技术领域
本发明涉及到一种添加了几种有益于人眼健康成份组合的配方食品,从而提供人体眼部所必需的叶黄素、玉米黄质、锌及硒微量元素的供给。属于食品加工领域。
发明背景
眼健康是一个持久而又重要的话题,人们对眼睛健康的关注一直没有停止过。目前市场上也出现了各种各样的护眼产品,长期以来,人们试图通过摄取或使用某些天然提取或合成的物质或一些微量元素来预防或治疗各种眼部不适或眼科疾病如视疲劳、弱视、近视、白内障、视黄斑衰退病(ARMD)等,保持眼睛健康,虽然这些产品的使用效果良莠不齐,但人们也一直在继续寻找最佳的药物组合以达到最好的护眼效果。
在所有的护眼成分中叶黄素(Lutein)和玉米黄质(Zeaxanthin)是一种十分重要的物质。叶黄素和玉米黄质是一种广泛存在于蔬菜、花卉、水果与某些藻类生物中的天然类胡萝卜素,被广泛应用于食品、保健品、化妆品、医药、烟草和动禽类饲料等多个领域。近年来的研究发现叶黄素和玉米黄质不仅与一种由年龄引起的视黄斑退化(age-related macular degeneration,ARMD)疾病有关,而且能有效地预防视力衰退。叶黄素和玉米黄质是唯一存在于人类眼视网膜上的一种类胡萝卜素,它们选择性地沉积在黄斑区和整个视网膜,以黄斑的中心凹周围密度为最高,视网膜周边部分逐渐减少。这些黄斑色素能有效地防止视网膜上氧化反应的发生。可以认为,治疗和预防眼底黄斑衰退或损伤的有效办法为补充叶黄素和玉米黄质。而且,叶黄素和玉米黄质在无ARMD时可以用于提高视觉功效,如对青少年近视及老年的肌肉退化症等所导致的老年人盲眼病,以及日光、电脑等所发射的紫外线对眼睛的损害有较好的保健作用。大量临床检测的结果表明:人体每日的推荐的叶黄素和玉米黄质摄入量为6mg,一般认为,叶黄素晶体可接受的日摄入量(ADI)为25mg/人/天,而不会引起胡萝卜素黄皮肤病。
叶黄素和玉米黄质独特的生理功能,已经引起了人们越来越大的兴趣。目前国外出现了大量含以上两种天然色素的健康食品和强化食品。美国FDA1995年批准了叶黄素和玉米黄质作为食品补充剂用于食品中,以提高其营养价值。如用于益视饮料和婴幼儿食品中。中国国内在2006年年底也批准了叶黄素作为着色剂在饮料、焙烤食品、冷冻食品、果冻和果酱类食品上的应用,添加量为50-150mg/kg;同时也批准了叶黄素作为营养强化剂使用。新的市场推动使叶黄素2004年的市值达到了1.39亿美元,而1999年只有6400万美元。强劲的增长势头还将继续,BCC认为,到2009年每年的增长率将达6.1%。
除了叶黄素和玉米黄质外,其它类物质如锌、硒对眼健康也很有益处。研究表明,锌能增强视觉神经的敏感度,是视网膜组织细胞中视黄醇还原酶的主要成份,强化锌时能够直接影响维生素A对视黄质的合成和代谢,有利于视黄醛的作用,从而增强人对弱光的适应能力,缺锌还会影响视网膜细胞的辩色能力。硒是维持视力的重要微量元素,是谷胱苷肽过氧化物酶的主要成分,缺硒可导致弱视。因此叶黄素类类胡萝卜素和锌、硒是很好的搭档,四者配伍有促进强化作用,从而共同对眼睛的营养保护作用,对预防青少年近视,上班族的视疲劳及老年性视力衰退有很好的护眼保健功能。
