Abstract

技术领域
本发明涉及一种生产神经干细胞的方法。
背景技术
神经干细胞在治疗神经障碍例如脊髓损伤的移植疗法中可用作供体细胞,并且预期其可应用于再生医学中(Okano H,Ogawa Y,Nakamura M,kaneko S,Iwanami A,Toyama Y.(2003)."Transplantation of neural stemcells into the spinal cord after injury".Seminars in Cell&DevelopmentalBiology14(3):191-198)。虽然已知神经干细胞可从胚胎干细胞(ES细胞)生产(Okada Y,Matsumoto A,Shimazaki T,Enoki R,Koizumi A,Ishii S,Itoyama Y,Sobue G,Okano H.Stem Cells.2008Dec;26(12):3086-98.Epub2008Aug28.),但是人ES细胞的使用从伦理的观点看仍在争论中。

发明内容
技术问题
然而,建立iPS细胞将花费相当长的时间,例如对小鼠将花费约1个月,对人将花费3-4个月。因此,从成纤维细胞经iPS细胞生产神经干细胞要花费较长时间,例如对于人,将花费半年以上。因此,考虑到在再生医学中的应用,希望开发更容易且更快速地产生神经干细胞的方法。
因此,本发明的目标是提供直接从体细胞而不建立iPS细胞的更容易且更快速地生产神经干细胞的新方法。
技术方案
通过对从成纤维细胞生产神经干细胞进行大量尝试,本申请的发明人发现:通过以下方式可以使成纤维细胞分化成为神经球(neurosphere)形式的神经干细胞:将4种去分化因子(Oct3/4、Sox2、Klf4和c-Myc)导入成纤维细胞并使它们生长数日,然后在存在生长因子的情况下漂浮培养所述成纤维细胞。因此已经实现了本发明。
应注意,当在本文中单独使用所述因子(例如“Sox2”、“Oct3/4”、“Klf4”和“c-Myc”)的名称而无诸如术语“基因”或“cDNA”时,它们是指作为来源自这些基因的基因产物的蛋白。
在本发明的一个实施方案中,所述生产神经干细胞的方法包括以下步骤:
(i)将一种或多种去分化因子导入体细胞;并且
(ii)在存在生长因子的情况下悬浮培养所述导入去分化因子的体细胞,以产生神经球。在将所述一种或多种去分化因子导入所述体细胞后,优选将所述细胞培养2-14天。可以将所述去分化因子(例如Sox2、Oct3/4和Klf4,还可包括c-Myc)导入所述体细胞中。可以将所述导入去分化因子的体细胞在存在生长因子(例如LIF和FGF)的情况下培养。此外,可以将所述导入去分化因子的体细胞在存在cAMP的情况下培养,并且优选在无NAC(N-乙酰基-L-半胱氨酸)的情况下培养。所述体细胞优选来源自哺乳动物,特别是来源自小鼠或人,优选来源自皮肤或肝,并且更优选为成纤维细胞或肝细胞。

==将去分化因子导入体细胞==
为了从体细胞生产神经干细胞,首先将去分化因子导入所述体细胞。
本文所用的“体细胞”是指组成动物身体的分化细胞,生殖系中的细胞(例如卵细胞、精子细胞、卵原细胞、精原细胞和它们的前体细胞)和来源自早期发育阶段的胚胎的全能未分化细胞(例如胚胎干细胞)除外。体细胞优选来源自哺乳动物,并且可以是任何动物物种例如小鼠或人。所述动物可以是成体、胎儿或胚胎,不包括人胎儿和人胚胎。体细胞可以来源自已建立的细胞系或从组织分离的原代培养的细胞;优选地,它的染色体数等处于正常状态。对作为体细胞来源的器官或组织没有特别的限制,包括皮肤、肝脏和血液。对体细胞的特征没有特别的限制,只要它已经失去受精细胞所具有的全能性的至少一部分,体细胞包括成纤维细胞、上皮细胞、肝细胞和血细胞。当用本发明的方法生产的神经干细胞用于治疗患者时,优选使用已从所述患者自身分离的体细胞。
