技术背景
已知疲劳分为病态疲劳和生理性疲劳,疾病时或过度训练时出现的病态疲劳情况,血中氨值上升。而与此相反,健康人生理性疲劳时血中氨值并没有观察到升高。
此外,疲劳感即疲劳症状的检查方法,使用由日本产业卫生学会产业疲劳研究会(Industrial Fatigue Research Committee)制作的自觉症状调查表(非专利文献1)。该自觉症状调查表将疲劳感分为三个因素,第1组为“困倦和疲倦”、第2组为“注意力难以集中”、第3组为“身体不适感”。
鸟氨酸作为通过促进生长激素分泌而增强肌肉形成,或通过提高基础代谢防止肥胖的食品材料,主要在美国和日本使用。此外,在欧洲鸟氨酸作为改善肝脏损伤药品以L-鸟氨酸L-天冬氨酸盐的形式使用。
与血中氨值上升相关的病态疲劳,已知可通过给药鸟氨酸或其盐,降低血中氨值,以治疗病态疲劳(专利文献1~3、非专利文献2)。但是,对于健康人生理性疲劳所致的疲劳感即疲劳自觉症状,还未见有通过使用鸟氨酸或其盐来改善的报道。

发明内容
发明要解决的问题
希望研制出能够改善有疲劳自觉症状人的自觉症状、能够创造充实生活的医药品、功能性食品等。也就是说,本发明的目的为提供改善疲劳自觉症状用的组合物。
解决问题的方法
本发明涉及下述(1)~(12)项内容。
(1)改善疲劳自觉症状用组合物,其中含有鸟氨酸或其盐为有效成分。
(2)上述(1)组合物,其中疲劳自觉症状为生理性疲劳的自觉症状。
(3)上述(1)或(2)的组合物,其中疲劳自觉症状为与困倦和疲倦,或注意力难以集中有关的症状。
(4)上述(1)~(3)任一项的组合物,其还含有蒜或蒜提取物。
(5)一种疲劳自觉症状的改善方法,该方法包括向需要的对象给药或使其摄取有效量的鸟氨酸或其盐。
(6)上述(5)中记载的方法,其中疲劳自觉症状为生理性疲劳的自觉症状。
(7)上述(5)或(6)中记载的方法,其中疲劳自觉症状为与困倦和疲倦,或注意力难以集中有关的症状。
(8)上述(5)~(7)中任一项记载的方法,其中还包括给药蒜或蒜提取物或使蒜或蒜提取物被摄取。
(9)鸟氨酸或其盐在制备用于改善疲劳自觉症状的组合物。
(10)上述(9)中记载的用途,其中疲劳自觉症状为生理性疲劳的自觉症状。
(11)上述(9)或(10)中记载的用途,其中疲劳自觉症状为与困倦和疲倦,或注意力难以集中有关的症状。
(12)上述(9)~(11)中任一项记载的用途,其还包括蒜或蒜提取物的用途。
发明的效果
通过本发明,可以提供含有鸟氨酸或其盐为有效成分,安全有效的改善疲劳自觉症状用组合物。

发明的最佳实施方案
本发明使用的鸟氨酸,可以是L-鸟氨酸和D-鸟氨酸,优选L-鸟氨酸。
鸟氨酸可通过化学合成方法、发酵生产方法等得到。此外,也可以通过购入市售品得到鸟氨酸。作为化学合成L-鸟氨酸的方法,可以举例,Coll.Czechoslov.Chem.Commun.,24,1993(1959)记载的方法。作为发酵生产L-鸟氨酸的方法,可以举例,特开昭53-24096号公报、特开昭61-119194号公报记载的方法。此外也可以通过例如Sigma Aldrich Company购买L-鸟氨酸和D-鸟氨酸。
作为鸟氨酸盐,可以举例,酸加成盐、金属盐、铵盐、有机胺加成盐、氨基酸加成盐等。作为酸加成盐,可以举例,盐酸盐、硫酸盐、硝酸盐和磷酸盐等无机酸盐;醋酸盐、马来酸盐、富马酸盐、枸橼酸盐、苹果酸盐、乳酸盐、α-酮戊二酸盐、葡糖酸盐、辛酸盐等有机酸盐。作为金属盐,可以举例,钠盐和钾盐等碱金属盐;镁盐和钙盐等碱土金属盐;铝盐、锌盐等。
