Abstract
本发明涉及LK8-Fc融合蛋白,其能增加抑制血管生成活性和体内稳定性。更具体地说,本发明涉及其中具有抑制血管生成活性的LK8蛋白与人免疫球蛋白IgG1的Fc区域融合的LK8-Fc融合蛋白,以及用于治疗癌症的组合物,所述组合物含有该融合蛋白。LK8-Fc融合蛋白不仅具有起到抗肿瘤的抑制血管生成活性和抑制转移活性,而且在体内的半衰期非常长,因此可用作更有效和经济的肿瘤治疗剂或者肿瘤抑制剂。
Description
技术领域
本发明涉及LK8-Fc融合蛋白,其能增加抑制血管生成(angiogenesis)活性和体内稳定性,更具体地说,本发明涉及其中具有抑制血管生成活性的LK8蛋白与人免疫球蛋白IgG1的Fc区域融合的LK8-Fc融合蛋白,以及用于治疗癌症的组合物,所述组合物含有该融合蛋白。
背景技术
血管生成指的是通过其由已有血管形成新血管的过程。已经知道在正常生理条件下,血管上皮细胞保持在难以增殖的状态,并且血管生成仅仅在极其有限的情况下发生,这包括女性月经周期。在控制血管生成机制方面的失调可引起许多病理性疾病,包括癌症,糖尿病、视网膜病、风湿性关节炎、银屑病等。在肿瘤的情况下,众所周知的是虽然在没有血管的帮助下肿瘤可生长到数mm3的体积,但血管生成对于肿瘤来说是必须的(N.Eng.J.Med.,333:1757,1995;Folkman,J.,New Engl.J.Med.,285:1182,1971)。
为了开始肿瘤血管生成,应当满足两个条件,即刺激血管生成因子的增加和抑制血管生成因子的减少。内源性抑制血管生成因子的典型例子是血管生长抑制因子(angiostatin)。已经报道血管生长抑制因子是与血液凝集相关的酶血纤维蛋白溶酶原的一部分,其由kringle结构组成,并具有在体外和体内条件下抑制血管生成的能力(O′Reilly,M.S.等人,Cell,79:315,1994)。kringle结构是由大约80个氨基酸和三个分子间二硫键组成的蛋白质结构域并组成独立的折叠单元。kringle结构在许多蛋白质中被发现,诸如凝血素、尿激酶、肝细胞生长因子和载脂蛋白(a)。特别是,已经报道各种kringle,诸如凝血素kringle和尿激酶kringle结构显示了抑制血管生成的能力(Lee,T.H.等人,J.Biol.Chem.,273:28805,1998;Kim等人,J.Biol.Chem.,278:11449,2003)。
糖蛋白载脂蛋白(a)与作为低密度脂蛋白(LDL)的主要蛋白成分的apo B-100共价结合形成脂蛋白(a)(Fless,G.M.,J.Biol.Chem.,261:8712,1986)。已知脂蛋白(a)参与胆固醇在体内输送,并且血浆中脂蛋白(a)的浓度增加与动脉粥样硬化以及心脏疾病有关(A rmstrong,V.W.等人,Artherosclerosis,62:249,1986;Assmann,G.,Am.J.Cardiol.,77:1179,1996)。载脂蛋白(a)包括显示与血纤维蛋白溶酶原IV和V以及失活蛋白酶状结构域同源的两种类型的kringle结构域。根据氨基酸序列同源性,载脂蛋白(a)kringleIV-结构域被分成10个亚型(IV1-IV10),并且除在各种人类载脂蛋白(a)基因的等位基因中具有3-42个拷贝数以外的IV2kringle以外,它们中的每个都仅仅具有一个拷贝。最后kringleV具有与血纤维蛋白溶酶原kringleV 83.5%的同源的氨基酸序列。
