本发明涉及化合物含量测定方法技术领域,具体涉及蒺藜提取物中呋甾皂苷含量的测定方法。本发明要提供一种蒺藜提取物中呋甾皂苷含量的测定方法,以克服现有技术存在的测量数值不能直观地反映蒺藜提取物中呋甾皂苷含量的问题。为了克服现有技术存在的问题,本发明的技术解决方案是:一种蒺藜提取物中呋甾皂苷含量的测定方法,取待测样品,加甲醇进行超声处理,放冷后加甲醇,作为供试品溶液;另取恒重的蒺藜呋甾皂苷化合物对照品,加甲醇作为对照品溶液;照分光光度法(《中国药典》2000年版一部附录VB)于516nm波长处测定吸收度,计算,即得。

技术领域:
本发明涉及化合物含量测定方法技术领域,具体涉及蒺藜提取物中呋甾皂苷含量的测定方法。
背景技术:
根据研究资料及文献资料表明,蒺藜总皂苷有治疗心脑血管疾病的作用,是蒺藜草的主要有效成分。由于蒺藜总皂苷中呋甾皂苷含量较低,水溶性较大,不易结晶,经药理试验及药效实验证明对抗心脑血管有明显的预防和治疗作用,因此对蒺藜甾体皂苷进行准确的含量控制是一项重要的工作。目前现有测定方法是用CoCl2作为参照物,比较颜色的深浅来判断呋甾皂苷的含量。其存在的问题是:测量数值不能直观地反映蒺藜提取物中呋甾皂苷含量,对于蒺藜总皂苷在治疗心脑血管疾病药物中的应用不具备指导意义。
发明内容:
本发明要提供一种蒺藜提取物中呋甾皂苷含量的测定方法,以克服现有技术存在的测量数值不能直观地反映蒺藜提取物中呋甾皂苷含量的问题。
为了克服现有技术存在的问题,本发明的技术解决方案是:一种蒺藜提取物中呋甾皂苷含量的测定方法,取待测样品,精密称定,加甲醇进行超声处理,放冷后加甲醇至刻度,摇匀,作为供试品溶液;另取恒重的蒺藜呋甾皂苷化合物对照品,精密称定,加甲醇溶解、摇匀,作为对照品溶液;分别精密吸取供试品溶液和对照品溶液,精密加改良对二甲氨基苯甲醛溶液,加甲醇至刻度,密塞,摇匀,水浴后迅速冷却至室温,以甲醇代替供试品溶液同样处理的溶液为空白,照分光光度法(《中国药典》2000年版一部附录VB)于516nm波长处测定吸收度,计算,即得;

本发明中,我们以蒺藜总皂苷中呋甾皂苷为主要控制指标,选用呋甾环专属性颜色反应(与盐酸对二甲氨基苯甲醛溶液显红色),以蒺藜呋甾皂苷化合物为对照品,制定了蒺藜呋甾皂苷的含量测定方法。我们通过研究发现该方法显色稳定,能够准确、直观地测定样品中呋甾皂苷的含量,对于蒺藜总皂苷在治疗心脑血管疾病药物中的应用具有指导意义。
具体实施方式:
下面将通过具体实施例对本发明做详细的说明。
一种蒺藜提取物中呋甾皂苷含量的测定方法:
取待测样品约30mg,精密称定,置25ml量瓶中,加甲醇20ml超声处理(功率250W,频率50kHz)20分钟,放冷后加甲醇至刻度,摇匀,作为供试品溶液。另取60℃减压干燥至恒重的蒺藜呋甾皂苷化合物对照品约20mg,精密称定,置25ml量瓶中,加甲醇使溶解并稀释至刻度,摇匀,作为对照品溶液。分别精密吸取供试品溶液和对照品溶液各2ml,置10ml量瓶中,精密加改良对二甲氨基苯甲醛溶液5ml,加甲醇至刻度,密塞,摇匀,置58±2℃水浴中加热2小时,迅速冷却至室温,以甲醇代替供试品溶液同样处理的溶液为空白,照分光光度法(《中国药典》2000年版一部附录VB)于516nm波长处测定吸收度,计算,即得。

1、一种蒺藜提取物中呋甾皂苷含量的测定方法,包括下述步骤:取待测样品,精密称定,加甲醇进行超声处理,放冷后加甲醇至刻度,摇匀,作为供试品溶液;另取恒重的蒺藜呋甾皂苷化合物对照品,精密称定,加甲醇溶解、摇匀,作为对照品溶液;分别精密吸取供试品溶液和对照品溶液,精密加改良对二甲氨基苯甲醛溶液,加甲醇至刻度,密塞,摇匀,水浴后迅速冷却至室温,以甲醇代替供试品溶液同样处理的溶液为空白,照分光光度法(《中国药典》2000年版一部附录VB)于516nm波长处测定吸收度,计算,即得;
