技术领域
本发明涉及一种防治血栓性静脉炎的营养配方纳米颗粒,更具体的说,是涉及一种防治血栓性静脉炎的营养配方纳米颗粒及其制备方法技术领域。
背景技术
静脉炎(全称血栓性静脉炎)是指静脉血管的急性无菌性炎症,根据病变部位不同,静脉炎可分为浅静脉炎和深静脉炎。少数病人可有发热、白细胞总数增高等,患者常常陈诉疼痛肿胀。西医认为静脉输入强刺激性、高浓度药物或使用时间较长,损伤静脉内皮细胞;浅表静脉曲张、血液淤滞;肥胖、吸烟、外伤造成静脉内皮损伤;细菌感染等。静脉血管内膜损伤后,形成血栓,迅速导致整条浅静脉壁的炎症反应,甚至累及静脉周围组织,并有渗出液,局部表现有疼痛,肿胀和压痛的索状硬条或串珠状硬结,全身反应不明显。引起静脉血栓形成的病因很多,如创伤、手术、妊娠、分娩、心脏病、恶性肿瘤、口服避孕药及长期站立、下蹲、久坐、久卧等,较常见的是外科手术后引发本病。
中医认为血栓性静脉炎为湿热下注、气郁血瘀、湿热毒邪阻于络脉所致,通过活血化瘀、清热祛湿、通络利湿防治血栓性静脉炎。
从营养学角度来说,血栓性静脉炎除了采取正规的治疗方法进行治疗外,还要注意饮食治疗,供给充足的营养元素,合理搭配饮食结构。本发明正是结合饮食营养调节和中药配方制成一种具有补充营养和辅助治疗双重功能的保健食品颗粒,同时提高人体免疫力、抗衰老、排毒养颜、滋养脾胃、降三高、防治糖尿病和防癌、抗癌等方面的功能。

本发明针对现有产品的不足,而提供一种具有补充营养、辅助治疗、提高自身增加肠胃的蠕动,治病同时增强人体免疫力等优点的一种防治血栓性静脉炎的营养配方纳米颗粒。
一种防治血栓性静脉炎的营养配方纳米颗粒,所述营养配方纳米颗粒由以下原料:紫薯、大米、小米、玉米淀粉、黑米、黑芝麻、氨基酸、中药浓缩液、维生素加工制成;
所述营养配方纳米颗粒原料按照重量份配比为:紫薯5-10份、大米1-5份、小米1-15份、玉米淀粉5-10份、黑米1-10份、黑芝麻1-10份、氨基酸0.1-0.5份、中药浓缩液2-5份、维生素0.1-0.5份;
所述营养配方纳米颗粒中的中药浓缩液的组方按照重量份配比为:银花藤25-35份、元参15-30份、当归5-10份、川芎10-16份、赤芍5-15份、红花6-15份、牛膝5-10份、甘草5-10份、威灵仙5-10份、茵陈25-30份、赤豆5-10份、薏仁5-10份、泽泻5-10份、木通5-10份、鱼鳔5-10份、新鲜石斛10-20份、葡萄10-20份、蜂蜜、水适量;
所述氨基酸的原料按照重量份配比为:异亮氨酸0.01-0.08份、酪氨酸0.01-0.05份、苯丙氨酸0.01-0.04份、赖氨酸0.01-0.03份、精氨酸0.01-0.05份、组氨酸0.01-0.07份、色氨酸0.01-0.06份、瓜氨酸0.01-0.05份、鸟氨酸0.01-0.05份、L-亮氨酸0.01份、L-异亮氨酸0.01份、L-缬氨酸0.01份;
维生素为维生素A、维生素D、维生素E、维生素K、维生素K1、维生素C、维生素B1、维生素B2、维生素B6、维生素B12、维生素Be、维生素Bu、烟酸、叶酸、泛酸、胆碱、生物素其中的一种、几种或全部混合;通过添加补充维持身体健康所必需。
本发明还公开了一种防治血栓性静脉炎的营养配方纳米颗粒及制备加工方法,包括以下步骤;
A、将新鲜石斛和葡萄清洗干净,筛选优质石斛鲜条和葡萄,晾干备用;
B、把新鲜石斛放入旋转离心榨汁机内进行石斛汁水和纤维分离待用;再将葡萄放入吹风连续式速冻装置内,通过温度为-40~-30℃的空气按照3~6m/s的速度使葡萄的表面结冰,再进行葡萄肉、葡萄皮、葡萄籽分离作业;将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油备用;
