发明背景

聚合矿物质表面活性剂包括对牙齿表面具有强亲和力、在牙齿表面沉积成聚合物层或覆盖层并且产生所需表面改性效果的任何试剂。这类聚合物的合适的实例为高分子电解质,例如缩聚磷酸化聚合物、聚膦酸酯、含磷酸根-或膦酸酯的单体或聚合物与如下物质的共聚物:乙烯基化的未取代单体和氨基酸或其它聚合物,例如蛋白质、多肽、多糖、聚丙烯酸酯、聚丙烯酰胺、聚甲基丙烯酸酯、聚乙基丙烯酸酯、聚甲基丙烯酸羟基烷基酯、聚乙烯醇、聚马来酸酐、聚马来酸酯、聚酰胺、聚乙烯胺、聚乙二醇、聚丙二醇、聚乙酸乙烯酯和聚乙烯基苄基氯、聚羧酸酯和羧基取代的聚合物、以及它们的混合物。适宜的聚合矿物质表面活性剂包括均授予Degenhardt等人的美国专利5,292,501、5,213,789、5,093,170、5,009,882和4,939,284中所述的羧基取代的醇聚合物,和授予Benedict等人的美国专利5,011,913中所述的二膦酸酯衍生的聚合物;合成阴离子聚合物,包括聚丙烯酸酯以及马来酸酐或马来酸与甲基乙烯基醚的共聚物(如Gantrez),如授予Gaffar等人的美国专利4,627,977中所述。优选的聚合物是二膦酸酯改性的聚丙烯酸。具有活性的聚合物具有足够的表面粘附特性以将薄膜蛋白质解吸并保持固定在牙釉质表面上。对于牙齿表面,具有末端或侧链磷酸盐或膦酸酯官能团的聚合物是优选的,然而具有矿物质结合活性的其它聚合物可根据吸附亲和力来证明有效性。
适宜的含膦酸酯聚合矿物质表面活性试剂的其它实例包括授予Degenhardt等人的US4,877,603中公开的作为抗牙结石剂的偕二膦酸酯聚合物;授予Dursch等人的US4,749,758和GB1,290,724(均转让给Hoechst)中公开的含膦酸酯基团的共聚物,其适用于洗涤剂和清洁组合物中;和授予Zakikhani等人的US5,980,776和授予Davis等人的US6,071,434中公开的可用于抑制牙垢和腐蚀应用中的共聚物和共调聚物、覆盖物、接合剂和离子交换树脂。其它聚合物包括GB1,290,724中公开的乙烯基磷酸和丙烯酸及其盐的水溶性共聚物,其中所述共聚物包含按重量计约10%至约90%的乙烯基磷酸和按重量计约90%至约10%的丙烯酸,更具体地讲,其中所述共聚物具有的乙烯基磷酸与丙烯酸的重量比率为70%的乙烯基磷酸比30%的丙烯酸;50%的乙烯基磷酸比50%的丙烯酸;或30%的乙烯基磷酸比70%的丙烯酸。其它适宜的聚合物包括由Zakikhani和Davis公开的水溶性聚合物,其通过二膦酸酯或聚膦酸酯单体与至少一种具有不饱和C=C键的其它化合物(如,丙烯酸酯和甲基丙烯酸酯单体)的共聚化制得,所述聚膦酸酯单体具有一个或多个不饱和的C=C键(如,亚乙烯基-1,1-二膦酸和2-(羟基氧膦基)亚乙基-1,1-二膦酸),所述聚合物例如具有如下结构的那些:

合适的聚合物包括二膦酸酯/丙烯酸酯聚合物,其由Rhodia以商品名ITC1087(平均分子量为3000至60,000)和Polymer1154(平均分子量为6000至55,000)提供。
优选的聚合表面活性试剂在口腔护理组合物中其它组分例如氟离子和金属离子的存在下是稳定的。还优选在水含量高的制剂中具有有限水解度的聚合物,从而有可能得到简单的单相洁齿剂或漱口水制剂。如果聚合表面活性试剂不具有这些稳定特性,一种选择是双相制剂,其中使聚合矿物质表面活性剂与氟化物或其它不相容的组分分开。另一种选择是配制非水性、基本非水性或具有有限量的水的组合物,以将聚合表面活性试剂与其它组分间的反应最小化。
优选的聚合表面活性试剂为多磷酸盐。尽管存在一些环状聚磷酸盐衍生物,但通常认为聚磷酸盐由主要以线性构型排列的两个或多个磷酸盐分子组成。尽管焦磷酸盐(n=2)从技术上说是多磷酸盐,但是所需的多磷酸盐为具有约三个或更多个磷酸根基团的那些,以使有效浓度的表面吸附能够产生足够的非结合磷酸根官能团,这增强了表面的阴离子表面电荷以及亲水特性。在所有磷酸盐中,期望的无机磷酸盐包括三聚磷酸盐、四聚磷酸盐和六偏磷酸盐。比四聚磷酸盐大的聚磷酸盐通常以无定形玻璃质材料出现。在本发明中优选具有下式的直链多磷酸盐:
XO(XPO3)nX
其中X为钠、钾或铵,并且n平均值为约3至约125。优选的多磷酸盐是n平均值为约6至约21的那些,例如由FMC Corporation和Astaris生产的以商品名Sodaphos(n≈6)、Hexaphos(n≈13)和Glass H(n≈21)市售的那些。这些聚磷酸盐可以单独使用或者组合使用。已知多磷酸盐在酸性pH下,尤其是低于pH5时,易于在水含量高的制剂中水解。因此,优选使用长链多磷酸盐,具体地讲是平均链长为约21的Glass H。据信,当经历水解时,此类长链多磷酸盐可产生短链多磷酸盐,其仍可有效地沉积在牙齿上,并且提供防色斑有益效果。
