发明背景
免疫球蛋白(也称为抗体)是糖蛋白,可特异识别外源分子。这些被识别的外源分子称为抗原。当抗原侵入人体或动物时,启动免疫应答,包括由B淋巴细胞产生抗体。通过免疫应答,可使微生物、较大的寄生虫、病毒和细菌毒素成为无害的。抗体特异识别并以高亲和力结合与各种类型抗原的特性使其成为医学和科学研究中的有用分子。
在脊椎动物中有5种免疫球蛋白类型,包括IgG、IgM、IgA、IgD和IgE,其在免疫系统中的功能各不相同。IgGs是血液中最丰富的免疫球蛋白。其基本结构是含有2个相同的重(H)链多肽和两个相同的轻(L)链多肽。H和L链通过二硫桥和非共价键结合在一起。链本身可分为可变区和恒定区。氨基酸序列高度变化的重链和轻链可变区(VH和VL)位于抗体分子的N末端部分。VH和VL一起形成特异的抗原识别位点。其余的C末端区的氨基酸序列变化较小,称为CH1、CH2、CH3和CL。

重链抗体构成了由camelids——例如骆驼和美洲驼所产生的IgG抗体的约四分之一部分(Hamers-Casternian C,et al.(1993))。这些抗体由两个重链形成,但不含轻链。因此,可变抗原结合部分是指VHH结构域,是最小的天然发生的完整的抗原结合位点,长度约120个氨基酸((Desmyter,A.,et al.(2001))。可通过免疫作用产生针对各种抗原的具有高特异性和亲和性的重链抗体(van der Linden,R.H.,et al.(1999)),VHH部分可在酵母中克隆和表达(Frenken,L.G.J.,et al.(2000))。其表达水平、可溶性和稳定性比传统的F(ab)或Fv片段高((Ghahroudi,M.A.et al.(1997))。鲨鱼也在其抗体中也有单VH样结构域,称为VNAR(Nuttall et al.(2003));(Dooley et al.(2003));(Nuttall et al.(2004))。
Holt等(2003)综述了称为“结构域抗体”或dAbs的抗原结合片段,它仅含抗体的VH或VL结构域,因此比例如Fab和scFv更小。DAbs是已知最小的抗体的抗原结合片段,从11kDa到15kDa。它们在微生物细胞培养物中高度表达。每个dAb含抗体的6个天然存在的互补决定区(CDRs)中的3个。
由于单克隆抗体技术的发展,生成用于传统免疫治疗的抗体成为可能。因此抗体可应用于包括研究、医学及最近的消费者应用在内的多种领域。
遗憾的是,传统的免疫治疗存在很多问题。这种应用依赖于抗体的大规模生产并涉及作为从例如抗体表达系统收获的蛋白的抗体或抗体片段本身的使用。因此,包括在给药前需要进行抗体纯化等的相关生产花费都是非常昂贵的,从而阻碍了其作为免疫制剂的广泛应用。另外,为了治疗大部分群体,需要大量传统的免疫治疗产物。例如,如果治疗基于初乳和/或免疫的鸡蛋,则很难得到大规模所需量的免疫治疗产物。
另外,在最近的一篇文献中″In situ delivery of passive immunityby lactobacilli producing single-chain antibodies″NatureBiotechnol.(2002)20,702-706,Kruger等报道了通过革兰氏阳性食品级细菌玉米乳杆菌(Lactobacillus zeae)产生针对变异链球菌的scFv抗体。该治疗包括在口腔黏膜部位通过被动免疫而进行原位递送,其中只有递送的单链抗体片段在大鼠中产生针对龋齿的保护作用。
从1960年代,已报道乳杆菌具有抗腹泻特征(Beck,C,et al.Beneficial effects of administration of Lactobacillus acidophilus indiarrhoeal and other intestinal disorders.Am.J.Gastroenterol(1961)35,522-30)。最近有限的可控试验已表明乳杆菌的特定种在急性病毒性肠胃炎中具有治疗和预防特性(Mastretta,E.,et al.Effect ofLactobacillus CG and breast-feeding in the prevention of rotavirusnosocomial infection.J.Pediatr.Gastroenterol.Nutr.(2002)35,527-531)。已表明植物乳杆菌(Lactobacillus casei)和干酪乳杆菌(Lactobacillus plantarum)的选择菌株在黏膜和全身免疫应答中具有强烈的佐剂作用。
乳杆菌是一种已知用于产生食品的细菌。例如,酸奶酪通常是通过用乳杆菌发酵牛奶而制备的。然后在合适的时间将仍含活乳杆菌的发酵的酸性产物冷却消耗。
乳杆菌在食品中的另一个用途是生成肉类食品例如香肠。在包装前将乳杆菌加到肉中,然后通过一段时间进行发酵过程而成熟。
乳杆菌在食品生产中的另一个用途是腌制诸如卷心菜(泡菜)、胡萝卜、橄榄或甜菜这样的蔬菜。可通过加入适量的乳杆菌起始培养物来控制天然的发酵过程。
乳杆菌在食品中的用途通常与几种健康效果相关,参见例如A.C.Ouwehand等在Int.Dairy Journal 8(1998)749-758中的描述。特别是食品的用途与几种健康效果相关,例如与诸如IBS(肠易激综合征)、降低乳糖消化不良、腹泻的临床症状、免疫刺激、抗肿瘤活性和增强矿物质吸收这样的肠道健康相关。
WO 99/23221描述了使抗菌素失活的多价抗原结合蛋白。宿主是用于产生此后收获并使用的抗体结合片段的乳酸菌。WO 99/23211描述了加入收获的抗体片段而产生抗腹泻作用。
WO 00/65057是通过单价抗原结合蛋白抑制病毒感染。抗原结合蛋白可以是来源于诸如WO 94/04678中所述的来源于camelids的这样的天然无轻链的免疫球蛋白的重链可变结构域。WO 00/65057描述了用编码单价抗原结合蛋白的基因来转化宿主。合适的宿主可包括乳酸菌。该发明涉及发酵过程领域及阻碍发酵的噬菌体感染问题。具体地,美洲驼VHH片段本身可通过中和乳酸乳球菌(Lactoccoccuslactis)噬菌体P2来用于解决噬菌体感染问题。
WO 00/65057和WO 99/23221涉及从细菌表达系统所收获的抗体片段的用途。
US 6,605,286涉及用革兰氏阳性菌向机体递送诸如细胞因子这样的有生物活性的多肽的用途。US 6,190,662和EP 0 848 756 B1涉及获得所需蛋白或多肽的表面表达的方法。Monedero等2004涉及关于通过干酪乳杆菌表达识别轮状病毒VP8和外部衣壳片段,从而体外阻断轮状病毒感染的单链抗体(scFv)的体外研究。但这些文献均没有揭示VHH或VNAR型治疗免疫球蛋白或其片段或结构域抗体的用途。
这些已知系统的一个主要不利之处是在治疗人类疾病时使用抗体或抗体片段本身(即收获的蛋白),这可能造成抗体在提供所需的健康用途前,甚至在到达所需部位前被降解或消化。另外,需要保证抗体或抗体片段在机体的特定部位具有活性。这依赖于所治疗的特定感染类型。
导致肠道疾病的微生物是在肠道产生疾病的微生物。这种微生物的例子包括大肠杆菌和沙门氏菌。
导致肠道疾病的病原菌的另一个例子是轮状病毒。轮状病毒是世界上婴儿腹泻唯一的最常见原因,多数儿童在最初5年被感染。在发展中国家,轮状病毒诱导的腹泻每年导致600,000到870,000例死亡,在发达国家,轮状病毒疾病造成极大的经济损失。在1999年七月肠套叠相关的恒河猴轮状病毒四价疫苗取消之后,已通过几种策略来发展安全的疫苗。目前有5种口服活性衰减的轮状病毒疫苗正在进行应用于儿童的临床试验,例如Barnes,G.L.et al.(1997)。但特别是在发展中国家的年已通过被动免疫治疗进行了几项研究,例如Offit,P.A.et al.(1985)。
分泌性免疫球蛋白是指抵抗包括轮状病毒在内的多种黏膜病原体感染的第一道防线。已表明临床疾病的保护作用主要依赖于产生抗外部壳蛋白VP4或VP7的中和抗体(Ruggeri,F.M.at al.(1998));(Giammarioli,A-M.et al.(1996))。但最近已表明非中和性VP6特异IGA抗体可抑制轮状病毒复制(Feng,N.et al.(2002));Schwartz-Cornil,I.et al.(2002))。已表明超免疫牛初乳和鸡蛋黄来源的抗体可有效治疗轮状病毒腹泻(Davidson et al.(1989));(Sarker,S.A.,etal.(2001))。已证明用来源于免疫的牛初乳免疫球蛋白可成功治疗儿童的轮状病毒腹泻(Sarker S.A.et al.(1998))。但生产这些免疫球蛋白制剂的高额费用限制了其作为免疫治疗法的广泛的大规模应用。
