℃烘干后冷却,研磨成细粉b,将细粉a与b混合后制成散剂。
为了更好的实现上述发明目的,本发明提供了一种用于防治羊白肌病的中药组合物的制备方法,当所述中药组合物的剂型为片剂,其制备方法包括以下步骤:
第一步,将陈皮、木香、甘松、神曲、麦芽和鸡矢藤混合,加相对于混合物3倍~5倍量醇浓度为55%~65%的乙醇回流提取2次~4次,每次2小时~3小时,过滤,滤液合并,回收乙醇,浓缩至70℃相对密度为1.30~1.32的膏体,备用;
第二步,将党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉和菟丝子按比例混合,加相对于混合物3~5倍量水,煮沸2~4小时,过滤,浓缩至70℃相对密度为1.27~1.29的膏体;
第三步,将第一步、第二步获得的膏体混合,55℃~65℃减压真空干燥,得干膏粉;

中兽医认为,饮食物的消化和营养物质的吸收、转输,是在脾胃、肝胆、大小肠等多个脏腑共同参与下的一个复杂的生理活动,其中脾起主导作用:脾的运化功能主要依赖脾气升清和脾阳温煦的作用:脾宜升则健。“人纳水谷,脾气化而上升”(《医学三字经·附录·脏腑》),“脾升而善磨”(《四圣心源》),水谷入胃,全赖脾阳为之运化。故“脾有一分之阳,能消一分之水谷;脾有十分之阳,能消十分之水谷”(《医原》)。脾的运化功能,统而言之曰运化水谷,分而言之,则包括运化水谷和运化水液两个方面。
因此,本发明治疗白肌病以健脾消食,益气养血为主要治疗原则,故选用中草药:陈皮、木香、甘松、神曲、麦芽、鸡矢藤、党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉、菟丝子、沙苑子、海马、女贞子、当归、熟地黄、何首乌、龙眼肉、酸枣仁、野料豆、蚂蝗七、人面子、卷茎蓼和独脚柑;优选地,本发明治疗白肌病以清热解毒为辅助治疗原则,还可以包括苣荬菜、蔊菜、漏芦、三叶委陵菜和苦葵鸦葱。
各原料药材的药理如下:
陈皮:性味苦、辛,温。归肺、脾经。具有理气健脾,燥湿化痰的功用。可用于胸脘胀满,食少吐泻,咳嗽痰多。故陈皮有三大类作用,一是导胸中寒邪,二破滞气,三益脾胃。
木香:辛,苦,性温.归脾,大肠,三焦经。行气止痛;调中导滞。主胞胁胀满足,脘腹胀痛,嗳吐泄泻,痢疾后重。用于胸脘胀痛、泻痢后重、食积不消、不思饮食。
甘松:味辛,甘,性温.归脾,胃经。甘松具有芳香行散,温中散寒,理气止痛,醒脾开胃的功效。主治脘腹冷痛胀满,不思饮食,胸痛,牙痛,跌仆伤痛,脚气浮肿,霍乱转筋。
神曲:性味苦,温。归脾;胃经。可健脾消食,理气化湿,解表。治伤食胸痞,腹痛吐泻,痢疾,感冒头痛,小儿伤饥失饱。神曲中有酵母菌。
麦芽:性味甘,平。归脾、胃经、肝经。具有行气消食,健脾开胃,退乳消胀的功效。主治食积不消,脘腹胀痛,脾虚食少,乳汗郁积,乳房胀痛,妇女断乳。
鸡矢藤:性平,味甘、微苦。心;肝;脾;肾经。祛风活血,止痛解毒,消食导滞,除湿消肿。治风湿疼痛,腹泻痢疾,肮腹疼痛,气虚浮肿,头昏食少,肝脾肿大,瘰疬,肠痈,无名肿毒,跌打损伤。
党参:性平,味甘、微酸。归脾、肺经。补中益气,健脾益肺。用于脾肺虚弱,气短心悸,食少便溏,虚喘咳嗽,内热消渴。
黄芪:甘,微温。归肺、脾、肝、肾经。生用固表,无汗能发,有汗能止;炙用补中,益元气,壮脾胃。生血,生肌,排脓内托,疮痈圣药。痘症不起,阳虚无热者宜之。功用固表敛汗,利水消肿,托疮排脓,通调水道。
