Abstract
本发明属于生物技术领域,涉及分离的禽流感病毒新毒株,本发明公开了一种H9N2禽流感病毒。本发明还公开了一种检测H9N2禽流感病毒的试剂盒,以及一种用于预防的H9N2禽流感病毒的疫苗,以及抗H9N2禽流感病毒的抗体。通过本发明的试剂盒,可以迅速的检测或者排除是否是由本发明的这种基因型的H9N2禽流感病毒所致的禽流感,从而尽快的控制禽流感疫情。同时,接种本发明的疫苗,可以预防本发明的这种基因型的H9N2禽流感病毒的感染。并且本发明还可以为禽流感病毒监测系统提供生物信息。

技术领域:
本发明属于生物技术领域,涉及三种H9N2禽流感病毒的新毒株及用于检测、预防禽流感病毒的试剂盒和疫苗。
背景技术:
禽流感(Avian influenza,AI)是由A型流感病毒引起的一种严重危害禽类健康的传染性疾病。禽类感染禽流感病毒(Avian influenza virus,AIV)后,症状可表现为非显性感染、亚临诊感染,或轻度呼吸道疾病、产蛋量降低直至急性全身致死性疾病等多种形式。根据AIV对易感鸡致病性的差异,可将AIV分为高致病性AIV和低致病性AIV。高致病性AIV(HPAIV)感染禽类所导致的禽流感对养禽业具有毁灭性的打击,故被国际兽疫局规定为A类疾病;而一些低致病性AIV,如H9AIV的感染虽然对感染禽的致死率低,但可引起产蛋量的减少和淘汰率的上升,并造成免疫抑制,引起其他疫病的发生或混合感染,对养禽业造成的经济损失也相当可观。因此,AI一直是威胁世界养禽业的主要疾病之一,世界各国都十分重视对AIV的研究工作。已有研究表明来自于禽类的H9N2禽流感病毒能偶尔从禽类传染给哺乳动物,包括人和猪。Ck/Bei-like和G1-like H9N2禽流感病毒在1990底首次从人和猪中分离到。2003年分离到的人H9N2禽流感病毒是一个新的重组体,很可能来源于当地的活禽市场。这种跨种间传播表明目前的H9N2禽流感病毒对人仍有潜在的感染性。AIV存在着广泛的抗原漂移和抗原转变现象,其抗原变异频率很高。因此测定AIV的全基因序列,对于研究禽流感病毒的检测、分布、流行规律以及控制禽流感的流行、蔓延均具有重要的意义。本研究分离鉴定了2002-2008年上海地区活禽市场的H9N2亚型禽流感病毒,经全基因序列测定分为三种基因型,对这三种基因型毒株的8个基因片段进行关键位点的分析以及基因进化关系分析,表明这三种基因型与目前报道的基因型存在差异,为H9N2禽流感病毒Ck/Bei系中的三种新的基因型,分别命名为Y1,Y2,Y3。
发明内容:
本发明提供了三种H9N2禽流感病毒及用于检测、预防禽流感病毒的试剂盒和疫苗,所述的这三种H9N2禽流感病毒及用于检测、预防禽流感病毒的试剂盒和疫苗要解决现有技术中无法确认和预防新的禽流感病毒从而导致新的禽流感疫情在禽类或者人类传播的技术问题。
本发明提供了一种分离的禽流感病毒(Y1),其基因组由SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4、SEQ ID NO:5、SEQ ID NO:6、SEQ ID NO:7和SEQID NO:8所示的RNA片段组成。
本发明还提供了一种分离的禽流感病毒,其保藏号为CGMCC NO:3117。

b),一个抗原液灭活的步骤,将福尔马林溶液加入病毒液中,检验合格后作为制备抗原液;
c),一个制备油相佐剂的步骤;
d),一个制备疫苗水相的步骤;
e),一个配制疫苗的步骤,将油相与水相预混后,再将混合物注入到高压匀浆泵内乳化,制成油包水乳剂即为疫苗。

