本发明公开了一种治疗奶牛缺乳症的药物组合物及其制备方法,其药物组合物包括以下原料药材:千屈草、木香、荃皮、乌药、甘松、倒水莲、九香虫、大荨麻、青皮、大枣、柴胡、鸡矢藤、香附、苤蓝、灵芝、娑罗子、菱蒂、郁金、万丈深、玫瑰花、乌蔹莓、莱菔子、白术、武靴藤、山楂、曼陀罗子、麦芽、谷芽、葛蕈、党参、芜菁、山药和薜荔果。本发明的有益效果是:本发明的药物组合物以健脾理气为主,以补气养血为辅,提高奶牛的免疫力,还能够提高泌乳量、乳脂率、乳蛋白、乳糖和乳中干物质,促进乳房饱满和乳头正常发育,乳汁饱满,不再有食欲骤减或不食的现象;还具有毒副作用小、疗效显著的优势,而且还能够很好的保证奶牛乳腺及其身体其他器官的正常生长。

技术领域
本发明涉及兽药领域,尤其涉及一种治疗奶牛缺乳症的药物组合物及其制备方法。
背景技术
奶牛场常出现青年牛到达配种年龄仍然不发情、成奶牛产后长期不发情、产量锐减等现象,有些牛因此而淘汰造成不小的损失。母畜年幼,乳腺发育不完善而缺奶。表现为乳房紧小,乳头短缩,外观幼稚,乳汁缺乏,拒绝哺乳。奶牛产后缺乳,是由于营养失调、体瘦气虚,或体壮腰好,或产后半虚半热,或产后感染疾病痊愈后等多种原因引起的常见的疾病。
一般奶牛产后缺乳的主要原因有:①饲养管理不善或饲喂霉烂变质饲料造成奶牛消瘦 ;②严寒或炎热过度刺激 ;③乳腺发育不良或腺体机能紊乱 ;④年老或过度劳役 ,致使乳腺机能减退或停止 ;⑤早配早产、产后热或乳腺炎 ;⑥产乳奶牛工期舍饲料造成运动不足或其它疾病等。乳期间受到某种因素影响而致乳腺机能紊乱,造成泌乳量减少或无乳。临床上多发于营养不良、老龄及初产奶牛。病因:营养不良是导致奶牛缺乳的主要原因。如炎夏高温季节奶牛食欲骤减或不食,冬春奶牛妊娠后期舍饲缺乏青饲料和优质干草、精料供给不足等均能造成奶牛产后缺乳。
目前,西医治疗奶牛缺乳是大量应用抗生素,垂体素等化学合成药物。西药相对于祖国传统中药而言,西医所用的药物,一般用化学合成方法制成或从天然产物提制而成。疗程较长,药物残留量较大,效果不理想,对奶牛和消费者的健康具有一定程度的影响。
发明内容
本发明所要解决的技术问题是在于,提供一种具有治疗奶牛缺乳症的药物组合物,能克服现有技术中的中药和西药的各种缺陷,以健脾理气为主,以补气养血为辅,提高奶牛的免疫力,还能够提高泌乳量、乳脂率、乳蛋白、乳糖和乳中干物质,促进乳房饱满和乳头正常发育,乳汁饱满,不再有食欲骤减或不食的现象;还具有毒副作用小、疗效显著的优势,而且还能够很好的保证奶牛乳腺及其身体其他器官的正常生长。

为了更好的实现上述发明目的,本发明提供了一种具有治疗奶牛缺乳症的药物组合物的制备方法,当药物组合物的剂型为粉针剂时,其制备方法包括以下步骤:
所述药物组合物中各原料药材的剂型为粉针剂,其制备方法包括以下步骤:
第一步,将所述原料药材按比例混合,放入超微粉碎机中粉碎成0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第二步,在第一步获得的混合粉料中加入相对于所述混合粉料的质量3~6倍的醇浓度为80%~95%的乙醇,搅拌溶解获得乙醇溶液,将乙醇溶液在5℃~10℃的条件下静置24~48小时,采用渗漉法以每分钟1~2.5ml的速度缓缓渗漉,收集渗漉液,浓缩并干燥,并在此放入超微粉碎机中粉碎30~60分钟,获得0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第三步,取第二步获得的混合粉料10g~20g,加入丙二醇30g~50g,加200ml注射用水,搅拌使其溶解,进一步加注射用水至1500ml,再加入1g~1.5g针用活性炭,充分搅拌30~60分钟;脱炭过滤;用0.22~0.27μm微孔滤膜过滤;冷冻干燥得无菌粉末,分装灌封。
当药物组合物的剂型为散剂时,其制备方法包括以下步骤:
第一步,将香附、苤蓝、灵芝、娑罗子、菱蒂和郁金按所述比例混合,用相对于获得的混合物质量的3~5倍的醇浓度为80%~90%的乙醇溶解,加热回流3~5小时后提取,过滤,得第一过滤液;将滤渣再次用相对于获得的混合物质量的2~4倍的醇浓度85%~95%的乙醇溶解,加热回流1~2小时后提取,过滤,得第二过滤液;合并第一过滤液和第二过滤液,减压回收乙醇并浓缩至药液浓度为0.6~0.9g生药/mL,经体积为5L的大孔吸附树脂柱洗脱,先用5倍树脂柱体积的去离子水或蒸馏水洗脱,再用3倍树脂柱体积的醇浓度为90%~95%乙醇洗脱,收集乙醇洗脱液,减压浓缩除去乙醇,干燥并粉碎成粉末;