至今仍没有获得广泛处置导致肠道疾病的微生物和病毒的特异治疗方法。例如,当前处置轮状病毒诱导的腹泻主要包括预防和口服再水合。因此,需要通过治疗和/或预防来提供处置导致肠道疾病的微生物的替代方法。
本发明涉及治疗和/预防导致肠道疾病的微生物的新方法。

发明的详细描述
本发明将通过实施例和技术方案进行描述。
总体上,本发明涉及将VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段递送到胃肠道(GIT),适于处置由肠道微生物感染所导致的疾病。
本发明也涉及含将抗体递送到GIT的递送系统的食品或药物成分,其中抗体在肠道中是有活性的。
可通过本领域已知的技术获得VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段。优选地,免疫球蛋白或其片段具有一下特征:i)在存在于GIT中的条件下具有很好的结合亲和性和所需的抑制作用;及ii)具有很好的蛋白水解稳定性,在蛋白水解酶的降解下是稳定的。
优选地,VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段,特异针对导致肠道疾病的微生物。

Holt等″Domain antibodies:proteins for therapy″Trends inBiotechnology(2003):Vol.21,No.11:484-490综述了称为“结构域抗体”或dAbs的抗原结合片段,它仅含有抗体的VH或VL结构域,因此比例如Fab和scFv更小。Dabs是抗体最小的已知的抗原结合片段,从11kDa到15kDa。它们在微生物细胞培养物中高度表达。每个dAb含抗体5个天然存在的互补决定区(CDRs)中的3个。
免疫球蛋白可以是多价的,即二价、三价、四价,其中含有多个可以结合位点。抗原结合位点可以来源于相同的亲本抗体或其片段或来源于结合相同抗原决定簇的不同抗体。如果所有结合位点具有相同的特异性,则可产生单特异性的免疫球蛋白。选择性地,可产生与相同抗原的不同抗原决定簇或甚至与不同抗原结合的多特异性免疫球蛋白。
VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段可天然产生,即对用所需抗原免疫的动物体内采血或通过基因工程技术合成。
抗体或其片段可天然产生或通过本领域已知的基因工程技术获得。
可筛选对多种不同抗原,包括微生物、较大的寄生虫、病毒和细菌毒素有活性的抗体。
也设计了VHH、VNAR和dAbs的功能等价物。功能等价物是指与全长序列有相同抗原结合亲和性的序列。例如,功能等价物包括添加或缺失不引起功能改变的氨基酸。
合成基因、将其整合到微生物宿主中并在微生物中表达基因的技术是本领域已知的,技术人员可通过常用的通用技术实现本发明。考虑了复制或整合载体的用途。
根据本发明的一个技术方案,一种包括将抗体递送到GIT的递送系统的食品或药物制剂,其中抗体是VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段,它们在肠道中是有活性的。优选地,递送系统是微生物,免疫球蛋白是美洲驼来源的抗体或其片段。我们惊奇的发现这些转化的微生物可在其表面表达骆驼重链抗体或其片段,能降低病毒装载,使病理学规范化,并在轮状病毒感染的动物模型中减轻腹泻。另外,骆驼的重链抗体或其片段可在轮状感染的体外和体内模型中有效降低感染。
本发明也包含不表现出抗原结合亲和性的整个抗体片段或部分的用途。片段优选地是功能片段。重链的功能片段是指免疫球蛋白重链的片段,该片段对同全长序列类似的抗原具有结合亲和性。当解离常数大于10exp5时具有结合亲和性。这种片段可利于治疗。例如,它可能具有很小的免疫原性,如果需要,它由于具有更小的体积而可以穿过组织。
抗体适于递送到GIT中,而且必须在胃和/和肠道中是有活性的。本发明的递送系统的一个优点包括体内产生或释放含VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段,定位于GIT中避免了口腔给药抗体在胃中降解的实际问题。
为了测定VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段是否适于本发明,可进行下列检验。
产生的抗体在低pH,优选地从1.5到3.5,并存在胃蛋白酶(一种在胃中含量丰富的蛋白酶)的特定条件下进行筛选,以获得在GIT中很好工作的非常有用的分子,从而适于本发明的用途。
本发明通常用于处置导致肠道疾病的微生物。优选地,本发明涉及处置导致肠道疾病的病毒和/或导致肠道疾病的细菌。处置可理解为包括治疗和/或预防。
导致肠道疾病的微生物包括例如沙门氏菌、Campilobacter、大肠杆菌或螺杆菌。导致肠道疾病的病毒包括例如Noro病毒(诺沃克类病毒)、肠道腺病毒、冠状病毒、杯状病毒和小病毒。轮状病毒和诺沃克家族的病毒是病毒性胃肠炎的主要原因,但许多其它病毒也可能与爆发有关。最优选地,本发明涉及处置轮状病毒感染。
本发明也用于治疗其它非导致肠道疾病,例如病毒性肝炎。
选择性地,递送系统包括益生微生物。益生微生物优选地可在通过GIT后存活,并在胃/肠道中具有活性。优选地,微生物可在GIT中产生短暂克隆;在GIT中表达基因;并可刺激肠道免疫系统。

实用来说,微生物可以是乳酸细菌(lactic acid bacterium)。更优选地,微生物从乳杆菌属(Lactobacillus)或双歧杆菌属(Bifidobacterium)选择。更优选地,微生物是乳杆菌(Lactobacillus)。具体地,乳杆菌是干酪乳杆菌393pLZ15。干酪乳杆菌最近重新鉴定为类干酪乳杆菌(Lactobacillus paracasei)(Perez-Martinez,2003)。
微生物也可以是益生菌芽孢杆菌种,如Le H.Due et al.,Appliedand Environmental Microbiology,2004,p.2161)所述,更优选地是蜡质芽孢杆菌(Bacillus cereus)、克劳氏芽孢杆菌(Bacillus clausii)和微小芽孢杆菌(Bacillus pumilus)。
基于益生菌的系统是安全的、吸引人的方法,是最便宜的抗体产生系统。微生物——优选地是表达的抗体相对较容易的乳杆菌的大规模应用,需要最小的处理和储存费用,是比较经济的。
选择性地,微生物可以是酵母。合适的酵母包括烘烤用的酵母
酿酒酵母。其它的酵母例如博伊丁假丝酵母(Candida boidinii)、多形汉逊酵母(Hansenula polymorpha)、甲醇毕赤酵母(Pichiatnethanolica)和巴斯德毕赤酵母(Pichia pastoris),也是已知的产生异源蛋白的系统,可用于本发明中。
丝状真菌、特别是来自木霉和曲霉属的种可在其培养基中分泌大量蛋白、代谢物和有机酸。该特性广泛应用于食品和饮品工业,其中由这些丝状真菌种分泌的化合物已使用多年。
应理解这些微生物可以是遗传修饰的微生物。
根据本方面的递送系统包含用编码VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段的基因转化的微生物。这种基于益生菌的系统是一种安全的吸引人的将抗体递送到GIT的方法。益生菌在本领域是已知的,是一种最便宜的抗体产生系统。因此,大规模应用乳杆菌表达抗体相对较容易,需要最小的处理和储存费用。另外,益生菌至少可在肠道中产生短暂克隆数天、数周或几个月,可在肠道中保持更长的时间,从而持续产生抗体,以更持续并延长对导致肠道疾病的微生物或病毒的保护作用。本发明基于以下发现:本发明的VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段可用于治疗或预防由导致肠道疾病的微生物所造成的感染。