虽然对将要导入体细胞的去分化因子没有特别的限制,但是它们可以是生产所述iPS细胞时使用的重编程因子。所述去分化因子优选包括选自以下基因家族每个成员的基因的基因产物结合物:Oct基因家族、Klf基因家族和Sox基因家族。考虑到生产神经球的效率,更优选的是还包含来自myc基因家族的基因的基因产物的结合物。属于Oct基因家族的基因包括Oct3/4、Oct1A和Oct6;属于Klf基因家族的基因包括Klf1、Klf2、Klf4和Klf5;属于Sox基因家族的基因包括Sox1、Sox2、Sox3、Sox7、Sox15、Sox17和Sox18。属于myc基因家族的基因包括c-myc、N-myc和L-myc。myc基因家族的基因产物可以被细胞因子(例如SCF和bFGF)或者化合物(例如阿扎胞苷(azacitidine)和丙戊酸钠(VPA))代替。
除了来自Oct基因家族的基因和来自Sox基因家族的基因之外,非前述结合物的去分化因子还包括包含Nanog基因和lin-28基因等的结合物。当将所述因子导入细胞中时,除了上述结合物中的基因之外,还可以将另一类型的基因产物导入。所述另外的基因产物的实例包括无限增殖化诱导因子例如TERT。应注意,如果一种或多种前述的去分化因子已在将要使用的体细胞中表达,那么可以不必导入一种或多种这类去分化因子。同时,如果存在能够代替一种具体的去分化因子功能的化合物,那么该化合物可用于代替所述去分化因子。所述化合物包括反苯环丙胺、CHIR99021、SB431542、PD0325901、thiazovivin,但不限于此。
由于编码上述去分化因子的所有基因在脊椎动物中都高度保守,除非指出具体的动物名称,否则本文中提及的每种所述基因均包括其同源物。而且,还涵盖包括多形基因在内的突变基因,只要其产物具有与野生型基因产物相当的功能。
将这些去分化因子导入所述体细胞的方法没有特别限制;例如,可以导入所述去分化因子的蛋白质本身(蛋白质转导方法),或者可以导入编码所述去分化因子的DNA以在体细胞中表达(基因转移技术)。
用于将所述去分化因子的蛋白导入所述体细胞的方法没有特别限制,可以应用本领域技术人员已知的任何方法。例如,可以将市售产品例如SAINT-Ph和Cellvader或者阳离子脂质用于导入,并且通过将被称为蛋白转导结构域(PTD)的肽与所述去分化因子的蛋白融合并将它们加入培养基中,可以将所述去分化因子的蛋白导入细胞中。
同时,如果基因转导方法将要被应用于将所述去分化因子导入所述体细胞,那么可以通过本领域技术人员已知的任何方法首先构建这样的重组表达载体,即其中编码所述去分化因子的DNA被插入至用于在所述体细胞中表达的合适启动子的下游。可以将两种或多种类型的所述去分化因子插入至单个载体中。对将要使用的表达载体没有特别的限制,实例包括pMX反转录病毒载体等。然后,将如上述构建的重组表达载体导入所述体细胞。用于导入的方法可以是本领域技术人员已知的任一方法,例如电穿孔法、磷酸钙法、脂转染法和利用反转录病毒感染的方法。因此,通过将能够表达所述去分化因子的表达载体导入所述体细胞并使所述去分化因子在所述体细胞中表达,可以将所述去分化因子导入所述体细胞。
将因此得到的所述导入去分化因子的体细胞在与培养成纤维细胞的条件相同的正常条件下培养以下时间:2至14天,优选3至10天,最优选对于小鼠3至6天,或者对于人7至10天。例如,可以在35至40℃,优选37℃下,在存在5%CO2的情况下使用含10%FBS的DMEM培养基培养所述细胞。
==诱导导入去分化因子的体细胞分化成神经球==
接下来,通过在存在生长因子的情况下漂浮培养如上处理的体细胞,可以诱导它们分化成为神经球形式的神经干细胞。将要使用的培养基优选是无血清培养基例如补加葡萄糖、谷氨酰胺、胰岛素、转铁蛋白、孕酮、腐胺和氯化硒的DMEM:Ham F-12培养基(F-12)。对将要加入培养基的生长因子没有特别限制,条件是它是能够诱导体细胞分化成神经球的因子或因子结合物,优选FGF、LIF、B27或它们的结合物。优选的FGF是FGF-2或FGF-8,FGF在培养基中的浓度可以是5至50ng/ml,优选10至40ng/ml。LIF在培养基中的浓度优选是约1000U/ml。依照来自Invitrogen的说明书,B27可以以大约50倍稀释使用。培养优选在以下条件下进行:5%CO2、在35至40℃,更优选37℃。通过将大约100uM的cAMP加入培养基,可以显著改善诱导分化成为神经球的效率。这优选在不存在N-乙酰基-L-半胱氨酸(NAC)的情况下进行。用于漂浮培养的培养皿没有特别限制,但优选用于细菌培养的未包被的塑料培养皿。