本发明借助MEGA3.1分析软件,对这三种基因型H9N2禽流感病毒的8个基因片段分别进行了系统发育树的绘制和分析。这三种基因型毒株属于Ck/Bei系,均为四重重组体,并且这三种基因型与目前报道的Ck/Bei系中的基因型不同,因此为RT-PCR检测这三种新基因型8个基因片段提供了模板序列。通过本发明的试剂盒,可以迅速的检测或者排除是否是新的禽流感由本发明的三种基因型的H9N2禽流感病毒所致,从而尽快的控制禽流感疫情。同时,接种本发明的疫苗,可以预防本发明的三种基因型的H9N2禽流感病毒的感染。使用本发明的抗体,可以治疗本发明的三种基因型的H9N2禽流感病毒的感染。

本发明还可以对禽流感病毒监测系统提供生物信息。对这三种基因型病毒关键位点的分析能提供这三种基因型是否有向人类感染的威胁以及8个片段重排的规律,尤其是有无H5片段的存在等,提供是否有向高致病性禽流感转变的可能性,并为我国H9N2禽流感病毒种子库提供储备,并为病毒的追踪溯源提供序列信息。

具体实施方式:
实施例1病毒的获得
于2002年至2008年从上海地区的活禽批发市场获得商品代三黄鸡,采集活禽样品包括气管或泄殖腔拭子,将采集样品放在含有抗菌素的pH值7.0~7.4的等渗磷酸盐缓冲液(PBS)内。将鉴定为阳性样品的棉拭子充分捻动、拧干后弃去拭子,样品液经1000r/min离心10min,取上清液作为接种材料。取处理好的样品以0.2mL/胚的量经尿囊腔途径接种9日龄~11日龄SPF鸡胚,每个样品接种5个胚,于35℃~37℃孵化箱内孵育,18h后每8h观察鸡胚死亡情况。无菌收取18h以后的死胚及96h仍存活鸡胚的鸡胚尿囊液,测血凝活性,通过下面的实施例对从鸡胚尿囊液中的病毒进行基因测序和鉴定,是三种新的甲型流感病毒H9N2禽流感病毒,对此进行了保藏,保藏号分别为:CGMCC NO:3117(Ck/SH/Y1/06)、CGMCC NO:3118(Ck/SH/Y1/07)、CGMCC NO:3119(Ck/SH/Y1/08)。
实施例2三种基因型H9N2禽流感病毒的全基因测序
2.1于2002年至2008年从上海地区的活禽批发市场获得商品代三黄鸡,共分离、鉴定了三种基因型H9N2禽流感病毒:A/Chicken/Shanghai/Y1/06(简写为Ck/SH/Y1/06,基因型Y1);A/Chicken/Shanghai/Y1/07(简写为Ck/SH/Y1/07,基因型Y2);
A/Chicken/Shanghai/Y1/08(简写为Ck/SH/Y1/08,基因型Y3)。病毒鉴定用的H9亚型血清购自中国农科院哈尔滨兽医研究所。设计用于扩增H9亚型AIV所有8个基因片段的特异引物由宝生物(大连)工程公司合成。