白术:味甘、苦,性温。归脾、胃经。有补脾利气,燥湿利水,固表止汗,运脾润燥的功效,主治脾失健运、气不化水所致之痞满腹泻,痰饮水肿;中气不足之心烦懒言,崩中漏下,久痢脱肛;脾虚气弱之奏里不固、虚汗自汗等证。
山药:味甘,性平。归肺、脾、肾经。具有健脾补肺、益胃补肾、固肾益精、聪耳明目、助五脏、强筋骨、长志安神、延年益寿的功效,主治脾胃虚弱、倦怠无力、食欲不振、久泄久痢、肺气虚燥、痰喘咳嗽、肾气亏耗、腰膝酸软、下肢痿弱、消渴尿频、遗精早泄、带下白浊、皮肤赤肿、肥胖等证。
刺五加:味辛、苦、微甘,性温。归脾、肾、心经。具有益气健脾,补肾安神,祛风除湿的功效,用于风寒湿痹、腰膝疼痛、筋骨痿软、行动迟缓、体虚羸弱、跌打损伤、骨折、水肿、脚气、阴下湿痒等证。
绞股蓝:味甘、苦,性微寒。归肺经、脾经、肾经。能益气,安神,降血压,清热解毒,止咳祛痰。
红景天:甘,苦,平。归肺、心经。改善睡眠、生血活血、抗脑缺氧、抗疲劳、活血止血、清肺止咳、化淤消肿、解热退烧、滋补元气等功效。
沙棘:味酸;涩;性温。止咳化痰;健胃消食;活血散瘀。主咳嗽痰多;肺脓肿;消化不良;食积腹痛;胃痛;肠炎;闭经;跌打瘀肿。
杜仲:味甘、微辛,性温。归肝、肾经。杜仲具有补肝肾,强筋骨,清除体内垃圾,加强人体细胞物质代谢,防止肌肉骨骼老化,平衡人体血压,分解体内胆固醇,降低体内脂肪,恢复血管弹性,利尿清热,广谱抗菌,兴奋中枢神经,提高白血球药理作用。
肉苁蓉:甘、咸,温。归肾、大肠经。补肾阳,益精血,润肠通便。用于阳痿,不孕,腰膝酸软,筋骨无力,肠燥便秘。
菟丝子:气味辛,甘,平,无毒。归肝、肾、脾经。补肾益精,养肝明目。适用于肝肾不足的腰膝筋骨酸痛,腿脚软弱无力、阳痿遗精、呓语、小便频数、尿有余沥、头晕眼花、视物不清、耳鸣耳聋以及妇女带下、习惯性流产等症。
沙苑子:性温,味甘。归肝、肾经。具有温补肝肾,固精,缩尿,明目的作用,用于肾虚阳痿,遗精早泄,尿频,或白带过多,腰膝酸软,腰痛;肝肾不足,目昏目暗,视力减退等证。
海马:甘温。归肝、肾经。具有补肾壮阳、温通血脉、镇静安神、散结消肿、舒筋活络、止咳平喘之功效。
女贞子:味甘、苦,微寒。归肝、肾经。有强肝益肾,清热明目的作用,主要用于治疗肝肾阴亏,头目失养之头昏耳鸣,阴虚内热等症状。
当归:味甘、辛、苦,性温。归肝、心、脾经。具有补血活血,调经止痛、润汤通便的作用,用于血虚萎黄、眩晕心悸、月经不调、经闭痛经、虚寒腹痛、肠燥便秘、风湿痹痛、跌扑损伤、痈疽疮疡等证
熟地黄:味甘,性微温。归肝、肾经。具有补血滋阴,益精填髓的作用,用于血虚萎黄;眩晕,心悸,失眠,月经不调,崩不止;肝肾阴亏,盗汗,足膝疼痛,腰膝酸软,耳鸣耳聋,头目昏花,遗精阳痿,不育不孕,月经不调,崩漏下血,须发早白,消渴,肾虚喘促等证。
何首乌:味甘、微苦,性平。归心、肝经。具有能养心安神、祛风通络的功效,主治失眠多梦,血虚身痛,风湿痹痛;皮肤瘙痒等证,首乌藤能够活血、通经、扩张血管、促进血液循环、下乳消肿。具有促进造血功能,增强免疫功能,降血脂与抗动脉粥样硬化,保肝。
龙眼肉:甘,温。归心、脾经。养血安神,长智敛汗,开胃益脾。壮阳益气,补脾胃。治妇人产后浮肿,气虚水肿,脾虚泄泻。补益心脾,养血安神。用于气血不足、心悸怔忡、健忘失眠、血虚萎黄。
