**WARNUNG** Anfang DESC Feld konnte Ende CLMS uberlappen **. PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Theaspiron, dadurch gekennzeichnet, dass man 4(2,6,6-Trimethyl-2-cyclohexen- 1 -yliden)-butan-2-ol mit einer Säure behandelt und das erhaltene Theaspiran durch Oxydation in Theaspiron überführt. 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Säure eine Protonsäure verwendet. Gegenstand der vorliegenden Erfindung ist ein neues, fortschrittliches Verfahren zur Herstellung von Theaspiron. Dieses Verfahren ist dadurch gekennzeichnet, dass man 4(2,6,6-Trimethyl-2-cyclohexen- l-yliden)-butan-2-ol (vgl. die Verbindung III des nachfolgenden Schemas) mit einer Säure behandelt und das erhaltene Theaspiran durch Oxydation in Theaspiron überführt. (Theaspiran (2,6,10, 10-tetramethyl- l-oxa-spiro[4,5]-dec- -6-en) der im nachstehenden Reaktionsschema angegebenen Formel IV stellt eine bekannte Verbindung dar FTetrahedron Letters 1995 (1969)1). Als Säuren sind insbesondere Protonsäuren, wie anorganische und organische Protonsäuren, wie Schwefelsäure, Phosphorsäure, p-Toluolsulfonsäure usw.; oder Lewissäuren wie BF5, SnCl4, ZnCl2, usw. geeignet. Bevorzugt ist p-Toluolsulfonsäure. Die Cyclisierung der Verbindung III zum Theaspiran (IV) kann mit oder ohne Lösungsmittel durchgeführt werden. Geeignet sind inerte Lösungsmittel, wie Hexan, Benzol, Nitromethan, chlorierte Kohlenwasserstoffe, wie Chloroform, usw., Äther, wie Dioxan, usw. Bevorzugt sind Benzol und Toluol. Die Temperatur ist nicht kritisch (Raumtemperatur, oder höhere oder niedere Temperaturen). Da sich Theaspiran bekanntermassen zum Geschmackstoff Theaspiron (vgl. die Formel V des Reaktionsschemas) oxydieren lässt (vgl. z.B. die US-Patentschrift Nr. 3 645 755), ist mit dem erfindungsgemässen Verfahren ein neuer, vorteilhafter Zugang zum Theaspiron eröffnet worden. Die Herstellung von Theaspiron über das (erfindungsgemäss herge steIlte) Theaspiran durch Oxydation des letzteren ist im nachfolgenden Reaktionsschema veranschaulicht. Reaktionsschema EMI1.1 ss-Jonon 4(2,6,6-Trimethyl-2-cyclohexen- - I-yliden)-2-acyloxy-but-2-en R = Acyl wie niederes Alkanoyl, z.B. Acetyl oder Aroyl, z.B. Benzoyl. EMI1.2 2,6,10, l0-Tetramethyl-l- 4(2,6,6-Trimethyl-2-cyclohexen- -oxaspiro[4,5]-dec-6-en - l-yliden)-butan-2-ol EMI2.1 2,6,10, 10-Tetramethyl- 1- -oxaspiro[4,5]-dec-6-en-8-on Der Alkohol III kann, wie im Reaktionsschema gezeigt, aus der Acyloxyverbindung II und diese aus p-Jonon (I) erhalten werden. Die Acyloxyverbindung II kann z.B. mit einem komplexen Hydrid, wie Lithiumborhydrid, Natriumborhydrid, Kaliumborhydrid, Lithiumaluminiumhydrid, usw. in den Alkohol III übergeführt werden. Man arbeitet dabei zweckmässigerweise in einem Alkohol, beispielsweise einem Alkanol, einem Alkohol/Äthergemisch oder einem Äther als Lösungsmitteln. Die Reaktionstemperatur ist nicht kritisch. Bevorzugt ist der Temperaturbereich von ca. -10 C - 80"C. Die Acyloxyverbindung II kann durch Umsetzung von t3- Jonon (I) mit einem Enolacylat erhalten werden. Geeignete Enolacylate sind Isopropenylacetat, Isobutenylacetat, usw. Die Bildung der Verbindung III erfolgt zweckmässigerweise in Anwesenheit katalytischer Mengen von Säuren, z.B. einer der oben angegebenen. Bevorzugt ist p-Toluolsulfonsäure. Das Enolacylat wird zweckmässigerweise im Überschuss, d.h. als Lösungsmittel eingesetzt. Man arbeitet vorzugsweise bei der Rückflusstemperatur des Reaktionsgemisches und entfernt das bei der Bildung der Verbindung II entstehende Keton (Aceton im Falle von Isopropenylacetat) laufend destillativ. Die Formel II soll sämtliche 4 möglichen Stereoisomeren, die Formel III beide Stereoisomeren umfassen. Desgleichen sollen die Formeln IV und V beide diastereomeren Verbindungen, d.h. die beiden Enantiomerenpaare umfassen. Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben. Beispiel a) 96 g ss-Jonon werden in 500 ml Isopropenylacetat gelöst und mit 0,6 g p-Toluolsulfonsäure-monohydrat versetzt. Das Reaktionsgemisch wird unter Inertgasatmosphäre 24 Stunden bei Rückflusstemperatur gerührt. Unter Vakuum wird das überschüssige Isopropenylacetat aus dem Reaktionsgemisch abdestilliert (Temperatur c 50 ), dann letzteres zur Befreiung von Restmengen mehrfach mit Hexan versetzt und wieder eingeengt. Auf diese Weise erhält man 108 g braunrotes 4(2,6,6-Trimethyl-2-cyclohexen- 1 -yliden)-2-acetoxy-but- -2-en (II mit R = Acetyl). UV (Äthanol): : 279 nm, log e: 4, 265. IR(Film): 1755, 1650, 1580, 1370, 1220/1205, 1150/1140, 1040, 1020, 945, 925, 885/875/865, 818 cm-1. NMR (CDCI3 + TMS): 8 = 6,65 - 5,85 (2H, m); 6 = 5,78 (1H, breites t); 6 = 2,20 und 2,15 (3H, je s); 8 = 2,05 (3H, breites S); 8 = 1,85 (3S, schmales m); 8 = 1,28 ppm (6H, S). MS: m/e: 234, Fragmente bei 192, 177, 159, 149, 136, 121, 107, 91, 81, 77, 71, 55, 43 = Basispeak. Die erhaltene rohe Acetoxyverbindung In(108 g) wird in 400 ml Äthanol (96%) gelöst und zu einer Suspension von 20 g Natriumborhydrid in 600 ml Äthanol (96%) innert 10 Min. bei 20-30 unter leichter Kühlung zugetropft. Hierauf wird das Reaktionsgemisch bis zu leichtem Rückfluss aufgewärmt und 15 Min. bei dieser Temperatur gerührt. Das Ende der Reaktion kann an einem spontanen Farbumschlag von dunkelgelb nach zitronengelb erkannt werden. Nach Abkühlung auf Raumtemperatur wird das trübe Reaktionsgemisch auf gesättigte Ammoniumchloridlösung/Eis gegossen und das Gemisch mit Hexan extrahiert. Nach üblichem Neutralwaschen mit Wasser und Trocknen mit wasserfreiem Natriumsulfat wird das Lösungsmittel unter Vakuum abgedampft. Man erhält 96 rohes 4(2,6, 6-Trimethyl-2-cyclohexen- l-yliden)-butan-2ol (III). UV (Äthanol): 4 239 nm (log e = 4). IR(Film): 3300, 1380/1370/1360, 1125, 1085, 950, 880, 830 cm-1. NMR (CDCl3 + TMS): 8 = 5,7 (1H, breites t); 8 = 5,4 (lH,tmitj = 7Hz);8 = 3,9 (IH, m mit J = 6Hz);8 = 2,55 (2H, breites t mit J = 7 Hi); 8 = 1,85 (3H, schmales m); 8 = 1,25 (3H, d mit J = 6 Hz); 8 = 1,23 ppm (6H, S). MS: m/e: 194, Fragmente bei 189, 161, 150, 135 = Basis, 121, 107,93,79,69,55,45,41. 96 g der rohen Verbindung III werden in 1,3 Liter abs. Benzol in Gegenwart von 1 g p-Toluolsulfonsäure-monohydrat 10 Stunden auf Rückflusstemperatur erhitzt. Die Reak tionslösung wird auf kalte, gesättigte Bicarbonatlösung gegossen und mit Hexan extrahiert. Nach dem Neutralwaschen und Trocknen des Extraktes mit Na2SO4 wird das Lösungsmittel im Vakuum abgedampft. Die resultierenden 96 g des braunen öligen rohen Theaspirans IV werden durch Kurzwegdestillation von Vorlauf und Rückstand abgetrennt. Ausbeute: 77 g Verbindung IV, Sdp.: 750/0,2 mm Hg, nD20 = 1,492. IR(Film): 1475, 1455, 1380, 1360, 1285, 1195, 1160/1150, 1130, 1110/1085/1080/1060, 1040, 1005, 990, 975, 930, 910/ 900, 880, 825, 775, 725 cm-1. NMR (CDCl3 + TMS): Isomeres A 8 = 5,25 (IH, schmales m); 8 = 4,1 (1H, breites m); 8 = 1,75 (3H, schmales m); 8 = 1,26 (3H, d mit J = 6 Hz); 8 = 0,95 und 0,88 ppm (je 3H, s). Isomeres B 8 = 5,40 (1H, schmales m); 8 = 4,05 (1H, breites m); 8 = 1,7 (3H, schmales m); 8 = 1,28 (3H, d mit J = 6 Hz); 8 = 1,00 und 0,88 ppm (je 3H, s). MS: m/e: 194, Fragmente bei 179, 151, 135 = Basispeak, 123, 109, 96, 82, 77, 67, 55, 41. b) 5,7 g (30 mMol) Theaspiran werden in 60 ml wasser -freiem tert.