实施例19:结合分子的结合动力学
使用OctetRED系统和来自ForteBio的链霉抗生物素蛋白生物传感器测量CR8020和CR8043的木瓜蛋白酶裂解的Fab片段的亲和性。将H3亚型A/Wisconsin/67/2005(ProteinScience)和A/Brisbane/10/2007(ProteinScience)流感病毒血凝素抗原生物素酰化以固定在链霉抗生物素蛋白生物传感器(ForteBio)上。使用浓度范围分别在2.3-150nM和0.16-30nM之间的CR8020和CR8043,在动力学缓冲液(ForteBio,18.5032)中重复进行5次Fab结合实验。在Octet上的亲和性测量实验设置如下:将所述生物素酰化的血凝素固定至链霉抗生物素蛋白生物传感器上1800秒,随后结合系列稀释的FabCR8020和CR80431200秒,及随后在动力学缓冲液中解离1800秒。结合数据使用Octet分析软件使用1∶1模型分析。