第二步,将乌药、甘松、倒水莲、九香虫和大荨麻按所述比例混合,加相对于获得的混合物质量的2~4倍的醇浓度85%~95%的乙醇,加热回流提取1~2次,提取液合并,用管式离心机离心除杂,将除杂后的提取液于60℃~80℃的条件下减压浓缩至60℃时相对密度为1.21~1.25的膏体,将浓缩后的膏体用喷雾干燥器干燥成粉末;
第三步,将剩余原料药材按所述比例混合,加相对于获得的混合物质量的2~4倍的水溶解,加热煎煮3~5小时,过滤;将滤渣再次用相对于获得的混合物质量的1~2.5倍的水溶解,再次加热煎煮1~1.5小时,过滤;合并两次的过滤液,减压浓缩除去水分,干燥并粉碎成粉末;
第四步,将第一步、第二步和第三步获得的粉末混合,随后过筛成80~120目的细粉,将获得的细粉投入混合搅拌机中搅拌,即获得散剂。

本发明的药物组合物遵循健脾理气,补气养血的原则,提供了一种具有治疗奶牛缺乳症的药物组合物,包括以下原料药材:千屈草、木香、荃皮、乌药、甘松、倒水莲、九香虫、大荨麻、青皮、大枣、柴胡、鸡矢藤、香附、苤蓝、灵芝、娑罗子、菱蒂、郁金、万丈深、玫瑰花、山楂、曼陀罗子、麦芽、谷芽、葛蕈、党参、芜菁、山药和薜荔果;优选地,还可以包括破骨风、牛耳大黄、蚯蚓、匍伏堇和蜂房。
上述各原料药材的药理特性如下:
木香:辛,苦,性温;归脾,大肠,三焦经。《本草纲目》云:“木香,乃三焦气分之药,能升降诸气。诸气膹郁,皆属于肺,故上焦气滞用之者,乃金郁则泄之也;中气不运,皆属于脾,故中焦气滞宜之者,脾胃喜芳香也;大肠气滞则后重,膀胱气不化则癃淋,肝气郁则为痛,故下焦气滞者宜之,乃塞者通之也”。功效:行气,止痛,健脾,消食。主治:行气止痛;调中导滞。主胞胁胀满足,脘腹胀痛,嗳吐泄泻,痢疾后重。用于胸脘胀痛、泻痢后重、食积不消、不思饮食。木香辛温香散,能升能降,通理三焦之气,尤其善行胃肠之气而止痛,兼有健脾消食之功,凡脾胃大肠气滞所致诸证均为常用之品。
乌药:性味辛,温。入胃,肾经。舒气,温中,散寒,止痛。1.寒凝气滞之胸腹诸痛证。该品味辛行散,性温祛寒,入肺而宣通,入脾而宽中,故能行气散寒止痛。治胸腹胁肋闷痛。2.尿频,遗尿。该品辛散温通,入肾与膀胱而温肾散寒,缩尿止遗。
甘松:味辛,甘,性温;归脾,胃经。《本草纲目》:“甘松,芳香能开脾郁,少加入脾胃药中,甚醒脾气”。《日华子本草》:“治心腹胀,下气。”甘松具有芳香行散,温中散寒,理气止痛,醒脾开胃的功效。主治脘腹冷痛胀满,不思饮食,胸痛,牙痛,跌仆伤痛,脚气浮肿,霍乱转筋。宽叶甘松挥发油有微弱的抗菌、驱风及解痉作用,对皮肤、粘膜无局部刺激性。
九香虫:性温,味咸;归肝、脾、肾经。《本草新编》:“九香虫,虫中之至佳者、入丸散中,以扶衰弱最宜。但不宜入于汤剂)以其性滑,恐动大便耳。九香虫亦兴阳之物,于胃寒胀痛,肝胃气痛,肾虚阳痿,腰膝酸痛。九香虫对金黄色葡萄球菌、伤寒杆菌、副伤寒杆菌、福氏痢疾杆菌有较强的抗菌作用;并有促进机体新陈代谢作用。
青皮:性味苦辛、温。入肝、胆、胃经。可疏肝破气、消积化滞。用于胸肋脘胀痛,乳痈、疝痛,食积气滞。现代药理实验研究表明:青皮注射液能降低离体豚鼠胃、肠、胆囊及小鼠子宫的紧张性收缩,并使膀胱平滑肌兴奋。对乙酰胆碱引起的豚鼠离体胃、肠及家兔在体胃平滑肌以及氨甲酰胆碱引起的胆囊收缩,有显著的解痉作用。对组胺引起的豚鼠离体肠和水杨酸毒扁豆碱引起的家兔在体肠紧张性收缩,有显著的抑制作用。能对抗脑垂体后叶素引起的小鼠子宫紧张性收缩。对豚鼠在体膀胱及家兔主动脉条的兴奋效应,主要通过兴奋肾上腺素能α受体,且对主动脉的兴奋作用比去甲肾上腺素弱。能显著拮抗豚鼠肺因组胺引起的灌流量减少,并能解除组胺对支气管链的痉挛作用。也能显著增加大鼠的胆汁流量,并使胆道张力增大。
柴胡:性微寒、味苦、辛、归肝经、胆经,《本草图经》载:“(柴胡)今关、陕、江湖间,近道皆有之,以银州者为胜。二月生苗,甚香,茎青紫,叶似竹叶稍紫……七月开黄花……根赤色,似前胡而强。芦头有赤毛如鼠尾,独窠长者好。二月八月采根。”具疏肝利胆、疏气解郁、散火之功效。功能:透表泄热,疏肝解郁,升举阳气。主治:治感冒发热、寒热虚劳发热。小儿痘疹余热,面黄肌瘦腹部膨大,疟疾,肝郁气滞,胸肋胀痛,脱肛,子宫脱落,月经不调。柴胡挥发油对伤寒、副伤寒疫苗、大肠杆菌液、发酵牛奶、酵母等所致发热有明显解热作用,且能使动物正常体温下降。对多种致炎剂所致踝关节肿和结缔组织增生性炎症均有抑制作用。
