技术领域
[0001]本发明涉及抗炎剂,尤其涉及以2-氨基苯酚或其衍生物作为有效成分的抗炎剂。
背景技术
[0002]炎症反应是个体防御病原体的重要的个体反应之一,对个体组织也产生损伤,所以过量的炎症反应反而有害。尤其对于自体免疫疾病或过敏性疾病等不存在病原体的炎症反应完全有害。因此,为抑制炎症反应开发出了多种多样的抗炎剂,例如阿司匹林、双氯芬酸、吲哚美辛、甲灭酸等非甾类抗炎剂,泼尼松龙、醋酸氢化可的松、二氟泼尼酯等甾类抗炎剂,这些均被用作药品。
[0003]炎症反应的主要症状是“痛”和“肿”。伴随炎症反应的“痛”由前列腺素引起。前列腺素的生成必需有环氧化酶(COX),所以其阻遏剂(COX阻遏剂)被用作发挥镇痛效果的抗炎剂。COX是催化由花生四烯酸生成前列腺素的酶,存在两种同种型,其中COX-1在消化道、肾、血小板等中组成型表达,是维持正常生理功能中必不可少的酶。而COX-2由白细胞介素-1α、肿瘤坏死因子α(TNF-α)等炎性细胞因子一次性诱导产生,是在炎症时过量表达的酶,被报道与风湿、关节炎等炎性疾病或癌症、胃溃疡、阿尔茨海默病、排卵分娩相关。作为抗炎剂的COX阻遏剂是以阻遏COX-2活性为目的,而由于多种抗炎剂也阻遏COX-1活性,所以产生腹痛等副作用。因此以欧美为中心开发出了COX-2选择性阻遏剂,有望成为胃肠障碍或肾障碍等副作用少的抗炎剂。但根据シンムラ等著,“心血管系におけるシクロオキシゲナ—ゼ(COX)-2は敵か,味方か?-COX-2ハザ—ドの分子メカニズム-”,インフイラメ—ション·アンド·レジェネレ—ション,第25卷、第6号、第517至524页、2005年中报道,施用了COX-2选择性阻遏剂罗非昔布的结直肠息肉患者发生心肌梗塞等心血管疾病的概率高,从而使COX-2选择性阻遏剂的安全性受到质疑。所以在日本至今未得到药品认可。
[0004]而伴随炎症的“肿”是病灶血管扩张,白细胞、淋巴细胞、巨噬细胞等免疫活性细胞局部聚集而引发的。血管扩张受一氧化氮(NO)诱导,因此为缓解“肿”,可使用阻遏NO产生的制剂。NO由一氧化氮合成酶(NOS)氧化L-精氨酸而产生。存在非衍生型和衍生型2种NOS,其中衍生型NOS(iNOS)大量存在于巨噬细胞、内皮细胞、平滑肌细胞等,是炎症反应中的重要因子。即,为抑制NO的产生,iNOS活性阻遏剂或抑制iNOS活性的制剂是有效的,所述iNOS活性阻遏剂如NG-硝基-L-精氨酸-甲基酯、异硫脲衍生物、2-亚氨基哌啶、L-刀豆素等。
[0005]已报导在多数炎症疾病中COX-2和iNOS同时过量表达,因此认为炎症反应与所述2种酶的活性大小相关。因此,为有效缓解炎症症状,有必要同时抑制引起“痛”和“肿”的COX-2和iNOS的活性。然而,已知COX阻遏剂或NOS阻遏剂的酶特异性高,仅可阻遏COX或iNOS各自的活性。因此有待开发可同时缓解“痛”和“肿”的抗炎剂。
[0006]而2-氨基苯酚(别名:寻霉素B)是下式1所示化合物,较易经氧化聚合而转化为下式2所示2-氨基吩噁嗪-3-酮(别名:寻霉素A)。已知2-氨基苯酚或其衍生物是具有抗菌活性的物质,尤其已知2-氨基吩噁嗪-3-酮是作为强力抗癌剂的放线菌素D的基本骨架。モトハシ等著、『ポテンシヤル·アンチツモア·フェノキサジンズ』、メデイカル·リサ—チ·レビユ—、第11卷、第250乃至254页、1991年中公开了2-氨基吩噁嗪-3-酮及其衍生物具有抗肿瘤活性,而シマモト等著、『アンチツモア·エフェクツ·オブ·ア·ノベル·フェノキサジン·デリバティブ·オン·ヒユ—マン·リユ—ケミア·セル·ラインズ·イン·ビトロ·アンド·イン·ビボ』、クリニカル·キヤンサ—·リサ—チ、第7卷、第704乃至708页、2001年中尤其公开了作为其衍生物之一的2-氨基-4,4α-二氢-4α,7-二甲基-吩噁嗪-3-酮对各种肿瘤细胞表现出细胞损伤活性。近年来,特开2004-143101号公报中公开了2-氨基吩噁嗪-3-酮可有效治疗病毒性疾病,特开2005-272334号公报中公开了2-氨基吩噁嗪-3-酮可有效治疗衣原体病,特开2005-60325号公报中公开了2-氨基吩噁嗪-3-酮可有效治疗螺旋菌属相关的消化器官疾病。但还不知2-氨基苯酚衍生物具有抗炎作用。