基于对眼健康的日益重视及对上面述及物质护眼功能的认同,目前市场上也出现了富含叶黄素或硒或锌或三者都含有的食品包括饮料、牛奶、酸奶等,但这些产品中对玉米黄质的功能或添加往往比较忽略,实际上,从某种意义上来说玉米黄质有着与叶黄素相同甚至更好的护眼效果,如在人类视网膜中心视黄斑处,玉米黄质的含量往往是叶黄素含量的两倍左右,越往视网膜边缘叶黄素的含量越高,玉米黄质的含量相对越少,在人眼视网膜最边缘处叶黄素的含量是玉米黄质的三倍左右,人体血浆中叶黄素含量也是玉米黄质含量的三倍,这说明在人体内存在着叶黄素向玉米黄质转换的机制,同时其它的研究也发现,人体摄入的部分叶黄素会转化为玉米黄质,所以说玉米黄质在人眼健康中有着不可忽略的作用。我们的人体预试验结果也证明补充玉米黄质与叶黄素的混和物,并辅以一定的硒,锌时,人眼的视觉功效特别是明视持久度和眼部自觉症状能到得很好的改善。
如前所述,目前市场上出现了各类护眼产品,但许多是这四者成分中一种或两种或三种成分的组合物,如CN1625395A提到用叶黄素和玉米黄质组合物用于眩光保护;CN1253091提及一种含叶黄素、锌、硒的有利于视力的乳制品或饮料;US2005/0147648通过补充玉米黄质、多不饱和脂肪酸、植物提取物等的组合来保持眼健康;US7,267,830提到补充类胡萝卜素和维生素A、C,DHA及一些微量元素来提高眼睛的健康水平的目的,在揭示的配方中叶黄素和玉米黄质比例大概固定在3∶1水平,没有特别意识到玉米黄质的功能;US7,282,225通过膳食补充维生素A、C、E,类胡萝卜素(beta-胡萝卜素、叶黄素、玉米黄质),微量元素来提高视觉功能和敏锐度,在这个配方中也没有突出玉米黄质在改善眼睛明视持久度方面的独特作用。
本发明提供了一种含有上述四种成分特定比例组合的配方食品,此类食品供给了人眼所需的叶黄素、玉米黄质、锌及硒等微量元素,有益于人眼健康,特别有利于人眼明视持久度的提高,有助于缓解视疲劳,改善眼部自觉症状。
发明内容
叶黄素和玉米黄质是人类视网膜中唯一存在的两种类胡萝卜素,与视黄斑衰退症息息相关,锌和硒能增强人眼对弱光的适应能力和辨色能力,四种物质的特定比例的组合物应用于配方食品中有利于人眼健康,本发明的目的就是说明引入了上述组合物的配方食品。
本发明中所用术语“视觉功效”指视觉功能如敏度、对比敏感性、眩光恢复、暗适应、明视持久度、光应激恢复及色觉等,特别关注的是明视持久度的提高,例如缓解视疲劳、改善眼部自觉症状等。
本发明中的术语“明视持久度”是用于评价视疲劳的一种指标。当人大脑皮质兴奋性降低时,视觉分析功能下降,眼睛注视对象物的过程中,不能明视的时间增加,能明视时间减少。这种明视时间对注视时间的百分比称为明视持久度,它是综合反应视功能和心理功能的一种指标。明视持久度(%)=明视时间/总视物时间*100。
本发明中定义的“配方食品”指的是各种为了强化某种营养功能的食品,如配方奶粉,包括配方婴儿奶粉或配方老年奶粉等,各种功能性饮料如含乳饮料、橙汁饮料、可乐、碳酸饮料、悬浮饮料等,焙烤类食品如蛋糕、饼干等,也包括用于营养补充为目的的各种膳食补充片剂、胶囊等。
本发明提供了一种有益于眼健康的配方食品,所述配方食品包括叶黄素、玉米黄质、锌、和硒。
其中,所述锌为无机锌盐或有机锌盐,所述无机锌盐为硫酸锌或亚硫酸锌,所述有机锌盐为乳清酸锌或抗坏血酸锌。
所述硒为无机硒盐或有机硒盐,所述无机硒盐为硒代磷酸盐、或亚硒酸盐,所述有机硒盐为硒代氨基酸或含硒或富含硒酵母;其中,硒代磷酸盐为硒代磷酸钠,所述亚硒酸盐为亚硒酸钠或硒酸钠,所述硒代氨基酸为L-硒代甲硫氨酸,所述含硒或富含硒酵母为啤酒酵母或面包酵母。
