脂肪酶,即三酰基甘油酰基水解酶,是一种特殊的酯键水解酶,它可以在油水界面上催化油脂的水解反应,生成脂肪酸和甘油、甘油单酯或二酯。脂肪酶通常被用于在水相体系中催化酯类化合物的水解反应,在非水相体系中催化醇类化合物的酯化、转酯反应化,在非水相体系中催化氨酰化反应等。(Current Opinion in Chemical Biology 2002, 6,145-150.)
南极假丝酵母( Candida antarctica) 首先是在南极洲分离得到的,该南极假丝酵母可以产生两种性质完全不同的脂肪酶,分别为南极假丝酵母脂肪酶A(CALA)和脂肪酶B(CALB)。Patkar等人1993年成功分离纯化分别得到了CALA和CALB(Indian. J. Chem. 199332B. 76-80)。1994年,Uppenberg等研究出CALB自由酶的三维立体结构和氨基酸顺序(Structure 1994, 2, 293-308),发现CALB由317个氨基酸残基组成,分子量为33kD,与其它脂肪酶的氨基酸序列对比发现它们之间没有明显的同源性。1995年CALB被成功克隆并在米曲霉(Aspergillns oryzae)(Canadian Journal of Botany 1995, 73, 869-875.)中得到表达,该专利为诺和诺德公司所拥有。为了进一步研究CALB的性质及提高产量,CALB又陆续在许多种宿主中被克隆表达,如毕赤酵母(Pichi apastoris),酿酒酵母(Saccharomyces cerevisiae)和大肠杆菌(Escherichia Coli)等。CALB对水溶性和非水溶性物质都表现出较高的催化活性,是用于许多反应的催化剂,并且被用于例如立体选择性转化和聚酯合成,特别是对于仲醇的高对映选择性使得CALB目前在生物技术的使用中成为最重要的酶。
天然的CALB仍存在一定的缺陷,如热稳定性较差产量较低等,使其在生产实践中的应用受到一定限制,成功修饰和改造CALB使其具有更加良好的性质是一个研究的热点。2005年Magnusson等人在Trp104处引入点突变,Trp104Ala突变株对4-庚醇和5-壬醇的特异性比野生型酶分别提高了270倍和5500倍(Chembiochem 2005, 6, 1051-1056.)。2009年Seo等将CALB分别与苹果酸脱氢酶、亚精胺腐胺周质结合蛋白质和肽基脯氨酰顺反异构酶进行融合并在大肠杆菌中表达,融合表达的CALB溶解性得到大大提高,从而增加了其表达量(BBA - Proteins & Proteomics 2009, 1794, 519-525.)。
布洛芬类化合物( profens) 是一类重要的药物,主要用于治疗疼痛、发烧、炎症等,是非甾体类抗炎药物( NSAIDs) 中重要的一类。这类药物临床已广泛应用包括萘普生、布洛芬、酮洛芬、氟比洛芬等(图1)。布洛芬类药物的α位含有一个手性碳原子,存在一对光学异构体,尽管目前布洛芬类药物以消旋形式存在,旦其中S构型布洛芬具有明显较高的临床效果。
利用生物催化方法合成布洛芬类化合物可通过多种生物催化剂催化的反应如水解、酯化、氧化、还原等反应来进行,其中通过脂肪酶催化的动力学拆分反应来制备光学纯的布洛芬类化合物便是可行路线之一,如脂肪酶通过水解或醇解羧酸酯、酯化羧酸等反应来拆分已有相关报导(Green Chem, 2011, 13, 2607.),但是南极假丝酵母脂肪酶B对布洛芬类化合物的酯衍生物几乎没有或仅有很低的对映选择性(Enzyme Microb Tech, 2006,39, 924; J Org Chem, 1994, 59, 4410; Biotechnol Bioeng, 2001, 75, 559.),这就的需要对现有的方法路线进行改进,以提高脂肪酶对布洛芬类化合物的对映选择性至理想水平。
发明内容