本发明的发明人观察到构成载脂蛋白(a)的一部分kringle(kringleKV38,此后称为“LK8蛋白”)具有在体外和体内条件下抑制血管生成的能力,这起到抗肿瘤和抑制肿瘤细胞转移作用(WO 2001/019868,题为“Angiogenesis inhibitor comprising LK6,LK7,LK8and LK68”;WO2004/073730,题为“Anticancer agent containing LK8”)。但是,发现LK8蛋白在猴子体内仅仅具有7-11小时的半衰期,因此其具有的问题在于需要以较短的间隔重复给药以便显示抗肿瘤效力。而且,血管生成抑制因子常常抑制细胞生长而不是对细胞有毒性,并且必须连续给药很长一段时间才会显示其抗肿瘤效果(Jain,R.K.等人,Nat.Clin.Pract.Oncol.,3:24,2006)。
因此,对于长期连续给药来说,需要大量的LK8重组蛋白的生产和所导致的生产成本的增加,并且高治疗成本和长期治疗时间给患者造成很大的负担。出于这种原因,在技术上难以使用LK8蛋白开发抗肿瘤剂。
同时,为了增加抗原性、纯化的容易性并增加在血液中的半衰期等,已经进行了免疫球蛋白或其片断与活性蛋白的融合蛋白的制备。这些例子包括:作为蛋白质药物的融合蛋白的白介素受体,其具有免疫球蛋白片断本身的功能和具有免疫球蛋白Fc的有用蛋白质功能的两种功能(韩国专利249572);通过将INF-α与Fc融合以便增加INF-α的血液半衰期得到的融合蛋白等等。然而,虽然INF-α和Fc的融合蛋白具有非常加长的半衰期,但其缺点在于INF-α的活性被降低(US 5,723,125)。
因此,本发明的发明人付出了许多努力来发现当将LK8蛋白质体内给药时能够使LK8保持长半衰期的同时其抑制血管生成效果不被降低的方法。出于这种目的,本发明的发明人通过将IgG1的Fc区域与LK8蛋白的C-末端融合构建了LK8-Fc融合蛋白并检查了其效果。结果,本发明的发明人发现,该融合蛋白显示了完全出乎意料的效果,因为与LK8蛋白的半衰期相比,由于Fc融合伴侣没有影响LK8蛋白本身的效果,该融合蛋白的半寿期增加了40-50倍,从而完成了本发明。
发明内容
本发明的目的在于,提供使用基因融合技术通过将人免疫球蛋白IgG1的Fc区域与LK8蛋白的C-末端融合得到的具有增加的生物可利用性的LK8-Fc融合蛋白,所述LK8蛋白的C-末端为具有抗肿瘤和抑制肿瘤细胞转移效果的人载脂蛋白(a)的kringle片断。
本发明的另一目的在于提供用于治疗癌症的组合物,所述组合物含有LK8-Fc融合蛋白。
本发明的另一目的在于提供用于抑制血管生成的组合物,所述组合物含有LK8-Fc融合蛋白。
为了实现上述目的,在一个方面,本发明提供了LK8-Fc融合蛋白,其中将LK8蛋白与人免疫球蛋白IgG1的Fc区域融合。在本发明中,LK8-Fc融合蛋白优选包括另外的Igκ前导序列以便进行融合蛋白的胞外分泌。
在另一方面,本发明提供了编码所述LK8-Fc融合蛋白的基因,含有所述基因的重组载体和使用所述重组载体转染的重组细胞。在本发明中,细胞优选动物细胞。动物细胞优选CHO/LK8-Fc细胞。
在又一方面,本发明提供了用于治疗癌症的组合物和用于抑制血管生成的组合物,所述组合物含有所述LK8-Fc融合蛋白。在本发明中,癌症优选自由直肠癌、胰腺癌、前列腺癌、肾癌、黑色素瘤、前列腺癌骨转移和卵巢癌组成的组中。
通过下列详细描述和所附的权利要求书,本发明的其他特征和方面将是非常明显的。

具体实施方式
在本发明中,构建了包括编码LK8-Fc融合蛋白的基因序列的重组质粒pMSG/LK8-Fc,并且建立了使用该基因转染以产生重组LK8-Fc融合蛋白的CHO/LK8-Fc细胞系。而且,使用由所建立的CHO/LK8-Fc细胞系产生的LK8-Fc融合蛋白处理小鼠,结果,可以观察到融合蛋白抑制肿瘤的生长和转移。
而且,已经发现,与LK8蛋白相比,由于Fc融合伴侣没有反过来影响LK8蛋白本身,根据本发明的LK8-Fc融合蛋白的半衰期增加大约40-50倍,因此,达到了给药蛋白质总量和给药频率降低的效果。
