Abstract

Description
发明背景
本发明涉及一种生产人造装置的方法,该人造装置具有生物膜和膜蛋白质的性能和功能,以及涉及这样的装置的结构。
生物膜蛋白质具有大的多种功能,包括充当泵、通道、阀门、能量转化器、和机械、热和电子传感器等等。因为这些蛋白质是纳米尺寸的和高效的,因此它们对于用于人造装置中来说是非常吸引人的。但是,它们的天然脂质膜环境具有缺点例如低强度,需要含水环境,和易于化学或者细菌降解。
发明内容

随着工艺革新产生的装置小型化使其更小、更轻和更有效,但是用于这些装置的功率源的进步并未进行的这样快。二十一世纪的功率源面临着这样的挑战,即,为越来越多数目的装置提供能量,同时降低尺寸和重量。另外,纳米工艺和生物工艺明日的产品将需要这样的电源,其甚至不类似于今天所用的电源的形式或者功能。
这里迫切的需要一种更轻和更紧凑的电功率源来用于广泛的多种新兴应用。这样的功率源将能够具有比现代的电池工艺能够实现的更大范围的游戏目标,使得功率密度最大,进行给定功率需求所需的重量最小。重量需求是至关重要的,因为对于常规燃料源来说,该燃料源必须接近于所述装置,并且如果是可移动的时,还要随着该装置传输。还可以发生燃料耗尽,因此必需更换供给。这会对用户的范围和移动性产生限制。

仅仅最近开发第一台纳米生物工艺装置所必需的工艺和知识才变得能够实现,并且已经报到了由生物化学燃料ATP提供能量的纳米尺寸有机/无机混杂装置的工程化和构造(Soong,R.K.,Bachand,G.D.,Neves,H.P.,Olkhovets,A.G.,Craighead,H.G.,和Montemagno,C.D.(2000),Science 290,第1555-1558页)。用于这些装置的ATP的产生以及这些装置为其他机器提供功率的用途使得人们开始关注宏观和纳米尺寸之间的功率转移以及不同类型能量的互相转化。

本发明的方法涉及制造一种薄织物,其组成为囊泡-线结合体形成的生物模拟膜和薄膜。在一种实施方案中,该囊泡是一种生物相容的聚合物膜,植入有两种能量转化蛋白质,细菌视紫红质和细胞色素氧化酶,其将光能量转化为电能,并且将这种能量传递给外部负荷。这些蛋白质已经被分离,并且通过数百万年的自然选择而优化来将光能和电能转化成电化学能。在加入到装置中时,它们提供了有用的不确定的功率量,并且提供了足够的轻,紧凑性和耐用性,用于在不利的和有利的环境二者应用中所需的高灵活性。

本发明涉及一种图1所示的囊泡-线结合体(1),其可以形成生物模拟膜或者薄膜(8)。在一种优选的实施方案中,薄膜(8)是通过将该囊泡-线结合体(1)编织到织物中而形成。在一种可选择的实施方案中,薄膜(8)是通过将该囊泡-线结合体(1)沉积成“造纸”布置而形成的,在其中该结合体相互粘附的。图1表示了一种囊泡或者聚合物(2),其具有植入到该囊泡膜中的任何种类的蛋白质(7)。囊泡(1)可以包含ABA三嵌段共聚物(3),该共聚物具有将它们连接到线(5)上的交联官能团(4)。同样还表示的是薄膜或者膜(8),其是通过多个这样的囊泡-线结合体(1)形成的。该结合体是通过提供功能化囊泡表面和功能化的线表面而形成的,如图2所示。