作为铵盐,可以举例,铵、四甲基铵等的盐。作为有机胺加成盐,可以举例,吗啉、哌啶等的盐。作为氨基酸加成盐,可以举例,甘氨酸、苯丙氨酸、赖氨酸、天冬氨酸、谷氨酸等的盐。
上述鸟氨酸的盐优选使用盐酸盐、枸橼酸盐、苹果酸盐、α-酮戊二酸盐、天冬氨酸盐,也可以是一种其它盐(remaining salts)或者将两种以上上述盐任意组合使用。
本发明的组合物,除了含有鸟氨酸或其盐为有效成分外,还可含有蒜(Allium sativum L.)或蒜提取物。本发明使用的蒜或蒜提取物可以是任意的形式,只要其含有公认的作为对于恢复疲劳有效的成分的蒜氨酸或其分解物蒜素。作为该蒜提取物,也可向例如備前化成株式会社(Bizen Chemical Co.Ltd.)等购买。
本发明的组合物含有蒜或蒜提取物时,鸟氨酸或其盐与蒜氨酸或蒜素的组成比为干燥重量比1∶10000~10000∶1,优选1∶3000~3000∶1,特别优选1000∶1~1∶1000。
本发明的组合物,可任选向鸟氨酸或其盐中添加适宜的适合于各用途的添加剂。
作为该添加剂,可以举例,缬氨酸、亮氨酸、异亮氨酸、精氨酸、赖氨酸、谷氨酰胺、丙氨酸、丝氨酸、甘氨酸、半胱氨酸、苏氨酸等氨基酸等。
本发明的组合物,可作为医药或食品添加剂(以下,也称为本发明的医药或食品添加剂)使用。
本发明的组合物作为医药使用时,可将鸟氨酸或其盐直接给药,但通常以各种制剂的形式提供。
制剂含有作为有效成分的鸟氨酸或其盐,还可含有其它任意治疗用有效成分。此外,这些医药制剂是将有效成分与一种或多种药理学上可使用的载体一起混合,按照制剂学技术领域中已知的任意方法制造的。
制剂的给药途径,优选使用治疗时效果最好的途径,口服给药,或可以举例静脉、腹膜内或皮下给药等胃肠外给药途径,优选口服给药。
作为给药剂型,可以举例,片剂、散剂、颗粒剂、丸剂、混悬剂、乳剂、浸剂、煎剂、胶囊、糖浆、溶液剂、酏剂、浸膏剂(extracts)、酊剂、流浸膏(fluid extracts)等口服制剂、注射剂、滴注剂、乳剂、栓剂等胃肠外制剂中的任一种,口服制剂优选使用。
可向适于口服给药例如糖浆剂这样的液体制剂中添加水、蔗糖、山梨醇、果糖等糖类,聚乙二醇、丙二醇等二元醇,芝麻油、橄榄油、大豆油等油类,对羟基苯甲酸酯(p-hydroxybenzoate ester)类等防腐剂、对羟基苯甲酸衍生物(p-oxybenzoate derivative)(例如对羟基苯甲酸甲酯)或苯甲酸钠等保存剂,草莓香料、薄荷等矫味剂等使其制剂化。
此外,可向适于口服给药,例如可向片剂、散剂以及颗粒剂等中添加乳糖、白糖、葡萄糖、蔗糖、甘露醇、山梨醇等糖类,马铃薯、小麦、玉米等的淀粉,碳酸钙、硫酸钙、碳酸氢钠、氯化钠等无机物,微晶纤维素、甘草粉末(licorice root powder)、龙胆粉末等植物粉末等赋形剂,淀粉、琼脂、明胶粉末、微晶纤维素、羧甲基纤维素钠、羧甲基纤维素钙、碳酸钙、碳酸氢钠、藻酸钠等崩解剂,硬脂酸镁、滑石、氢化植物油、聚乙二醇、硅油等润滑剂,聚乙烯醇、羟丙基纤维素、甲基纤维素、乙基纤维素、羧甲基纤维素、明胶、淀粉浆等粘合剂,脂肪酸酯等表面活性剂,甘油等增塑剂使其制剂化。
适于非口服给药,例如注射剂优选包含鸟氨酸或其盐的灭菌水性剂,所述鸟氨酸或其盐与受试者血液等张的。例如在注射剂的情况下,可用含有盐溶液、葡萄糖溶液或盐溶液与葡萄糖溶液的混合物的载体制备注射用溶液。