酸枣仁:甘、酸,性平。归心经,脾经,肝经,胆经。主烦心不得眠,脐上下痛,血转久泄,虚汗烦渴,补中,益肝气,坚筋骨,助阴气,令人肥健。
野料豆:味甘苦咸,气寒,无毒。归肺经,脾经。补益肝肾,祛风解毒。治阴亏目昏,肾虚腰痛,盗汗,筋骨疼痛,产后风痉,小儿疳疾。
蚂蝗七:微苦,凉,无毒。健脾消食,清热除湿。治小儿疳疾,肝炎。
人面子:酸,凉。健脾消食,生津止渴。主治消化不良,食欲不振,热病口渴。外用治烂疮,褥疮。
卷茎蓼:辛;性温。健脾消食。主消化不良;腹泻。
独脚柑:味甘;微苦;性凉。归肝;脾;胃经。健脾消积;清热杀虫。主小儿伤食;疳积黄肿;夜盲;夏季热;腹泻;肝炎。
苣荬菜:味苦;性寒。 清热解毒;利湿排脓;凉血止血。主咽喉肿痛;疮疖肿毒;痔疮;急性菌痢;肠炎;肺脓疡;急性阑尾炎;叶血;衄血、咯血;尿血;便血;崩漏。
蔊菜:味辛;苦;性微温。归肺;肝经。 祛痰止咳;解表散寒;活血解毒;利湿退黄。主咳嗽痰喘;感冒发热;麻疹透发不畅;风湿痹痛;咽喉肿痛;疔疮痈肿;漆疮;经闭;跌打损伤;黄疸;水肿。
漏芦:味苦;性寒。归胃;大肠;肝经。清热解毒;活血通乳。主疮疖肿毒;乳痈;肋腺炎;淋马结结核;痔瘘;疥癣痒疹;目赤肿痛;痢疾;蛔早腹痛;风湿痹痛;闪腰岔气;跌打损伤;产后乳汁不下。
三叶委陵菜:苦,微寒。清热解毒,散瘀止血。治骨结核,口腔炎,瘰疬,跌打损伤,外伤出血。
苦葵鸦葱:苦辛,寒。清热解毒。治疔毒恶疮,近有试用于治疗胃癌,甲状腺癌。
以下采用实施例来详细说明本发明的实施方式,借此对本发明如何应用技术手段来解决技术问题,并达成技术效果的实现过程能充分理解并据以实施。
实施例1 中药组合物1
本发明实施例提供了一种用于防治羊白肌病的中药组合物,由以下重量份数的原料药材制成:取陈皮18重量份、木香27重量份、甘松15重量份、神曲26重量份、麦芽11重量份、鸡矢藤30重量份、党参36重量份、黄芪33重量份、白术14重量份、山药26重量份、刺五加8重量份、绞股蓝28重量份、红景天23重量份、沙棘29重量份、杜仲14重量份、肉苁蓉23重量份、菟丝子33重量份、沙苑子20重量份、海马15重量份、女贞子15重量份、当归24重量份、熟地黄34重量份、何首乌9重量份、龙眼肉28重量份、酸枣仁18重量份、野料豆23重量份、蚂蝗七9重量份、人面子20重量份、卷茎蓼12重量份和独脚柑18重量份。
其制备方法包括以下步骤:
第一步,将陈皮、木香、甘松、神曲、麦芽和鸡矢藤混合,加相对于混合物3倍量醇浓度为65%的乙醇回流提取3次,每次2小时,过滤,滤液合并,回收乙醇,浓缩至70℃相对密度为1.30的膏体,备用;
第二步,将党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉和菟丝子按比例混合,加相对于混合物4倍量水,煮沸2小时,过滤,浓缩至70℃相对密度为1.27的膏体;
第三步,将第一步、第二步获得的膏体混合, 65℃减压真空干燥,得干膏粉;
第四步,将干膏粉与淀粉进行混匀处理,得到膏体混合物;膏体和淀粉的重量比为1∶0.01;将膏体混合物进行制粒处理、干燥处理,得到的膏体混合物颗粒;
第五步,将膏体混合物颗粒与滑石粉、硬脂酸镁依次进行混匀处理、压片处理、包衣处理,即得中药制剂片剂成品;颗粒、滑石粉、硬脂酸镁的重量比为100∶1∶0.15。
