本发明公开了一种盐酸聚六亚甲基胍在防治柑桔酸腐病上的应用及其保鲜剂。该柑桔保鲜剂,按总质量分数100%计,由0.05%~0.5%盐酸聚六亚甲基胍,0.05%~0.2%咪唑类杀菌剂、苯并咪唑类杀菌剂、嘧霉胺、氟咯菌腈、嘧菌酯中的一种或数种的混合物,0.02%~0.15%聚氧乙烯醚类或季胺盐类乳化剂,以及余量的水组成。本发明的柑桔保鲜剂能明显抑制沙糖桔等柑桔类果实在贮藏期间腐烂,特别是酸腐病的发生。如沙糖桔果实用本发明的柑桔保鲜剂处理后在常温(15~25)贮藏45天后,酸腐病发病率低于10%,而对照高达60%,因此本发明的柑桔保鲜剂显著的降低宽皮桔类在贮藏过程中酸腐病的发生,大大提高了柑桔的经济价值。

盐酸聚六亚甲基胍在防治柑桔酸腐病上的应用及其保鲜剂
技术领域
本发明属于水果保鲜领域,具体来说涉及盐酸聚六亚甲基胍在防治柑桔酸腐病上的应用及其保鲜剂。
背景技术
柑桔是世界第一大水果,有138个国家栽培,面积约667万公顷,年总产量1亿余吨,约占世界水果总产量的22%。据统计,2008年我国柑橘栽培面积达3045万亩,总产量2331万吨,占世界柑桔产量约23%,为世界柑桔第一生产大国。广东省是我国柑桔主产区之一,沙糖桔为我省柑桔品种中最具特色的品种,在国内外水果市场上极具竞争力。然而由于沙糖桔独特的生物学特性,与柑桔类其它水果相比难保鲜,采后损耗相当严重,每年的损失都在数亿元以上,严重制约沙糖桔产业的发展。
柑桔属非呼吸跃变型果实,但采后呼吸代谢旺盛导致营养与风味品质迅速下降。沙糖桔皮薄汁多,采后病害发生严重,更不耐贮藏。沙糖桔贮藏期间发生的主要病害有酸腐病、青绿霉病、黑腐病和蒂腐病。目前,控制沙糖桔果实病害的最有效的途径是低温贮藏结合化学杀菌剂处理,所用的杀菌剂有多菌灵、甲基托布津、抑霉唑、双胍盐(百可得)等,但对腐烂特别是酸腐病的控制方面仍不够理想。
酸腐病是目前沙糖桔贮运中最常见、最难防治的病害之一,其致病菌为半知菌亚门丝孢纲内的白地霉(Geotrichum candidum),病菌从伤口侵入。酸腐病只危害果实,一般发生于成熟或过熟果,特别是贮藏期的果实。酸腐病的发病率受采前栽培措施、成熟度、采收方法、果实水分状况、贮运措施等多种因素影响,如广东省沙糖桔主产区2007年酸腐病发病率在10%以下,而2010年初因天气原因发病率高达50%以上。对于柑桔酸腐病的防治,咪唑类和苯并咪唑类防腐剂均无效。双胍盐(百可得)对防治酸腐病效果较好,但存在农药残留和安全性问题,在美国双胍盐(百可得)因致癌作用已被禁止使用。因此,寻求安全高效而又操作简单的防酸腐病技术势在必行。
