Abstract

Description
技术领域
本申请要求于2011年10月10日提交的韩国专利申请第10-2011-0103306号的优先权和权益,在此为了所有目的通过参考将其并入,如同其完全在此阐述。
本发明涉及使用甘氨酰-tRNA合成酶(GRS)和钙粘蛋白(CDH)筛选用于预防或治疗癌症的试剂的新方法。更具体而言,其涉及筛选调节GRS或其片段与CDH的结合水平的测试试剂的方法。
背景技术
作为源自遗传密码发展中的一部分而出现的古老蛋白,氨酰基tRNA合成酶(AARS)是翻译机构的关键组分。20种酶(每种对应每种氨基酸)在细胞质中催化各氨基酸附着至其同源tRNA,其中带电的tRNA继而用于核糖体蛋白合成。令人惊奇的是,对于许多tRNA合成酶已经发现外翻译功能(ex-translational function),包括大肠杆菌中的基因调节、粗糙脉孢菌(N.crassa)的线粒体中的RNA剪接(Kamper等,1992.Mol.Cell.Biol.12,499-511)以及脊椎动物中功能多样性,该多样性包括对炎性响应和血管形成的调节等(Park等,2005.Trends.Biochem.Sci.30,569-574)。这些扩展功能与特异化基序和结构域(例如内部短序列基序和附加GST、亮氨酸拉链和螺旋-转角-螺旋结构域)的增加相关(Guo等,2010.FEBS Lett.584,434-442)。特异化基序和结构域的添加促进了赋予新功能的新的蛋白质-蛋白质相互作用。据认为,AARS以及哺乳动物细胞中作为多重-tRNA合成酶复合体一部分的蛋白质与许多疾病的关联中的某一些是由于其外翻译功能的破坏或改变所致(Park等,2008;Sampath等,2004)。实际上,存在在酪氨酰-和甘氨酰-tRNA合成酶中的引起进行性神经病性腓骨肌萎缩病(Charcot-Marie-Tooth disease)的显性突变,该突变不破坏氨酰化活性(Jordanova等,2006.Nat.Genet.38,197-202;Nangle等,2007.Proc.Natl.Acad.Sci.USA104,11239-11244)。
对于翻译机构的必需组分还令人惊奇的是观察到以下:酪氨酰-和色氨酰-tRNA合成酶(YRS和WRS)的特定片段(通过选择性剪接或天然蛋白水解产生)与胞外受体的结合以及通过所述胞外受体的信号传导,所述胞外受体包括PMN细胞上的CXCR1和CXCR2(YRS)(Wakasugi和Schimmel,1999.Science284,147-151)以及内皮细胞上的VE-钙粘蛋白(WRS)。这两种合成酶由哺乳动物细胞在特定条件下分泌,这使他们的外翻译功能成为可能(Wakasugi等,2002.Proc.Natl.Acad.Sci.USA99,173-177;Yang等,2007.Structure15,793-805)。总而言之,这些观察结果提出了这样一种可能性:发现tRNA合成酶的外翻译功能的一种方式可以是通过对在不进行翻译的生理环境下存在的那些tRNA合成酶进行注释来进行(Park等,2008,Proc.Natl.Acad.Sci.USA105,11043-11049;Sampath等,2004.Cell119,195-208)。这样的考虑可以使我们检查人血清中特定合成酶的存在。
发明内容
技术问题
部分地由于我们对AARS和临床观察的结构-功能关系进行的不间断的研究,我们专注于GRS。这些观察使我们理解GRS作为癌微环境中循环蛋白的可能作用,由此通过确认CDH充当GRS的受体并且调节ERK途径和调控细胞的死亡和生长而完成本发明。
因此,本发明的目的在于提供一种筛选用于预防或治疗癌症的试剂的方法,所述方法包括以下步骤:
(a)在存在或不存在测试试剂时使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在存在或不存在测试试剂时测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂时GRS或其片段与CDH的结合亲和力与不存在测试试剂时GRS或其片段与CDH的结合亲和力进行比较;和
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂。
本发明的另一目的在于提供一种筛选用于预防或治疗癌症的试剂的方法,所述方法包括以下步骤:
(a)在存在或不存在测试试剂时使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在存在或不存在测试试剂时测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂时GRS或其片段与CDH的结合亲和力与不存在测试试剂时GRS或其片段与CDH的结合亲和力进行比较;
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂。
(e)使步骤(d)中鉴定出的测试试剂与表达CDH的细胞接触;和
(f)测定所述表达CDH的细胞的凋亡。
技术方案
为了实现所述目的,本发明提供一种筛选用于预防或治疗癌症的试剂的方法,所述方法包括以下步骤:
(a)在存在或不存在测试试剂时使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在存在或不存在测试试剂时测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂时GRS或其片段与CDH的结合亲和力与不存在测试试剂时GRS或其片段与CDH的结合亲和力进行比较;和
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂。
为了实现另一目的,本发明提供一种筛选用于预防或治疗癌症的试剂的方法,所述方法包括以下步骤:
(a)在存在或不存在测试试剂时使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在存在或不存在测试试剂时测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂时GRS或其片段与CDH的结合亲和力与不存在测试试剂时GRS或其片段与CDH的结合亲和力进行比较;
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂;
(e)使步骤(d)中鉴定出的测试试剂与表达CDH的细胞接触;和
(f)测定所述表达CDH的细胞的凋亡。

定义

本文所用的“表达”是指蛋白质或核酸在细胞中的形成。
本文所用的“宿主细胞”是指含有通过任意手段(例如电穿孔、磷酸钙沉淀、微注射、转化和/或病毒感染等)引入至细胞中的异源DNA的原核细胞或真核细胞。
术语“多肽(单数)”在本文可以与术语“多肽(复数)”以及“蛋白质”相互替换地使用,并且是指例如通常在自然状态的蛋白质中发现的氨基酸残基的聚合物。