香附:性味辛微苦甘,平。入肝、三焦经。《本草纲目》曰:“香附之气平而不寒,香而能窜,其味多辛能散,微苦能降,微甘能和”。《本草衍义补遗》:“香附子,必用童便浸,凡血气药必用之,引至气分而生血,此阳生阴长之义也。”可理气解郁;调经止痛;安胎。主胁肋胀痛;乳房胀育;疝气疼痛;月经不调;脘腹痞满疼痛;嗳气吞酸,呕恶;经行腹痛;崩漏带下;胎动不安。用于肝郁气滞,胸、胁、脘腹胀痛,消化不良,胸脘痞闷,寒疝腹痛,乳房胀痛,月经不调,经闭痛经。乙醇提取物不影响肾上腺素和乙酰胆碱对血压的作用,但能部分阻断组织胺的作用。
苤蓝:性味: 甘辛,凉。主治:治小便淋浊,大便下血,肿毒,脑漏。
娑罗子:性温,味甘;归肝、胃经。《纲目拾遗》:“葛祖遗方,治心胃寒痛,虫痛。宽中下气,治胃脘肝膈膨胀,疳积疟痢,吐血劳伤,平胃通络。”理气宽中,和胃止痛。用于胸腹胀闷、胃脘疼痛。娑罗子皂苷治疗老年支气管哮喘,能迅速有效地改善临床症状,增加呼吸流速峰值,娑罗子提取物娑罗子皂苷16-18mg加入10%葡萄糖液中静滴治疗脑外伤、良性颅内高压症、脑手术后,有效率达91%。可用于治疗心绞痛、高脂血症,本品既能疏肝解郁以行滞,又能理气宽中以和胃。治疗肝胃气滞之胸闷胁痛、脘腹胀痛等证。
郁金:辛、苦,寒。归肝、心、肺经。《唐本草》:“主血积,下气,生肌,止血,破恶血,血淋,尿血,金疮。”《本草纲目》:“治血气心腹痛,产后败血冲心欲死,失心颠狂。”活血止痛,行气解郁,清心凉血,利胆退黄。
玫瑰花:性温,味甘微苦,归肝脾经。《本草纲目拾遗》:“和血,行血,理气。治风痹。”《本草再新》:“舒肝胆之郁气,健脾降火。治腹中冷痛,胃脘积寒,兼能破血。”利气、行血,治风痹,散疲止痛。
莱菔子:消食除胀,降气化痰。主治食积气滞,脘腹胀满,嗳气,下痢后重,咳嗽痰多,喘促胸满。用于饮食停滞、脘腹胀痛、大便秘结、积滞泻痢、痰壅喘咳。入脾、胃、肺经,能消食除胀,功效显著,有“冲墙倒壁”之称。
白术:味甘、苦,性温,入脾、胃经,有补脾利气,燥湿利水,固表止汗,运脾润燥的功效,主治脾失健运、气不化水所致之痞满腹泻,痰饮水肿;中气不足之心烦懒言,崩中漏下,久痢脱肛;脾虚气弱之奏里不固、虚汗自汗等证,白术能促进胃液分泌,增强小肠的吸收作用,对胃肠功能有调节作用。具有明显而持久的利尿作用。
山楂:味酸、甘,性微温,归脾、胃、肝经,用于消食健胃,行气散瘀,现代研究山楂含糖类、蛋白质、脂肪、维生素C、胡萝卜素、淀粉、苹果酸、枸橼酸、钙和铁等物质,具有降血脂、血压、强心和抗心律不齐等作用。山楂内的黄酮类化合物牡荆素,是一种抗癌作用较强的药物,山楂提取物对癌细胞体内生长、增殖和浸润转移均有一定的抑制作用。山楂能入脾胃消积滞,散宿血,故治水痢及产妇腹中块痛也。大抵其功长于化饮食,健脾胃,行结气,消瘀血,故小儿产妇宜多食之。能防治心血管疾病,具有扩张血管、增加冠脉血流量、改善心脏活力、兴奋中枢神经系统、降低血压和胆固醇、软化血管及利尿和镇静作用;有强心作用,对老年性心脏病有益处;能开胃消食,对消肉食积滞作用更好;有活血化淤的功效,有助于解除局部淤血状态,对跌打损伤有辅助疗效;对子宫有收缩作用,在孕妇临产时有催生之效,并能促进产后子宫复原;所含黄酮类和维生素C、胡萝卜素等物质能阻断,减少自由基的生成,增强机体的免疫力,有防衰老、抗癌的作用;山楂中有平喘化痰、抑制细菌、治疗腹痛腹泻的成分。
麦芽:性味甘,平。归脾、胃经、肝经。《日华子本草》:“温中,下气,开胃,止霍乱,除烦,消痰,破症结,能催生落胎。”《滇南本草》:“宽中,下气,止呕吐,消宿食,止吞酸吐酸,止泻,消胃宽膈,并治妇人奶乳不收,乳汁不止。”具有行气消食,健脾开胃,退乳消胀的功效。主治食积不消,脘腹胀痛,脾虚食少,乳汗郁积,乳房胀痛,妇女断乳。麦芽可用于肝郁气滞,胸胁胀闷,及肝脾不各,嗳气少食等证。
谷芽:甘,平。归脾、胃经。《本经逢原》:“谷芽,启脾进食,宽中消谷,而能补中,不似麦芽之克削也。”《中药材手册》:“治脾虚,心胃痛,胀满,热毒下痢,烦渴,消瘦。”《本草纲目》:“快脾开胃,下气和中,消食化积。”消食和中,健脾开胃。用于食积不消,腹胀口臭,脾胃虚弱,不饥食少。炒谷芽偏于消食,用于不饥食少。焦谷芽善化积滞,用于积滞不消。该品消食和中,作用和缓,助消化而不伤胃气。
党参:性平,味甘、微酸;归脾、肺经。补中益气,健脾益肺。用于脾肺虚弱,气短心悸,食少便溏,虚喘咳嗽,内热消渴。该品为临床常用的补气药,功能补脾益肺,效近人参而为较弱,适用于各种气虚不足者。所含皂苷、菊糖、微量生物碱、淀粉等对人体多脏器有不同程度的强壮作用,能提高人体的适应性。有抗溃疡,能增加肠张力、调节胃运动,在调理肠胃制剂中重用党参的肠胃调神剂对慢性腹泻,溃疡性结肠炎,胃窦炎,慢性萎缩性胃炎等颇有效验。