发明内容
[0009]本发明旨在提供较之前抗炎剂副作用小,改善“痛”和“肿”等炎症症状的效果高的抗炎剂。
[0010]发明人经锐意研究发现,2-氨基苯酚或其衍生物具有通过阻遏环氧化酶(COX)活性而阻遏前列腺素E2合成的作用,通过抑制iNOS生成而阻遏由巨噬细胞合成一氧化氮的作用,及具有抑制肥大细胞的脱粒反应的作用。另外发现2-氨基苯酚或其衍生物具有抑制黑色素细胞的黑色素合成,及增强胶原产生的作用。
[0011]即,本发明通过提供含有2-氨基苯酚或其衍生物的抗炎剂解决上述课题。

具体实施方式
[0013]本发明所述2-氨基苯酚指下式1所示化合物,只要能达到本发明的效果,则不问其来源,可使用市售品。本发明所述2-氨基苯酚的衍生物是2-氨基苯酚的氧化聚合物,指下式2所示2-氨基吩噁嗪-3-酮及以其为基本骨架的衍生物,发挥等于或优于2-氨基苯酚的效果。2-氨基吩噁嗪-3-酮可通过合适的提取、分离方法从富含其的植物、细菌等中制备,也可将其前体2-氨基苯酚经氧化聚合而制成。例如,可通过特开2003-2878号公报记载的使2-氨基苯酚与3价铁离子(铁氰化钾等)反应的方法或特开平2-193984号公报记载的使2-氨基苯酚与人或牛的血红蛋白反应的方法进行合成。

[0021]如上所述,因为本发明所用2-氨基苯酚或其衍生物具有阻遏一氧化氮合成、阻遏环氧化酶合成或抑制嗜碱性粒细胞脱粒的作用,可作为抗炎剂、抗过敏剂、抗特应剂应用于各种食品、化妆品、准药品、医药品。另外,也因为具有促进向T淋巴细胞——Th2细胞分化的作用而被用作诸如银屑病或风湿等自体免疫疾病的预防剂和治疗剂。又因为具有阻遏黑色素合成或增强胶原产生的作用而取皮肤外用剂的形式时,可用作具有抗衰老作用的美白用和/或美肤用化妆品。本发明取皮肤外用剂的形式时,作为抗炎剂所含有效成分的2-氨基苯酚或其衍生物的含量通常为0.00002-1%(w/w),优选0.0001-0.5%(w/w),使用量可根据皮肤的症状适当选择,可每日1次,也可分成每日数次,每次使用量通常为0.1μg-10mg/1cm2皮肤,优选1μg-5mg/1cm2皮肤。