术语“GRS多肽”是指称为甘氨酰tRNA合成酶的多肽。所述GRS多肽可以是具有氨基酸序列SEQ ID NO:1(GenBank登录号AAA86443.1)或GenBank登录号AAA57001.1(SEQ ID NO:5)、EAL24449.1(SEQ ID NO:6)、BAA06338.1(SEQ IDNO:7)、NP_002038.2(SEQ ID NO:8)、AAH07755.1(SEQ ID NO:9)、AAH07722.1(SEQID NO:10)的多肽。并且GRS包括其功能等效物。
术语“GRS片段”是指全长GRS多肽的一部分,并且其包含C末端的反密码子结合域。例如,其可以是以下片段,所述片段由SEQ ID NO:1的全长GRS氨基酸的511位至最后685位氨基酸(SEQ ID NO:2)组成,并且包含C末端反密码子结合域(SEQID NO:11,为SEQ ID NO:1的511位~674位氨基酸)。此外,GRS片段包含其功能等效物。同时,GRS的线粒体形式(例如SEQ ID NO:7)与细胞质形式(例如SEQ ID NO:1)的不同之处在于添加了54个氨基酸。因此,C末端的结合域的位置可以被本领域技术人员认为反映出这一点。
术语“功能等效物”是指下述多肽,其包含的氨基酸序列与GRS或其片段的氨基酸序列展示出基本相同的生理活性,并与其具有至少70%氨基酸序列同源性(即同一性),优选至少80%,更优选至少90%,例如70%、71%、72%、73%、74%、75%、76%、77%、78%、79%、80%、81%、82%、83%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%和100%氨基酸序列同源性,并且“功能等效物”是指与由SEQ ID NO:1或SEQ ID NO:2表示的多肽具有基本相同的生理活性的多肽。“基本相同的生理活性”是指通过结合CDH、特别是CDH6或CDH18激活ERK途径,从而抑制或杀死癌细胞。功能等效物可以包括例如对SEQ ID NO:1的一些氨基酸进行添加、取代或缺失的结果而生成的肽。氨基酸取代优选是保守性取代。天然存在的氨基酸的保守性取代的实例如下:脂肪族氨基酸(Gly、Ala、Pro)、疏水性氨基酸(Ile、Leu、Val)、芳香族氨基酸(Phe、Tyr、Trp)、酸性氨基酸(Asp、Glu)、碱性氨基酸(His、Lys、Arg、Gln、Asn)和含硫氨基酸(Cys、Met)。此外,功能等效物还包括具有本发明的GRS的一些氨基酸序列的缺失的变体。氨基酸的缺失或取代优选位于不直接参与本发明多肽的生理活性的区域。并且氨基酸的缺失优选位于不直接参与GRS的生理活性的区域。此外,功能等效物还包括在GRS氨基酸序列的两个末端或在序列内添加数个氨基酸的变体。另外,本发明的功能等效物还包括对本发明多肽的一些化学结构进行修饰同时保持本发明多肽的基本骨架和生理活性的多肽衍生物。该修饰的实例包括用于改变本发明多肽的稳定性、贮存性、挥发性或溶解度的结构修饰。
本文将序列同一性或同源性定义为:在为了实现最大百分比序列同一性将序列进行比对和必要时引入空位后,并且在不将任何保守性取代(如上文所述)认为是序列同一性的部分情况下,候选序列中与GRS的氨基酸序列(SEQ ID NO:1)相同的氨基酸残基的百分比。GRS的氨基酸序列的N末端、C末端或内部延伸、缺失或插入都不解释为影响序列同一性或同源性。因此,序列同一性可以通过常用于比较两个多肽的氨基酸位置的相似性的标准方法确定。使用诸如BLAST或FASTA等计算机程序,比对两个多肽各自氨基酸的最佳匹配(沿着一个或两个序列的全长或沿着一个或两个序列的预定部分)。所述程序提供默认开放罚分和默认空位罚分,并且诸如PAM250(一个标准的评分矩阵,参见Dayhoff等,Atlas of Protein Sequence and Structure,第5卷,增刊3(1978))等评分矩阵(scoring matrix)可以与计算机程序联用。例如,可以如下计算百分比同一性。一致性匹配的总数乘以100,然后被匹配跨度(matched span)内的较长序列的长度与为了比对两个序列而引入至较长序列内的空位数之和除。

术语“核酸”、“DNA序列”或“多核苷酸”是指单链或双链形式的脱氧核糖核苷酸或核糖核苷酸聚合物,并且除非另外限定,包括以与天然存在的核苷酸类似的方式与核酸杂交的天然核苷酸的已知类似物。
“启动子”是指调节与特定宿主细胞中的启动子可操作地连接的核酸序列的表达的DNA序列,而术语“可操作地连接”是指一种核酸片段与其他核酸片段连接从而使其表达功能受其他核酸片段影响。另外,启动子可以包括用于控制转录的操纵子序列、编码合适的mRNA核糖体结合位点的序列和控制转录和翻译的终止的序列。另外,其可以是组成型诱导靶基因的表达的组成型启动子,或是在特定位点和特定时间诱导靶基因的表达的诱导型启动子。

本文使用术语“类似物”来指与参照分子结构类似但是已经通过用备选取代基替换参照分子的特定取代基而以靶向受控方式进行修饰过的分子。与参照分子相比,本领域技术人员预期类似物会展示相同、相似或改进的功用性。为了鉴定具有改进的性状(例如针对靶分子更高的结合亲和力)而进行的类似物的合成和筛选是药理化学领域公知的方法。
当指蛋白质和/或蛋白质序列时术语“同源物”是指他们天然或人工源自共同的祖蛋白或蛋白序列。类似地,当核酸和/或核酸序列天然或人工源自共同的祖核酸或核酸序列时他们是同源的。
如本文所用,“接触”具有其正常含义,并且是指将两个以上试剂(例如两个多肽)结合,或将试剂与细胞(例如蛋白和细胞)结合。接触可以发生在体外,例如在试管或其他容器中将两个以上试剂结合或将测试试剂与细胞或细胞裂解物结合。接触还可以在细胞内或原位发生,例如通过在编码两个多肽的重组多核苷酸的细胞中共表达而在细胞中或细胞裂解物中使两个多肽接触。
术语“试剂”或“测试试剂”是指任何物质、分子、元素、化合物、实体或他们的组合。其包括但不限于例如蛋白质、多肽、有机小分子、多糖和多核苷酸等。其可以是天然产物、合成化合物、化学化合物或两种以上物质的组合。除非另外指出,术语“试剂”、“物质”和“化合物”可以相互替换地使用。
更具体而言,可以用本发明方法鉴定的测试试剂包括多肽、β转角模拟物、多糖、磷脂、激素、前列腺素、类固醇、芳香族化合物、杂环化合物、苯二氮卓(benzodiazepine)、低聚N取代甘氨酸、低聚氨基甲酸酯、多肽、糖、脂肪酸、类固醇、嘌呤、嘧啶或者他们的衍生物、结构类似物或组合。一些测试试剂是合成分子,而其他是天然分子。测试试剂获自各种来源,包括合成或天然化合物的文库。许多种能够以逐步方式合成的化合物可以用于产生组合文库。可以通过WO95/12608、WO93/06121、WO94/08051、WO95/35503和WO95/30642中所述的编码合成文库(ESL)方法构建化合物的大组合文库。肽文库也可以通过噬菌体展示法产生(参见例如Devlin,WO91/18980)。细菌、真菌、植物和动物提取物形式的天然化合物的文库可以获自商业来源或现场收集。可以对已知的药理学试剂进行定向或随机化学修饰,例如酰化、烷基化、酯化、酰胺化,以产生结构类似物。
测试试剂可以是天然存在的蛋白质或其片段。此类测试试剂可以获自天然来源,例如细胞或组织裂解物。多肽试剂的文库可以例如由商购的cDNA文库制备,或用常规方法产生。测试试剂可以是肽,例如具有约5~约30个氨基酸、优选约5~约20个氨基酸,特别优选约7~约15个氨基酸的肽。肽可以是天然存在的蛋白质、随机肽或“偏性(biased)”随机肽。