2.2将阳性样本接种SPF鸡胚,取感染鸡胚尿囊液用TRIzol抽提试剂进行RNA的抽提。
2.3 8个基因片段RT-PCR扩增如下:采用20μL的cDNA合成体系置于0.2mL反应管中,取溶于DEPC处理水中的AIV RNA样品9μL,分别加入上游引物3μL,5倍RT-PCR缓冲液4μL,dNTPs(10mmol/L)2.5μL,RNA酶抑制剂0.5μL,AMV反转录酶2μL,用无RNase的超纯水加至总体积为20μL,置于PCR仪中进行cDNA合成,以30℃10min,42℃60min,进入PCR扩增。
PCR反应体系100μL:在PCR反应管中分别加入cDNA10μL,上、下游引物各3μL(20pmol/μL),10×LA PCR Buffer10μL,dNTPs(2.5mmol/L)10μL,MgCl2(25mmol/L)8μL,LA Taq DNA聚合酶0.5μL(5U/μL),灭菌超纯水加至100μL,置于PCR仪中进行PCR扩增。首先95℃预变性2min,然后进行以94℃30s,55℃60s(HA,PB1,M,NP这4个基因的退火温度为55℃,PB2,NS,PA,NA的退火温度为52℃),72℃180s的PCR循环,进行40个循环后于72℃延伸10min。
2.48个基因片段的cDNA克隆、鉴定和测序:RT-PCR产物电泳经凝胶回收后,按pMD18-T载体说明书进行连接、转化,用EcoRI和HindIII进行双酶切鉴定;鉴定为阳性克隆后送英骏生物有限公司测序。
其具体实施步骤如下:
2.4.1RT-PCR产物电泳
取RT-PCR产物加10×loading buffer,在1%琼脂糖凝胶(含0.1ug/ml的溴化乙锭)中,以TAE缓冲液(0.04M Tris—acetate,1mMEDTA)电泳0.5hr,电泳结束后,在紫外线下观察。
2.4.2RT-PCR产物回收
1)将目的片段切下;
2)先冻融,再捣碎;
3)加500ul平衡酚混匀,置-20℃,60min;
4)取出后立即离心,5000g×5min;
5)小心地将水相转移至另一EP管中;
6)在原EP管内加200ulTE(PH8.0),充分混匀。必要时可用玻棒再次捣碎,置-20℃,30min;
7)离心,500g×5min,将水相转移并合并至含有上次水相的EP管中;
8)加等体积酚、氯仿:异戊醇各抽提一次;
9)在转移出的水相中加1/10体积3mol乙酸钠(PH5.2)和2.5倍体积的无水乙醇置-20℃30min;
10)离心,12000g×15min。弃上清,70%乙醇洗涤一次,真空干燥;
11)沉淀用7ul TE缓冲液(PH8.0)溶解,-20℃保存备用;
上述操作也可通过相关公司生产的胶回收试剂盒进行回收。
2.4.3回收的产物与pMD18-T载体的连接
SolutionI:5ul;

PCR Product:4.5ul;
4℃连接12h-24h。
2.4.4感受态细胞的制备
1)划平板,尽量形成单一菌落,使性质均一(一定要熟悉E.coli的颜色、大小、亮度,什么时候最好),37℃过夜,16h。选择分散、单个菌落,一般8h可见菌落,16h差不多;
2)挑取单个菌落,接种到3-5mlLB培养过夜(37℃振摇,200-300rpm);
3)取1ml菌液到LB中,37℃,2-4h,200-300rpm振荡培养至OD550=0.3-0.5,此时正好是E.coli对数生长期早期,生命力最好;
4)放入冰水中,停止增殖,保持该状态10-15min;
5)离心,弃上清,留沉淀于冰浴中(4℃,4000g×10min);
6)0.1MCaCI2(4℃预冷),体积多一些没关系,作用20-30min,时间可以长一些;
7)4℃,4000g×10min离心,去CaCI2;
8)加CaCI2,量只有上一次的1/5到1/10;
9)分装200ul于EP管中。
置4℃冰箱,12-24h即可用于转化。
2.4.5细菌的转化
1)无菌操作取一定量的感受态细胞(200ul)置冰浴;
2)加入需1.4.3制备的连接物10ul,轻轻手腕旋转或吹打以充分混匀;
3)取出置42℃热休克90S,不要摇动,立刻冰浴1-2min;
4)每管加预热的LB800ul,37℃振摇45min-1h,以复苏抗性;
5)稍离心一下,弃去部分上清,剩200ul左右全部涂板,带有Amp抗性的平板;待液体被吸收后,置37℃培养12-18hr;
6)观察见有数个单、小菌落,继续培养,挑选单菌落接种到含Amp的LB液体培养基中
2.4.6重组质粒的提取及酶切鉴定
1)从摇床中取出上述培养物,取1.5ml于EP管中,1,2000rpm,1min;
2)弃上清,加预冷的溶液I 100ul,用枪吹起菌体;
3)加新鲜配制溶液II200ul,通过手腕混匀;
4)加预冷溶液III150ul,倒转混匀,12000rpm,5min;
5)上清移于另一EP管中,加等体积酚/氯仿,混匀,12000rpm,5min;
6)上清移于另一EP管中,并加2倍体积冷无水乙醇-20℃沉淀一段时间,12000rpm,5min;
7)弃上清,加1ml70%乙醇洗涤沉淀,弃去,完全干燥沉淀,并用20ul TE(ph8.0,含20ug/mlRNase)重悬沉淀。用EcoRI和HindIII进行双酶切鉴定。发现阳性克隆的送英骏公司测序(具体见序列表)。
实施例3HA基因氨基酸序列分析
推导的氨基酸序列分析表明,三种基因型毒株均含有7个潜在的糖基化位点(Asn-X-Ser/Thr,X为除Pro以外的任意氨基酸),其中5个位于HA1部分(29aa~31aa、141aa~143aa、218aa~220aa、298aa~300aa、305aa~307aa),2个位于HA2部分(492aa~494aa、551aa~553aa)。裂解位点序列分析发现,所有毒株均为RSSR↓GLF,对禽类为低致病力。构成受体结合位点的氨基酸(109aa、161aa、163aa、191aa、202aa、203aa)除198位氨基酸变化较大外,其它位点在所有毒株中都是相当保守的。此外本研究表明Ck/SH/Y1/06在HA基因226位点为Gln,证明为人类非易感性的H9亚型AIV,而其余2个毒株在该位点为Leu,表明有向人类感染的嗜性(如表1所示)。