其中,叶黄素在所述配方食品的添加量为0.3~250mg/kg,玉米黄质在所述配方食品的添加量为0.3~250mg/kg,叶黄素和玉米黄质的质量比例为0.05∶10.0~10.0∶0.05之间,优选叶黄素和玉米黄质的质量含量比例为0.1∶2.0~2.0∶0.1之间。具体地说,本发明中叶黄素和玉米黄质可以以一定的比例混合得到,也就是说在得到可应用于制备添加到配方食品中的剂型之前,它们就是按一定的比例混合的,其中之一叶黄素和玉米黄质的混合比例为0.05∶1.0到1.0∶0.05之间。
其中,锌在所述配方食品的添加量为4~80mg/kg,添加的锌为有机锌或无机锌时以纯锌计。硒在所述配方食品的添加量为30~350μg/kg,添加的硒为有机硒或无机硒时以纯硒计。
其中,叶黄素是从天然物中提取的,并以晶体或脂肪酸酯的形式存在。叶黄素添加在食品中的制剂形式为微胶囊粉末、或者均匀分散在可食性植物油中,或者水分散性乳液。其中,叶黄素在其相应制剂形式中质量百分含量为1~25wt%。
其中,玉米黄质是从天然物中提取的或通过化学合成得到的,并以为晶体或脂肪酸酯的形式存在。玉米黄质添加在食品中的制剂形式为微胶囊粉末、或者均匀分散在可食性植物油中、或者为水分散性乳液。其中,玉米黄质在其相应制剂形式中质量百分含量为1~25wt%。
其中,所述配方食品还包括至少一种抗氧化剂,所述抗氧化剂包括合成生育酚(维生素E)、天然生育酚(天然维生素E)、D-抗坏血酸钠(异Vc钠)、抗坏血酸(维生素C)、抗坏血酸棕榈酸酯、卵磷脂。
其中,所述配方食品应用于各种配方奶粉、各种功能性饮料、焙烤类食品、各种膳食补充片剂和胶囊。具体地说,配方食品可以应用于各种为了强化某种营养功能的食品,如配方奶粉,包括配方婴儿奶粉或配方老年奶粉等;各种功能性饮料如含乳饮料、橙汁饮料、可乐、碳酸饮料、悬浮饮料等;焙烤类食品如蛋糕、饼干等;也包括用于营养补充为目的的各种膳食补充片剂、胶囊等。
本发明中预先将要添加的叶黄素、玉米黄质、锌、硒适当的剂型溶解于水或油中,在配方食品加工过程中适当的步骤中加入。
应用在本发明所提供的范围内,添加了以上组分,不会影响到配方食品原有的口味。
在本发明的范围内,所述的按比例混合的四种物质的组合物也可制成软胶囊或硬胶囊或片剂的形式,直接经口服用,达到膳食补充的目的。
此外,在所述的组合物中,或者在叶黄素、玉米黄质、锌、硒单独组份的制剂中可以加入一定量的抗氧化剂如合成生育酚、天然生育酚、抗坏血酸棕榈酸酯、卵磷脂等。
本发明的有益效果是补充叶黄素、玉米黄质、锌、硒的混合物,并且相对于叶黄素,适当地扩大玉米黄质的量能大大提升这种药物组合的缓解视疲劳,改善眼部自觉症状的效果。本发明的配方食品有益于人眼健康,特别有利于人眼明视持久度的提高,有助于缓解视疲劳,改善眼部自觉症状。

具体实施方式
以下将结合实施例对本发明做进一步说明,本发明的实施例仅用于说明本发明的技术方案,并非限定本发明。
实施例1
叶黄素、玉米黄质、锌、硒缓解视疲劳实验研究
本试验按自愿原则选择年龄为17-23岁之间,视力易疲劳的长期使用视屏显示终端(video display terminals,VDT)操作者278名,挑选时排除下列受试者:患有某种眼科疾病可能影响到试验测试结果;长期或短期服用有关药物、保健品或作用其它治疗方法可能影响到对结果的判定;不符合纳入标准,未按规定食用受试药物者,或者资料不全等影响功效或安全判定者。
实验中设立对照组和试食组,其中按随机原则对照组选定48人,服用安慰剂,试食组又分为I、II、III、IV、V五组,每组46人,各组分别服用表1中所示的药物组合片剂,每日1次,每次2片,连续服用35天,试验采用自身前后对照和组间对照。