更具体地,VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段可用于治疗或预防由轮状病毒引起的病毒性胃肠炎或腹泻。
本发明的进一步优点是使用了益生菌微生物表达VHH或VNAR型重链免疫球蛋白或其功能片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其功能片段,可使微生物——例如乳杆菌能提供与该益生菌微生物相关的正常的健康益处,并在处置需要进行治疗的感染中具有预防/治疗的益处。该“双重作用”的治疗具有协同作用,从而比本领域已知的治疗方法对病人具有更大的健康益处可。
根据本发明的一个技术方案,VHH型重链免疫球蛋白或其功能片段来自camelids,包括美洲驼和骆驼。重链抗体的选择性来源是鲨鱼。最近表明鲨鱼在其抗体中也具有单个VH类似的结构域,称为VNAR。根据本发明可使用VNAR型免疫球蛋白片段。也可考虑使用前述功能等价物。选择性地,可使用重链或轻链免疫球蛋白的结构域抗体(dAbs)或其片段。优选地,免疫球蛋白是美洲驼来源的重链抗体或其片段。已在本领域发现多种美洲驼来源的重链抗体片段。更优选地是对轮状病毒,特别是轮状病毒株Wa、CK5、Wa1、RRV或CK5的结合亲和性的解离常数至少为10exp 5的重链免疫球蛋白或其片段。
因为已发现美洲驼重链抗体可有效治疗轮状病毒感染,因此它是特别有利的。已发现美洲驼VHH抗体片段在轮状病毒感染过程中可降低病毒装载、使病理学规范化,并可减轻腹泻。
特别优选地,在本发明的序列表,SEQ ID No 1到21中提供了对轮状病毒具有亲和性的美洲驼来源的VHH序列。选择性地,根据本发明,与SEQ ID No.1至少有70%、80%、85%、90%、95%、98%或99%氨基酸同源性并对轮状病毒颗粒或抗体具有亲和性的VHH序列也是优选的技术方案。如本领域所述,VHH序列可来源于骆驼,通过免疫和/或亲和性筛选获得,也可来源于其它诸如鼠或人这样的哺乳动物,和/或通过氨基酸替代使其骆驼化。在另一个技术方案中,VHH序列可被融合而产生具有2、3、4、5或多个VHH单位的多聚体单位,选择性地通过间隔分子连接。在另一个技术方案中,可将几个单独的或在多聚体分子中的VHH序列组合。优选地,VHH序列具有不同的特异性,例如可组合VHH序列而对特定病原体提供广谱亲和性。在特别优选的技术方案中,将对任何一个轮状病毒株Wa、CK5、Wa1、RRV或CK5具有亲和性的2、3、4、5或更多的VHH序列组合成单独的单体单位或载体上——例如在益生菌和/或多聚体分子上——的组合单位。
另外,也出乎意料地发现美洲驼重链抗体适于在GIT中给药。美洲驼重链抗体高度抵抗胃中蛋白酶降解,可抵抗胃中的酸性环境。尽管蛋白水解系统在GIT中比在例如口中活性更高。肠道中的活性被蛋白水解活性——包括蛋白酶和肽酶所抑制。我们已惊奇地发现在GIT中体内产生或释放抗体片段可解决口服抗体在胃中和肠道中降解的实际问题。本发明是第一个可在GIT中表达抗体的系统,适于治疗轮状病毒感染。
当选择益生菌微生物作为递送系统时,我们发现这些转化的微生物可在其表面表达美洲驼重链抗体或其片段,从而降低病毒装载、使病理学规范化,并在轮状病毒感染的动物模型中减轻腹泻。
美洲驼重链抗体在GIT中由微生物表达。美洲驼来源的VHH抗体片段可在微生物表面和/或作为微生物的分泌蛋白表达。优选地,VHH抗体片段的分泌形式是多聚体形式的,以增强病毒装载的聚集和清除。
优选地,微生物或更优选地益生菌,是用含美洲驼重链抗体或其片段的基因的表达载体来转化的。表达载体可包含组成型启动子以表达抗体或其片段。这种组成型启动子可支持在给药后,抗体由转化的乳杆菌进行持续的(至少短时间)原位表达。选择性地,可选择仅在GIT中和/或胃/肠道中有活性——例如仅适于GIT特异表达的启动子。这可保证在GIT中表达和/或分泌美洲驼重链抗体或其片段。多种乳杆菌组成型启动子在本领域是已知的,特异的在GIT中表达的诱导型启动子的例子是Pldh(Pouwels et al″Lactobacilli as vehicles fortargeting antigens to mucosal tissues by surface exposition of foreignantigens″Methods in Enzymology(2001)336:369-389)。
实施例中描述的表达载体可在转化的乳杆菌中复制,并表达抗体片段。应理解本发明不对这些复制表达载体进行限定。在本领域已知:可将整个表达盒插入到所谓的“整合”质粒中,从而使表达盒在转化后整合到乳杆菌染色体中。(Pouwels,P.H.and Chaillou,S.Geneexpression in lactobacilli(2003)Genetics of lactic acid bacteria page 143-188)。
根据选择的技术方案,如果选择胶囊作为递送系统,胶囊方法应该在通过胃和GIT后可存活,并可在一段时间内持续释放抗体。从而保证将美洲驼重链抗体或片段随时间递送到胃中。美洲驼重链抗体或VHH或VNAR片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段,由于在释放时可在肠中存活的能力,因此特别适于该方法。
根据本发明的技术方案,抗体递送到GIT中可通过使用食品和制药工业已知的胶囊而实现。可使用天然生物多聚体。例子包括Caalginate、角叉菜胶、结冷胶或明胶。递送系统可以是本领域已知的胶囊方法,将免疫球蛋白或其片段特异地递送到肠道中。因此胶囊必须存活,直到进入肠道中,然后进行释放。这种递送系统包含通用的保护系统,可避免抗体降解。这种技术包括脂质体引导法、转盘扫描和凝聚过程。可使用任何触发因子来促进胶囊化成分的释放,例如pH改变(肠包被)、机械压迫、温度、酶活性。这些技术在Sebastien Gouin″Microencapsulation:industrial appraisal of existing technologies andtrends″Food Science and Technology(2004)15:330-347中进行了扩展。选择性地,胶囊化方法可使抗体在肠道和/或胃中缓慢释放。从而使抗体或其功能片段或等价物在一段时间内持续释放。
美洲驼重链抗体或其片段可用于治疗或预防病毒感染,例如病毒性胃肠炎或轮状病毒感染。
具体地,抗体可通过已用美洲驼重链抗体转化的微生物递送到GIT中,包括步骤i)用编码美洲驼重链抗体的基因转化微生物,及ii)对需治疗的人或动物的GIT中给药转化的微生物。
递送系统可包含用在肠道中表达和/或分泌的抗体或抗体片段转化的微生物。因此,使用微生物作为递送系统具有在GIT环境中体内定位产生抗体片段的优点,从而解决了口服抗体在胃中降解的实际问题。这种基于益生菌的系统是一种安全的吸引人的将抗体递送到GIT中的方法。因此,大规模使用乳杆菌表达抗体是相对较容易的,需要最小的处理和储存费用,是非常经济的。另外,益生菌将在肠道中存活更长的时间,从而可持续产生抗体,以更持续地抵抗导致肠道疾病的微生物。
微生物在本发明的食品中合适的量是每份106至1011或者(例如,如果每份体积未知)为100克产物106至1011,更优选地,这些水平为每份或每100g产物108到109。
根据本发明的抗体必须在肠道/胃中有活性,即它们必须是有功能的,并保持灭活靶标的正常活性。根据本发明的活性抗体应与其靶标正常结合,因此,抗体对抗原的结合亲和性应该是正常的。当解离常数大于105时具有结合亲和性。
因此,根据本发明的食品或药物制剂可选择性地针对特定的疾病或疾病症状。抗体的选择取决于待治疗或减轻的疾病或症状。
应理解当产物是食品时可使用任何抗体。但当产物是药物制剂时,优选地是VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段。
优选地,抗体或其片段应具有一个或多个以下特征:
i)在存在于G/I通道的条件下具有较好的结合亲和性和所需的抑制功能;及ii)因为对蛋白水解酶的降解是稳定的,因此具有较好的蛋白水解稳定性。iii)抗体应该是热稳定的,从而使其可包含在各种食品中。可通过巴斯德消毒来制备食品,优选地是尽管进行热处理,但抗体的活性大部分保留。