实施例2中药组合物2
本发明实施例提供了一种用于防治羊白肌病的中药组合物,由以下重量份数的原料药材制成:取陈皮18重量份、木香27重量份、甘松15重量份、神曲26重量份、麦芽11重量份、鸡矢藤30重量份、党参36重量份、黄芪33重量份、白术14重量份、山药26重量份、刺五加8重量份、绞股蓝28重量份、红景天23重量份、沙棘29重量份、杜仲14重量份、肉苁蓉23重量份、菟丝子33重量份、沙苑子20重量份、海马15重量份、女贞子15重量份、当归24重量份、熟地黄34重量份、何首乌9重量份、龙眼肉28重量份、酸枣仁18重量份、野料豆23重量份、蚂蝗七9重量份、人面子20重量份、卷茎蓼12重量份、独脚柑18重量份、苣荬菜8重量份、蔊菜6重量份、漏芦15重量份、三叶委陵菜7重量份和苦葵鸦葱12重量份。
其制备方法如下:
将各原料药按重量分数比混合碎成粗粉,再进一步粉碎,过50目筛,使能通过50目筛的细粉量在达到60%,收集通过50目筛的细粉得细粉a;再将不能通过50目筛的粗粉投入多功能提取罐提取两次,第一次加6倍量水煎煮1.5小时,第二次加5倍量水煎煮1小时,合并两次煎液,过滤得滤液;加热浓缩至糊状,放入烘箱95℃烘干后冷却,研磨成细粉b,将细粉a与b混合后制成散剂。
本发明的中药组合物的毒性试验
取陈皮18g、木香27g、甘松15g、神曲26g、麦芽11g、鸡矢藤30g、党参36g、黄芪33g、白术14g、山药26g、刺五加8g、绞股蓝28g、红景天23g、沙棘29g、杜仲14g、肉苁蓉23g、菟丝子33g、沙苑子20g、海马15g、女贞子15g、当归24g、熟地黄34g、何首乌9g、龙眼肉28g、酸枣仁18g、野料豆23g、蚂蝗七9g、人面子20g、卷茎蓼12g和独脚柑18g;混合均匀后,粉碎,向粉碎后的混合物中加1200g的水,煎煮2.5小时,过滤,滤渣中加入500g的水再次煎煮1.5小时,过滤,将两次滤液混合,减压浓缩至生药浓度为3g/ml的煎剂。
选用SD大鼠,给予不同浓度(18.0、6.0、2.0g生药/kg)的本发明的中药添加剂的汤剂,每天灌胃一次,连续90天,末次给药后24小时各组活杀1/2动物(雌雄各半),其余1/2动物继续观察2周后活杀。试验期间观察动物的外 观、一般行为、摄食量、体重变化,给药后90天和停药2周进行血液学(RBC、 HB、网织红细胞、PLT、CT、WBC及分类)和血液生化(AST、ALT、ALP、 Glu、BUN、Crca、TP、T.BIL、ALB、CHOL)、尿液生化、脏器系数、病理 组织学等指标检查。试验结果表明:本发明的中药添加剂在高、中、低剂量组动物一般状态良好,外观体征、行为活动、进食量和体重增长均无异常变化; 三个剂量组及对照组血液学检查、血液生化学、尿液生化检查均在正常范围,组间无显著差异;各组主要脏器组织病理学检查未见明显异常。上述指标停药2周后也未见改变。本试验用药剂量分别为临床用药剂量的180、60、20倍,根据试验结果本发明的中药制剂在高、中、低三个剂量(18.0、6.0、2.0g 生药/kg)连续90天给药对大鼠无明显影响,无明确的毒性靶器官和敏感指标,恢复期观察也未见延迟性毒性反应,提示本发明的中药制剂临床应用的剂量安全性较高。
