降钙素基因相关的肽(在下文称为CGRP)是在一些物种中以两种形式(称为CGRP-α和CGRP-β(或分别为 CGRP-I和 CGRP-II))存在的肽。CGRP肽在物种内是高度保守的肽。例如,在Peptides

从文献中可明显得知,CGRP引发各种药理作用,例如∶1)血管舒张,2)肌肉和肝AMP激酶(AMPK)活化和脂解和/或脂肪氧化,3)降低食物摄入,4)抑制胃排空和改变肠管功能,和5)提高糖酵解和抑制糖原合成。对所提及的作用的纯生理意义还不完全了解,并且没有出现长期接触CGRP的结果(例如,空腹胰岛素、HbA1C和持续血管舒张)。然而,在新陈代谢病中,AMPK活化、脂肪氧化和降低食物摄入是有利的,已经建议糖酵解和抑制糖原合成调解抗胰岛素性。
天然的CGRP具有小于30分钟的半衰期,并且CGRP输注之后的药理作用持续期短是明显的。由于给予CGRP的血管舒张作用,天然CGRP的体内药理学研究受到血管舒张和补偿性血管收缩继发性的效果的影响。由此,血管舒张作用是CGRP的剂量限制性作用,其妨碍了对与代谢性综合症有关的作用的完全理解。由此,CGRP的药理学应用需要产生具有长期作用的CGRP类似物,并且甚至只可以用长效类似物来获得一些效果。

药物制剂
在一方面,按照本发明,给予受益于这种治疗的患者(例如,病人或动物)治疗有效量的本发明的化合物。该治疗可以例如是抗胰岛素性、II型糖尿病、高血压症、肥胖症、血脂异常、动脉粥样硬化和血栓。给予本发明化合物的剂量范围是大到足以产生预期效果的剂量范围。

树脂结合的肽的合成
SPPS方法A
SPPS方法A是指保护的肽基树脂的合成,该方法在Applied Biosystems 433肽合成仪(还称为ABI433A合成仪)上使用Fmoc化学过程,规模为0.25 mmol或1.0 mmol,使用生产商的FastMoc UV方案,该方案在NMP中使用HBTU或HATU介导的结合,UV检测Fmoc保护基的去保护状况。

SPPS方法B是指在基于微波的Liberty肽合成仪(CEM Corp.,
North Carolina)上、通过Fmoc化学过程的肽合成。合适的树脂是预载的、低填充量的王氏树脂,得自于Novabiochem(例如,低填充量的fmoc-Lys(Mtt)-王氏树脂,0.35
mmol/g)。在NMP中,在至高达70或75℃下,用5%哌啶进行Fmoc-脱保护。偶合化学过程是DIC/HOAt(在NMP中)。将氨基酸/HOAt溶液(0.3M,在NMP中,摩尔过量3-10倍)加入到该树脂中,而后加入相同摩尔当量的DIC(0.75M,在NMP中)。例如,对于下列规模的反应,每个偶合使用如下数量的0.3M氨基酸/HOAt溶液∶规模/mL,0.10 mmol/2.5 mL,0.25 mmol/5 mL,1

纤维素结合的肽的合成
在96孔板中,使用配备有AutoSpot模块的完全自动化MultiPep自动装置(rorobot)(得自于Intavis

Fmoc氨基酸/0.3M HOAt/NMP,每个SPOT 30 min)以三联形式(in triple)进行偶合。通过用20%哌啶(piperdine)/NMP(2

用降钙素受体类受体(CRLR)、受体活性调节蛋白-1(RAMP1)和CRE荧光素酶(cAMP响应成份,用于荧光素酶基因表达对照)稳定地转染中国仓鼠卵巢(CHO)细胞。荧光素酶是报告胞内cAMP积聚的指示基因。
将细胞在F12培养基中培养,该培养基补充有GlutaMAXTM(Gibco
cat no 31765-027)、FBS(10%,Gibco 16140-071)、青霉素-链霉素(1%,Lonza
DE-17-602E)、G418(400µg/ml,Gibco 10131-027)、灭瘟素(Blasticidine)(10µg/ml,Corning cat no 15205)、潮霉素(400µg/ml, Calbiochem cat no400052)。
在实验之前的当天,在100µl培养基中,将细胞播种在96孔板(Packard #6005180)中,20000个细胞/孔。实验的当天,用试验培养基(Dulbecco's培养基w/o酚红(Gibco cat no

