技术领域
本发明涉及一种4-二氟甲氧基苯胺的制备方法,属于药品制备领域。
技术背景
4-二氟甲氧基苯胺是泮托拉唑和拟除虫菊酯类杀虫剂重要的中间体,其合成都是采用扑热息痛为原始原料,以乙醇作溶剂,扑热息痛首先与氢氧化钠反应生成对乙酰氨基苯酚钠,然后在碱性条件下再和二氟一氯甲烷反应生成4-二氟甲氧基乙酰苯胺;以甲醇作为溶剂,4-二氟甲氧基乙酰苯胺在碱性条件下乙酰氨基发生水解,乙酰基脱保护得到4-二氟甲氧基苯胺,两步收率65%,尤其第二步水解反应收率仅为55%,此合成路线存在收率低,成本高,产品质量差,污染重等弊端。
发明内容
本发明公开了一种工艺收率高,成本低,质量好,污染轻,适宜工业化生产的4-二氟甲氧基苯胺。
一种4-二氟甲氧基苯胺的制备方法,该制备方法的步骤为:
首先,将4-硝基苯酚于氢氧化钠反应生产对乙酰氨基苯酚钠,然后在碱性条件下再和二氟一氯甲烷反应生成4-二氟甲氧基硝基苯;
4-二氟甲氧基硝基苯在三氧化铁和活性碳共同催化下,以水和肼为还原剂将硝基还原成为氨基生产4-二氟甲氧基苯胺。
本发明的优点在于该方法是将4-硝基苯酚为起始原料,二氧六环作为溶剂,两步总收率达90%,得到4-二氟甲氧基苯胺纯度达98.5%以上,此工艺收率高,成本低,质量好,污染轻,适宜工业化生产,与同行业相比极具竞争优势。

一种4-二氟甲氧基苯胺的制备方法,该制备方法的步骤为:
首先,将4-硝基苯酚于氢氧化钠反应生产对乙酰氨基苯酚钠,然后在碱性条件下再和二氟一氯甲烷反应生成4-二氟甲氧基硝基苯;
4-二氟甲氧基硝基苯在三氧化铁和活性碳共同催化下,以水和肼为还原剂将硝基还原成为氨基生产4-二氟甲氧基苯胺。该方法制备的4-二氟甲氧基苯胺两步总收率达90%,得到4-二氟甲氧基苯胺纯度达98.5%以上,此工艺收率高,成本低,质量好,污染轻,适宜工业化生产,与同行业相比极具竞争优势。

4-二氟甲氧基硝基苯在三氧化铁和活性碳共同催化下,以水和肼为还原剂将硝基还原成为氨基生产4-二氟甲氧基苯胺。