本发明的技术方案如下:通过对南极假丝酵母菌脂肪酶B进行突变,提高其对布洛芬类化合物的对映选择性,使通过脂肪酶动力学拆分路线高效、可行。
本发明由野生型南极假丝酵母菌脂肪酶B(Candida antarctica lipase B)基因(SEQ ID NO.1)通过突变得到具有改良性质的脂肪酶,该酶在毕氏酵母工程菌中表达,该脂肪酶突变体,能够将布洛芬类化合物的酯衍生物水解,动力学拆分得到具有光学活性的S构型布洛芬类化合物(图2)。在本发明的实施方案中,该脂肪酶与天然存在的CALB野生型相比,在将底物水解为产物方面具有一个或多个改善的性质。其能够以SEQ ID NO.02蛋白质(南极假丝酵母脂肪酶B,Candida antarctic lipase B,缩写为CALB)的至少1.5倍活性将布洛芬类化合物的酯衍生物水解为至少70%对映体过量的S构型的布洛芬类化合物。
在进一方面,该重组脂肪酶包括与SEQ ID NO. 02 至少85%同一性的氨基酸序列,并且具有对应于SEQ ID NO. 02的189位残基为芳香族氨基酸残基,对应于SEQ ID NO. 2的190位残基为非极性氨基酸残基,这些氨基酸突变对于改善脂肪酶的活性剂对映选择性至关重要。
在进一方面,所述脂肪酶突变体包含以下一项或多项突变,39残基位为非极性氨基酸残基;40位残基为非极性氨基酸残基;138位残基为极性不带电荷氨基酸残基;139位残基为极性不带电荷氨基酸残基;152位残基为极性带正电荷的氨基酸残基;154位残基为非极性氨基酸残基;157位残基氨基酸残基为极性氨基酸残基。
本发明中涉及的芳香族氨基酸为Phe、Tyr,非极性氨基酸为Ala、Val、Leu、Ile、Pro、Phe、Trp、Met,极性不带电荷氨基酸为Gly、Ser、Thr、Cys、Tyr、Asn、Gln,极性带正电荷的氨基酸残基为Lys、Arg、His。
进一方面,本发明的一些实施方案中,含有突变的脂肪酶变体针对布洛芬类化合物的酯衍生物展现出更高的活性及选择性,这些突变包含下列一项或者多项突变,氨基酸序列189位残基为Phe (SEQ ID NO. 4, 10, 12),190位残基为Leu或Ile (SEQ ID NO. 6,8, 10, 12),39位残基为Ala或Val (SEQ ID NO. 34),40位残基为Ala或Val (SEQ ID NO.34),138位残基为Ser、Thr或Cys (SEQ ID NO. 14, 16, 18, 36, 38, 40, 42),139位残基为Tyr、His者Arg (SEQ ID NO. 14, 16, 36, 38, 40, 42),152位残基为Lys或His (SEQID NO. 30, 32),154残基位为Gly或Ala (SEQ ID NO. 20, 22, 24, 26, 28, 30, 32,36, 38, 40, 42),157位残基为Ser、Thr、Asp或Gly (SEQ ID NO. 20, 22, 24, 26, 28,30, 32, 36, 38, 40, 42)。
本发明的实施方式包括编码所述脂肪酶变体的核酸,例如与编码本发明所述CALB变体的核酸序列具有至少85%的序列同一性。本发明的相关实施方式包括包含这些核酸的载体及包含该类载体的宿主细胞。
本发明还包括脂肪酶突变体在动力学拆分手性羧酸酯类化合物中的应用。所述的手性羧酸酯类化合物为如式(I)所示的化合物,
其中R1为烷基、苯基、带有取代的苯基,苯基的取代基为卤素、烷基、苯基或带有取代的苯基、杂环等;
R2为烷基;
R3为烷基、苯基或带有取代的苯基,苯基的取代基为卤素、烷基或硝基。
在pH 5-9的磷酸盐缓冲液溶液中,在本发明所述的脂肪酶突变体的催化下,手性羧酸酯类化合物进行水解,生成光学活性手性羧酸。
作为优选,所述的手性羧酸酯类化合物为布洛芬类化合物的酯衍生物,其结构如式(II)所述,布洛芬类化合物为布洛芬、氟比洛芬、阿利洛芬、非诺洛芬、萘普生或酮洛芬,