这个事实被认为是由所有本领域技术人员不能预测的效果,因为有用蛋白质与Fc的常规融合蛋白质显示了比亲本蛋白质低的效果,或者显示了在增加半衰期方面的不足的效果,相反本发明的LK8-Fc融合蛋白具有与现有LK8蛋白的相同效果,同时,其半衰期远长于现有LK8蛋白质的半衰期。
在本发明中,观察到LK8-Fc融合蛋白在体外条件下抑制由bFGF诱导的人内皮细胞的迁移,并具有在体内条件下抑制血管生成的作用。同时,观察到当LK8-Fc蛋白一旦被给药到SD鼠(6w,雄性,Charles River,日本)的肌肉细胞中时,与LK8蛋白的体内半衰期相比,由于Fc融合伴侣的原因,其体内半衰期增加40-50倍。这表明融合蛋白可显示高效力,甚至当给药频率和药物剂量降低时也是如此。因此,LK8-Fc融合蛋白可显示融合蛋白可显示抗肿瘤和抑制肿瘤细胞转移的效果,甚至当其以不到LK8蛋白的1/10的剂量作为有效量以最小7天一次的间隔给药时也是如此。但是,给药频率和剂量不必需如此限制,并可基于患者的年龄、性别和健康状况以及疾病的种类和严重程度来确定。
当根据本发明的LK8-Fc融合蛋白与在现有技术中已经使用的化学治疗或放射治疗结合使用时,可得到协同效应。而且,本发明的融合蛋白可与具有抑制血管生成效果的其他种类的制剂结合使用。另外,本发明的LK8-Fc融合蛋白可与具有与本发明的融合蛋白的机制不同的机制的抑制血管生成剂组合给药,在这种情况下,可实现有效抗肿瘤或者抑制转移的作用。
免疫球蛋白重链恒定区域包括由CH1-铰链区域-CH2-CH3(-CH4)组成的4或5个域。重链区的DNA序列在免疫球蛋白家族中具有交叉同源性。例如,IgG的CH2结构域与IgA和IgD的CH2结构域同源,并与IgM和IgE的CH3同源。
在本文中使用的术语“Fc区域”指的是免疫球蛋白链恒定区域,优选免疫球蛋白重链恒定区域或其部分的羧基端部分。例如,免疫球蛋白Fc区域可包括:(1)CH1域,CH2域和CH3域;(2)CH1域和CH2域;(3)CH1域和CH3域;(4)CH2域和CH3域;或者(5)两个或三个区与免疫球蛋白铰链区域的组合。在本发明的优选实施方式中,免疫球蛋白Fc区域至少包括:免疫球蛋白铰链区域、CH2域和CH3域,并缺乏CH1域。
重链恒定区域来自于其的免疫球蛋白的优选家族是IgG(Igγ)(γ亚族1,2,3或4)。其他免疫球蛋白家族IgA(Igα),IgD(Igδ),IgE(Igε)和IgM(Igμ)也可在本发明中使用。合适的免疫球蛋白重链恒定区域的选择在US5,541,087和US5,726,044中详细描述。认为本领域技术人员可从特定免疫球蛋白家族和亚族中选择特定免疫球蛋白重链恒定区域序列以便得到特定结果。编码免疫球蛋白Fc区域的DNA部分优选包括至少铰链区域的一部分和IgA,IgD,IgE或IgM中的任何一个的Fcγ的CH3域者同源区的一部分。
基于所需的用途,来自除人以外的其他物种(例如大鼠或小鼠)的恒定区域基因可被使用。在DNA构建体中被用作融合伴侣的免疫球蛋白Fc区域一般可从任何哺乳动物种类中得到。当需要在宿主细胞或动物中诱导对Fc区域的免疫应答时,Fc区域可来自与宿主细胞或动物相同的物种。例如,如果宿主细胞或动物是人,人免疫球蛋白Fc区域可被使用,如果宿主细胞活动物是大鼠,则啮齿动物免疫球蛋白Fc区域可被使用。
编码在本发明的实践中有用的人免疫球蛋白Fc区域的核酸序列和由其翻译的氨基酸序列在SEQ ID NO.2中阐明,但本发明的范围不限于此。例如,能够使用其他免疫球蛋白Fc区域。,诸如由GenBank或者EMBL数据库中现有的核苷酸序列编码的那些,例如AF045536.1(Macaca fuscicularis),AF045537.1(Macaca mulatta),AB016710(Felixcatus),K00752(Oryctolagus cuniculus),U03780(Sus scrofa),Z48947(Camelus dromedarius),X62916(Bos taurus),L07789(Mustela vision),X69797(Ovis aries),U 17166(Cricetulus migratorius),X07189(Rattusrattus),AF57619.1(Trichosurus vulpecula)或者AF035195(Monodelphisdomestica)。