[0031]实验1:2-氨基吩噁嗪-3-酮的合成
以2-氨基苯酚为原料合成了2-氨基吩噁嗪-3-酮。将550mg(5mmol)2-氨基苯酚(和光纯药工业株式会社售)悬浮于50ml蒸馏水,并向其中加入0.1N盐酸225ml后,用0.1N氢氧化钠水溶液调至pH7.0。在搅拌下向所述溶液中经5分钟滴加500ml 5mM铁氰化钾水溶液,再于26℃下反应30分钟。将经减压干燥反应液所得固体溶于450ml甲醇,通过离心除去未溶解的固体后,再进行减压干燥。将所得固体溶于200ml乙酸乙酯中,并加入200ml 0.005N盐酸水溶液,进行搅拌后,用分液漏斗回收乙酸乙酯层。重复此操作3次后,将所得乙酸乙酯层在减压浓缩至15ml体积,将其根据常规方法,用硅胶层析柱(商品名“ワコ—ゲルC200”,和光纯药工业株式会社售)和逆相C30层析柱(商品名“デベロシルC30”,野村化学株式会社售,或商品名“HW-40F”,东ソ一株式会社售)进行纯化而得到220mg 2-氨基吩噁嗪-3-酮。
[0032]实验2:抑制巨噬细胞产生NO、PGE2的活性
将小鼠巨噬细胞株RAW264.7悬浮于补充了10%(v/v)胎牛血清(以下简称FCS)的RPMI1640培养基中至细胞浓度达1×106个/ml,而制成细胞悬液。将其以50μl等份接种于96孔微平板中,加入2-氨基吩噁嗪-3-酮至终浓度1.6-100μM,再加入2μg/ml脂多糖和10IU/mlIFN-γ作为诱导剂,加入上述培养基至200μl体积。将其于37℃下培养2天后进行活细胞计数,对培养上清液通过常规方法——Griess法测定NO量,另外通过使用抗PGE2抗体的“PGE2EIA试剂盒”(アマシヤムバイオサイエンス公司售)测定PGE2量。作为比较例,使用作为COX阻遏剂的吲哚美辛(和光纯药工业株式会社售)、阿司匹林(和光纯药工业株式会社售),作为COX-2选择性阻遏剂的NS-398(和光纯药工业株式会社售)同上述进行了实验。另外将未加入上述样品的体系设为对照,结果显示于表1。表1中的“QA”表示2-氨基吩噁嗪-3-酮(寻霉素A)、“NS”表示NS-398、“IN”表示吲哚美辛、“AS”表示阿司匹林,显示了各样品的NO产生量、PGE2产生量、巨噬细胞增殖相比对照的测定值的相对值。

[0034]如表1的结果所示,2-氨基吩噁嗪-3-酮(表1中的“QA”)不仅降低巨噬细胞的增殖,还抑制NO的产生和PGE2的产生。而比较例中的其他抗炎剂尽管抑制PGE2产生的效果显著,但却不怎么发挥抑制NO产生的效果,表明对现有抗炎剂尽管可期待其改善“痛”的效果,但无法期待其改善“肿”的效果。
另外,在使用iNOS抗体的蛋白印记分析研究可通过本实验得到的各细胞提取液中所含iNOS的量时发现,iNOS的量随着2-氨基吩噁嗪-3-酮以浓度依赖的方式减少。因此认为2-氨基吩噁嗪-3-酮通过减少iNOS量来发挥其阻遏NO产生的活性。
[0035]实验3:对COX-1和COX-2阻遏活性
为验证上述实验2的结果,研究了PGE2的合成体系中必要的酶COX的酶活性是否被阻遏。使用“COXインヒビタ—スクリ—ニングアツセイキツト”(ケイマン公司售),在存在COX-1或COX-2和作为基质的花生四烯酸的反应体系中如下表2所示浓度加入作为实验样品的2-氨基吩噁嗪-3-酮或2-氨基苯酚,通过使用抗PGE2抗体的EIA测定生成的PGE2的量,作为了COX的活性量。设置作为对照的未加入实验样品的体系,通过与所述测定值进行比较来求出COX-1或COX-2的残留活性率。用残留活性率对实验样品的各浓度作图,酸楚IC50值,结果显示于表2。