其中R1为式(III)所述结构;
R2为烷基、苯基或带有取代的苯基,苯基的取代基为卤素、烷基或硝基。
在pH 5-9的磷酸盐缓冲液溶液中,加入定量的用助溶剂溶解的布洛芬类化合物的酯衍生物底物,之后加入定量的脂肪酶变体催化水解反应,该脂肪酶变体可以多种可行的方式如粗酶粉、酶溶液、固定化酶等多种形式存在。
本发明的有益技术效果:通过对南极假丝酵母菌脂肪酶B进行突变,提高了其对布洛芬类化合物的对映选择性。

具体实施方式
下面通过具体的实施方案描述本发明的脂肪酶突变体以及利用该酶动力学拆分布洛芬类化合物的酯衍生物。除特别指明,本发明中所用的实验方案均为本领域技术人员所公知的方法。此外,实施例应理解为说明性的,而不是限制本发明。
某些术语的定义。
动力学拆分实现对映体拆分的一种方法,在本发明中指脂肪酶催化布洛芬类化合物的酯衍生物中S构型的底物水解产生S构型的布洛芬类化合物,而R构型的底物不能反应或者反应较慢,从而达到拆分的效果。
对映体过量定义为对映体混合物中一个异构体A比另一个异构体 B多出来的量占总量的百分数,缩写为ee,公式为(A-B)/(A+B)*100%,对映体过量值用于表示一种手性化合物的光学纯度。ee值越高,光学纯度也越高。

实施例涉及培养基配方
a.YPD培养基:10g酵母提取物、20g蛋白质胨溶于900mL蒸馏水(如配置平板,可在灭菌前加入20g琼脂),115℃高压灭菌30min,使用前加入100 mL 20%葡萄糖溶液。
b.YPDS转化平板:YPD培养基+15g/l琼脂+1M山梨醇。
c.10*YNB:134g YNB溶于1000mL蒸馏水,过滤除菌,4℃保存。
d.500*生物素:20mg 生物素溶于10mL蒸馏水,取1mL稀释100倍,过滤除菌,4℃保存。
e.10*甲醇:5mL 甲醇与95mL蒸馏水混合,过滤除菌, 4℃保存。
f.20%葡萄糖:200g 葡萄糖溶于1000mL蒸馏水,过滤除菌, 4℃保存。
g.10*甘油:100mL 甘油与900mL蒸馏水混合,115℃高压灭菌30min,4℃保存。
h.BMGY培养基:10g酵母提取物、20g蛋白质胨溶于700mL蒸馏水,120,115℃高压灭菌30min,用前加入100mL 1M pH6.0磷酸钾缓冲液、100mL 10*甘油、100mL10*YNB、2mL 500*生物素。

实施例1南极假丝酵母(cgmcc 2.2366)基因组DNA的提取及野生型脂肪酶B基因的克隆
南极假丝酵母cgmcc 2.2366 用YPD液体培养过夜后,发酵液用1mL离心管3000r/min离心5min,弃上清收集菌体,可重复几次以得到足够量的细胞;b)菌体用足够量的DNA裂解液重悬,加入0.4g酸洗玻璃珠,再加入200μL酚:氯仿:异戊醇,盖紧管盖,以防涡旋时酚漏出;c)涡旋仪高速涡旋3min;d)12000 r/min离心5min,上清液(约200μL)转移至新的离心管;e)上清液加入700加入预冷的无水乙醇后,-20℃无水放置1h,12000 r/min离心15min沉淀DNA,倒掉乙醇,沉淀用800乙醇,沉淀乙醇混悬,12000 r/min离心15min沉淀DNA,真空干燥;f)DNA用100μL 1*TE重溶。

a)从种子培养基中取0.4mL接种于20mL LB液体培养基中培养3h;b)3000r/min,5min分两次富集2mL菌体于1.5mLEP管中,弃上清;c)加入100弃上冰冷TSS溶液,重新悬浮菌体,冰浴30min;d)加入20μL连接液(空质粒pPICZα A酶切片段、目的片段酶切片段及连接液)轻轻旋转混匀,冰浴30min;e)42℃热休克60s,冰浴2min,加入600μL LB液体培养基。37℃培养,150r/min振荡培养1h;f)分别取150μL涂布于LLB (ZeocinTM)抗性平板。

实施例5制备酵母感受态细胞