该囊泡优选是一种脂化聚合物或者三嵌段共聚物,如US专利7208089所述。图3表示了本发明的一种实施方案,在这里将该囊泡表面功能化来产生胺端接的PEtOz-PDMS-PEtOz囊泡,参见Joon-Sik Park等人Macromolecules 2004,37,6786-6792,其的内容在此明确引入作为参考。该囊泡表面可以使用其他已知的技术来功能化。图4表示了甲基丙烯酸酯端接的PEtOz-PDMS-PEtOz囊泡和NHS端接的PEtOz-PDMS-PEtOz囊泡。所选择的功能性可以根据囊泡-线结合体中所用的线的类型而变化。此外,聚合物的类型和功能化会取决于加入到囊泡中的蛋白质的类型。
该线可以选自多种可利用的材料,包括但不限于纤维素材料,羧甲基纤维素(CMC),氨基乙基纤维素(AE-纤维素)和基于尼龙的材料。该优选的纤维素材料是亲水性的,并且不溶于水和大部分有机溶剂。如图5所示,该纤维素材料上的多个羟基与另一个链上的氧分子形成氢键,将该链并排一起稳定保持,并且形成高拉伸强度的微纤。结晶纤维素在25 Mpa的压力下将在水中变成无定形的。纤维素的羟基可以部分的或者完全的与不同的反应物反应,来产生具有有用性能的衍生物。纤维素酯和纤维素醚是最重要的市售材料,例如醋酸纤维素,乙基纤维素,甲基纤维素,羟丙基纤维素,羧甲基纤维素,羟丙基甲基纤维素和羟乙基甲基纤维素,仅仅提到了少数几个。如图6所示,另外一种线材料可以是市售的羧甲基纤维素(CMC)。CMC是一种纤维素衍生物,其具有与羟基键合的羧甲基(-CH2-COOH)。极性(有机酸)羧基赋予了纤维素可溶性和化学反应性。低取代度(DS=0.2)的部分羧甲基化的纤维素保持了它的纤维特性,同时它的许多性能不同于初始纤维的这些性能。平均链长和取代度是非常重要的;高疏水性、低取代的CMC是触变性(摇溶)的,但是更大延伸的高取代的CMC是假塑性的。在低pH时,CMC能够通过在羧酸和游离羟基之间的内酯化而形成交联。图8表示了由氨基乙基纤维素(AE-纤维素)形成的线。AE-纤维素可以在氢氧化钠的存在下,通过纤维素与2-氨基乙基硫酸的反应来形成。AE-纤维素市售自Whatman,并且以前已经用于色谱柱和过滤器。
图7表示了用优选的线来结合优选的囊泡。在这种实施方案中,将羧甲基纤维素(CMC)线在二环己基碳二酰亚胺(DCCI)的存在下,与胺功能化的囊泡反应。可选择的,AE-纤维素将与卤化物例如三氯甲基嘌呤或者苯磺酰氯反应。它还将在碳二酰亚胺例如二环己基碳二酰亚胺的存在下与蛋白质和有机酸反应。

在本发明的一种形式中,将细菌视紫红质和细胞色素氧化酶整合到囊泡中,该囊泡进一步与线结合。将该囊泡-线结合体编织到织物中,该织物与微制作电极接触。在理解了细菌视紫红质,细胞色素氧化酶,和它们在脂质和聚合物膜中的整合之后,就能够最好的理解所提出的装置的运行。对全部这三种都进行了深入研究,并且已经有多个涉及它们的合成和功能的文献。关于能量-转化蛋白质和它们在脂质和聚合物膜中的加入的进一步的细节,参见US专利7208089。
因为离子在膜表面上的扩散是大的,并且可以通过合适的选择囊泡而变得更大,因此该囊泡表面本身到了这样的程度,即,需要对生物太阳能电池进行连续的功能化(Pitard等人,1996)。囊泡例如脂化聚合物或者任何一种诸多的生物相容性聚合物基质,包含蛋白质,并且充当了质子阻挡层。这些聚合物基质是非常通用的,优选仅仅需要:(a)它们形成了囊泡,当使用跨膜蛋白质,其将蛋白质分离成顶部和底部二等分,(b)它们形成了充分类似于天然脂质膜的环境,以使得蛋白质能够容易的插入到正确定向的囊泡中,和(c)蛋白质所经历的囊泡的局部化学环境不会导致蛋白质解开或者变形,以这样的方式来包含蛋白质的天然功能。