[0037]表2的结果表明,2-氨基苯酚和2-氨基吩噁嗪-3-酮阻遏COX-1和COX-2活性。用IC50值比较所述效果,则摩尔比为4-6倍、质量比为2-3倍左右,2-氨基吩噁嗪-3-酮更优。另外,COX-1/COX-2比是2-氨基苯酚约为1,2-氨基吩噁嗪-3-酮约为1.3,也比阿司匹林(COX-1/COX-2比=0.24)或吲哚美辛(COX-1/COX-2比=0.03)等非甾类COX阻遏剂的COX-2选择性高,当把2-氨基苯酚或2-氨基吩噁嗪-3-酮作为口服药施用于患者时,可期待胃痛等副作用较现有非甾类COX阻遏剂变轻,所以可用作抗炎剂、解热剂、镇痛剂等口服用医药品。
[0038]实验4:抑制嗜碱性粒细胞脱颗粒反应的活性
因为已证实2-氨基吩噁嗪-3-酮(寻霉素A)阻遏抗炎性介质从巨噬细胞游离,因此研究了对已知同样游离炎性介质的嗜碱性粒细胞的脱颗粒反应的影响。基于对通过脱颗粒反应排出细胞外的颗粒中所含β-氨基己糖苷酶活性的测定来计算出脱颗粒率(%)和脱颗粒抑制率(%)。
脱颗粒反应测定中使用的细胞
将源自已知通过IgE交联而引起脱颗粒反应的大鼠嗜碱性粒细胞的癌细胞RBL-2H3(ヒユ—マンサイエンス振兴财团·研究资源バンク售、目录号“J C R B 0023”)在补充有10%(v/v)FCS的MEM培养基(日水制药公司售)中培养并使用。
脱颗粒反应的诱发及β-氨基己糖苷酶活性的测定
将所述细胞根据常规方法用胰蛋白酶-EDTA处理并从培养皿回收,再悬浮于补充有10%(v/v)FCS的MEM培养基中至细胞浓度达5×105个/ml,而制成细胞悬液。将其以400μl等份接种于24孔微平板中,在5%(v/v)二氧化碳培养箱中培养3-5小时后,以100μl/孔加入0.625μg/ml的抗二硝基酚(以下简称DNP)的小鼠IgE(Sigma-Aldrich公司售,用补充有10%(v/v)FCS的MEM培养基稀释)而用IgE致敏细胞(抗DNP-IgE终浓度0.125μg/ml)。在5%(v/v)二氧化碳培养箱中过夜培养所述细胞后,吸去培养上清液,等份加入500μl/孔Siraganian缓冲液(119mM NaCl、5mM KCl、0.4mM MgCl2、25mM哌嗪-N,N”-双(2-乙磺酸)(PIPES)、40mM NaOH pH7.2)来洗净各孔2次。等份加入160μl/孔含0.1%牛血清白蛋白(BSA)、5.6mM葡萄糖、1mM CaCl2的Siraganian缓冲液(以下简称“含BSA的Siraganian缓冲液”),于37℃加热15分钟。用含BSA的Siraganian缓冲液稀释作为被检样品的2-氨基吩噁嗪-3-酮(寻霉素A)、具有美白作用的人血清白蛋白(アルブチン)或过敏性疾病治疗剂——奥沙米特(阳性对照)中的任一种,至达表3所示浓度,等份加入20μl/孔其中的任一种,于37℃加热15分钟(被检样品加入孔)。向加入所述被检样品中任一种的被检样品加入孔中等份加入20μl/孔用含BSA的Siraganian缓冲液稀释至50μg/ml的DNP-白蛋白(Sigma-Aldrich公司售),于37℃加热15分钟。在冰上冷却此24孔平板10分钟后,取上清,将其100μl分注于96孔微平板,根据下述方法测定所述上清液中β-氨基己糖苷酶的活性。另外,与此不同,作为阴性对照,除了对BL-2H3细胞使用了不含抗体、未补充FCS的MEM培养基之外,在与被检样品加入孔相同的条件下培养、处理,测定了所述上清液中β-氨基己糖苷酶的活性(IgE非致敏孔)。另外,吸去所述IgE非致敏孔的一部分的培养上清液后,加入含BSA的Siraganian缓冲液,洗净各孔2次后,等份加入200μl/孔含BSA的Siraganian缓冲液,于-80℃重复2此冻结、解冻,破碎细胞,回收含所述破碎细胞的全液,以1500rpm离心分离10分钟后,将其上清100μl分注于96孔微平板中,将其作为全颗粒提取物而测定β-氨基己糖苷酶活性。
β-氨基己糖苷酶活性测定法
向加入了取出的被检样品加入孔上清、IgE非致敏孔上清或全颗粒提取物的96孔微平板的各孔中,等份加入50μl含作为基质的1mM 4-硝基苯基-N-乙基-β-D-氨基葡萄糖苷(PNAG)的柠檬酸缓冲液(pH4.5),于37℃保持1小时,等份加入0.1M碳酸钠(pH10.5)50μl来中止反应,作为被检样品加入孔、IgE非致敏孔或全颗粒提取物孔的吸光度,测定了A405-650吸光度,各自根据下公式1和2求出脱颗粒率(%)和脱颗粒抑制率(%)。加入各被检样品时对脱颗粒反应的抑制程度用对脱颗粒的抑制作用“强”(抑制率为50%以上)、“弱”(抑制率为小于50%-30%以上)、“无”(抑制率为小于30%-30%以上)、“促进”(抑制率小于30%)的4个等级评价,其结果显示于表3。表3中“QA”表示2-氨基吩噁嗪-3-酮(寻霉素A)、“AB”表示人血清白蛋白,而“OX”表示奥沙米特。