测试试剂还可以是“核酸”。核酸测试试剂可以是天然存在的核酸、随机核酸或“偏性”随机核酸。例如,原核或真核基因组的消化物可以类似地如上所述用于蛋白。
在一些优选方法中,测试试剂是小分子(例如分子量不超过约1,000的分子)。优选的是,将高通量检验改良并用于筛选此类小分子。许多检验可用于此类筛选,例如如Schultz(1998)Bioorg Med Chem Lett8:2409-2414;The present inventorsller(1997)Mol Divers.3:61-70;Fernandes(1998)Curr Opin Chem Biol2:597-603;和Sittampalam(1997)Curr Opin Chem Biol1:384-91中所述。
用于本发明的筛选方法的测试试剂的文库可以通过对GRS、其片段或其类似物进行结构研究而制备。与GRS结合的测试试剂可以通过此类结构研究鉴定。
GRS的三维结构可以以许多方式(例如晶体结构和分子建模)来研究。使用x射线结晶学研究蛋白结构的方法是文献中公知的。参见Physical Bio-chemistry,Van Holde,K.E.(Prentice-Hall,New Jersey1971),第221-239页,和Physical Chemistry withApplications to the Life Sciences,D.Eisenberg&D.C.Crothers(Benjamin Cummings,Menlo Park1979)。GRS结构的计算机建模提供为设计用于筛选GRS的测试试剂提供了另一手段。分子建模方法已经在文献中描述,例如题目为"System and method formolecular modeling utilizing a sensitivity factor"的美国专利第5,612,894号和题目为"Molecular modeling method and system"的美国专利第5,583,973号。另外,蛋白结构还可以通过中子衍射和核磁共振(NMR)来确定。参见例如Physical Chemistry,第4版,Moore,W.J.(Prentice-Hall,New Jersey1972),和NMR of Proteins and Nucleic Acids,K.Wuthrich(Wiley-Interrscience,New York1986)。
本发明人查明GRS(甘氨酰tRNA合成酶)与CDH相互作用,并调节ERK途径,由此调节细胞死亡和生长。即,本发明人确认GRS与CDH6相互作用,并通过PP2A调节ERK途径,由此调节细胞死亡和生长。
因此,本发明提供一种筛选用于预防或治疗癌的试剂的方法,所述方法包括以下步骤:
(a)在测试试剂存在或不存在下使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在测试试剂存在或不存在下测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂下GRS或其片段与CDH的结合亲和力与不存在测试试剂下GRS或其片段与CDH的结合亲和力进行比较;和
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂。
更具体而言,所述癌包括但不限于恶性黑素瘤、白血病、结肠癌、肺癌、肝癌、胃癌、食道癌、胰腺癌、胆囊癌、肾癌、膀胱癌、前列腺癌、睾丸癌、子宫颈癌、子宫内膜瘤、绒毛膜瘤、卵巢癌、乳腺癌、甲状腺癌、脑肿瘤、头颈癌、皮肤癌、淋巴瘤,其还可以包括诸如前体B细胞赘生物等B细胞赘生物、诸如前体T细胞赘生物等T细胞和NK细胞赘生物以及诸如经典型霍奇金淋巴瘤等霍奇金淋巴瘤(霍奇金病)。
此外,与GRS或其片段相互作用的蛋白可以是CDH(钙粘蛋白),优选的是,其可以是CDH6(K-钙粘蛋白,钙粘蛋白-6)或CDH18(钙粘蛋白-18)。CDH6或CDH18可以是Genbank中公开的序列。更具体而言,在CDH6的情况下,其可以是SEQ IDNO:12(Genbank登录号AAH00019.1)、SEQ ID NO:13(Genbank登录号NP_004923.1)或SEQ ID NO:14(Genbank登录号BAA06562.1)。更具体而言,在CDH6的情况下,其可以是SEQ ID NO:15(Genbank登录号AAH31051.1)、SEQ ID NO:16(Genbank登录号EAW68851.1)或SEQ ID NO:17(Genbank登录号NP_001035519.1)。在本发明的实例中,对于CDH6和CDH18使用SEQ ID NO.12和SEQ ID NO.15。
同时,筛选方法可以使用与CDH6相互作用的GRS片段。通过使用包含GRS片段(例如由SEQ ID NO:2表示的片段)的反密码子结合域的片段,本发明提供一种筛选用于预防或治疗癌的试剂的方法,所述方法包括以下步骤:
(a)在测试试剂存在或不存在下使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在测试试剂存在或不存在下测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂下GRS或其片段与CDH的结合亲和力与不存在测试试剂下GRS或其片段与CDH的结合亲和力进行比较;和
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂。
同时,在本发明中,筛选方法还包括以下步骤:(e)使步骤(d)中鉴定出的测试试剂与表达CDH的细胞接触;和(f)测定所述表达CDH的细胞的凋亡。因此,本发明的方法可以是筛选用于预防或治疗癌症的试剂的方法,所述方法包括以下步骤:
(a)在测试试剂存在或不存在下使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在测试试剂存在或不存在下测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂下GRS或其片段与CDH的结合亲和力与不存在测试试剂下GRS或其片段与CDH的结合亲和力进行比较;
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂;
(e)使步骤(d)中鉴定出的测试试剂与表达CDH的细胞接触;和
(f)测定所述表达CDH的细胞的凋亡。
本发明中,由于GRS或其片段通过与CDH相互作用经由ERK途径而诱导癌细胞死亡,因此改变GRS或其片段与CDH的结合水平的测试试剂与CDH相互作用,并且其可以诱导表达CDH的细胞的凋亡。测试试剂可以模拟GRS对CDH的结合。优选的是,改变GRS或其片段与CDH的结合亲和力的测试试剂可以是GRS或其片段与CDH的结合的抑制剂。
本发明中,表达CDH的细胞可以是表达CDH6或CDH18的细胞,并且优选的是,其可以是癌细胞,更优选的是,其可以是肾癌细胞、肝癌细胞、肺癌细胞、结肠癌细胞或源自/分化自他们的细胞。