满足这些条件的囊泡包括但不限于脂化聚合物和具有亲水性外嵌段和疏水性内嵌段的一般性能的三嵌段共聚物。混有蛋白质的聚合物囊泡优选是US专利7208089和国际专利申请PCT/US08/74163中所述的这些。BR和COX在囊泡表面中定向和合并,并且由该囊泡-线结合体所形成的膜覆盖着电极。
这里有许多的方案可以用来提高电极与蛋白质的亲合性,例如在US专利7208089中所提供的这些方案。电极栅可以直接放在脂质的顶上,其处于外部连接用于电测量的细丝网的形式。在除去了上表面上的液体之后,铝或者镍的薄透明层可以直接喷涂到该膜上来形成反电极。可选择的,该电极可以通过触点阵列的栅格化而电化学沉积到脂质表面上。这种沉积将在膜上表面上产生数百万的纳米级细丝。重复和合并上述步骤,产生包含在脂质膜中的定向的COX和BR。
对于BR和COX的定向来说,这里有两种可能的情况:平行和反平行的偶极定向。如果该偶极是平行的,则对二者而言,所述排列可以通过同时施加单场而实现。如果它们是反平行的,则使用PM的大的聚集偶极矩。正确的定向将如下来实现:通过COX在高场中的初始定向,随后通过PM在这样的场中的定向,该场足够小来避免COX的扰动,但是足够大来控制PM的碎片。
在囊泡形成中使用聚合物膜因为下面的原因而是令人期望的:它们具有比脂质膜更长的寿命,它们更结实,和它们具有更容易调节的性能,例如电子和离子传导性和透过性。这些膜的内部必须是疏水性的和弹性的,以使得能够尽可能接近的模拟天然蛋白质环境。
存在着广泛的多种生物相容的聚合物,其具有宽范围的性能例如吸光率,极性,电和离子传导性等等。增强本发明的太阳能电池的性能的聚合物必须是与蛋白质和电极可相容的。质子的不可透过性也是重要的。涂覆聚合物表面的能力会是重要的,因为它会在质子传导性和跨膜电导方面起到主要作用。该聚合物的寿命以及它对于包含在它中的蛋白质寿命的影响也是相关的,并且是它选择中的因素。选择短寿命但是高性能的聚合物在特定的应用中会是有用的。
前述的用于由生物成分来生产高效和能够生产的太阳能功率源的方法证实了能量转化生物蛋白质与外部装置的整合,并且指出了制造能够大规模生产的生物太阳能电池(其能够为广泛的多种装置供能)的途径。
在本发明的另一方面,通过使用加入到三嵌段共聚物膜中的水通道蛋白族的蛋白质,能够生产稳定的膜,该膜仅仅通过水,因此其通过渗析促进了水净化,脱盐和分子浓缩。该水通道蛋白阻止了全部污染物的通过,包括细菌,病毒,矿物质,蛋白质,DNA,盐,清洁剂,溶解的气体,和甚至来自水溶液的质子,但是该水通道蛋白分子因为它们的结构而能够传输水。关于水通道蛋白族的蛋白质进一步的细节公开在US专利7208089中。水在特定方向上移动穿过该膜,这归因于水压或者渗透压。水净化/脱盐作用可以用两个腔室的装置来实现,该装置具有被加入到聚合物膜中的硬蛋白质在它的中心隔开的腔室,该腔室填充有水通道蛋白。这种膜本身是水不能透过的,并且将污染的水分离到第一腔室中,净化的水处于第二腔室中。仅仅纯净的水能够在两个腔室之间流动。因此,当膜一侧上的海水或者其他污染的水置于适当的压力下时,纯净的水自然流向另一腔室。