而且,在免疫球蛋白重链恒定区域中氨基酸的取代或缺失可在本发明的实践中使用。例如,氨基酸取代可被引入到上部CH2域以便产生具有对Fc受体的亲和力降低的Fc突变体(Cole等人,J.Immunnol.,159:3613,1997)。任何本领域技术人员可使用公知的分子生物学技术制备这些构建体。
在本发明中,常规重组DNA技术被用于制备在本发明的实践中有用的Fc融合蛋白。Fc融合蛋白构建体优选在DNA水平上产生,并且由此产生的DNA被插入到表达载体中并表达,从而制备出本发明的融合蛋白。
在本文中使用的术语“载体”指的是任何核酸,包括作为适于被结合到宿主细胞中和待重组并整合到宿主细胞基因组中的核苷酸序列,或者作为游离基因并可自动进行复制。所述载体包括线性核酸、质粒、噬粒(phagemids)、粘粒(cosmids)、RNA载体、病毒载体以及类似物。病毒载体的例子包括逆转录病毒,腺病毒和腺病毒-相关病毒,但本发明的范围不限于此。
在本文中使用的术语“基因表达”或者“目标基因的表达”指的是DNA序列的转录、mRNA转录翻译以及Fc融合蛋白产物的分泌。
合适的宿主细胞可使用本发明的DNA序列转化或转染,并用于目标蛋白的表达和/活分泌。在本发明中使用的优选宿主细胞包括不死的杂交瘤细胞,NS/O骨髓瘤细胞,293细胞,中国仓鼠卵巢(CHO)细胞,Hela细胞和COS细胞。
可被用于在哺乳动物细胞中高水平表达产生地融合蛋白的表达系统是编码分泌框的DNA构建体,包括从5’到3’方向的信号序列、目标蛋白和免疫球蛋白Fc区域。
在本文中使用的术语“前导序列”指的是定向LK8-Fc融合蛋白的分泌,然后在宿主细胞中翻译并裂解的序列。本发明中的前导序列为编码启动蛋白质跨过的内质网膜运输的氨基酸序列的多聚核苷酸。在本发明中有用的前导序列包括抗体轻链序列,例如抗体14.18(Gillies等人,J.Immunol.Meth.,125:191,1989),Igκ前导序列,抗体重链信号序列,例如MOPC141抗体重链前导序列(Sakano等人,Nature,286:5774,1980),以及本领域已知的其他前导序列(Watson等人,Nucleic Acids Research,12:5145,1984)。
本发明提供了通过将本发明的LK8-Fc融合蛋白给药到患有所述疾病的哺乳动物治疗各种癌症、病毒性疾病、相关疾病以及由其引起的疾病的方法。相关疾病可包括通过血管生成增殖并转移的各种实体瘤,但本发明的范围不限于此。
本发明中的癌症可以是直肠癌、胰腺癌、前列腺癌、肾癌、黑色素瘤、前列腺癌骨转移和卵巢癌,但本发明的范围不限于此。
本发明的组合物可通过适于特定分子的任何途径给药。本发明的组合物可通过任何合适的方式直接(例如典型地通过注射、皮下注射或者局部给药到组织位点)或者全身(例如胃肠外或口服)提供。当组合物经胃肠外提供时,诸如通过静脉、皮下、眼部、腹膜、肌肉、口腔、直肠、阴道、眼窝、脑内、颅内、脊柱内、心室内、鞘内、脑池内、囊内、鼻内提供,或者通过气雾给药时,组合物优选包括部分水或者生理上可接受的流体悬液或者溶液。因此,载体和赋形剂是生理上可接受的以便另外将所需组合物输送给患者,其不反过来影响患者的电解和体积平衡。用作试剂的流体介质因此可包括正常的生理盐水。