本发明中,GRS和CDH的结合水平的改变可以是增加或减少。例如,GRS和CDH的结合水平的改变为增加可以通过诱导ERK的信号转导而刺激(或增加)细胞死亡,GRS和CDH的结合水平的改变为减少可以通过模拟GRS的功能而刺激(或增加)细胞死亡。
为了实施本发明可以使用本领域公知的各种生物化学和分子生物学技术或检验。此类技术例如描述于Sambrook等,Molecular Cloning:A Laboratory Manual,ColdSpring Harbor Press,N.Y.,第2版(1989)和第3版(2000);和Ausubel等,CurrentProtocols in Molecular Biology,John Wiley&Sons,Inc.,New York(1987-1999)。
优选的是,首先针对测试试剂的调节GRS和CDH的生物活性的能力而对测试试剂进行检验(第一检测步骤)。特别是,在第一步骤中,可以在测试试剂的存在下通过检验分离GRS的生物活性而鉴定调节所述多肽的生物活性的调节剂。更优选的是,本发明可以包括:
(a)在测试试剂的存在下使测试试剂与GRS或CDH接触;和
(b)测定GRS或CDH的活性,以及选择改变GRS或CDH的活性的测试试剂。
GRS或CDH的不同生物活性的调节可以在第一步骤中得到检验。例如,可以针对调节GRS或CDH的表达水平(例如转录或翻译)的活性而对测试试剂进行检验。还可以针对调节GRS或CDH的细胞水平或稳定性(例如翻译后修饰或蛋白水解)而对测试试剂进行检验。
通过第一检验步骤鉴定了增加生物活性的测试试剂,然后对测试试剂进行进一步测试,以测试测试试剂是否具有改变GRS和CDH的相互作用的能力(第二测试步骤)。
在第一步骤和第二步骤中,都可以使用完整的GRS和亚基或他们的片段、类似物或功能衍生物。在这些检测中可以使用的片段通常保留GRS的一种或多种生物活性。含有此类片段或类似物的融合蛋白也可用于筛选测试试剂。GRS的功能衍生物通常具有氨基酸缺失和/或插入和/或取代,同时保留了一种或多种生物活性,因此也可用于实施本发明的筛选方法。
可以使用各种公知技术来鉴定调节GRS或CDH的测试试剂。优选的是,用基于细胞的检验系统筛选测试试剂。例如,在通常的基于细胞的检验(即,第二筛选步骤)中,在测试试剂的存在下测定报道基因的活性(即,酶活性),然后与在不存在测试试剂下的报道基因的活性进行比较。报道基因可以编码本领域已知的任何可检测的多肽(响应或报道多肽),例如所述可检测多肽可以通过荧光或磷光或借助于其拥有的酶促活性而检测。可检测响应多肽可以例如为荧光素酶、α-葡糖醛酸酶、α-半乳糖苷酶、氯霉素乙酰转移酶、绿色荧光蛋白、增强绿色荧光蛋白和人分泌的碱性磷酸酶。
在基于细胞的检验中,还可从也存在于宿主细胞中的不同载体表达测试试剂(例如,肽或多肽)。在一些方法中,测试试剂的文库由此类载体的文库(例如,cDNA文库;参见以下实施例)编码。使用本领域公知的方法产生此类文库(参见例如Sambrook等和Ausubel等,见上文),或获自各种商业来源。
除了上述基于细胞的检验之外,还可以用基于非细胞的方法进行筛选。这些方法包括例如迁移率DNA结合检验、甲基化和尿嘧啶干扰检验、DNA酶和羟基自由基足迹分析、荧光偏振和UV交联或化学交联剂。对于综述,参见例如Ausubel等,见上文(第12章,DNA-Protein Interactions)。用于分离共结合蛋白(包括核酸和DNA/RNA结合蛋白)的一种技术包括使用UV交联或化学交联剂,包括例如可切割交联剂二巯基双(琥珀酰亚氨基丙酸酯)和3,3'-二巯基双(磺基琥珀酰亚氨基丙酸酯);参见例如McLaughlin,Am.J.Hum.Genet.,59:561-569,1996;Tang,Biochemistry,35:8216-8225,1996;Lingner,Proc.Natl.Acad.Sci.U.S.A.,93:10712,1996;和Chodosh,Mol.Cell.Biol.,6:4723-4733,1986。

具体实施方式
以下参照附图描述本发明的优选实施方式。
在以下说明和附图中,相同的附图标记用于指代相同或相似的组件,因此会省略相同或相似的组件的描述的重复。
<实验方法>
1.分泌检验
将RAW264.7细胞在条件培养基中培养。收集条件培养基,并用10%TCA使蛋白质沉淀。通过SDS-PAGE分离沉淀的蛋白质,并将其转移至PVDF膜以用于免疫印迹。
2.共培养检验
通过将U937或RAW264.7细胞(0.125×106个细胞~1×106/孔)接种至已经在6孔板上培养12小时的H460或HCT116细胞层(0.25×106/孔)上进行共培养实验。然后将细胞在无血清培养基中共培养6小时。为了评估GRS的分泌,收获培养基,然后用10%TCA使蛋白质沉淀。为了证明共培养时巨噬细胞/单核细胞与肿瘤之间的物理相互作用是否对于GRS分泌是必须的,使用配备有具有0.4μm孔的插入体(insert)的24孔Transwell细胞培养室(Costar,#3470)。将H460细胞接种在该室中,将U937细胞接种在插入体中,并且以1:2H460:U937的比例分别温育12小时。然后将插入体转移至培养有所述H460细胞的该室中。培养24小时后,除去插入体,并收获培养基以用于检验分泌。
3.凋亡检验
将测试细胞用不同浓度的重组GRS在指定时间处理,并将其收获。用PBS洗涤后,将细胞固定在70%乙醇中1小时,并用50mg/ml的碘化丙啶的PBS溶液染色。通过使用CellQuest软件(BD Biosciences)的荧光激活细胞分选仪对每份样品读取2000个细胞。对于MTT检验,向150ml培养基添加20ml的MTT溶液(5mg/ml)。温育4小时后,添加200ml DMSO。用微板阅读器(TECAN,Mannedorf,Swiss)测定570nm处的吸光度。通过使用他们各自抗体(Cell Signaling,Beverly,MA)的免疫印迹确定从前半胱天冬酶3生成了活性半胱天冬酶3。
4.可溶性受体结合检验
为了鉴定GRS的受体,使用Maxisorp板(Nunc,Rochester,NY)来进行ELISA检测。将板用溶于磷酸盐缓冲盐水中的纯化的带有his标签的GRS(1μg/ml)或BSA(1μg/ml)涂布,然后封闭(PBS,4%脱脂乳)。向所述板添加Fc-融合钙粘蛋白家族蛋白(1μg/ml),用PBST(0.05%Tween20)洗涤后,将板与抗人IgG1Fc-HRP(Thermo,Waltham,MA)一起温育。然后将板洗涤,添加TMB(3,3’,5,5’-四甲基联苯胺)溶液,并且在微板阅读器(TECAN,Mannedorf,Swiss)于450nM读数。
为了确认相互作用,使经纯化的Fc-CDH2、6、18(1μg/ml)与His-GRS(1μg/ml)一起温育2小时。使反应进行蛋白A/G琼脂糖与Fc-融合钙粘蛋白的免疫沉淀,并且通过使用抗-GRS的免疫印迹进行分析,以检测复合体。
5.异种移植物小鼠模型