在200μl培养基中,将细胞播种在96孔板(Biological
Industries,cat no E-TCMT-F)中,50,000个细胞/孔,并培养过夜。在37℃,在包含DMEM(Gibco

诱导的食物摄入的减少
一旦雄性Spraque Dawley(SD)大鼠到达,每个笼中放两个大鼠,在它们之间具有分隔墙。将它们在相反的昼夜节律下放置,并使环境丰富。使大鼠适应相反的昼夜节律至少1周,而后转入实验系统笼(单独放置)中。使它们额外适应实验系统笼5-6天,而后开始实验。在驯化期间,使大鼠不限量地接触食物(Altromin
1324)和水。
开始研究之前的当天,基于体重(250-300 g)将大鼠分成三或四个组(n=5-8)。
开始研究之前16小时,使大鼠禁食。与给药结合恢复供给食物。使大鼠接受单一皮下注射(s.c.,1
ml/kg)赋形剂或CGRP类似物。在第一个实验中,腹膜内给予(i.p.,1 ml/kg)α1类似物。
在“FeedWIN”系统(Ellegaard
Systems A/S,Faaborg,Denmark)中检验类似物对食物和水消耗的效果。该系统包括32个工作站。在每个工作站,记录单个大鼠的持续的水和食物消耗。
一个工作站由带有金属盖的笼和放置在笼的每个面上的两个天平组成。天平记录从食物和水容器中减少的重量,由此记录个体分别消耗的食物和水的重量。将32个工作站与两个PC连接,PC通过特别设计的软件(FeedWIN)收集和处理数据。每15分钟收集数据,直至48小时,并表示为累积食物摄入(平均值±SEM)。

重量大约250 g的雄性Spraque
Dawley大鼠(n=24)用于合并的药物动力学(T1/2,Cmax等等)和药效(GLP-1释放)研究。将大鼠分成4个组,每个组n=6个大鼠。用单一皮下注射(1 ml/kg)赋形剂、α-CGRP(138
nmol/kg)、类似物β1(231 nmol/kg)或类似物α1(151 nmol/kg)来处理大鼠。在给药前和给药后0.5、1、3、7和24小时,从未麻醉大鼠的舌下静脉取出血液(200µl)样品。将血液收集在含有EDTA的管中。将样品在冰上保持,并离心。将血浆在-20℃下保持,直到分析CGRP类似物和总的GLP-1为止。
商业获得的试剂盒(人CGRP ELISA,cat.#A05481,SPIbio)用于监测CGRP-类似物的血浆水平。简要地说,该试验是使用两个抗人CGRP的抗体的两位点酶联免疫吸收试验(ELISA)。将单克隆抗体固定到微板孔的表面。将五微升血浆样品或校准物加到合适的孔中,并与100µL缓冲剂和100µL AChE-共轭的Fab'片段一起培养2小时。然后将孔排空,洗涤,并将Ellman's试剂加入到每个孔中,在黑暗中额外培养30-90分钟。利用分光光度计(在405 nm)测定颜色强度。信号水平与血浆样品中CGRP-类似物的浓度成正比。将CGRP-类似物在血浆(Bioreclamation,UK)中稀释,并用作校准物。在0至30000 pM范围内制作校准物的线。将校准物在-18℃下保存在Micronic管中。改进的CGRP试验的定量下限是大约750 pM,不精确性(%CV)<10%。

十八只Spraque Dawley大鼠用于该研究。当开始研究时,大鼠大约1岁,并且已经喂食了10个月的多脂饮食(Research Diet,D12451 45%)。在研究开始之前14天,使它们单独居住,并且不限量地接触食物和水。每天用类似物α1(100 nmol/kg,s.c.)或赋形剂处理它们一次,处理14天。