[0041]因为本实验中以β-氨基己糖苷酶活性为指标求了脱颗粒率,根据以下方法确认了发现具有强β-氨基己糖苷酶活性抑制的2-氨基吩噁嗪-3-酮(寻霉素A)无对β-氨基己糖苷酶活性的直接抑制作用。即在上述实验中使用的最高浓度的2-氨基吩噁嗪-3-酮(寻霉素A)用作被检样品,为与用于测定对脱颗粒反应的抑制时的浓度相同,将预先用含BSA的Siraganian缓冲液稀释5倍之物以50μl/孔分注于96孔微平板中。与此不同,将上述全颗粒提取样品以50μl/孔分注于96孔微平板中。向所述各孔中等份加入50μl/孔含1mM PNAG的0.1M柠檬酸缓冲液(pH4.5),于37℃加热1小时。等份加入50μl/孔0.1M碳酸钠(pH10.5)中止反应,测定了A405-650吸光度。仅加入含BSA的Siraganian缓冲液作为对照,同样进行测定。根据以下公式3求出对β-氨基己糖苷酶活性的阻遏率(%)。用被检样品和全颗粒提取样品各3孔进行测定,求出其平均值。

[0044]表3的结果表明,2-氨基吩噁嗪-3-酮浓度在5μM以上时才能检测到其脱颗粒抑制活性,在10-40μM时可检测到其对脱颗粒反应的强抑制活性。阳性对照奥沙米特在23μM以上时才能检测到其脱颗粒抑制活性,在94μM时可检测到其强抑制活性。检测不到人血清白蛋白的脱颗粒抑制活性。另外,加入40μM的2-氨基吩噁嗪-3-酮时的β-氨基己糖苷酶活性阻遏率达9.9%,将用作溶剂的DMSO用于测定的培养基中的浓度时达8.6%,因已证实所述物质本身或DMSO不阻遏所述酶的活性,明确了所述物质抑制嗜碱性粒细胞的脱颗粒反应本身。此结果提示,由于2-氨基苯酚或2-氨基苯酚衍生物(2-氨基吩噁嗪-3-酮等)抑制嗜碱性粒细胞或肥大细胞等的脱颗粒反应,因此可用作抗过敏剂。另外还提示,因为认识到也抑制脱颗粒反应引发的有关炎性的介质排出,2-氨基苯酚或2-氨基苯酚衍生物(2-氨基吩噁嗪-3-酮等)不仅抑制经由巨噬细胞的炎症反应,也可抑制经由嗜碱性粒细胞或肥大细胞的炎症反应。虽然未显示具体数据,检测不到用作实验样品的溶剂的DMSO在本实验的培养条件下具有对测定结果有影响的细胞障碍。
[0045]实验5:2-氨基吩噁嗪-3-酮或2-氨基苯酚对黑色素合成的阻遏效果
将小鼠黑色素瘤细胞株B16细胞(ATCC:CRL-6322)悬浮于补充有10%(v/v)FCS的RPMI1640培养基中,再将其以每孔2.0×104个细胞接种于6孔平板中。所述细胞贴壁后,加入2-氨基吩噁嗪-3-酮至浓度达0.5μM、1μM、2μM,加入2-氨基苯酚至浓度达1μM、2μM、4μM,另外加入作为阳性对照的曲酸至浓度达0.5mM、1mM、2mM,根据常规方法,在5%CO2气氛中于37℃下培养5天。另外,作为对照,使用了未加入样品之物。培养后回收细胞,对各种活细胞进行计数后,用磷酸缓冲液洗净,用1N氢氧化钠水溶液溶解,煮沸30分钟,根据常规方法,用市售的读板仪(plate reader)测定450nm波长下的吸光度,根据预先用黑色素标准品(Sigma公司售)在同一波长下测定吸光度而制成的标准曲线算出样品中的黑色素量。根据常规方法Bradford法测定蛋白质量,算出每1mg蛋白质中的黑色素量,再根据公式4算出黑色素量(%)再根据公式5算出活细胞数(%),结果显示于表4。