根据本发明的LK8-Fc融合蛋白的计量优选在0.03-300mg/m2,更优选0.3-30mg/m2。但是,最佳计量基于待治疗的疾病和副作用的存在而变化,但可通过常规实验来确定。融合蛋白的给药可通过周期性丸剂注射或者通过从置于患者体外(例如i.v.袋)或体内(例如可生物可蚀性物质的植入物)的静脉内或者腹膜内给药。而且,本发明的融合蛋白可于多种不同生物活性分子一起给药到目标受体。但是,融合蛋白与其他分子的最佳组合、给药模式和融合蛋白的剂量可由本领域技术人员通过常规实验来确定。
根据本发明的血管生成抑制因子可被用作治疗各种与血管生成有关的病变的试剂,包括各种肿瘤和肿瘤转移、糖尿病视网膜病、风湿性关节炎、银屑病以及类似疾病。在这种情况下,根据本发明的LK8-Fc融合蛋白还可以与相关疾病有关的其他治疗剂结合使用。
实施例
此后,将参照实施例进一步详细描述本发明。本领域技术人可以理解,这些实施例仅仅出于解释的目的,而不解释为对本发明的范围的限制。
实施例1:表达LK8-Fc融合蛋白的重组载体的构建
为了构建编码LK8和Fc的融合蛋白的载体,使用以前由本发明的发明人制备的含有LK8基因的pET11B载体(WO2001/019868)作为模板通过PCR得到LK8基因(SEQ ID NO:1)。另外,编码Fc的基因(SEQID NO:2)使用pRC13-Hpa载体(韩国专利467706)作为模板通过PCR得到。在每次PCR中使用的引物在下表1中显示。
特别是,在下列条件下进行PCR反应:模板DNA在94℃变性5min,然后在进行30个循环,每个循环94℃下30sec,56℃下30sec和72℃下1min,接着在72℃下延伸5min。而且,为了易于克隆,限制酶消化位点被插入每个引物中,使得到的PCR产物具有限制酶消化位点。
通过PCR反应产生的两个基因片段被插入到含有用于促进待产生的蛋白质的细胞外分泌的Igκ前导序列的pSecTag载体(Invitrogen,USA)中。特别是,LK8基因片段和pSecTag载体使用SfiI和BamHI消化,然后LK8基因片段与pSecTag载体连接以构建pSecTag-LK8。Fc基因片段使用BamHI和XhoI消化,然后与使用BamHI和XhoI消化的pSecTag-LK8连接,从而构建pSecTag/LK8-Fc。
在质粒pSecTag/LK8-Fc中,Igκ前导序列、LK8基因和Fc基因使用限制酶消化并被插入到哺乳动物细胞表达载体pMSG(KCCM 10202;韩国专利公开10-2002-0010327)中。也就是说,pMSG载体和pSecTag-LK8-Fc质粒使用限制酶NheI和XhoI消化,然后消化的Igκ-LK8-Fc片段被插入到pMSG载体中,从而构建pMSG/LK8-Fc(图1)。

实施例2:表达大量LK8-Fc融合蛋白的动物细胞系的建立
为了建立产生LK8-Fc融合蛋白的动物细胞系,使用Dosper(Roche,Switzerland)将在实施例1中构建的pMSG/LK8-Fc与二氢叶酸还原酶DHFR(dihydrofolate reductase)基因(Columbia University,USA)一起转染到剔除了DHFR基因的细胞系CHO DG44(Columbia university,USA)中。然后,通过该细胞系,初步选择适于含10%血清的MEM-α最低培养基的菌落,并通过逐渐增加MTX(甲氨喋呤Methotrexate;ChoongWaePharma Corporation,Korea)浓度(包含50nM和1μM)对所选菌落进行次级培养。在次级培养过程中,在显示耐MTX的菌落中,第二次选择分泌大量目标蛋白的细胞系。所选的细胞系在含有无血清培养基HyQ-SFM-CHO(Promega,USA)的旋转烧瓶中培养以便有利于蛋白质的大量产生,最终选择的细胞系被命名为“CHO/LK8-Fc”。
实施例3:LK8-Fc融合蛋白的纯化