山药:味甘,性平,入肺、脾、肾经,具有健脾补肺、益胃补肾、固肾益精、聪耳明目、助五脏、强筋骨、长志安神、延年益寿的功效,主治脾胃虚弱、倦怠无力、食欲不振、久泄久痢、肺气虚燥、痰喘咳嗽、肾气亏耗、腰膝酸软、下肢痿弱、消渴尿频、遗精早泄、带下白浊、皮肤赤肿、肥胖等证。麸炒山药补脾健胃。用于脾虚食少,泄泻便溏,白带过多。该品养阴能助湿,所以湿盛中满、或有积滞、有实邪者不宜。需要提醒大家的是,食用山药一般无明显禁忌症,但因其有收敛作用,所以患感冒、大便燥结者及肠胃积滞者忌用。
薜荔果:酸,平。《日华子本草》:“治发背,游风,风疹,妇人血经不匀及难产。”《本草纲目》:“固精消肿,散毒止血,下乳,治久痢肠痔,心痛阴颓。”补肾固精,通乳,活血消肿,解毒。治遗精、阳痿、久痢脱肛、乳汁不通等病症。用于肾亏腰酸,阳痿遗精等症。本品性味酸平,有补肾固精作用,用于肾亏腰瘦,阳痿遗精等症,本品又有活血消肿解毒的功效,对于痈肿初起之症。
大枣:味甘,性温,入脾、胃经。具有补气健脾、养血安神、调和营卫、缓和药性的作用,用于脾胃虚弱,体倦乏力,食少便溏,气血不足,头痛发热,汗出热风等证,大枣具有增强肌力的作用,具有增加血清总蛋白和白蛋白的功能。红枣所含有的环磷酸腺苷,是人体细胞能量代谢的必需成分,能够增强肌力、消除疲劳、扩张血管、增加心肌收缩力、改善心肌营养,对防治心血管系统疾病有良好的作用;红枣对慢性肝炎、肝硬化、贫血、过敏性紫癜等病症有较好疗效;红枣含有三萜类化合物及环磷酸腺苷,有较强的抗癌、抗过敏作用。大枣能润心肺、止咳、补五脏、治虚损、除肠胃癖气,还能安中养脾、平胃气、通九窍、助十二经等。
千屈草:性味:苦,凉。主治:清热解毒,凉血止血。用于肠炎,痢疾,便血;外用治外伤出血。
荃皮:性味:微苦、涩,温。主治:活血祛瘀,生肌,收敛。主治跌打损伤,瘀血内滞,骨折,刀伤。
倒水莲:性味:苦、涩,寒。主治:清热,燥湿,止痢。用于湿热黄疸,腹痛泻痢,疟疾寒热,目赤肿痛。
大荨麻:性味:苦、辛,温。主治:祛风除湿,活血止痛止痒。主治风湿麻木,关节痛,劳伤疼痛,疝痛,水肿,毒蛇咬伤,皮肤瘙痒,小儿惊风吐乳,妇女产后体虚。
鸡矢藤:性味:甘、微苦,平。主治:祛风利湿,消食化积,止咳,止痛。用于风湿筋骨痛,跌打损伤,外伤性疼痛,肝胆、胃肠绞痛,黄疸型肝炎,肠炎,痢疾,消化不良,小儿疳积,肺结核咯血,支气管炎,放射反应引起的白血球减少症,农药中毒;外用治皮炎,湿疹,疮疡肿毒。
灵芝:性味:性温,味淡。主治:滋补强壮。用于健脑、消炎、利尿、益肾。
菱蒂:性味:味微苦;性平。主治:解毒散溃疡。主胃溃疡;疣赘。
鹅臎:性味:辛;温。归经:脾;胃;肾经。主治:补肝。主《日华子》:“治聤耳及聋,纳之;亦疗手足皴。”
万丈深:性味:苦、微甘,平。主治:健脾消积,清热解毒,下乳。用于小儿疳积,乳汁不足,支气管炎,肺炎,肝炎,白带,肠风下血,筋骨疼痛,跌打损伤,疮疖痈肿。
乌蔹莓:性味:苦、酸,寒。主治:解毒消肿,活血散瘀,利尿,止血。用于咽喉肿痛、目翳,咯血,血尿,痢疾;外用治痈肿,丹毒,腮腺炎,跌打损伤,毒蛇咬伤。
武靴藤:性味:《实用中草药》:"性平,味苦。"主治:①《实用中草药》:"根,消肿解毒,清热凉血,治多发性脓肿,深部脓肿,乳腺炎,痈疮肿毒。"②《常用中草药彩色图谱》:"嫩枝叶,止痛,生肌,消肿。治枪弹伤。杀虱。"
曼陀罗子:性味;辛苦,温。归经:《四川中药志》:"入肝、脾经。"主治:平喘,祛风,止痛。治喘咳,惊痫,风寒湿痹,泻痢,脱肛,跌打损伤。①《分类草药性》:"治跌打损伤,遂瘀血,通经络。"②《贵州民间方药集》:"熏治牙痛。"③《四川中药志》:"能祛风胜湿,定喘消肿。治风寒湿痹,关节肿痛,惊痫脱肛,跌打损伤及泻痢等症。"
葛蕈:性味:苦甘,凉。①《纲目》:"苦甘,无毒。"②《纲目拾遗》:"性凉。"③《四川中药志》:"味涩苦,性寒,无毒。"主治:清热,解毒,醒酒。治风热癍疹,肺热咳嗽吐血,血崩,痔疮。①《纲目》:"醒酒,治酒积。"②《纲目拾遗》:"解肌热,散风火及阳明风热癍疹。"③《四川中药志》:"清肺热,解热毒。治咳嗽吐血,血崩及痔疮肿痛。"④《湖南药物志》:"指生蛇头疔,小儿阴茎肿,捣烂敷患处。"
芜菁:性味:苦辛甘,平。①《别录》:"味苦温,无毒。"②《饮膳正要》:"味甘,平,无毒。"③《纲目》:"辛甘苦。"主治:开胃下气,利湿解毒。治食积不化,黄疸,消渴,热毒风肿,疔疮,乳痈。①《千金·食治》:"主消风热毒肿。"②《食疗本草》:"下气,治黄疸,利小便。根:主消渴,治热毒风肿。""冬月作菹煮作羹食之,能消宿食,下气,治嗽。"③《饮膳正要》:"温中益气,去心腹冷痛。"④《本草备要》:"捣敷阴囊肿大如斗,末服解酒毒,和芸薹根捣汁,鸡子清调涂诸热毒。"⑤《医林纂要》:"利水解热,下气宽中,功用略同萝卜。"
