Abstract

公开了一种蜂王浆新鲜度测定的新方法。该方法主要通过分别采用三维荧光光谱法和ELISA(酶联免疫吸附)方法测定蜂王浆中的糖基化末端产物Pentodilysin和CML(羧甲基赖氨酸),来监测蜂王浆的新鲜度。迄今为止,国际上没有一个公认的可如实反映蜂王浆新鲜度并能在实际应用中推广的指标。

一种蜂王浆新鲜度测定的方法
技术领域
本发明涉及到蜂王浆的新鲜度的检测。
背景技术
新鲜蜂王浆中含有丰富的生物活性物质,对人体具有很高的营养保健作用,在临床应用上取得显著疗效,如调节免疫、抗疲劳、抗衰老、抗肿瘤、改善睡眠等。对消化系统、循环系统、心脑血管系统等均有良好的辅助治疗作用,被称为“人类健康之友”。
蜂王浆的成分极其复杂,含有多种活性物质,这些活性成分对温度及其敏感,若贮存方法不当,其物理性状、化学成分、生理功能等品质会发生显著变化。因此蜂王浆的质量,关键在于新鲜度。
蜂王浆的品质与新鲜度之间存在密切的关系。目前主要采取测定蜂王浆中的蛋白质,如57KDa;蜂王浆中的酶,超氧化物歧化酶等来测定蜂王浆的新鲜度指标。但这些测定方法均存在地域差异性,检测仪器昂贵等缺陷。迄今为止,国际上没有一个公认的可如实反映蜂王浆新鲜度并能在实际应用中推广的指标。
本发明的测定方法以荧光光谱为基础,测定蜂王浆不同储藏条件下的Pentodilysine和CML的含量。
发明内容
本发明中测定的Pentodilysine和CML,是蜂王浆糖基化末端产物。蜂王浆在储存过程中,蛋白质会与糖发生非酶促褐变,即美拉德反应,且美拉德反应的程度随着储存时间的延长而加剧。其美拉德反应产物,Pentodilysine和CML的含量变化明显,可通过三维荧光光谱扫描及ELISA法检测其含量变化。
本发明中样品制备简单,Pentodilysine和CML的检测方法可靠,可作为测定蜂王浆新鲜度的方法。
具体实施方式
一种蜂王浆新鲜度测定的方法,该方法分别通过三维荧光光谱方法和ELISA(酶联免疫吸附)法,测定蜂王浆中的Pentodilysine和CML(羧甲基赖氨酸)的含量变化。
所述三维荧光光谱方法是指波长扫描的方法。
所述ELISA法采用的是ELISA试剂盒。
所述Pentodilysine和CML(羧甲基赖氨酸)是蜂王浆储存过程中美拉德反应的产物□糖基化末端产物。
所述Pentodilysine的提取方法是蜂王浆∶水∶三氯乙酸的比例为1∶5∶5,12000rpm/min,10min,取上清。
所述CML的提取方法是,蜂王浆以1∶5的比例溶于水。
所述波长扫描的参数为,λex:320nm,λem:380□500nm,扫描速度:240nm/min,延迟时间:0.0s。
所述新鲜蜂王浆的Pentodilysine谱图为:当λex=320nm时,新鲜王浆中Pentodilysinee在λem=445.8nm处有一个最大荧光峰,在λem=422nm处有一个肩峰。
新鲜蜂王浆的CML的含量为9.04ng±0.04ng/mL。

1.一种蜂王浆新鲜度测定的方法,该方法分别通过三维荧光光谱方法和酶联免疫吸附法,测定蜂王浆中的Pentodilysine和羧甲基赖氨酸的含量变化;
所述Pentodilysine的提取方法是蜂王浆∶水∶三氯乙酸的比例为1∶5∶5,12000rpm/min,10min,取上清;
所述羧甲基赖氨酸的提取方法是,蜂王浆以1∶5的比例溶于水。
2.根据权利要求1所述的方法,其特征在于,所述三维荧光光谱方法是指波长扫描的方法。
3.根据权利要求1所述的方法,其特征在于,所述酶联免疫吸附法采用的是酶联免疫吸附试剂盒。
4.根据权利要求1所述的方法,其特征在于,所述Pentodilysine和羧甲基赖氨酸是蜂王浆储存过程中美拉德反应的产物-糖基化末端产物。
5.根据权利要求2所述的方法,其特征在于,所述波长扫描的参数为,λex:320nm,λem:380-500nm,扫描速度:240nm/min,延迟时间:0.0s。
6.根据权利要求1所述的方法,其特征在于,新鲜蜂王浆的Pentodilysine谱图为:当λex=320nm时,新鲜王浆中Pentodilysine在λem=445.8nm处有一个最大荧光峰,在λem=422nm处有一个肩峰。
7.根据权利要求1所述的方法,其特征在于,新鲜蜂王浆的羧甲基赖氨酸的含量为9.04ng±0.04ng/mL。