通过小组评定研究了本发明的皮肤外用剂是否对已晒黑的皮肤发挥抗炎作用和阻遏黑色素合成的效果。给20人的评定小组成员的腕部贴上约10cm2的遮光胶带,以防止日光照射,在实施海水浴半天后,揭下所述遮光胶带,将所述皮肤部分作为晒黑前的状态。将适当量的配含2-氨基吩噁嗪-3-酮(样品A)或2-氨基苯酚(样品B)的下述实施例1的乳液或下述实施例1中作为对照的未配含2-氨基吩噁嗪-3-酮或2-氨基苯酚的乳液分别涂于照射日光的腕部皮肤上,每日2次。由医生肉眼观察3日后的皮肤炎症状态(有无痛、肿),7日后的黑色素沉积状态(皮肤黑度),以“有效果”(恢复到晒黑前的状态)、“多少有效果”(虽不能使恢复到晒黑前的状态,但比对照乳液效果佳)、“无效果”(与对照乳液程度相同)、“恶化”(比对照乳液恶化)的4个等级进行评价,结果显示于表6。

通过小组评定研究了本发明的抗炎剂是否对牙龈炎发挥治疗效果。将仅通过每次餐后刷牙而症状得不到改善的牙龈炎患者18人分成3组,在每次餐后刷牙后用配含2-氨基吩噁嗪-3-酮(样品A)或2-氨基苯酚(样品B)的下述实施例4的液剂10m1漱口1分钟。另外,用未配含2-氨基吩噁嗪-3-酮或2-氨基苯酚的液剂作为对照同样进行实验。2周后,由医生通过目测诊断牙龈炎状况,以“有效果”(恢复到无牙龈炎的状态)、“多少有效果”(牙龈炎较实验前缓和)、“无效果”(相比实验前无变化)、“恶化”(比实验前还恶化)的4个等级进行评价,结果显示于表7。

通过小组评定研究了本发明的抗炎剂是否对胃炎发挥治疗效果。将胃炎患者12人分成3组,在每次晚餐后服用配含2-氨基吩噁嗪-3-酮(样品A)或2-氨基苯酚(样品B)的下述实施例5的锭剂。另外,用未配含2-氨基吩噁嗪-3-酮或2-氨基苯酚的锭剂作为对照同样进行实验。1周后,由医生通过问诊诊断胃炎状况,以“有效果”(恢复到无胃炎的状态)、“多少有效果”(胃炎较实验前缓和)、“无效果”(相比实验前无变化)、“恶化”(比实验前还恶化)的4个等级进行评价,结果显示于表8。

