Abstract
本发明公开了一种新的植物组织特异性表达启动子——GIF1启动子及其应用,所述的启动子分离自作物籽粒灌浆基因GIF1。本发明还公开了含有所述启动子序列的载体以及宿主细胞。本发明还公开了所述启动子的用途,所述启动子对于指导目的基因在植物中组织特异性表达,尤其是在植物的种子、节间和/或根部特异性表达,从而对改良植物(尤其是作物)品质特别有用。
Description
技术领域
本发明属于植物学和基因工程领域。具体而言,本发明涉及新的植物(优选作物)组织特异性表达启动子,本发明还涉及所述启动子的用途,尤其在启动目的基因在植物组织中特异性表达的基因工程中的用途。
背景技术
当前,对于农作物产量改良的研究主要集中在以下几个方面:1.增加作物的源,即加强作物的光合作用;2.增加作物库的大小;3.提高作物光合产物由源向库运输的能力。
在很多作物中,籽粒是判断该作物优劣的标准,直接关系到作物的产量和品质。尽管人们采取了许多方式来提高农作物的产量和对农作物进行改良,然而,目前还缺乏有效的手段。例如,我国的主要粮食作物水稻,当前的许多水稻高产栽培品种尤其超级杂交稻和大穗大粒品种存在籽粒灌浆不饱满的情况,这在很大程度上影响了水稻产量的进一步提高。
植物的根部对其生长、营养吸收、能量传递等均具有重要作用。改善作物根部的吸收、传输、生长等能力有助于提高作物的总体生长和品质。植物的节间是物质、能量运输的枢纽,也对植物的支撑起到一定作用。
对于除作物以外的其它植物而言,种子、节间或根本身或整株植物可能存在其经济价值或实用价值,例如观赏等。因此,组织特异性地改造这些植物,是提高其品质的有用手段。
组织特异性表达启动子可以驱动靶基因在特定的组织或细胞表达,而不影响植物其它组织的生理活动。
因此,为了改良植物(尤其是作物)的品质性状或调控作物籽粒次生代谢,本领域迫切需要开发新的特异性良好的植物组织(尤其是在种子、根部、节间等部位)特异性表达启动子,从而实现农作物或其它经济植物产量和品质的提高。
发明内容
本发明的目的在于提供一种新的植物(尤其是作物)组织特异性表达启动子,所述启动子可指导目的基因特异性地在植物籽粒、根部或节间等组织中表达。
本发明的目的还在于提供所述启动子的用途。
在本发明的第一方面提供了一种分离的多核苷酸,其选自下组:
(1)由SEQ ID NO:3的1-2379位所示的序列构成的核苷酸序列;
(2)具有SEQ ID NO:3中1-2379位所示的核苷酸序列且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(3)SEQ ID NO:3中1-2379位所示的核苷酸序列的片段,所述片段能启动目的基因在植物中组织特异性表达;
(4)在严格条件下能够与(1)、(2)或(3)限定的核苷酸序列杂交且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(5)与(1)、(2)或(3)限定的核苷酸序列有70%、80%、85%、90%、95%以上相同性且能启动目的基因在植物中组织特异性表达的核苷酸序列;或
(6)与(1)-(5)任一项所限定的核苷酸序列互补的核苷酸序列。
在一个优选例中,所述多核苷酸的序列如SEQ ID NO:3的1-2379位所示。
在一个优选例中,所述植物是作物,优选禾本科植物,更优选所述的禾本科作物选自:水稻、小麦、大麦、玉米、或高粱。
在一个优选实施方式中,所述目的基因选自:编码具有SEQ ID NO:2氨基酸序列的作物籽粒灌浆蛋白、以及与作物淀粉转运、积累相关的基因。

在本发明的第五方面中,提供了一种制备转基因植物的方法,所述转基因植物的组织特异性表达目的基因,所述的方法包括:
(a)提供含有作为启动子元件的本发明的多核苷酸以及与所述多核苷酸可操作地连接的目的基因的构建物;
(b)将所述构建物导入植物细胞、组织或器官;
(c)选出导入了所述构建物或染色体中整合有所述构建物的植物细胞、组织或器官;和
(d)使所述植物细胞、组织或器官再生成植株。
在一个优选例中,所述植物为作物。
在一个优选例中,所述目的基因特异性表达的组织是种子、根部和/或节间,优选种子。
在本发明的第六方面中,提供了一种转基因植物。

具体实施方式
本发明人经过广泛而深入的研究,首次从植物籽粒灌浆基因(GIF1,其序列如SEQ ID NO:3所示)中分离到一个能够指导目的基因在作物中组织特异性表达的启动子,该启动子来源于GIF1基因(GIF1,Grain Incomplete Filling1)的上游区域,因此本发明人将之命名为GIF1启动子。所述的启动子对于定点地改良植物品质是特别有用的。在此基础上完成了本发明。
术语
如本文所用,所述的“植物”包括但不限于:作物、具有经济价值或实用价值的其它植物等。
如本文所用,所述的“作物”包括但不限于:禾本科植物。更优选的,所述的禾本科植物包括但不限于:水稻,小麦、大麦、玉米、高粱等。
如本文所用,“分离的”是指物质从其原始环境中分离出来(如果是天然的物质,原始环境即是天然环境)。如活体细胞内的天然状态下的多聚核苷酸和多肽是没有分离纯化的,但同样的多聚核苷酸或多肽如从天然状态中同存在的其他物质中分开,则为分离纯化的。
如本文所用,所述的“可操作地连接”是指两个或多个核酸区域或核酸序列的功能性的空间排列。例如:启动子区被置于相对于目的基因核酸序列的特定位置,使得核酸序列的转录受到该启动子区域的引导,从而,启动子区域被“可操作地连接”到该核酸序列上。
如本文所用,所述的“启动子”或“启动子区(域)”是指一种核酸序列,其通常存在于目的基因编码序列的上游(5’端),能够引导核酸序列转录为mRNA。一般地,启动子或启动子区提供RNA聚合酶和正确起始转录所必需的其它因子的识别位点。在本文中,所述的启动子或启动子区包括启动子的变体,其通过插入或删除调控区域,进行随机或定点突变等来获得。
如本文所用,术语“特异性表达”是指目的基因在特定的时间和/或特定的组织表达。“组织特异性启动子”又称“器官特异性启动子”,在这类启动子调控下,基因往往只在某些特定的器官或组织部位表达,并表现出发育调节的特性。本发明中,所述的“组织特异性启动子”是指导基因在植物中组织特异性表达启动子,所述组织选自籽粒、根部或节间。
通常,如果在某组织或器官中mRNA以比在其它组织或器官中高至少1倍,优选高至少5倍,优选至少高10倍,更优选至少高100倍,最优选至少高1000倍水平被表达,则该启动子被认为是组织或器官特异性的。
如本文所用,“外源的”或“异源的”是指来自不同来源的两条或多条核酸或蛋白质序列之间的关系。例如,如果启动子与目的基因序列的组合通常不是天然存在的,则启动子对于该目的基因来说是外源的。特定序列对于其所插入的细胞或生物体来说是“外源的”。
如本文所用,“顺式调控元件”是指对基因的转录起始和转录效率起调节作用的保守性碱基序列。
如本文所用,“目的基因”是指可由本发明的启动子指导表达的基因。合适的目的基因包括但不限于:改良作物籽粒品质、产量、性状或代谢相关的基因(例如编码具有SEQ ID NO:2氨基酸序列的作物籽粒灌浆蛋白)、以及与作物淀粉转运、积累相关的基因。
植物组织特异性表达启动子
本发明提供一种植物组织特异性表达的多核苷酸(启动子),所述的核酸具有选自下组的序列:
(1)由SEQ ID NO:3的1-2379位所示的序列构成的核苷酸序列;