此外,这些胃肠外制剂中还可添加口服制剂中所示例的防腐剂、保存剂、矫味剂、赋形剂、崩解剂、润滑剂、粘合剂、表面活性剂和增塑剂等中的一种或多种辅料。
本发明药物组合物中的鸟氨酸或其盐的浓度,根据制剂种类、该制剂给药所期待的效果等进行适当选择,鸟氨酸或其盐,通常为0.1~100重量%,优选0.5~80重量%,特别优选1~70重量%。
本发明药物组合物的给药量和给药次数根据给药形式,患者的年龄、体重、待治疗症状的性质或轻重不同而不同,通常成人一日给药鸟氨酸或其盐50mg~30g,优选100mg~10g,特别优选200mg~3g,一日给药一次至数次。
给药周期没有特别的限制,通常为1日~1年,优选1周~3个月。
本发明的组合物作为食品添加剂使用时,可直接使用鸟氨酸或其盐,也可按照与上述药物制剂同样的方法制成含有鸟氨酸或其盐为有效成分的食品添加剂。
本发明的食品添加剂必要时可与其它食品添加剂混合或溶解于其它食品添加剂中,例如加工制造成粉末、颗粒、小丸、片和各种液体制剂的形式。
含有本发明食品添加剂的饮料或食品(以下简称为本发明的饮料或食品),除了在饮料或食品中添加本发明的食品添加剂外,可按一般饮料或食品的制造方法进行加工制造。
此外,本发明的饮料或食品还可采用例如流化床制粒、搅拌制粒、挤出制粒、旋转制粒、气流制粒、压缩成形制粒、粉碎制粒、喷雾制粒、喷射制粒等制粒方法,锅包衣、流化床包衣、干法包衣等包衣方法,膨化干燥(puffdry)、过剩水蒸气法、泡沫垫(foam mat)方法、微波加热方法等膨化方法,用挤出制粒机或挤出机等挤出方法等制造。
本发明饮料或食品例如可以是果汁类、清凉饮料、茶类、乳酸菌饮料、发酵乳、冰激凌、黄油、奶酪、酸奶酪、加工乳、脱脂乳等乳制品,火腿、香肠、汉堡等畜肉制品,煮熟的鱼酱(蒲鉾)、日本鱼香肠(竹輪)、带有蔬菜的油炸鱼丸(satsumaage)等鱼肉加工制品(fish paste foods),煎蛋饼(だし巻き)、蛋豆腐(tamago-dofu)等蛋制品,甜点、果冻、口香糖、糖果、快餐点心(スナツク菓子)等点心类,面包类、面条类、酱菜类、熏制品、干鱼(干物)、用大酱汤和汤慢火煮肉(佃煮)、盐腌品、汤类、调味料等中的任一形式。
此外,本发明的饮料或食品还可以是例如粉末状食品、片状食品、瓶装食品、罐装食品、软罐头食品(retort food)、胶囊状食品、药片状食品、流食(liquid food)、饮料制品等形式。
本发明的饮料或食品可作为改善疲劳自觉症状用健康食品、功能性食品、营养补食品、特定保健用食品等饮料或食品使用。
本发明的食品添加剂或饮料或食品,为饮料或食品中通常使用的添加剂,例如可添加食品添加剂表示手册(日本食品添加物协会,平成9年1月6日发行)(Japan’s Specifications and Standards For Food Additives(Japan FoodAdditivess Association,issued on January 6,1997)中记载的甜味剂、着色剂、防腐剂、增粘稳定剂、抗氧化剂、显色剂、漂白剂、抑菌剂(anti-fungal agent)、胶基、苦味剂、酶、光亮剂、酸味剂、调味料、乳化剂、增强剂、生产用剂、香料、香辛料提取物等。
向本发明饮料或食品中添加食品添加剂的添加量,根据饮料或食品的种类、摄取该饮料或食品期待达到的效果等进行适宜的选择,通常含有鸟氨酸或其盐0.1~90重量%,优选0.5~80重量%,特别优选1~70重量%。
本发明饮料或食品的摄取量,根据摄取形式、摄取者的年龄、体重等的不同,成人一天通常摄取鸟氨酸或其盐50mg~30g,优选100mg~10g,更优选200mg~3g,一日一次至数次。