除了多磷酸盐以外或作为多磷酸盐的替代,可使用其它多磷酸化的化合物,具体地讲是多磷酸化肌醇化合物,例如植酸、肌醇五(二氢磷酸酯)、肌醇四(二氢磷酸酯)、肌醇三(二氢磷酸酯)和它们的碱金属盐、碱土金属盐或铵盐。本文优选的是植酸以及它的碱金属盐、碱土金属盐或铵盐,植酸还被称为肌醇1,2,3,4,5,6-六(二氢磷酸酯)或肌醇六磷酸。在本文中,术语“肌醇六磷酸”包括植酸和它的盐以及其它多磷酸化肌醇化合物。
牙齿亲和剂所需量为提供预防色斑功效的有效量。有效量按所述总口部组合物的重量计通常为约0.1%至约35%。在洁齿剂制剂中,所述量优选为约2%至约30%,更优选约5%至约25%,并且最优选约6%至约20%。在漱口水组合物中,牙齿亲和剂的量优选为约0.1%至5%,并且更优选约0.5%至约3%。
除了产生表面改性效果以外,牙齿亲和剂还可用于稳定不溶解的盐。例如,已发现Glass H可溶解不溶解的亚锡盐。因此,在包含例如氟化亚锡的组合物中,Glass H有助于降低亚锡的色斑促生效应。
牙齿漂白剂
所述口部组合物中的第二基本组分为牙齿漂白剂。适于漂白的活性物质选自由下列组成的组:过氧化物、过硼酸盐、过碳酸盐、过氧酸、过硫酸盐、以及它们的组合。适宜的过氧化物包括过氧化氢、过氧化脲、过氧化钙、过氧化钠、过氧化锌、以及它们的混合物。优选的过碳酸盐是过碳酸钠。优选的过硫酸盐是过硫酸氢钾。
优选用于洁齿剂制剂中的过氧化物源是过氧化钙和过氧化脲。过氧化氢和过氧化脲优选用于漱口水制剂中。以下的量代表过氧化物原料的量,尽管过氧化物来源也可包含过氧化物原料之外的成分。本发明的组合物可包含按所述组合物的重量计约0.01%至约30%,优选约0.1%至约10%,并且更优选约0.5%至约5%的过氧化物源。
除了美白以外,所述过氧化物还可向口腔提供其它有益效果。早已认识到,过氧化氢和其它过氧化合物可对于龋齿、牙斑、齿龈炎、牙周炎、口腔异味、牙齿色斑、复发性口腔溃疡、假牙致痛、矫正器具损伤、拔牙后和牙周手术后疼痛、创伤性口服损伤和粘膜感染、病毒性口腔炎等进行有效地治疗性和/或预防性处理。包含过氧化物的试剂在口腔中施以化学机械作用,生成通过与组织酶和唾液酶交互作用而产生的数千微小氧气泡。漱口水的漱动作用可增强这种固有的化学机械作用。已推荐,以此类作用来将其它试剂递送到被感染的齿龈裂缝中。因此,过氧化物漱口水可预防已知与牙周病有关的厌氧细菌定殖和增殖。然而,包含过氧化氢的组合物或其它释放过氧化物化合物一般提供不良的味道和口感,通常被描述为针刺感、刺痛感和刺激感。此外,过氧化物与其它常见赋形剂在其中相互作用,并且在储存时趋于不稳定,在较短时间内不断丧失释放活性物质或初生态氧的能力,并且趋于缩减或破坏所期望的此类赋形剂功能。在此类赋形剂中加入风味剂、感觉材料和着色剂,以增强需要口腔过氧化或漂白处理的那些人对口腔护理产品的接受性。本发明的组合物在储存期间风味剂特性改变方面令人惊奇地稳定。
除了上面描述的组分之外,本组合物还可包含统称为口部可接受载体材料的附加任选组分,这些附加组分描述于以下段落中。
口部可接受的载体材料
所述口部可接受的载体包括一种或多种适于局部口服的相容的固体或液体赋形剂或稀释剂。本文所用术语“相容的”是指组合物的组分能够混合但不相互作用,所述相互作用会显著降低组合物的稳定性和/或功效。
本发明的载体或赋形剂包括常见且常规组分,如洁齿剂、非研磨剂凝胶、龈下凝胶、漱口水或洗口水、口喷剂、口香糖、药糖块和口气清新薄荷,它们将在下文被更详细地描述。
待使用的载体的选择基本上是按照组合物引入口腔的方法来确定。可用于牙膏、牙胶或类似产品包括磨料、起泡剂、粘合剂、湿润剂、风味剂和甜味剂等的载体材料,如公开于授予Benedict的美国专利3,988,433中。用于两相洁齿剂制剂的载体材料公开于下述文献中的物质:1993年5月23日公布的美国专利5,213,790、1992年9月8日公布的5,145,666和1994年1月25日公布的5,281,410,上述专利均授予Lukacovic等人;和授予Schaeffer的美国专利4,849,213和4,528,180。漱口水、洗口水或口喷剂载体材料通常包括水、风味剂和甜味剂等等,例如,在授予Benedict的美国专利3,988,433中所公开。药糖块载体材料通常包括一种糖果基质;口香糖载体材料包括一种树胶基质、风味剂和甜味剂,这公开于例如授予Grabenstetter等人的美国专利4,083,955中。小袋载体材料通常包括香袋、风味剂和甜味剂。对于用于递送活性物质至牙周袋或牙周袋周围的龈下凝胶,按例如1993年3月30日公布的美国专利5,198,220和1993年9月7日公布的美国专利5,242,910选择“龈下凝胶载体”,上述两项专利均授予Damani。适于制备本发明组合物的载体在本领域已为人们所熟知。它们的选择取决于如口味、成本和架藏稳定性等次要考虑因素。