动物实验遵守首尔国立大学的大学动物管理及使用委员会指南。通过使用20号针头将3×107个细胞皮下注入BALB/c裸雌鼠中并使其生长而测试HCT116、SN12和RENCA的致瘤性。每2天检查动物中的肿瘤生长,并且如果观察到肿瘤形成,则使用卡尺测定肿瘤(肿瘤体积计算为长度×宽度2×0.52)。使肾肿瘤(SN12,RENCA)生长5天,并每天腹膜内注射His-GRS蛋白持续4天。对于回归模型,使肿瘤细胞生长9天,然后通过肿瘤内注射来注射His-GRS蛋白。对于生长模型,使用或不使用His-GRS蛋白来注射肿瘤细胞。牺牲动物后,测定肿瘤重量,并将肿瘤包埋在最佳切断温度(OCT)化合物中以用于进行免疫荧光染色。将冰冻切片(10μm)附着至载玻片上,用PBS处理并用4%多聚甲醛固定,用含有2%CAS的PBS封闭,并用Yo-Pro-1(Invitrogen)在37℃染色2小时。用含有0.1%Tween20(Sigma)的PBS洗涤载玻片,并用DAPI(Invitrogen)在37℃对细胞核染色20分钟。将切片固定,并通过共聚焦免疫荧光显微镜(Nikon C-1共聚焦显微镜)进行观察。
6.细胞培养和材料
使RAW264.7、SH-SY5Y、HSF(人皮肤成纤维细胞)、HEK293、BV2、MCF-7、SN12、SK-BR-3、BT474和HeLa细胞在含有10%胎牛血清和50mg/ml链霉素以及青霉素的达尔伯克氏改良伊格尔培养基(Dulbecco's modified Eagle's medium,DMEM)生长。为了培养A549、H322、H460、HCT116、Caki-1、RENCA、HT1080和Jurkat细胞,使用具有相同如上所述补充物的RPM1640培养基。使用针对全长人GRS的兔多克隆抗体(Abcam,Cambrage,UK)进行蛋白质免疫分析。从Invitrogen(Carlsbad,CA)获得靶向CDH6的5′-UUUCAUAGAACUCA GCAAAUUCUGG-3′和5′-UAAUAAUGAAGAGAUCUCCUGCUCC-3′的Si-RNA。使用Stealth universal RNAi(Invitrogen)作为阴性对照。
7.重组人GRS的制备
将编码685个氨基酸的人GRS cDNA在EcoRI和XhoI限制性酶的位点处亚克隆至pET-28a(Novagen,Madison,WI),通过IPTG诱导在大肠杆菌Rosetta(Novagen)中过表达。然后使用镍亲和色谱(Invitrogen)按照制造商说明书对经His标记的GRS进行纯化。通过IPTG诱导在大肠杆菌Rosetta中表达GST-GRS片段。在含有0.5%TritonX-100的PBS缓冲液中使用谷胱甘肽琼脂糖对GST融合蛋白进行纯化。为了除去脂多糖(LPS),在含有100mM NaCl的10mM无热原磷酸钾缓冲液(pH6.0)对蛋白质溶液进行透析。透析后,含有GRS的溶液加载至已经用相同缓冲液预平衡过的多粘菌素树脂(Bio-Rad),温育2小时,然后进行洗脱。为了进一步除去残留LPS,再次在含有20%甘油的PBS中对溶液中进行透析,并用Mustang E膜(Pall Gelman Laboratory,Ann Arbor,美国)通过Acrodisc单元进行过滤。
8.GRS分泌检验
将RAW264.7细胞培养至70%汇合。然后将细胞洗涤两次,并在无血清或葡萄糖耗尽的DMEM培养基中进一步培养。另外,向无血清培养基添加阿霉素(1mg/ml,Calbiochem,San Diego,CA)、具有环己酰亚胺(1μg/ml,Sigma St.Louis,MO)的TNF-α(1ng/ml,BD Pharmingen,San Diego,CA)、Fas-配体(10ng/ml,Millipore)或抗-Fas抗体(5μg/ml,克隆体CH11,Millipore,Billerica,MA)。在指定时间收集培养基,以500g离心10分钟,并以20,000g再次离心15分钟,以除去污染物。用10%TCA从上清液中使蛋白在4℃沉淀12小时,然后以18,000g离心15分钟。收获球团,用100mMHEPES缓冲液(pH8.0)再悬浮,通过10%SDS/PAGE分离,并将蛋白质转移至PVDF膜以用于使用多克隆抗-GRS抗体进行免疫印迹。用抗-IgG抗体预先除去一部分上清液,并与抗-GRS抗体在4℃一起温育2小时。然后添加蛋白A琼脂糖(Invitrogen),在4℃将混合物温育4小时。接下来使蛋白A琼脂糖珠沉淀,用含有150mM NaCl、1%Triton X-100、10mM NaF、1mM原钒酸钠、10%甘油和蛋白酶抑制剂(Calbiochem)的50mM Tris-HCl缓冲液(pH7.5)洗涤3次,并使用样品缓冲液将结合蛋白洗脱。
9.细胞坏死检验和血浆膜完整性评估
将RAW264.7细胞接种到6孔皿上,并温育12小时。然后将细胞洗涤两次,并与指定的条件培养基一起温育。为了测定培养基的LDH浓度,收集条件培养基,然后以2,000g离心15分钟。收获上清液,并使用LDH-细胞毒性检验试剂盒(BioVision,Mountain view,CA)按照制造商说明书测定LDH酶活性。通过将释放的LDH表示为总细胞LDH的百分比,可以计算细胞存活率。为了通过免疫荧光染色监视膜完整性,将细胞接种在22×22mm盖玻片上,并温育24小时。用PBS洗涤培养皿2次,并与指定浓度的条件培养基一起温育。在4%多聚甲醛中将细胞固定10分钟,并用冷PBS冲洗2次。将盖玻片与溶于PBS中的3%CAS一起温育30分钟,然后与溶于PBS中的50mg/ml碘化丙啶、5μM Yo-Pro-1(Invitrogen)溶液一起温育1小时。使用4’,6-二脒基-2-苯基吲哚二盐酸盐(DAPI)对细胞核进行染色。将细胞固定并通过荧光显微镜(Nikon C-1共聚焦显微镜)观察。
10.细胞结合检验
将细胞接种到6孔皿上,并温育12小时。然后向培养基添加生物素化GRS,并进一步在指定时间中温育。将细胞用冷PBS洗涤4次,然后在含有150mM NaCl、2mM EDTA、1%Triton X-100、1%脱氧胆酸钠、10mM NaF、1mM原钒酸钠、10%甘油和蛋白酶抑制剂的50mM Tris-HCl(pH7.4)裂解缓冲液中裂解。通过SDS/PAGE对所提取的蛋白质(30mg)进行解析,并通过链亲和素缀合的辣根过氧化物酶(HRP)(Pierce,Rockford,IL)进行检测。对于生物素化,将重组GRS(1.5mg)与0.25mg溶于PBS中的硫代-NHS-SS-生物素(Pierce)一起在4℃温育2.5小时。为了通过免疫荧光染色监视GRS的细胞结合,将细胞接种在22×22mm盖玻片上,并温育12小时。然后将培养皿与生物素化GRS和生物素化BSA一起温育1小时。在4%多聚甲醛中将细胞固定10分钟,并用冷PBS冲洗2次。将盖玻片与溶于PBS的3%CAS一起温育30分钟,然后用Alexa488-缀合的链亲和素(Invitrogen)捕获标记有经结合生物素的GRS。将细胞固定,并通过共聚焦免疫荧光显微镜观察。为了通过流式细胞仪监视GRS的细胞结合,将细胞用特异性si-RNA转染,并温育48小时。将进行了如上所述处理的细胞用PBS洗涤进行3次,并用溶于FACS缓冲液(含有2%BSA的PBS)中的Alexa488-缀合链亲和素(Invitrogen)染色。然后,用PBS洗涤细胞3次。使用CellQuest软件(BD Biosciences,Mountain view,CA)通过流式细胞仪对细胞进行分析。
11.表面等离子共振分析