为了纯化LK8-Fc融合蛋白,将CHO/LK8-Fc细胞系在HyQ-SFM-CHO培养基中以实施例2中相同的方式旋转培养。如图2所示。细胞被培养同时在培养的第六天细胞的生长和发育能力被观察,通过离心收集上清液。然后,上清液中含有的LK8-Fc融合蛋白以下列方式纯化。在LK8-Fc融合蛋白的Fc区域具有对蛋白G琼脂糖(sepharose)(Amersham Pharmacia,USA)的亲和力的事实的基础上,进行亲和柱层析。特别是,在含有20-100mM磷酸钠的结合缓冲液(pH 6-8)中,在上清液中含有的LK8-Fc融合蛋白被连接到蛋白G琼脂糖(sepharose)柱上,然后使用甘氨酸缓冲液(pH 2-5)将其从柱上洗脱(图3)。
纯化的LK8-Fc融合蛋白最后使用PBS透析,然后其纯度通过使用4-20%的浓度梯度的凝胶电泳SDS-PAGE和免疫印迹来检查(图4)。在电泳结果中,融合蛋白的分子量在还原条件下大约为37kDa,在非还原条件下大约为75kDa。为什么在非还原条件下分子量大约是在还原条件下的两倍的原因是,由于融合蛋白在非还原条件下在LK8-Fc融合蛋白的Fc区域中存在的二硫键,在非还原条件下融合蛋白以二聚体存在(图4)。
实施例4:LK8-Fc融合蛋白抑制内皮细胞迁移的能力分析
为了分析重组蛋白LK8-Fc是否具有抑制血管生成活性,使用人脐静脉内皮细胞(HUVEC;Cambrex,USA)在体外进行受伤迁移测定(wounding migration assay)(Kim等人,J.Biol.Chem.,278:29000,2003)。具体地说,在EGM-2培养基(Cambrex,USA)中悬浮的HUVEC细胞被设置在包被了1.5%的明胶的24-孔组织培养板的每个孔中然后培养到至少90%的培养克隆率,然后将培养基被含有0.1%FBS的EBM-2(Cambrex,USA)培养基替换。在细胞在上述条件下培养大约15小时之后,使用微量吸管头将细胞刮下并将从培养板上除下的细胞使用PBS洗涤两次将它们除去。刮下部分成像并使用参考线标记。刮下的受伤HUVEC单层细胞在LK8-Fc存在或不存在下被接种bFGF,并且在下8小时后观察迁移到剥落区域中的HUVEC。然后,细胞迁移的抑制通过对迁移超过参考线的细胞数计数进行观察。上述实验重复三次,实验结果在图5和图6中显示。
在图5中,X-轴表示处理量样品的种类和浓度,Y轴表示迁移超过参考线的细胞数。图6是显示由图5的数据计算的百分比的示意图。具体地说,图5显示了使用各种不同浓度的LK8-Fc融合蛋白处理的细胞迁移的相关抑制情况,其中相关抑制情况通过从每个数据中减去仅仅使用PBS处理而没有使用样品处理的组中迁移的细胞数来确定,并计算百分比(100%=仅仅使用bFGF处理的组中的迁移细胞数)。LK8蛋白被用作阳性对照组。
当使用bFGF处理内皮细胞时,细胞的迁移极大地被诱导。如图5和图6中所示,当使用LK8蛋白处理细胞时,由bFGF诱导的迁移被抑制,并且处理的LK8蛋白质的浓度增加导致其抑制活性增加。在其中以与LK8蛋白相同的摩尔浓度使用LK8-Fc融合蛋白处理内皮细胞的情况下,HUVEC细胞的迁移被有效地抑制到与LK8蛋白相似的程度。与单独使用bFGF处理的组相比,在使用每种1μM的LK8蛋白和LK8-Fc融合蛋白处理细胞的情况下,LK8蛋白处理的组和LK8-Fc融合蛋白处理的组分别显示了大约68%(p<0.005)和大约64%(p<0.05)的内皮细胞迁移抑制活性。通过上述结果可以看出,LK8-Fc融合蛋白在体外条件下显示了与LK8类似水平的抑制内皮细胞迁移活性。
实施例5:LK8-Fc融合蛋白体内抑制血管生成的能力分析