其中:
1、配方中除了列出的活性成分外,其中叶黄素为CarolGoldTM10%TAB,玉米黄质为CarolZeaTM10%TAB,锌为抗坏血酸锌,硒为硒代氨基酸如L-硒代甲硫氨酸,其余为片剂所需的辅料如羟甲基纤维素、变性淀粉等,总片剂重为300mg。
2、安慰剂的外观颜色、片剂重量与试食片剂完全一致。
各项指标于试食试验开始及结束进各测定一次,检测指标分为安全性指标和功效性指标。前者包括一般体格检查、血、尿、便常规检查,血生化指标检查,B超、胸部X线和心电图检查等其它相应的检查。后者包括眼部症状检查:①详细询问病史,观察眼部自觉症状:眼胀、眼痛、畏光、视物模糊、眼干涩等。按症状轻重(重症3分,中度2分,轻度1分)在试食前后统计积分值,并就主要症状改善(每一症状改善1分为有效,改善2分为明显),计算症状改善率。②明视持久度测定:明视时间对注视时间的百分比称为明视持久度,测定时间为3min,测定2次取平均值。
数据处理:用统计软件STATE6.0计算分析数据。凡自身对照资料采用配对t检验,两组均数比较采用成组t检验,后者需进行方差齐性检验,对非正态分布或方差不齐的数据进行适当的变量转换,待满足正态方差齐性后,用转化的数据进行t检验;若转换的数据仍不能满足于正态方差齐性要求,改用t检验或秩和检验;但变异系数太大,如CV>50%的资料,应用秩和检验。功效指标用X2检验。
对照组和试验组试食后的明视持久度变化率见图1。试食后试验组受试者明视持久度比试食前均有显著提高(P<0.05),单纯补充叶黄素者提高10.27±8.56%;补充叶黄素与玉米黄质的混合物时根据其中叶黄素和玉米黄质比例的不同,受试者明视持久度提高的程度也不一样,在总量不变的情况下,叶黄素与玉米黄质比例为3∶1时明视持久度提高14.56±10.21%,而当叶黄素与玉米黄质的比例调整为1∶2时,这一数值为18.23±7.25%,二者间存在显著性差异(P<0.05);当再补充一定量的微量元素锌、硒时,受试者的明视持久度进一步得到显著性提高,而且当叶黄素和玉米黄质的比例较小时,这一提高效果更为明显(P<0.01)。
以眼部自觉症状(临床症状积分)为指标(参见图2),也可以看出这种改善视觉的效果,试食前后的结果具有统计学意义(P<0.001),而且,当补充叶黄素、玉米黄质、锌、硒四者混合物并且其中玉米黄质的比例高于叶黄素时,总有效率最高,达到59.45%,与叶黄素比例高时(总有效率为47.27%)有显著性差异(P<0.05)。
而且试食前后试验组和对照组人群的一般体格检查、血液生化指标以及尿常规检查结果均在正常范围内。试食前试验组和对照组人群胸透、心电图、腹部B超检查均在正常范围内。
此试验很好地证明了本发明的优点,也就是补充叶黄素、玉米黄质、锌、硒的混合物,并且相对于叶黄素,适当地扩大玉米黄质的量能大大提升这种药物组合的缓解视疲劳,改善眼部自觉症状的效果。
实施例2
含叶黄素、玉米黄质、锌、硒的健视饮料
此饮料中含有白砂糖、柠檬酸、柠檬酸钠、B族维生素、叶黄素、玉米黄质、锌、硒、其它植物提取物等。按总量为1000g计,其中含叶黄素60mg,玉米黄质0.3mg,锌40mg,硒其余为别的组份和水,用一定量的合成生育酚干粉作抗氧化剂。用到的叶黄素为5%CWS-B微粒形式,玉米黄质为5%CWS干粉形式,锌为乳清酸锌,硒为L-硒代甲硫氨酸。
按实施例1相似的试验设计进行人体试验,试验结果分析见表2。