通过在这些条件下继续培养所述导入去分化因子的体细胞并不时更换培养基,原代神经球开始形成,对于小鼠成纤维细胞和人成纤维细胞通常在约7天内。可以适当地更换培养基,例如每14天一次。培养的时间没有特别的限制,条件是所述神经球即准备好被收集,小鼠成纤维细胞和人成纤维细胞通常在约20天内即准备好被收集。应注意,通过按照如上所述诱导所述体细胞分化而最初得到的神经球被称为原代神经球(PNS)。
可以将如此得到的原代神经球解离(dissociate)并培养,以使得可在相同条件下再次形成次代神经球。该再次形成的神经球以及通过重复神经球解离-神经球形成过程形成的神经球统称为次代神经球(SNS)。
因此,通过依照本发明的方法,可以在短时间内从体细胞形成大量神经球。
==诱导从神经球分化成神经元细胞==
如此得到的原代神经球和次代神经球具有如神经干细胞的功能。例如,通过在普通的分化培养基中培养,它们不仅可以分化成为神经元,而且可以分化成为神经胶质细胞(例如星形胶质细胞、少突胶质细胞和施万氏细胞(Schwann cell))。在该情形下,用于诱导分化的优选培养基是补加葡萄糖、谷氨酰胺、胰岛素、转铁蛋白、孕酮、腐胺和氯化硒的DMEM:Ham F-12培养基(即省略FGF和肝素的用于增殖神经干细胞的培养基)。音猬因子蛋白(Sonic hedgehog protein)可以存在或不存在。培养优选在5%CO2、35至40℃下条件下进行5至7天。
在另一方面,在其中首先使用ES细胞形成胚状体(EB)并随后将EB诱导分化成神经干细胞的情况下,如果在正常分化条件下培养次代神经球,那么不仅分化成神经元而且分化成神经胶质细胞;而如果在相同分化条件下培养原代神经球,那么仅诱导分化成神经元,包括运动神经元和GABA能神经元(日本专利公开文本No.2002-291469)。
因此,通过如下本发明的方法,不仅可以在短时间内从所述体细胞分化神经干细胞,而且可以从原代神经球的阶段分化神经胶质细胞。
实施例
实施例1
该实施例显示,本发明的生产神经干细胞的方法可以诱导从成纤维细胞分化成为神经干细胞。
==小鼠原代成纤维细胞的制备==
为了建立来源自小鼠皮肤的成纤维细胞,在颈脱位后去除成年小鼠(8周,ICR小鼠,雄性)的毛并收集腹部皮肤。将所分离的皮肤在含青霉素/链霉素(50U,50mg/ml)和0.6%谷氨酰胺的磷酸盐缓冲盐水(PBS)中洗涤4次,并从所述分离的皮肤取下真皮。将所述真皮置于细胞培养皿中,以盖玻片覆盖,并在原代细胞起始培养基(TOYOBO)中培养。
在培养开始后的第6-7天,将贴附于所述塑料皿和盖玻片的成纤维细胞通过以0.25%胰蛋白酶-EDTA洗涤脱附,以50000细胞/ml接种于新塑料皿上,并在37℃和5%CO2的条件下在原代细胞起始培养基中培养10至14天。
==人原代成纤维细胞的制备==
在37℃和5%CO2的条件下,使用含10%FBS的DMEM,通过与用于小鼠的相同方法从收集自人皮肤组织的真皮得到成纤维细胞。将从所述组织块培养的成纤维细胞通过胰蛋白酶处理接种,并作为原代成纤维细胞培养。然后,为了增加反转录病毒的感染效率,通过以表达Slc7a1的慢病毒感染所述原代成纤维细胞(Takahashi and Yamanaka,Cell2006vol.126p.663-676)过夜并使它们生长,来诱导它们表达Slc7a1——一种反转录病毒的受体,将所得到的成纤维细胞用于如下的实验中。
==将去分化因子导入已建立的成纤维细胞==