为了检查LK8-Fc融合蛋白在体内是否抑制血管生成,观察了LK8-Fc融合蛋白对鸡胚胎绒毛膜尿囊膜(此后缩写为“CAM”)中血管生成的影响(Kim等人,J.Biol.Chem.,278:29000,2003)。具体地说,将部分鸡胚胎卵清蛋白除去,然后在鸡胚胎中制备用于蛋白处理和观察的窗口。然后,将得到的鸡胚胎在37℃下培养48小时。将LK8-Fc融合蛋白和LK8蛋白放置在Thermanox盖玻片(Nunc,USA)上并干燥,其后将每种蛋白质注射到胚胎CAM中并且胚胎另外继续培养48小时。然后,将脂肪乳液注射到胚胎绒毛膜鸟囊膜中,并且观察theramanox周围的血管生成。在该实施例中,每组使用60个鸡胚胎(图7)。
结果,在使用盐水处理的胚胎作为阴性对照组中,显示了大约39.2±5.6%的血管生成抑制。与此对照,在分别使用每种10μg的LK8蛋白和LK8-Fc融合蛋白处理的情况下,观察到大约66.2%(p<0.05与对照组相比)和大约63.2%(p<0.05与对照组相比)的血管生成抑制。也就是说,该结果显示了使用每种样品处理显著地抑制了血管生成,并且两种样品之间没有观察到效果方面的差别。因此确定,LK8-Fc融合蛋白不仅在实施例3中的体外条件下显示血管生成抑制活性,而且也在体内条件下显示血管生成抑制活性。
实施例6:LK8-Fc融合蛋白的药物动力学(PK)分析
为了观察LK8-Fc融合蛋白的药效动力学,将LK8-Fc融合蛋白和LK8蛋白的每种一次给药到六周龄雄性SD小鼠(Charles River,Japan)然后在各个时间点测定LK8-Fc融合蛋白在血浆中的浓度。特别是,LK8-Fc融合蛋白和LK8蛋白使用FITC(Sigma,USA)标记,并且180μg的每种LK8-Fc-FITC和LK8-FITC蛋白被一次肌肉注射到SD小鼠(每组三个动物)用于蛋白检测。在蛋白质给药后,以0.017,0.051,0.085,0.17,0.51,1,2,4,6,8,24,48,72,120和168小时为间隔,通过眼部出血从动物中取200μl的血液样品,然后从血样中提取血浆。通过使用Fluorometer(PerkinElmer,USA)测定490nm(FITC的激发波长)和535nm(FITC的发射波长)处的吸收来测定血浆中蛋白的浓度并使用标准曲线基于测定的吸收值计算蛋白质浓度(表2)。

蛋白的药物动力学通过表2中显示的数据进行分析。结果,在以LK8-Fc融合蛋白给药的组中,LK8-Fc融合蛋白的半衰期(t1/2)显示为大约177小时,作为体内暴露指示的AUC(0-t)和AUC(inf)被分析分别为103,001h·pmol/mL和176,759h·pmol/mL(表3和图8)因此可以确认,LK8-Fc融合蛋白的半衰期由于Fc融合伴侣而增加,因此其在体内的生物稳定性显著增加。

实施例7:通过使用LK8-Fc融合蛋白处理的抑制实体瘤生长
异体移植有人结肠癌细胞的肿瘤模型被用于观察LK8-Fc融合蛋白是否具有对实体瘤生长的抑制效果。具体地说,在添加了10%FBS(GIBCO,USA)的DMEM(GIBCO,USA)培养的大约5x 106LS174T人结肠癌细胞(ATCC,USA)被皮下接种到BALB/c裸鼠(Charles River,Japan)的背部中央部分。植入结肠癌细胞10天后,将LK8-Fc融合蛋白和LK8蛋白的每种分别以35mg/kg/time和10mg/kg/time的剂量给药,使它们以相同的摩尔浓度给药。在给药方案中,以在实施例6中得到的PK测试结果为基础上将LK8-Fc融合蛋白以7天为间隔给药一次。为了比较LK8-Fc融合蛋白和LK8蛋白之间的效力,以LK8蛋白给药的动物被分成两组,一组是以7天为间隔给药一次的LK8蛋白给药组,另一种组是每天给药一次蛋白的阳性对照组,其中用于阳性对照组的给药方案通过前面的实验被确信有效。每组使用5个动物,并且在肿瘤细胞移植一个月后观察肿瘤的生长。处理过程持续20天,并以3-4天为间隔测定一次肿瘤的尺寸。肿瘤尺寸通过方程宽度2×长度×0.52来计算。这些实验被重复两次,具有相似的结果。