破骨风:性味: ①《贵州民间药物》:"性平,味苦辛。"②《四川中药志》:"性温,味苦,无毒。"主治:祛风除湿,活血止痛。治风湿腰腿骨节疼痛,跌打损伤,疮毒,痈疽。①《分类草药性》:"治风湿,和血脉骨节痛;清热,治头风疼痛。"②《中国药植志》:"治跌打损伤,有钻筋透骨之效:主治腰痛,腿痛等,亦有去骨中风寒之效能。"③《四川中药志》:"疗风湿筋骨痛、腰痛,散骨破后的积血,并治头风。"
匍伏堇:性味:苦、微辛,寒。主治:清热解毒,消肿排脓。用于肝炎,百日咳,目赤肿痛;外用治急性乳腺炎,疔疮,痈疖,带状疱疹,毒蛇咬伤,跌打损伤。
蚯蚓: 咸,寒。入肝、脾、肺经。入肝、脾、肺三经。功能主治:清热,平肝,止喘,通络。治高热狂躁,惊风抽搐,风热头痛,目赤,中风半身不遂,喘息,喉痹,关节疼痛,齿衄,小便不通,瘰疬。痄腮,疮疡。
蜂房:甘,平。归胃经。功能主治:祛风,功毒,杀虫,止痛。用于龋齿牙痛,疮疡肿毒,乳痈,瘰疬,皮肤顽癣,鹅掌风。
牛耳大黄:性味:苦、酸,寒。主治:清热解毒,凉血止血,通便,杀虫。用于肺结核咳血,急性肝炎,痢疾,便秘,功能性子宫出血,痔疮出血,痈疖疮疡;外用治腮腺炎,神经性皮炎,疥癣,烧伤,外伤出血。
以下采用实施例来详细说明本发明的实施方式,借此对本发明如何应用技术手段来解决技术问题,并达成技术效果的实现过程能充分理解并据以实施。
实施例1 药物组合物1
本发明实施例提供了一种具有治疗奶牛缺乳症的药物组合物,由以下重量份数的原料药材制成:屈草15重量份、木香18重量份、荃皮22重量份、乌药17重量份、甘松19重量份、倒水莲21重量份、九香虫18重量份、大荨麻20重量份、青皮22重量份、大枣15重量份、柴胡10重量份、鸡矢藤20重量份、香附17重量份、苤蓝13重量份、灵芝17重量份、娑罗子16重量份、菱蒂12重量份、郁金17重量份、万丈深18重量份、玫瑰花17重量份、山楂15重量份、曼陀罗子22重量份、麦芽18重量份、谷芽15重量份、葛蕈16重量份、党参14重量份、芜菁13重量份、山药18重量份和薜荔果20重量份。
其制备方法如下:
当药物组合物的剂型为粉针剂,其制备方法包括以下步骤:
第一步,将所述原料药材按比例混合,放入超微粉碎机中粉碎成0.7μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第二步,在第一步获得的混合粉料中加入相对于所述混合粉料的质量5倍的醇浓度为85%的乙醇,搅拌溶解获得乙醇溶液,将乙醇溶液在10℃的条件下静置35小时,采用渗漉法以每分钟2.0ml的速度缓缓渗漉,收集渗漉液,浓缩并干燥,并在此放入超微粉碎机中粉碎40分钟,获得0.8μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第三步,取第二步获得的混合粉料20g,加入丙二醇50g,加200ml注射用水,搅拌使其溶解,进一步加注射用水至1500ml,再加入1.5g针用活性炭,充分搅拌50分钟;脱炭过滤;用0.27μm微孔滤膜过滤;冷冻干燥得无菌粉末,分装灌封。
实施例2药物组合物2
本发明实施例提供了具有治疗奶牛缺乳症的药物组合物,由以下重量份数的原料药材制成:屈草12重量份、木香17重量份、荃皮16重量份、乌药20重量份、甘松20重量份、倒水莲19重量份、九香虫13重量份、大荨麻22重量份、青皮20重量份、大枣18重量份、柴胡9重量份、鸡矢藤13重量份、香附21重量份、苤蓝11重量份、灵芝20重量份、娑罗子15重量份、菱蒂12重量份、郁金15重量份、万丈深17重量份、玫瑰花12重量份、山楂10重量份、曼陀罗子22重量份、麦芽16重量份、谷芽14重量份、葛蕈16重量份、党参15重量份、芜菁13重量份、山药17重量份、薜荔果21重量份、破骨风9重量份、牛耳大黄15重量份、蚯蚓3重量份、匍伏堇18重量份和蜂房12重量份。
其制备方法如下:
当药物组合物的剂型为散剂,其制备方法包括以下步骤:
第一步,将香附、苤蓝、灵芝、娑罗子、菱蒂和郁金按所述比例混合,用相对于获得的混合物质量的4倍的醇浓度为90%的乙醇溶解,加热回流5小时后提取,过滤,得第一过滤液;将滤渣再次用相对于获得的混合物质量的3倍的醇浓度90%的乙醇溶解,加热回流1.5小时后提取,过滤,得第二过滤液;合并第一过滤液和第二过滤液,减压回收乙醇并浓缩至药液浓度为0.7g生药/mL,经体积为5L的大孔吸附树脂柱洗脱,先用5倍树脂柱体积的去离子水或蒸馏水洗脱,再用3倍树脂柱体积的醇浓度为90%乙醇洗脱,收集乙醇洗脱液,减压浓缩除去乙醇,干燥并粉碎成粉末;