以下文的方式,将包含各个去分化因子DNA中的每一个的重组表达载体pMXs-Oct3/4、pMXs-Sox2、pMXs-Klf4和pMXs-c-Myc(都来自Addgen)用于制备包含表达4种因子的组合(Oct3/4、Sox2、Klf4和c-Myc)、3种因子的组合(Oct3/4、Sox2和Klf4)或3种因子的组合(Oct3/4、Sox2和c-Myc)的反转录病毒的培养基,而将pMX-GFP(Cell BiolabsInc.)用于制备含GFP的培养基,作为转化的指示物和针对因子插入组的阴性对照组。用于制备载体的方法详细记载于Takahashi andYamanaka,Cell2006vol.126p.663-676中,该文献以引用的方式纳入本文。
在37℃下,将Platinum-E反转录病毒包装细胞在含10%FBS的DMEM中培养14天,然后包装所述重组表达载体。将所述重组表达载体的各个混合物转染至所述Platinum-E细胞中,并且在24小时后更换培养基。在5%CO2和35至40℃的条件下,将所述细胞进一步在含10%FBS的DMEM中培养24小时,然后回收培养上清液。依赖于用于包装的重组表达载体,所回收的培养上清液的每一种包含如下之一:表达4种因子(Oct3/4、Sox2、Klf4和c-Myc)中每一种的反转录病毒混合物、表达除c-Myc之外的3种因子(Oct3/4、Sox2和Klf4)中每一种的反转录病毒混合物、表达除Klf4之外的3种因子(Oct3/4、Sox2和c-Myc)中每一种的反转录病毒混合物或者表达GFP的反转录病毒。
将如此得到的培养上清液以合适的量加入原代成纤维细胞的培养基中,目的是以所述反转录病毒感染它们,以使得在所述小鼠原代成纤维细胞中表达所述4种因子、所述3种因子或GFP,在所述人原代成纤维细胞中表达所述4种因子或所述GFP。
将这些反转录病毒转染的成纤维细胞接种于塑料培养皿上,并在5%CO2和35至40℃的条件下在含10%FBS的DMEM中培养24小时至5天。
==诱导分化成神经球==
将如上述培养的所述导入去分化因子的成纤维细胞通过以0.25%胰蛋白酶EDTA洗涤解离,以50细胞/ul的密度再悬浮于补加LIF(重组人LIF;Chemi-Con,100U/ml)和FGF(PeproTech,20ng/ml)的10ml无血清DMEM/F-12培养基(1:1,Invitrogen;含0.6%D-葡萄糖、5mMHEPES、3mM NaHCO3、2mM谷氨酰胺、25ug/ml胰岛素、100ug/ml转铁蛋白、20nM孕酮、60uM腐胺和30nM硒酸钠)中,接种于细菌塑料培养皿中,并在37℃和5%CO2的条件下悬浮培养,每14天更换培养基。在悬浮培养的第3天和第20天,观察细胞的状态。
图1示出在第3天(A、C、E)和第20天(B、D、F),从已导入所述4种去分化因子(Oct3/4、Sox2、Klf4、c-Myc)(A、B)、已导入所述除c-Myc之外的3种去分化因子(C、D)或者已导入GFP(E、F)的小鼠细胞形成神经球,所述神经球为所述导入去分化因子的成纤维细胞的细胞簇形式。如图1中所示,不论所导入的去分化因子是所述除c-Myc之外的3种因子或者是所有所述4种因子,在悬浮培养所述成纤维细胞后都可以得到神经球。
图1还示出在悬浮培养所述导入去分化因子的成纤维细胞期间的第3天,从已导入所有所述4种去分化因子(Oct3/4、Sox2、Klf4和c-Myc)(G)或者已导入GFP(H)的人细胞形成神经球。如图1中所示,甚至在使用人细胞时,在悬浮培养所述成纤维细胞后也可以得到神经球。
图2示出对在其中使用小鼠细胞的各个条件下产生的神经球数目进行计数得到的结果。已导入所述4种因子的成纤维细胞每10ml培养基产生了平均7.5个神经球,已导入所述除c-Myc之外的3种因子的成纤维细胞产生了平均1个神经球,而已导入所述除Klf4之外的3种因子或已导入GFP的成纤维细胞未产生神经球。应注意,在测量中,对于球形细胞簇,计数直径等于或大于50uM的细胞簇。