本发明的组合物还可以为含水或非水性的非研磨性凝胶和龈下凝胶的形式。在另一个方面,本发明还提供注有本组合物的牙具。牙具包括用于和口腔中牙齿和其它组织接触的器具,所述器具注入有本文所述组合物。牙具可被注入纤维,该纤维包括牙线或牙带、薄片、薄带、薄膜和聚合物纤维。
在一个优选的实施例中,本主题发明的组合物为洁齿剂形式,例如牙膏、牙胶和牙粉。这些牙膏和牙胶的组分通常包括一种或多种牙齿研磨剂(约6%至约50%)、表面活性剂(约0.5%至约10%)、增稠剂(约0.1%至约5%)、湿润剂(约10%至约55%)、调味剂(约0.04%至约2%)、甜味剂(约0.1%至约3%)、染色剂(约0.01%至约0.5%)和水(约2%至约45%)。此类牙膏或牙胶还可包括一种或多种防龋剂(约0.05%至约0.3%作为氟离子)和抗牙结石剂(约0.1%至约13%)。当然,牙粉包含基本上所有非液体的组分。
本主题发明的其它优选实施方案是液体产品,包括漱口水或漱口液、口喷剂、牙科用溶液和冲洗流体。这些漱口剂和口喷剂的组分通常包括一种或多种的水(约45%至约95%)、乙醇(约0%至约25%)、湿润剂(约0%至约50%)、表面活性剂(约0.01%至约7%)、调味剂(约0.04%至约2%)、甜味剂(约0.1%至约3%)和着色剂(约0.001%至约0.5%)。此类漱口水和口腔喷剂还可包括一种或多种防龋剂(约0.05%至约0.3%作为氟离子)和抗牙结石剂(约0.1%至约3%)。牙科用溶液的组分一般包括水(约90%至约99%)、防腐剂(约0.01%至约0.5%)、增稠剂(0%至约5%)、调味剂(约0.04%至约2%)、甜味剂(约0.1%至约3%)和表面活性剂(0%至约5%)中的一种或多种。
可被包括在本发明的组合物中的口部可接受的载体或赋形剂的类型,与具体的非限制性实例一起,将在下面的章节中论述。
氟化物源
洁齿剂和其它口腔组合物中通常具有足量的水溶性氟化物化合物,以便为组合物提供氟离子浓度,和/或当所述氟化物化合物以按重量计约0.0025%至约5.0%,优选约0.005%至约2.0%的浓度使用时能提供防龋效果。多种产生氟离子的物质可被用作本发明组合物中适宜的可溶性氟化物源。适宜产生氟离子材料的实例公开于1970年10月20日授予Briner等人的美国专利3,535,421和1972年7月18日授予Widder等人的美国专利3,678,154中。代表性的氟离子源包括:氟化亚锡、氟化钠、氟化钾、单氟磷酸钠、氟化铟以及其它多种来源。氟化亚锡和氟化钠以及它们的混合物是优选的。
研磨剂
可用于本主题发明组合物的局部口腔载体中的牙齿研磨剂包括许多不同的物质。所选的材料必须是与组合物中相关的物质相容的那个,并且不会过分磨损牙质。合适的研磨剂包括例如含有凝胶和沉淀剂的二氧化硅、不溶解的聚偏磷酸钠、水合氧化铝、碳酸钙、二碱式磷酸钙二水合物、焦磷酸钙、磷酸三钙、聚偏磷酸钙和树脂研磨物质,例如尿素和甲醛的颗粒缩合产物。
用于本组合物中的另一类研磨剂是颗粒状热固性聚合树脂并描述于1962年12月25日授予Cooley和Grabenstetter的美国专利3,070,510中。合适的树脂包括例如三聚氰胺、苯酚、脲、三聚氰胺-脲、三聚氰胺-甲醛、脲-甲醛、三聚氰胺-脲-甲醛、交联环氧化物和交联聚酯。
优选多种类型的二氧化硅牙齿研磨剂,因为它们具有使牙齿特别清洁但又不会过度磨损牙釉或牙质的磨光性能的独特有益效果。本发明的二氧化硅磨料抛光物质,以及其它研磨剂的平均粒径通常在约0.1至约30微米,并且优选约5至约15微米的范围内。所述研磨剂可以是沉淀二氧化硅或硅胶,如在公布于1970年3月2日授予Pader等人的美国专利3,538,230和公布于1975年1月21日授予DiGiulio的美国专利3,862,307中所描述的二氧化硅干凝胶。实例包括W.R.Grace&Company,Davison Chemical Division以“Syloid”为商品名销售的二氧化硅干凝胶和例如J.M.Huber Corporation以商品名销售的沉淀二氧化硅,尤其是具有119、118、109和129命名的二氧化硅。可用于本发明牙膏中的这类二氧化硅牙齿研磨剂更详细地描述于1982年7月29日授予Wason的美国专利4,340,583;和普通转让的公布于1997年2月18日的美国专利5,603,920;公布于1996年12月31日的美国专利5,589,160;公布于1997年8月19日的美国专利5,658,553;公布于1997年7月29日的美国专利5,651,958和公布于2004年5月25日的美国专利6,740,311。