第二步,将乌药、甘松、倒水莲、九香虫和大荨麻按所述比例混合,加相对于获得的混合物质量的3倍的醇浓度90%的乙醇,加热回流提取2次,提取液合并,用管式离心机离心除杂,将除杂后的提取液于70℃的条件下减压浓缩至60℃时相对密度为1.22的膏体,将浓缩后的膏体用喷雾干燥器干燥成粉末;
第三步,将剩余原料药材按所述比例混合,加相对于获得的混合物质量的3倍的水溶解,加热煎煮4小时,过滤;将滤渣再次用相对于获得的混合物质量的2倍的水溶解,再次加热煎煮1.5小时,过滤;合并两次的过滤液,减压浓缩除去水分,干燥并粉碎成粉末;
第四步,将第一步、第二步和第三步获得的粉末混合,随后过筛成100目的细粉,将获得的细粉投入混合搅拌机中搅拌,即获得散剂。
药物组合物的毒性试验:
1、试验药物
选取屈草15 g、木香18 g、荃皮22 g、乌药17 g、甘松19 g、倒水莲21 g、九香虫18 g、大荨麻20 g、青皮22 g、大枣15 g、柴胡10 g、鸡矢藤20 g、香附17 g、苤蓝13 g、灵芝17 g、娑罗子16 g、菱蒂12 g、郁金17 g、万丈深18 g、玫瑰花17 g、山楂15 g、曼陀罗子22 g、麦芽18 g、谷芽15 g、葛蕈16 g、党参14 g、芜菁13 g、山药18 g和薜荔果20 g混合,粉碎成粉末;将获得粉末加入1500g冷水中浸泡24小时,以文火煎煮,煎煮时间为1小时,将煎煮液过滤,获得第一滤液,收集滤渣;将过滤获得的滤渣再次加入1500g冷水中,以文火煎煮3次,每次煎煮1.5小时,过滤并合并过滤液,获得第二滤液;合并两次滤液,减压浓缩除去部分水,获得汤剂,汤剂的生药浓度为3g/ml。
2、试验方法:选用SD大鼠,给予不同浓度(18.0、6.0、2.0g生药/kg)的本发明的中药添加剂的汤剂,每天灌胃一次,连续90天,末次给药后24小时各组活杀1/2动物(雌雄各半),其余1/2动物继续观察2周后活杀。试验期间观察动物的外 观、一般行为、摄食量、体重变化,给药后90天和停药2周进行血液学(RBC、 HB、网织红细胞、PLT、CT、WBC及分类)和血液生化(AST、ALT、ALP、 Glu、BUN、Crca、TP、T.BIL、ALB、CHOL)、尿液生化、脏器系数、病理 组织学等指标检查。试验结果表明:本发明的药物组合物在高、中、低剂量组动物一般状态良好,外观体征、行为活动、进食量和体重增长均无异常变化; 三个剂量组及对照组血液学检查、血液生化学、尿液生化检查均在正常范围,组间无显著差异;各组主要脏器组织病理学检查未见明显异常。上述指标停药2周后也未见改变。本试验用药剂量分别为临床用药剂量的180、60、20倍,根据试验结果本发明的药物组合物在高、中、低三个剂量(18.0、6.0、2.0g 生药/kg)连续90天给药对大鼠无明显影响,无明确的毒性靶器官和敏感指标,恢复期观察也未见延迟性毒性反应,提示本发明的药物组合物临床应用的剂量安全性较高。
对比试验
山西省和顺县淼福养殖场,2012年12月底,发现将近300头奶牛,年龄16-20月龄,平均为18月龄,在初次泌乳期出现不同程度的乳房紧小,乳头短缩,外观幼稚,乳汁缺乏,拒绝哺乳,体瘦气虚,或遇到炎夏高温季节奶牛食欲骤减或不食。
从上述300头奶牛中,选取180头,将180头奶牛随机分成三组,分别为实施例组1、实施例组2和对照组,三组的患病奶牛在体重、病情等方面均无显著差异,具有可比性。实施例组1、实施例组2和对照组分别使用本发明实施例1的粉针剂、实施例2的散剂和传统药物进行治疗;其中,粉针剂采用皮下注射,每次2ml/次,每天两次,连续注射7天;散剂的使用方法是将散剂按照2:100的重量比例传统饲料中,连续饲喂7天;传统药物为皮下静脉注射脑垂体后叶素50单位,连续7天,饲喂传统饲料。实验周期为7天,实验前后分别称取各组患病奶牛体重,期间记录患病奶牛的痊愈情况及死亡数,计算出痊愈率,以及每头奶牛的泌乳量和乳成分,结果如表1和表2所示。其中,治愈:乳房和乳头正常,乳汁饱满,不再有食欲骤减或不食的现象,乳房饱满,乳头恢复正常;无效:临床症状没有改善。

从表1的数据可知:与传统药物相比,本发明实施例1和2的药物在治疗奶牛缺乳症方面具有疗效显著、价格低廉、无毒副作用的优势。