实施例3
含叶黄素、玉米黄质、锌、硒的婴儿配方奶粉
此婴儿配方奶粉中除含有常规碳水化合物、脂肪、蛋白质、维生素A、D、E、K、B族维生素、微量元素、低聚糖外,还含有叶黄素、玉米黄质、锌、硒等有益眼健康的成分,按总量为1000g计,其中叶黄素含量为0.3mg,玉米黄质60mg,锌4mg,硒150μg。用到的叶黄素为1%CWS干粉形式,玉米黄质为5%CWS干粉形式,锌为硫酸锌,硒为富含硒的面包酵母,天然生育酚为抗氧化剂。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例4
含叶黄素、玉米黄质、锌、硒的含乳饮料
此饮料中含有白砂糖、全脂奶粉、柠檬酸钠、B族维生素、稳定剂、叶黄素、玉米黄质、锌、硒等。按总量为1000g计,其中含叶黄素250mg,玉米黄质12.5mg,锌80mg,硒180μg,添加一定量的抗坏血酸棕榈酸酯,其余为别的组份和水。用到的叶黄素为10%CWS-B微粒形式,玉米黄质为1%CWS干粉形式,锌为乳清酸锌,硒为硒代磷酸钠。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例5
含叶黄素、玉米黄质、锌、硒的中老年配方奶粉
此配方奶粉中除含有常规碳水化合物、脂肪、蛋白质、维生素A、D、E、K、B族维生素、微量元素、低聚糖外,还含有叶黄素、玉米黄质、锌、硒等有益眼健康的成分,按总量为1000g计,其中叶黄素含量为12.5mg,玉米黄质250mg,锌40mg,硒并添加适量的卵磷脂。用到的叶黄素为1%CWS干粉形式,玉米黄质为5%CWS干粉形式,锌为硫酸锌,硒为硒酸钠。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例6
含叶黄素、玉米黄质、锌、硒的面包
此配方面包中除含有常规中高筋面粉、白砂糖、搅拌用黄油、酵母、一定量的面包改良剂外,还含有叶黄素、玉米黄质、锌、硒等有益眼健康的成分,按总量为1000g计,其中叶黄素含量为54.7mg,玉米黄质19.4mg,锌60mg,硒并添加了适量的D-抗坏血酸钠。用到的叶黄素为10%CWS干粉形式,玉米黄质为12%CWS干粉形式,锌为亚硫酸锌,硒为富含硒的啤酒酵母。得到的面包织构均匀,色泽金黄,口感舒适。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例7
含叶黄素、玉米黄质、锌、硒的果冻
此配方果冻中除含有常规中果冻胶、白砂糖、乳酸钙、柠檬酸外,还含有叶黄素、玉米黄质、锌、硒等有益眼健康的成分,另添加一定量的混合生育酚、异Vc钠。按总量为1000g计,其中叶黄素含量为15.0mg,玉米黄质1.4mg,锌40mg,硒250μg,抗坏血酸为抗氧化剂。用到的叶黄素为25%干粉形式,玉米黄质为5%CWS干粉形式,锌为乳清酸锌,硒为亚硒酸钠。得到的果冻颜色诱人,澄清透亮。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例8
富含叶黄素、玉米黄质、锌、硒的明胶软胶囊
按每个软胶囊重量为200mg计,其包含以下组分:叶黄素脂肪酸酯12mg,玉米黄质2mg,维生素E 10mg,维生素C 15mg,乳清酸锌3.6mg,亚硒酸钠24μg,卵磷脂18mg,大豆油30mg。其中,用到的叶黄素脂肪酸酯为20%OS形式,玉米黄质为25%OS形式。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。