可应用的研磨剂混合物如上面所列的的不同等级的二氧化硅研磨剂。在本主题发明的洁齿剂组合物中,研磨剂的总量按重量计通常在约6%至约70%的范围内。牙膏优选包含按所述组合物的重量计约10%至约50%的研磨剂。本主题发明的牙齿溶液、口腔喷剂、漱口水和非研磨性凝胶组合物通常包含很少的研磨剂或完全不含研磨剂。
抗牙结石剂
本组合物可任选地包括附加的抗牙结石剂,例如可作为焦磷酸离子源的焦磷酸盐。本发明组合物中使用的焦磷酸盐包括焦磷酸二碱金属盐、焦磷酸四碱金属盐、以及它们的混合物。焦磷酸二氢二钠(Na2H2P2O7)、焦磷酸四钠(Na4P2O7)和焦磷酸四钾(K4P2O7)的未水合和水合形式是优选物质。在本发明的组合物中,焦磷酸盐能够以三种方式中的一种存在:基本上溶解、基本上不溶解,或溶解与不溶解的焦磷酸盐的混合物。
包括基本上溶解的焦磷酸盐的组合物指其中至少一种焦磷酸离子源具有足够的量以提供至少约1.0%游离焦磷酸离子的组合物。游离焦磷酸盐离子的含量可为约1%至约15%,在一个实施方案中为约1.5%至约10%,在另一个实施方案中为约2%至约6%。根据组合物pH值的不同,游离焦磷酸盐离子可以各种质子化状态存在。
包括基本上不溶解的焦磷酸盐的组合物指包含低于约20%,优选小于约10%的总焦磷酸盐溶解于组合物中的组合物。在这些组合物中,焦磷酸四钠是优选的焦磷酸盐。在洁齿剂组合物中,焦磷酸四钠可为无水形式或十水合物形式,或任何其它稳定于固体形式的种类。盐为其固体颗粒形式,该形式可为其结晶和/或非晶形状态,盐的粒径优选足够小以在美学上可接受和在使用中易溶解。在制备这些组合物中使用的焦磷酸盐量为任何能有效控制牙结石的量,该量按所述洁齿剂组合物的重量计通常为约1.5%至约15%,优选约2%至约10%,并且最优选约3%至约8%。
组合物还可包括溶解和不溶解的焦磷酸盐的混合物。可使用上述提及的任何焦磷酸盐。
焦磷酸盐更详细地描述于Kirk-Othmer Encyclopedia ofChemical Technology,第三版,第17卷,Wiley-IntersciencePublishers(1982)。
可以代替或者可以与焦磷酸盐组合使用的任选试剂包括已知物质,例如合成的阴离子聚合物,包括聚丙烯酸酯以及马来酸酐或马来酸与甲基乙烯基醚的共聚物(例如Gantrez),例如在授予Gaffer等人的美国专利4,627,977中所描述;以及例如聚氨基丙磺酸(AMPS)、二膦酸盐(例如EHDP;AHP)、多肽(例如聚天冬氨酸和聚谷氨酸)、以及它们的混合物。
螯合剂
另一种任选试剂是螯合剂,也称多价螯合剂,如葡糖酸、酒石酸、柠檬酸和其可药用的盐。螯合剂能络合存在于细菌细胞壁中的钙。螯合剂也可通过从钙桥中除去钙而破坏牙斑,所述钙桥有助于保持生物量完整。然而,不希望使用对钙有过高亲和力的螯合剂,因为这会导致牙齿脱矿质,这与本发明的目的和意图相背离。合适的螯合剂的典型钙结合常数为约101至105,以提供减少牙斑和牙结石的改善的清洁作用。螯合剂也具有与金属离子复合的能力,因此有助于预防产品在稳定性或外观方面的不利影响。螯合例如铁或铜的离子有助于延迟成品的氧化变质。
适宜的螯合剂实例是葡糖酸钠或钾和柠檬酸钠或钾;柠檬酸/柠檬酸碱金属盐组合;酒石酸二钠;酒石酸二钾;酒石酸钾钠;酒石酸氢钠;酒石酸氢钾;以及它们的混合物。适用于本发明中螯合剂的量为约0.1%至约2.5%,优选为约0.5%至约2.5%,并且更优选为约1.0%至约2.5%。
适用于本发明的其它螯合剂是阴离子聚合的聚羧酸酯。这些物质是本领域是众所周知的,它们以其游离酸、或部分或优选完全中和的水溶性碱金属(例如钾,优选钠)盐或铵盐的形式应用。实例是马来酸酐或酸与另一种可聚合的烯键式不饱和单体的1:4至4:1共聚物,所述单体优选具有约30,000至约1,000,000分子量(MW)的甲基乙烯基醚(甲氧基乙烯)。这些共聚物例如以GAF Chemicals Corporation的商品名Gantrez AN 139(分子量500,000)、AN119(分子量250,000)和S-97药品等级(分子量70,000)获得。
其它有效的聚合聚羧酸酯包括马来酸酐与丙烯酸乙酯、甲基丙烯酸羟乙酯、N-乙烯基-2-吡咯烷酮或乙烯的1:1共聚物,后者可以商品名例如,Monsanto EMA No.1103,MW10,000和EMA Grade61获得,和丙烯酸与甲基丙烯酸甲酯或甲基丙烯酸羟乙酯、丙烯酸甲酯或丙烯酸乙酯、异丁基乙烯基醚或N-乙烯基-2-吡咯烷酮的1:1共聚物。