通过对比试验可知:与传统药物相比,本发明实施例1和2的药物组合物在治疗奶牛缺乳症方面具有毒副作用小、疗效显著的优势,而且还能够很好的保证奶牛乳腺及其身体其他器官的正常生长。
表2各实验组中每头奶牛每天的泌乳量和乳成份的治疗效果对照表
从表2的数据可知:实施例组1和实施例组2的泌乳量比对照组均有增长,分别提高了14.54%和13.61%。实验组1和2乳成分中乳脂率、乳蛋白、乳糖和乳中干物质也分别比对照组高3.23和3.01、2.74和2.31、0.5和0.65、4.64和4.4个百分点。
通过对比试验可知:与传统药物相比,本发明实施例1和2的药物组合物在治疗奶牛缺乳症方面具有泌乳量高,乳脂率明显提高、乳蛋白含量高、乳糖和乳中干物质也相对有所提高,乳房饱满和乳头正常,乳汁饱满,不再有食欲骤减或不食的现象。
典型病例:
2014年1月山西华鑫养殖场的450只18月龄的奶牛,将近160只奶牛在初次泌乳期出现不同程度的乳房紧小,乳头短缩,外观幼稚,乳汁缺乏,拒绝哺乳,体瘦气虚,或遇到炎夏高温季节奶牛食欲骤减或不食等症状。选取其中的50只患病奶牛,开始饲喂添加有本发明实施例2散剂的饲料,散剂按照2:100重量比混入传统饲料中,喂养7天,第2天患病奶牛症状明显改善,第3天50只奶牛均痊愈,继续饲喂4天后,与健康奶牛相比,乳房和乳头正常,乳汁饱满,不再有食欲骤减或不食的现象,乳房饱满,乳头恢复正常,与健康奶牛相比,泌乳量高,乳脂率明显提高、乳蛋白含量高、乳糖和乳中干物质也相对有所提高,平均体重基本相同。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

1.一种治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物包括以下原料药材:千屈草、木香、荃皮、乌药、甘松、倒水莲、九香虫、大荨麻、青皮、大枣、柴胡、鸡矢藤、香附、苤蓝、灵芝、娑罗子、菱蒂、郁金、万丈深、玫瑰花、山楂、曼陀罗子、麦芽、谷芽、葛蕈、党参、芜菁、山药和薜荔果。

5.根据权利要求1-4任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物中各原料药材的重量份数为:屈草10~18重量份、木香12~20重量份、荃皮14~25重量份、乌药13~22重量份、甘松11~21重量份、倒水莲15~22重量份、九香虫12~20重量份、大荨麻14~23重量份、青皮10~25重量份、大枣10~24重量份、柴胡8~16重量份、鸡矢藤11~23重量份、香附12~25重量份、苤蓝9~17重量份、灵芝10~20重量份、娑罗子13~18重量份、菱蒂8~19重量份、郁金10~19重量份、万丈深11~21重量份、玫瑰花10~22重量份、山楂9~18重量份、曼陀罗子13~23重量份、麦芽12~20重量份、谷芽11~17重量份、葛蕈8~17重量份、党参12~20重量份、芜菁11~17重量份、山药12~19重量份、薜荔果10~22重量份、破骨风8~17重量份、牛耳大黄10~18重量份、蚯蚓3~7重量份、匍伏堇12~21重量份和蜂房10~22重量份。
6.根据权利要求1-5任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物中各原料药材的重量份数为:屈草12重量份、木香17重量份、荃皮16重量份、乌药20重量份、甘松20重量份、倒水莲19重量份、九香虫13重量份、大荨麻22重量份、青皮20重量份、大枣18重量份、柴胡9重量份、鸡矢藤13重量份、香附21重量份、苤蓝11重量份、灵芝20重量份、娑罗子15重量份、菱蒂12重量份、郁金15重量份、万丈深17重量份、玫瑰花12重量份、山楂10重量份、曼陀罗子22重量份、麦芽16重量份、谷芽14重量份、葛蕈16重量份、党参15重量份、芜菁13重量份、山药17重量份、薜荔果21重量份、破骨风9重量份、牛耳大黄15重量份、蚯蚓3重量份、匍伏堇18重量份和蜂房12重量份。
7.根据权利要求1-6任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物中各原料药材的剂型为粉针剂,其制备方法包括以下步骤:
