如此处所用,“核酸”指编码可重组产生的多肽的天然发生或部分或完全非天然发生的核酸。核酸可由分离或通过化学手段合成的DNA片段构成。核酸可整合至另一核酸中,例如整合至表达质粒或真核宿主细胞的基因组/染色体中。质粒包括穿梭质粒和表达质粒。通常,所述质粒也包含原核增殖单元,其包含分别用于质粒在原核细胞中复制和选择的复制起点(例如复制的ColE1起点)和选择标记(例如氨苄青霉素或四环素抗性基因)。

“选择标记”是基因,其允许携带该基因的细胞在相应“选择剂”存在下特异性选择。有用的阳性选择标记是抗生素抗性基因。该选择标记允许转化有该基因的宿主细胞在相应抗生素例如选择剂存在下进行阳性选择;未转化的宿主细胞将不能够在存在选择剂的选择性培养条件下生长或存活。选择标记可以是阳性的、阴性的或双功能的。阳性选择标记允许选择携带标记的细胞,而阴性选择标记允许携带标记的细胞被选择性消灭。通常,选择标记将赋予对药物的抗性或补偿宿主细胞中的代谢或分解代谢缺陷。对真核细胞重要的选择标记包括,例如氨基葡糖苷磷酸转移酶(APH)基因,如潮霉素磷酸转移酶(hyg)、新霉素和G418APH、二氢叶酸还原酶(DHFR)、胸苷激酶(tk)、谷氨酰胺合成酶(GS)、天冬酰胺合成酶、色氨酸合成酶(吲哚)、组氨醇脱氢酶(组氨醇D)的基因,以及提供对嘌呤霉素、博来霉素、腐草霉素、氯霉素、Zeocin和霉酚酸抗性的基因。其它选择标记基因描述于WO 92/08796和WO 94/28143中。

为了产生分泌多肽,目的结构基因包括编码“信号序列”或“前导肽”的DNA片段。所述信号序列指导新合成的多肽到达并通过内质网(ER)膜,其中所述多肽可被安排用于分泌。所述信号序列在蛋白质通过ER膜的过程中被信号肽酶切割掉。宿主细胞分泌机器的识别对信号序列的功能是至关重要的。因此所用的信号序列必须被宿主细胞分泌机器的蛋白质和酶所识别。
翻译调节元件包括翻译起始密码子(AUG)和翻译终止密码子(TAA、TAG或TGA)。在一些构建体中可包括内部核糖体进入位点(IRES)。

“多肽”是天然产生或合成产生的由肽键连接的氨基酸残基的聚合物。少于约20个氨基酸残基的多肽可称为“肽”。“蛋白质”是包含一条或更多条多肽链的多肽,其中至少一条链具有100个氨基酸或更多个氨基酸的长度。蛋白质还包含非肽组分,如碳水化合物基团。碳水化合物和其它非肽取代基可通过产生蛋白质的细胞向该蛋白质添加,并根据细胞类型而有所变化。本文中蛋白质以它们的氨基酸主链结构进行定义;添加如碳水化合物基团通常没有详细说明,但仍然存在。
“异源DNA”或“异源多肽”指在给定宿主细胞中不是天然存在的DNA分子或多肽,或DNA分子群体或多肽群体。对特定宿主细胞异源的DNA分子可含有来自宿主细胞物种的DNA(即内源DNA),只要该宿主DNA与非宿主DNA(即外源DNA)组合。例如,认为含有非宿主DNA片段的DNA分子是异源DNA分子,所述非宿主DNA片段编码有效连接包含启动子的宿主DNA片段的多肽。相反,异源DNA分子可包含与外源启动子有效连接的内源结构基因。由非宿主DNA分子编码的多肽是“异源”多肽。
“克隆质粒”是核酸,如质粒、粘粒、噬菌粒或细菌人工染色体(BAC),其具有在宿主细胞中进行自主复制的能力。克隆质粒通常含有一个或少数限制性内切核酸酶识别位点(其允许在不破坏质粒的基本生物学功能的情况下以可决定的方式插入核酸),以及编码选择标记的核苷酸序列,所述选择标记适合用于鉴定和选择转化有克隆质粒的细胞。抗性基因通常包括提供四环素、氨苄青霉素或新霉素抗性的基因。

