Abstract

本发明公开了一种含复方氨基酸的脂质体制剂的配方及其制备方法,确定了脂质体制剂的原料质量配比,制备方法包括:(1)称取大豆磷脂、胆固醇,加水,加热搅拌,制成油相;(2)称取盐酸半胱氨酸、色氨酸,加工艺用水溶解,再依次加入缬氨酸、异亮氨酸、亮氨酸、醋酸赖氨酸、蛋氨酸、苯丙氨酸、苏氨酸、精氨酸、甘氨酸、脯氨酸中的几种或全部;再依次加入辅料维生素A、维生素C、维生素E、维生素K的一种或几种和成膜材料,最后加入山梨酸钾或尼泊金乙酯,搅拌,制成水相;(3)将油相与水相混合,用高速剪切机剪切成脂质体制剂。本发明开创了不采用任何有机溶剂制备脂质体的技术,所得脂质体制剂包封率高、稳定性好,能实现产业化生产。

Description

含复方氨基酸的脂质体制剂配方及其制备方法
技术领域
本发明涉及以特殊物理形状为特征的医药配制品,具体来说,涉及含复方氨基酸成分的脂质体制剂。
背景技术
脂质体技术的发展有近50年的历史,目前的脂质体制备方法主要有四大类:1)去污剂透析法,但该法很难工业化生产,且不适合包封水溶性药物;2)以薄膜分散法为基础,即将用于形成脂质体的磷脂溶于溶于有机溶媒中——通常是氯仿、氯仿和甲醇的混合物、无水乙醚、叔丁醇等,之后通过减压的条件下除去溶媒形成干燥的磷脂膜。将磷脂膜水化即可形成多层脂质体(multilamellar vesicles)。此法虽是最经典的方法,但是却存在许多缺点,例如:使用毒性很大的有机溶媒;无法应用于工业化生产;当用含药水溶液水化时,形成的多层脂质体(multilamellar vesicles)层与层之间药物浓度不均一,必须通过反复冻融处理等等;3)以常规w/o乳或w/o/w复乳为基础的。此种制备方法虽然可以大规模生产,但存在一些问题,如除去成乳时的毒性溶媒需要特殊的技术;由于有机溶媒的存在,包裹蛋白质类或核酸类药物常常会导致药物丧失活性。4)以注入法为基础,如乙醇注入法。虽可大规模生产,但是如果采用被动载药法制备脂质体,对于水溶性药物通常包封率比较低。
在中国专利数据库中,涉及医用脂质体的专利和专利申请件有ZL96122373.1号《医用脂质体软膏》、02125280.7号《皮肤用辣椒碱脂质体制剂》、ZL200810107876.0号《一种添加珍珠水解液脂质体的祛痘精华液》、200910048164.0号《脂质体硫酸钙复合纳米人工骨、制备方法和应用》、200910205088.X号《一种添加珍珠水解液脂质体的祛痘啫喱》等。然而迄今为止,还没有不采用任何有机溶剂、脂质体包封率高、制剂的稳定性好、能实现产业化大生产的脂质体制剂的制备技术问世。
发明内容
本发明的目的是提供一种含复方氨基酸的脂质体制剂的配方,为制备该制剂奠定基础。
本发明的又一目的是提供上述制剂的制备方法,克服现有制备技术的不足,该技术不采用任何有机溶剂,所制得的制剂脂质体包封率高、制剂的稳定性好、能实现产业化大生产。
发明人提供的含复方氨基酸的脂质体制剂的配方为:
大豆磷脂10~150份,胆固醇6~80份,盐酸半胱氨酸0.3~1.2份,色氨酸0.03~0.6份,缬氨酸 0.4~1.3份,异亮氨酸0.5~2.0份,亮氨酸0.8~2.5份,醋酸赖氨酸0.2~1.2份,蛋氨酸0.06~0.5份,苯丙氨酸0.09~0.7份,苏氨酸0.1~0.6份,精氨酸0.1~1.6份,甘氨酸0.2~2份,脯氨酸0.05~0.3份,维生素A 0.03~2份,维生素C 0.3~2份,维生素E 0.4~3份,维生素K 0.01~0.1份,成膜材料65~200份,山梨酸钾或尼泊金乙酯 0.5~4份,余为工艺用水。
上述各种原料均为市售产品,所述成膜材料是海藻酸钠(褐澡酸钠)或海藻酸(褐澡酸)的衍生物或海藻多糖复合胶。
上述工艺用水是注射用水或者纯化水。
发明人提供的含复方氨基酸的脂质体制剂的制备方法,包括:
(1) 按配方比例称取大豆磷脂、胆固醇置洁净容器中,加配方所需的工艺用水,加热至40℃~90℃,搅拌25~300min,搅拌浆转速控制在1500~6000r/min,制得油相; 
(2) 按配方比例称取盐酸半胱氨酸、色氨酸置洁净容器中,加配方所需的工艺用水溶解后,再依次加入缬氨酸、异亮氨酸、亮氨酸、醋酸赖氨酸、蛋氨酸、苯丙氨酸、苏氨酸、精氨酸、甘氨酸、脯氨酸中的几种或全部;再依次加入辅料维生素A、维生素C、维生素E、维生素K的一种或几种和成膜材料,最后加入山梨酸钾或尼泊金乙酯,搅拌使其溶解混匀,制得水相;
(3) 将油相与水相混合,利用高速剪切机剪切,制备成粒径小于200nm的脂质体制剂。
发明人为考察产品的稳定性,将产品分别置于-28℃、-5℃、20℃、55℃,相对湿度均不大于75%的环境条件下放置14天。进行相应指标的检测,结果为:
产品生物学评价:细胞毒性小于或等于1级,无皮肤及黏膜刺激性、无致敏性;
产品在偏光显微镜下观察,脂质体形态完好,粒径小于200nm;