附加的有效聚合多磷酸盐公开于1979年2月6日授予Gaffar的美国专利4,138,477和1980年1月15日授予Gaffar等人的美国专利4,183,914,并包括马来酸酐与苯乙烯、异丁烯或乙基乙烯基醚的共聚物;聚丙烯酸、聚亚甲基丁二酸和聚马来酸;和商品名为Uniroyal ND-2的分子量低到1,000的丙烯磺酸低聚物。
其它活性剂
本发明可任选地包括其它试剂,如抗微生物剂。包括在这种试剂中的是水不溶性的非阳离子抗微生物剂如卤代二苯基醚、苯酚化合物,所述化合物包括苯酚及其同系物、一烷基和多烷基与芳族的卤代酚、间苯二酚及其衍生物、双酚化合物和卤代N-水杨酰苯胺、安息香酯和卤代N-碳酰苯胺。水溶性抗微生物剂包括季铵盐和双-二胍盐以及三氯生一磷酸盐。季铵试剂包括其中季氮上的一个或两个取代基的碳链长度(通常为烷基)为约8至约20、通常为约10至18个碳原子,同时剩余取代基(通常为烷基或苄基)具有更低的碳原子数如约1至约7个碳原子,通常为甲基或乙基的那些。典型的季铵抗菌剂实施例有十二烷基三甲基溴化铵、氯化十四烷基吡啶、溴化度米芬、氯化N-十四烷基-4-乙基吡啶鎓、十二烷基二甲基(2-苯氧乙基)溴化铵、苄基二甲基硬脂基氯化铵、氯化十六烷基吡啶、季铵化的5-氨基-1,3-双(2-乙基己基)-5-甲基六氢嘧啶、烷基苄基二甲基氯化铵、苄索氯铵和甲基苄索氯铵。其它化合物是公开于1980年6月3日公布的授予Bailey的美国专利4,206,215中的双[4-(R-氨基)-1-吡啶鎓烷烃。也可包括其它抗微生物剂如铜盐、锌盐和二价锡盐。还有用的是酶,包括糖苷内切酶、木瓜蛋白酶、葡聚糖酶、变异酶(mutanase)、以及它们的混合物。上述试剂公开于1960年7月26日授予Norris等人的美国专利2,946,725和1977年9月27日授予Gieske等人的美国专利4,051,234中。优选的抗微生物剂包括锌盐、亚锡盐、西吡氯铵、氯己定、三氯生、三氯生一磷酸盐、和风味油例如百里酚。三氯生和此类其它试剂公开于1991年5月14日公布的Parran,Jr.等人的美国专利5,015,466和公开于1990年1月16日公布的授予Nabi等人的美国专利4,894,220。这些试剂可提供抗色斑有益效果,并且通常含量按所述组合物的重量计为约0.01%至约5.0%。
另一种可加入到本发明组合物中的任选活性剂是控制超敏性的牙质脱敏剂,例如钾、钙、锶和锡的盐,包括硝酸盐、氯化物、氟化物、磷酸盐、焦磷酸盐、多磷酸盐、柠檬酸盐、草酸盐和硫酸盐。

本组合物还可包含表面活性剂,其通常也被称为起泡剂。合适的表面活性剂为那些在整个宽的pH值范围内具有适度稳定性和泡沫的物质。表面活性剂可以是阴离子的、非离子的、两性的、两性离子的、阳离子的、或它们的混合物。
可用于本发明的阴离子表面活性剂包括烷基中含有8至20个碳原子的烷基硫酸的水溶性盐(如烷基硫酸钠)和含有8至20个碳原子的脂肪酸的磺化单酸甘油酯的水溶性盐。这类阴离子表面活性剂的实例是月桂基硫酸钠(SLS)和椰子基单酸甘油酯磺酸钠。其他合适的阴离子表面活性剂为肌氨酸盐如月桂酰肌氨酸钠、牛磺酸盐、月桂基磺基乙酸钠、月桂酰羟乙基磺酸钠、月桂基聚氧乙烯醚羧酸钠和十二烷基苯磺酸钠。也可使用阴离子表面活性剂的混合物。许多合适的阴离子表面活性剂公开于1976年5月25日授予Agricola等的美国专利3,959,458中。本组合物中阴离子表面活性剂的典型含量在约0.025%至约9%,在一些实施方案中约0.05%至约5%,在其它实施方案中约0.1%至约1%的范围内。
另一种合适的表面活性剂选自由肌氨酸盐表面活性剂、羟乙基磺酸盐表面活性剂和牛磺酸盐表面活性剂组成的组。优选可用于本文的是这些表面活性剂的碱金属或铵盐,如下列的钠盐和钾盐:月桂酰肌氨酸盐、肉豆寇酰肌氨酸盐、棕榈酰肌氨酸盐、硬脂酰肌氨酸盐和油酰肌氨酸盐。所述肌氨酸盐表面活性剂按所述总组合物的重量计可以约0.1%至约2.5%,优选约0.5%至约2.0%的含量存在于本发明的组合物中。
用于本发明的阳离子表面活性剂包括脂族季铵化合物的衍生物(所述化合物具有一个包含约8至18个碳原子的长链烷基链)如月桂基三甲基氯化铵;氯化鲸蜡基吡啶鎓;鲸蜡基三甲基溴化铵;二异丁基苯氧乙基二甲基苄基氯化铵;椰子基烷基三甲基亚硝酸铵;氟化鲸蜡基吡啶鎓等。优选的化合物是季铵氟化物,其描述于1970年10月20日公布的Briner等人的美国专利3,535,421中,其中所述季铵氟化物具有洗涤剂性质。在本发明公开的组合物中,某些阳离子表面活性剂也可用作杀菌剂。阳离子表面活性剂如洗必太尽管也适用于本发明,但由于它们会沾污口腔硬组织,因此不是优选的。本领域的技术人员知道这种可能性,并在掺入阳离子表面活性剂时必须清楚这种限制。