C、将石斛汁水和葡萄肉放入搅拌筒内得基体溶液I;
D、在密封罐内加入相当于石斛纤维和葡萄皮总重量20-30%的蜂蜜及5-10倍重量的蒸馏水,搅拌打碎至100目,在温度37.5℃~38.9℃下密封保存24h,灭菌,过滤,放入无光密封罐,保持密封罐内的温度,在加入相当于石斛纤维和葡萄皮总重量的0.2%-0.5%的α-淀粉酶和蛋白酶的混合酶制剂;pH6.5条件下酶解60分钟;酶解后的料液过滤后,滤液浓缩去蛋白后醇洗,滤渣去离子水洗涤;酶解后的料液在80~140MPa的超高压下均质,离心分离去除不溶物,得基体溶液II;
E、将银花藤、元参、当归置于在40-50℃的蒸馏水中,浸泡20-30min分钟,加热到100℃,保持压力在0.2MPa微煮15min,再将上述溶液过滤;过滤溶液在0.05MPa条件下加热浓缩;将过滤溶液却至50℃以下,将过滤溶液泵入含有质量含量为85%的乙醇的醇沉罐, 30分钟后,将醇沉液通过0.3um过滤器过滤,将上述得到的醇沉液在0.04MPa条件下加热收醇,得中药提取液a;
将牛膝、赤豆、薏仁混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;
取川芎、赤芍、红花、甘草、威灵仙、茵陈、泽泻、木通、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉加水煎煮提取3次,第一次加2倍量水,温度保持100-120℃,进行熬煮20分钟;第二次加4倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加2倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,保温浓缩至原来体积的1/2,加入中药提取液a,得基体溶液III备用;
F、将基体溶液I、基体溶液II、基体溶液III混合过滤浓缩加入葡萄籽油得到中药浓缩液;
G、将紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分放入超微粉碎机,得颗粒粗粉,并通过100℃微波加热15分钟进行灭菌,再次将颗粒粗粉放入纳米处理设备进行纯物理粉碎处理粉碎至1-100nm;颗粒粗粉加入氨基酸、维生素、中药浓缩液、及辅料搅拌均匀进行湿法制粒或一步制粒制取颗粒。
进一步:所述辅料选自糊精、β-环糊精、可溶性淀粉、微晶纤维素、碳酸镁或碳酸钙中的任一种。
进一步:所述步骤G中的湿法制粒的步骤如下:在混合制粒机中加入粘合剂进行制湿颗粒,浆叶转速 70~150 转/分钟,飞刀转速200~800转/分钟,运行10~30分钟,将湿颗粒置于沸腾干燥机,进风温度45~85℃,干燥5~30分钟,得颗粒,控制颗粒水分4.0%~9.0%;
进一步:所述步骤G中的一步制粒的步骤如下:控制颗粒温度在50±5℃,干燥,水分控制在4.0%~9.0%。
紫薯:其富含蛋白质、淀粉、果胶、纤维素、氨基酸、维生素及多种矿物质,同时还富含硒元素和花青素,紫薯营养丰富具特殊保健功能,其中的蛋白质氨基酸都是极易被人体消化和吸收的;其中富含的维生素 A 可以改善视力和皮肤的粘膜上皮细胞,维生素 C 可使胶元蛋白正常合成,防治坏血病的发生,花青素是天然强效自由基清除剂,具有普通红薯的营养成分外,还富含硒元素、铁元素和花青素。