[0061]如表8所示,样品A和样品B发挥了治疗胃炎的效果。实验7和实验8的结果显示,2-氨基吩噁嗪-3-酮或2-氨基苯酚作为诸如牙龈炎或胃炎等炎性疾病的治疗剂有效。
[0062]实验10:增强胶原产生的作用
在抗坏血酸存在下,使用源自人胎儿的正常成纤维细胞NHDF研究了2-氨基吩噁嗪-3-酮对增强胶原产生的作用。另外,一并研究了2-氨基吩噁嗪-3-酮是否单独发挥增强胶原产生的作用。又研究了2-氨基吩噁嗪-3-酮对NHDF细胞无细胞障碍。
实验材料
用补充有10(v/v)%FCS的Dulbecco氏MEM培养基(日水制药公司售,以下简称“D-MEM”)稀释4mM 2-氨基吩噁嗪-3-酮(溶于DMSO),在将其用于细胞培养时的培养基中的浓度如表9所示配制实验样品。
胶原产生量的测定方法
将NHDF细胞(クラボウ公司,目录号“KF-4001”)悬浮于补充有10(v/v)%FCS的D-MEM中制成5×105细胞/ml,以50μl/孔等份接种于96孔微平板中。作为抗坏血酸,使用了在培养基中的稳定性比抗坏血酸高的L-抗坏血酸-2-葡糖苷(株式会社林原商事售,商品名“アスコフレツシユ”,以下简称“AA-2G”)。在37℃培养1天后,除去培养上清液,以各100μl/孔等份加入实验样品中的任一种与含100μM AA-2G(终浓度50μM)的补充有10(v/v)%FCS的D-MEM培养基或不含AA-2G的补充有10(v/v)%FCS的D-MEM培养基,于37℃,5(v/v)%CO2存在下培养3天后,除去培养上清液,新配制与最初加入的物质相同浓度的被检样品与含100μM AA-2G或不含AA-2G的D-MEM培养基,以各100μl/孔等份加入,于37℃,5(v/v)%CO2存在下再培养3天。作为对照,仅加入了补充有10(v/v)%FCS的D-MEM培养基或含AA-2G的补充有10(v/v)%FCS的D-MEM培养基,与加入实验样品的情况同样进行培养。将所有细胞培养液均调成用于溶解2-氨基吩噁嗪-3-酮的DMSO的浓度为定值(终浓度0.05(v/v)%)。除去这些细胞的培养上清液后,以50μl/孔等份加入含1mg/ml胃蛋白酶(Sigma公司售)的1M醋酸溶液,使用平板混合器于室温在振摇下用4小时胃蛋白酶进行消化。将此胃蛋白酶消化物边用移液器反复抽打边回收到1.5ml容积的离心管中,以200μl/管加入Sircol胶原测定试剂盒(Biocolor公司售)的DyeReagnt后,于室温下颠倒混匀30分钟进行反应,离心反应液(4℃,15,000rpm,10分钟)后,完全除去上清液,向沉淀加入50μl 1N NaOH进行溶解后,测定A560-650的吸光度。与此不同,向96孔微孔平板加入经2倍稀释的胶原(KOKEN公司售)至5μg/孔-0.0395μg/孔,用胰蛋白酶处理,与除去培养上清液的细胞同样处理所述消化物,并测定A560-650的吸光度,从而绘制标准曲线。根据所述标准曲线算出细胞的胰蛋白酶消化物中所含胶原量,其结果显示于表9。每个样品浓度以3个平行孔进行实验,结果用其平均值表示。表9中的“QA”表示2-氨基吩噁嗪-3-酮(寻霉素A)。
细胞障碍实验
与上述胶原测定同样,将在实验样品和含或不含AA-2G的补充有10(v/v)%FCS的D-MEM培养基中培养6天的细胞的培养上清液除去后,用磷酸缓冲生理盐水(PBS(-))洗净,以200μl/孔加入用D-MEM培养基稀释10倍的alamarBlue液(TREK Diagnostic Systems公司售),于37℃培养90分钟后,测定荧光强度(激发光:544nm,荧光:590nm)。通过用加入实验样品的孔的荧光强度除于加入对照的孔的荧光强度并乘以100求出细胞生存率(%)。其结果显示于表9。

[0064]如表9所示,在培养基中加入2-氨基吩噁嗪-3-酮和AA-2G进行培养时,胶原产生量以与至0.125或0.25μM的2-氨基吩噁嗪-3-酮的加入量成比例增加,在加入至0.5和0.25μM时,产生几乎等量的胶原。另外,在加入1μM以上时,相比加入0.5或0.25μM的情况,胶原产生量减少。与此相比,在未加入AA-2G时,未检测到胶原产生增强。另外,2-氨基吩噁嗪-3-酮的浓度在0.5μM以下时,无论何种情况细胞生存率均达92%以上,与对照仅有8%左右的差异,判断为无细胞障碍;而与此相比,1μM以上的浓度可检测到依赖于浓度的细胞障碍。因此认为,2-氨基吩噁嗪-3-酮的浓度在1μM以上时,发生伴随对NHDF细胞的细胞障碍的增强胶原产生的作用的下降。
[0065]此结果表明,2-氨基苯酚或2-氨基苯酚的衍生物(2-氨基吩噁嗪-3-酮等)在抗坏血酸存在下,在0.125μM的浓度下增强胶原的产生,其效果在0.25μM的浓度时达到显著。另外还认识到,含2-氨基苯酚或其衍生物(2-氨基吩噁嗪-3-酮等)的皮肤外用剂在增强胶原产生中表现优,不仅用于抗炎或美白,还可用于抗皱或抗衰老。

