技术背景
随着社会的进步和发展，人们生活节奏加快，日益激烈的社会竞争使人们的生活和工作压力越来越大，睡眠障碍已成为生活中常见的精神系统疾病，严重威胁着人类的身体健康。有关统计数据表明，全球有约45％的人群有睡眠质量不佳的问题。
目前在临床上使用的具有镇静催眠作用的化学合成药，因有嗜睡、恶心、呕吐、高热、肌张力增高及激动不安、意识模糊等严重的毒副作用，同时容易产生一定的耐药性和依赖性，其应用受到了一定的限制。由于中医对改善睡眠的独特理论，在失眠的治疗上有独特的特点，中药作为我国传统疾病治疗手段，对人体多方面进行整体调节功能，标本兼治，又较少有毒副作用。因此，目前人们积极的以中医药基本理论为指导，根据辨证施治的原则，从中药传统方剂中寻找和开发一种具有改善睡眠治疗效果好，不良反应少，应用范围广的中药组合物。
发明内容
本发明的目的在于提供一种改善睡眠的组方其制备方法。该工艺制得的产品不仅服用安全方便，生物利用度高，对人体副作用很小，能够显著改善人体睡眠质量，而且提供的制备方法能尽量保存组方成分活性，易于大型工业化生产，有很好的市场开发应用前景。
本发明的技术方案提供了一种具有改善睡眠的组合物，由下列重量份数的原料组成：冬虫夏草8-16份、玛咖8-14份、枸杞6-12份、大枣6-12份、灵芝12-18份、手掌参6-12份、石斛5-11份、雪莲花4-10份、红景天6-12份、蜂蜜2-7份。

(2)将冬虫夏草单独进行超微粉碎；
(3)将(1)所得二氧化碳超临界萃取物、(2)所得虫夏草超微粉与蜂蜜、可压性淀粉和羧甲基淀粉钠混合均匀，制备成片剂剂型。
在一些实施方案中，上述实施方案的组合物的制备方法，由以下步骤组成：
(1)将玛咖、大枣、枸杞、灵芝、手掌参、石斛、雪莲花及红景天混合后进行超微粉碎，循环冷却温度设置为-10--20℃，粉碎时间为10-15min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取，其中二氧化碳超临界萃取的条件为：压力30-40MPa，温度50-60℃，时间3-4h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的30-40％，混合物粉末装入量占二氧化碳超临界萃取釜体积的10-20％，萃取结束后收料；
(2)冬虫夏草采用冷冻冻干技术进行干燥，时间设置为48-72h，然后超微粉碎，过200目筛；
(3)将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜、及可压性淀粉和羧甲基淀粉钠混合均匀，制粒；
(4)将步骤(3)中所得制品在高速旋转式压片机上进行常规方法压片，即得。
在另一些实施方案中，上述实施方案的组合物的制备方法，由以下步骤组成：
(1)将玛咖、大枣、枸杞、灵芝、手掌参、石斛、雪莲花及红景天混合后进行超微粉碎，循环冷却温度设置为-10℃，粉碎时间为12min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取，其中二氧化碳超临界萃取的条件为：压力35MPa，温度55℃，时间3.5h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的35％，混合物粉末装入量占二氧化碳超临界萃取釜体积的15％，萃取结束后重复收料；
(2)冬虫夏草采用冷冻冻干技术进行干燥，时间设置为60h，然后超微粉碎，过200目筛；
(3)将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜及可压性淀粉和羧甲基淀粉钠混合均匀，制粒；
(4)将步骤(3)中所得制品在高速旋转式压片机上进行常规方法压片，即得。
本发明的显著优点在于：
1.本发明配方合理科学，发挥几种药材的协同作用，其中富含多糖类、核苷类、皂甙类、黄酮类、维生素、微量元素等生物活性物质。具有睡眠障碍人群服用结果显示，本发明的组合物能够显著的缩短入睡时间，延长睡眠时间，提高生活质量。

组合物的制备方法为：
(1)、称取清洗和冷冻干燥后的玛咖11份、大枣9份、枸杞9份、灵芝15份、手掌参9份、石斛8份、雪莲花7份及红景天9份进行超微粉碎，循环冷却温度设置为-10℃，粉碎时间为12min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取：压力35MPa，温度55℃，时间3.5h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的30％，混合物粉末装入量占二氧化碳超临界萃取釜体积的15％，萃取结束后重复收料；
(2)、冬虫夏草用纯净水洗净之后，采用冷冻冻干技术进行干燥，时间设置为60h，然后超微粉碎，过200目筛，称取12份备用；
(3)、将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜4份，可压性淀粉15份，羧甲基淀粉钠3份混合均匀，制粒；
(4)、将步骤(3)中所得制品采用常规湿法制粒压片技术在高速旋转式压片机上进行压片，干燥温度设定为50℃，干燥时间为30min，每5min翻动一次。然后将所得压片使用除粉机除去片上粉末即得合格成型片。