第一步,将所述原料药材按比例混合,放入超微粉碎机中粉碎成0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第二步,在第一步获得的混合粉料中加入相对于所述混合粉料的质量3~6倍的醇浓度为80%~95%的乙醇,搅拌溶解获得乙醇溶液,将乙醇溶液在5℃~10℃的条件下静置24~48小时,采用渗漉法以每分钟1~2.5ml的速度缓缓渗漉,收集渗漉液,浓缩并干燥,并在此放入超微粉碎机中粉碎30~60分钟,获得0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第三步,取第二步获得的混合粉料10g~20g,加入丙二醇30g~50g,加200ml注射用水,搅拌使其溶解,进一步加注射用水至1500ml,再加入1g~1.5g针用活性炭,充分搅拌30~60分钟;脱炭过滤;用0.22~0.27μm微孔滤膜过滤;冷冻干燥得无菌粉末,分装灌封。
8.根据权利要求1-6任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物的剂型为散剂,其制备方法包括以下步骤:
第一步,将香附、苤蓝、灵芝、娑罗子、菱蒂和郁金按所述比例混合,用相对于获得的混合物质量的3~5倍的醇浓度为80%~90%的乙醇溶解,加热回流3~5小时后提取,过滤,得第一过滤液;将滤渣再次用相对于获得的混合物质量的2~4倍的醇浓度85%~95%的乙醇溶解,加热回流1~2小时后提取,过滤,得第二过滤液;合并第一过滤液和第二过滤液,减压回收乙醇并浓缩至药液浓度为0.6~0.9g生药/mL,经体积为5L的大孔吸附树脂柱洗脱,先用5倍树脂柱体积的去离子水或蒸馏水洗脱,再用3倍树脂柱体积的醇浓度为90%~95%乙醇洗脱,收集乙醇洗脱液,减压浓缩除去乙醇,干燥并粉碎成粉末;
第二步,将乌药、甘松、倒水莲、九香虫和大荨麻按所述比例混合,加相对于获得的混合物质量的2~4倍的醇浓度85%~95%的乙醇,加热回流提取1~2次,提取液合并,用管式离心机离心除杂,将除杂后的提取液于60℃~80℃的条件下减压浓缩至60℃时相对密度为1.21~1.25的膏体,将浓缩后的膏体用喷雾干燥器干燥成粉末;
第三步,将剩余原料药材按所述比例混合,加相对于获得的混合物质量的2~4倍的水溶解,加热煎煮3~5小时,过滤;将滤渣再次用相对于获得的混合物质量的1~2.5倍的水溶解,再次加热煎煮1~1.5小时,过滤;合并两次的过滤液,减压浓缩除去水分,干燥并粉碎成粉末;
第四步,将第一步、第二步和第三步获得的粉末混合,随后过筛成80~120目的细粉,将获得的细粉投入混合搅拌机中搅拌,即获得散剂。
9.根据权利要求1-6任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物的剂型为粉针剂,其制备方法包括以下步骤:
第一步,将所述原料药材按比例混合,放入超微粉碎机中粉碎成0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第二步,在第一步获得的混合粉料中加入相对于所述混合粉料的质量3~5倍的醇浓度为85%~95%的乙醇,搅拌溶解获得乙醇溶液,将乙醇溶液在5℃~10℃的条件下静置24~36小时,采用渗漉法以每分钟1~2ml的速度缓缓渗漉,收集渗漉液,浓缩并干燥,并在此放入超微粉碎机中粉碎40~60分钟,获得0.1~10μm体积平均粒径的微米级颗粒和粒径小于0.1μm体积平均粒径的纳米级颗粒组成的混合粉料,所述混合粉料的得粉率至少为95%;
第三步,取第二步获得的混合粉料10g~20g,加入丙二醇30g~50g,加200ml注射用水,搅拌使其溶解,进一步加注射用水至1000ml,再加入1g~1.5g针用活性炭,充分搅拌30~50分钟;脱炭过滤;用0.22~0.24μm微孔滤膜过滤;冷冻干燥得无菌粉末,分装灌封。
10.根据权利要求1-6中任一项所述的治疗奶牛缺乳症的药物组合物,其特征在于,所述药物组合物的剂型为散剂,其制备方法包括以下步骤:

第二步,将乌药、甘松、倒水莲、九香虫和大荨麻按所述比例混合,加相对于获得的混合物质量的3~5倍的醇浓度80%~95%的乙醇,加热回流提取1~2次,提取液合并,用管式离心机离心除杂,将除杂后的提取液于60℃~70℃的条件下减压浓缩至60℃时相对密度为1.20~1.23的膏体,将浓缩后的膏体用喷雾干燥器干燥成粉末;
第三步,将剩余原料药材按所述比例混合,加相对于获得的混合物质量的2~4倍的水溶解,加热煎煮2~4小时,过滤;将滤渣再次用相对于获得的混合物质量的1~2倍的水溶解,再次加热煎煮1~2小时,过滤;合并两次的过滤液,减压浓缩除去水分,干燥并粉碎成粉末;
第四步,将第一步、第二步和第三步获得的粉末混合,随后过筛成80~120目的细粉,将获得的细粉投入混合搅拌机中搅拌,即获得散剂。