小米:小米亦称粟米,古代叫禾,是谷子去壳后的产物。除了丰富的铁质外, 小米也有蛋白质,复合维他命B、钙质、 钾、纤维素等。小米内含有多种对性有益的功能因子、能壮阳、滋阴、优生。
大米中医认为性味甘平,有补中益气、健脾养胃、益精强志、和五脏、通血脉、聪耳明目、止烦、止渴、止泻的功效,认为多食能令人“强身好颜色”。

川芎,味辛,温,入肝、脾、三焦三经,行气开郁,法风燥湿,活血止痛。
赤芍,苦,微寒。归肝经。有清热凉血,活血祛瘀的功效。
红花,辛,温,无毒,入心、肝经,通经、活血。
牛膝,生用散瘀血,消痈肿。
甘草,和中缓急,润肺,解毒,调和诸药。
威灵仙,味辛、咸,性温,具有祛风湿、通经络的功效,能治疗风湿痹痛、腰膝冷痛、扁桃体炎等症。
茵陈,保肝作用,滋阴补肾,清热解。
赤豆有除热毒、散恶血、通乳、消胀满、利小便的功效。
薏仁,健脾渗湿,除痹止泻。
泽泻,甘、淡,性寒。归肾、膀胱经。利水渗湿,泄热,化浊降脂。
木通,泻火行水,通利血脉,治小便赤涩,淋浊,水肿,胸中烦热,喉痹咽痛。

实施例 1
一种防治血栓性静脉炎的营养配方纳米颗粒,所述营养配方纳米颗粒由以下原料:紫薯、大米、小米、玉米淀粉、黑米、黑芝麻、氨基酸、中药浓缩液、维生素及水适量加工制成;
所述营养配方纳米颗粒原料按照重量份配比为:紫薯8份、大米2份、小米5份、玉米淀粉8份、黑米8份、黑芝麻8份、氨基酸0.1份、中药浓缩液5份、维生素0.1份;
所述中药浓缩液的组方按照重量份配比为:银花藤30份、元参25份、当归10份、川芎12份、赤芍12份、红花12份、牛膝8份、甘草8份、威灵仙8份、茵陈30份、赤豆8份、薏仁8份、泽泻8份、木通8份、鱼鳔8份、新鲜石斛15份、葡萄15份、蜂蜜、水适量;
所述氨基酸的原料按照重量份配比为:异亮氨酸0.02份、酪氨酸0.03份、苯丙氨酸0.01份、赖氨酸0.01份、精氨酸0.01份、组氨酸0.01份、色氨酸0.01份、瓜氨酸0.01份、鸟氨酸0.01份、L-亮氨酸0.01份、L-异亮氨酸0.01份、L-缬氨酸0.01份;

一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,包括以下步骤;
A、将新鲜石斛和葡萄清洗干净,筛选优质石斛鲜条和葡萄,晾干备用;
B、把新鲜石斛放入旋转离心榨汁机内进行石斛汁水和纤维分离待用;再将葡萄放入吹风连续式速冻装置内,通过温度为-40~-30℃的空气按照3~6m/s的速度使葡萄的表面结冰,再进行葡萄肉、葡萄皮、葡萄籽分离作业;将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油备用;
C、将石斛汁水和葡萄肉放入搅拌筒内得基体溶液I;
D、在密封罐内加入相当于石斛纤维和葡萄皮总重量20-30%的蜂蜜及5-10倍重量的蒸馏水,搅拌打碎至100目,在温度37.5℃~38.9℃下密封保存24h,灭菌,过滤,放入无光密封罐,保持密封罐内的温度,在加入相当于石斛纤维和葡萄皮总重量的0.2%-0.5%的α-淀粉酶和蛋白酶的混合酶制剂;pH6.5条件下酶解60分钟;酶解后的料液过滤后,滤液浓缩去蛋白后醇洗,滤渣去离子水洗涤;酶解后的料液在80~140MPa的超高压下均质,离心分离去除不溶物,得基体溶液II;葡萄皮中富含花青素,能够保护人体免受自由基的损伤,能够增强血管弹性、松弛血管,增加全身血液循环,具有一定的降血压功效;能增强免疫系统能力,具有强抗氧化、抗突变、减轻肝机能障碍、保护心血管等功能,葡萄皮和石斛鲜条纤维一起搅拌打碎经蜂蜜水提起花青素,可最大程度的提取出花青素的同时可保护花青素的品质。将水溶性膳食纤维加入基体溶液得到基体溶液II,可增加肠胃的蠕动的功效,保证代谢废物正常排出体外;吸附各种代谢废物,使之正常排出体外,提高石斛的综合利用价值同时提高人体对中药成分的吸收。