实施例4绘制系统发育树
借助MEGA3.1分析软件,对三种基因型毒株的8个基因片段分别进行了系统发育树的绘制和分析(如图1、图2、图3、图4、图5、图6、图7和图8所示)。分析结果表明这三种基因型毒株属于Ck/Bei系,均为四重重组体,这三种基因型不同于以往报道的Ck/Bei系中的H9N2禽流感病毒基因型,本研究中这三种基因型组成情况见表2,目前已报道的Ck/Bei系中的基因型组成情况见表3,本研究中这三种基因型其内源性基因中含有H5N1的片段(表2中的Gs/GD系为H5N1病毒)。

实施例5三种基因型H9N2禽流感病毒8个基因片段RT-PCR诊断试剂盒
试剂盒的组成包括:病毒RNA抽提液、RT反转录反应液和PCR反应液。
其中,病毒RNA的抽提采用TRIzol法,由于TRIzol法抽提病毒RNA为本领域的常规技术,在这里不再赘述。
RT反转录反应液组成如下:随机引物,5倍RT-PCR缓冲液,dNTPs(10mmol/L),RNA酶抑制剂,AMV反转录酶,无RNase的超纯水。
PCR反应液组成如下:上、下游引物(20pmol/μL),10×LA PCR Buffer,dNTPs(2.5mmol/L),MgCl2(25mmol/L),LA Taq DNA聚合酶(5U/μL)。
这三种基因型H9N2禽流感病毒8个基因片段的上、下游引物情况如下表:
5.1RT-PCR诊断试剂盒使用说明
取11μL的RT反转录反应液和9μL的病毒RNA置于0.2mL反应管中,以30℃10min,42℃60min,置于PCR仪中进行cDNA合成;取5μL的上述cDNA产物和45μL的PCR反应液置于0.2mL反应管中,首先95℃预变性2min,然后进行以94℃30s,55℃60s(HA,PB1,M,NP这4个基因的退火温度为55℃,PB2,NS,PA,NA的退火温度为52℃),72℃180s的PCR循环,进行40个循环后于72℃延伸10min。RT-PCR产物经琼脂糖电泳后判定结果,8个片段长度见上表。
实施例6三种基因型H9N2禽流感病毒的灭活疫苗
6.1疫苗抗原液制备及毒价测定
将这三种基因型H9N2禽流感病毒用灭菌生理盐水稀释后,接种9日龄非免疫鸡胚,37℃培养72h(弃去24h内死亡鸡胚),4℃冷藏24h后,收取尿囊液,测定其毒价:在109.6-109.8ELD50,HA价在1:256-512。病毒液置-18℃冰箱中保存备用。
6.2抗原液灭活
将福尔马林溶液加入病毒液中,福尔马林溶液终浓度为0.2%,置37℃温箱内摇动作用1.6小时后,经接种9日龄SPF鸡胚检验合格后作为制备抗原液。
6.3油相佐剂制备
以注射用专用白油(杭州炼油厂)、司本-80(上海大众药业)和硬脂酸铝(上海远航试剂厂),按一定比例(94%轻质白油,6%司本-80,再加2%硬脂酸铝)配制混合,经高温灭菌后备用。
6.4疫苗水相制备
将一定量的吐温-80(上海大众药业)经高温灭菌后,加入到己灭活抗原液中,吐温-80的终浓度为3%,经充分溶解混合即为水相。
6.5疫苗配制
将一定比例的油相与水相(比例为1:1)在立式胶体磨机中预混后,再将混合物注入到高压匀浆泵内乳化,制成油包水乳剂。
6.6无菌及安全检验
随机抽取4瓶疫苗,分别接种鲜血琼脂培养基,37℃培养48小时,无菌生长。另取免疫剂量的3倍量,颈部皮下注射10日龄非免雏鸡6羽,观察2周,无任何不良反应。
6.7理化性状检查
疫苗色泽正常,3000rpm离心15分钟不分层,37℃放置21天不破乳。
6.8免疫效果评估
取SPF4周龄公鸡,在清洁条件下隔离饲养。为预防试验期间被感染并模仿实际生产条件,所以鸡在接种这三种基因型H9N2禽流感油乳剂灭活苗前均用Lasota株NDV弱毒疫苗及鸡传染性法氏囊弱毒疫苗免疫,5d后注射这三种基因型H9N2禽流感油乳灭活苗。在接种这三种基因型H9N2禽流感油乳剂灭活苗后2、3、4、5周后分别采血,测定对H9亚型AIV HI抗体效价。
以0.5ml一次肌肉注射后2、3、4和5周采血,分离血清测定其对H9亚型AIV的滴度。2周后大多数鸡都已表现出不同程度的HI抗体。在3周后,所有免疫鸡都产生抗H9的HI抗体,且较2周时显著上升,这一HI抗体滴度在5周龄时上升至最高值,这三种基因型H9N2禽流感油乳剂灭活苗:Y1平均可达10log2,Y2平均可达8.6log2,Y3平均可达9.6log2。而且个体间差异减少。
据有关资料表明,当HI抗体的平均值达到4log2就有保护力,但有时能分离到病毒。当达到5log2以上时其保护率达100%并且分离不到病毒。以上试验说明这三种基因型H9N2禽流感油乳剂灭活苗免疫效果较好,能抵抗AIV的侵袭,性能稳定。