结果,肿瘤的生长由于使用LK8蛋白和LK8-Fc融合蛋白处理而被抑制。而且,在使用LK8蛋白一天给药一次的组和使用LK8F-c融合蛋白以7天为间隔给药一次的组中,与对照组相比观察到显著的肿瘤生长抑制效果(图9)。但是,在其中如用于LK8-Fc融合蛋白的给药方案那样将LK8蛋白以7天为间隔给药一次的情形中,没有观察到肿瘤生长抑制效果。特别是,在植入细胞系之后第21天观察的结果中,以盐水给药的对照组的肿瘤体积平均为2409±591mm3(±SD),而以LK8蛋白每天给药一次的组中肿瘤体积为1188±1022mm3(±SD),以LK8蛋白以7天为间隔给药一次的组中肿瘤体积为3203±3284mm3(±SD),以LK8-Fc融合蛋白以7天为间隔给药一次的组中肿瘤体积为899±773mm3(±SD)。也就是说,与以盐水给药的对照组的肿瘤生长速度的平均值相比,以LK8蛋白每天给药一次显示了大约50%的肿瘤生长抑制,以LK8-Fc融合蛋白以7天为间隔给药一次的组显示大约63%的肿瘤生长抑制。以以LK8蛋白每天给药一次的组与LK8-Fc融合蛋白以7天为间隔给药的组显示了类似的肿瘤抑制活性,这归因于LK8-Fc融合蛋白的增加的半衰期。
实施例8:LK8-Fc融合蛋白的抑制转移活性分析
为了观察LK8-Fc融合蛋白是否具有针对抑制结肠癌细胞系的肝转移的效果,通过将结肠癌细胞植入到BALb/c裸鼠(Charles River,Japan)脾脏中得到的动物模型被用于观察肝转移。特别是,BALb/c裸鼠使用克他命(Sigma,USA)麻醉,然后将3x105LS174T人结肠癌细胞转移到脾脏中,一天后,开始以LK8-Fc蛋白给药。以与实体瘤模型的情形相同的方式,在实施例6中得到的PK检测结果的基础上将蛋白以7天为间隔以35mg/kg/时间的浓度给药。在肿瘤移植第14天之后,将鼠杀死,取出肝脏并且对转移的肿瘤小结的数目计数,从而确定肿瘤转移。
结果,在使用盐水处理的对照组中通过转移到肝脏表面产生的肿瘤小结数为每单位区域120.3±35.1(±SD),而在LK8-Fc融合蛋白处理的组中该数目为56.8±31.9(±SD),表明在LK8-Fc融合蛋白给药的组中,与在对照组中相比通过转移产生的小结数明显减少(图10)。
工业实用性
如上面详细描述的那样,本发明提供了其中LK8蛋白与人免疫球蛋白IgG的Fc区域融合的LK8-Fc融合蛋白。而且,本发明提供了用于治疗肿瘤的含有LK8-Fc融合蛋白的组合物。本发明的LK8-Fc融合蛋白不仅具有引起抗肿瘤的抑制血管生成活性和抑制肿瘤细胞转移活性,而且在体内的半衰期非常长,因此可用作更有效和经济的肿瘤治疗剂或者肿瘤抑制剂。
虽然已经参照具体特征对本发明进行了详细描述,但本领域技术人员将会理解,该描述仅仅是优选实施方式,而不是限制本发明的范围。因此,本发明的实际范围将由所附的权利要求书及其等同物来限定。

1、一种LK8-Fc融合蛋白,其特征在于:LK8蛋白与人免疫球蛋白IgG1的Fc区域融合。
2、根据权利要求1所述的LK8-Fc融合蛋白,其特征在于:另外含有将融合蛋白分泌到细胞外的Igκ前导序列。
3、编码权利要求1或2所述的LK8-Fc融合蛋白的基因。
4、含有权利要求3所述的基因的重组载体。
5、使用权利要求4所述的重组载体转染的重组细胞。
6、根据权利要求5所述的重组细胞,其特征在于:所述细胞是动物细胞。
7、根据权利要求5所述的重组细胞,其特征在于:所述细胞是CHO/LK8-Fc细胞。
8、一种制备LK8-Fc融合蛋白的方法,其特征在于:所述方法包括:培养权利要求5所述的重组细胞。
9、一种治疗癌症的组合物,其特征在于:含有权利要求1或2所述的LK8-Fc融合蛋白。
10、根据权利要求9所述的治疗癌症的组合物,其特征在于:癌症选自由直肠癌、胰腺癌、前列腺癌、肾癌、黑色素瘤、前列腺癌骨转移和卵巢癌所组成的组中。
11、一种抑制血管生成的组合物,其特征在于:含有权利要求1或2所述的LK8-Fc融合蛋白。