可用于本发明组合物中的非离子表面活性剂包括由氧化亚烷基团(本身亲水的)与本身可为脂族或烷基芳族的有机疏水化合物缩合而产生的化合物。合适的非离子表面活性剂的实例包括聚丙二醇与环氧乙烷的加聚物(聚醚)、烷基酚的聚环氧乙烷缩合物、衍生自环氧乙烷与氧化丙烯和1,2-乙二胺的反应产物的缩合产物、脂族醇的环氧乙烷缩合物、长链叔胺氧化物、长链叔膦氧化物、长链二烷基亚砜、以及这些物质的混合物。
用于本发明的两性离子合成表面活性剂包括脂族季铵、鏻鎓和锍化合物的衍生物,其中该脂族基团可以是直链或支链的,并且其中一个脂族取代基包含约8至18个碳原子,而一个脂族取代基包含阴离子水增溶基团,例如羧基、磺酸根、硫酸根、磷酸根或膦酸根。
合适的甜菜碱表面活性剂公开于1993年1月19日授予Polefka等人的美国专利5,180,577中。典型的烷基二甲基甜菜碱包括癸基甜菜碱或2-(N-癸基-N,N-二甲基胺)乙酸酯、椰油基甜菜碱或2-(N-椰油基-N,N-二甲基胺)乙酸酯、十四烷基甜菜碱、棕榈基甜菜碱、月桂基甜菜碱、鲸蜡基甜菜碱、硬脂基甜菜碱等等。酰氨基甜菜碱的实施例有椰油酰氨基乙基甜菜碱、椰油酰氨基丙基甜菜碱、月桂酰氨基丙基甜菜碱等。精选的甜菜碱优选为椰油酰氨基丙基甜菜碱,并且更优选为月桂酰氨基丙基甜菜碱。
增稠剂
在制备牙膏或凝胶中添加增稠剂以提供:所需的组合物稠度、使用时所需的活性物质释放特性、架藏稳定性和组合物的稳定性等。合适的增稠剂包括一个或羧乙烯基聚合物、角叉菜胶、羟乙基纤维素(HEC)、天然和合成粘土(即胶体硅酸镁铝和合成锂皂石)和纤维素醚的水溶性盐如羧甲基纤维素钠(CMC)和羧甲基羟乙基纤维素钠的组合。也可使用天然树胶如刺梧桐树胶、黄原胶、阿拉伯树胶和黄蓍胶。胶态硅酸镁铝或细分的二氧化硅可用作部分增稠剂以进一步改善稠度。
可用作增稠剂或胶凝剂的合适的羧乙烯基聚合物包括卡波姆,它是丙烯酸与季戊四醇烷基醚或蔗糖烷基醚交联的一类均聚物。卡波姆可以系列从B.F.Goodrich商购获得,包括Carbopol 934、940、941、956、以及它们的混合物。
增稠剂可以按所述总牙膏或凝胶组合物的计通常约0.1%至约15%,优选约2%至约10%,更优选约4%至约8%的量使用。更高的浓度可用于口香糖、药用糖块和口气清新薄荷糖、小药囊、非研磨性凝胶和牙龈下凝胶。
湿润剂
本组合物中另一种任选的载体材料是湿润剂。湿润剂用于防止牙膏组合物在接触空气后发生硬化,并使组合物赋予口腔潮湿感,并且对于特定湿润剂来说,湿润剂还可用来赋予牙膏组合物理想的甜味。以纯湿润剂为基础,该湿润剂的含量按本发明组合物的重量计通常为约0%至约70%,优选约5%至约25%。可用于本主题发明组合物中的适宜湿润剂包括可食用的多元醇,如甘油、山梨醇、木糖醇、丁二醇、聚乙二醇、丙二醇和三甲基甘氨酸。
风味剂和甜味剂
本发明组合物中也可添加调味剂。然而,许多典型的风味剂与漂白剂、具体地讲与过氧化物是化学不相容的。授予Clipper等人的美国专利4,684,517中公开的适宜风味剂的实例包括薄荷醇、水杨酸甲酯、肉桂醛和丁香油。调味剂通常以按所述组合物的重量计约0.001%至约5%的含量用于该组合物中。
可使用的甜味剂包括蔗糖、葡萄糖、糖精、三氯蔗糖、右旋糖、左旋糖、乳糖、甘露糖醇、山梨醇、果糖、麦芽糖、木糖醇、糖精盐、非洲甜果素、天冬甜素、D-色氨酸、二氢查尔酮、丁磺氨和环磺酸盐,尤其是环磺酸钠、三氯蔗糖和糖精钠、以及它们的混合物。组合物优选包含按所述组合物的重量计约0.1%至约10%,优选约0.1%至约1%的这些试剂。
除了调味剂和甜味剂之外,冷却剂、流涎剂、加温剂和麻木剂也可用作本发明的组合物的任选组分。这些试剂以按所述组合物的重量计约0.001%至约10%,优选约0.1%至约1%的含量存在于组合物中。
冷却剂可以是任何各种各样的物质。包括在本发明中的这些物质是羧酰胺、薄荷醇、缩酮、二醇、以及它们的混合物。本组合物中的优选冷却剂是对孟烷氨基甲酰试剂,如N-乙基-对-孟烷-3-酰胺,商品名为“WS-3”;N,2,3-三甲基-2-异丙基丁酰胺,商品名为“WS-23”;以及它们的混合物。其它优选的冷却剂选自由下列组成的组:薄荷醇、由Takasago生产的称为TK-10的3-1-薄荷氧基丙烷-1,2-二醇、由Haarmann和Reimer生产的称为MGA的薄荷酮甘油缩醛和由Haarmann和Reimer生产的称为的乳酸薄荷酯。本文所用术语薄荷醇和薄荷基包括这些化合物的右旋和左旋异构体、以及它们的外消旋混合物。TK-10描述于7/10/84公布的授予Amano等人的美国专利4,459,425中。WS-3和其它试剂描述于1979年1月23日公布的授予Watson等人的美国专利4,136,163中。