使用ProteOn XPR36蛋白相互作用检验系统(BioRad)通过SPR技术确定GRS与钙粘蛋白-Fc融合蛋白的结合。根据制造商说明书通过胺偶联方法将CDH6、18和IgG(阴性对照)固定在GLC金芯片上(各自为约1000RU)。将各种浓度的纯化GRS以100mL/min施加至含有0.005%Tween20的基于磷酸盐的盐水中的流动细胞60秒,然后解离600秒。通过共振单位的变化确定结合,其中将一个共振单位定义为1pg/mm2。通过从对照表面减去记录的结合反应而对Sensogram进行处理。使用ProteonManagerTM软件(版本2.1)计算平衡解离常数。使用Langmuir1:1结合模型对数据进行评价。
12.RT-PCR检验
将细胞在六孔皿上温育12小时,洗涤两次,然后进行刺激。通过使用具有随机六聚物的RNeasy小型试剂盒(QIAGEN,Valencia,CA)提取总RNA,并用对GRS具有特异性的引物和GAPDH进行RT-PCR。
13.细胞色素C释放检验
使用蛋白质免疫分子检查细胞色素C的易位。将HeLa细胞与150nM GRS一起温育指定时间,并将收获的细胞再悬浮于在冰上的含有10mM氯化钾、1.5mM MgCl2、0.5mM EDTA、1mM DTT和蛋白酶抑制剂(Calbiochem)的20mM的HEPES(pH7.5)的低渗缓冲液中5分钟,然后匀质化6次。以10,000g将样品离心10分钟。对上清液中的蛋白进行SDS-PAGE,然后用针对细胞色素C和微管蛋白的多克隆抗体进行探测。
<结果>
1.从巨噬细胞分泌GRS
在3个不同人受试对象和2个不同CL57BL/6小鼠的血清中检测GRS(图1A)。与细胞裂解物的缺少一致,在相同样品中都没有检出β-微管蛋白、LDH或GAPDH。这些观察促使发明人了解关于分泌的GRS的生理功能的更多信息。为了抓住癌微环境中的分泌GRS的生理功能的线索,检查了是否能够从培养细胞中检出GRS的分泌,以及如果能的话,该分泌是否对细胞类型具有特异性。使六种不同的细胞系(H322、A549、HEK293、HCT116、RAW264.7和HeLa)在无血清培养基中温育12小时,并且使用TCA使分泌蛋白从培养基中沉淀出。使用抗GRS多克隆抗体(α-GRS)通过蛋白质印迹分析确定沉淀蛋白。在6种测试细胞系中,仅在鼠类巨噬细胞系RAW264.7的培养基中检出GRS(图1A)。在相同条件下,我们观察到没有微管蛋白释放至培养基中,表明培养基中GRS的存在不归因于细胞裂解(图1A)。
为了进一步确认GRS的分泌,使培养的RAW264.7细胞的无血清培养基中的蛋白免疫沉淀,并用α-GRS进行印迹。全长GRS在用GRS获得的免疫沉淀物中特异性检出,而在用模拟(mock)IgG获得的免疫沉淀物中未检出(图1B)。分泌的时间曲线分析显示,对于RAW264.7细胞,GRS在饥饿12小时后在培养基中检出,并在随后时间某种程度增加(图1C)。
既然GRS分泌具有明显特异性,我们调查了其他免疫细胞,例如T淋巴细胞、Jurkat细胞、源自小神经胶质细胞的BV2细胞和巨噬细胞样U937单核细胞。GRS仅在U937和RAW264.7细胞的条件培养基中检出,而在Jurkat或BV2细胞中未检出(图1D)。因此,GRS在饥饿条件下从巨噬细胞/单核细胞分泌。
2.不同的凋亡胁迫诱导GRS的分泌
为了调查GRS分泌是否通过除了饥饿之外的其他刺激物诱导,用信号传导分子处理RAW264.7细胞。这些刺激物包括调节巨噬细胞中细胞因子产生的雌激素(Carruba等,2003)、影响巨噬细胞分化的的TNF-α(Witsell和Schook,1992)和能够从巨噬细胞和肿瘤细胞分泌的EGF。另外,测试了阿霉素通过DNA损伤诱导细胞死亡。用这四种“配体”中的每一种处理6小时后,仅阿霉素诱导GRS的分泌(图S1B)。这些结果表明凋亡胁迫对于GRS分泌是重要的。考虑到该可能性,用其他凋亡胁迫处理RAW264.7细胞,并比较GRS随时间的分泌。除了阿霉素之外,葡萄糖耗尽或具有环己酰亚胺的TNF-α处理导致GRS从RAW264.7的时间依赖性分泌,但是分泌自人结肠癌HCT116或子宫颈癌HeLa细胞(图8C)的GRS不是时间依赖性分泌的。这三种凋亡胁迫也诱导GRS从U937细胞分泌(图S1D)。与GRS相反,在阿霉素处理的U937细胞中未检出KRS或YRS以及其他已知的AARS细胞因子的分泌(图S1E)。
由于在这三种不同凋亡胁迫中任一种开始后立即观察到GRS分泌,GRS分泌不可能是由细胞裂解引起。为了确认该结论,通过监视sub-G1细胞周期群和通过进行MTT(3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐)检测而确定细胞裂解。发现所有凋亡胁迫都没有严重地影响细胞周期和存活率(图S2A和B)。还测定了条件培养基中细胞溶质乳酸脱氢酶(LDH)的活性。培养基中的LDH活性小于细胞提取物中活性的10%,并且在条件培养基中不增加(图S2C)。由于细胞膜完整性在凋亡或坏死时被破坏,对RAW264.7细胞用膜渗透性Yo-Pro染色,也用膜不渗透性碘化丙啶染色(Bhatnagar等,2007)。在诱导凋亡胁迫后没有染料使细胞核结构染色。但是,当对巨噬细胞用紫外线照射(200J/m2)然后温育18小时,两种染料都使细胞核DNA染色(图S2D)。这些结果与GRS分泌不是由细胞裂解或膜破坏产生相一致。
3.GRS分泌可以由源自癌细胞的Fas配体诱导
除了其他功能,巨噬细胞在癌细胞微环境的免疫监视中起重要作用。为了探索巨噬细胞分泌GRS是否受与肿瘤细胞的接近程度的影响,建立了使用H460人类大细胞肺肿瘤和U937细胞的共培养体系。显著地观察到GRS而不是KRS、YRS和WRS的分泌,并且分泌的量随着所添加的U937细胞的数量的增加而增加(图2A)。
在使用HCT116和U937细胞以及H460和RAW264.7细胞的共培养体系中也检测到GRS分泌。此外,将源自骨髓的巨噬细胞(BMDM)用于共培养体系。在该离体检验中,也看到GRS分泌(图2D)。接下来,为了确定GRS分泌是否需要肿瘤和巨噬细胞之间的物理接触,使用之前所述的条件培养基和Transwell培养皿(Khodarev等,2002)。显著的是,收获自H460细胞的条件培养基包括来自RAW264.7和U937细胞的GRS分泌。还通过使插入体中的U937细胞与Transwell皿的小室内的H460细胞一起温育而将U937和H460细胞分离。GRS的分泌随着U937细胞的数量而增加(图2F)。这些结果表明GRS的分泌不需要癌和巨噬细胞之间的物理接触。
由于肿瘤细胞可以通过分泌Fas配体来逃避免疫监视(Igney和Krammer,2002),并且由于GRS的分泌由凋亡胁迫和由共培养的肿瘤细胞诱导,检查了Fas配体是否参与GRS分泌。为了测试该可能性,向条件培养基中的U937细胞添加激动性Fas抗体,并且确定该添加是否触发GRS的分泌。实际上,处理后,GRS在培养基中以时间依赖性方式累积(图2G)。为了确认Fas配体和GRS分泌之间的关系,还向共培养体系中添加激动性Fas抗体。在中和效应后,GRS分泌在培养基中减少(图2H)。但是,如通过RT-PCR分析所确定,没有证据表明Fas影响GRS基因的转录(图S2E)。最后,用环己酰亚胺封闭重新蛋白合成不抑制GRS的分泌(图S2F)。这些结果与源自癌细胞的Fas配体分泌的GRS的模型一致。