(2)具有SEQ ID NO:3中1-2379位所示的核苷酸序列且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(3)SEQ ID NO:3中1-2379位所示的核苷酸序列的片段,所述片段能启动目的基因在植物中组织特异性表达;
(4)在严格条件下能够与(1)、(2)或(3)限定的核苷酸序列杂交且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(5)与(1)、(2)或(3)限定的核苷酸序列有70%、80%、85%、90%、95%以上相同性且能启动目的基因在植物中组织特异性表达的核苷酸序列;或
(6)与(1)-(5)任一项所限定的核苷酸序列互补的核苷酸序列。
在本发明的一个实施方式中,所述多核苷酸的序列如SEQ ID NO:3的1-2379位所示。
在本发明的另一个实施方式中,所述植物是作物,优选禾本科植物,包括但不限于水稻、玉米、小麦、大麦、或高粱等。
在本发明的另一个实施方式中,特异性表达所述目的基因的组织是种子、根部和/或节间,优选种子。
也可通过基因工程方法在该启动子中加入增强子等顺式作用元件达到进一步调控基因表达的作用。
多核苷酸的杂交是本领域技术人员熟知的技术,特定的一对核酸的杂交特性指示它们的相似性或同一性。因此,本发明还涉及与前述指定的核苷酸序列杂交且两个序列之间具有至少50%,较佳地至少70%,更佳地至少80%(例如85%、90%、95%、96%、97%、98%、或99%)相同性的多核苷酸。
本发明特别涉及在严格条件下与本发明所述多核苷酸可杂交的多核苷酸。“严格条件”是指:(1)在较低离子强度和较高温度下的杂交和洗脱,如0.2×SSC,0.1%SDS,60℃;或(2)杂交时加有变性剂,如50%(v/v)甲酰胺,0.1%小牛血清/0.1%Ficoll,42℃等;或(3)仅在两条序列之间的相同性至少在50%,优选60%以上、65%以上、70%以上、75%以上、80%以上、85%以上或90%以上,更优选是95%以上时才发生杂交。并且,可杂交的多核苷酸也具有指导目的基因在植物中组织特异性表达的功能。
本发明还包括与本发明的任一种启动子序列具有50%或以上(优选60%以上,70%以上,80%以上,更优选90%以上,最优选95%以上)相同性的核酸,所述核酸也具有指导目的基因在植物中组织特异性表达的功能。“相同性”是指按照位置相同的百分比,两条或多条核酸之间的相似水平(即序列同源性、相似性或同一性)。
在本发明启动子的指导下,可以使β-葡萄糖苷酶(GUS)基因特异地在水稻、小麦、大麦、玉米、高粱等禾本科植物的籽粒、根部或节间中表达。因此可见,本发明的启动子是一种组织或器官特异性的启动子。所述的启动子对于定点地改良植物的品质是特别有用的。
β-葡萄糖苷酶(GUS)能催化裂解一系列的β-葡萄糖苷,产生具有发色团或荧光的物质,可用分光光度计、荧光计或组织化学等方法对GUS活性进行定量和空间定位分析。在本技术领域中,GUS基因已被广泛地用作转基因植物、细菌和真菌的报告基因,特别是其可被用于研究外源基因表达的具体细胞和组织部位。
本发明的组织特异性启动子在理论研究和作物改良中具有重要的应用价值。这些启动子可以被应用于标记特定组织,引导特定的功能基因在特定的组织中表达,以及应用于特有组织的生长发育研究和针对性改良。本发明的启动子对于改良植物的品质或表型(尤其是作物籽粒(例如水稻籽粒))是特别有用的。
目的基因
本发明的启动子可以被可操作地连接到目的基因上,该目的基因相对于启动子而言可以是外源(异源)的。对所述目的基因的核酸序列没有特别的限制(如一种结构性核酸序列),所述的目的基因优选编码具有特定功能的蛋白,例如某些在农业或植物改良上具有重要特性或功能的蛋白。