Claims (6)
1.一种分离的禽流感病毒,其基因组由SEQ ID NO:9、SEQ ID NO:10、SEQ ID NO:11、SEQ ID NO:12、SEQ ID NO:13、SEQ ID NO:14、SEQ ID NO:15和SEQ ID NO:16所示的RNA片段组成。
2.如权利要求1所述的一种分离的禽流感病毒,其保藏号为CGMCC NO:3118。
3.一种用于预防权利要求1或2所述的禽流感病毒的疫苗,其特征在于:含有灭活的权利要求1或2所述的禽流感病毒。
4.权利要求3所述的用于预防禽流感病毒的疫苗的制备方法,其特征在于包括如下步骤:a),一个疫苗抗原液制备及毒价测定的步骤,将权利要求1或2所述的禽流感病毒用灭菌生理盐水稀释后,接种非免疫鸡胚,收取尿囊液,测定其毒价:在109.6-109.8ELD50,HA价在1:256-512之间,病毒液置冰箱中保存备用;
b),一个抗原液灭活的步骤,将福尔马林溶液加入病毒液中,检验合格后作为制备抗原液;
c),一个制备油相佐剂的步骤;
d), 一个制备疫苗水相的步骤;
e), 一个配制疫苗的步骤,将油相与水相预混后, 再将混合物注入到高压匀浆泵内乳化,制成油包水乳剂即为疫苗。
5.如权利要求4所述的用于预防禽流感病毒的疫苗的制备方法,其特征在于:所述的油相佐剂由白油 、司本-80 和硬脂酸铝配制混合而成。
6.如权利要求4所述的用于预防禽流感病毒的疫苗的制备方法,其特征在于:在制备疫苗水相的步骤中,将吐温-80经高温灭菌后,加入到己灭活抗原液中,吐温-80的终浓度在2%-5%之间,经充分溶解混合即为水相。