总之,根据本发明的方法,可以不经建立iPS细胞而直接从体细胞生产神经球。
==诱导神经球分化==
接下来,将证明如此得到的神经球具有作为神经干细胞的潜能。
将在悬浮培养第14天或之后形成的神经球(直径>75uM)逐一转移至具有Poly-O和纤连蛋白双包被的小室的载玻片上,并在37℃和5%CO2的条件下,在补加2%FBS的0.5ml无血清DMEM/F-12培养基(1:1,Invitrogen;含0.6%D-葡萄糖、5mM HEPES、3mM NaHCO3、2mM谷氨酰胺、25ug/ml胰岛素、100ug/ml转铁蛋白、20nM孕酮、60uM腐胺和30nM硒酸钠)中贴壁培养7天。将产生的分化细胞以4%的低聚甲醛固定15分钟。在以PBS洗涤后,将所述分化的细胞在4℃下以抗人βIII-微管蛋白的单克隆抗体(Cat.No.T8660,Sigma,1000倍稀释)、兔抗胶质细胞原纤维酸性蛋白(GFAP)多克隆抗体(Cat.No.Z0334,DAKO,400倍稀释)或抗-O4(少突胶质细胞)的单克隆抗体(Cat.No.MAB345,Chemicon,1000倍稀释)处理过夜。这里,βIII-微管蛋白被用作神经元的标记物,GFAP被用作星形胶质细胞的标记物,O4被用作少突胶质细胞的标记物。然后,将Alexa488标记的抗小鼠IgG山羊抗体(Invitrogen,500倍稀释)、Alexa350标记的抗兔IgG山羊抗体(Invitrogen,500倍稀释)或Alexa555标记的抗小鼠IgM山羊抗体(Invitrogen,1000倍稀释)适当地用作第二抗体,并且在荧光显微镜下观察标本。
如图3A中所示,贴壁培养的细胞分化成为神经元、星形胶质细胞或少突胶质细胞,因此确认了所得到的神经球具有分化成为神经元细胞以及神经胶质细胞的潜能。
接下来,在直接来源自成纤维细胞的神经球和来源自iPS细胞(38C2)的神经球之间比较生成多个分化的细胞类型的潜能。如上所述,使直接来源自成纤维细胞的神经球分化。在另一方面,在存在10-8M视黄酸的情况下,使38C2细胞(Okita et al.,Nature vol.448,pp.313-317,2007)形成胚状体(EB),然后在补加20ng/ml FGF-2(Okada et al.,Dev.Biol.vol.275,pp.124-142,2004)的无血清培养基中培养。EB在所述培养基中在7天内形成原代神经球,这使它们以与那些直接来源自成纤维细胞的神经球相同的方式分化。在显微镜下观察所述分化细胞,并对所述分化细胞计数,同时分辨其细胞类型。图3B示出了所述分化的神经元细胞和神经胶质细胞中的每一种的百分比。如图3B中所示,在所有来源自iPS细胞的原代神经球(11个神经球)中,仅分化出神经元细胞,而在65%的直接来源自成纤维细胞的神经球(26个神经球中的17个神经球)中,分化出神经元细胞以及神经胶质细胞。
因此,通过本发明的方法得到的原代神经球具有分化成为神经胶质细胞的潜能,因此它们本身可作为移植源使用。
应注意,通过本发明的方法还能够从收集自小鼠胚胎的皮肤的成纤维细胞以几乎相同的效率生产神经球。
实施例2
该实施例显示,在实施例1中描述的生产神经干细胞的方法中,通过在存在cAMP的情况下培养所述导入去分化因子的成纤维细胞,可以显著改善诱导分化成神经球的效率。
为了检查细胞死亡抑制剂N-乙酰基-L-半胱氨酸(NAC)和8-氯苯基硫(8-chlorophenylthio,pCPT-cAMP)——一种cAMP的可透过膜的类似物——对诱导分化成神经球的效率的改善,将补加1mM的NAC(Sigma)和100uM的pCPT-cAMP(Sigma)中的一种或两种的培养基用于实施例1中描述的悬浮培养中。接下来的其他步骤以与实施例1中所述相同的方式进行,对培养第20天的神经球数目计数。