本发明适宜的流涎剂包括由Taka sago生产的加温剂的实例是辣椒和烟酸酯,如烟酸苄酯。适宜的麻木剂包括苯佐卡因、利多卡因、丁香芽油和乙醇。
混杂载体材料
在制备商业上合适的口腔组合物时使用的水应该优选是低离子含量的并且不含有机杂质。水的含量按本文所述含水组合物的重量计一般最多为约99%。这些水的量包括加入的游离水和由其他物质如由山梨醇带入的水。
本发明也可包括碱金属碳酸氢盐,它可以提供许多功能,所述功能包括研磨、除臭、缓冲和调节pH值。碱金属碳酸氢盐是水溶性的,除非被稳定化,它在含水体系中易于释放二氧化碳。碳酸氢钠也称为发酵粉,它是常用的碱金属碳酸氢盐。本组合物可包含约0.5%至约30%,优选约0.5%至约15%,最优选约0.5%至约5%的碱金属碳酸氢盐。
通过使用缓冲剂可调节本组合物的pH值。如本文所用,缓冲剂涉及用于调节含水组合物例如漱口水和牙科用溶液pH的试剂,鉴于过氧化物稳定性,优选将pH调节到约pH4.0至约pH6.0的范围内。缓冲剂包括碳酸氢钠、磷酸一钠、磷酸三钠、氢氧化钠、碳酸钠、焦磷酸钠、柠檬酸和柠檬酸钠。按本组合物的重量计,缓冲剂的典型用量为约0.5%至约10%。
泊洛沙姆可用于本组合物中。泊洛沙姆属非离子表面活性剂也可按功能分为乳化剂、粘合剂、稳定剂和其它相关的功能。泊洛沙姆是末端为伯羟基的双官能嵌段聚合物,分子量从1,000到超过15,000。泊洛沙姆由BASF以商品名Pluronics和Pluraflo出售。本发明合适的泊洛沙姆是Poloxamer 407和Pluraflo L4370。
二氧化钛也可加入到本组合物中。二氧化钛是能给组合物提供不透明性的白色粉末。二氧化钛的含量按所述洁齿剂组合物的重量计一般为约0.25%至约5%。
其它可用于本组合物的任选试剂包括选自烷基聚二甲基硅氧烷共聚多元醇和烷氧基聚二甲基硅氧烷共聚多元醇的聚二甲基硅氧烷共聚多元醇,如C12-C20烷基聚二甲基硅氧烷共聚多元醇以及它们的混合物。高度优选是商品名为Abil M90的鲸蜡基聚二甲硅氧烷共聚多元醇。通常,聚二甲基硅氧烷共聚多元醇的含量按重量计为约0.01%至约25%,优选约0.1%至约5%,更优选约0.5%至约1.5%。该聚二甲基硅氧烷共聚多元醇辅助提供积极的牙齿感觉有益效果。
使用方法
本发明还涉及美白牙齿并且防止牙齿变色的方法。当反复使用所述组合物时,这些组合物的有益效果可随时间而增加。
本文的处理方法包括使受试者的牙釉质表面和口腔粘膜与如本发明所述的口部组合物接触。处理方法可以是用洁齿剂刷洗,或用洁齿剂浆液或漱口水漱洗。其它方法包括将局部用口腔凝胶、假牙产品、口喷剂或其它形式与患者的牙齿和口腔粘膜接触。受试者可以是其牙齿表面接触所述口部组合物的任何人或动物。“动物”是指包括家庭宠物或其它家畜,或关在笼子中的动物。
例如,处理方法可包括个人使用一种洁齿剂组合物来刷洗狗的牙齿。另一个实例包括用口腔组合物将猫的嘴漂洗足够长时间以看到有益效果。可以配制包含本口腔组合物的宠物护理产品,如咀嚼物和玩具。将所述组合物包含在相对柔软但坚固耐用的材料中,如生皮、由天然或合成纤维制成的绳子、和由尼龙、聚酯或热塑性聚氨酯制成的聚合物制品。当动物咀嚼、口舔或啃咬上述产品时,所含的活性组分被释放到动物的口腔中的唾液基质中,这相当于有效的刷洗或漂洗。

下列实施例进一步描述和证明了本发明范围内的具体实施方案。所给出的这些实施例仅仅是为了举例说明,不应被认为是对本发明的限制,因为在不背离本发明的精神和范围的条件下,对其进行许多改变是可能的。
实施例I漱口水组合物
如本发明所述的漱口水组合物(IA-IE)示于下文中,其中组分量以重量百分比(%)为单位。这些组合物可使用常规方法来制备。

实施例II色斑预防功效
使用体外方法来测定本发明组合物的色斑预防功效。称取(200±10mg)羟基磷灰石(HAP)粉末,并且与处理溶液(或为纯的口腔漱口液,或为25%w/w的洁齿剂和水的上清液)接触30秒。然后通过离心(15,000rpm,15分钟)除去HAP,在去离子水中经由再悬浮洗涤,并且通过离心除去。重复离心/再悬浮步骤。通过将处理过并且洗涤过的HAP粉末加入到新煮好的茶水(1袋/100ml,50℃)中,放置30秒,然后洗涤两次,来实施HAP粉末的染色。使洗涤过的HAP在10ml水中再悬浮,并且使用0.45μm孔的过滤器,真空过滤通过过滤器。将其干燥和层压。采用数字式摄影技术,使用白光成像体系(Fuji 2000照相机),拍摄HAP的颜色读数。使用Giant 2.0.1软件包,测定颜色值(L*a*b*)。