实施例9
富含叶黄素、玉米黄质、锌、硒的明胶软胶囊
按每个软胶囊重量为200mg计,其包含以下组分:叶黄素3.6mg,玉米黄质12mg,维生素E 8mg,维生素C 10mg,乳清酸锌1.8mg,亚硒酸钠32μg,卵磷脂24mg,大豆油21mg。其中,用到的叶黄素为25%OS形式,玉米黄质为20%OS形式。
按实施例1相似的试验设计进行人体试验,试验结果分析见表3。

本发明通过上面的实施例进行举例说明,但是,本发明并不限于这里所描述的特殊实例和实施方案。任何本领域中的技术人员很容易在不脱离本发明精神和范围的情况下进行进一步的改进和完善,因此本发明只受到本发明权利要求的内容和范围的限制,其意图涵盖所有包括在由附录权利要求所限定的本发明精神和范围内的备选方案和等同方案。
Claims (19)
1、一种有益于眼健康的配方食品,所述配方食品包括叶黄素、玉米黄质、锌、和硒。
2、如权利要求1所述的配方食品,其中,所述锌为无机锌盐、或有机锌盐。
3、如权利要求2所述的配方食品,其中,所述无机锌盐为硫酸锌或亚硫酸锌,所述有机锌盐为乳清酸锌或抗坏血酸锌。
4、如权利要求1所述的配方食品,其中,所述硒为无机硒盐或有机硒盐。
5、如权利要求4所述的配方食品,其中,所述无机硒盐为硒代磷酸盐或亚硒酸盐,所述有机硒盐为硒代氨基酸、或含硒或富含硒酵母。
6、如权利要求5所述的配方食品,其中,所述硒代磷酸盐为硒代磷酸钠,所述亚硒酸盐为亚硒酸钠或硒酸钠,所述硒代氨基酸为L-硒代甲硫氨酸,所述含硒或富含硒酵母为啤酒酵母或面包酵母。
7、如权利要求1所述的配方食品,其中,叶黄素在所述配方食品的添加量为0.3~250mg/kg,玉米黄质在所述配方食品的添加量为0.3~250mg/kg。
8、如权利要求1或7所述的配方食品,其中,叶黄素和玉米黄质的质量含量比例为0.05∶10.0~10.0∶0.05之间。
9、如权利要求1或7所述的配方食品,其中,叶黄素和玉米黄质的质量含量比例为0.1∶2.0~2.0∶0.1之间。
10、如权利要求1所述的配方食品,其中,锌在所述配方食品的添加量为4~80mg/kg。
11、如权利要求1所述的配方食品,其中,硒在所述配方食品的添加量为30~350μg/kg。
12、如权利要求1所述的配方食品,其中,叶黄素是从天然物中提取的,并以晶体或脂肪酸酯的形式存在。
13、如权利要求11所述的配方食品,其中,叶黄素添加在食品中的制剂形式为微胶囊粉末、或者均匀分散在可食性植物油中,或者水分散性乳液。
14、如权利要求1、12和13任一所述的配方食品,其中,叶黄素在其相应制剂形式中质量百分含量为1~25wt%。
15、如权利要求1所述的配方食品,其中,玉米黄质是从天然物中提取的或通过化学合成得到的,并以为晶体或脂肪酸酯的形式存在。
16、如权利要求14所述的配方食品,其中,玉米黄质添加在食品中的制剂形式为微胶囊粉末、或者均匀分散在可食性植物油中、或者为水分散性乳液。
17、如权利要求1、15和16任一所述的配方食品,其中,玉米黄质在相应制剂形式中质量百分含量为1~25wt%。
18、如权利要求1所述的配方食品,其中,所述配方食品还包括至少一种抗氧化剂,所述抗氧化剂包括合成生育酚、天然生育酚、D-抗坏血酸钠、抗坏血酸、抗坏血酸棕榈酸酯、卵磷脂。
19、一种如权利要求1~18所述的配方食品在制备各种配方奶粉、各种功能性饮料、焙烤类食品、各种膳食补充片剂和胶囊的应用。