抗体或其片段应可在肠道中表达和分泌。已知本领域中的几种模拟GIT条件的检测法用于例如筛选可在GIT条件下存活的合适的益生菌。在Picot,A.and Lacroix,C.(International Dairy Journal 14(2004)505-515)中描述了测定抗体是否在GIT条件下存活的检测法。
为了测定抗体是否适于本发明,可使用下述检测方法。产生的抗体在低pH、优选地1.5到3.5,存在胃蛋白酶(一种在胃中含量丰富的蛋白酶)特定的条件下进行筛选,从而得到在G/I通道中工作,适于本发明的高效分子。
本发明可用于处置导致肠道疾病的微生物。导致肠道疾病的微生物包括病毒或导致肠道疾病的细菌。疗处置是指治疗和/或预防。
本发明的进一步的技术方案是含上述递送系统的食品或药物制剂。可根据本发明制备几种食品,例如膳食替代物、汤、面条、冰激凌、调料、调味品、涂抹食品、快餐、谷物、饮料、面包、饼干、其它烘烤食品、甜点、bars、巧克力、树脂、奶制品、减肥食品,例如减肥食品或膳食替代物等。虽然上述类型在食品应用中是优选的,但本发明的食品可用作饮食的补充。

含用抗体或其功能片段转化的微生物的给药递送系统的替代方法包括给药食品——在其至少一个表面含包被的递送系统——的分散工具。术语分散工具包括管、吸管、匙或棒或其它用于向用户递送液体或半固体食品的工具。分散工具也用于向用户递送固体食品。分散用的管或吸管特别适于特定的饮料,其中过高或过低的pH和/或温度是指不推荐直接向饮料中加入微生物。
当本发明的递送系统含胶囊化抗体或甚至抗体本身时也可使用分散工具。
分散工具含至少一个包被微生物、胶囊化抗体或抗体本身的表面。分散工具用这些颗粒包被后,将工具储存于不透水和其它污染物的外膜中。含这些颗粒的包被材料是对人和细菌无毒性的,可使用诸如玉米油或蜡这样的油。在US 6,283,294 B1中描述了这方面的内容。当含颗粒的分散工具插到饮料或半固体食品中,则颗粒整合到食品中,对一份食品给予所需剂量的抗体。
优选地,胶囊化抗体、抗体本身或微生物可悬浮在水中,然后使用分散工具并进行蒸发干燥。使用该方法可使分散工具包被胶囊化颗粒、抗体本身或微生物,然后当分散工具与液体或半固体食品接触时进行释放。
本发明的进一步技术方案涉及制备含将抗体递送到GIT中的递送系统的食品或药物制剂的方法,其中抗体在肠道中是有活性的,包括将递送系统在制备食品时加入到食品或药物制剂中。
微生物在食品中必须是有活性的,因此,例如,如果在加工时对食品加热,则微生物必须在加热步骤后加入(后给料)。但如果用微生物对食品进行发酵,则不能进行发酵后的加热步骤。如果产物是液体产物,则可用分散工具例如吸管进行微生物给药。
本发明的进一步技术方案涉及含根据本发明的递送系统并在给药后对病人肠道产生健康效果的食品或药物制剂的用途。这种健康效果包括抗体提供的特定的健康效果。微生物本身可提供几种健康效果,例如对肠道有益的IBS(肠道易激综合征)、降低乳糖消化不良、腹泻的临床症状、免疫调节、抗肿瘤活性、佐剂效果及增强矿物质吸收。
根据本发明的食品或药物制剂可用于处置,包括治疗或预防导致肠道疾病的细菌或病毒引起的感染。其它包括在本发明中的抗体也可提供多种其它的健康效果。
本发明基于以下认识:本发明的VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段可用于治疗或预防由导致肠道疾病的微生物所造成的感染。另外,VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段可用于治疗或预防由导致肠道疾病的微生物轮状病毒引起的病毒性胃肠炎或腹泻。
本发明的进一步优点是含表达VHH或VNAR型重链免疫球蛋白或其片段,或免疫球蛋白重链或轻链的结构域抗体(dAbs)或其片段的益生菌的食品或药物制剂的用途,可使微生物,例如乳杆菌提供与该微生物相关的正常健康效果,以及在需要进行治疗的感染的处置中提供预防/治疗效果。该“二元效果”治疗可对病人提供比本领域已知方法更大的健康效果。
根据本发明的另一个技术方案,VHH型重链免疫球蛋白或其片段来源于camelids、包括美洲驼和骆驼。已在本领域中发现许多美洲驼来源的治疗抗体片段。更优选地是对轮状病毒,特别是轮状病毒株Wa、CK5、Wa1、RRV或CK5具有解离常数至少为105的结合亲和性的重链免疫球蛋白或其片段。
我们惊奇地发现美洲驼重链抗体可有效治疗轮状病毒感染。当使用的抗体是美洲驼重链抗体时,产生的健康效果包括抗腹泻作用。因此,美洲驼重链抗体可用于治疗轮状病毒感染,包括治疗或预防轮状病毒感染。我们发现美洲驼VHH抗体片段在轮状病毒感染时可降低病毒滴度,使病理正常化并减轻腹泻。轮状病毒仍然是世界上婴儿腹泻的单一的最常见的原因,大多数儿童在5岁前感染。在发展中国家,轮状病毒诱导的腹泻每年可造成600,000到870,000例死亡,在发达国家,轮状病毒疾病造成大量经济损失。
另外,也出乎意料地发现美洲驼重链抗体适于在肠道中给药。我们惊奇地发现美洲驼重链抗体在胃中高度耐受蛋白酶的降解,而且可耐受胃中的酸性环境。尽管在肠道/胃中的蛋白水解系统比例如口中的环境更有活性。在GIT定位体内产生抗体片段可解决口服的给药抗体在胃中降解的实际问题。本发明是第一个可在GIT中表达抗体的系统,适于治疗轮状病毒感染。
因此,含表达美洲驼重链抗体的食品或药物制剂的用途可使乳杆菌提供正常的与该益生菌相关的健康作用,并在轮状病毒感染的处置中提供预防/治疗作用。本发明是第一个可在GIT中表达抗体的系统,适于治疗轮状病毒感染。
应理解可给药食品或药物制剂以对病人提供健康效果和/或对抗特定的疾病或感染。抗体的选择依赖于待治疗的疾病。
优选地,用含美洲驼重链抗体或其片段的表达载体转化微生物。可使用整合型或复制型载体。
如果选择胶囊作为递送系统,则胶囊方法应在通过GIT时在胃中存活,应该在一段时间内提供持续的抗体释放。从而保证美洲驼重链抗体或片段在一段时间内递送到胃中。美洲驼重链抗体或其重链由于其在释放时具有可在肠道中存活的能力,因此特别适于胶囊化方法。
具体地,可使用用美洲驼重链抗体转化的微生物将抗体递送到GIT中,包括步骤i)用编码美洲驼重链抗体的基因转化微生物;及ii)对需治疗的人或动物的GIT给药转化的微生物。
人造黄油和其它涂抹食品
典型地,它们是水包油或油包水的乳液,也可以是覆盖的基本不含脂肪的涂抹食品。典型地,这些产品在温度例如2-10℃是易涂开的,但流动不通畅。脂肪水平在很大范围变化,即全脂的人造黄油含60-90wt%脂肪,中等脂肪的黄油含30-60wt%脂肪,低脂肪黄油含10-30wt%脂肪,更低或无脂肪黄油含0到10wt%脂肪。
人造黄油和其它涂抹食品的脂肪可以是任何食用脂肪,通常为豆油、葵花籽油和棕榈油。可使用修饰形式的脂肪,例如氢化、酯化、提纯等。其它合适的油是本领域已知的,可根据需要进行选择。
人造黄油或涂抹食品的pH优选地为4.5到6.5。
除了人造黄油外的其它涂抹食品的例子是干酪涂抹食品、甜味涂抹食品、酸奶酪涂抹食品等。
涂抹食品进一步可选的成分为乳化剂、着色剂、维生素、防腐剂、树脂、增稠剂等。产物通常在水中平衡。
人造黄油或其它涂抹食品平均一份的典型大小是15克。优选地,每份人造黄油或涂抹食品中产VHH的乳杆菌是106至1011,更优选地为每份108至1010。必须在加热过程后无菌加入乳杆菌菌株。选择性地,可向食品中加入胶囊化VHH。优选地每份加入25到5000μg,更优选地每份为50到500μg。最优选地,每天食用2到3份。
冷冻的甜点食品
对于本发明而言,术语冷冻的甜点食品包括含诸如冰淇淋、冷冻黄油、冰冻果子露果汁冰糕、冰牛奶和冷冻奶油冻、冰水物、格兰尼它冰糕及冷冻的水果汤这样的冷冻甜点的牛奶。优选地,冷冻甜点中固体的水平(例如糖、脂肪、调料等)为大于3wt%,更优选地为10到70wt%,例如40到70wt%。
冰激凌典型地含2到20wt%脂肪、0到20wt%甜料、2到20wt%非脂肪奶成分和诸如乳化剂、稳定剂、防腐剂、香料成分、维生素、矿物质等这样的选择性成分,用水平衡。典型地冰激凌是充气的,例如使其扩大20到400%,更通常为40到200%,并在-2到-200℃,更优选地-10到-30下冷冻。冰激凌通常含0.1wt%的钙。

饮料、例如基于茶的产品或食品替代物