合适的目的基因包括但不限于:改良作物品质、性状或代谢相关的基因。例如,所述的目的基因选自:与作物籽粒的品质、产量、抗性或代谢相关的基因(例如作物灌浆蛋白基因(GIF1基因)),以及与作物淀粉转运、积累相关的基因。
本发明的启动子还可以被可操作地连接到被改进的目的基因序列上,该目的基因相对于启动子是外源(异源)的。所述的目的基因可以被改进来产生各种期望的特性。例如,目的基因可以被改进来增加必需氨基酸的含量,提高氨基酸序列的翻译,改变翻译后的修饰(如磷酸化位点),将翻译产物转运到细胞外,改善蛋白的稳定性,插入或删除细胞信号等。
此外,启动子和目的基因可以设计成下调特定基因。这一般是通过将启动子连接到目的基因序列上来实现,该序列以反义反向被引导。本领域的普通技术人员熟悉这种反义技术。任何核酸序列可以以这种方式被调节。
载体和宿主细胞
本发明的任何一种前述的启动子和/或目的基因序列可被包含在重组载体中。
作为一种方式,所述的重组载体包括本发明的启动子,在所述启动子的下游包含多克隆位点或至少一个酶切位点。当需要表达目的基因时,将目的基因连接入适合的多克隆位点或酶切位点内,从而将目的基因与启动子可操作地连接。
作为另一种方式,所述的重组载体包括(从5’到3’方向):引导目的基因转录的启动子,和目的基因。如果需要,所述的重组载体还可以包括3’转录终止子,3’多聚核苷酸化信号,其它非翻译核酸序列,转运和靶向核酸序列、抗性选择标记、增强子或操作子。
用于制备重组载体的方法是本领域熟知的。术语“重组表达载体”指本领域熟知的细菌质粒、噬菌体、酵母质粒、植物细胞病毒、哺乳动物细胞病毒或其他载体。总之,只要其能够在宿主体内复制和稳定,任何质粒和载体都是可以被采用的。
本领域的技术人员熟知的方法能用于构建含有本发明所述的启动子和/或目的基因序列的表达载体。这些方法包括体外重组DNA技术、DNA合成技术、体内重组技术等。表达载体还包括翻译起始用的核糖体结合位点和转录终止子。
此外,表达载体优选地包含一个或多个选择性标记基因,以提供用于选择转化的宿主细胞的表型性状,如二氢叶酸还原酶、新霉素抗性、潮霉素抗性以及绿色荧光蛋白(GFP)等。
重组载体中除了含有本发明的启动子,还可含有一种或多种其它启动子。所述的其它启动子例如是:组织特异性的、组成型的或诱导型的。例如甘露氨酸合成酶的花椰菜花叶病毒19S和35S(CaMV19S CaMV35S)、增强的CaMV、烟草RB7等。
包含上述适当的启动子和目的基因的载体,可以用于转化适当的宿主细胞,以使其能够表达蛋白质。
宿主细胞可以是原核细胞,如细菌细胞;或是低等真核细胞,如酵母细胞;或是高等真核细胞,如植物细胞。代表性例子有:大肠杆菌,链霉菌属、农杆菌;真菌细胞如酵母;植物细胞等。本领域一般技术人员都清楚如何选择适当的载体和宿主细胞。
用重组DNA转化宿主细胞可用本领域技术人员熟知的常规技术进行。当宿主为原核生物如大肠杆菌时,能吸收DNA的感受态细胞可在指数生长期后收获,用CaCl2法处理,所用的步骤在本领域众所周知。另一种方法是使用MgCl2。如果需要,转化也可用电穿孔的方法进行。当宿主是真核生物,可选用如下的DNA转染方法:磷酸钙共沉淀法,常规机械方法如显微注射、电穿孔、脂质体包装等。转化植物也可使用农杆菌转化或基因枪转化等方法,例如叶盘法、幼胚转化法、花芽浸泡法等。对于转化的植物细胞、组织或器官可以用常规方法再生成植株,从而获得转基因的植物。
例如,可通过叶圆片转化-再生程序(如Horsch等人(1985)Science 227:1229所述)将融合基因导入植物内。也可采用其它转化方法,如PEG法、基因枪法、电激法(Horsch等人,1984,Scienc 223:496,Barton等人,1983,Cell 32:1033,Liu等人,Acta Phytophysiol Sin,21:195-205),这也在本发明范围内。但是这些方法有操作复杂、外源基因插入的多拷贝性和不完整性以及较高比例的不育植株等缺点(Potrykus 1990,Bio/Technol,8:535-542;Finnegan和McElroy,1994,Bio/Technol.12:883-888)。