如图4A中所示,当仅加入pCPT-cAMP时,与两种细胞死亡抑制剂都不加入的情况(对照组)相比,每10ml培养基的神经球数目增加超过100倍。在另一方面,当仅加入NAC时,与所述对照组相比未观察到差异。在NAC和pCPT-cAMP两者都加入的组中,与仅加入pCPT-cAMP的情况相比所得到的神经球数目显著降低。因此,NAC抑制了pCPT-cAMP改善诱导分化的效率的作用。
因此,当通过培养所述导入去分化因子的成纤维细胞来诱导分化成神经球时,如果存在cAMP则将显著提高所述诱导分化的效率。优选在不存在NAC的情况下进行诱导。
应注意,当使用从小鼠胚胎皮肤得到的成纤维细胞时,通过使用本发明的方法可产生类似的cAMP效应。
对于人成纤维细胞的情况,培养基补充有替代LIF的cAMP或者补充有cAMP和LIF,或者补充有cAMP、LIF和ROCK抑制剂Y27632,对从250,000个细胞得到的神经球数目计数。如图4B中所示,LIF和cAMP对神经球形成有加性作用,而Y27632无作用。
实施例3
在该实施例中,使用携带Oct4-GFP转基因的转基因小鼠的胚胎和成体生成神经球,并基于在未分化细胞中表达的Oct4基因的表达检查它们的分化阶段。
依照实施例1的方法制备成体小鼠的成纤维细胞。如下所述制备小鼠胚胎成纤维细胞:
然后,依照实施例1的方法,使用4种因子(Oct3/4、Sox2、Klf4和c-Myc)以这些成纤维细胞生成神经球。在显微镜下对包含GFP阳性细胞的神经球(GFP阳性神经球)数目和包含非GFP阳性细胞的神经球(GFP阴性神经球)数目计数。GFP阳性神经球和GFP阴性神经球的比显示于图5A中。
在来源自胚胎成纤维细胞和来源自成体成纤维细胞的神经球中,分别有85%(100个神经球中的85个)和6%(100个神经球中的6个)的神经球为GFP阳性神经球,表明来源自胚胎的神经球的分化程度比来源自成体的神经球的分化程度更低。从移植后肿瘤发生的角度来看,处于高分化状态的神经球危险更小;因此,来源自成体的神经球优于来源自胚胎的神经球。
类似地,检查了来源自iPS的神经球中GFP阳性神经球的百分数,并且与来源自成体的神经球进行了比较。如图5B中所示,97%的来自iPS的神经球为GFP阳性神经球,表明来自iPS的神经球未分化程度更高。该事实说明,从移植后肿瘤发生的角度来看,依照本发明生产的神经球优于来源自iPS的神经球。
Claims (10)
1.一种生产神经干细胞的方法,包括以下步骤:
(i)将基因Oct3/4、Klf4和Sox2的基因产物结合物作为去分化因子导入体细胞;
(ii)将所述导入去分化因子的体细胞培养2至14天;并且
(iii)在存在LIF和FGF的情况下漂浮培养在所述步骤(ii)中培养的所述导入去分化因子的体细胞,以产生神经球。
2.权利要求1的生产神经干细胞的方法,其中
在所述步骤(ii)中,将所述导入去分化因子的体细胞培养3至10天。
3.权利要求1的生产神经干细胞的方法,其中
在所述步骤(ii)中,将所述导入去分化因子的体细胞培养3至6天。
4.权利要求1的生产神经干细胞的方法,其中
在所述步骤(ii)中,将所述导入去分化因子的体细胞培养7至10天。
5.一种生产神经干细胞的方法,包括以下步骤:
(i)将基因Oct3/4、Klf4和Sox2的基因产物结合物作为去分化因子导入体细胞;
(ii)将所述导入去分化因子的体细胞培养24小时至5天;并且
(iii)在存在LIF和FGF的情况下漂浮培养在所述步骤(ii)中培养的所述导入去分化因子的体细胞,以产生神经球。
6.权利要求1或5的生产神经干细胞的方法,其中
在所述步骤(i)中,还将c-Myc导入所述体细胞。
7.权利要求1或5的生产神经干细胞的方法,其中
在所述步骤(iii)中,在存在cAMP的情况下培养所述导入去分化因子的体细胞。
8.权利要求7的生产神经干细胞的方法,其中
在所述步骤(iii)中,在不存在NAC的情况下培养所述导入去分化因子的体细胞。
9.权利要求1或5的生产神经干细胞的方法,其中
所述体细胞来源自小鼠或人。
10.权利要求1或5的生产神经干细胞的方法,其中
所述体细胞为成纤维细胞或肝细胞。