PHMG(盐酸聚六亚甲基胍,Polyhexamethylene guanidine)是国际上新合成的一种绿色环保型广谱抗菌剂,PHMG属胍类衍生物,是一种阳离子聚合物。它通过作用于病原菌的细胞壁和细胞质的膜系统,与细胞膜磷脂中的脂肪酸结合,迅速击穿它们的细胞质膜,攻破病菌细胞的核心,使病菌体内的生命物质外流,从而达到最佳的杀菌效果。PHMG是高分子聚合物,不易被动物组织吸收,因而大大降低了它的毒性。据俄罗斯检疫部门的结果,PHMG对小鼠类的半致死量LD50为25000mg/kg,为实际无毒级。由于PHMG具有无毒、高效、稳定等特点,而被广泛用于医院消毒、饮用水和食品加工设备的消毒。PHMG对细菌和病毒有较强的杀灭及抑制作用,对真菌抑制作用的研究和应用较少。目前,未有PHMG应用于采后果蔬真菌病害研究和应用的报道。
发明内容
本发明的第一个目的是提供盐酸聚六亚甲基胍在防治柑桔酸腐病上的应用。
本发明通过对柑桔采后酸腐病致病菌白地霉Geotrichum candidum离体和活体研究,筛选到了一种能明显抑制白地霉Geotrichum candidum生长,有效降低沙糖桔果实酸腐病发生抗菌物质-盐酸聚六亚甲基胍,结合防治青绿霉病杀菌剂,得到的保鲜剂能有效抑制宽皮桔贮藏和流通过程中腐烂,特别是酸腐病的发生。
因此本发明的第二个目的是提供一种专门针对柑桔采后腐烂,特别是控制酸腐病发生的柑桔保鲜剂。
本发明的柑桔保鲜剂,按总质量分数100%计,由0.05%~0.5% 盐酸聚六亚甲基胍,0.05%~0.2% 咪唑类杀菌剂、苯并咪唑类杀菌剂、嘧霉胺、氟咯菌腈、嘧菌酯中的一种或数种的混合物,0.02%~0.15% 聚氧乙烯醚类或季胺盐类乳化剂,以及余量的水组成。
所述的咪唑类杀菌剂优选为咪鲜胺和抑霉唑。
所述的苯并咪唑杀菌剂优选为多菌灵、甲基托布津和特克多。
所述的盐酸聚六亚甲基胍优选质量分数为0.1%~0.5%,进一步优选为0.25%~0.5%。
本发明的柑桔保鲜剂的制备方法如下,先将乳化剂和盐酸聚六亚甲基胍混合,再加入咪唑类杀菌剂、苯并咪唑类杀菌剂、嘧霉胺、氟咯菌腈、嘧菌酯中的一种或数种的混合物作为防腐剂和少量水,搅拌均匀得到产物,使用时再加入水稀释到上述浓度而得到本发明的柑桔保鲜剂。
本发明的柑桔保鲜剂能明显抑制沙糖桔等柑桔类果实在贮藏期间腐烂,特别是酸腐病的发生。如沙糖桔果实用本发明的柑桔保鲜剂处理后在常温(15~25)贮藏45天后,酸腐病发病率低于10%,而对照高达60%,因此本发明的柑桔保鲜剂显著的降低宽皮桔类在贮藏过程中酸腐病的发生,大大提高了柑桔的经济价值。

具体实施方式:
以下是对本发明的进一步说明,而不是对本发明的限制。
下述实施例中多菌灵即2-苯并咪唑基氨基甲酸甲酯,甲基托布津即1,2-二(3-甲氧碳基-2-硫脲基)苯,特克多即2-(噻唑-4-基)苯并咪唑,咪鲜胺即N-丙基-N-[2-(2,4,6-三氯苯氧基)乙基]-咪唑-1-甲酰胺,抑霉唑即1-「2-(2,4-二氯苯基)-2-(2-烯丙氧基)」-1H-咪唑。
一、离体抑菌实验
1 材料与方法
(1)菌株:从发生酸腐病沙糖桔中分离纯化的柑桔白地霉Geotrichum candidum.