乳杆菌可有益用于饮料例如果汁、软饮料等中。根据本发明的一种非常有益的饮料是基于茶的产品或食品替代物饮品。这种产品在下面进行详细描述。显然可对其它含由乳杆菌细菌产生的维生素的饮料中使用同样的水平和成分。
根据本发明的目的,术语基于茶的产品是指含茶或替代茶的草药成分——例如茶包、茶叶、草药茶包、草药液、茶粉、草药茶粉、冰茶、冰草药茶、碳酸冰茶、碳酸草药液等的产品。典型地,本发明中这些基于茶的产品在使用前需进行制备步骤,例如从茶包、茶叶、草药茶包或草药液进行茶的酿造或对茶粉或草药茶粉进行增溶。对这种产品,优选地是调整产品中乳杆菌的水平,以使一份最终产品中含有上述乳杆菌所需的水平。

对提取而获得终产物的食品而言,通常的目标是保证200ml或200克的每份食品含有如上文所述的所需的量。在本文中,应理解到,通常存在于基于所提取产品的茶中的乳杆菌仅有一部分最终被提取到最后的茶饮料中。为了补偿这种作用,通常向待提取的食品中掺入2倍提取物所需的量。
对茶叶或茶包而言,典型地使用1-5克茶来制备一份200ml的饮料。
如果使用茶包,可将乳杆菌掺入到茶成分中。但对某些用途而言,将茶与乳杆菌分离是有利的,例如插入单独的茶包间隔或在茶包纸上使用。
色拉调味品或蛋黄酱
通常调味品或蛋黄酱是水包油乳液,乳液中的油相通常占产物的0到80wt%。对非脂肪还原产物来说,脂肪水平典型地为60到80%,对色拉调味品而言,脂肪水平通常为10-60wt%,更优选地为15-40wt%,低或无脂肪调味品可含例如甘油三酸酯的水平为重量的0、5、10、15%。
调味品和蛋黄酱通常是低pH产物,优选地pH为2-6。
调味品或蛋黄酱选择性地可含有其它成分,例如乳化剂(例如蛋黄)、稳定剂、成酸剂、多聚物、散装成分、香料、着色剂等。成分的平衡液是水,存在的水平是0.1到99,9wt%,更优选地为20-99wt%,最优选地为50到98wt%。

食品替代物快餐或bars

柠檬粉
乳杆菌也可以以小袋用于干粉中,通过速溶在水中而得到凉爽的柠檬水。这种粉末可含有基于食品的载体,例如麦芽糊精或其它载体。选择性的其它成分可以是着色剂、维生素、矿物质、防腐剂、树脂、增稠剂等。
平均一份食品或人造黄油或其它调味品的典型大小是30到50克。优选地产VHH的乳杆菌在柠檬粉中为每份106至1011,最优选地为每份108至1010。根据本领域技术人员已知的方法,必须将乳杆菌菌株喷洒在载体上,从而使其保持活性。选择性地,可向食品中加入胶囊化VHH。优选地每份加入25到5000μg,更优选地每份加入50到500μg。
在所有上述食品中,可加入作为活体培养的(湿的)生物量或作为干燥成分——但仍含本领域已知的活微生物——的转化微生物。

实施例2
体外抑制轮状病毒
从苏格兰中洛锡安郡Moredun研究所获得牛轮状病毒ComptonCK5,BS-C1细胞购自欧洲动物细胞保藏中心(European Animal CellCulture Collection)。
将BS-C1细胞培养在补加10%热灭活的胎牛血清、1%MEM氨基酸溶液(100×)、20mmol I-1L-谷氨酸、100I.U.ml-1青霉素、100μg ml-1链霉素和2.5μg ml-1两性霉素B(均购自Sigma,US)的Earles改进基本培养基中。
将CK5轮状病毒储液在添加1%MEM氨基酸溶液(100×)、20mmol I-1L-谷氨酸和0.5μg/ml结晶胰岛素的无血清的培养基(SFM)EMEM中稀释,然后将稀释的5ml种液加到培养在162cm2组织培养烧瓶(Costar,UK)中的BS-C1细胞的汇合细胞单层中。在37℃使病毒在细胞上吸收1小时,然后将培养基加到75ml。将烧瓶在37℃温育至观察到完全的细胞病变作用。将培养物冻(-70℃)融2次,然后集中并于1450g,4℃离心15分钟,除去细胞碎片。将上清轻轻倒出,并等份储存在-70℃。
将单层BS-C1细胞于37℃,95%空气和5%二氧化碳下在12孔组织培养板中培养。在使用前至少2小时的时候,除去培养基,用SFM替代。在SFM中稀释CK5病毒至50pfu/ml。在SFM中稀释筛选的抗轮状病毒片段,然后将等体积病毒和片段稀释液混合(总体积为200μl),于37℃温育1小时。
然后将病毒片段混合物铺在单层细胞上(每种重复3个孔)。将平板于37℃,95%空气和5%二氧化碳下温育1小时。然后,除去病毒,并加入含100I.U.ml-1青霉素、100μg ml-1链霉素、2.5μg ml-1两性霉素B和0.1μg/ml结晶胰岛素,及0.75%Sea Plaque琼脂(FMC)的EMEM覆盖层。然后将平板于37℃,95%空气和5%二氧化碳下温育4天。用溶在10%甲醛中的1%结晶紫染色后,除去琼脂糖,用水洗涤孔,并对斑进行计数。
根据上述方法,从23个检测的克隆中得到9个可产生中和该轮状病毒株的抗体片段。在斑检测实验中,片段2B10和1D3中和轮状病毒的作用最有效(图1)。
图1所示的是通过体外斑检测法测定的轮状病毒CK5的病毒中和作用。在每个数据点显示了4个测定值的平均中和速率。如果在数据点的伸展超过10%,则表示有两个较大的测定值(虚线)。将检测的抗体片段分为2个图,A和B。A的阴性对照是或者省略病毒(无病毒),或者没有加入非轮状病毒特异的VHH。非特异VHH片段对人孕激素hCG是特异的。该片段的分离如(Spinelli,S.,Frenken,L.,Bourgeois,D.,de Ron,L.,Bos,W.,Verrips,T.,Anguille,C,Cambillau,C.and Tegoni,M.″The crystal structure of a Llama heavy chain domain″.Nat.Struct.Biol.(1996)3:752-757;Frenken,L.G.J.,et al.″Isolation of antigenspecific Llama VHH antibody fragments and their high level secretionby Saccharomyces cerevisiae″.J.Biotechnol.(2000)78,11-21)所述。
因此,通过本方法可鉴定多种可在体外系统中抑制轮状病毒感染的VHH片段。
实施例3
轮状病毒在鼠体内的抑制
将某些通过实施例2所述的方法筛选的VHH片段用于体内实验中,以研究这些抗体片段在鼠中预防或治疗轮状病毒诱导的腹泻的效率。该模型系统常用于研究轮状病毒感染(Ebina,T,Ohta,M,Kanamaru,Y,Yamamotoosumi,Y,Baba,K.″Passive immunisations ofsuckling mice and infants with bovine colostrum containing antibodies tohuman rotavirus″.J Med Virol.(1992)38:117-123)。
从丹麦获得怀孕14天、无轮状病毒的BALB/c鼠。将鼠在Huddinge医院的动物中心进行单独培养。从瑞典Huddinge医院Karolinska研究所的当地伦理委员会获得批准。正常的块状食物和水随意提供。为了检测片段在轮状病毒(RV)结合后是否可抑制感染,将筛选的VHH片段在用于感染的一天前与滴定量的RRV(2×107ffu)预混合。
用VHH片段每天处理4天龄的幼鼠,包括第0天(感染前一天)到第四天(图2),对腹泻进行评估。对抗体片段2B10观察到腹泻发生明显降低,如图2所示。与未处理组相比较,用片段2B10处理的组在第二天产生腹泻的幼鼠数目显著降低。另外,与其它RRV处理的组的多数幼鼠相比较,在第3、4、5天,2B10处理组的幼鼠没有一只表现出腹泻症状(图2)。与未处理组相比较,没有发现无关VHH片段RR6(针对含氮染料;Frenken,L.G.J.,et al.″Isolation of antigen specificLlama VHH antibody fragments and their high level secretion bySaccharomyces cerevisiae″.J.Biotechnol.(2000)78,11-21)具有统计学显著的作用。
另外,对每个处理组计算了每只幼鼠腹泻的平均天数,以每只幼鼠的腹泻天数除以每个处理组幼鼠的总数目表示。与未处理组2.87±0.29天相比较,片段2B10处理的组为0.33±0.21天,是显著降低的。