4.分泌GRS的细胞结合
为了确定分泌GRS的靶细胞,将人乳腺癌MCF7、HeLa、HCT116和RAW264.7细胞与不同浓度的生物素化GRS一起温育。使用该检验,经检测生物素化GRS与HCT116、HeLa和RAW264.7细胞以剂量依赖性方式结合,但是不与MCF7细胞结合(图3A)(在相同条件下,未检测到生物素化BSA与4种细胞类型中任一种结合)。接下来,确定GRS与HeLa和HCT116细胞的结合的时间进程。与生物素化GRS的结合在与这些细胞一起温育5分钟后检出,并在15分钟至30分钟时增加至最大(图S3A)。为了确认细胞被GRS结合的特异性,将细胞与未经标记的GRS一起预温育15分钟,然后添加生物素化GRS。当细胞用未经标记的GRS预温育时,生物素信号显著受抑制(图S3B)。通过免疫荧光染色进一步监视GRS的细胞结合。染色强度随着GRS的量增加,但是通过添加未经标记的GRS而减少(图S3C),因此染色强度特别归因于GRS结合。总之,结果表明特定肿瘤细胞和巨噬细胞是分泌的GRS的靶标。
5.GRS对肿瘤细胞的促凋亡效果
由于GRS分泌由诸如Fas等凋亡配体(由肿瘤细胞分泌)诱导,并且分泌的GRS与肿瘤细胞结合,我们调查了GRS是否发挥旁分泌效果。分别地,通过使用MTT检验监视细胞存活率和死亡,和通过利用流式细胞仪监视sub-G1细胞群。当用GRS处理时,HCT116细胞的存活率以剂量依赖性方式降低,而RAW264.7细胞的存活率并非如此(图3B)。此外,由于用GRS处理,HCT116的sub-G1群增加,但HCT116的sub-G1群不增加(图3C)。通过半胱天冬酶3的激活而确认由GRS处理导致的凋亡性细胞死亡(图3D)。还通过监视Bax和Bcl-2的细胞水平以及细胞色素C的释放而测试了GRS的凋亡活性。虽然GRS不影响Bax的细胞水平,其减少抗凋亡介质Bcl-2的水平(图S3D),并且增加作为细胞死亡的已知特征物(signature)的细胞色素C的释放(图S3E)。由于在这些检验中热失活的GRS不发挥任何促凋亡效果(图3C和D),GRS的天然构象似乎对于其促凋亡活性而言是重要的。
为了调查GRS是否能够诱导生理环境中的细胞死亡,通过使用具有或不具有抗GRS抗体的Transwell小室来共培养HCT116和U937细胞。与U937细胞共培养时HCT116细胞的存活率减少。但是,α-GRS的添加损害了GRS对U937细胞的存活率的效果(图3E)。在进一步的实验中,GRS减少了不同肿瘤细胞系(包括人肺泡腺癌A549、H460、HCT116和HeLa细胞)的存活率。但是,GRS对MCF-7、成神经细胞瘤SH-SY5Y细胞或对人皮肤成纤维细胞(HSF)、人胚肾HEK293或RAW264.7细胞(图3F)无影响。因此,GRS对特定组的癌细胞发挥其促凋亡活性。
6.反密码子-结合结构域负责凋亡效果
为了鉴定负责GRS的前凋亡活性的结构域,我们使用人GRS的已知X射线晶体结构来指导4种不同片段的构造(图S4A和B)。在4种不同片段中,我们获得3种(F2~4)可溶形式(由于低稳定性和溶解度,我们未能获得GRS(F1)的N末端片段)。将F2~4片段分别添加至HeLa和RAW264.7细胞。浓度为150nM时,GRS和175个氨基酸的C末端反密码子结合结构域(ABD)都减少HeLa细胞的存活率。与之相反,F2或F3都不能与GRS或ABD的活性相比较。该结果表明ABD包含了GRS的促凋亡细胞因子活性(图S4C)。
7.作为GRS的功能受体的钙粘蛋白-6
既然分泌的N末端截短的WRS与VE-钙粘蛋白结合从而抑制血管发生(Tzima等,2005),并且已知数种钙粘蛋白(CDH)与肿瘤细胞存活和恶性有关(Cavallaro和Chrostofori,2004),我们测试了GRS是否能够与11种不同的CDH蛋白中的任一种结合。His-GRS与各自融合至人IgG1的Fc片段的11种不同的钙粘蛋白的细胞外结构域之间的相互作用通过ELISA检验来确定。该筛选鉴定了GRS与CDH6或CDH18特异性结合,但不与其余CDH特异性结合(图4A)。通过体外拉下(pull-down)检验确了GRS与CDH6和CDH18结合,而不与CDH2结合(图4B)。
GRS与CDH6和CDH18的结合亲和力通过表面等离子共振检验来测定。GRS与CDH6和CDH18结合的KD分别为3.4和1.25nM(图4C和图S5A),但GRS不与IgG蛋白结合(图S5B)。在进一步研究中,在拉下检验中,仅由片段F4编码的ABD结构域与CDH6结合,但由片段F2或F3编码的ABD结构域不与CDH6结合(图4D)。并且还使用ELISA检验确认了仅F4片段与CDH6结合,而F2或F3片段不与CDH6结合(F1片段不可溶)(图4I)。
由于CDH6似乎涉及肝细胞癌、肾癌和小细胞肺癌(SCLC)的病因(Shimoyama等,1995;Li等,2005),调查了CDH6是否能够充当肿瘤细胞中的GRS的功能受体。首先,确定了在测试的肿瘤细胞系中的CDH6的表达水平。发明人发现CDH6的表达在GRS-敏感性癌细胞系(HCT116、H460和HeLa)中上调,但在GRS-非敏感性癌细胞系(MCF7和SH-SY5Y)中不上调(图4E)。使用蛋白质印迹分析(图4F)和流式细胞仪,发现当CDH6转录被其特异性si-RNA抑制时GRS与HCT116细胞的结合减少,但是不被非特异性对照siRNA减少(图4G和S5C)。然后测试了CDH6的可溶性细胞外结构域是否能够抵消GRS的凋亡活性。发现GRS对HCT116细胞的存活率的影响由于添加可溶性CDH6受体蛋白而消失(图4H)。结果表明CDH6是肿瘤细胞中的GRS的功能受体。
8.GRS通过抑制ERK激活而诱导细胞死亡
为了调查GRS处理诱导细胞死亡的机制,用GRS处理HCT116细胞,并且确定了对3种有丝分裂原激活的蛋白激酶ERK、p38MAPK和JNK的影响。发现磷酸化的ERK和p38MAPK而不是JNK的水平被GRS处理以剂量和时间依赖性方式减少(图5A和S6A)。由于已知ERK的激活涉及癌细胞增殖和存活(Warner和Mclntosh,2009),测试了癌细胞对GRS的凋亡易感性是否由ERK的激活状态确定。令人感兴趣的是,GRS-敏感性HCT116、H460和HeLa癌细胞显示了增强的ERK的磷酸化,而GRS-非敏感性SH-SY5Y和MCF7癌细胞不显示(图5B)。该结果提供了ERK的激活状态与癌细胞对GRS的敏感性之间的正相关。
在3种GRS-敏感性细胞系中,已知HCT116和H460细胞含有导致ERK激活的KRAS突变。Ras-Raf-MEK-ERK级联是癌细胞增殖和存活的关键信号传导途径(Roberts和Der,2007)。因此测试了ERK被致癌性Ras突变的激活是否对GRS处理赋予凋亡敏感性。对于该目的,选择通常对GRS处理不敏感的HEK293细胞。将3种不同的Ras活性突变(KRAS、HRAS和NRAS)引入HEK293细胞中,建立了稳定的细胞系。所选择的稳定细胞系表达转染Ras突变体的每一种,并且在引入突变后,显示了急剧增加的ERK的磷酸化。然后用GRS处理细胞系,并研究对ERK磷酸化的影响。引人注目的是,在所有3种细胞系中,GRS抑制了ERK的磷酸化(图5C)。接下来,检查细胞存活率和Ras转染的HEK293细胞是否受GRS处理影响。分别使用MTT检验和流式细胞仪,发现GRS处理抑制三种Ras转染体的细胞存活率和增加其死亡。与之相反,GRS处理对对照HEK293细胞没有影响(图5D和S6B)。结果与以下假设一致,即,通过激活的ERK的去磷酸化,GRS能够诱导Ras激活的肿瘤细胞的凋亡。