摄取周期没有特别的限制,通常为1日~1年,优选1周~3个月。
通过给药或摄取本发明的医药或饮料或食品可以改善疲劳自觉症状。
本发明所述的疲劳自觉症状与疲劳感同义。
疲劳分为生理性疲劳和病态疲劳,本发明的医药或饮料或食品优选用于改善生理性疲劳的自觉症状。
生理性疲劳意味着人维持健康的一种防御反应。具体说,意味着工作、家务、闲暇时运动等日常活动时表现出的疲劳,不仅是肉体上的疲劳,也包括精神上的疲劳。
而病态疲劳是伴随心脏病、支气管哮喘(気管支喘息)、肝炎、贫血、代谢性疾病、肌肉疾病、各种感染症、癌症等基础疾病出现的疲劳症状,以及慢性疲劳综合征、精神原因引起的抑郁症、运动过度综合征(overtrainingsyndrome)等引起的疲劳。
对于可经给药或摄取本发明的医药或饮料或食品而改善的疲劳自觉症状没有特别的限制,优选与困倦和疲倦、注意力难以集中相关症状。
困倦和疲倦相关症状,可以举例,头重、全身无力、下肢无力、打哈欠、头脑迷糊、困倦、眼睛疲劳、站立辛苦(立つていゐのが辛い)等。
注意力难以集中相关症状,可以举例,难以思考、不愿讲话、焦躁、精神涣散、对事情没有兴趣、健忘、易于出错等。
以下为鸟氨酸疲劳自觉症状改善效果的调查试验例。
试验例1
试验如下进行。14名健康的45岁至64岁男女按每组7名分为2组,使其服用实施例1的片剂(含有鸟氨酸的片剂)或比较例1的片剂(不含有鸟氨酸的片剂),1日6片,服用三周进行试验。试验开始时以及试验结束时根据受试者的疲劳自觉症状,用直观模拟标度尺(VAS)法进行评价。
也就是说,将线段的两端记为基准表现,将受试者的情况标记于图1或图2所示询问项目内容相关线段的相应部分。在疲劳自觉症状中,问题1~8为与“困倦和疲倦”相关的指标,问题9~15为与“注意力难以集中”相关的指标。
测定由线段左端至标记点的距离(mm),求算试验前与试验后的差值。将该差值所占试验前值的比例用百分率表示,计算每组的平均值和标准偏差。此外,用鸟氨酸组的平均值减去安慰剂组的平均值作为平均改善率(%)。此外,确认试验开始时两组间无差异。
此外,测定试验开始时和试验结束时7名受试者血氨值,确认各受试者血中氨值没有变动,在正常范围内。
而且,在随机分配下,采用双盲平行组间比较进行试验。两组间的统计学差异,通过试验开始时和试验结束时的差的双侧分布的非配对t检验进行测定。
“困倦和疲倦”相关结果如图3所示。此外,“注意力难以集中”相关结果如图4所示。所有项目中摄取鸟氨酸后显示了改善效果,其中关于“难以思考”项安慰剂组和鸟氨酸摄取组有明显差异。
以上结果表明通过摄取鸟氨酸可以明显改善疲劳自觉症状。
试验例2
健康的20岁至60岁男女27名按每组6~7名分为4组,服用表1的胶囊剂,每天4粒,服用1周进行试验。

*胶囊使用白色纤维素第0号硬胶囊,在空隙处填充结晶纤维素。
*无味蒜提取物中,含有约1%干重的蒜氨酸或蒜素。
对试验开始时和试验结束时受试者的疲劳自觉症状按照试验例1同样的方法进行评价。
其结果,“注意力难以集中”考察项中“焦躁”项,鸟氨酸组的平均改善率为7.2,蒜组的平均改善率为5.1,与其相比鸟氨酸+蒜组为9.6,因此,鸟氨酸与蒜组合得到更明显的改善的效果。
此外,“注意力难以集中”考察项中“精神涣散”项,鸟氨酸组的平均改善率为3.4,蒜组的平均改善率为4.2,与其相比鸟氨酸+蒜组为21.2,鸟氨酸与蒜的组合得到更明显的改善效果。