需要注意的是,从现在开始,在以下实施例中,含编码片段2B10的pUR5071来源的质粒命名为pUR655,编码的抗体重新命名为VHH1。
实施例4(a到k)
如下进行分别与实施例1到3类似的进一步实验:
a)构建抗轮状病毒scFv-2A10和VHH1的表达载体。
从抗轮状病毒mAb 2A10(IgA类型)分泌杂交瘤(Giammarioli etal.(1996)Virol.225:97-110))中提取总RNA。用5′RACE试剂盒(5′RACE System for Rapid Amplification of cDNA Ends(第二版,InvitrogenTM life technologies,Carlsbad,CA)扩增编码重链(VH)和轻链(VK)可变区的序列。VH 5′RACE的引物如下:

用含ClaI限制性位点的正向引物和含EcoRI限制性位点的反向引物从pUR655扩增VHH1,然后将其插入到pBS-E-tag载体中。为了产生表面表达的抗体片段,将scFv-2A10-E-tag和VHH1-E-tag从pBS-E-tag载体上通过ClaI和Xhol限制性位点切割,并与干酪乳杆菌蛋白酶P蛋白的最后244个氨基酸编码的锚定序列融合(Kriiger et al,Nature Biotechnol(2002)20:702-706),然后插入到乳杆菌表达载体pLP502中(图3A和3C)。为了产生分泌的抗体片段,通过PCR扩增在E标记后插入一个终止密码子(TAA),将产物插入到pLP502的ClaI和Xhol限制性位点之间(图3B和3D)。pLP502载体含乳糖脱氢酶基因的组成型启动子(Pldh)(Pouwels et al.″Lactobacilli as vehicles for targeting antigens to mucosal tissues bysurface exposition of foreign antigens″Methods in Enzymology(2001)336:369-389)。对类干酪乳杆菌的转化如前所述进行(上述Kriiger etal(2002))。
b)比较转化的乳杆菌中抗体特异的转基因的表达水平。
从培养至OD600为0.8的不同乳杆菌构建体中提取总RNA(QIAGEN),用RQ1DNase(Promega)消化残余的DNA后进行反转录。通过qPCR core试剂盒及green I(MedProbe,Oslo,Norway)和ABI PRISM 7000序列检测系统(PE Applied Biosystems,Foster City,CA)测定不同抗体片段的mRNA的量。典型的时间分布为:起始步骤95℃,10分钟,然后进行第二个步骤,95℃15秒,58℃1分钟,40个循环。产生的cDNA用于产生16S rRNA和抗体插入片段的标准曲线,对16S rRNA使用引物p0(5′GAGAGTTTGATCCTGGCTCAG 3′)和p6(5′CTACGGCTACCTTGTTACGA3′),对插入片段使用引物prtpsp(5′TCTTGCCAGTCGGCGAAAT 3′)和XhoI-VHH(5′CCGCTCGAGTGCGGCACGCGGTTCC 3′)。
c)分泌的抗体片段的纯化。
将含构建体的类干酪乳杆菌培养至OD600为0.8,以纯化分泌的VHH1抗体片段。离心后,将上清的pH调整为7,并通过0.45μm过滤器进行过滤。随后根据RPAS纯化系统(Amersham-Bioscience,Little Chalfont,Buckinghamshire,UK)提供的说明书将分泌的抗体片段进行纯化。用Spectra/Porμ膜MWCO 6-8000(Spectrum MedicalIndustries,Inc.,Los Angeles,CA)对1×PBS于8℃进行过夜透析。将纯化的抗体片段进行15%SDS-聚丙烯酰胺凝胶电泳,验证样品的纯度,并通过BioRad蛋白测定法(BioRad Laboratories,Hercules,CA)测定总蛋白的浓度。
d)蛋白提取和蛋白浓度测定。
将含构建体2A10-锚定物、VHH1-锚定物、分泌型2A10、分泌型VHH1、分泌型无关物质及无关物质锚定物的类干酪乳杆菌在含3μg/ml红霉素的MRS肉汤中培养至OD600为0.8。将细菌在含10mg/ml溶菌酶的10mM Tris-HCl,pH 8.0中,于37℃裂解1小时,然后通过占空比为60%的超声进行破碎(6×30s开/关循环)(Digital Sonifier,model 250,Branson Ultrasonics coorporation,Danbury,CT)。通过离心除去碎片。通过超滤(Amicon,Beverly,MA)将上清浓缩50倍。通过BioRad蛋白测定法如上所述测定蛋白浓度。
e)酶联免疫吸收测定法和流式细胞技术。
用兔抗人轮状病毒血清(1/1000)包被ELISA 96孔板。用1∶100稀释的恒河猴轮状病毒储液(RRV)进行第二次包被。封闭后,将平板与蛋白提取液或浓缩的上清一起温育。加入鼠抗E标记的抗体(Amersham Pharmacia Biotech,Bucks,UK)或兔抗美洲驼抗体、辣根过氧化物酶耦联的羊抗鼠抗体或猪抗兔抗体(DAKO AJS,Glostrup,Denmark)和3,3′,5,5′-四甲基联苯胺(TMB),用Vmax微板读数仪(Molecular Devices,Sunnyvale,CA)测定630nm处的吸收值。所有抗体进行1/1000稀释。以纯化的VHH1-E-标记和单克隆2A10抗体为标准,测定不同乳杆菌转化子产生的抗体片段的浓度。
根据标准程序用抗E标记抗体进行流式细胞计数,通过FACS计数仪(Becton Dickinson,Stockholm,Sweden)分析样品。
结果如图4a所示。
f)电子显微技术SEM TEM。
对扫描电子显微镜(SEM)而言,将在表面表达VHH1的乳杆菌转化子和未转化类干酪乳杆菌与RRV混合,温育后,固定并加到包被RC58聚L-赖氨酸包被的玻片上。通过SEM(JEOL JSM-820,Tokyo,Japan)在15kV分析玻片。
对透射电子显微镜(TEM)而言,将RRV加到载网上,滴加来自表达分泌型VHH1的上清(25倍浓缩)或来自未转化的类干酪乳杆菌的上清。然后加入鼠抗E标记抗体(1∶1000)和10nm金标记的羊抗鼠IgG抗体(1∶1000)(Amersham Biosciences)。通过TEM在80KV分析载网(Tecnai 10透射电子显微镜,Fei Company,TheNetherlands)。
结果如图4b和图11所示。
g)病毒的产生和纯化。
将恒河猴轮状病毒如前所述培养在MA104细胞中。在整个研究中使用斑纯化的RRV。通过用RRV在每毫升含0.5μg胰岛素(SigmaChemical Co.,St.Louis,Mo.)的无血清M199培养基(GibcoLaboratories,Grand Island,N.Y.)以0.1的感染复数(MOI)感染MA104细胞,从而产生用于整个研究过程的单个病毒储液。当细胞病变作用达到单层的75%时,将细胞冻融两次,并通过低速离心除去细胞裂解物。将病毒上清分成小份,储存在-80℃备用。通过免疫过氧化物酶聚焦还原实验测定病毒的滴度。产生单个病毒储液用于整个实验中。
h)体外中和检测法。
通过如前所述的微中和检测法(Giammarioli et al 1996,如上)对表达抗体的乳杆菌进一步检测其对轮状表达的抑制作用。对锚定的抗体片段,将细菌在MEM培养基中系列稀释,并与200ffu胰岛素激活的RRV(100μl)于37℃温育1小时。温育完毕后,通过离心除去细菌,用上清接种到培养在96孔板上的MA104细胞单层。将纯的或稀释在MEM中的浓缩的来自分泌抗体片段的乳杆菌的培养上清与RRV温育,并接种MA104细胞单层。将接种的平板于37℃温育1小时,用MEM培养基洗涤,补充含抗生素(庆大霉素和青霉素/链霉素)的新鲜MEM培养基,并在37℃,在CO2环境下培养18小时。将细胞单层固定,并用免疫过氧化物酶如(Svensson et al 1991,如上)所述进行染色。与对照孔数目相比较,RRV感染的细胞数目降低大于60%说明具有中和作用。以乳杆菌产生的纯化VHH1片段作为阳性对照。