作为一种方式,制备转基因植物的方法是:将携带启动子和目的基因(两者可操作地连接)的双元载体转入农杆菌,农杆菌再将含启动子和目的基因的载体片段整合到植物的染色体上。涉及的转基因受体植物例如是水稻、小麦等。
在本发明的一个实施方式中,本发明人构建GIF1基因的启动子连β-葡糖醛酸酶基因(GUS)的克隆,并转化水稻ZH11,观察GIF1启动子启动GUS报告基因和GIF1基因的组织特异性表达。
本发明的主要优点
揭示一种可指导目的基因在植物中组织特异性表达的启动子,所述的启动子对于定点地改良植物品质特别有用。
实施例
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件,如分子克隆实验手册(Molecular cloning:Alaboratory manual,3rd ed.,Sambrook等,Cold Spring Harbor Laboratory,2001)和植物分子生物学实验手册(Plant Molecular Biology-A LaboratoryMannual,Clark等,Springer-Verlag,1997)中所述的条件,或按照制造厂商所建议的条件。
实施例1.GIF1基因的获得
本发明人从水稻中花11(ZH11)诱变突变体库中,发现了一个水稻突变体,该突变体的种子的灌浆受到较为严重的影响,营养生长和野生型相比没有变化,但籽粒灌浆受到严重影响,种子千粒重减少15-30%,并且米质变差,本发明人将之命名为gif1(grain incomplete filling 1),说明GIF1基因是一个重要的通过灌浆控制产量和米质的基因。
本发明人通过gif1突变体和珍汕97杂交得到基因定位群体。利用BSA(Bulked Segregant Analysis)分析法,以均匀分布在水稻12条染色体上的130对SSR引物对GIF1位点进行扫描,将GIF1位点初步定位在4号染色体长臂SRD5附近,最终将GIF1基因定位在caps-4和caps-8之间的32Kb中,经预测这段区域共包含三个基因,本发明人通过对突变体和野生型的DNA的测序,发现在突变体gif1中有一个碱基的缺失(GIF1基因组DNA序列第4588位)。
通过对GIF1的精细定位,通过测序和功能验证,得到野生型GIF1的基因组(DNA)序列见SEQ ID NO:3(包括启动子);GIF1编码区的序列(cDNA序列)见SEQ ID NO:1;GIF1蛋白的序列见SEQ ID NO:2。
当发生前述的突变(GIF1基因组DNA序列第4588位缺失)后,突变型不表达GIF1蛋白。
实施例2.GIF1启动子序列的获得
本发明人根据已经克隆的GIF1基因,在NCBI(http://www.ncbi.nlm.nih.gov/)数据库搜索该基因的基因组区域,经过生物软件预测(http://www.cbs.dtu.dk/services/Promoter/)启动子,最终选择了2379bp作为启动子序列。
实施例3.载体构建和农杆菌转化
本发明人构建GIF1基因的启动子连β-葡糖醛酸酶基因(GUS)的克隆,并转化水稻ZH11,观察GIF1启动子启动GUS报告基因和GIF1基因的组织特异性表达。
具体的构建方法如下:
采用以下引物:
正向引物tataagcttgatcggccatactcc(SEQ ID NO:4)和
反向引物taggatccctttgctctcacacttg(SEQ ID NO:5),