因此,净化的水可以获自不可饮用的来源,或者如果该水源包含了感兴趣的化学品,则可以选择性的除去水,在输入腔室中留下高浓度的该想要的化学品。但是,重要的是该水通道蛋白海因为除了它们专门的水选择性之外的其他原因而适于本发明。这个蛋白质族的许多成员例如水通道蛋白Z(AqpZ)是极其结实的,并且能够经受着污染来源的水的苛刻条件,而不丧失功能。AqpZ抗由于曝露于酸、电压、清洁剂和热而引起的变性或者分解。所以,该装置能够用于净化被这样的材料污染的水源,该材料会淤塞或者破坏另外一种膜,并且它可以用于经历了持续高温的这样的区域。
AqpZ还是易变的。因为这种蛋白质根据一种遗传顺序而明确的以主细菌表达(其影响它的最终形状和功能),因此本领域技术人员能够容易的改变它的遗传代码来改变该蛋白质的特性。所以该蛋白质可以工程化来完成期望的应用,该应用可以不同于蛋白质的初始功能。例如,通过将接近于水通道中心的具体的氨基酸残留物简单的改变成半胱氨酸,所产生的水通道蛋白将结合该溶液中任何自由的汞,并且由于堵塞而停止传输水。因此,当有毒物质的浓度升高过高时,膜装置中所用的这些变异蛋白质将通过简单的停止流动来检测水样品中的汞污染。
本发明优选的形式具有常规过滤盘的形式,因为它最容易化验功能性的方式。为了制作这样的盘,使用 Langmuir-Blodgett槽将5nm厚的单层合成的三嵌段共聚物和蛋白质沉积到25mm市售超滤盘表面上。该盘上的单层然后曝露于254nm的UV光来交联该聚合物和提高它的耐久性。最后,将220nm孔径的PVDF膜用环氧胶合到该盘表面上,来确保安全操作和防止端部裂纹。
该装置是如下来化验的:将它安装到腔室中,其驱动加压的源水穿过膜。该装置在下列情况中被认为是起作用的:当仅仅纯水穿过到膜的另一侧,并且污染的溶质浓缩保留在初始的腔室中。污染的溶液必须加压,来克服纯水流入具有更高数目的溶解粒子的腔室中的自然倾向。水通道蛋白Z膜的目的是反渗透和将纯水与污染溶质分离。系统的这种倾向或者渗透压力可以以磅/平方英寸(psi)来表示。例如,海水的渗透压大约是360psi。
这里有几种能够用来使得所述装置承受这些类型压力的方法。聚合物的一些变体自然的比其他变体具有更大的耐久性,并且可以用UV光交联来提供额外的刚度。另外一种方法是将高浓度的无毒和易于除去的溶质加入到新水腔中,来保持跨膜的正常的渗透,同时还因为腔室加压而发生反渗透。最后,反渗透所需的压力可以通过在串联的密封的、连接的含有逐渐降低的浓度的污染物的腔室中,使用多个AqpZ装置来降低。在每对腔室中净化水所需的形成压力是反渗透所需总压力的一部分。所以,每个膜仅仅必须承受小的压力,并且具有剩余物完整性更大的变化。这样,如果每对腔室之间的浓度差仅仅是10%而非100%,则在每个接点处净化源水所需的压力仅仅是上述高压的10%。纯水将在最后的腔室中恒压恒流连续生产。
该水通道蛋白反渗透膜能够在仅仅一个步骤中净化具有几种不同类型的污染的水。传统高纯度净化系统需要几种部件,所述部分可以包括水软化器,碳过滤器,离子交换器,UV或者化学杀菌,和在水(其没有水通道蛋白净化的水那样干净)能够生产之前配合使用的两通路反渗透过滤器组。这种精制的装置不能如同水通道蛋白膜那样,从源水中除去小于150道尔顿的溶解的气体或者物质。此外,全部这些部件需要维护。UV灯需要更换和提供能量。离子交换器在它们饱和时需要化学再生。软化器需要盐。碳和反渗透筒当它们结垢时必须更换。最后,单步装置将需要远小于典型的净化系统的较少的空间和重量,并且这种优点使得本发明的水通道蛋白水净化装置是便携的。