L*代表y轴上的光亮度,a*代表x轴上的色度(红-绿),而b*代表z轴上的色度(黄-蓝)。通过从未处理和未染色的HAP粉末L*a*b*测量中减去已处理HAP粉末的L*a*b*测量,计算出L*、a*和b*单独成分的变化(Δ值)。计算Δ值平方和的平方根,作为总颜色变化(ΔE)。结果综述于下文表1中,显示了总光亮度ΔL*变化、黄色Δb*变化和总颜色ΔE。预防牙齿变色的本发明组合物显著优于阴性对照(水和Crest Cavity Protection),并且程度与包含多磷酸盐的Crest Dual Action Whitening相同。制备出的原样产品与40℃下老化后的产品相比,在性能上无差别,证明在加速老化条件下的储存期间内可保持产品的功效。

实施例III组合物的稳定性
通过在25℃和40℃的储存条件下测定过氧化物含量、pH和多磷酸盐水解度,评定本发明组合物的稳定性。
使用水容性PeroXOquantTM定量过氧化物检测分析法来测定过氧化氢,所述分析法基于在二甲苯酚橙的存在下亚铁氧化成高铁离子来检测过氧化物。过氧化物首先与山梨醇反应,将其转变成过氧基,其继而引发Fe2+至Fe3+的氧化反应。Fe3+与二甲苯酚橙染料复合以生成紫色产物。在595nm波长下使用微孔板分光光度计测定此复合物来测定样本中的过氧化氢。所述方法具有最多约10%的误差幅度。
使用电位仪(Orion720A型)和Orion 组合pH电极,在室温下测定实施例IC漱口水产品的pH。
经由从直链多磷酸盐链中分离使余链分别缩短n=1或n=3的正磷酸盐(n=1)和三偏磷酸盐(n=3,环状)的含量来测定多磷酸盐的水解度。正磷酸盐和三偏磷酸盐的增量表明多磷酸盐的水解度。
稳定性测试结果综述于下文表2中,表明本发明组合物在25℃和40℃的储存期间,在过氧化物降低时是稳定的。pH在储存期间趋于降低,而多磷酸盐水解度随着pH的降低而增加。然而,如实施例II中所示,未观察到老化产品色斑预防功效的变化。

实施例IV两相洁齿剂组合物
如本发明所述的两相洁齿剂组合物由优选以50:50比率分散的包含过氧化物的第一洁齿剂组合物(IVA至IVC)和第二洁齿剂组合物(IVD至VE)构成。这些组合物使用常规方法制备。

Claims (10)
1.一种口腔护理组合物,所述组合物提供增强的牙齿美白和色斑预防功效,所述组合物包含
(a)按所述组合物的重量计0.01%至30%的牙齿漂白活性物质;
(b)按所述组合物的重量计0.1%至35%的多磷酸化化合物,所述多磷酸化化合物具有对牙齿的亲和性以提供色斑预防,并选自平均链长为6或更长的直链多磷酸盐;植酸;肌醇五(二氢磷酸酯);肌醇四(二氢磷酸酯);肌醇三(二氢磷酸酯);它们的碱金属盐、碱土金属盐或铵盐或它们的混合物;和
(c)口部可接受的含水载体,
其中所述组合物具有4.0至6.0范围内的pH,所述组合物在40℃的储存条件下防止漂白活性物质损失可保持稳定至少三个月。
2.如权利要求1所述的口腔护理组合物,其中所述多磷酸盐具有6至125的平均链长。
3.如权利要求1所述的口腔护理组合物,其中所述多磷酸盐具有6至21的平均链长。
4.如权利要求1所述的口腔护理组合物,其中所述多磷酸盐具有21的平均链长。
5.如权利要求1所述的口腔护理组合物,其中所述牙齿漂白活性物质选自:过氧化物、过硼酸盐、过碳酸盐、过氧酸、过硫酸盐、或它们的混合物。
6.如权利要求5所述的口腔护理组合物,其中所述牙齿漂白活性物质选自:过氧化氢、过氧化脲、过氧化钙、过碳酸钠、或它们的混合物。
7.如权利要求1所述的口腔护理组合物,所述组合物为洁齿剂或漱口水的形式。
8.如权利要求1所述的口腔护理组合物,所述口腔护理组合物包含风味剂组分,所述风味剂组分包含一种凉爽剂或凉爽剂的混合物,所述凉爽剂选自薄荷醇、羧酰胺、缩酮、二醇、或它们的混合物。
9.一种口腔护理组合物,所述组合物提供增强的牙齿美白和色斑预防功效,所述组合物包含
(a)按所述组合物的重量计0.01%至10%的过氧化物牙齿漂白活性物质;
(b)0.1%至5%的多磷酸化化合物,所述多磷酸化化合物具有对牙齿的亲和性以提供防色斑,并选自平均链长为6至125的直链多磷酸盐;植酸;肌醇五(二氢磷酸酯);肌醇四(二氢磷酸酯);肌醇三(二氢磷酸酯);它们的碱金属盐、碱土金属盐或铵盐或它们的混合物;和
(c)口部可接受的含水载体,
其中所述组合物具有4.0至6.0范围内的pH,并且其中所述组合物在40℃的储存条件下防止过氧化物损失可保持稳定至少三个月。
10.如权利要求9所述的口腔护理组合物,所述组合物为漱口水的形式,包含作为牙齿漂白活性物质的过氧化氢或过氧化脲,以及具有6至21平均链长的直链多磷酸盐。