本发明的中药组合物的毒性试验
取陈皮18g、木香27g、甘松15g、神曲26g、麦芽11g、鸡矢藤30g、党参36g、黄芪33g、白术14g、山药26g、刺五加8g、绞股蓝28g、红景天23g、沙棘29g、杜仲14g、肉苁蓉23g、菟丝子33g、沙苑子20g、海马15g、女贞子15g、当归24g、熟地黄34g、何首乌9g、龙眼肉28g、酸枣仁18g、野料豆23g、蚂蝗七9g、人面子20g、卷茎蓼12g、独脚柑18g、苣荬菜8g、蔊菜6g、漏芦15g、三叶委陵菜7g和苦葵鸦葱12g;混合均匀后,粉碎,向粉碎后的混合物中加1200g的水,煎煮2.5小时,过滤,滤渣中加入500g的水再次煎煮1.5小时,过滤,将两次滤液混合,减压浓缩至生药浓度为3g/ml的煎剂。

对比试验
(1)供试动物
选取新疆奇台县某白肌病高发的山羊养殖场确诊后的病羊中随机选取150头2月龄病羊作为治疗对比试验对象;另从新疆奇台县某羊养殖场中随机选取180头健康妊娠羊作为预防对比试验对象。
(2)预防对比实验:
将随机挑选的180头妊娠羊随机分成3组(实施例组1、实施例组2和对照组),其中,对照组使用传统羊饲料饲喂妊娠羊,喂母羊9个月,生产的仔羊母乳喂养2月后改为传统羊饲料饲喂仔羊;实施例组1使用1000kg饲料中添加5kg实施例1中的中药组合物1饲喂妊娠羊,喂母羊9个月,生产的仔羊母乳喂养2月后改为使用1000kg饲料中添加5kg实施例1中的中药组合物1饲喂仔羊;实施例组2使用1000kg饲料中添加5kg实施例2中的中药组合物2饲喂妊娠羊,喂母羊9个月,生产的仔羊母乳喂养2月后改为使用1000kg饲料中添加5kg实施例2中的中药组合物2饲喂仔羊,观察记录各组仔羊的发病率及死亡率,结果如表1所示。

从表1的数据可知:本发明含有中药组合物的饲料对羊白肌病有明显的预防效果。
(4)治疗对比试验:
将随机选取的150只2月龄病羊随机分成3组(中药治疗组、西药治疗组、空白对照组),用不同药进行治疗,其中,中药治疗组选用实施例2的中药组合物2,药量为每日0.6g/kg,连续服用4天;西药治疗组选用硒酸钠维生素E注射液肌肉注射,用量为每只3-5毫升,每天1次,连续4天;空白对照组不喂任何药物。观察记录各组羊的治愈率和死亡率,结果如表2所示。

从表2的数据可知:本发明中药组合物对羊白肌病治疗效果明显优于西药治疗组,降低死亡率,提高治愈率。
2012年冬季,邯郸市某养殖场羔羊精神不振,食欲减少,被毛粗乱,背拱起,四肢无力,颈部强直,偏向一侧,轻者走路摇摆,关节僵直,重则站立不稳或举步跌倒,逐渐消瘦贫血,心跳加快,节律不齐,第一音增强,呼吸频数,听诊有罗音,尿频而量多,呈红褐色,羔羊一般多在母羊出牧前或归牧后,因奔跑寻找母羊吃奶而突然倒地死亡。
根据剖检的6个仔山羊病例来看,最显著而一致的病变为心脏极度扩张,心肌厚薄不均,颜色淡。心肌变性,心肌和乳头肌周围有灰黄色条纹,状似虎斑。骨骼肌变性,尤其是前后肢肌和背最长肌变性比较明显,肌纤维粗糙,颜色淡白,其中夹杂着颗粒性增生物,并有淤血小点。肠系膜淋巴结肿胀,柔软,切面多汁,压之有大量乳白色液体流出;切面有小粒状突出物。第四胃发炎、出血;十二指肠、空肠、回肠和部分盲肠黏膜呈紫红色,充血或出血,其内容物呈红色粥状。大部分病例的肠壁滤泡肿胀,确诊为羊白肌病。
