Abstract

本发明公开了一种注射级土霉素生产工艺,将口服级土霉素加入到甲醇和纯化水的混合溶剂中,再加入试剂盐酸使之溶解;然后依次加入黄血盐和硫酸锌,搅拌、过滤;加入氨水调节pH值在4.5~5.0之间出结晶;结晶液用离心机分离得湿产品,湿产品经过纯化水洗涤真空干燥得注射级土霉素。本发明的生产工艺简单,成本低廉;注射级土霉素甲醇中溶解度合格,杂质含量低吸收度高。

注射级土霉素生产工艺
技术领域
本发明属于医药制造技术领域,具体的说是一种注射级土霉素生产工艺。
背景技术
土霉素是一种广谱抗生素,现主要以兽用为主,随着畜牧业的快速发展,土霉素的给药方式也在发生变化,注射级土霉素的需求增多。
现有技术是发酵法生产土霉素,在发酵法生产土霉素的后工序中,通过优化工艺条件来提高产品质量;但所得产品存在甲醇中溶解度不合格、杂质多、吸收低等问题。
发明内容
本发明的目的在于克服现有技术的缺点,提供一种甲醇中溶解度合格,杂质含量低吸收度高的注射级土霉素生产工艺。
本发明的目的是这样实现的:一种注射级土霉素生产工艺,包括如下工艺步骤:
(1)将口服级土霉素加入到甲醇和纯化水的混合溶剂中,再加入试剂盐酸在40~45℃的温度下使之溶解;然后依次加入用少量纯化水溶解的黄血盐和硫酸锌,搅拌30分钟后经0.2um微孔滤膜过滤;
(2)将上述滤液降温至15~20℃,加入10%氨水调节PH值在4.5~5.0之间出结晶;结晶液静置30分钟后用离心机分离,滤饼用纯化水洗涤至出口水PH值为7;继续用离心机分离2小时后出湿产品;
(3)湿产品通过真空干燥机干燥,温度80~90℃,真空度0.08MPa,干燥4~6小时,得注射级土霉素;
其中,上述工艺步骤所用的原料配方比例为:土霉素0.8~1.2kg、甲醇48~56L、纯化水6~10L、试剂盐酸0.2~0.4L、黄血盐0.012~0.020kg、硫酸锌0.010~0.018kg。
作为本发明更进一步优化的技术方案:所用的原料配方比例为:土霉素1kg、甲醇52L、纯化水8L、试剂盐酸0.3L、黄血盐0.016kg、硫酸锌0.014kg。
为了提高步骤(1)的过滤效果,在进行0.2um微孔滤膜过滤之前还要进行一次尼龙滤布过滤。
所述的真空干燥机采用双锥回转真空干燥机。
本发明的优点在于:本发明的生产工艺简单,成本低廉;注射级土霉素甲醇中溶解度合格,杂质含量低。
具体实施方式
实施例1:(1)将50kg口服级土霉素加入到2600L甲醇和400L纯化水的混合溶剂中,再加入15L试剂盐酸在40℃的温度下使之溶解;然后依次加入用少量纯化水溶解的0.8kg黄血盐和0.7kg硫酸锌,搅拌30分钟后经8222尼龙滤布过滤后,再经0.2um微孔滤膜过滤;(2)将上述滤液降温至18℃,加入10%氨水调节PH值在4.5出结晶;结晶液静置30分钟后用离心机分离,滤饼用纯化水洗涤至出口水PH值为7;继续用离心机分离2小时后出湿产品;(3)湿产品通过真空干燥机干燥,温度80~90℃,真空度0.08MPa,干燥5小时,得注射级土霉素。
实施例2:(1)将42kg口服级土霉素加入到2500L甲醇和380L纯化水的混合溶剂中,再加入16L试剂盐酸在45℃的温度下使之溶解;然后依次加入用少量纯化水溶解的0.7kg黄血盐和0.6kg硫酸锌,搅拌30分钟后经8222尼龙滤布过滤后,再经0.2um微孔滤膜过滤;(2)将上述滤液降温至16℃,加入10%氨水调节PH值在4.5出结晶;结晶液静置30分钟后用离心机分离,滤饼用纯化水洗涤至出口水PH值为7;继续用离心机分离2小时后出湿产品;(3)湿产品通过真空干燥机干燥,温度80~90℃,真空度0.08MPa,干燥5小时,得注射级土霉素。
实施例3:(1)将56kg口服级土霉素加入到2700L甲醇和420L纯化水的混合溶剂中,再加入18L试剂盐酸在42℃的温度下使之溶解;然后依次加入用少量纯化水溶解的0.9kg黄血盐和0.8kg硫酸锌,搅拌30分钟后经8222尼龙滤布过滤后,再经0.2um微孔滤膜过滤;(2)将上述滤液降温至15℃,加入10%氨水调节PH值在4.5出结晶;结晶液静置30分钟后用离心机分离,滤饼用纯化水洗涤至出口水PH值为7;继续用离心机分离2小时后出湿产品;(3)湿产品通过真空干燥机干燥,温度80~90℃,真空度0.08MPa,干燥5小时,得注射级土霉素。

Claims (4)

1.一种注射级土霉素生产工艺,包括如下工艺步骤:
(1)将口服级土霉素加入到甲醇和纯化水的混合溶剂中,再加入试剂盐酸在40~45℃的温度下使之溶解;然后依次加入用少量纯化水溶解的黄血盐和硫酸锌,搅拌30分钟后经0.2um微孔滤膜过滤;
(2)将上述滤液降温至15~20℃,加入10%氨水调节PH值在4.5~5.0之间出结晶;结晶液静置30分钟后用离心机分离,滤饼用纯化水洗涤至出口水PH值为7;继续用离心机分离2小时后出湿产品;
(3)湿产品通过真空干燥机干燥,温度80~90℃,真空度0.08MPa,干燥4~6小时,得注射级土霉素;
其中,上述工艺步骤所用的原料配方比例为:土霉素0.8~1.2kg、甲醇48~56L、纯化水6~10L、试剂盐酸0.2~0.4L、黄血盐0.012~0.020kg、硫酸锌0.010~0.018kg。
2.按照权利要求1所述的注射级土霉素生产工艺,其特征在于:所用的原料配方比例为:土霉素1kg、甲醇52L、纯化水8L、试剂盐酸0.3L、黄血盐0.016kg、硫酸锌0.014kg。
3.按照权利要求1或2所述的注射级土霉素生产工艺,其特征在于:在进行0.2um微孔滤膜过滤之前还要进行一次尼龙滤布过滤。
4.按照权利要求1或2所述的注射级土霉素生产工艺,其特征在于:所述的真空干燥机采用双锥回转真空干燥机。