其余理化指标及无菌指标均未发生变化。
本发明提供了一种含复方氨基酸的脂质体制剂的配方,在此基础上提供了一种不采用任何有机溶剂制备脂质体的技术,该脂质体对氨基酸的包封率大于90%,而且该制剂的各种组分及适宜配比绿色环保,对人体无任何毒害、无刺激性成分,患者使用的舒适感比市面上任何一种外用伤口及创面制剂强。各种促进伤口及创面愈合的有效成分被脂质体包封后,能够通过吸附融合的方式将细胞营养物质转运至创面细胞内,参与细胞的新陈代谢,快速促进创面愈合;本发明还采用了成膜技术,产品涂于伤口及创面后会在几分钟内成膜,这种膜能够保持伤口湿润、阻隔外界尘埃及病原体对伤口的二次感染,同时能够自由通透氧离子,为伤口的愈合创造良好的微环境。
具体实施方式
现通过下列实施例详细说明本发明的具体实施方式。
实施例l  
先称取大豆磷脂25g、胆固醇7g置洁净容器中,加工艺用水161ml,加热至60℃,搅拌100min,搅拌浆转速控制在3500r/min,制得油相; 
接着称取盐酸半胱氨酸0.3g、色氨酸0.15g置洁净容器中,加工艺用水646ml溶解后,再依次加入缬氨酸1.25g、异亮氨酸0.83g、亮氨酸1.62g、醋酸赖氨酸0.79g、蛋氨酸0.48g、苯丙氨酸0.53g、苏氨酸0.15g、精氨酸0.15g、甘氨酸0.21g、脯氨酸0.09g;再依次加入辅料维生素A0.3g、维生素C0.82g、维生素E0.50g、维生素K0.03g,海藻酸钠(褐澡酸钠)151.9g,最后加入山梨酸钾0.9g,搅拌使其溶解混匀,制得水相; 
之后将油相与水相混合,利用高速剪切机剪切,制成粒径小于200nm的脂质体制剂。
实施例2.
先称取大豆磷脂140g、胆固醇80g置洁净容器中,加工艺用水350ml,加热至72℃,搅拌120min,搅拌浆转速控制在5100r/min,制得油相; 
接着称取盐酸半胱氨酸0.30g、色氨酸0.31g置洁净容器中,加工艺用水340ml溶解后,再依次加入缬氨酸0.41g、异亮氨酸0.84g、亮氨酸1.93g、醋酸赖氨酸0.99g、蛋氨酸0.15g、苯丙氨酸0.16g、苏氨酸0.36g、精氨酸0.47g、甘氨酸0.58g、脯氨酸0.09g;再依次加入辅料维生素A0.21g、维生素C0.45g、维生素E0.31g、维生素K0.04g,海藻酸钠(褐澡酸钠)81.9g,最后加入山梨酸钾0.5g,搅拌使其溶解混匀,制得水相; 
之后将油相与水相混合,利用高速剪切机剪切,制成粒径小于200nm的脂质体制剂。
实施例3
先称取大豆磷脂25g、胆固醇9g置洁净容器中,加工艺用水250ml,加热至70℃,搅拌60min,搅拌浆转速控制在3500r/min,制得油相; 
再称取盐酸半胱氨酸0.61g、色氨酸0.39g置洁净容器中,加工艺用水573 ml溶解后,再依次加入缬氨酸1.01g、异亮氨酸0.66g、亮氨酸1.95g、醋酸赖氨酸0.36g、蛋氨酸0.27g、苯丙氨酸0.38g、苏氨酸0.44g;再依次加入辅料维生素C0.41g,海藻酸钠(褐澡酸钠)135.2g,最后加入山梨酸钾1.32g,搅拌使其溶解混匀,制得水相; 
之后将油相与水相混合,利用高速剪切机剪切,制成粒径小于200nm的脂质体制剂。
实施例4
先称取大豆磷脂47g、胆固醇36g置洁净容器中,加工艺用水320ml,加热至82℃,搅拌200min,搅拌浆转速控制在1800r/min,制得油相; 
接着称取盐酸半胱氨酸0.39g、色氨酸0.24g置洁净容器中,加工艺用水428ml溶解后,再依次加入缬氨酸0.62g、异亮氨酸0.89g、亮氨酸2.11g、醋酸赖氨酸0.59g、蛋氨酸0.43g、精氨酸1.14g;再依次加入辅料维生素C0.97g、维生素K0.18g,海藻多糖复合胶160g,最后加入尼泊金乙酯1.44g,搅拌使其溶解混匀,制得水相; 
之后将油相与水相混合,利用高速剪切机剪切,制成粒径小于200nm的脂质体制剂。
实施例5
先称取大豆磷脂11g、胆固醇7g置洁净容器中,加工艺用水120ml,加热至75℃,搅拌60min,搅拌浆转速控制在4000r/min,制得油相; 
接着称取盐酸半胱氨酸0.48g、色氨酸0.09g置洁净容器中,加工艺用水670ml溶解后,再依次加入亮氨酸1.96g、醋酸赖氨酸1.29g、蛋氨酸0.37g、苯丙氨酸0.29g、苏氨酸0.54g、精氨酸0.55g、脯氨酸0.29g;再依次加入辅料维生素A1.2g、维生素C1.92g、维生素E1.22g,海藻酸(褐澡酸)的衍生物180g,最后加入山梨酸钾1.8g,搅拌使其溶解混匀,制得水相; 
之后将油相与水相混合,利用高速剪切机剪切一定时间,制成粒径小于200nm的脂质体制剂。
上述实施例仅列举了部分含复方氨基酸的脂质体制剂的制备方法实例,凡与本发明方法相似,无须付出创造性劳动的技术方案均在本申请件的保护范围之内。