以下为本发明的实施例。
实施例1
含有鸟氨酸的片剂的制备
将鸟氨酸盐酸盐136.2kg(商品名:L-鸟氨酸盐酸盐,协和发酵工业社制)、微晶纤维素36.0kg(商品名:Avcel FD101,Asahi Kaei Chemicals Co.,Ltd.)、蔗糖脂肪酸酯6.6kg(商品名:DK酯F-20W,第一工业制药社制)、磷酸钙1.2kg(商品名:磷酸三钙,太平化学产业社制)以及β-环糊精20.0kg(商品名:Seldex B-100,日本食品化工社制)用锥形混合器(CB-1200混合器,日本干燥机株式会社制)进行混合。将得到的混合物用辊压缩成形机(VIRGO524SS1AY,菊水制作所社制)在压缩成形压力为10kN下压缩成形,制备直径8mm,250mg的片剂。
实施例2
含有鸟氨酸的肠溶胶囊的制备
将实施例1制备的混合物20kg与0.2kg二氧化硅混合搅拌得到的混合物投入胶囊填充机中,填充明胶制2号硬胶囊20000枚,得到硬胶囊。将得到的硬胶囊表面通过High Coater HCT-48型(Freund Corpoarion)用玉米蛋白溶液包衣,制备20000枚含有鸟氨酸盐酸盐的肠溶胶囊。
实施例3
含有鸟氨酸的肠溶片剂的制备
将实施例1制得的片剂表面通过High Coater HCT-48型(FreundCorpoarion)用虫胶溶液包衣,制备肠溶片剂。
实施例4
含有鸟氨酸的饮料的制备
将鸟氨酸盐酸盐1.28kg(商品名:L-鸟氨酸盐酸盐,协和发酵工业社制)、赤藓醇3kg(日研化学社制)、枸橼酸0.05kg(Kyowa Hi Foods Co.,Ltd.)、人工甜味料3g、香料0.06kg以液温70℃搅拌溶解于水50L中,用枸橼酸调pH至3.3后,将溶液用板式灭菌(plate sterilization)进行灭菌并装入瓶中,然后将瓶进行巴氏灭菌,制成饮料。
实施例5
含有鸟氨酸和蒜提取物的饮料的制备
将鸟氨酸盐酸盐0.64kg、无味蒜提取物0.6kg、赤藓醇3kg、枸橼酸0.05kg、人工甜味料3g、香料0.06kg以液温搅拌溶解于70℃水50L中,用枸橼酸调pH至3.3后,将溶液用板式灭菌进行灭菌并装入瓶中,然后将瓶巴氏灭菌,制成饮料。
比较例1
实施例1的鸟氨酸盐酸盐用等量的乳糖代替,制成不含有鸟氨酸的片剂。
工业适用性
通过本发明,可以提供含有鸟氨酸或其盐为有效成分、安全有效改善疲劳自觉症状用的组合物。
Claims (12)
1.改善疲劳自觉症状用组合物,其中含有鸟氨酸或其盐为有效成分。
2.权利要求1记载的组合物,其中疲劳自觉症状为生理性疲劳的自觉症状。
3.权利要求1或2记载的组合物,其中疲劳自觉症状为与困倦和疲倦、或注意力难以集中相关的症状。
4.权利要求1~3任一项记载的组合物,其还含有蒜或蒜提取物。
5.一种疲劳自觉症状的改善方法,其包括向需要的对象给药或使对象摄取有效量的鸟氨酸或其盐。
6.权利要求5记载的方法,其疲劳自觉症状为生理性疲劳的自觉症状。
7.权利要求5或6记载的方法,其疲劳自觉症状为与困倦和疲倦、或注意力难以集中相关的症状。
8.权利要求5~7任一项记载的方法,其还包括给药或摄取蒜或蒜提取物。
9.鸟氨酸或其盐在制备用于改善疲劳自觉症状的组合物中的用途。
10.权利要求9记载的用途,其中疲劳自觉症状为生理性疲劳的自觉症状。
11.权利要求9或10记载的用途,其中疲劳自觉症状为与困倦和疲倦、或注意力难以集中相关的症状。
12.权利要求9~11任一项记载的用途,其还包括使用蒜或蒜提取物。