材料与方法

实施例2
表达质粒的构建
基于T5RNA聚合酶的细菌pQE80表达载体购自Qiagen(Hilden,德国)。
EcoRI和HindIII限制性位点用于插入编码C1qA(SEQ ID NO:6)的核酸序列以生成表达质粒pQE80_C1qA(注释质粒图谱见图1)。EcoRI和HindIII限制性位点用于插入编码在N末端至C末端方向包含抗融合肽T-1357和C1qA(SEQ ID NO:49、50)的缀合物的序列以生成表达质粒pQE80_Rob 1(注释质粒图谱见图2)。EcoRI和HindIII限制性位点用于插入编码在N末端至C末端方向包含C1qA和T-1357(SEQ ID NO:51)的缀合物的序列以生成表达质粒pQE80_Rob 2(注释质粒图谱见图3)。
实施例3
多肽缀合物的产生和纯化
用实施例2中获得的表达质粒转化大肠杆菌细胞。通过氨苄青霉素抗性选择转化的细菌。接种OD为0.1OD/mL的起始培养物。培养物在37℃下添加有0.5μg/ml氨苄青霉素的SB培养基(32g蛋白胨、20g酵母提取物、5g NaCl和5mL 1M NaOH加1L水)中生长。OD600nm高于0.8-1.0后完成培养。将培养液离心并在含有12.11g/l TRIS羟基甲基氨基甲烷(TRIS)、1mM MgSO4、用25%(w/v)HCl调整pH值至7.0的缓冲液中用高压破碎沉淀物中的细胞。每100mg生物量使用500ml缓冲液。细胞破碎后离心悬浮液。用含200ml/l 30%(w/v)Brij、1.5M NaCl和60mMEDTA,pH值调整为7.0的溶液洗涤沉淀物。在第二次离心步骤之后,用100mM TRIS、20mM EDTA、pH 6.5洗涤IB。经过最后的离心步骤后,将IB离心并储存于-20℃。用SDS-PAGE确认样品的同质性。无需额外的纯化步骤。
室温下通过在pH值为11.5-12.0的30mM KOH中搅拌30分钟来溶解IB。当样品完全溶解后,通过在50mM pH值为8.5的硼酸盐缓冲液中对溶解物脉冲进行复性,至最大浓度为0.3mg/ml。

使用SDS-PAGE的表达分析
根据Schagger,H.,和von Jagow,G.,AnalBiochem.166(1997)368-379通过十二烷基硫酸钠(SDS)聚丙烯酰胺凝胶电泳(SDS-PAGE)处理复性的缀合物。

LDS样品缓冲液,四倍浓缩液(4x):4g甘油、0.682g TRIS-Base、0.666g TRIS盐酸盐、0.8g LDS(十二烷基硫酸锂)、0.006g EDTA(乙二胺四乙酸)、按质量(w/w)计0.75ml的1%Serva Blue G250水溶液、按质量(w/w)计0.75ml的1%酚红溶液,加水至总体积为10ml。