Claims (4)

1. 一种含复方氨基酸的脂质体制剂的配方,其特征在于该配方为:
大豆磷脂10~150份,胆固醇6~80份,盐酸半胱氨酸0.3~1.2份,色氨酸0.03~0.6份,缬氨酸 0.4~1.3份,异亮氨酸0.5~2.0份,亮氨酸0.8~2.5份,醋酸赖氨酸0.2~1.2份,蛋氨酸0.06~0.5份,苯丙氨酸0.09~0.7份,苏氨酸0.1~0.6份,精氨酸0.1~1.6份,甘氨酸0.2~2份,脯氨酸0.05~0.3份,维生素A 0.03~2份,维生素C 0.3~2份,维生素E 0.4~3份,维生素K 0.01~0.1份,成膜材料 65~200份,山梨酸钾或尼泊金乙酯 0.5~4份,余为工艺用水。
2.  如权利要求1所述的配方,其特征在于所述各种原料均为市售产品;所述成膜材料是海藻酸钠(褐澡酸钠)或海藻酸(褐澡酸)的衍生物或海藻多糖复合胶。
3. 如权利要求1所述的配方,其特征在于所述工艺用水是注射用水或者纯化水。
4. 制备权利要求1所述含复方氨基酸的脂质体制剂的方法,其特征包括以下步骤:
(1) 按配方称取大豆磷脂、胆固醇置洁净容器中,加工艺用水,加热至40℃~90℃,搅拌25~300min,搅拌浆转速控制在1500~6000r/min,制得油相;
(2) 按配方称取盐酸半胱氨酸、色氨酸置洁净容器中,加工艺用水溶解后,再依次加入缬氨酸、异亮氨酸、亮氨酸、醋酸赖氨酸、蛋氨酸、苯丙氨酸、苏氨酸、精氨酸、甘氨酸、脯氨酸中的几种或全部;再依次加入辅料维生素A、维生素C、维生素E、维生素K的一种或几种和成膜材料,最后加入山梨酸钾或尼泊金乙酯,搅拌使其溶解混匀,制得水相;
(3)  将油相与水相混合,利用高速剪切机剪切,制成粒径小于200nm的脂质体制剂。