实施例4
组合物的制备方法为：
(1)、称取清洗和冷冻干燥后的玛咖12份、大枣10份、枸杞10份、灵芝16份、手掌参10份、石斛9份、雪莲花8份及红景天10份进行超微粉碎，循环冷却温度设置为-20℃，粉碎时间为14min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取：压力40MPa，温度60℃，时间4h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的40％，混合物粉末装入量占二氧化碳超临界萃取釜体积的20％，萃取结束后重复收料；
(2)、冬虫夏草用纯净水洗净之后，采用冷冻冻干技术进行干燥，时间设置为72h，然后超微粉碎，过200目筛，称取14份备用；
(3)、将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜5份，可压性淀粉16份，羧甲基淀粉钠4份混合均匀，制粒；
(4)、将步骤(3)中所得制品采用常规湿法制粒压片技术在高速旋转式压片机上进行压片，干燥温度设定为50℃，干燥时间为30min，每5min翻动一次。然后将所得压片使用除粉机除去片上粉末即得合格成型片。
实施例5

(2)、冬虫夏草用纯净水洗净之后，采用冷冻冻干技术进行干燥，时间设置为72h，然后超微粉碎，过200目筛，称取16份备用；
(3)、将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜7份，可压性淀粉18份，羧甲基淀粉钠5份混合均匀，制粒；
(4)、将步骤(3)中所得制品采用常规湿法制粒压片技术在高速旋转式压片机上进行压片，干燥温度设定为50℃，干燥时间为30min，每5min翻动一次。然后将所得压片使用除粉机除去片上粉末即得合格成型片。
实施例6临床功效评价
1、资料和方法
研究对象与分组
收集失眠和精神抑郁时间达3个月以上的患者50例，男30例，女20例，年龄35-63岁。治疗患者35例，对照患者15例。
2、方法
2.1治疗方法
治疗患者选用本发明实施例1中提供的药物组合物，用量为每次0.6g(0.2g/片，3片)，3次/日。
2.2观察方法
分别观察治疗前后患者晚上入睡时间，睡眠持续时间，及其表现出来的烦躁、焦虑等症状，并记录服药4周后的表现。
3、结果
治疗组患者服药4周之后，睡眠质量得到显著改善，入睡时间缩短，持续睡眠时间也延长到大于7小时，之前多梦多醒现象明显减少，烦躁、焦虑等现象基本消失，有效率达到71％。停药3个月之后未见复发。
以上临床实验结果表明，本发明提供的药物组合物具有很好的改善睡眠的作用，且实验中患者未见明显的不良反应，临床用药安全，且患者情况好转之后复发率很低。
以上所述仅为本发明的优选实施方案，应该指出，对于本领域的技术人员来说，在不脱离本发明原理的前提下，还可以做一些改进，这些有限的改进也应该被视为本发明所保护的范围。

1.一种改善睡眠的组合物，其特征在于，由下列重量份数的原料制成：冬虫夏草8-16份、玛咖8-14份、枸杞6-12份、大枣6-12份、灵芝12-18份、手掌参6-12份、石斛5-11份、雪莲花4-10份、红景天6-12份、蜂蜜2-7份；
所述组合物进一步包括可压性淀粉12-18份，羧甲基淀粉钠1-5份；
所述组合物的制备方法，包括以下步骤：
(1)将玛咖、大枣、枸杞、灵芝、手掌参、石斛、雪莲花及红景天混合后进行超微粉碎，再进行二氧化碳超临界萃取；
(2)冬虫夏草采用冷冻冻干技术进行干燥，时间设置为48-72h，然后超微粉碎，过200目筛；
(3)将(1)所得二氧化碳超临界萃取物、(2)所得冬虫夏草超微粉及蜂蜜、可压性淀粉和羧甲基淀粉钠混合均匀，制备成片剂剂型；
其中，步骤(1)所述的超临界萃取的温度为50-60℃，步骤(1)所述超微粉碎的温度为-10--20℃。

4.一种如权利要求1所述组合物的制备方法，其特征在于，由以下步骤组成：
(1)将玛咖、大枣、枸杞、灵芝、手掌参、石斛、雪莲花及红景天混合后进行超微粉碎，循环冷却温度设置为-10--20℃，粉碎时间为10-15min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取，其中二氧化碳超临界萃取的条件为：压力30-40MPa，温度50-60℃，时间3-4h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的30-40％，混合物粉末装入量占二氧化碳超临界萃取釜体积的10-20％，萃取结束后收料；
(2)冬虫夏草采用冷冻冻干技术进行干燥，时间设置为48-72h，然后超微粉碎，过200目筛；
(3)将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜、可压性淀粉和羧甲基淀粉钠淀粉混合均匀，制粒；
(4)将步骤(3)中所得制品在高速旋转式压片机上进行常规方法压片，即得。
5.根据权利要求4所述的制备方法，其特征在于，由以下步骤组成：
(1)将玛咖、大枣、枸杞、灵芝、手掌参、石斛、雪莲花及红景天混合后进行超微粉碎，循环冷却温度设置为-10℃，粉碎时间为12min，将得到的混合物粉末过200目筛之后进行二氧化碳超临界萃取，其中二氧化碳超临界萃取的条件为：压力35MPa，温度55℃，时间3.5h，使用乙醇为夹带剂，乙醇用量为萃取釜体积的35％，混合物粉末装入量占二氧化碳超临界萃取釜体积的15％，萃取结束后重复收料；
(2)冬虫夏草采用冷冻冻干技术进行干燥，时间设置为60h，然后超微粉碎，过200目筛；
(3)将步骤(1)所得超临界萃取物和步骤(2)中的冬虫夏草粉末混合，再加入蜂蜜及可压性淀粉和羧甲基淀粉钠淀粉混合均匀，制粒；
(4)将步骤(3)中所得制品在高速旋转式压片机上进行常规方法压片，即得。