表达质粒的构建

实施例6
细胞-细胞融合试验
描述了用于评估1型人免疫缺陷病毒(HIV-1)中和作用的基于细胞系的测定。该测定基于CEM.NKR细胞,其被转染来表达HIV-1共同受体CCR5以补充CD4和CXCR4共同受体的内源表达。获得的CEM.NKR-CCR5细胞有效地复制了R5和X4表型的原代HIV-1分离株。在使用来自HIV-1感染个体的血清或特异性抗HIV-1单克隆抗体的中和测定中,CEM.NKR-CCR5细胞与含有促分裂原激活的外周血液单核细胞(PBMC)的比较表明HIV-1中和作用的灵敏性在两种细胞类型中是相似的。
第1天,将表达gp160的HeLa细胞(2×104细胞/50μl/孔)接种到白色96孔微量滴定板上补充10%FCS和2μg/ml多西环素的DMEM培养基中。第2天,在透明96孔微量滴定板中每孔加入100μl上清液样品或抗体对照。随后加入100μl含8×104CEM-NKr-Luc悬浮细胞的培养基并在37℃温育30分钟。将HeLa细胞培养基从96孔板中吸出,加入200μl抗体/CEM-NKr-Luc混合物中的100μl并在37℃温育过夜。第3天,按100μl/孔加入Bright-GloTM萤光素酶测定底物(1,4-二硫苏糖醇和连二亚硫酸钠;Promega Corp.,美国)并且在室温最少温育15分钟后测定发光。
材料
在含营养物和10%FCS以及400μg/ml G418和200μg/ml潮霉素B的DMEM培养基中培养HeLa-R5-16细胞(在多西环素诱导下表达HIVgp160的细胞系)。CEM.NKR-CCR5-Luc(目录号:5198,从NIH AIDSResearch&Reference Reagent Program McKesson BioServicesCorporation Germantown,MD 20874,美国可获得的T细胞系)。细胞类型:转染(电穿孔)CEM.NKR-CCR5(Cat.#4376)以在HIV-2LTR的转录调控下表达荧光素酶基因并且在含10%胎牛血清、4mM谷氨酰胺、青霉素/链霉素(100U/mL青霉素、100μg/mL链霉素)和0.8mg/ml遗传霉素硫酸盐(G418)的RPMI 1640中进行繁殖。生长特征:圆形淋巴样细胞,形态变化不大。细胞以单细胞在悬浮液中生长,其可形成小团块。每周两次按1∶10分传培养。特殊特征:HIV-2LTR转活后表达荧光素酶活性。适合用原代HIV分离株感染、中和作用和药物敏感性测定(Spenlehauer,C.,等,Virology 280(2001)292-300;Trkola,A.,等,J.Virol.73(1999)8966-8974)。通过NIH AIDS Research and Reference ReagentProgram,NIAID,NIH from Drs.John Moore and Catherine Spenlehauer获得细胞系。Bright-GloTM萤光素酶测定缓冲液(Promega Corp.USA,PartNo E2264B)、Bright-GloTM、萤光素酶测定底物(Promega Corp.USA,partNo EE26B)。
实施例7
多肽结合亲和力的测定
在25℃使用3000仪器(Pharmacia,Uppsala,瑞典)通过表面等离振子共振(SPR)测定基于HIV-1gp41蛋白质的HR1-HR2相互作用(HR,七残基重复1和2区)的多肽的结合亲和力。
很好地建立了用于研究分子相互作用的体系。其允许连续实时监测配体/分析物结合并因此测定结合速率常数(ka)、解离速率常数(kd)和平衡常数(KD)。SPR技术基于测定金包被的生物传感器芯片表面附近的折射指数。折射指数的改变表明由固定化的配体与溶液中注入的分析物间的相互作用引起的表面上质量的改变。如果分子结合表面上固定化的配体,则质量增加,在解离的情况下则质量减少。

待分析的多肽缀合物首先以约1mg/mL的浓度溶解于pH值为9的50mM NaHCO3中,并随后在HPS-P缓冲液中稀释至25到1.95nM的多个浓度。样品接触时间为5分钟(结合相)。此后用HBS-P洗涤芯片表面5分钟(分离相)。在精确的25℃下(标准温度)进行所有相互作用。在测定周期内,样品储存于12℃。以每秒一个信号的检测速率检测信号。将样品以逐渐增加的浓度,50μL/分钟的流速注入偶联HR1的生物传感器元件。用0.5%(w/v)SDS溶液以50μL/分钟的流速将表面再生1分钟。
通过使用BIAevaluation 4.1软件包分析由几种不同浓度获得的sensorgram曲线来测定定义为ka/kd的平衡常数(KD)。通过从HR2-HR1相互作用的值中减去包含HR2的多肽与游离链霉抗生素蛋白表面相互作用的反应值来校正非特异性结合。数据的拟合按照1∶1 Langmuir结合模型。