结果如图5所示。
i)体内检测法。
所有动物实验由瑞典Huddinge医院Karolinska institutet的当地伦理委员会批准。怀孕的BALB/c雌鼠购自丹麦Mollegard。研究中使用4天龄的幼鼠。对幼鼠每天喂养一次10μl不同的处理物,从第一天直到第三天。乳杆菌在轮状病毒感染前一天给药一次。在第0天用10μl体积的2×107pfu的RRV口服进行感染。
每天记录腹泻的发生,直到第四天。在第五天对幼鼠进行腹腔内注射戊巴比妥使其安乐死。将小肠切片在(QIAGEN)中固定进行RNA提取,或在中性的缓冲福尔马林中固定进行组织病理学分析,或悬浮在无菌PBS中进行乳杆菌存活率分析。用各种lactobodies进行四个独立的实验,最初检测了产VHH1锚定的VHH1片段的细菌的剂量应答行为,然后检测了在合适剂量的其它lactobodies。在每个实验中包括表达无关抗体片段的对照lactobodies或未中和的乳杆菌。在每个实验中也包括只进行感染的组。
为了估计乳杆菌在鼠肠道中的存活率,将幼鼠在第一天喂食一次表达锚定的VHH1的乳杆菌,其中一半在第0天用RRV感染。在第1、3、7、14天对每组的2只幼鼠进行安乐死,并除去形成切片。通过将肠道提取物培养在含红霉素(3μg/ml)的Rogosa平板上来测定转化子的存在。用PCR检测VHH1的插入。
结果如图6所示。
j)肠道样品的分析。
在第四天获得小肠切片,并用4%中性缓冲的福尔马林灌注,在切片后根据标准程序进行苏木伊红染色。每个玻片进行轮状病毒感染的典型迹象的盲测。
从小肠只分离总细胞RNA,并在用无RNase的消化后进行实时分析。用EZ RT-core试剂盒(PE AppliedBiosystems,Foster City,CA,USA)进行实时PCR。用含克隆在NcoI和XhoI限制性位点的RRV vp7基因的pet28a(+)载体绘制标准曲线。在58℃,存在600nM引物、300nM探针、5mM Mn的条件下扩增轮状病毒vp7mRNA或病毒的基因组RNA(ABI 7000cycler,Applied Biosystems),得到一个长121bp的扩增产物。正向引物为(VP7f:5’-CCAAGGGAAAAT GTAGCAGTAATTC-3’)核苷酸(nt)791-815),反向引物为(VP7r:5’-TGCCACCATTCTTT CCAATTAA-3’)(nt 891-912),探针为(5’-6FAM-TAACGGCTGATCCAACCACAGCACC-TAMR-3’(nt 843-867),根据恒河猴轮状病毒vp7基因序列(登录号AF295303设计这些引物和探针。PCR检测的最低水平是10个病毒RNA拷贝。对来自每个动物的RNA样品以内部的持家基因对照GAPDH进行归一化(Overbergh et al,(1999)Cytokine 11:305-312)。无病毒和少于10个病毒的检测表示感染被清除。
结果如图7所述。
k)粪便中VHH1片段在乳杆菌表面的原位表达。
在每天结束时收集来自获得表达VHH1锚定片段的乳杆菌的组、无转化的乳杆菌的组和未处理组的3只动物的粪便样品。将样品在Super frost的包被玻片上涂片,并用甲醇∶丙酮(1∶1)在冰上固定10分钟。加入鼠抗E标记抗体(1/200),然后向玻片上加入cy2标记的猴抗鼠抗体(1/200),在湿润条件下温育1小时。通过荧光显微镜检测表面表达的VHH1片段。
统计
根据粪便的一致性估计幼鼠中腹泻情况。水样腹泻的分值为2,松散的粪便分值为1,无粪便或正常粪便分值为0。每天计算腹泻分值的百分比。在处理组的总的分值与未处理组进行Fisher′s精确检验。烈度定义为在研究过程中每组的腹泻分值的总和,持续时间定义为腹泻的总天数。烈度和持续时间通过Kruskal Wallis和Dunn检验进行分析。

图4a所示的是2A10-scFv和VHH1由乳杆菌进行表面表达,以抗E标记抗体进行的流式细胞计数表示。与表达锚定的VHH1和无关scFv及VHH对照片段的细菌相比较,在产2A10锚定片段的乳杆菌中可观察到更低水平的E标记的检测(数据未显示)。
通过ELISA和电子显微技术分析抗体片段的结合活性。对于ELISA,用E标记检测2A10-锚定和VHH1-锚定的转化乳杆菌的匀浆和来自分泌2A10和分泌VHH1的转化乳杆菌的上清。从乳杆菌表达的VHH1-锚定、VHH1-分泌和2A10-锚定的抗体片段,结合到包被轮状病毒的平板上。对锚定型和分泌型美洲驼VHH1片段(纯化或来自上清)可观察到更高水平的结合作用。分泌型2A10的量太低,不能用ELISA检测。未转化的类干酪乳杆菌、来自表达锚定型或分泌型scFv及锚定型或分泌型VHH的转化乳杆菌的无关抗体片段不与轮状表达结合(数据未显示)。由锚定型VHH1转化子产生的抗体片段的量计算为约104VHH1片段/细菌,及6002A10片段/细菌(胞内和表面)。VHH1分泌型转化子在上清中产生约1μg/ml VHH1片段。
图4b和11a所示的是其表面表达VHH1抗体片段的乳杆菌,将其与轮状病毒温育,然后通过SEM进行分析。结果表明病毒在细菌表面结合(图4ba和11b),但未转化的类干酪乳杆菌菌株没有结合(图11b)。通过TEM(负染色),可验证来自用分泌型VHH1载体转化的乳杆菌上清的美洲驼抗体片段与病毒的结合,其中未转化的类干酪乳杆菌菌株对照上清不与病毒结合(数据未显示)。
产抗体片段的乳杆菌在轮状病毒中和检测法中的作用如图5进行分析。该图的实线表示产E标记纯化的VHH1抗体(20μg/ml)的乳杆菌达到的中和水平。点线表示2A10单克隆杂交瘤上清(147ng/ml)的中和水平。通过不同浓度的锚定型VHH1乳杆菌(■),2A10锚定的乳杆菌和未转化乳杆菌(□)获得的中和作用。图5所示的是存在表达表面结合的VHH1的乳杆菌或存在含分泌型VHH1的上清时,剂量依赖型感染的显著降低。也观察到来自未转化乳杆菌的上清的轻微的中和能力。即使含150ng/ml抗体的2A10单克隆杂交瘤细胞的上清具有95%的保护性,2A10转化的乳杆菌(分泌型和锚定型)也没有保护作用。
图6所示的是用表达VHH1锚定片段的乳杆菌处理的鼠腹泻的传播。与未转化乳杆菌的组相比较,表面VHH1表达的乳杆菌在第二天显著地降低腹泻的传播(P=0.0172)。
图7所示的是在未处理组中,十二指肠和空肠切片的组织化学揭示了轮状病毒感染的典型迹象;绒毛顶端扩大、液泡化、绒毛基部收缩及细胞中的细胞核不极化(a)。获得类干酪乳杆菌的组(b)或病毒锚定型VHH1的乳杆菌(c)及未感染组(d)表现出相当归一化的组织化学特性。
图8所示的是锚定型VHH1处理组的平均病毒载量至少比未处理组低200倍。也观察到无关乳杆菌对照的益生菌作用(病毒载量降低10倍)。病毒的清除定义为未检测到vp7或少于10个vp7RNA分子。与未处理组的9%相比较,锚定型VHH1处理组中有27%动物清除了病毒。
图9所示的是,在第二天,与未处理组相比较,剂量为108和109CFU表达锚定型VHH1片段的乳杆菌对腹泻传播有显著降低,P<0.0001及P=0.0024。每组的幼鼠数目为:在107和108CFU/剂量每组有7只,在109和未处理组各有8只。
图10所示的是用表达锚定型VHH1片段的乳杆菌与RRV温育而产生感染的组也包括在内。在该组中处理按常规进行。与未处理组相比较,在第二天,冻干形式的表面表达VHH1的乳杆菌可显著降低腹泻的传播(P=0.0317)。在第三天,与未处理组相比较(n=10),在获得预温育的表达锚定型VHH1的乳杆菌的组(n=7;P=0.0004)、冻干的表达锚定型VHH1的乳杆菌的组(n=7;P=0.0072)、表达锚定型VHH1的乳杆菌的组(n=10;P=0.0022)中腹泻的传播显著降低。与未处理组相比较,当给药冻干形式的表面表达VHH1的乳杆菌(n=10;P<0.01)时,或当用RRV与新鲜的细菌培养物预温育2小时而进行感染(P<0.05)时,疾病的烈度也降低了,
在粪便中于乳杆菌表面原位表达VHH1片段(实施例4(k)的结果)
在第四天结束,收集用表达锚定型VHH1片段的乳杆菌处理的组、未处理组和阴性对照鼠的3只动物的粪便样品,测定锚定型VHH1片段的原位表达,可通过荧光抗E标记抗体在处理的鼠中检测乳杆菌表达的VHH1。在对照组中没有观察到染色(数据未显示)。