以GIF1的基因组DNA为模板,PCR得到GIF1基因的启动子(其序列如图1所示),并克隆载体pBI101(购自Clonetech公司,带有GUS),再用EcoR I、Hind III酶切,回收片断,将该片断连入用相同酶切的pCAMBIA1300便得到了pCAMBIA1300+启动子+GUS的相应克隆(组织显色方法见Jeferenson,RA(1987)Plant Mol Biol Rep)。
结果见图2A-K,可知GUS基因在植株的根部、节间、种子的背腹部的微管束有特异性表达。
此外,采用RT-PCR的方法,从所述的野生型或突变型植株中抽提mRNA,通过RT-PCR分别扩增GIF1,将获得的扩增产物进行琼脂糖电泳检测。
在该实验中,用来扩增GIF1的引物为:

用来扩增ubi-1的引物为:

结果见图3A-C,其中,图3A是突变体gif1和野生型的mRNA的RT-PCR检测结果;图3B是水稻不同组织中GIF1 mRNA的RT-PCR检测结果;图3C是不同时期(DAF)水稻穗部组织中GIF1 mRNA的RT-PCR检测结果。

Claims (12)
1.一种分离的多核苷酸,其选自下组:
(1)由SEQ ID NO:3的1-2379位所示的序列构成的核苷酸序列;
(2)具有SEQID NO:3中1-2379位所示的核苷酸序列且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(3)SEQ ID NO:3中1-2379位所示的核苷酸序列的片段,所述片段能启动目的基因在植物中组织特异性表达;
(4)在严格条件下能够与(1)、(2)或(3)限定的核苷酸序列杂交且能启动目的基因在植物中组织特异性表达的核苷酸序列;
(5)与(1)、(2)或(3)限定的核苷酸序列有70%、80%、85%、90%、95%以上相同性且能启动目的基因在植物中组织特异性表达的核苷酸序列;或
(6)与(1)(5)任一项所限定的核苷酸序列互补的核苷酸序列。
2.如权利要求1所述的多核苷酸,其特征在于,所述目的基因选自:编码具有SEQ ID NO:2氨基酸序列的作物籽粒灌浆蛋白、以及与作物淀粉转运、积累相关的基因。
3.如权利要求1所述的多核苷酸,其特征在于,特异性表达所述目的基因的组织是种子、根部和/或节间。
4.权利要求1所述的多核苷酸作为启动植物组织特异性地表达目的基因的启动子的用途。
5.一种载体,其特征在于,所述的载体含有权利要求1-3中任一项所述的多核苷酸,作为启动子元件。
6.如权利要求5所述的载体,其特征在于,所述的载体还含有与所述的核酸可操作地连接的目的基因。
7.如权利要求6所述的载体,其特征在于,所述的目的基因选自:编码具有SEQID NO:2氨基酸序列的作物籽粒灌浆蛋白,以及与作物淀粉转运、积累相关的基因。
8.一种遗传工程化的宿主细胞,其特征在于,所述的细胞:
含有权利要求5-7中任一项所述的载体;或
其基因组中整合有外源的权利要求1-3中任一项所述的多核苷酸。
9.一种制备转基因植物的方法,其特征在于,所述的方法包括:
(a)提供含有作为启动子元件的权利要求1-3中任一项所述的多核苷酸以及与所述多核苷酸可操作地连接的目的基因的构建物;
(b)将所述构建物导入植物细胞、组织或器官;
(c)选出导入了所述构建物或染色体中整合有所述构建物的植物细胞、组织或器官;和
(d)使所述植物细胞、组织或器官再生成植株。
10.用权利要求9所述的方法制得的转基因植物。
11.含有权利要求1-3中任一项所述的多核苷酸或权利要求5-7中任一项所述的载体的植物。
12.如权利要求11所述的植物,其特征在于,所述植物为作物,优选为禾本科植物,更优选所述的禾本科植物选自:水稻、小麦、大麦、玉米、或高粱。