泳道8-奇异标记(magic mark)
转移缓冲液:192mM甘氨酸、25mM TRIS、20%甲醇(v/v)。
在SDS-PAGE后根据Burnette(Burnette,W.N.,Anal.Biochem.112(1981)195-203)的“Semidry-Blotting-Method”将分离的缀合物电泳转移(25V、1小时)至PVDF滤膜(孔径:0.45μm、Invitrogen Corp.)。
印迹后将膜浸入TBST(1l 10mM TRIS缓冲液,补充有150mMNaCl、1ml20,调节pH值为7.5)中的5%的膜封闭剂(AmershamBiosciences)中,在室温振荡约30分钟并随后在4℃过夜。封闲步骤完成后用TBST将膜洗涤三次。
为了检测,将封闭后的膜与生物素标记的肽HIV-gp41P2(593-621)-Bi以及5μg/ml TBST、0.15mM CaCl2和12mM MgCl2振荡下温育3小时。
显色步骤结束后用TBST洗涤膜并与Lumi-LightPLUsWestern-Blotting底物温育,并随后显影(SDS凝胶见图4且Western印迹见图5)。
Claims (14)
1.多肽缀合物,其特征在于所述缀合物包含
a)选自SEQ ID NO:01的人补体因子C1q的A亚基的第一多肽,
b)选自抗融合肽的第二多肽,所述抗融合肽抑制与膜融合相关事件或膜融合事件本身。
2.权利要求1的缀合物,其特征是所述抗融合肽抑制病毒由于膜融合对未感染细胞的感染。
3.根据权利要求1的缀合物,其特征在于所述第一多肽和所述第二多肽具有选自以下顺序的顺序:
N-末端-第一多肽-第二多肽-C-末端,或
N-末端-第二多肽-第一多肽-C-末端。
4.根据前述权利要求任何一项的缀合物,其特征在于所述缀合物在所述第一多肽和所述第二多肽之间包含接头多肽。
5.根据权利要求1-3任何一项的缀合物,其特征在于所述第二多肽选自包含SEQ ID NO:08到SEQ ID NO:19的抗融合肽。
6.权利要求1-3任何一项的多肽缀合物,其氨基酸序列为SEQ IDNO:50或SEQ ID NO:51。
7.多肽缀合物,其特征在于所述缀合物包含
a)选自SEQ ID NO:01的人补体因子C1q的A亚基的第一多肽(1stpp),
b)选自抗融合肽的第二多肽(2nd pp),所述抗融合肽抑制与膜融合相关事件或膜融合事件本身,和
c)选自抗CCR5抗体的抗原结合片段,或抗HIV-1抗体的抗原结合片段,或抗CD4抗体的抗原结合片段的第三多肽(3rd pp),
d)连接所述第一、第二和/或第三多肽的接头多肽(link pp),
其中所述多肽缀合物的多肽从N到C末端具有顺序:
[1st pp]a-[link pp]m-[2nd pp]b-[link pp]n-[3rd pp]c-[link pp]o-[1st pp]d-[link pp]p-[2nd pp]e-[link pp]q-[3rd pp]f-[link pp]r-[1stpp]g
其中a、b、c、d、e、f、g、m、n、o、p、q、r均为整数0或1,并且
a+d+g=1,
b+e=1,
c+f=0或1,
m、n、o、p、q、r彼此独立地是0或1,

8.权利要求7的缀合物,其特征在于所述抗融合肽抑制病毒由于膜融合对未感染细胞的感染。
9.根据权利要求4或7的缀合物,其特征在于所述接头多肽选自包含SEQ ID NO:20到SEQ ID NO:48的多肽。
10.用于产生根据权利要求1或7的多肽缀合物的方法,其特征在于所述方法包括
a)在适合表达所述多肽缀合物的条件下培养包含编码权利要求1或7的多肽缀合物的核酸的宿主细胞,并
b)从所述细胞或培养基中回收所述多肽缀合物。
11.药物组合物,其含有根据权利要求1到9中任何一项的多肽缀合物,或其药学上可接受的盐,以及药学上可接受的赋形剂或载体。
12.根据权利要求1到9中任何一项的多肽缀合物用于制备治疗病毒感染的药物的用途。
13.根据权利要求12的用途,其特征在于所述病毒感染是HIV感染。
14.根据权利要求1到9中任何一项的多肽缀合物在制备治疗需要抗病毒治疗的患者的药物中的用途。