小鼠肠道中乳杆菌的存活
对幼鼠喂食表达抗轮状病毒的锚定型VHH1乳杆菌一次(在第0天),其中一半在第一天用RRV感染。在各自的第二天,将每组中的两只幼鼠杀死,通过培养肠道物质来检测乳杆菌转录子的存在。处理48小时后,可在十二指肠和回肠检测到细菌,而且轮状病毒感染组和未感染组没有差异,而在处理后96小时,没有检测到转化子(数据未显示)。
乳杆菌转化子在轮状病毒感染模型中降低腹泻的效率。
在轮状病毒诱导的腹泻的幼鼠模型中检测了转化的乳杆菌的治疗效果。幼鼠在5天内(第一天到第三天)口服表达2A10-scFv或分泌型或锚定型的VHH1的乳杆菌,并在第0天用RRV感染。对照组包括未转化的类干酪乳杆菌和表达无关的锚定型VHH抗体片段的细菌。108CFU作为腹泻干涉的合适剂量(图9)。与用未转化类干酪乳杆菌处理的鼠相比较,表面表达VHH1的细菌缩短了约0.9天(P<0.01)和0.7天(P<0.05)的疾病持续时间(通常持续1.2天)。在用表面表达VHH1的乳杆菌处理的鼠中,与未处理的幼鼠相比较,腹泻的烈度也从3.7降低到1.7(P<0.001),与用非转化的类干酪乳杆菌处理的幼鼠相比较,降低了1.2倍(P<0.01)。也观察到非转化乳杆菌轻微的益生菌作用(表2)。另外,在第二天和第三天,用表面表达VHH1的乳杆菌处理的鼠与未处理的鼠相比较(对这两天,P<0.0001)或与非转化类干酪乳杆菌处理的鼠相比较(对第二天,P<0.02),腹泻的传播显著降低(图6a)。表达分泌型或锚定型2A10和分泌型VHH1的构建体并不比未转化的乳杆菌组诱导出显著更高的保护作用(图6a,b)。当给药冻干形式的表面表达VHH的乳杆菌(P<0.01)或当通过RRV与细菌的新鲜培养预温育2小时而产生感染时(P<0.05),与未处理组相比较,疾病的烈度也降低了(表2和图5)。在第四天,近端小肠切片的组织学检验表明,在用表面表达VHH1的乳杆菌处理的轮状病毒感染的动物中,病理学改变显著降低,而且在某些鼠中,组织学是完全归一化的。在没有乳杆菌处理的组中的组织学表明具有典型的轮状表达感染迹象,绒毛顶端扩大、绒毛基部收缩、液泡化及细胞核不规则排列(图7a、b、c、d)。为了估计是否在肠细胞中病毒的复制降低了,进行实时PCR检测轮状病毒vp7基因的表达。在获得表面表达VHH1抗体片段的乳杆菌处理的动物中的平均病毒载量比未处理的鼠低至少250倍。表达无关的VHH片段的乳杆菌也降低了病毒vp7的载量(达10倍)。在获得表面表达锚定型VHH1的乳杆菌的组中的清除速率是27%,未处理组为9%(图8)。
这些实验表明在干酪乳杆菌393pLZ15表面和成功表达了作为分泌蛋白的抗轮状病毒的美洲驼来源的VHH抗体片段(VHH1)和scFv(scFv-2A10)。也揭示了这些重组的乳杆菌在幼鼠感染模型中通过体外中和来治疗轮状病毒的效果。
实施例5:
抗轮状病毒的VHH的藻酸钙胶囊化
将2%藻酸钠溶液逐滴加到含1%VHH的0.1M氯化钙溶液中,结果会形成藻酸钙小珠(体积约为2mm)。将分散的液体静置30分钟,使藻酸钙小珠沉到烧杯底部。然后用滤网收集小珠,并用水洗涤一次。
实施例6:
含胶囊化抗轮状病毒VHH或产VHH的乳杆菌的冰淇淋的组成和制备。
制备含以下配方的冰激凌成分:
总可溶性固体:占重量的35%
冰含量在-18℃:占重量的54%
所有冰淇淋的成分用高速混合仪混合3分钟。在80℃加入水。冰水混合物的温度在混合后约为55-65℃。
然后将混合物匀浆化(2000psi),并在81℃通过板式热交换器25秒,使其进行巴斯德消毒。然后将混合物在板式热交换器中冷却至4℃备用。
选择性地,可加入产抗轮状病毒的VHH的乳杆菌菌株来代替胶囊化VHH溶液,优选地是每份的浓度为109或更高。
然后将冰淇淋预混合物通过Technohoy MF 75刮板式热交换器进行冷冻,在冰淇淋中不引入膨胀率。将冰淇淋在4.4℃到-5.4℃下进行挤压成形。然后将产物放在-35℃的急速冷冻箱中进行硬化,然后储存在-25℃。
含以下成分的冰水溶液如下制备:
总可溶性固体:占重量的25.5%
冰含量在-18℃:占重量的62%
所有冰水成分用高速混合仪混合3分钟。在80℃加入水。冰水混合物的温度在混合后约为55-65℃。
然后将混合物匀浆化(2000psi),并在81℃通过板式热交换器25秒,使其进行巴斯德消毒。然后将混合物在板式热交换器中冷却至4℃备用。
此时可加入产抗轮状病毒的VHH的乳杆菌菌株来代替胶囊化VHH溶液,优选地是每份的浓度为109或更高。
然后将冰淇淋溶液通过具有30%膨胀率(空气的体积百分比)的Technohoy MF 75刮板式热交换器进行冷冻。将冰淇淋在-3.8℃到-4.5℃下进行挤压成形。然后将产物放在-35℃的急速冷冻箱中进行硬化,然后储存在-25℃。
实施例7:
含胶囊化抗轮状病毒VHH或产VHH的乳杆菌的涂抹食品的成分。
根据本领域已知的标准程序用表3中的成分制备涂抹食品。

在最后加热步骤之后,可无菌加入产抗轮状病毒的VHH的乳杆菌来代替胶囊化VHH溶液,优选地是每份的浓度为109或更高。
实施例8:
含胶囊化抗轮状病毒VHH的调味品的成分和制备。
为了制备调味品,将细菌菌株培养在合适的食品生料(例如牛奶)中,以获得高密度培养,并充分产生VHH。将培养的生料与调味品的典型成分(例如油、醋、蛋黄、盐)于可保持VHH作用的条件下混合,并进行蛋黄酱的标准制备过程,例如混合和匀浆化。选择性地,首先将调味品的典型成分例如油、醋、蛋黄、盐进行混合,然后进行加热处理,然后加入培养的生料进行匀浆化。可根据实施例5加入胶囊化抗轮状病毒的VHH来代替产VHH的微生物。
Claims (21)
1.VHH型重链免疫球蛋白或其片段,其具有与SEQ ID NO:1至少85%同一的氨基酸序列并对轮状病毒颗粒或抗原有亲和性。
2.权利要求1的重链免疫球蛋白或其片段,其中所述的重链免疫球蛋白或其片段具有与SEQ ID NO:1至少90%同一的氨基酸序列。
3.权利要求2的重链免疫球蛋白,其中所述的重链免疫球蛋白具有SEQ ID NO:1的氨基酸序列。
4.VHH型重链免疫球蛋白或其片段,其具有选自SEQ IDNO:2,3,4,12,14,17,19或20的氨基酸序列。
5.权利要求1-4中任一项的重链免疫球蛋白或其片段,其中所述的重链免疫球蛋白或其片段与轮状病毒具有解离常数至少10exp5的结合亲和力。
6.权利要求1-4中任一项的重链免疫球蛋白或其片段,其中所述的轮状病毒选自轮状病毒株Wa、CK5、Wa1、RRV或CK5。
7.重链免疫球蛋白,其结合与权利要求3或4的重链免疫球蛋白相同的表位。
8.多聚体分子,其包含权利要求1-7中任一项的VHH型重链免疫球蛋白或其片段,其中VHH序列可被融合而产生具有2个或以上VHH单位的多聚体单位,选择性地通过间隔分子连接。
9.多聚体分子,其包含根据权利要求1-7中任一项的2个或以上的VHH序列,所述VHH序列可被融合而产生具有2、3、4、5或多个VHH单位,选择性地通过间隔分子连接。
10.权利要求8或9的多聚体分子,其中,所述的VHH具有不同的特异性。
11.权利要求8-10中任一项的多聚体分子,其中,每一VHH具有对选自轮状病毒株Wa、CK5、Wa1、RRV或CK5中任一种的亲和力。
12.权利要求11的多聚体分子,其中,所述的VHH结合成单独的单体单位或载体上的组合单位。
13.权利要求1-7中任一项的重链免疫球蛋白或其片段在制备用于治疗或预防轮状病毒感染的药物中的用途。
14.编码权利要求1-7中任一项的VHH型重链免疫球蛋白或其片段的基因。
15.表达载体,其包含权利要求14所述的编码VHH型重链免疫球蛋白或其片段的基因。
16.权利要求15的表达载体,其中,所述的表达载体包含组成型启动子或适于GIT特异表达的启动子,优选pldh。
17.宿主,其包含权利要求15或16任一项所述的表达载体。
18.权利要求17的宿主,其中所述的宿主为乳杆菌(Lactobacillus)。
19.多聚体免疫球蛋白,其包含权利要求1-7中任一项所述的重链免疫球蛋白或其片段。
20.权利要求19的多聚体免疫球蛋白,其中所述的重链免疫球蛋白或其片段包含来自相同亲本抗体或其片段的抗原结合位点。
21.权利要求19的多聚体免疫球蛋白,其中所述的重链免疫球蛋白或其片段包含来自结合相同表位的不同抗体的抗原结合位点。