根据常规方法均匀加热混合A成分后冷却,并加入B成分得到皮肤外用剂。
本产品抗炎效果优,具有抑制嗜碱性粒细胞或肥大细胞的脱颗粒反应的作用,因此在特应性症的治疗中也有效。另外,本产品阻遏黑色素合成,可增强胶原的产生,通过适用于皮肤可抑制晒黑所致的皮炎、灼伤、斑点、雀斑、皱纹、小皱纹的发生,所以是至具有透明感的美丽皮肤的乳液。

混合成分A,在70℃加热溶解,混合成分B并溶解。冷却后均匀分散成分C,得到盒装化妆材料。
本产品抗炎效果优,且具有抑制嗜碱性粒细胞或肥大细胞的脱颗粒反应的作用,因此在特应性症的治疗上有效。另外,本产品阻遏黑色素合成,可增强胶原的产生,通过适用于皮肤可抑制晒黑所致的皮炎、灼伤、斑点、雀斑、皱纹、小皱纹的发生,所以是至具有透明感的美丽皮肤的盒装化妆材料。
实施例3

本产品抗炎效果优,具有增强胶原产生的作用,因此可减轻牙龈炎,适用于治疗或预防牙周病、牙槽脓溢。
实施例4

作为稳定剂,向含1%(w/w)海藻糖的生理盐水中溶解2-氨基吩噁嗪-3-酮至1μg/ml,或溶解2-氨基苯酚至0.1μg/ml,根据常规方法通过精密过滤除菌得到液剂。
本产品具有COX阻遏活性,阻遏一氧化氮合成的活性和抑制脱颗粒的活性,因此可作为内服液、注射液、漱口药被用作诸如过敏(包括风湿、牙周病、骨质疏松症、胃炎、特应性症)等各种炎性疾病,癌症或阿尔茨海默病的治疗或预防剂、镇痛剂、解热剂等。
实施例5
[0071]锭剂
向无水结晶α-麦芽糖粉末(商品名“ファイント—ス”,株式会社林原商事售)均匀混合2-氨基吩噁嗪-3-酮或2-氨基苯酚,根据常规方法压制,得到每200mg的锭中含1μg 2-氨基吩噁嗪-3-酮或0.1μg 2-氨基苯酚的锭剂。
本产品具有COX阻遏活性,阻遏一氧化氮合成的活性和抑制脱颗粒的活性,因此可经口被用作诸如过敏(包括风湿、牙周病、骨质疏松症、胃炎、特应性症)等各种炎性疾病,癌症或阿尔茨海默病的治疗或预防剂、镇痛剂、解热剂等。
工业实用性
[0072]如述,2-氨基苯酚或其衍生物具有阻遏NO合成的作用,阻遏PGE2合成的作用及抑制脱颗粒反应的作用,因此可被用作抗炎剂、抗过敏剂等。另外,因为还具有阻遏黑色素合成及增强胶原产生的作用,可用作旨在美白、抗皱、抗衰老等的皮肤外用剂。
Claims (4)
1.含有2-氨基苯酚或其衍生物作为有效成分的抗炎剂。
2.权利要求1的抗炎剂,其具有阻遏一氧化氮合成、阻遏环氧化酶活性作用及抑制脱粒反应的作用。
3.权利要求1或2的抗炎剂,其特征在于是呈美白用皮肤外用剂的形式和/或抗皱用皮肤外用剂的形式。
4.权利要求1-3中任一项的抗炎剂,其特征在于所述2-氨基苯酚衍生物是2-氨基吩噁嗪-3-酮。