E、将银花藤、元参、当归置于在40-50℃的蒸馏水中,浸泡20-30min分钟,加热到100℃,保持压力在0.2MPa微煮15min,再将上述溶液过滤;过滤溶液在0.05MPa条件下加热浓缩;将过滤溶液却至50℃以下,将过滤溶液泵入含有质量含量为85%的乙醇的醇沉罐, 30分钟后,将醇沉液通过0.3um过滤器过滤,将上述得到的醇沉液在0.04MPa条件下加热收醇,得中药提取液a;
将牛膝、赤豆、薏仁混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;
取川芎、赤芍、红花、甘草、威灵仙、茵陈、泽泻、木通、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉加水煎煮提取3次,第一次加2倍量水,温度保持100-120℃,进行熬煮20分钟;第二次加4倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加2倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,保温浓缩至原来体积的1/2,加入中药提取液a,得基体溶液III备用;
F、将基体溶液I、基体溶液II、基体溶液III混合过滤浓缩加入葡萄籽油得到中药浓缩液;
G、将紫薯、大米、小米、玉米淀粉、黑米、黑芝麻放入超微粉碎机,得颗粒粗粉,并通过100℃微波加热15分钟进行灭菌,再次将颗粒粗粉放入纳米处理设备进行纯物理粉碎处理粉碎至1-100nm;颗粒粗粉加入氨基酸、维生素、中药浓缩液、及辅料搅拌均匀进行湿法制粒,浆叶转速 70~150 转/分钟,飞刀转速200~800转/分钟,运行10~30分钟,将湿颗粒置于沸腾干燥机,进风温度45~85℃,干燥5~30分钟,得颗粒,控制颗粒水分4.0%~9.0%。
对比实施例2
一种防治血栓性静脉炎的营养配方纳米颗粒,加工方法及其他营养成分同实施例1,所述中药浓缩液的组方按照重量份配比为:银花藤30份、元参25份、当归10份、川芎12份、赤芍12份、红花12份、牛膝8份、甘草8份、威灵仙8份、茵陈30份、赤豆8份、薏仁8份、泽泻8份、木通8份、新鲜石斛15份、葡萄15份、蜂蜜、水适量;和实施例1相比不含鱼鳔成分。
对比实施例3
一种防治血栓性静脉炎的营养配方纳米颗粒,其他营养成分同实施例1,中药浓缩液的组方按照重量份配比为:银花藤30份、元参25份、当归10份、川芎12份、赤芍12份、红花12份、牛膝8份、甘草8份、威灵仙8份、茵陈30份、赤豆8份、薏仁8份、泽泻8份、木通8份、鱼鳔8份、水适量。和实施例1相比不含新鲜石斛、葡萄、蜂蜜成分及加工制成的基体溶液I、基体溶液II。加工制备方法为:实施例1加工步骤的E和G。
对比实施例4
一种防治血栓性静脉炎的营养配方纳米颗粒,和实施例1相比不含紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分;其他均与实施例1相同。
对比实施例5
一种防治血栓性静脉炎的营养配方纳米颗粒,和实施例1相比不含氨基酸、维生素等微量元素;其他均与实施例1相同。
对比实施例6