立即对出现症状的20只2月龄以上仔羊和30只未出现症状的妊娠母羊,在1000kg饲料中添加5kg实施例2中的中药组合物2饲喂羊,均喂养7天;第3天20只患病仔羊症状明显改善,继续饲喂4天痊愈;跟踪观察,30只妊娠母羊所生产的仔羊未出现羊白肌病。

6.根据权利要求1-5任一项所述的用于防治羊白肌病的中药组合物,其特征在于,所述中药组合物中各原料药材的重量份数为:陈皮18重量份、木香27重量份、甘松15重量份、神曲26重量份、麦芽11重量份、鸡矢藤30重量份、党参36重量份、黄芪33重量份、白术14重量份、山药26重量份、刺五加8重量份、绞股蓝28重量份、红景天23重量份、沙棘29重量份、杜仲14重量份、肉苁蓉23重量份、菟丝子33重量份、沙苑子20重量份、海马15重量份、女贞子15重量份、当归24重量份、熟地黄34重量份、何首乌9重量份、龙眼肉28重量份、酸枣仁18重量份、野料豆23重量份、蚂蝗七9重量份、人面子20重量份、卷茎蓼12重量份、独脚柑18重量份、苣荬菜8重量份、蔊菜6重量份、漏芦15重量份、三叶委陵菜7重量份和苦葵鸦葱12重量份。
7.根据权利要求1-6任一项所述的防治羊白肌病的中药组合物,其特征在于,所述中药组合物的剂型为片剂,其制备方法包括以下步骤:
第一步,将陈皮、木香、甘松、神曲、麦芽和鸡矢藤混合,加相对于混合物3倍~5倍量醇浓度为55%~65%的乙醇回流提取2次~4次,每次2小时~3小时,过滤,滤液合并,回收乙醇,浓缩至70℃相对密度为1.30~1.32的膏体,备用;
第二步,将党参、黄芪、白术、山药、刺五加、绞股蓝、红景天、沙棘、杜仲、肉苁蓉和菟丝子按比例混合,加相对于混合物3~5倍量水,煮沸2~4小时,过滤,浓缩至70℃相对密度为1.27~1.29的膏体;
第三步,将第一步、第二步获得的膏体混合,55℃~65℃减压真空干燥,得干膏粉;
第四步,将所述的干膏粉与淀粉进行混匀处理,得到膏体混合物;所述的膏体和淀粉的重量比为1∶0.01~0.02;将所述膏体混合物进行制粒处理、干燥处理,得到的膏体混合物颗粒;
第五步,将所述的膏体混合物颗粒与滑石粉、硬脂酸镁依次进行混匀处理、压片处理、包衣处理,即得中药制剂片剂成品;所述的颗粒、滑石粉、硬脂酸镁的重量比为100∶1~3∶0.15~0.3。
8.根据权利要求1-6任一项所述的用于防治羊白肌病的中药组合物,其特征在于,所述药物组合物的剂型为散剂,其制备方法包括以下步骤:
将各原料药按重量分数比混合碎成粗粉,再进一步粉碎,过50目筛,使能通过50目筛的细粉量在达到60%,收集通过50目筛的细粉得细粉a;再将不能通过50目筛的粗粉投入多功能提取罐提取两次,第一次加6倍量水煎煮1.5-2小时,第二次加5倍量水煎煮1-2.0小时,合并两次煎液,过滤得滤液;加热浓缩至糊状,放入烘箱95℃烘干后冷却,研磨成细粉b,将细粉a与b混合后制成散剂。
9.一种如权利要求1-6任一项所述的用于防治羊白肌病的中药组合物的制备方法,其特征在于,所述中药组合物的剂型为片剂,其制备方法包括以下步骤:
第一步,将陈皮、木香、甘松、神曲、麦芽和鸡矢藤混合,加相对于混合物3倍~5倍量醇浓度为55%~65%的乙醇回流提取2次~4次,每次2小时~3小时,过滤,滤液合并,回收乙醇,浓缩至70℃相对密度为1.30~1.32的膏体,备用;