-Butanol gelöst und innert 2 Stunden bei 40 mit 120 ml (ca. 30 mMol CrO3) tert.-Butylchromatlösung (150 g CrO3, 400 ml tert.-Butanol, 140 ml Acetanhydrid) versetzt und das Gemisch bei 400 gerührt. Nach 8 Tagen werden weitere 20 ml tert.-Butylchromatlösung zugetropft; dieselbe Menge wird nach weiteren 10 Tagen nochmals zugetropft und nach insgesamt 16 Tagen wird das Reaktionsgemisch aufgearbeitet. Das Reaktionsgemisch wird in 1 Liter Methylenchlorid auf genommen, mit Eis überschichtet und mit 1 Liter Sulfit-/Bi sulfitlösung (40 g NaHSO3, 50 g Na2SO3, 1 Liter Wasser) wäh rend 1 Stunde verrührt. Es wird mit gesättigter NaHCO3-Lö sung und Wasser neutral gewaschen, über MgSO4 getrocknet und zur Trockne verdampft. Man erhält 3,0 g gelbes öl, das an der 30-fachen Menge Silikagel (Körnung 0,063-0,200 mm) mit Hexan/Äther-Gemi schen (5-10% Äther) säulenchromatographisch gereinigt wird; Ausbeute 20% Theaspiron. Sdp.: 880/0,12 mm Hg. UV (Äthanol): ax = 235 mm, (e = 11740). MS: m/e: 208 = M +, 193, 175,152, 110, 96, 82, 69,55,41 IR (Film): 1675, 1630, 1480, 1450, 1390/80/70, 1345, 1310, 1280, 1270, 1160, 1090, 980, 920, 890 cm-1. NMR (CDCl3 + TMS): 1H bei Ï = 5,72 (schmales Quadruplett mit J = 1,5 H2); 1H bei 8 = 4,2 zentriert (Multiplett); 3H bei 8 = 2,01 u. 8 = 1,99 (je Singulett für die zwei Diastereomeren); 3H bei 8 = 1,30 (Dublett mit J = 6 Hz); 6H bei 8 = 0,99 u. 1,02 (Singulette für die gem. Dimethylgruppen).

- PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Theaspiron, dadurch gekennzeichnet, dass man 4(2,6,6-Trimethyl-2-cyclohexen- 1 -yliden)-butan-2-ol mit einer Säure behandelt und das erhaltene Theaspiran durch Oxydation in Theaspiron überführt.2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Säure eine Protonsäure verwendet.Gegenstand der vorliegenden Erfindung ist ein neues, fortschrittliches Verfahren zur Herstellung von Theaspiron.Dieses Verfahren ist dadurch gekennzeichnet, dass man 4(2,6,6-Trimethyl-2-cyclohexen- l-yliden)-butan-2-ol (vgl. die Verbindung III des nachfolgenden Schemas) mit einer Säure behandelt und das erhaltene Theaspiran durch Oxydation in Theaspiron überführt.(Theaspiran (2,6,10, 10-tetramethyl- l-oxa-spiro[4,5]-dec- -6-en) der im nachstehenden Reaktionsschema angegebenen Formel IV stellt eine bekannte Verbindung dar FTetrahedron Letters 1995 (1969)1).Als Säuren sind insbesondere Protonsäuren, wie anorganische und organische Protonsäuren, wie Schwefelsäure, Phosphorsäure, p-Toluolsulfonsäure usw.; oder Lewissäuren wie BF5, SnCl4, ZnCl2, usw. geeignet. Bevorzugt ist p-Toluolsulfonsäure.Die Cyclisierung der Verbindung III zum Theaspiran (IV) kann mit oder ohne Lösungsmittel durchgeführt werden. Geeignet sind inerte Lösungsmittel, wie Hexan, Benzol, Nitromethan, chlorierte Kohlenwasserstoffe, wie Chloroform, usw., Äther, wie Dioxan, usw. Bevorzugt sind Benzol und Toluol.Die Temperatur ist nicht kritisch (Raumtemperatur, oder höhere oder niedere Temperaturen).Da sich Theaspiran bekanntermassen zum Geschmackstoff Theaspiron (vgl. die Formel V des Reaktionsschemas) oxydieren lässt (vgl. z.B. die US-Patentschrift Nr. 3 645 755), ist mit dem erfindungsgemässen Verfahren ein neuer, vorteilhafter Zugang zum Theaspiron eröffnet worden.