一种防治血栓性静脉炎的营养配方纳米颗粒,和实施例1相比:不含紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分及氨基酸、维生素等微量元素;其他均与实施例1相同。
对比实施例7
一种防治血栓性静脉炎的营养配方纳米颗粒,和实施例1相比:不同的是,制取中药浓缩液,通过药材加水直接熬制方法制得。
临床实验分三部分:
(一)动物急慢性毒性实验:
1、动物急性毒性试验
药物:本发明实施例1和其余对比实施例所得营养粉。
试验动物 :普通级 NIH 小鼠,体重 18g±2g,雌雄各半。
方法与结果:
将营养颗粒各50g,加入水,制成的营养颗粒粉糊。将实验小鼠随机分成7组雌雄各半,分别喂食7种营养颗粒粉糊,容积 1ml/10g 体重,密切观察8小时并作记录,继续观察7天;7天中,小鼠无死亡,行为活泼,摄食量如常,毛色光亮无疏松污,无惊厥嗜唾,眼、口、鼻、耳及肛周无异常分泌物,无断尾烂趾。分别从7组中选择2 只处死,肉眼所见:心、肝、肺、胃内及胸腺等重要脏器,未见异常改变,表明两种该营养粉均无急性毒性。
2、动物长期毒性试验
药物:本发明实施例1和其余对比实施例所得营养颗粒加水制成的营养颗粒粉糊。
动物:普通级 SD 大鼠,体重 200g±10g,雌雄各半。
方法与结果:
7种营养颗粒粉糊的配置:均分为高、中、低三个剂量组,分别为患者食用量的200、100、50g;将实验鼠随机分成7组,分别连续喂食调制好的营养颗粒粉糊,连续喂食40 天,观察动物全身毒性反应和严重程度,处死后按操作规程检查各部位,并进行血液学、ALT、BUN 及心、肝、脾、肺、肾、胃等主要脏器的病理学检查;
经过长期喂食,两组大鼠均未出现毒性反应。生长发育良好。肉眼外观及主要脏器未见异常。外周血象及血清ALT、BUN与对照组比较无病理性改变。病理报告心、肝、脾、肺、肾、胃等均未有意义的改变,因此可以认为,经病理证实,7种营养颗粒粉糊对动物无慢性毒性表现。
通过以上两个毒性实验,证明本发明按实施例1和对比实施例制作的营养颗粒是安全的,无毒副作用的,可以用来对临床患者进行临床试验。
3、本发明实施例使用效果调查:
(1)调查方式 :
① 使用人群:普通人群
② 使用时间:90天
③ 使用人数:100人
④ 使用方法:每人每天泡水500 mL
⑤ 指标:湿热下注、气郁血瘀、湿热毒邪、浅表静脉曲张、患肢局部红肿,疼痛、容易疲劳等临床症状消失,且口感清酸甜为显效;饮用后上述临床症状缓解,血糖水平恢复正常,且口感清酸甜则记为有效。饮用后无明显舒适感则记为无效。
(2)调查结果
调查结果见表1。

结论
由对比实施例 2的数据可看出,无效人数的比例和实施例 1相比,增加了8%,表明鱼鳔在中药配方中起到了引药归经,增强疗效之功用,提高人体对药物的吸收效果;鱼鳔补肾,肾具有主司摄藏、主藏精、主生长发育和生殖、主水液化谢的功能。肾功能正常则精气充盈,全身诸脏之阴阳皆旺;反之,由于各种致病因素影 响或年老体衰,可致肾功能失常。由于鱼鳔是一种富有粘性的物质,含有极丰富的蛋白质、维他命、矿物质等营养成分,所以,鱼泡具有补肾虚、健腰膝、养阴益精及滋润养颜的作用。对于体质虚弱、真阴亏损、精神过劳、劳累过度的人士至为合适。