然后检查ERK磷酸化受CDH6的负调节是否普遍适用于所有细胞类型。在所测试的细胞系中,HCT116、肾癌SN12、乳腺癌细胞SK-BR-3和肾癌Caki-1显示了高水平的CDH6和磷酸化ERK(图S6C)。但是,肾癌细胞系RENCA、纤维肉瘤细胞系HT1080显示了高水平的ERK磷酸化以及不可检测水平的CDH6水平。与之相反,虽然存在高水平的CDH6,乳腺癌细胞系BT474显示了低水平的ERK磷酸化。另外,用K-、N-和HRAS转染HEK293细胞不影响CDH6的细胞水平(图S6D)。所有这些结果都暗示CDH6的细胞水平与ERK激活不直接相关。
测试了各种细胞系对所施用GRS的凋亡胁迫的易感性。仅具有高水平的CDH6和ERK磷酸化的细胞对GRS诱导的凋亡具有易感性(图5D)。GRS诱导的凋亡与高水平的CDH6蛋白和ERK磷酸化的相关性扩展至8个细胞系。
ERK的磷酸化不仅受MEK1和2激酶调节,还受磷酸盐调节(Keyse,2000;Gronda等,2001;Wang等,2003)。但是,MEK的调节不因GRS处理而减少(数据未示出)。为了调查GRS是否能够通过激活磷酸酶而抑制ERK磷酸化,我们使用4种不同的磷酸酶抑制剂处理HCT116细胞,并且检查他们会如何影响GRS诱导的ERK磷酸化。在4种磷酸酶中,冈田酸磷酸酶(PP2A)抑制剂特异性封闭ERK的GRS依赖性抑制(图5E)。GRS处理还减弱了CDH6和PP2A之间的相互作用(图5F和G),并且减少了PP2A的磷酸化(图S6E),从而由此增强PP2A的活性(Chen等,1992;Janssens和Goris,2001)。同时,PP2A和ERK之间的相互作用增加(图5H)。这些结果表明GRS能够从CDH6释放PP2A,然后诱导PP2A活性,其进而使ERK去磷酸化。
9.体内GRS的抗肿瘤效果
为了调查GRS在体内也具有活性的可能性,在使用HCT116细胞的异种移植物模型中测试了GRS。在第一实验中,将HCT116细胞注入Balb/C裸小鼠中,使肿瘤生长直到他们达到100mm2的平均尺寸。在第9天,将仅载体(vehicle)或者10mg或20mg GRS直接递送至肿瘤。在第21天在载体处理组中肿瘤体积增加约2.5倍。与之相反,GRS注射以剂量依赖性方式减少肿瘤体积(图6A和B)。在分别用10μg和20μg的GRS处理后,肿瘤重量减少至56%和34%。此外,动物体重和体型没有变化,表明没有因为施用GRS而发生明显的毒性(图6B和D)。使用免疫荧光分析来调查在用20mg GRS注射的肿瘤中是否诱导凋亡。在经GRS处理的肿瘤组织中比在对照组织中出现更多数量的Yo-Pro-1阳性肿瘤细胞(图6E)。
接下来,检查了GRS对肿瘤发生起始阶段的影响。将HCT116细胞与或不与GRS(20μg)一起注入裸小鼠。虽然在载体对照中肿瘤体积增加至185mm2,当GRS与所述细胞共注射时肿瘤没有生长(图6F和G)。与载体对照相比,在用GRS处理后肿瘤重量减少了46%(图6H)。在经GRS处理的动物中的凋亡可以清楚地看到(图6J)。再次由动物体重和体型不变化可以明显看出,未观察到由于施用GRS导致的明显毒性(图6G和I)。
GRS的ABD(F4)而不是催化性结构域(F2)足以在基于细胞的检验中引发凋亡活性。为了测试GRS的两个结构域之间的该区别是否也在体内看到,将HCT116细胞与20mg的催化性结构域(F2)或ABD片段一起注入Balb/C裸小鼠中。片段F4抑制肿瘤生长,但F2没有(图S7A和B)。与单独注射GRS相比,肿瘤重量的减少37%和46%(图S7C)。两个片段都几乎不给出由于其注射而导致的毒性(图S7D)。因此,在体外基于细胞的检验中看到的ABD的凋亡活性也在体内重演。
10.体内GRS的CDH6依赖性抗肿瘤效果
由GRS对引发了CDH6和ERK磷酸化标记的致瘤性细胞诱导的强健凋亡活性,增加了体内GRS的抗肿瘤活性可能具有CDH6依赖性的可能性。为了测试该可能性,检查了肾癌细胞中GRS的细胞结合,其与CDH6相关。GRS仅与表达高水平的CDH6的肾癌细胞SN12结合,但不与表达低水平的CDH6的RENCA细胞结合(图7A和C),表明GRS的活性依赖于CDH6的表达水平。该观点进一步被使用利用这两种细胞系的异种移植物模型的GRS体内抗肿瘤活性测试确认。为此,我们将SN12和RENCA细胞移植到裸小鼠中,并通过腹膜内注射以两个不同剂量(2mg/kg和6mg/kg)注射GRS蛋白4次。在这两个模型中,GRS仅抑制SN12的生长(图7B),而不抑制RENCA细胞的生长(图7D)。在分别用2mg/kg和6mg/kg的GRS处理后,SN12的肿瘤重量减少了36%和58%。但是,在相同条件下RENCA的肿瘤重量仅减少了7%和15%。动物体重不改变表明在两个异种移植物小鼠模型中都没有发生明显的毒性(图S7E和F)。为了确定GRS对ERK处理状态的影响,还通过蛋白质印迹监视ERK的磷酸化。在SN12异种移植物样品中,在GRS处理组中清楚地观察到ERK的去磷酸化(图7F),进一步验证了使用体外基于细胞的检验看到的GRS的CDH6依赖性凋亡活性也在体内重演。
产业可利用性
如上可以看出,本发明涉及GRS和CDH的新用途,并提供筛选用于预防或治疗癌症的试剂的方法。所述方法可以用于开发用于治疗各种癌症的新试剂。
Claims (4)
1.一种筛选用于预防或治疗癌的试剂的方法,所述方法包括以下步骤:
(a)在存在或不存在测试试剂时使GRS(甘氨酰-tRNA合成酶)或其片段与CDH(钙粘蛋白)接触;
(b)在存在或不存在测试试剂时测定GRS或其片段与CDH的结合亲和力;
(c)将在存在测试试剂时GRS或其片段与CDH的结合亲和力与不存在测试试剂时GRS或其片段与CDH的结合亲和力进行比较;
(d)鉴定改变GRS或其片段与CDH的结合亲和力的测试试剂;
(e)使步骤(d)中鉴定出的所述测试试剂与表达CDH的细胞接触;和
(f)测定所述表达CDH的细胞的凋亡,
其中所述CDH是CDH6或CDH18,
其中所述片段由SEQ ID NO:2或SEQ ID NO:11表示的氨基酸序列组成,
其中所述癌选自结肠癌、肺癌、肝癌和肾癌组成的组。
2.如权利要求1所述的方法,其中,所述GRS是选自由SEQ ID NO:1和SEQ IDNO.5~SEQ ID NO:10表示的氨基酸组成的组中的氨基酸序列。
3.如权利要求1所述的方法,其中,所述CDH6是选自由SEQ ID NO:12、SEQID NO.13和SEQ ID NO:14表示的氨基酸组成的组中的氨基酸序列。
4.如权利要求1所述的方法,其中,所述CDH18是选自由SEQ ID NO:15、SEQID NO.16和SEQ ID NO:17表示的氨基酸组成的组中的氨基酸序列。