a)挑取P. pichia X33单菌落至5mL YPD培养基中,30℃振荡培养过夜。次日取适量活化菌液,接种50mL YPD,30mL振荡培养至OD600为1.3~1.5。b)3000 r/min ,离心5min收获菌体。c)用50mL的灭菌去离子水洗涤菌体,3000 r/min,400离心5min。d)用25mL的灭菌去离子水洗涤菌体,3000 r/min,离心15min。e)将菌体重悬于20mL 1mol/L sorbital中,3000 r/min,5min。f)菌体重悬于1mL 1mol/L sorbital中,并分别取80μl至1.5mL离心管,冰水中放置,当天使用。
实施例6线性化质粒转化酵母细胞
a)取80μl感受态细胞,加入5~10μl线性化浓缩质粒混匀。b)转入2mm电转杯中,0℃培养5min。c)电转条件:电压1800V,脉冲时间一次(5~10sec)。d)立即加入1mL的1 mol/L 山梨醇溶液到电转杯中,并转移到1.5mL消毒管中,30℃静止培养2~3h。e)分别取50-200μl电转化菌液到YPDS培养皿中涂布,30℃培养2~4天,可见单菌落。
实施例7 突变库表达
取单菌落接于96孔板深度培养(每个孔1 ml BMMY培养基zeocin (100 µg/mL),220 rpm 28 °C培养4天,每天加1%甲醇诱导表达。后离心取上清液进行筛选。测序,得到如序列表所述的序列。
实施例8 4 -硝基苯基酯的制备
4 -硝基苯基酯的制备体系如下:0 °C,20ml二氯甲烷中加入布洛芬化合物相应酸(2.4 mmol),4-硝基苯酚 (0.36 g, 2.6 mmol),N, N'-二环己基碳化二亚胺(0.70 g, 3.4mmol)。搅拌反应4小时后,过滤出去沉淀物,滤液真空干燥。液相检测。
实施例9 动力学拆分
反应体系:0.2M pH7.0磷酸钠缓冲盐200μl,如实施例7所述的上清液100 µL,底物20μl, 28 °C反应。反应液用二氯甲烷萃取2次。
实施例10高效液相色谱分析底物和产物
检测布洛芬及布洛芬对硝基酚酯的条件为:色谱柱:OJ-H,流动相:正己烷:异丙醇:TFA=93:7:0.1,流速:0.7ml/min,波长:240nm,柱温:30°C,检测器:UV检测器。
检测氟比洛芬及布氟比洛芬对硝基酚酯的条件为:色谱柱:AD-H,流动相:正己烷:乙醇:TFA=90:10:0.1,流速:0.8ml/min,波长:254nm,柱温:30°C,检测器:UV检测器。
检测酮洛芬及酮洛芬对硝基酚酯的条件为:色谱柱:AD-H,流动相:正己烷:异丙醇:TFA=87:13:0.1,流速:0.8ml/min;波长:254nm,柱温:30°C,检测器:UV检测器。

1.一种脂肪酶突变体,其氨基酸序列如SEQ ID NO.10,14,16,20,30。
2.一种核酸,其能够编码权利要求1所述的脂肪酶突变体。
3.如权利要求2所述的核酸,其核酸序列如SEQ ID NO.9,13,15,19,29。
4.一种表达载体,该载体含有权利要求2或3所述的核酸且能在宿主细胞中进行表达。
5.一种宿主细胞,其含有权利要求2或3的核酸或权利要求4所述的表达载体。
6.如权利要求5所述的宿主细胞,该宿主细胞是毕氏酵母。
7.权利要求1所述的脂肪酶突变体在动力学拆分手性羧酸酯类化合物中的应用。
8.如权利要求7所述的应用,其特征在于,所述的应用包括下述方法:在pH 5-9的磷酸盐缓冲液溶液中,在权利要求1所述的脂肪酶突变体的催化下,手性羧酸酯类化合物进行水解,生成光学活性手性羧酸。
9.如权利要求8所述的应用,其特征在于,所述的手性羧酸酯类化合物为布洛芬类化合物的酯衍生物,其结构如式(II)所述,布洛芬类化合物为布洛芬、氟比洛芬、阿利洛芬、非诺洛芬、萘普生或酮洛芬,
其中R1为式(III)所述结构;
R2为烷基、苯基或带有取代的苯基,苯基的取代基为卤素、烷基或硝基。