由对比实施例 3和实施例 1相比的数据可看出,不含新鲜石斛15份、葡萄15份、蜂蜜成分及加工制成的基体溶液I、基体溶液II;无效人数的比例和实施例 1相比,增加了7倍;且显效率的平均天数,比实施例 1相比多了3天(即感觉有效果,临床症状消失的平均天数多了3天;这表明本发明的加工制备方法使中药浓缩液组方和水溶性膳食纤维及葡萄籽油等成分之间产生了相互协同的药理作用,水溶性膳食纤维增加肠胃的蠕动的功效促进人体对药物成分的吸收,保证代谢废物正常排出体外,另外葡萄皮中含有的花青素、白藜芦醇,具有强抗氧化、抗突变、减轻肝机能障碍、保护心血管等功能,增强免疫系统能力;与中药为伍提高吸收效果,同时改善中药的口感,经过本发明的精心配置和最大程度药物成分的提取,本发明中药浓缩液配方和制备方法出现了出人意料的技术效果。
由对比实施例 4的数据和实施例 1相比可看出,不含紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分,无效人数的比例和实施例 1相比,增加了10倍,且显效率的人数也下降了,表明本发明的紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分中药浓缩液的药效之间产生了相互协同的药理作用, 供给人体充足的营养元素,饮食结构合理搭配,营养成分补充人体必须的能量,达到均衡营养,促进身体对药物的吸收。
由对比实施例 5和实施例 1相比,氨基酸、维生素等微量元素;补充人体所需要的营养元素及微量元素调节生理机能、催化代谢吸收药物的过程中有十分重要的作用。
由对比实施例 6和实施例 1相比的数据可看出,不含紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分及氨基酸、维生素等微量元素;无效率人数将近15倍且有效率人数也下降近20%;且显效率的平均天数,比实施例 1相比多了10天(即感觉有效果的人,临床症状消失的平均天数多了10天;说明营养成分及微量元素在人体药物吸收的过程中发挥了及其重要的协同作用,大大促进人体对药物和营养的吸收,加快人体恢复。
由对比实施例 7和实施例 1相比的数据可看出,药材加水直接熬制方法和本发明的中药制取方法相比,本发明的制备方法,在药效的提取上远远大于传统的加水直接熬制方法,药效成分的提取程度有了巨大的提高。
综上所述,本发明提供的一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,活血化瘀、清热祛湿、通络利湿防治血栓性静脉炎和营养成分相互协同的药理作用,供给人体充足的营养元素,饮食结构合理搭配,营养成分补充人体必须的能量,达到均衡营养,促进身体对药物的吸收,所有原料均来自药食两用天然植物,无毒副作用。

所述营养配方纳米颗粒原料按照重量份配比为:紫薯5-10份、大米1-5份、小米1-15份、玉米淀粉5-10份、黑米1-10份、黑芝麻1-10份、氨基酸0.1-0.5份、中药浓缩液2-5份、维生素0.1-0.5份;
所述中药浓缩液的组方按照重量份配比为:银花藤25-35份、元参15-30份、当归5-10份、川芎10-16份、赤芍5-15份、红花6-15份、牛膝5-10份、甘草5-10份、威灵仙5-10份、茵陈25-30份、赤豆5-10份、薏仁5-10份、泽泻5-10份、木通5-10份、鱼鳔5-10份、新鲜石斛10-20份、葡萄10-20份、蜂蜜、水适量。

3.一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,其特征在于,包括以下步骤;
A、将新鲜石斛和葡萄清洗干净,筛选优质石斛鲜条和葡萄,晾干备用;