水通道蛋白膜还快于常规的系统。常规高速反渗透装置每分钟能够制造大约28.4L(7.5加仑)的净水。最新的研究表明水分子以54 μmol/s的速率跨AqpZ饱和的脂质膜(0.0177mm.sup.2)移动。(Pohl,P.,Saparov,S.M.,Borgnia,M.J.,和Agre,P.,(2001),Proceedings of the National Academy of Sciences 98,第9624-9629页),因此,表面积1.0平方米的理论水通道蛋白Z反渗透膜每分钟能够过滤高到3295L的纯水。该速率比常规的净化器快了116倍。
最后,新的蛋白质基膜的生产也是非常廉价的。该方法的核心,AqpZ容易以毫克量从工程化大肠埃希菌菌株来获得。平均2.5mg的纯蛋白质可以由生产它的每升培养液来获得。10mg的蛋白质可以由大约5美元的生长介质来生产。这对于几种实际大小的装置来说是足够的蛋白质。同样,其中植入了AqpZ的聚合物可以在同样的实验室中,以对于每个装置来说仅仅几便士价值的化学品来生产。该水通道蛋白Z反渗透膜是一种新的、有效的和廉价的水净化装置。
因此,这里已经公开了这样的方法和设备,其利用生物部件来实现由结垢的、盐化的或者污染的水来高效生产完全纯净的水。本发明证实了水传输生物蛋白质与外部装置的整合,并且指出了制造能够大规模生产的水净化装置的途径。
虽然已经描述了本发明的优选的实施方案,但是应当理解可以对此处公开的方法和装置进行众多的改变和改进,而不脱离在下面的权利要求中所提出的本发明的实际主旨和范围。
Claims (12)
1.一种囊泡-线结合体,其中所述囊泡-线结合体是一种聚合物囊泡,其中该囊泡被结合到线上,来形成囊泡-线结合体,
其中该结合体是通过提供功能化的囊泡表面和功能化的线表面来形成的,
其中将蛋白质加入到该囊泡的膜中,
其中该囊泡是由具有亲水性外嵌段和疏水性内嵌段的三嵌段共聚物形成的,
其中该线包括纤维素材料或基于尼龙的材料。
2.根据权利要求1的囊泡-线结合体,其中所述蛋白质是水通道蛋白。
3.根据权利要求1的囊泡-线结合体,其中将大于一种的蛋白质加入到该囊泡的膜中。
4.根据权利要求3的囊泡-线结合体,其中将在协同作用时由光来产生电的两种不同蛋白质加入到该囊泡的膜中。
5.根据权利要求4的囊泡-线结合体,其中所述两种蛋白质为细菌视紫红质和细胞色素氧化酶。
6.根据权利要求1的囊泡-线结合体,其中该线包括纤维素、纤维素酯或纤维素醚。
7.根据权利要求1的囊泡-线结合体,其中该线包括羧甲基纤维素、氨基乙基纤维素、纳米-结晶纤维素、或纤维素纳米纤维。
8.一种生物模拟膜,其包括多个根据权利要求1-7中任一项的囊泡-线结合体。
9.根据权利要求8的生物模拟膜,是通过将多个根据权利要求1-7中任一项的囊泡-线结合体的线编织成织物而形成的,和/或
其是通过将根据权利要求1-7中任一项的囊泡-线结合体沉积成“造纸”布置而形成的,其中该结合体是彼此粘附的。
10.根据权利要求8或9的生物模拟膜,其是水过滤膜。
11.根据权利要求8或9的生物模拟膜,其是用光来产生电的膜。
12.一种功率源,其包含根据权利要求11的生物模拟膜,其中将该生物模拟膜与微制作电极接触。