(2)孢子萌发试验:采用孢子液法,将计算好的一定量PHMG药剂加入已灭菌的1/2 PBD培养基中,配制成9个浓度梯度:0,0.1,1,2,5,10,20,50,100 mg/L。分别取5ml含不同浓度PHMB的培养基至10 ml已灭菌的试管中,加入0.1ml 已配制好的标准孢子悬浮液,于26℃恒温条件下250rpm/min振荡培养,培养8小时后取出1-2菌液,显微镜下观察,测定孢子萌发数,统计孢子萌发率。
(3)菌丝生长试验:采用滤纸抑菌圈法,把已配好并经过高压蒸汽灭菌的PDA培养基冷却45-50 °C,加入Geotrichum candidum孢子悬浮液至培养基中,孢子浓度为106个/L,将培养基倒入已灭菌的培养皿中制成平板。取直径5 mm的灭菌滤纸片放入平板中央,在滤纸上加入10μl  1g/L的PHMG溶液(用无菌水代替PHMG作为对照),然后在26℃恒温培养箱内培养5天,测量滤纸抑菌圈的直径并进行分析。
(4)菌丝体扫描电镜观查:将已配好含有10mg/L PHMG并经高压蒸汽灭菌的PDA培养基冷却至45-50 °C,倒入已灭菌的培养皿中制成平板,接种上Geotrichum candidum孢子悬浮液,在26℃恒温培养箱内培养5天,用直径为0.4 cm 的打孔器在上述菌落边缘切下菌丝体按常规方法进行扫描电镜观查。
试验结果
如图1所示,PHMG显著抑制了Geotrichum candidum离体孢子萌发,并且表现出浓度效应,当PHMG的浓度为5 mg/L时,Geotrichum candidum孢子萌发率仅为2.13%,而10mg/L PHMG则几乎完全抑制了沙糖桔Geotrichum candidum孢子萌发。滤纸抑菌圈法试验表明,含10μl  1g/L PHMG的滤纸片对Geotrichum candidum菌丝生长显著的抑制作用,在26℃培养培养5天后,抑菌圈直径为2.19 cm (图2)。从图3可看出,在不含PHMG的PDA培养基,Geotrichum candidum菌丝生长正常,菌丝呈棒状、均匀、表面光滑,而在含PHMG培养基上生长的菌丝,菌丝体严重扭曲,表面凹陷,进一步证实了PHMG对菌丝伸长的抑制作用。
综上所述,盐酸聚六亚甲基胍(PHMG)能应用于防治柑桔酸腐病上。
二、柑桔保鲜剂
实施例1:
将PHMG 5g和聚季铵盐-7 2g混合,然后再加入多菌灵10g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例2:
将PHMG 25g和二癸基二甲基氯铵2g混合,再加入甲基托布津10g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例3:
将PHMG 50g和蓖麻油聚氧乙烯醚类农乳BY120  15g混合,再加入特克多20g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例4:
将PHMG 10g和十二烷基三甲基氯化铵4g混合,再加入咪鲜胺10g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例5:
将PHMG 10g和壬基酚聚氧乙烯醚农乳100号5g混合,再加入抑霉唑10g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例6:
将PHMG 5g和蓖麻油聚氧乙烯醚类农乳剂BY120 5g,再加入特克多5g、抑霉唑5g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
实施例7:
将PHMG 40g和蓖麻油聚氧乙烯醚类乳化剂 EL-80 12g,再加入咪鲜胺5g和少量水,搅拌均匀,最后加水至总重量为10 kg,搅拌均匀,得到本实施例的柑桔保鲜剂,用该柑桔保鲜剂浸泡沙糖桔整果。
本发明柑桔保鲜剂的实施效果试验
按上述实施例1~7配制保鲜剂。挑选适期采收、无病虫害和机械伤的沙糖桔果实,在上述保鲜剂中分别浸泡30秒后稍晾干,以不做浸泡处理的沙糖桔果实作对照。各实验组和对照组的沙糖桔都用0.03mm厚的聚乙烯薄膜袋包装,置于塑料筐中,每筐15kg,常温(25℃) 贮藏45天,统计各组的腐烂情况,结果见表1。

Claims (6)

1.盐酸聚六亚甲基胍在防治柑桔酸腐病上的应用。
2.一种柑桔保鲜剂,其特征在于,按总质量分数100%计,由0.05%~0.5% 盐酸聚六亚甲基胍,0.05%~0.2% 咪唑类杀菌剂、苯并咪唑类杀菌剂、嘧霉胺、氟咯菌腈、嘧菌酯中的一种或数种的混合物,0.02%~0.15% 聚氧乙烯醚类或季胺盐类乳化剂,以及余量的水组成。
3.根据权利要求2所述的柑桔保鲜剂,其特征在于,所述的盐酸聚六亚甲基胍的质量分数为0.1%~0.5%。
4.根据权利要求3所述的柑桔保鲜剂,其特征在于,所述的盐酸聚六亚甲基胍的质量分数为0.25%~0.5%。
5.根据权利要求2、3 或4 所述的柑桔保鲜剂,其特征在于,所述的咪唑类杀菌剂为咪鲜胺和抑霉唑。
6.根据权利要求2、3 或4 所述的柑桔保鲜剂,其特征在于,所述的苯并咪唑杀菌剂为多菌灵、甲基托布津和特克多。