B、把新鲜石斛放入旋转离心榨汁机内进行石斛汁水和纤维分离待用;再将葡萄放入吹风连续式速冻装置内,通过温度为-40~-30℃的空气按照3~6m/s的速度使葡萄的表面结冰,再进行葡萄肉、葡萄皮、葡萄籽分离作业;将葡萄籽经真空脱气,然后采用CO2超临界流体萃取,CO2超临界流体萃取的条件为:萃取压力45MPa,萃取温度35-45℃,萃取时间30min,分离得到葡萄籽油备用;
C、将石斛汁水和葡萄肉放入搅拌筒内得基体溶液I;
D、在密封罐内加入相当于石斛纤维和葡萄皮总重量20-30%的蜂蜜及5-10倍重量的蒸馏水,搅拌打碎至100目,在温度37.5℃~38.9℃下密封保存24h,灭菌,过滤,放入无光密封罐,保持密封罐内的温度,在加入相当于石斛纤维和葡萄皮总重量的0.2%-0.5%的α-淀粉酶和蛋白酶的混合酶制剂;pH6.5条件下酶解60分钟;酶解后的料液过滤后,滤液浓缩去蛋白后醇洗,滤渣去离子水洗涤;酶解后的料液在80~140MPa的超高压下均质,离心分离去除不溶物,得基体溶液II;
E、将银花藤、元参、当归置于在40-50℃的蒸馏水中,浸泡20-30min分钟,加热到100℃,保持压力在0.2MPa微煮15min,再将上述溶液过滤;过滤溶液在0.05MPa条件下加热浓缩;将过滤溶液却至50℃以下,将过滤溶液泵入含有质量含量为85%的乙醇的醇沉罐, 30分钟后,将醇沉液通过0.3um过滤器过滤,将上述得到的醇沉液在0.04MPa条件下加热收醇,得中药提取液a;
将牛膝、赤豆、薏仁混合搅拌放入超微粉碎机,超微粉碎机加热保持温度80度5-6分钟,进行超微粉碎得到300-400目的超微混合细粉;
取川芎、赤芍、红花、甘草、威灵仙、茵陈、泽泻、木通、鱼鳔,混合均匀加30-60℃温水浸泡10分钟后,放入冷冻柜内,在-10~-20℃冷冻10-20分钟,形成冷冻药块,将冷冻药块和超微混合细粉加水煎煮提取3次,第一次加2倍量水,温度保持100-120℃,进行熬煮20分钟;第二次加4倍量水,温度保持80-100℃,进行熬煮10分钟;第三次加2倍量水,温度保持120-150℃,进行熬煮30分钟,过滤,保温浓缩至原来体积的1/2,加入中药提取液a,得基体溶液III备用;
F、将基体溶液I、基体溶液II、基体溶液III混合过滤浓缩加入葡萄籽油得到中药浓缩液;
G、将紫薯、大米、小米、玉米淀粉、黑米、黑芝麻等营养成分放入超微粉碎机,得颗粒粗粉,并通过100℃微波加热15分钟进行灭菌,再次将颗粒粗粉放入纳米处理设备进行纯物理粉碎处理粉碎至1-100nm;颗粒粗粉加入氨基酸、维生素、中药浓缩液、及辅料搅拌均匀进行湿法制粒或一步制粒制取颗粒。
4.根据权利要求3所述的一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,其特征在于,所述辅料选自糊精、β-环糊精、可溶性淀粉、微晶纤维素、碳酸镁或碳酸钙中的任一种。
5.根据权利要求3所述的一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,其特征在于,所述步骤G中的湿法制粒的步骤如下:在混合制粒机中加入粘合剂进行制湿颗粒,浆叶转速 70~150 转/分钟,飞刀转速200~800转/分钟,运行10~30分钟,将湿颗粒置于沸腾干燥机,进风温度45~85℃,干燥5~30分钟,得颗粒,控制颗粒水分4.0%~9.0%。
6.根据权利要求3所述的一种防治血栓性静脉炎的营养配方纳米颗粒的制备加工方法,其特征在于,所述步骤G中的一步制粒的步骤如下:控制颗粒温度在50±5℃,干燥,水分控制在4.0%~9.0%。
